 When plotted in linear coordinates, the dose-response curves of microorganisms exposed to a lethal agent, such as radiation or a toxic substance, often have a characteristic sigmoid shape. Irrespective of whether they are very narrow or broad they can be described by the Fermi function, which is a mirror image of the logistic function, i.e. S(X)=1/{1+ exp [(XX c)/a]} where S(X) is the fraction of the surviving organisms, X the dose of the lethal agent, X c a characteristic dose marking the inflection point of S(X), which corresponds to 50% mortality, and a a measure of the steepness of the survival curve around X c. It is demonstrated that, if the susceptibilities of the individual organisms, expressed in terms of a characteristic lethal dose, have a symmetric unimodal distribution, the dose-response curve of the population has a Fermian sigmoid shape. It is also shown that the mode and variance of the distribution can be estimated from the shape parameters of the Fermian survival curve, X c and a. The conditions under which the output,γ b (t), of a biological system is related to the input,γ a (t), by an integral equation of the typeγ b (t) = ∫ 0 t γ a (ω)w(t−ω)dω, where ω(t) is a transport functioncharacteristic of the system, are analyzed in detail. Methods of solving this type of integral equation are briefly discussed. The theory is then applied to problems in tracer kinetics in which input and output are sums of exponentials, and explicit formulae, which are applicable whether or not the pool is uniformly mixed, are derived for “turnover time” and “pool” size.  相似文献

For any essentially nonlinear system of reaction-diffusion equations of the generic form ∂ci/∂t=Di2ci+Qi(c,x,t) supplemented with Robin type boundary conditions over the surface of a closed bounded three-dimensional region, it is demonstrated that all solutions for the concentration distributionn-tuple function c=(c 1(x,t),...,c n (x,t)) satisfy a differential variational condition. Approximate solutions to the reaction-diffusion intial-value boundary-value problem are obtainable by employing this variational condition in conjunction with a Galerkin-Ritz procedure. It is shown that the dynamical evolution from a prescribed initial concentrationn-tuple function to a final steady-state solution can be determined to desired accuracy by such an approximation method. The variational condition also admits a systematic Galerkin-Ritz procedure for obtaining approximate solutions to the multi-equation elliptic boundary-value problem for steady-state distributions c=−c(x). Other systems of phenomenological (non-Lagrangian) field equations can be treated by Galerkin-Ritz procedures based on analogues of the differential variational condition presented here. The method is applied to derive approximate nonconstant steady-state solutions for ann-species symbiosis model.  相似文献

An important criterion used to detect adaptive evolution in DNA sequence data is ωi > 1, where ωi is the ratio of nonsynonymous to synonymous substitution rates in lineage i. However, the evaluation of multiple ωi within a phylogenetic tree can easily inflate the statistical type I error rate. We developed two rigorous methods of analysis that avoid this and other potential pitfalls. We applied these methods to four published examples of adaptive evolution. One case was strongly supported by our reanalysis (abalone sperm lysin), and one was weakly supported (baboon α-globin), but two examples (primate lysozyme and Antarctic fish β-globin) did not show significant evidence of adaptive evolution. Our first method is a “bottom-up” hierarchical maximum likelihood approach, which (1) tests for significant heterogeneity in ω across the phylogeny, (2) locates its source using a sequence of planned comparisons, and (3) tests homogeneous groups of ω for ω > 1, using a modified level of significance that incorporates the pretesting. The second method is a “top-down” log-linear analysis based on estimates of nonsynonymous and synonymous substitutions in pairs of lineages. The log-linear test is applied to pairs of lineages joined at progressively deeper nodes. For each pair, the analysis simultaneously tests for adaptive evolution (ω > 1), a shift in natural selection (ω1 ≠ω2), and unequal evolution rate (the relative rate test). In both tests, we emphasized that the criterion ω1 ≠ ω2 is an important additional indicator of a phylogenetic shift in the balance between natural selection and genetic drift between two related lineages. To evaluate how the land carbon reservoir has been responding to the rising CO2 concentration of the atmosphere, it is important to study how plants in natural forests adjust physiologically to the changing atmospheric conditions. Many experimental studies have addressed this issue, but it has been difficult to scale short-term experimental observations to long-term ecosystem-level responses. This paper derives carbon-isotope-related variables for the past 100–200 years from measurements on trees from natural forests. Calculations show that the c i/c a ratios [c i/c a is the ratio of the CO2 concentration (μmol mol−1) in the intercellular space of leaves to that in the atmosphere] of the trees were constant or increased slightly before the 20th century, but changed more rapidly in the 20th century; some increased, some decreased, and some stayed constant. Proceeding from three previously derived expressions for the intensity of nitrification in soil as a function of time (logΣN=K.logt+q), as a function of incubation moisture (logΣN=A.pF i+B), as a function of initial moisture (logΣN=C.pF v+D), it was shown that the nitrification intensity as a function of time and of moisture can be expressed by the bilinear function log ΣN=a.pF i.logT+b.pF i+c.logt+d; as a function of time and of initial moisture by the bilinear function logΣ=N=a.pF v.logt+b.pF v+c.logt+d; as a function of initial and incubation moisture by the bilinear function log ΣN=a.pF ipF v+b.pF i+c.pF v+d. The intensity of nitrification as a function of time, incubation moisture and initial moisture may be expressed by the multilinear function log ΣN=a.pF i.pF v.logt+b.pF i.pF v+c.pF i.logt+d.pF v.logt+e .pF i+f.pF v=g.logt+h. Resonance Raman (RR) spectroscopy was used to investigate conformational characteristics of the hemes of several ferricytochromes of the cytochrome c 3 family, electron transfer proteins isolated from the periplasm and membranes of sulfate-reducing bacteria. Our analysis concentrated on the low-frequency region of the RR spectra, a fingerprint region that includes vibrations for heme-protein C–S bonds [ν(CaS)]. It has been proposed that these bonds are directly involved in the electron transfer process. The three groups of tetraheme cytochrome c 3 analyzed, namely Type I cytochrome c 3 (TpIc 3s), Type II cytochrome c 3 (TpIIc 3s) and Desulfomicrobium cytochromes c 3, display different frequency separations for the two ν(CaS) lines that are similar among members of each group. These spectral differences correlate with differences in protein structure observed among the three groups of cytochromes c 3. Two larger cytochromes of the cytochrome c 3 family display RR spectral characteristics for the ν(CaS) lines that are closer to TpIIc 3 than to TpIc 3. Two other multiheme cytochromes from Desulfovibrio that do not belong to the cytochrome c 3 family display ν(CaS) lines with reverse relative areas in comparison with the latter family. This RR study shows that the small differences in protein structure observed among these cytochrome c 3 correlate to differences on the heme–protein bonds, which are likely to have an impact upon the protein function, making RR spectroscopy a sensitive and useful tool for characterizing these cytochromes.  相似文献

In connection with previous studies (Rashevsky,Mathematical Biology of Social Behavior, chap. xii), a situation is investigated in which the two mutually exclusive possible behaviors of a society consist of the desire to keep the present socioeconomical situation and the desire to change it inany way. The psychophysiological tendency ϕ towards either of the behaviors is considered to be proportional to the difference between the actual incomei of the individual and his needsi′. Assuming that the distribution functionN 1(i′) of the needs is a given characteristic of the population, it is shown that the distribution functionN(ϕ) of ϕ in the society can be derived fromN 1(i′) and from the distributionN 2(i) of the incomesi. The study comprises a data set of CTD, optical properties—K 0(PAR), c p, a(PAR), b(PAR)—and optical constituents—Chl a, SPM, CDOM—from 72 shelf and off-shelf stations in the Faroe Islands (62°N, 7°W) North East Atlantic, in early spring 2005. Results showed that shelf waters surrounding the islands were cold and low saline, whereas off-shelf waters were warmer (~1°C) and more saline (~0.05) PSU. A pronounced oceanographic front separated the two waters, and diffuse light attenuation K 0(PAR), beam attenuation c p, Chl a, absorption a(PAR), and scattering coefficient b(PAR) were all significantly higher on the shelf. Analyses showed that off-shelf light attenuation K 0(PAR) was governed by Chl a, shown by a high (r 2 = 0.64) Chl aK 0(PAR) correlation, whereas light attenuation on the shelf was governed by both Chl a, SPM, and CDOM in combination. A Chl a specific diffuse attenuation coefficient K0* ( \textPAR ) K_{0}^{*} \left( {\text{PAR}} \right) of 0.056 (m2 mg−1 Chl a) and a Chl a specific beam attenuation ( c\textp* c_{\text{p}}^{*} ) of 0.27 (m2 mg−1 Chl a) coefficients were derived for the off-shelf. It is pointed out that Chl a is the single variable that changes over time as no rivers with high SPM and CDOM enter the shelf area. Data were obtained in early spring, and Chl a concentrations were low ~0.5 mg Chl a m−3. Spring bloom Chl a are about 10 mg Chl a m−3 and estimations showed that shelf K 0(PAR) will increase about 5 times and beam attenuation about 10 times. The Faroe Islands shelf–off-shelf waters is a clear example where physical conditions maintain some clear differences in optical properties and optical constituents. The purpose of this study was to develop a method to determine the power output at which oxygen uptake (O2) during an incremental exercise test begins to rise non-linearly. A group of 26 healthy non-smoking men [mean age 22.1 (SD 1.4) years, body mass 73.6 (SD 7.4) kg, height 179.4 (SD 7.5) cm, maximal oxygen uptake (O2max) 3.726 (SD 0.363) l · min−1], experienced in laboratory tests, were the subjects in this study. They performed an incremental exercise test on a cycle ergometer at a pedalling rate of 70 rev · min−1. The test started at a power output of 30 W, followed by increases amounting to 30 W every 3 min. At 5 min prior to the first exercise intensity, at the end of each stage of exercise protocol, blood samples (1 ml each) were taken from an antecubital vein. The samples were analysed for plasma lactate concentration [La]pl, partial pressure of O2 and CO2 and hydrogen ion concentration [H+]b. The lactate threshold (LT) in this study was defined as the highest power output above which [La]pl showed a sustained increase of more than 0.5 mmol · l−1 · step−1. The O2 was measured breath-by-breath. In the analysis of the change point (CP) of O2 during the incremental exercise test, a two-phase model was assumed for the 3rd-min-data of each step of the test: X i =at i +b i for i=1,2,…,T, and E(X i )>at i +b for i =T+1,…,n, where X 1, … , X n are independent and ɛ i ∼N(0,σ2). In the first phase, a linear relationship between O2 and power output was assumed, whereas in the second phase an additional increase in O2 above the values expected from the linear model was allowed. The power output at which the first phase ended was called the change point in oxygen uptake (CP-O2). The identification of the model consisted of two steps: testing for the existence of CP and estimating its location. Both procedures were based on suitably normalised recursive residuals. We showed that in 25 out of 26 subjects it was possible to determine the CP-O2 as described in our model. The power output at CP-O2 amounted to 136.8 (SD 31.3) W. It was only 11 W – non significantly – higher than the power output corresponding to LT. The O2 at CP-O2 amounted to 1.828 (SD 0.356) l · min−1 was [48.9 (SD 7.9)% O2 max ]. The [La]pl at CP-O2, amounting to 2.57 (SD 0.69) mmol · l−1 was significantly elevated (P<0.01) above the resting level [1.85 (SD 0.46) mmol · l−1], however the [H+]b at CP-O2 amounting to 45.1 (SD 3.0) nmol · l−1, was not significantly different from the values at rest which amounted to 44.14 (SD 2.79) nmol · l−1. We revisit the relationship between plant water use efficiency and carbon isotope signatures (δ13C) of plant material. Based on the definitions of intrinsic, instantaneous and integrated water use efficiency, we discuss the implications for interpreting δ13C data from leaf to landscape levels, and across diurnal to decadal timescales. Previous studies have often applied a simplified, linear relationship between δ13C, ratios of intercellular to ambient CO2 mole fraction (C i/C a), and water use efficiency. In contrast, photosynthetic 13C discrimination (Δ) is sensitive to the ratio of the chloroplast to ambient CO2 mole fraction, C c/C a (rather than C i/C a) and, consequently, to mesophyll conductance. Because mesophyll conductance may differ between species and over time, it is not possible to determine C c/C a from the same gas exchange measurements as C i/C a. On the other hand, water use efficiency at the leaf level depends on evaporative demand, which does not directly affect Δ. Water use efficiency and Δ can thus vary independently, making it difficult to obtain trends in water use efficiency from δ13C data. As an alternative approach, we offer a model available at to explore how water use efficiency and 13C discrimination are related across leaf and canopy scales. The model provides a tool to investigate whether trends in Δ indicate changes in leaf functional traits and/or environmental conditions during leaf growth, and how they are associated with trends in plant water use efficiency. The model can be used, for example, to examine whether trends in δ13C signatures obtained from tree rings imply changes in tree water use efficiency in response to atmospheric CO2 increase. This is crucial for predicting how plants may respond to future climate change. High internal resistance is a key problem limiting the power output of the microbial fuel cell (MFC). Therefore, more knowledge about the internal resistance is essential to enhance the performance of the MFC. However, different methods are used to determine the internal resistance, which makes the comparison difficult. In this study, three different types of MFCs were constructed to study the composition and distribution of internal resistance. The internal resistance (R i) is partitioned into anodic resistance (R a), cathodic resistance (R c), and ohmic resistance () according to their origin and the design of the MFCs. These three resistances were then evaluated by the “current interrupt” method and the “steady discharging” method based on the proposed equivalent circuits for MFCs. In MFC-A, MFC-B, and MFC-C, the R i values were 3.17, 0.35, and 0.076 Ω m2, the values were 2.65, 0.085, and 0.008 Ω m2, the R a values were 0.055, 0.115, and 0.034 Ω m2, and the R c values were 0.466, 0.15, and 0.033 Ω m2, respectively. For MFC-B and MFC-C, the remarkable decrease in R i compared with the two-chamber MFC was mainly ascribed to the decline in and R c. In MFC-C, the membrane electrodes’ assembly lowered the ohmic resistance and facilitated the mass transport through the anode and cathode electrodes, resulting in the lowest R i among the three types.  相似文献

Rising atmospheric [CO2], ca, is expected to affect stomatal regulation of leaf gas‐exchange of woody plants, thus influencing energy fluxes as well as carbon (C), water, and nutrient cycling of forests. Researchers have proposed various strategies for stomatal regulation of leaf gas‐exchange that include maintaining a constant leaf internal [CO2], ci, a constant drawdown in CO2 (ca ? ci), and a constant ci/ca. These strategies can result in drastically different consequences for leaf gas‐exchange. The accuracy of Earth systems models depends in part on assumptions about generalizable patterns in leaf gas‐exchange responses to varying ca. The concept of optimal stomatal behavior, exemplified by woody plants shifting along a continuum of these strategies, provides a unifying framework for understanding leaf gas‐exchange responses to ca. To assess leaf gas‐exchange regulation strategies, we analyzed patterns in ci inferred from studies reporting C stable isotope ratios (δ13C) or photosynthetic discrimination (?) in woody angiosperms and gymnosperms that grew across a range of ca spanning at least 100 ppm. Our results suggest that much of the ca‐induced changes in ci/ca occurred across ca spanning 200 to 400 ppm. These patterns imply that ca ? ci will eventually approach a constant level at high ca because assimilation rates will reach a maximum and stomatal conductance of each species should be constrained to some minimum level. These analyses are not consistent with canalization toward any single strategy, particularly maintaining a constant ci. Rather, the results are consistent with the existence of a broadly conserved pattern of stomatal optimization in woody angiosperms and gymnosperms. This results in trees being profligate water users at low ca, when additional water loss is small for each unit of C gain, and increasingly water‐conservative at high ca, when photosystems are saturated and water loss is large for each unit C gain.  相似文献

Two respiratory-deficient nuclear petites, FY23Δpet191 and FY23Δcox5a, of the yeast Saccharomyces cerevisiae were generated using polymerase-chain-reaction-mediated gene disruption, and their respective ethanol tolerance and productivity assessed and compared to those of the parental grande, FY23WT, and a mitochondrial petite, FY23ρ0. Batch culture studies demonstrated that the parental strain was the most tolerant to exogenously added ethanol with an inhibition constant. K i, of 2.3% (w/v) and a specific rate of ethanol production, q p, of 0.90 g ethanol g dry cells−1 h−1. FY23ρ0 was the most sensitive to ethanol, exhibiting a K i of 1.71% (w/v) and q p of 0.87 g ethanol g dry cells−1 h−1. Analyses of the ethanol tolerance of the nuclear petites demonstrate that functional mitochondria are essential for maintaining tolerance to the toxin with the 100% respiratory-deficient nuclear petite, FY23Δpet191, having a K i of 2.14% (w/v) and the 85% respiratory-deficient FY23Δcox5a, having a K i of 1.94% (w/v). The retention of ethanol tolerance in the nuclear petites as compared to that of FY23ρ0 is mirrored by the ethanol productivities of these nuclear mutants, being respectively 43% and 30% higher than that of the respiratory-sufficient parent strain. This demonstrates that, because of their respiratory deficiency, the nuclear petites are not subject to the Pasteur effect and so exhibit higher rates of fermentation. The parameters estimated from traditional A/C i curve analysis are dependent upon some underlying assumptions that substomatal CO2 concentration (C i) equals the chloroplast CO2 concentration (C c) and the C i value at which the A/C i curve switches between Rubisco- and electron transport-limited portions of the curve (C i-t) is set to a constant. However, the assumptions reduced the accuracy of parameter estimation significantly without taking the influence of C i-t value and mesophyll conductance (g m) on parameters into account. Based on the analysis of Larix gmelinii’s A/C i curves, it showed the C i-t value varied significantly, ranging from 24 Pa to 72 Pa and averaging 38 Pa. t-test demonstrated there were significant differences in parameters respectively estimated from A/C i and A/C c curve analysis (p<0.01). Compared with the maximum ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) carboxylation rate (Vcmax), the maximum electron transport rate (Jmax) and Jmax/Vcmax estimated from A/C c curve analysis which considers the effects of g m limit and simultaneously fits parameters with the whole A/C c curve, mean Vcmax estimated from A/C i curve analysis (Vcmax-C i) was underestimated by 37.49%; mean Jmax estimated from A/C i curve analysis (Jmax-C i) was overestimated by 17.8% and (Jmax-C i)/(Vcmax-C i) was overestimated by 24.2%. However, there was a significant linear relationship between Vcmax estimated from A/C i curve analysis and Vcmax estimated from A/C c curve analysis, so was it Jmax (p<0.05).  相似文献

Following our established theoretical model to deal with the second-harmonic generation (SHG) excited by a linearly polarized focused beam in type I collagen, in this paper, we further quantitatively characterize the differences between SHG emissions in type I collagen excited by collimated and focused beams. The effects of the linear polarization angle (α) and the fibril polarity characterized by the hyperpolarizability ratio ρ on SHG emission has been compared under collimated and focused beam excitation, respectively. In particular, SHG emission components along the i axis ( I2w,i )\left( {I_{2\omega {,}i} } \right) (i = x,y,z), the induced SHG emission deviation angle γ ij , and the detected SHG signals (I 2ω,ij ) in the ij plane by rotating the applied polarizer angle φ ij have been investigated (i = x, x, y; j = y, z, z). Results show that under our simulation model, SHG emission in the xy plane, such as I 2ω,x ,I 2ω,y ,γ xy and I 2ω,xy varying as polarization angle (α) under collimated and focused light, presents no significant difference. The reverse of the fibril polarity has induced great impact on I 2ω,x ,γ xy and I 2ω,xy in both collimated and focused light. I 2ω,x and γ xy show similarity, but I 2ω,xy at α = 30° demonstrates a slight difference in focused light to that in collimated light. Under focused light, the reverse of fibril polarity causes obvious changes of the collected SHG intensity I 2ω,xz and I 2ω,yz at a special polarization angle α = 60° and γ xz , γ yz along α.  相似文献

We investigated the tree growth and physiological response of five pine forest stands in relation to changes in atmospheric CO2 concentration (ca) and climate in the Iberian Peninsula using annually resolved width and δ13C tree‐ring chronologies since ad 1600. 13C discrimination (Δ≈ci/ca), leaf intercellular CO2 concentration (ci) and intrinsic water‐use efficiency (iWUE) were inferred from δ13C values. The most pronounced changes were observed during the second half of the 20th century, and differed between stands. Three sites kept a constant ci/ca ratio, leading to significant ci and iWUE increases (active response to ca); whereas a significant increase in ci/ca resulted in the lowest iWUE increase of all stands at a relict Pinus uncinata forest site (passive response to ca). A significant decrease in ci/ca led to the greatest iWUE improvement at the northwestern site. We tested the climatic signal strength registered in the δ13C series after removing the low‐frequency trends due to the physiological responses to increasing ca. We found stronger correlations with temperature during the growing season, demonstrating that the physiological response to ca changes modulated δ13C and masked the climate signal. Since 1970 higher δ13C values revealed iWUE improvements at all the sites exceeding values expected by an active response to the ca increase alone. These patterns were related to upward trends in temperatures, indicating that other factors are reinforcing stomatal closure in these forests. Narrower rings during the second half of the 20th century than in previous centuries were observed at four sites and after 1970 at all sites, providing no evidence for a possible CO2‘fertilization’ effect on growth. The iWUE improvements found for all the forests, reflecting both a ca rise and warmer conditions, seem to be insufficient to compensate for the negative effects of the increasing water limitation on growth.  相似文献

A pale yellow-colored, moderately halophilic, Gram-negative, catalase- and oxidase-positive, non-sporulating, rod-shaped, motile, aerobic bacterium, designated strain JSM 073008T, was isolated from a sea anemone (Anthopleura xanthogrammica) collected from Naozhou Island, Leizhou Bay, South China Sea. The organism was able to grow with 1–20% (w/v) total salts (optimum, 5–10%), at pH 6.0–10.0 (optimum, pH 7.5) and 10–40°C (optimum, 25–30°C). The major cellular fatty acids were C16:0, C16:1 ω7c/iso-C15:0 2-OH and C18:1 ω7c. The polar lipids consisted of phosphatidylethanolamine, phosphatidylglycerol and an unidentified phospholipid. The predominant respiratory quinone was Q-8 and the genomic DNA G + C content was 47.5 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain JSM 073008T should be assigned to the genus Alteromonas, being most closely related to Alteromonas hispanica F-32T (sequence similarity 96.9%), followed by Alteromonas genovensis LMG 24078T (96.6%) and Alteromonas litorea TF-22T (96.4%). The sequence similarities between the novel isolate and the type strains of other recognized Alteromonas species ranged from 95.9% (with Alteromonas stellipolaris ANT 69aT) to 94.5% (with Alteromonas simiduii BCRC 17572T). The combination of phylogenetic analysis, phenotypic characteristics and chemotaxonomic data supported the view that strain JSM 073008T represents a new species of the genus Alteromonas, for which the name Alteromonas halophila sp. nov. is proposed.
