A laboratory experiment was performed to study the responsiveness of selected condition measures to starvation in herring, Clupea harengus, larvae in relation to temperature and ontogeny. The larvae at two intervals of development, i.e. stage 1 larvae with initial exogenous feeding and stage 2 larvae with established feeding prior to notochord flexion, were reared at three temperatures (5, 8 and 11°C) and subjected to sub-lethal durations of starvation. Temporal changes in standard length, dry weight, DNA concentration (% of dry weight), RNA concentration (% of dry weight), Fulton's condition factor (CF) and RNA/DNA were assessed and compared with fed controls. Starvation led to decreases in dry weight, CF, RNA concentration and RNA/DNA, while it led to an increase in DNA concentration. Higher responsiveness to starvation was observed at higher temperatures, and the magnitude of the changes was higher in stage 2 larvae. The shortest latency in starvation response was found with respect to RNA/DNA which was length independent in the size range studied. RNA/DNA was also significantly related to average DNA growth rate, and the model for DNA growth rate was, SGRDNA=4.49 RNA/DNA + 7.14 T – 0.42 T2 – 37.5 ; n=32, r2=0.85, p < 0.001). While the model seemed to adequately represent the average temperature dependent DNA growth, a relatively low classification success made it unsuitable for depicting individually starving larvae. Critical levels in DNA concentration can be used (2.2% for stage 1 larvae, 2.9% for stage 2 larvae) to differentiate starving larvae (after 3 – 5 days) from feeding larvae. RNA/DNA was the most sensitive and suitable condition index studied in detecting early starvation of herring larvae.  相似文献

研究通过对岩原鲤仔鱼在饥饿和再投喂条件下其生存、生长率、RNA/DNA和RNA/蛋白质比率的测定,评估了仔鱼对饥饿的耐受能力和恢复能力。在(19.5±0.5)℃水温下,将岀膜后第16天的岩原鲤仔鱼随机分成6个组:1个持续投饲对照组,实验组分别禁食1、2、3、4、5d后再投喂,实验共进行10d。每天分别从各组取9尾鱼测定体重、体长、RNA、DNA、蛋白质含量。实验结果显示,饥饿处理组仔鱼存活率和以上各项生长指标均随饥饿时间的增加而下降,在恢复投喂后均表现不同程度的补偿生长,其中饥饿1、2、3d的仔鱼在恢复投喂后显示出完全补偿生长,几乎弥补了饥饿所产生的影响,平均终体重与对照组比较无显著差异。饥饿4、5d的仔鱼显示部分补偿生长,恢复投喂只少量减轻了饥饿的影响,平均终体重与对照组相比存在显著差异。饥饿1、2、3d的仔鱼和4、5d的仔鱼在恢复投喂后分别需要1—2d和4d时间才能达到与对照组无显著差异水平。仔鱼生长率变动范围从0.59%到8.00%WW/day,仔鱼RNA/DNA比率、RNA/蛋白质比率与生长率的回归方程为:GR=3.63RNA/DNA 1.74(R2=0.80)和GR=120.14RNA/Protein 2.33(R2=0.31),两种比率均与生长率呈显著线性相关,RNA/DNA比率对生长变化的拟合度更好。结果表明,仔鱼阶段食物缺乏很可能是影响岩原鲤仔鱼存活、生长的主要因素。RNA/DNA更适合作为评定岩原鲤仔鱼营养条件和生长的指标。  相似文献

Starved Japanese flounder Paralichthys olivaceus larvae were characterized by relatively lower levels of RNA content throughout their early life stages. Significant differences in the RNA: DNA ratios were found between fed and starved fish, and appeared to increase as starvation proceeded. Ontogenetic changes in RNA: DNA ratios were clearly observed during metamorphosis, especially decreasing during the period from the late-metamorphic to postmetamorphic stages. The criteria established from these laboratory experiments, were applied to the nutritional condition of wild larvae and juveniles collected in Wakasa Bay, Sea of Japan in 1994 and 1995 by measuring RNA and DNA content. Starved fish were mainly found in stage I (settling stage) fish during the late season of settlement in 1995. This suggests that starvation could be associated with settlement in Japanese flounder.  相似文献

RNA/DNA ratio is a useful and reliable indicator of the nutritional status of fish larvae and juveniles. In order to assess the nutritional status of field-caught larval Pacific bluefin tuna Thunnus orientalis (Temminck et Schlegel), starvation experiments of hatchery-reared larvae were conducted and changes in the RNA/DNA ratio of fed and starved larvae were analyzed. Starvation experiments were conducted every 3 days after first feeding. The survival rate of Pacific bluefin tuna larvae ranged 10-50% after 1 day of starved conditions and growth retardation was observed immediately. These results suggest that Pacific bluefin tuna larvae have a very low tolerance to starvation. The RNA/DNA ratios of fed larvae were approximately 2.0-4.0. On the other hand, the value of starved larvae significantly decreased to 1.0-3.0. The nutritional status of 3 cohorts of field-caught tuna larvae collected in the northwestern Pacific Ocean was examined based on the value of the RNA/DNA ratio of the 1 day starved larvae. 4.35-25.77% of the cohorts were regarded as the “starving condition”, which was negatively correlated to the ambient prey densities. These findings suggest that the nutritional condition of larval Pacific bluefin tuna was influenced by the ambient prey density, and starvation itself and starvation-induced predation could greatly contribute to mortality in the larval period of Pacific bluefin tuna.  相似文献

In order to clarify the nutritional conditions of larval milkfish in the surf zone, the following parameters were examined: 1) DNA and RNA content and RNA/DNA ratio of fed and unfed larvae collected from the surf zone and reared in the laboratory; 2) survival rate of the unfed larvae; and 3) total length, otolith increment counts and RNA/DNA ratio of wild larvae collected daily from the surf zone. The DNA and RNA content of the unfed larvae decreased, but increased in fed larvae. The RNA/DNA ratio decreased in unfed larvae, whereas in the fed larvae it decreased for the first three days after capture and increased thereafter. These results indicated that the values of DNA and RNA content and RNA/DNA ratio could be used as an indicator of nutritional condition of milkfish larvae after 6 days of starvation. Although total length of the wild-larvae did not show serial changes, their otolith increment counts showed continuous increases, indicating that the larvae sojourned in the surf zone for several days. In the same period, RNA/DNA ratios of the wild larvae decreased continuously, the ratios of larvae with fewer otolith increment counts being relatively higher than those of larvae with greater increment counts. Based on these results, the milkfish larvae remaining in the surf zone were concluded as being under insufficient nutritional conditions.  相似文献

Nutritional condition and vertical distribution of Baltic cod larvae   总被引:5,自引:0,他引:5  
Newly hatched Baltic cod Gadus morhua larvae are typically found at depths >60 m. This is a region of low light and prey availability, hence generating the hypothesis that larvae have to migrate from hatching depth to the surface layer to avoid starvation and improve their nutritional condition. To test this hypothesis, Baltic cod larvae were sampled during the spawning seasons of 1994 and 1995 with depth-resolving multiple opening/closing nets. Each larva was aged by otolith readings and its RNA/DNA ratio was determined as a measure of nutritional condition. The RNA/DNA ratios of these larvae aged 2-25 days (median 10 days) ranged from 0.4 to 6.2, corresponding to levels exhibited by starving and fast-growing larvae in laboratory calibration studies (starvation, protein growth rate, Gpi= -12.2% day−1; fastgrowing larvae, Gpi=14.1%day−1) respectively. Seventy per cent of the field caught larvae had RNA/DNA ratios between the mean values found for starving and fed laboratory larvae. Only larvae aged 8-11 days had higher mean RNA/DNA ratios above 45 m than below ( t -test, P<0.05). However, the instantaneous protein growth rates were significantly higher for all larval age groups in the surface layers ( t -test, P<0.05). Starving larvae were found in all depths sampled (10-85 m), whereas growing larvae (positive Gpi) were restricted to samples taken shallower than 45 m. These superior growth rates above 45 m corroborate the hypothesis and imply that migration to the shallow water layers is a prerequisite for good nutritional condition, growth and survival of Baltic cod larvae. The frequent occurrence of cod larvae older than 8 days in the deep water in poor condition suggests that a proportion of the larvae will die from Starvation in the deep layers of the Baltic Sea.  相似文献

Hatchery reared larval striped bass, Morone saxatilis , 8-days-post-hatching were subjected to various feeding/starvation regimes over a period of 14 days.
Batches of larvae from each treatment were sampled over the 14-day period and subdivided for determination of notochord length and RNA:DNA ratio. The best growth was found in fully fed F1000 larvae (exposed to 1000 Artermia nauplii l−1), which reached 8.2 mm after 11 days and 9.6 mm after 14 days. Starved animals after 11 days had notochord lengths of 4.9 mm. Growth curves from feeding-delayed larvae indicated that animals fed after up to 5 days starvation were capable of complete recovery. F100 larvae (exposed to 100 Anemia nauplii 1−l) had a slower growth rate than F1000 larvae, reaching a notochord length of 7.3 mm after 14 days. RNA:DNA ratios over time closely followed notochord growth curves, with clear differences between starved, F100 and F1000 larvae being established after only 2 days. Equilibrium RNA:DNA ratios of 3.0 and 2.25 were established in F1000 and F100 larvae, respectively, 6.8 days after the beginning of the experiment. The average lag time between a change from the starved to the fed condition and a change in RNA:DNA ratio as determined by the divergence of the nucleic acid curve from the starved condition was 0.66 days.
In treatments where starvation followed various periods of feeding, larvae regressed in notochord length such that the final length at 14 days reflected the degree of feeding. RNA:DNA ratios in these animals again closely followed growth curves with a lag time of 0.81 days.
It was concluded that RNA:DNA ratios provided very accurate indices of growth in striped bass larvae which were highly sensitive to feeding status.  相似文献

Fish larvae were collected monthly between March and September 1997 in the Mira and Guadiana estuaries (southern Portugal). Hydrological parameters were registered and zooplankton samples were obtained simultaneously. Densities of fish larvae (ind·100 m–3) were calculated from 211 samples and larval nutritional condition measured as RNA/DNA ratios were obtained for 346 individuals, using a fluorimetric method for nucleic acid quantification. Correlating variables were further studied using multiple regression analysis in order to assess the relative importance of abiotic and biotic factors affecting within-year trends in abundance and nutritional condition of estuarine fish larvae. Results indicated that: 1) the abundance of fish larvae seems conditioned by temperature and predation; and 2) their nutritional condition is dependent on temperature and prey availability. Temperature is an important variable structuring estuaries and therefore conditions the behaviour and physiology of fish larvae. Furthermore, the co-occurrence of predators and larvae might be related to similar feeding patterns or comensalism. Whenever feeding conditions are suitable, they usually determine enhanced growth and nutritional condition. However, predation seems to control this latter relationship through its effect on larval mortality.  相似文献

Six condition indices based on RNA, total soluble protein and two metabolic enzymes [lactate dehydrogenase (LDH) and citrate synthase (CS)] were analysed in muscle tissue of individual larvae, post-flexion reared sea bass Dicentrarchus labrax using DNA and total soluble protein as standards for size. In addition, the effect of 2 days of food deprivation on the cell proliferation rates was assessed. The RNA:DNA best reflected short-term changes in feeding conditions. If standardized by DNA content, LDH activity was a better indicator of condition than any other index but RNA:DNA. Further, the analysis of cell proliferation rates in muscle from 26 day-old larvae proved useful in distinguishing continuously fed larvae from individuals subjected to 2 days of fast.  相似文献

Experiments were carried out to evaluate the influence of rearing temperature and food concentration (20 and 30 °C, 1×105 and 2×105 cells ml−1) on the starvation threshold and nucleic acid content of the larvae of Balanus amphitrite. The larvae were also field-reared using micro-enclosures. Laboratory-reared larvae were larger in size than the field-reared larvae. An increase in size, DNA content and instar index of the starved II instar larvae was observed indicating that the absence of food may not be fatal to this early instar. The temperature at which larvae were raised and the food concentration had variable influence on the capacity to withstand starvation. Exposure to increased temperatures during starvation eliminated the effect of doubling food concentration during their feeding period prior to starvation. The larvae reared at 20 °C had comparatively lower nucleic acid content. The laboratory-reared larvae had ca. 1.7 times greater RNA:DNA ratio than larvae raised at comparable temperature in the field.  相似文献

Nucleic acid assessments and especially the RNA/DNA ratio are used widely as indices of growth rate or nutritional condition of larval fish. Methodological aspects are considered with the aim of defining a most reliable analytical procedure. Laboratory calibrations can give conflicting results, but general and common features may be drawn from a global examination. A high variability of RNA content, and thus RNA/DNA ratio, was revealed by estimates at the individual level and seems to be more related to feeding condition in late larval or juvenile stages than in yolk-sac and first-feeding larvae. An alternative is suggested, based on the relative DNA content which appears more stable and sensitive to starvation during these early stages. The consistency of such a pattern is strongly supported by findings about regulation of ribosomal RNA content of tissues, and especially white muscle, by nutrition. Determination of a threshold founded on a simple observation of morphological development stages is proposed as a base to define the best way to assess the larval nutritional condition. Some field applications are reviewed briefly. Finally an appraisal of the advantages and drawbacks of the different nucleic acid-based indices is attempted, which permits delineation of some prospects for future research.  相似文献

The Argentine hake, Merluccius hubbsi, is one of the most important fishing species in the Argentine Sea due to its great abundance and high-quality meat. The study of the nutritional condition is widely used to determine the physiological state of the fish larvae and to estimate their survival possibilities. The larval nutritional condition reflects the environmental conditions to which they have been exposed and represents a useful instrument to determine favourable nursery areas. It also provides tools for the comprehensive management of a population subjected to fishing exploitation. This study aimed to determine potential differences in the nutritional condition of M. hubbsi larvae from the two fishing stocks (Northern and Southern) of the species. The authors assessed the nutritional condition of larvae captured during the 2012 main reproductive peak in the nursery areas of each population. Two different methodologies were applied: a morphometrical approach, by recording five morphometric variables, and a biochemical technique, employing the RNA/DNA index (RDs) and its derived index of growth performance. The morphometrical indexes did not show differences in the larval condition between both stocks. Nonetheless, the RDs index did detect differences in the nutritional condition of larvae from different stocks. The RDs index of larvae in pre-flexion and flexion stages showed significant differences between stocks, indicating that these stages are more susceptible to starvation. The results suggest that the biochemical indexes prove to be more sensitive than the morphometric indexes to detect slight differences in hake larvae nutritional condition. The scope and limitations of these techniques for the analysis of the nutritional condition of larvae are discussed.  相似文献

Synopsis The relation between the feeding and mortality of fish larvae is mediated not only by starvation but also by their growth. In this work, daily otolith ring deposition was analysed in laboratory-reared larvae of Odontesthes microlepidotus (Jenyns), and their growth was studied in relation to ring number and otolith diameter. The nutritional condition of these larvae was assessed by means of morphometric comparisons between fed and starved larvae in order to develop a tool to evaluate the nutritional status of larvae in breeding areas. Changes in body shape have been found as a consequence of starvation. No dependence was found between mean hepatocyte nuclear area and fasting.  相似文献

The small-scale variability in condition and growth of dab Limanda limanda larvae was investigated in relation to a tidal-mixing front (Manx West Coast front) off the west coast of the Isle of Man, Irish Sea. RNA:DNA ratios and otolith microincrement analysis were used to estimate nutritional status and recent growth rates of larvae captured on 5 May 1999 in mixed, frontal and stratified waters associated with this front. RNA:DNA ratios were significantly higher at the Manx West Coast front compared to mixed waters, although recent growth rates did not vary significantly with water column stability. More favourable feeding conditions associated with frontal waters from previously reported large-scale investigations in the Irish Sea were reflected on a smaller spatial scale. Although frontal accumulation of dab larvae was not observed in the current study, dab larvae found in the stratified and frontal waters were significantly larger than those captured in the mixed waters. This pattern could suggest an offshore movement of dab larvae into more favourable waters as they develop.  相似文献

Understanding the nutritional condition and survival of fish larvae is of primary importance in mass larva culture because intensive mortality is concentrated during the larval period. In order to estimate growth and nutritional status based on biochemical indices of slime flounder, Microstomus achne, larvae reared under starved and fed conditions in the hatchery for 58 days and the changes of RNA, DNA, and protein contents were described with the progress of growth and developmental. DNA contents increased gradually throughout the experimental period until 12 and 58 days post‐hatching (DPH) in starved and fed groups respectively. Although they fluctuated and decreased around 12 and 46 DPH, the RNA contents of the fed group increased gradually after hatching; however, in the starved group, they decreased after yolk absorption and 7 DPH. Subsequently, the RNA/DNA ratios in the starved group remained constant until 6 DPH and then decreased rapidly. In the fed group, this decreased slightly from hatching to 14 DPH, then increased gradually until the end of the experiment, except at the lower level of around 46 DPH. Namely, temperature shocks (around 14 DPH) and the dramatic changes in body shape (around 46 DPH) were accompanied by the decrease of the RNA/DNA ratios. Total protein/DNA in both groups decreased rapidly during yolk absorption 0–7 DPH, then decreased continuously until death in the starved group; in the fed group total protein/DNA increased again with feeding. It is suggested that the changes in these biochemical values reflect yolk absorption, feeding, morphogenetic changes, growth, and environmental conditions.  相似文献

Larvae of Coregonus spp. were reared in the laboratory under different temperature (4, 6 and 8°C) and feeding conditions ( ad libitum , limited, with starvation intervals). Their RNA/DNA ratios were determined with a highly sensitive fluorescence technique. After resorption of their yolk reserves (about 2 weeks after hatching), well fed larvae (RNA/DNA >2.5) could be significantly distinguished from larvae reared under limited food supply (RNA/DNA < 2.5), both at the 6 and 8°C levels. At 4°C, no differences due to the feeding regime were found. During a second series of experiments, larvae were affected by an intestinal disease, which was caused by the ingestion of unsuitable copepod plankton. This disease provoked high mortality, decreased growth and RNA/DNA ratios which were almost as low as in temporarily starved larvae from the first series. Coregonid larvae sampled in Lake Constance during spring 1990 showed RNA/DNA ratios which were unexpectedly low when judged on the basis of mean body length and average ambient temperature. It was obvious from macroscopic observations that some of these wild larvae were severely attacked by the intestinal disease. The low RNA/DNA ratios in field samples are, therefore, interpreted as a sublethal result of this disease.  相似文献

【目的】分析家蚕Bombyx mori受饥饿胁迫后的蛋白质谱变化,探索其耐受饥饿的机理。【方法】以家蚕品种932为实验材料,利用双向电泳和质谱技术检测5龄起蚕经过24 h饥饿胁迫的蛋白质谱差异变化,利用荧光定量PCR技术分析BmLp-c 6的转录表达。【结果】经比对饥饿蚕和正常取食蚕的血淋巴蛋白谱,饥饿蚕有62个特异蛋白点。蛋白点的等电点在4.22~6.98之间,分子量分布在20.81~144.69 kDa间。选取只在饥饿时出现的特异蛋白点No. 7111进行质谱鉴定,根据其氨基酸序列进行引物设计,获得了目的基因BmLp-c 6,经与载体pET-21d(+)连接重组后,成功获得诱导表达。经实时荧光定量PCR分析,当5龄起蚕受到饥饿胁迫影响时,BmLp-c 6基因在血淋巴中大量转录表达,但在中肠中的转录表达水平却极低。【结论】家蚕5龄起蚕在饥饿胁迫下,血淋巴中的蛋白质谱发生变化,BmLp-c 6会大量转录表达。  相似文献

Abstract

One of the key factors affecting larval survival is food availability. This study investigated the influence of starvation on the nutritional condition of zoea I larvae of Pleuroncodes monodon. Experimental treatments with differential periods of starvation and subsequent feeding (point of no return: PNR) in addition to treatments with differential periods of feeding and subsequent starvation (point of reserve saturation: PRS) were used to quantify larval survival and the occurrence of lipid droplets in the hepatopancreas. Larval survival differed significantly depending on the starvation and feeding treatment administered. A high percentage of survival was found for the starvation treatment until day 1 (S1-PNR), for the feeding treatment until day 4 (F4-PRS), and for the continuously fed control groups (FC). Survival was minimal for the starvation treatment lasting until day 7 (S7-PNR) and for the continuously starved control groups (SC). In turn, similar tendencies were observed in the utilization of energy reserves; the lipid droplets significantly decreased throughout the PNR treatment, while the presence of lipid droplets gradually increased during the PRS treatment. All these larval condition parameters can be used in fishery models of population dynamics, which estimate the nutritional status of the offspring and their effects on survival.  相似文献

The nutritional condition and protein growth rates of Japanese temperate bass larvae and juveniles were studied in relation to prey distribution and feeding habits in the nursery grounds in Chikugo estuary, Ariake Sea, Japan. Samples were collected from a wide spatial area covering the nursery grounds of the fish in March and April 2003. Food habits of the fish were analyzed by examining the gut contents. Fish condition was evaluated by using RNA/DNA ratio and other nucleic acid-based indices and protein growth rates. The nucleic acid contents in individually frozen larvae and juveniles were quantified by fluorometric method. Two distinguished feeding patterns, determined by the distribution of prey copepods, were identified along the nursery ground. The first pattern showed the dependency of the fish on the calanoid copepod Sinocalanus sinensis, which was the single dominant prey in low saline upper river areas and the second pattern involved a multi-species dietary habit mainly dominated by Acartia omorii, Oithona davisae and Paracalanus parvus. Values of RNA, DNA, total protein, growth rates and for all the nucleic acid-based indices were higher in upstream areas than in the downstream areas. The proportion of the starving fish was higher in the downstream areas than in the upstream areas. Condition of juvenile sea bass was not equal throughout the nursery grounds; fish in the upper river were in better condition than those in the lower estuary. We speculated that utilization of S. sinensis, which appears a suitable prey item and provide a better foraging environment in the upstream nursery ground, is one of the key factors for early survival and growth of Japanese temperate bass larvae and juveniles in the Chikugo estuary.  相似文献
