The rate of dry matter accumulation by seeds of Vicia faba L. cv. Minica increases with temperature in the range of 16 to 26°C. The duration of dry matter accumulation decreases with temperature, resulting in a decrease of final seed dry weight. In this study we test the hypothesis that a diffusion barrier for O2, located in the seed coat, inhibits seed respiration and growth. The rate of O2 uptake of intact seeds and of excised embryos and seed coats (separated seeds) was measured in air and buffer at 16, 20, and/or 26°C at various O2 concentrations and developmental stages. Oxygen uptake rates of intact seeds in buffer were only 9 to 15% of those in air. In buffer, the respiration rate of intact seeds decreased at a pO2 below air saturation (21 kilopascals), whereas separated seeds showed a decline of O2 uptake only below 80% of air saturation. In air, embryo excision had no effect on the sensitivity of seed respiration to pO2, at both 20 and 26°C. In air at 20°C, separated and intact seeds showed similar rates of O2 uptake. Oxygen uptake by intact seeds, both halfway and beyond the linear growth phase, showed a temperature coefficient Q10 of 2.3 and was insensitive to pO2 in the range of 80 to 100% of ambient. These results indicate that V. faba seed respiration in air is not limited by the diffusion of O2 into the seed.  相似文献

After learning how to recognize the parts of a cell, differentiate between types of cells, and identify the nucleus, the students study blood samples from both familiar and “unknown” animals. Oxygen Diffusion from the Roots of Woody Species   总被引:4,自引:0,他引:4  
Application of the‘polarographic’ technique for studying oxygen diffusion from roots has provided preliminary results which establish that oxygen passes through the woody species Salix atrocinerea Brot., Salix fragilis L., Salix repens L., and Myrica gale L., in the gaseous phase as it does in other wetland species. Entry into the shoots occurs through the bark directly above the water table and in the willow cuttings the effective length of shoot for gas intake was the basal three centimetres above the water table. The length of shoot involved was longer in Myrica gale and the roots were of the normal (un-nodulated) type. Trials on Alnus glutinosa (L.) Gaertn. provided no conclusive results and it is thought that this was perhaps because measurements were on nodulated roots only.  相似文献

The supply of oxygen to proliferating cells within a scaffold is a key factor for the successful building of new tissue in soft tissue engineering applications. A recent in vivo model, where an arteriovenous loop is placed in a scaffold, allows a vascularising network to form within a scaffold, establishing an oxygen source within, rather than external, to the scaffold. A one-dimensional model of oxygen concentration, cell proliferation and cell migration inside such a vascularising scaffold is developed and investigated. In addition, a vascularisation model is presented, which supports a vascularisation front which moves at a constant speed. The effects of vascular growth, homogenous and heterogenous seeding, diffusion of cells and critical hypoxic oxygen concentration are considered. For homogenous seeding, a relationship between the speed of the vascular front and a parameter defining the rate of oxygen diffusion relative to the rate of oxygen consumption determines whether a hypoxic region exists at some time. In particular, an estimate of the length of time that a fixed point in the scaffold will remain under hypoxic conditions is determined. For heterogenous seeding, a Fisher-like travelling wave of cells is established behind the vascular front. These findings provide a fundamental understanding of the important interplay between the parameters and allows for a theoretical assessment of a seeding strategy in a vascularising scaffold.  相似文献

Characterization of the Resistance to Oxygen Diffusion in Legume Nodules   总被引:1,自引:0,他引:1  
A method for characterizing the resistance to oxygen diffusionin legume nodules has been developed. This is based on the assumptionsthat diffusion can be described using a simple resistance analogueand that the respiratory response of the bacteriod-containingcells to external oxygen concentrations can be analysed as adiffusion-limited process. Applying this analysis to experimentaldata from infact white clover plants allowed the total diffusionresistance to be separated into (a) a minimum resistance and(b) the extent to which this resistance can be increased. Whenthe carbohydrate status of the nodules was reduced by dark treatments,the minimum diffusion resistance increased, and after 24–28h darkness equalled the maximum resistance. Of production by homogenates and isolated membranes of E. coli has been examined. Approximately one-fourth of the O2-generated by extracts in the prescence of NAD (P) H is attributable to the membranes. The autoxidizable membrane component is a member of the respiratory chain, since O2-production is NADH-specific, amplified by cyanide, and absent from membranes lacking the respiratory NADH dehyd-rogenase. Other respiratory substrates (succinate, I -phosphoglycerol, D-lactate. and L-lactate) supported Or production at efficiencies between 3 and 30 O2-released per 10.000 electrons transferred, under conditions of substrate saturation.

Membranes from quinoneless mutants quantitatively retain the ability to evolve O2-. indicating that the dehydrogenases are the sites of O2-production. Relative O2-production was greater at low substrate concentrations, probably reflecting the facilitation of unpairing of electrons that may occur when enzymes with multiple redox centers are only partially reduced.

Respiration rate, cell volume, rates of membraneous and cytosolic O2-production, and SOD levels were used to calculate a steady-state concentration of O2-between 10--10 and 10--9 M in well-fed, aerobic, SOD-proficient cells.  相似文献

Oxygen Diffusion Pathways and Nitrogen Fixation in Legume Root Nodules   总被引:1,自引:0,他引:1  
The theoretical analysis presented in this paper suggests thatthe nature of the diffusion pathway from the surface of a noduleto the infected zone depends on the morphology of the nodule;in particular the cross-sectional area of the intercellularspaces in the inner cortex. If the diffusion barrier containscontinuous pathways there will be no pressure difference betweenthe atmosphere and the infected zone. The conditions under whichthe intercellular spaces of the inner cortex could be air-filled,water-filled, or a combination of both are explored. An experimentto resolve this issue is suggested. Information obtained usingcryo-scanning electron microscopy and oxygen electrode datahave been used to illustrate various points. Expressions arederived for the diffusion resistance of the nodule and its componentparts. To provoke further discussion a simple mechanism forthe control of diffusion is described in general terms. Models of diffusion MRI within a voxel are useful for making inferences about the properties of the tissue and inferring fiber orientation distribution used by tractography algorithms. A useful model must fit the data accurately. However, evaluations of model-accuracy of commonly used models have not been published before. Here, we evaluate model-accuracy of the two main classes of diffusion MRI models. The diffusion tensor model (DTM) summarizes diffusion as a 3-dimensional Gaussian distribution. Sparse fascicle models (SFM) summarize the signal as a sum of signals originating from a collection of fascicles oriented in different directions. We use cross-validation to assess model-accuracy at different gradient amplitudes (b-values) throughout the white matter. Specifically, we fit each model to all the white matter voxels in one data set and then use the model to predict a second, independent data set. This is the first evaluation of model-accuracy of these models. In most of the white matter the DTM predicts the data more accurately than test-retest reliability; SFM model-accuracy is higher than test-retest reliability and also higher than the DTM model-accuracy, particularly for measurements with (a) a b-value above 1000 in locations containing fiber crossings, and (b) in the regions of the brain surrounding the optic radiations. The SFM also has better parameter-validity: it more accurately estimates the fiber orientation distribution function (fODF) in each voxel, which is useful for fiber tracking.  相似文献

Proportions of respiring bacteria determined with a 2-(p-iodophenyl)-3-(p-nitrophenyl)-5-phenyl tetrazolium chloride dye-epifluorescent technique were significantly elevated in the 300-μm surface layer of a salt marsh estuary. Almost all the detectably respiring bacteria in the particle-laden surface layer and a significant proportion in subsurface waters were attached to particles.  相似文献

A method is reported that combines the microscopic determinations of specific, individual, respiring microorganisms by the detection of electron transport system activity and the total number of organisms of an estuarine population by epifluorescence microscopy. An active cellular electron transport system specifically reduces 2-(p-iodophenyl)-3-(p-nitrophenyl)-5-phenyl tetrazolium chloride (INT) to INT-formazan, which is recognized as opaque intracellular deposits in microorganisms stained with acridine orange. In a comparison of previously described sample preparation techniques, a loss of >70% of the counts of INT-reducing microorganisms was shown to be due to the dissolution of INT-formazan deposits by immersion oil (used in microscopy). In addition, significantly fewer fluorescing microorganisms and INT-formazan deposits, both ≤0.2 μm in size, were found for sample preparations that included a Nuclepore filter. Visual clarity was enhanced, and significantly greater direct counts and counts of INT-reducing microorganisms were recognized by transferring microorganisms from a filter to a gelatin film on a cover glass, followed by coating the sample with additional gelatin to produce a transparent matrix. With this method, the number of INT-reducing microorganisms determined for a Chesapeake Bay water sample was 2-to 10-fold greater than the number of respiring organisms reported previously for marine or freshwater samples. INT-reducing microorganisms constituted 61% of the total direct counts determined for a Chesapeake Bay water sample. This is the highest percentage of metabolically active microorganisms of any aquatic population reported using a method which determines both total counts and specific activity.  相似文献

The Schwann cell, basement membrane, and connective tissue layers that surround the squid giant axon and constitute barriers to diffusion, were modeled in a number of ways to analyze various experimental results. The experiments considered are (a) the time-course of the potassium concentration in the space between the Schwann cell and the axon membrane (from now on referred to as the F-H space) after an initial loading, (b) the time-course of sodium concentration in the F-H space after a sudden change in the sodium concentration in the external fluid; (c) the time-course of the concentration of tetrodotoxin (TTX) or saxitoxin (STX) in the F-H space after a sudden change in external concentration, including (or not) the effects of specific binding of TTX or STX to sites on the axon membrane and nonsaturable binding to sites in the F-H space or in the spaces (clefts) between Schwann cells; (d) the effects of the F-H space, clefts, and diffusion into the clefts from the outside (from now on referred to as convergence into the clefts) on the measured series resistance.

The analysis shows that (1) in no case is it necessary to include the effects of the convergence into the clefts from the outside; (2) in case a, the basement membrane, connective tissue layers, and the unstirred layer may be neglected, i.e., the clefts are rate limiting; (3) in case b the clefts may be neglected, i.e., the unstirred layer is rate limiting; (4) in most cases the clefts may be replaced by an equivalent thin diffusion barrier.

A method for direct identification and quantitative measurementsof mixed or pure gases diffusing through seed coats was devisedto test the hypothesis that the dormancy of Xanthium pennsylvanicumseeds is caused by oxygen-impermeable seed coats. The diffusionof oxygen through seed coats of X. pennsylvanicum was shownto obey Fick's first law. Oxygen diffused through the lowerand upper seed coats at the same rate. Imbibed lower and upperseeds showed essentially equal oxygen uptake rates before radicleemergence. This uptake was lower than the rate at which oxygencan diffuse into the seed. Therefore upper seeds are not dormantbecause of seed coat restriction of oxygen diffusion. The relationshipsof oxygen with other factors involved in seed dormancy are discussed.  相似文献

Three rice varieties, cv. Norin 36, cv. Norin 37 and cv. Yubae, were grown in a loam with a 20 cm water-table which gave aerobic conditions to a depth of not less than 15 to 17 cm. Under these conditions Norin 36 grew more vigorously and tillered more frequently than the other two varieties. The rates of oxygen diffusion at 23°C from roots up to 11 cm in length were however appreciably lower for Norin 36 (4.3 × 10?8g · cm?2 of root surface · min?1) than for Norin 37 or Yubae (c. 7.8 × 10?8g). A considerable increase (up to 200 %) in the oxygen diffusion rate (ODR) from the roots occurred if they were cooled to 3°C, and at this temperature differences in ODR between the varieties were not significant. For a purely physical system, because of the decrease in the diffusion coefficient of oxygen in water, and, the increase in oxygen solubility, a drop of c. 20 % in ODR should accompany the above 20°C drop in temperature. A 16 % drop was recorded for artificial ‘roots’ under these conditions. It was concluded that respiratory activity at the higher temperature must have been responsible for the low readings and intervarietal differences observed at 23°C. By increasing the 3°C values by 25 % a mean value of 14.2 × 10?8g · cm?2 of root surface · min?1 was recorded for the three varieties, being the probable ODR at 23°C in the absence of a respiratory factor. Calculations show that respiratory activity removed enough oxygen to reduce the ODR for Norin 36 by more than 9 × 10?8g, and for Norin 37 and Yubae by c. 6.7 × 10?8g · cm?2 of root surface · min?1. Anatomical investigations showed that cortical breakdown was always extensive at 4 to 4.5 cm from the apex of the roots. In some cases however breakdown had not occurred in the basal segment of the root. No opinion could be formed as to whether differences in the amount of cortical breakdown between the varieties might have occasioned the respiratory differences observed. An interesting feature of the root anatomy was the failure of breakdown in those regions of the roots through which lateral roots emerged.  相似文献

The oxygen diffusion resistance of Lupinus albus (L.) cv. Multoluparoot nodules was increased by subjection to short-term stresses;lowering rhizosphere temperature from 25 to 16 °C (2 h),detopping plants (3 h), darkening plants (21 h) or exposingroots to 20 mol m–3 KN03 for 2, 4 or 6 d. Microscopicobservations and measurements showed that this resulted in thearea of open intercellular spaces within the inner cortex beingreduced due to both cell expansion and increased productionof an occluding glycoprotein. Electrophoretic and Western Blotanalysis using the monoclonal antibodies MAC236 and MAC265 showedtwo distinct glycoprotein antigens with molecular weights of240 and 135 kDa, respectively. Both antigens are localized withinintercellular spaces of the inner cortex. The amount of glycoproteinwas determined using either ELISA, with MAC265, or quantificationof immunolabelling with MAC236. This immunolabelling also localizedthe glycoprotein within globules adhering to the inside of theinner cortical cell walls. Root nodules of Lupinus albus (L.) cv. Multolupa were subjectedto short- and medium-term stresses by lowering rhizosphere temperaturefrom 25 to 16°C (2 h), detopping plants (3 h), darkeningplants (21 h) or exposing roots to 20 mol m–3 KNO3 for4 d. All experimental treatments produced increases in oxygendiffusion resistance, compared with control plants. These correlatedwith structural changes in the nodule cortex, which is describedin detail for the first time. The most noticeable change isthe occlusion of intercellular spaces by a glycoprotein whichwas identified using the monoclonal antibody MAC236. This glycoproteinwas also found surrounding bacteria in intercellular spacesof the cortex of control nodules.
