从河南省临颖县采集的病毒感染的甜瓜样本经ELISA检测和接种分离获得黄瓜花叶病毒(Cucunbermosaicvirus,CMV)分离物。把该分离物接种西葫芦,从发病的叶片中提取总RNA,并以此为模板经RTPCR扩增获得CMV的外壳蛋白(cp)基因,将其克隆到pUCmT质粒上。经序列测定和分析,结果表明该cp基因由657个核苷酸组成,编码218个氨基酸。其核苷酸序列与黄瓜花叶病毒亚组I的分离物有较高的同源性,达92.2%~93.9%,与亚组II的同源性仅为76.8%~77.8%,与我国报道的CMV分离物的cp基因序列比较,除香蕉株系XB外核苷酸序列的同源性达91.8%~93.4%。根据这些分析,该CMV分离物属于亚组I。将cp基因通过中间载体pJIT163定向克隆到植物表达载体pBINPLUS中(重组双元载体质粒命名为pBCP),并经冻融法导入农杆菌中,经PCR及酶切鉴定,证实质粒已被导入。利用该植物表达载体对西瓜的遗传转化工作目前正在进行中。  相似文献

应用ELISA的方法检测到感染了黄瓜花叶病毒(Cucumber mosaic virus,CMV)的万寿菊。根据已报道的CMV外壳蛋白(CP)基因的保守序列设计并合成引物,提取万寿菊叶片总RNA为模板,进行cDNA合成和PCR扩增,得到约875bp的片断,与预期片断大小相符。序列分析显示:该分离物CMV-WSJ CP基因全长657bp,编码218个氨基酸,其核苷酸和氨基酸序列与CMV亚组Ⅱ的分离物有很高的同源率,分别达到98.33%~99.24%和98.63%~100%;与CMV亚组I分离物的同源率分别仅为74.43%~76.26%和77.17%~78.99%。因此,从万寿菊叶片上检测出的黄瓜花叶病毒分离物CMV-WSJ应归属于亚组Ⅱ。  相似文献

通过间接酶联免疫法(ID-ELISA)检测到染病落葵病样中存在黄瓜花叶病毒(Cucumber Mosaic Virus,CMV)。从病叶中提取总RNA,用RT-PCR方法扩增得到657bp的CMV CP基因片断,将扩增产物与T载体连接并进行测序。用DNA MAN将得到的CP基因序列与GenBank收录的黄瓜花叶病毒两亚组部分株系或分离物的CP基因序列进行比较,结果表明该CP基因与CMV亚组Ⅰ、亚组Ⅱ之间的核苷酸序列同源性分别为91.17~95.43%和75.30~75.76%,推导氨基酸序列同源性分别为95.41~97.71%和81.28~81.74%,表明CMV-Ba与亚组Ⅰ同源关系密切。  相似文献

在对云南省烟草病毒病的研究中,分离到一种直径约26~30nm的球形病毒。提纯病毒进行的SDSPAGE发现一条55kD蛋白带。55kD蛋白N端10个氨基酸与CMV亚组II的Q株系外壳蛋白N端氨基酸同源性为100%。以CMVQ抗血清对55kD蛋白进行了Western blot检测,发现55kD蛋白与CMV Q株系抗血清有血清学反应。根据已报道的CMV亚组II外壳蛋白基因序列合成引物,采用RTPCR技术扩增到一条约09kb的cDNA条带,并进行了克隆及序列测定,经Genbank比较,发现此09kb cDNA包含一657bp的外壳蛋白基因,其核苷酸序列及其推导的氨基酸序列与CMV亚组II分离物有极高的同源性,分别达97%~98%和96%~99%,而与亚组I分离物的同源性仅为75%~76%和78%~79%。因此,该球形病毒应为CMV亚组II的一个分离物,命名为CMVYnb。  相似文献

从吉林长春感病辣椒上获得一黄瓜花叶病毒(Cucumber mosaic virus,CMV)分离物(CMV-CC),根据GenBank中已登录的CMV外壳蛋白(coat protein,CP)基因核苷酸序列设计简并引物,通过RT-PCR的方法克隆到了长度为657 bp的目的片段。序列分析表明,CMV-CC与CMVI组各分离物核苷酸同源性为93.2%-97.9%。根据完整CP基因核苷酸序列构建的系统进化树显示:38个CMV分离物可分为3个组,CMV-CC属于CMV的IB亚组。将CMV-CC CP基因与原核表达载体pET-22b(+)连接,在大肠杆菌BL21(DE3)诱导表达出分子量约27 kD的融合蛋白。表达的融合蛋白经树脂纯化后免疫家兔制备了抗血清。用间接ELISA测定抗血清效价为1/4 096。Western blotting分析表明制备的抗血清对CP有高度特异性,为准确、快速地检测CMV奠定了基础。  相似文献

黄瓜绿斑驳花叶病毒辽宁分离物全基因组序列测定   总被引:7,自引:0,他引:7  
以感病组织总RNA为模板,采用RT-PCR方法扩增并测定黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)辽宁分离物(CGMMV-LN)的基因组全序列。CGMMV-LN基因组全长6 422 nt,5'非编码区(noncoding region,NCR)和3'NCR分别为59 nt和175 nt。CGMMV-LN编码的4个蛋白依次是186 kD和129kD的复制酶,29 kD的移动蛋白和17.4 kD的外壳蛋白。CGMMV-LN与其他4个CGMMV分离物基因组核苷酸序列同源性为97.6%~99.3%,与同属其他3种病毒基因组核苷酸序列同源性仅为61.7%~62.8%。基于186kD复制酶和外壳蛋白氨基酸序列的同源树显示:侵染葫芦科作物的烟草花叶病毒属病毒可分为2个亚组,亚组I包括所有CGMMV分离物,亚组II包括Kyuri绿斑驳花叶病毒(Kyuri green mottle mosaic virus,KGMMV)、黄瓜果实斑驳花叶病毒(Cucumber fruit mottle mosaic virus,CFMMV)和小西葫芦绿斑驳花叶病毒(Zucchini ...  相似文献

利用非序列依赖性扩增(Sequence-independent amplification,SIA)方法对所采柴胡病样进行分子鉴定。序列测定及分析发现,伴有花叶症状的柴胡受到黄瓜花叶病毒(Cucumber mosaic virus,CMV)和蚕豆萎蔫病毒2号(Broad bean wilt virus 2,BBWV2)的复合侵染。为明确柴胡CMV分离物(CMV-SXCH)和柴胡BBWV2分离物(BBWV2-CH)的分类地位,进一步克隆CMV-SXCH的CP、MP和BBWV2-CH的LCP、SCP;序列比对发现,CMV-SXCH与CMV亚组ⅠB中株系XJ2相似性最高,其核苷酸和氨基酸序列同源性分别为99.1%、99.5%;BBWV2-CH与BBWV2中的K株系相似性最高,核苷酸和氨基酸序列同源性分别为95.4%、99.2%。系统进化树分析表明,CMV-SXCH与大多数CMV中国分离物聚类为一簇,同属于CMV亚组ⅠB;BBWV2-CH与BBWV2韩国K株系聚类为一簇,亲缘关系最近。  相似文献

为澄清浙江省豇豆病毒病病原及其分类,采用马铃薯Y病毒属通用引物Sprimer扩增了浙江豇豆线状病毒基因组3′-末端序列。同时又根据已知CMV RNA3序列设计引物pCMV-44-63/1200-1181,扩增了混合的CMV-ZJ运动蛋白基因全序列。外壳蛋白氨基酸序列分析表明,产中仅存在一种线状病毒(CV-ZJ),该病毒与一泰国豇豆蚜传花叶病毒(CABMV)具有最高的同源性(98.6%),与花生条纹病毒(PStV)、石斛花叶病毒(DeMV0.)、赤豆花叶病毒(AZMV)、黑眼豆豆花叶病毒(BICMV)和菜豆普通花叶病毒(BCMV)分离物间的同源性为88.5%-94.4%,而与其它CABMV分离物的同源性均低于70%。进化树分析表明,PStV、AZMV、DeMV、BlCMV和BCMV为同种异名,应统称为BCMV,而CABMV为另一种病毒;CV-ZJ和泰国CABMV分离物应分类为BCMV豇豆株系。CMV-ZJ的运动蛋白序列分析表明,该分离物属于CMV亚组Ⅰ,与其它分离物氨基酸序列同源性为93.1%-97.8%。  相似文献

目的:克隆芜菁花叶病毒(Turnip mosaic virus,TuMV)的3'末端序列,并进行CP基因序列分析.方法:以TuMV杭州榨菜分离物(TuMv-HZZC)接种病叶为材料,利用病毒粒子吸附法制备病毒RNA模板,经RT-PCR扩增获得了TuMV-HZZC 3'末端序列,将其克隆到PMD 18-T质粒上进行序列分析.结果:TuMV-HZZC分离物3'末端序列包括部分的Nib基因、完整的TuMVCP基因和3'-UTR,CP基因为864bp,分别编码288个氨基酸,3'-UTR序列(不包括PolyA尾巴)为213bp.经过与其他TuMV分离物的CP基因核苷酸和氨基酸比较,同源性分别达到88.0%~97.6%和91.0%-96.5%.结论:TuMV的系统进化具有典型的地域和寄主关联性.  相似文献

烟草环斑病毒(Tobacco ringspot virus,TRSV)是我国二类进境检疫危险性有害生物,对农业生产危害较大.本研究依据TRSV外壳蛋白基因cp序列设计合成了2条引物,通过RT PCR扩增得到长约1500bp的目的片段.将目的片段与质粒pET-22b(+)连接,构建了含TRSV cp基因的融合蛋白原核表达载体pETRSV-CP.序列分析表明,TRSV-SD1的cp基因全长1548bp,编码515个氨基酸与GenBank中其它TRSV分离物cp基因相比,核苷酸及推导的氨基酸序列同源性为90.7%~94.6%.将pETRSV-CP转入大肠杆菌,诱导表达.SDS-PAGE结果显示,表达的TRSV CP融合蛋白的相对分子质量约为58kDa.以此融合蛋白制备的抗血清的效价为1/1024,抗血清与TRSV具有良好的特异性反应.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Fluid collected from hatching eggs of Haemonchus contortus contained a lipase which hydrolysed 2-naphthyl laurate (about 0·7 μmol naphthol freed /h/106 eggs). The fluid also hydrolysed l-leucinamide (about 2·3 μmol leucine freed/h/106 eggs). The fluid when added to normal or heated eggs caused ‘hatching’. ‘Hatching’ also occurred in exsheathing fluid from infective juveniles and in a preparation of pancreatic lipase containing leucine aminopeptidase. A purified mammalian leucine aminopeptidase in combination with several different lipases did not attack egg shells.The ‘spontaneous’ hatching of eggs of H. contortus was strongly inhibited by 1,10-phenanthroline, 10?3M, and this inhibition was reversed by Zn2+. However, the inhibition of ‘hatching’ of eggs in externally applied hatching fluid, or the hydrolysis of leucinamide in hatching fluid was generally less marked.  相似文献
