  1. As part of a general investigation into the exchange of gasesbetween the bogbean, Menyanthes trifoliata, and its environment,determinations have been made of the composition of the internalatmosphere of the plant at five different levels, viz. leafyshoot, three successive portions of stem, and roots.
  2. Procedureis described for the analysis of small gas samplesusing a modificationof the Bonnier and Mangin apparatus.
  3. In darkness with thelower part of the plant surrounded by anatmosphere of nitrogen,it has not been possible to show thepresence of a regular downwardsgradient of oxygen, such aswould be expected on a basis ofnormal gaseous diffusion.
  4. Under these Air Top/Nitrogen Bottomconditions the roots areshown to contain between 12?5 and 17?5percent, of oxygen intheir intercellular gas system. The portionof stem immediatelyadjacent to the roots showed an oxygen levelof from 14?0 to18?0 per cent.
  5. In plants kept under NitrogenTop/Air Bottom conditions in thedark for 48 hours, the concentrationof oxygen in the rootsfell to a level of 3?1 per cent, or belowin three plants andto 6?o per cent, in a fourth. The oxygenconcentration in theadjacent lowermost part of the stem wasas high as 18?0 percent., depending on the degree of aerationof the solution bathingthe stem. It is suggested that thisdistribution of oxygen canbe related to the relative impermeabilityof roots and of stemendodermis to gaseous diffusion from without,and it is thoughtthat the major part of the oxygen supply tothe roots is transportedto them through the stelar air passages.
  6. Some evidence is presented to show that oxygen passes fromthelower parts of the plant into the medium surrounding it.

  1. Investigations were made on the influence of inorganic nitrogenouscompounds upon the the germination of tobacco seeds (Nicotianatabacum L. var. virginica (AGDH.) COM. "Bright Yellow") inducedby GA3, kinetin and ammonium salts of organic acids. Potassiumnitrate, ammonium chloride, ammonium sulfate and ammonium nitrategreatly increase the germination of the seeds induced by theabove reagents, while these inorganic salts, given alone, arealmost ineffective in causing germination.
  2. Kinetin was shownto induce germination of tobacco seeds inthe dark. The discrepancywith the results of previous investigationsin this respectwas discussed.
  3. It was inferred that nitrogenous metabolismis involved in theprocess of dark-germination of tobacco seedsas induced by theabove-stated stimulating factors and promotedby inorganic nitrogenoussubstances.

  1. Seed germination of Begonia Evansiana ANDR. was investigatedat 29?C.
  2. The germination was induced under long-day conditions,the criticaldaylength being about 8 hours. Exposure to at least2 or 3 cyclesof long days was necessary for germination. Theseeds couldgerminate under otherwise non-inductive photoperiods,when thedark period was interrupted with a short period ofillumination.Thus the photoperiodic behaviour of Begonia seedsin germinationis similar to that of typical long-day plantsin flowering.
  3. The application of gibberellin brought aboutno germinationin complete darkness, but markedly reduced thecritical daylengthfor germination, even 1-minute photoperiodsbeing inductive.The germination under continuous light wasalso favoured bygibberellin application. The action of gibberellinin germinationof Begonia seeds may be to intensify the lightaction or tosubstitute for a part of it.

  1. Apprehension over the adequncy of current techniques stimulateda detailed study of the time factor in the arsenate inhibitionof growth and respiration in excised stem and root sectionsof Pisum sativum.
  2. Growth inhibition by arsenate sets in veryslowly, its rateof onset being related to the molar concentration(C) of arsenateate by the relation where T50 is the time taken in hours to reduce the growthrateto 50 per cent of the control and K is a constant. An explanationof the physiological basis of this relationship is attempted.
  3. Estimates were made of the final steady growth rate (relativeto control) in various arsenate concentrations. The inhibitionscalculated from this rate are held to approximate to the truearsenate effect and are shown to be very different from thosecalculated from ‘total growth’ measures.
  4. Respirationof growing stem sections is not inhibited by thelow arsenateconcentrations that inhibit growth. Some inhibitionis indicatedat high concentrations (3 ? 10–4M. and over)but onlyafter 15-20 hours of exposure.
  5. Two per cent sucrose has noeffect on the arsenate inhibiitionof stem growth. Sucrose,however, markedly stimulates respirationin stem sections, butthis stimulation is prevented by arsenate.
  6. The misinterpretationswhich may arise as a result of ignoringthe time factor in inhibitionstudies in excised organ sectionsare discussed and the desirabilityof constructing completegrowth curves in all such studies isstressed.

Germination of spinach (Spinacia oleracea L. var. grabra cv.Nobel) seeds was inhibited at a high temperature (35?C). Effectsof KCN on the respiration of seeds incubated at 20 and 35?Cwere compared in order to investigate the mechanism of inhibitionof seed germination by high temperature. Respiration of germinatingseeds incubated at 20?C was inhibited about 50% by 5 mM. KCNsolution, whereas it hardly inhibited the weak respiration ofthe seeds at 35?C. Germination of seeds was delayed by exogenousKCN. When the KCN solution was renewed daily, germination wascompletely inhibited. Pericarp removal promoted germinationat 35?C, but atypical germination (cotyledons emerging earlierthan a radicle) took up more than half of the total germination.The inhibitory action of KCN on the respiration of seeds wasnot altered by pericarp removal. A KCN addition, even at 20?C,elicited atypical germination in the pericarp-less seeds. Theseresults show that cyanide-sensitive respiration is needed toinduce typical spinach seed germination (root emergence), butis rendered inoperative by high temperatures thus bringing aboutpoor germination and atypical germination.   1. A survey of potential fat-producing moulds has shown that atleast 40 strains from 10 species are of interest.
  2. These havebeen grown on five different media and A. nidulans,P. spinu-losum,P. javanicum, P. piscarium, P. flavo-cinereum,P. oxalicum,A. flavus, A. flavipes have shown the most promise.
  3. The fatcontent on felt weight was maximal at 39?7 per cent,with A.flavipes and 34?6 per cent, with F. lini, and on usedsugarwas maximal at 9?3 per cent, with A. flavipes and 6?7per cent,with A. nidulans.
  4. The potential value of micro-organisms asfat producers is discussedwith reference to the moulds.

  1. Investigation was made on the influence of inorganic phosphateupon the germination of positively photoblastic tobacco seed(Nicotiana tabacum L. var. uirginica (AGDH.) COM. "Bright Yellow")induced by GA3, GA3M, kinetin, red light, and ammonium saltsof various organic acids.
  2. Inorganic phosphate increases theGAs-induced germination, andinhibits the germination causedby ammonium citrate, while itdoes not influence the germinationbrought about by GA3M, kinetin,and red light.
  3. The optimumpH for the GA3-induced germination lies in the acidicpH range,indicating that the undissociated form of GA3 is operative.The stimulatory effect of phosphate is, however, not ascribedmerely to the pH control in the mediurr. Phosphate exerts somespecific influence for which the presence of the free carboxylgroup of GAs is required.
  4. The observed contrasting effectsof phosphate on the GA3-inducedgermination (i.e., acceleration),on the one hand, and on theammonium citrate-induced germination(i.e., inhibition), onthe other, were explained by assumingthat the phosphate effectsultimately consist in acceleratingthe uptake of the carboxylicacid into the seeds.
  5. GA3M alsohas an activity of inducing the germination of tobaccoseedwithout light.

An attempt has been made to elucidate some of the changes whichtake place in seeds of Striga hermonthica (immature, ripe andoverripe), (a) during moisture-treatment at three temperatures(15°C., 22°C., and 32°C.), and (b) during after-ripeningin dry storage. Observations on the drift of respiration of immature seed throughoutpretreatment at 15°C. show that an inverse relationshipexists between germination and respirational activity. Thissuggests that germination depends on the accumulation of somemetabolite during pretreatment. It would seem that this metabolitemay also be used as a substrate for respiration during pretreatment. After-ripening of the immature seeds has been shown to changethe nature of the seeds in some way so that their peak respirationon exposure to moisture is at first progressively reduced. Thisreduction has been ascribed to the presence of some barriercapable of restricting gaseous diffusion. The data presented, togetherwith those of the previous paperof this series, suggest that the specific metabolite requiredfor germination might be proteinaceous. Certain other data appearto rule out the suggestion that germination capacity is conditionedby the sugar content of the seeds. Reasons are given which supportthe view that one of the main effects of the stimulating solutionon the seed is to increase the permeability of the seeds togaseous diffusion.  相似文献

  1. The loss of carbohydrate, acid, and cell-wall material, andthe production of carbon dioxide and alcohol, have been studiedin apples under aerobic and anaerobic conditions.
  2. The changesin all these metabolites, except the cell-wall material,proceedin a regular manner.
  3. The presence of oxygen has a conservingeffect on the loss ofcarbohydrate.
  4. The presence or absenceof oxygen is without effect on the rateof loss of acid.
  5. Therate of loss of acid is proportional to the logarithm ofitsconcentration and is constant for a given variety of apple.
  6. The loss of carbohydrate plus acid accounts quantitativelyforthe production of carbon dioxide and alcohol, both in airorin nitrogen.
  7. The anaerobic respiration of carbohydrateby apples is identical,so far as the nature and quantity ofthe end products are concerned,with the alcohol fermentationof yeast.
  8. The carbon dioxide which is produced in nitrogen,over and abovethat produced in fermentation, is equivalentto that which wouldbe produced by oxidation of the acid.

The effect of ethylene on quantitative and qualitative aspectsof respiration in Spergula arvensis L. seeds was studied. Oxygenconsumption and CO2 evolution from seeds increased 60 h afteraddition of ethylene to the dry seed; these increases were consideredto be a function of the number of seeds that were germination,rather than a direct effect of ethylene. The respiratory quotient(RQ) on day 1 (0.5) was lower than on subsequent days, indicatingan absence of anaerobic processes in the early stages of germinationand the use of a different respiratory substrate to that oxidizedon days 2–4. Measurements of C4/C1 ratios showed thatethylene did not break dormancy by initiating a switch fromthe Embden-Meyerhof-Parnas pathway (EMPP) to the pentose phosphatepathway (PPP). On the contrary, some evidence was obtained fora switch from the PPP to the EMPP after 3–6 h in pre-imbibedand non pre-imbibed seeds treated with ethylene.   1. Investigations were carried out to examine the effects of certainnucleic acid antimetabolites upon the germination of leaf mustardseed. Antimetabolites tested were 2 barbituric acid, variousuracil analogues, azaadenine, azaxanthine, azaguanine and benzimidazol.
  2. Among the antimetabolites tested, only 2-thiouracil showedastrong inhibitory action on the germination of Brassica junceavar. foliosa (BAILEY) Corn. "Akatirimen".
  3. This inhibitionby 2-thiouracil was relieved markedly by suchpyrimidine derivativesas uracil, thymine, orotic acid, uridylicacid and cytidylicacid, though these compounds did not showany promotive effecton the germination by themselves. On the other hand, sucha relieving effect was not shown bypurine derivatives suchas adenine, hypoxanthine and xanthine.
  4. Among other kinds ofcole-wort seeds tested, a similar thiouracilinhibition on seedgermination was observed only in a limitednumber of speciesor varieties.

  1. Formyltetrahydrofolate synthetase (E. C. 6. 3. 4. 3) was foundto be widely distributed in higher plants and the high enzymeactivity was observed in green leaves of Brassica and Alliumspecies, spinach, and in pea seedlings. In pea seedlings, theenzyme activity changed during the course of germination, andmost of the enzyme activity was located in a soluble fractionof the cytoplasm.
  2. The enzyme was labile and lost the activityrapidly, even whenstored at 5 in the presence of 0.1 M mercaptoethanol.It was,however, found that ammonium sulfate was very effectivein stabilizingthe enzyme activity.
  3. The enzyme has been purifiedapproximately 500-fold from extractsof pea seedlings by treatmentswith ammonium sulfate, protaminesulfate, hydroxylapatite, calciumphosphate gel, and DEAE-cellulosecolumn chromatography.
  4. Thepurified enzyme was specific for formate, ATP and FAH4,andthe Michaelis constants for these reactants were 2.1 10–2M, 5.1 10–4 M, and 5.6 10–3 M, respectively.
  5. The optimum pH was found to be 8.0, and the optimal temperaturewas observed at 37. Both NH4$ and a divalent cation (MgSS orMnSS) were required for the optimal activity.

Three kinds of discs were taken from tobacco leaves whose lowerepidermis had been peeled off, half-peeled or unpeeled. Therole of the epidermis and its relation to the kinetin effecton chlorophyll degradation during senescence were studied. Ourresults follow.
  1. Chlorophyll degradation due to kinetin was retarded only whenthe lower epidermis was present.
  2. The decrease in chlorophyllcontent in leaf discs on water duringsenescence was nearlyproportional to the size of the lowerepidermis attached tothe discs; i.e., unpeeled discs>half-peeleddiscs>peeleddiscs.
  3. Cellular fractions possessing activity which induceschlorophylldegradation were extracted from the isolated lowerepidermis(i, ii) and its acetone powder (iii): (i) L-2 fraction(1.14d1.16)was separated by stepwise sucrose density-gradientcentrifugationfrom the 10,000?g pellet of the cell homogenate.(ii) The A-fraction(M.W.5,000) was precipitated with 0–80%saturation ofammonium sulfate from 105,000 ? g supernatantof cell homogenateand eluted in the void volume by SephadexG-25 column chromatography.(iii) The fraction precipitatedwith 0–30% saturationof ammonium sulfate from the 105,000?gsupernatant, containeda large amount of DNA and its activityremained even if DNAwas removed.
  4. Activity was not retainedwhen the fractions were obtained fromisolated lower epidermispretreated with 2?10–5 M kinetinfor 2 hr in darknessat 25?C.

The effects of kinetin and gibberellin were examined under anaerobicconditions (0% oxygen) and aerobic conditions (20% oxygen) onthe germination of dehusked seeds of indica and japonica ricecultivars that had been harvested at different times duringthe formation of seeds. Surjamkhi was used as a representativeof deep dormant indica cultivars and Assam IV as a less dormantindica cultivar. Sasanishiki was used as the japonica rice cultivar.Both phytohormones were applied at a concentration of 10-3Mwhichproved to have the greatest stimulatory effect in preliminarywork at concentrations of 10-3–10-5M. Under aerobic conditions,inhibition of germination by dehusking of Sasanishiki seedsthat had been harvested either 30 or 60 d after anthesis wasovercome by kinetin and all seeds germinated. Complete germinationinduced by kinetin under aerobic conditions was also achievedwith the dehusked seeds of the indica rice cultivar Assam IVthat had been harvested on two occasions and of Surjamkhi thathad been harvested 28 d after anthesis. In contrast, germinationof dehusked japonica seeds stimulated by anaerobiosis was inhibitedby kinetin. The stimulatory effects of gibberellin on the germinationof indica and japonica rice seeds were observed under aerobicand anaerobic conditions. Under anaerobic conditions, the responsesof dehusked indica and japonica rice seeds to kinetin and gibberellindiffered, being negative with kinetin and positive with gibberellin.Under aerobic conditions, the stimulatory effects of kinetinon germination of dehusked seeds were greater than those ofgibberellin. Thus, treatment with kinetin appears to be usefulfor breaking the considerable dormancy commonly observed inthe dehusked seeds of indica rice. Mechanisms are proposed toexplain the stimulatory effects of these phytohormones on thegermination of dehusked seeds of indica and japonica rice underaerobic and anaerobic conditions. Two neutral plant hormones, one isolated recently from plants(3-indolylacetonitrile) and the other (3-indolylacetaldehyde)reported to be present in plants, are avaible as pure syntheticcompounds for investigation of their biological activities.This paper is mainly concerned with their effects on cellelongationin the Avena coleoptile
  2. 3-Indolylacetaldehyde is considerablyless active than 3-indolylaceticacid in the Avena straight-growthtest; for example, a 1.0 mg./l.solution of the aldehyde showsan activity equivalent to thatof a 0.1 mg./l. solution of theacid
  3. An acidic substance is produced in solutions of the aldehydeduring the period of assay. In some experiments it accountsfor all of the activity shown by the aldehyde solutions, onthe assumption that it is 3-indoylacetic acid, and in otherexperiments it shows a greater activity than that of the aldehydesolutions from which it was obtained. Therefore, it is concludedthat 3-indolylacetaldehyde, itself is either inactive or inhibitory.Acid production in aldehyde solutions in vitro is much lower,a fact which suggests that there is enzymatic oxidation of aldehydeto acid in the presence of coleoptiles.
  4. The activities of3-indolylacetaldehyde in the pea test andin root-inhibitionand of 3-indolylacetone in the straight-growthtest are brieflyreported.
  5. 3-Indolylacetonitrile is considerably more activethan 3-indolylaceaticacid in the Avena straight-growth test;for erample, a 0.1 mg./l.solution of the nitrile shows an activityequivalent to a 1.0mg./l. solution of the acid. The inhibitoryeffect at concentrationsabove 1.0–10.0 mg./l. is lesswith the nitrile than withthe acid.
  6. There is negligible productionof acid in solutions of the nitrileboth in vitro and in thepresence of Avena coleoptiles at temperaturesranging from –18?to 25? C. for varying lengths of time.The possibility of enzymaticconvesion of nitrile to acid insidethe cells of the coleoptileis discussed
  7. The activities of 3-indolylacetamide and of 2:4-dichlorophenoxyaceticacid and the corresponding nitrile are considered in this connexion
  8. The nitrile is destroyed by treatment with alkali but notbyacid. In the light of these results, it is desirable to re-examineprevious work on identification of auxins in plants by theiracid and alkali sensitivity. Evidence for the existence of thenitrile in a number of other plants is presented.

  1. The variety Phulwa was worked upon for respiratory studiesandgrowth response to dormancy-breaking hormones. The hormoneswere used in single and double doses.
  2. The respiratory studiesreveal that ethylene chlorhydrin affectsonly the bud areasand not the non-bud areas.
  3. The intensity of the effect ofthe hormone progressively decreasesin the deeper layers ofcells, till finally no effect is visiblein the cells of thepith areas.
  4. A distinct correlation exists between the respiratoryintensityand dormancy.
  5. Of the two hormones used the respiratoryrates recorded forN. ethylene chlorhydrin treated tubers, givenin single anddouble doses, were the highest. Similarly thegrowth responseto both the doses showed the maximum reductionof the dormancyperiod, 01 N. ethylene chlorhydrin treatmentswere not effectivein breaking the dormancy or in significantlyaltering the rateof respiration.
  6. Treatments with 05 percent, ammonium thiocyanate given insingle dose showed the lowestrespiratory rates and the germinationrecords showed no differenceover the control. With the doubledose a gradual rise in therespiratory rates was observed. Thegermination records alsoshowed an earliness of 20 and 22 daysover the control and single-dosetreatments respectively.
  7. Treatments with 2 per cent, ammoniumthiocyanate depressed germination.The respiratory rates recordedwere also low.
  8. In general the double-dose treatments showedbetter resultsthan the single-dose treatments. Also, treatmentson 3 weeksold tubers were more effective than on 6 weeks oldtubers.

  1. A close parallelism in the drift of the rate of respirationand the protein-N/ non-protein-N ratio is shown to occur bothin apple fruits attached to the tree and when detached fromthe tree at various stages of development and stored for severalmonths at 12 C.
  2. In detached fruits the fall in respirationwhich occurs immediately(during the first 48 hours) after pickingis only accompaniedby a concomitant fall in net protein invery young fruits inwhich active cell division is taking place.Subsequently, infruit of all ages when a climacteric rise inrespiration occursit is accompanied by a net increase in protein.
  3. It is argued that the climacteric rise in respiration is aresultof increase in the level of protein which will be expectedtoreduce the ATP/ADP ratio.
  4. Over the climacteric period,although rate of respiration andnet protein content both rise,R rises more rapidly than proteinand, subsequently, falls ata faster rate than P. It is suggestedthat this may be due tothe ‘new’ protein containinga higher proportionof enzyme(s) directly involved in respirationand leading, forexample, to a reduction in the ATP/ADP ratio.

Seed priming (imbibition in water or osmotic solutions followedby redrying) generally accelerates germination rates upon subsequentre-imbibition, but the response to priming treatments can varyboth within and among seed lots. Seed maturity could influenceresponsiveness to priming, perhaps explaining variable primingeffects among developmentally heterogeneous seed lots. In thecurrent study, muskmelon (Cucumis melo L.) seeds at two stagesof development, maturing (40 d after anthesis (DAA)) and fullymature (60 DAA), were primed in 0?3 M KNO3 for 48 h at 30 ?C,dried, and imbibed in polyethylene glycol 8000 solutions of0 to –1?2 MPa at 15, 20, 25, and 30 ?C. Germination sensitivitiesto temperature and water potential () were quantified as indicatorsof the influence of seed maturity and priming on seed vigour.Germination percentages of 40 and 60 DAA control seeds weresimilar in water at 30 ?C, but the mean germination rate (inverseof time to germination) of 40 DAA seeds was 50% less than thatof 60 DAA seeds. Germination percentages and rates of both 40and 60 DAA seeds decreased at temperatures below 25 ?C. Reductionsin also delayed and inhibited germination, with the 40 DAAseeds being more sensitive to low than the 60 DAA seeds. Primingsignificantly improved the performance of 40 DAA seeds at lowtemperatures and reduced , but had less effect on 60 DAA seeds.Priming lowered both the minimum temperature (Tb) and the minimum (b) at which germination occurred. Overall, priming of 40 DAAseeds improved their germination performance under stress conditionsto equal or exceed that of control 60 DAA seeds, while 60 DAAseeds exhibited only modest improvements due to priming. Asthe osmotic environment inside mature fruits approximates thatof a priming solution, muskmelon seeds may be ‘primed’in situ during the late stage of development after maximum dryweight accumulation.   1. A new technique for studying extension growth in the root isdescribed which is based on excising a zone which extends 1·5–3·0mm. from the tip. Large numbers of these segments are culturedwith different nutrient fluids in the dark at 25° C. withcontinual shaking.
  2. The effects of a large number of nutrientson the growth ofthe segments have been studied, but only two,sugar and potassiumions, have been found to have stimulatingeffects.
  3. The effects of water, three concentrations of sugar,and oneof potassium in air, and with an atmosphere containing5 percent. oxygen have been studied in detail in connexionwith lengthincrease, sugar absorption, content of free sugar,cellulosecontent, dry weight, and respiration.
  4. It has beenshown that with increasing concentration of sugarin the medium,the rate of growth, the time during which growthproceeds, theinternal concentration, respiration, dry weight,and celluloseformation all increase. Also that potassium stimulatesthe rateof growth and respiration, and that with per cent,oxygen allthe aspects studied are depressed.
  5. It is suggested that thestimulation due to sugar may be attributedto an accelerationof water absorption with a complementaryincrease in celluloseformation. It is further suggested thatsugar accelerates waterabsorption by accumulating in the vacuoleand thus sustainingthe osmotic pressure of the vacuolar sap.It is further suggestedthat potassium stimulates growth byincreasing water absorptionthrough an effect on respiration.The effect of respirationin this connexion may be to promotethe transport of water directlyto enhance the osmotic pressureof the sap by inducing an accumulationof inorganic ions inthe vacuole.

  1. Polyphenoloxidase is present in ivy leaves but not in thoseof Aucuba or Euonymus.
  2. Respiration of intact ivy leaves wasmarkedly stimulated bycatechol (R.Q. approximately=I), gallicacid, caffeic acid,and dihydroxyphenylalanine. The stimulationwas not relatedto injury as far as could be detected and wasnot followed byinhibition. The extra oxygen consumption representsa many timesrepeated oxidation of the amount of catechol supplied.
  3. The effect of catechol on the respiration of Aucuba and Euonymusleaves was very small.
  4. Cupferron and phenylthiourea, whichinhibit polyphenoloxidasein vitro, nevertheless increased respirationwhen administeredto leaves through the petiole. On the otherhand, when appliedby infiltration, cupferron did cause inhibition,but in Aucubaand Euonymus as much as in ivy.
