Patterns of puffing activity in the salivary gland chromosomes of Drosophila   总被引:28,自引:0,他引:28  
Exposure of Drosophila melanogaster larvae to high temperature for short periods of time results in marked changes in the puffing patterns of salivary gland chromosomes. Temperature shock induces puffing at 9 specific loci; this pattern of induced puffs shows little developmental specificity and is similar in three strains of D. melanogaster (including the mutant lethal giant-larvae) and in D. simulans. Temperature shock also (i) retards the regression of some developmentally specific puffs and (ii) results in the regression of all other puffs normal to development. The effect of temperature treatment is similar in vivo and after in vitro treatment of salivary glands. The in vitro response is not sensitive to cycloheximide. A similar pattern of induced puffs to that found after temperature treatment is found during recovery of larvae from anoxia, but additional puffs are induced after anoxia. The size and duration of activity of the induced puffs is dependent upon the magnitude of the treatment.  相似文献

Titers of ecdysone and 20-OH ecdysone were measured separately in both hemolymph and salivary glands of metamorphosing Rhynchosciara larvae. Gland titers were consistently higher than hemolymph titers. Although 20-OH ecdysone was the most prominent form of the hormone, measurable quantities of ecdysone were also observed throughout development in both tissues. Changes in salivary gland replication and puffing activity could be correlated with changes in gland 20-OH ecdysone titers. This was true for both developmentally changing RNA puffs and DNA puffs, which occur during the prepupal period. The DNA puffs are tied to the final DNA replication cycle, and both this cycle and the period of amplification can be correlated with increases in gland 20-OH ecdysone content. Effect of mutations at the ebony or black locus on induction of heat shock puffs in polytene nuclei of salivary glands ofDrosophila melanogaster larvae were examined by [3H]uridine autoradiography. The levels of-alanine in the body are known to be increased by mutation at the ebony locus but decreased by mutation at the black locus. The presence of mutant allele/s at either locus in the homo- or heterozygous condition prevented induction of the 93D puff by heat shock. Elimination of the mutant allele at the ebony or black locus by recombination or by reversion of a P element insertion mutant allele of ebony restored the heat shock inducibility of the 93D puff. In vivo or in vitro administration of excess-alanine to salivary glands of wild-type larvae also resulted in the 93D site being refractory to heat shock induction. In agreement with earlier results, noninduction of the 93D puff during heat shock due to the-alanine effect was accompanied by unequal puffing of the 87A and 87C loci. The selective inducibility of the 93D puff by benzamide was not affected by ebony or black mutations or by excess-alanine in wild-type larvae  相似文献

Summary Patterns of protein synthesis in the salivary glands ofDrosophila melanogaster have been studied throughout late larval and prepupal development by pulse labelling the tissues with35S-methionine. Specific changes to the pattern of proteins synthesized during development are found and the significance of these changes is discussed in view of the known changes in gene (puffing) activity which occur at the same times. Incubation of Chironomus salivary glands with -amanitine in concentrations from 1 to 10 /ml results in the suppression of puffing and chromosomal 3H-uridine incorporation after 30 to 60 min in 80% of the cells. Nucleolar 3H-uridine incorporation remains completely unaffected. Even 4 h after the injection of high doses of -amanitine into living larvae, nucleolar incorporation is still pronounced. The distribution of resistant cells within the salivary glands suggests that the uptake of -amanitine is subject to physiological restrictions.—A puff typically induced during in vitro incubation of salivary glands was found to be less sensitive to -amanitine than the Balbiani rings.  相似文献

Summary Salivary glands of 9–10 day oldlethal giant larvae (l(2)gl, 2–0.0) respond to culture with ecdysone in a similar manner to mid third instar glands from wild type larvae. In particular, the ecdysone dependent puffs characteristic of pupariation, not previously observed in this mutant, are immediately induced. The secretion of the stored glue granules into the lumen of the salivary glands is also induced by ecdysone inl(2)gl.  相似文献

The hormone ecdysone induces a large number of changes in the puffing pattern of mid third instar larvae of Drosophila hydei. The pattern of changes occurring after experimental administration of the hormone are identical with those observed in normal development during a 6 hour period before puparium formation. After administration of the hormone a considerable number of puffs react with a change in activity within 15–20 min. During this period 3 puffs arise newly, 12 puffs show a strong increase in activity, 6 puffs show a less pronounced increase in activity and 12 puffs show a decrease in activity. At a period of 4–6 hours after administration of the hormone another 5 puffs arise newly. The effect of the hormone was identical in both in vivo and in vitro experiments. — Diameter measurements on several puffs reacting within 30 min with an increase in diameter showed that these puffs reacted simultaneously. Most of the puffs that showed a decrease in activity reacted with some delay. — A study of the effect of different hormone concentrations revealed that the kinetics of 4 puffs with respect to the relationship between concentration and puff size was identical over a range of concentrations from 33·10–5 to 33CU/l. Three of these puffs showed a reaction with even lower concentrations. Maximum puff size is attained by all puffs at a concentration of 33·10–4CU/l. Among the puffs studied no difference in their reaction threshold was found. — A study of the behavior of 5 puffs of the group reacting within 15–20 min and one of the group reacting after 4–6 hours in midintestine and Malpighian tubules revealed that these puffs showed the same reaction after injection of the hormone as observed in the salivary glands. — All puffs activated by administration of the hormone showed particularly strong uptake of tritiated uridine and accumulation of acidic protein. — It is concluded that the hormone ecdysone induces a pattern of changes in gene activity that is far more complex in Drosophila hydei than in Chironomus tentans.  相似文献

The effect of injection of various concentrations (4 ng to 0.5 g/larva) of -amanitin on chromosomal RNA synthesis in larval salivary glands of D. hydei was investigated at subsequent time intervals (90 min to 20 hrs) after injection. — As revealed by autoradiography, 3H-uridine incorporation into the polytene chromosomes was greatly reduced as compared with that in control larvae. In -amanitin injected larvae, new chromosome puffs could be induced by a temperature treatment. These puffs never attained a size comparable to that in the controls. The newly induced puffs did not show specific incorporation of 3H-uridine. — Puffs which were active before the toxin was applied undergo a reduction in size and incorporation of 3H-uridine.  相似文献

Summary The late larvae of Drosophila gibberosa Patterson and Mainland choose different pupariation sites than the larvae of Drosophila melanogaster Meigen. Since the larvae of D. gibberosa do not attach themselves to the substratum, the salivary glands contain only a small amount of the glue proteins before pupariation. Proteins comprising the salivary gland secretions of late larvae of these two species were compared and found to be qualitatively quite different. Only five polypeptides with the same molecular masses were identified in both species. The rate of protein synthesis in the salivary glands of D. gibberosa continued to increase through the late larval stage and pupariation. As a consequence, the total amount of protein contained in the salivary glands also continued to increase after pupariation. To demonstrate temporal changes in protein synthesis from 48 h before pupariation to 28 h after pupariation, newly synthesized polypeptides were pulse labeled by culturing salivary glands in vitro. The patterns of polypeptide synthesis fell into four major groups depending upon whether the synthesis of a protein stopped shortly after pupariation, stopped during late pupariation, increased at pupariation, or was initiated after pupariation. Changing patterns of protein synthesis are correlated with the known changes in gene puffing during this developmental period.  相似文献

A portion of the X chromosome was examined for puffing activity shortly before puparium formation in several species of Hawaiian Drosophila. A large puff was induced by incubating excised salivary glands with ecdysone. In two of the seven species studied, the region in question has been moved by inversion to a new position relative to the banding sequence in the other species, but the reaction to ecdysone remained unchanged. Another puff, not affected by short term incubations with ecdysone, displayed different degrees of activity in some homosequential species, affording a cytological means for distinguishing these species.  相似文献

Modeling of morphologically unusual dark puffs was conducted using Drosophila melanogaster strains transformed by construct P[ry; Prat:bw], in which gene brown is controlled by the promoter of the housekeeping gene Prat. In polytene chromosomes, insertions of this type were shown to form structures that are morphologically similar to small puffs. By contrast, the Broad-Complex (Br-C) locus, which normally produce a dark puff in the 2B region of the X chromosome, forms a typical light-colored puff when transferred to the 99B region of chromosome 3R using P[hs-BRC-z1]. A comparison of transposon-induced puffs with those appearing during normal development indicates that these puff types are formed via two different mechanisms. One mechanism involves decompaction of weakly transcribed bands and is characteristic of small puffs. The other mechanism is associated with contacts between bands adjacent to the puffing zone, which leads to mixing of inactive condensed and actively transcribed decondensed material and forming of large dark puffs.  相似文献

Four distinctly crossbanded, stout polytene chromosomes are present in the nuclei of both the basal reservoir region and gland proper region of salivary glands of young larvae of the Cecidomyid Dasyneura crataegi. In older larvae, asynchronous progressive splitting of the chromosomes into oligotene fibrils occurs, underlining their truly polytene nature. Three nucleoli are present, located on two of the chromosomes. A series of massive puffs is also organised by one of the nucleolar chromosomes. Three other features of interest shown by the chromosomes of this species are (a) the centromeric association of only two, the nucleolar organising, chromosomes of the four present; (b) the high breakability of the centromeric regions of these two chromosomes; and (c) the marked heterochromatin proliferation which is found at these regions in older larvae. As in most Cecidomyids, the salivary glands are of complex structure with anterior basal reservoir and posterior gland proper zones. Marked differences in the relative and absolute sizes of these two regions are found during the development of the glands, which indicate that their names are inappropriate to their probable functions.  相似文献

Summary Morphological and metabolic changes on the salivary chromosomes of Sciara coprophila were followed during the later half of the fourth larval instar.Cytological maps were prepared for five successive stages from mid-fourth instar to the prepupal stage. These maps, which constitute a revision of those published earlier by Crouse, summarized our cytological findings and were the basis for studies on DNA replication of these chromosomes.Similar to earlier studies in Chironomidae, differences in the puffing pattern were noted between the anterior and the posterior portions of the salivary gland. The most striking difference was noted in region 2B on chromosome III which produces a large puff only in nuclei from the anterior part of the gland. Other autosomal puffs, although present in both parts of the gland, showed constant differences in size.An increase in the number of bands from mid-fourth to late fourth instar was observed. The new bands are all of the light-staining kind.In Sciara the puffed area may include a large number of bands in addition to the bands which originated the puff. The maximal extent of puffs was determined in terms of chromosomal map regions and the number of bands subject to obliteration.In the autoradiographic experiments use was made of H3-thymidine as DNA precursor. The aim of these studies was to detect any asynchronies in the replication time of bands. In fact, marked differences in the relative rates of uptake of H3-thymidine of a number of bands in a certain proportion of chromosomes have been observed, while others showed uniform incorporation. Since these latter were found with higher frequency the period of uniform labeling must comprise a larger part of the replication cycle then the periods of localized labeling. To assess the validity and constancy of the observed patterns of unequal incorporation, a semiquantitative analysis was carried out. It showed that the bands showing localized uptake may be separated into two broad groups. In one of these groups are the centromere regions and certain chromosomal ends, which are presumably heterochromatic. The other group comprises most of the puff sites and bulbs. Since late replication is characteristic of heterochromatin, we assumed that bands of the former group (C) replicate late in the cycle, while puffs and bulbs start replication early, and the period of equal labeling is intermediate. Other intermediate labeling patterns were observed and are described.It is known that in the fourth instar from two to three DNA replications occur in the salivary gland nuclei, the last of which coincides with puffing. Several stages may be distinguished in the puffing process based on morphology and rates of isotope uptake of the puffs. The first sign of puffing is a very high rate of incorporation at puffs. It is maintained throughout this last DNA synthesis period and only declines when all other chromosomal regions have ceased to replicate. A pattern of high and exclusive uptake at the heterochromatic sites (pattern C) was never observed in this replication; instead puffs are the last regions to terminate DNA synthesis.These results are discussed in relation to several current problems, such as, asynchronous DNA replication, the problem of metabolic DNA, and the concept of the heterochromatic state.Submitted in partial fulfillment of the requirements for the degree of Doctor of Philosophy, in the Faculty of Pure Science, Department of Zoology, Columbia University, New York. The N-banding patterns of the polytene chromosomes of Drosophila melanogaster, Chironomus melanotus, Ch. th. thummi and Ch. th. thummi x Ch. th. piger were studied. In Chironomus the polytene N-banding patterns correspond to the polytene puffing patterns. This is revealed by comparison of the puffing and N-banding patterns of identical chromosomes. Size and staining intensity of the N-bands reflect the size of the puffs as shown by puff induction. There is no evidence that the N-bands are also located in Chironomus heterochromatin or are restricted to the nucleolar organizer regions. In Drosophila the -heterochromatin is strongly N-positive, whereas the -heterochromatin, as well as the Chironomus heterochromatin is not N-banded. Contrary to Chironomus, the puffs in Drosophila polytene chromosomes do not give rise selectively to well stained N-bands. — The N-banding method is interpreted to stain specifically non-histone protein which is (1) accumulated in genetically active chromosome regions and (2) present in a specific type of heterochromatin (-heterochromatin of Drosophila).  相似文献

Modifications in the synthesis of salivary gland RNA were induced by treatments with 10 /ml cycloheximide (CHM) on 4th instar larvae of Chironomus pallidivitattus. After 3, 6 and 24 h CHM treatment, RNA was labeled in vitro, by incubating the salivary glands in a medium containing H3-uridine. The electrophoretical analyses corresponding to the 3 and 6 h treatment showed a stimulation of the non-ribosomal components of the newly synthesized RNA, while preribosomal RNA synthesis appeared depressed. This fact was also confirmed at cytological level, since autoradiograms made after 3 h of CHM treatment showed a reduced H3-uridine label over the nucleolus and an increase of diffuse labeling over the chromosomes. Longer treatments (24 h) causes a considerable inhibition of the synthesis of all RNA species. The role played by protein synthesis inhibition in the aforementioned effects is discussed. — Some of the morphological implications of CHM treatment, such as modifications of the nucleolar structure (nucleolar segregation) are also reported. The use of a squash technique based on glutaraldehyde fixation of the salivary glands, considerably facilitates such studies.  相似文献

Summary Parotid, submandibular and sublingual glands were removed from rats and investigated histochemically. Pyruvate oxidase, iso-citric dehydrogenase, -ketoglutarate oxidase, succinic dehydrogenase, malate dehydrogenase and furfuryl alcohol dehydrogenase activity were observed in the salivary ducts which may be interpreted as significant of high metabolic activity.The 11 -hydroxysteroid dehydrogenase in these ducts displayed marked substrate specificity utilizing 11-hydroxyandrostenedione and cortisol but not 11 -hydroxyoestrone or 11 -hydroxyprogesterone. The relationship between corticosteroids and salivary electrolyte concentrations is discussed.  相似文献

Liver homogenate-supernatants from most Japanese exhibit an atypical pH optimum for ethanol oxidation at pH 8.8 instead of 10.5, the typical pH-activity optimum. It has been proposed that atypical livers contain alcohol dehydrogenase isozymes with 2 subunits while typical livers contain isozymes with 1 subunits, both produced by the ADH 2 gene. Because it is difficult to differentiate the atypical ADH2 2-2 phenotype from the ADH2 2-1 phenotype by starch gel electrophoresis, an agarose isoelectric focusing procedure was developed that clearly separated the atypical Japanese livers into two groups, A1 and A2. The isozymes in A1 and A2 livers were purified. Type A1 livers contained a single isozyme with an atypical pH-rate profile; it was designated 22. Three isozymes were isolated from A2 livers, two of which corresponded to 11 and 22. A third, absent from the typical and the atypical A1 livers, had an intermediate mobility; it was designated 21. Type A1 livers are, therefore, the homozygous ADH2 2-2 phenotype, and type A2 livers, the heterozygous ADH2 2-1 phenotype. The ADH2 2-2 phenotype was found in 53% of 194 Japanese livers, and the ADH2 2-1 phenotype, in 31%. During five hours after the injection of -ecdysone into the hemolymph of D. virilis late third instar larvae the formation of larval glycoproteins in the salivary glands is terminated and the synthesis of a different set of glycoproteins which is characteristic for the prepupal gland is initiated. The data presented suggest that products from early puffs inhibit the formation of larval glycoproteins while the induction of late puffs may be responsible for the appearance of prepupal glycoproteins.  相似文献

Summary Endo--galactosidase from Escherichia freundii cleaves polylactosaminyl structures as follows: R-GlcNAc1-3Gal1-4GlcNAc1-R + H2O R-GlcNAc1–3Gal + GlcNAc1-R. By staining with Griffonia simplicifolia agglutinin-II following the enzyme digestion, the distribution of R-GlcNAc1–3Gal1–4GlcNAc can be demonstrated in tissue sections. This carbohydrate chain is one of the backbone structures carrying the blood-group-related antigens and, thus, localization of this structure may provide detailed information about the distribution of variants with different backbone structures. Various formalin-fixed, paraffin-embedded tissue sections were stained by Griffonia simplicifolia agglutinin-II with or without prior enzyme digestion and the reactivity of the agglutinin imparted by enzyme digestion was studied in the following tissues and cells: pancreatic acinar cells, gastric surface mucosae, duct cells and mucous cells of salivary glands and tracheal glands, surface epithelium of trachea, goblet cells of large intestine, columnar epithelium of uterine cervical glands, distal and collecting tubules of kidney, certain cells of anterior lobe and colloid of middle lobe of pituitary glands, epithelial reticular cells and Hassall's corpuscles of thymus and Kupffer cells of liver. In gastric surface mucosae, the reactivity of the agglutinin appeared in non-secretor individuals but not in the secretor individuals, and in mucous cells of salivary and tracheal glands the reactivity appeared in Le(a - b -) non-secretor individuals but not in Le(a + b -) non-secretor or secretor individuals. In pancreatic acinar cells and duct cells of salivary glands from fetuses and newborn infants, prior fucosidase digestion markedly enhanced the Griffonia simplicifolia agglutinin-II reactivity elicited by endo--galactosidase digestion. Prior fucosidase digestion was also a prerequisite for revealing the reactivity of this agglutinin by endo--galactosidase digestion in gastric surface mucosae from secretor individuals. -Galactosidase digestion disclosed reactivity of this agglutinin in pancreatic acinar cells and duct cells of salivary glands even after the removal of endo--galactosidase-labile lactosamine structures by sequential digestion with endo--galactosidase and -N-acetylhexosaminidase. These results demonstrate that the procedures developed in this study provide a useful means for detecting different types of lactosamine structures which carry blood-group antigens in humans tissues.  相似文献

The avermectins, broad-spectrum anti-parasitic agents which are effective against ticks, act principally by stimulating -aminobutyric acid (GABA) receptors directly or indirectly. GABA is known to potentiate dopamine-induced fluid secretion in the salivary glands of female ixodid ticks. We thus tested wheter 10 M ivermectin would have an effect similar to GABA in the isolated salivary gland preparation ofDermacentor andersoni.
