Amphitetranychus quercivorus (Ehara and Gotoh, 1990) was first identified in Mongolian oak tree Quercus mongolica in Daegu, Korea. In comparison to A. viennensis (Zacher), A. quercivorus was much shorter in the distal, dorsally directed portion of the male aedeagus, as well as smaller and less folded in the distal anastomosed portion of the female peritreme. Comparison of the nucleotide sequences of ITS2 and COI revealed 90% and 89% similarities, respectively, between the two species. In addition, species-specific primer sets for each species were designed using unique ITS2 sequences and then used to diagnose these two species under plant quarantine. We report the Korean name of this species as “Sin-gal-na-mu-eung-ae”.  相似文献

Leaf blight symptom caused by Gibberella moniliformis was observed in the seedlings of Kentucky bluegrass (cv. Perfection) in Jinju, Korea, in January 2013. The symptoms first appeared as brown to yellow lesions on leaves, which later became white and bleached. The pathogen was isolated from turfgrass leaves and identified as G. moniliformis by using the internal transcribed spacer (ITS)1 and ITS4 sequences of rRNA. G. moniliformis was confirmed by a pathogenicity test, and the causal agent was reisolated from infected turfgrass.  相似文献

The population genetic structure of the razor clam Sinonovacula constricta was investigated between populations collected from China, Korea and Vietnam using cytochrome oxidase subunit 1 (COI) mtDNA and internal transcribed spacer 2 (ITS2) rDNA markers. A total of 622 bp of COI and 495 bp of ITS2 were sequenced. Highly significant Fst values and low rates of migration were observed from populations between Vietnam and China, (Fst = 0.7578, P < 0.001, Nm = 0.05 for COI; Fst = 0.91389, P < 0.001, Nm = 0.07 for ITS2) and Korea (Fst = 0.79783, P < 0.001, Nm = 0.07 for COI; Fst = 0.74143, P < 0.001, Nm = 0.03 for ITS2). However, lower Fst values and higher gene flow were detected between populations from China and Korea (Fst = 0.25733, P < 0.001, Nm = 0.73) based on COI analysis. The similar pattern was also captured by ITS2 rDNA marker. In addition, Neighbor-joining phylogenetic tree analysis of the two markers resulted in one cluster consisting of haplotypes from Vietnam, and a second group comprising haplotypes from China and Korea. This research revealed a closer genetic relationship between clam population from China and Korea but less similarity to population from Vietnam.  相似文献

The locally extinct dung beetle, Gymnopleurus mopsus Pallas, 1781 (Coleoptera: Scarabaeidae), has not been found in South Korea since the 1970s. This research was conducted to understand the genetic divergence between the South Korean and Mongolian populations of G. mopsus as a part of its reintroduction program in South Korea. The genetic distance and diversity were determined using the mitochondrial cytochrome c oxidase subunit I (COI) gene sequence (658 bp ) corresponding to the DNA barcode region. The mitochondrial COI gene sequences of 64 individuals of G. mopsus collected in South Korea (7 individuals) and Mongolia (57 individuals) showed a relatively high level of genetic diversity (nucleotide diversity, 0.0078 ± 0.0007; Haplotype diversity, 0.965 ± 0.017). The genetic distances between the South Korean and Mongolian populations lay within the intraspecific level and the phylogenetic reconstruction using the neighbor‐joining (NJ) method showed that all individuals belonged to a single clade. This result indicates that the current Mongolian population of G. mopsus is a good candidate source population to restore the locally extinct population of the species in South Korea.  相似文献

In the present study, specimens of Dendrolimus superans collected from South Korea suggest the presence of D. sibiricus, instead of D. superans. Comparisons of the wing morphology, female and male genitalia, 3′‐end region of the cytochrome c oxidase I (COI) gene sequence, and internal transcribed spacer (ITS) 2 sequence of the Korean specimens with those of the D. superans specimens from Japan consistently supported the presence of D. sibiricus in South Korea. Phylogenetic analyses of the concatenated sequences of COI and ITS2 from the available sequence types of D. sibiricus and D. superans, along with South Korean specimens, were conducted using the Bayesian inference (BI) and maximum likelihood (ML) methods. These phylogenetic analyses placed the South Korean specimens with the Russian D. sibiricus as an inclusive group, excluding the Japanese D. superans, indicating the distribution of D. sibiricus in South Korea. Nevertheless, D. superans formed a distinct group only by BI analysis (Bayesian posterior probabilities = 0.89), whereas D. sibiricus, including the South Korean samples, formed a distinct group only by the ML analysis (99%), suggesting a low genetic divergence between the two species.  相似文献

The dung beetle, Copris tripartitus (Coleoptera: Scarabaeidae), is an endangered insect in Korea. In order to establish a conservation strategy, a preliminary investigation on the genetic diversity of Korean populations was performed using mitochondrial COI (658 bp), CytB (433 bp), and nuclear ITS2 (411 nucleotide positions). Sequencing of 69 individuals collected from five localities showed substantially higher variability (5.02% for COI, 4.62% for CytB, and 8.03% for ITS2). The resulting networks for mitochondrial DNA (mtDNA) haplotypes exhibited two star‐like phylogenies, which might indicate that Korean populations have recently expended from two small populations. The ITS2 network, which was presented in the form of a star‐like phylogeny, confirmed that a recent population expansion occurred. Considering the high genetic diversity and gene flow in C. tripartitus populations, one issue regarding conservation seems to be the recovery of previous habitats.  相似文献

With about 5000 known species, the Vespidae is a large family belongs to order Hymenoptera. The genus Vespa with 22 species is one of the four genera of the subfamily Vespinae. In Korea, 10 species and subspecies are recognized. Because of their social behavior, their treat to human health and their impact in apiculture, the reliable and sometimes automated identification of these insects to species level are important. To test the efficacy of DNA barcoding method for identification of species of the genus Vespa in Korea, 30 samples of eight Korean species of genus Vespa were collected and mitochondrial DNAs of 658 bp fragment cytochrome oxidase subunit 1 (CO1) region were sequenced. A Bayesian Inference based on COI gene of the Korean Vespa species was constructed. The phylogenetic tree shoed that identification of all specimens is possible based on COI gene and we found strong relation between the sequences of the collected species from different localities in South Korea which clustered together with 100% support with sequences of the same species in GenBank. The results demonstrate that DNA barcoding is a useful technique for rapid and accurate species recognition in Korean Vespa species. The DNA barcode part of COI for V. binghami is provided for the first time that can help for identification of this species through DNA barcoding. Also, the genetic diversity among Korean Vespa velutina was zero suggests that the invasion might have occurred in a single event with small number of founders.  相似文献

To understand the evolutionary relationship among six Tetranychus species in Korea, 598 nucleotides of cytochrome oxidase subunit I (COI) were sequenced and analyzed. The sequences were extremely rich in A+T (77% on average). The nucleotide differences in pairwise comparison among species ranged from 6.7% to 14.9%. Nucleotide substitutions based on all pairwise comparison showed 55% of transversions. The largest genetic distance was found between species T. viennensis and T. piercei. The inferred phylogenetic tree indicates that T. urticae, T. cinnabarinus and T. kanzawai are clustered together and showing sister-group relationship with T. truncatus at the highest bootstrap support. T. viennensis showed the largest genetic distance from all other species. T. piercei, a species recently found in Korea, was also distinct from the group composed of four species and also from T. viennensis. The restriction sites of Alu I, Dde I and Sau 3A were searched in the sequences of COI. The restriction fragment length polymorphisms (RFLPs) in COI genes and intraspecific variabilities of three restriction sites in diverse colonies of each species collected from different locations and various host plants were shown.  相似文献

DNA barcoding involves the use of one or more short, standardized DNA fragments for the rapid identification of species. A 648‐bp segment near the 5′ terminus of the mitochondrial cytochrome c oxidase subunit I (COI) gene has been adopted as the universal DNA barcode for members of the animal kingdom, but its utility in mushrooms is complicated by the frequent occurrence of large introns. As a consequence, ITS has been adopted as the standard DNA barcode marker for mushrooms despite several shortcomings. This study employed newly designed primers coupled with cDNA analysis to examine COI sequence diversity in six species of Pleurotus and compared these results with those for ITS. The ability of the COI gene to discriminate six species of Pleurotus, the commonly cultivated oyster mushroom, was examined by analysis of cDNA. The amplification success, sequence variation within and among species, and the ability to design effective primers was tested. We compared ITS sequences to their COI cDNA counterparts for all isolates. ITS discriminated between all six species, but some sequence results were uninterpretable, because of length variation among ITS copies. By comparison, a complete COI sequences were recovered from all but three individuals of Pleurotus giganteus where only the 5′ region was obtained. The COI sequences permitted the resolution of all species when partial data was excluded for P. giganteus. Our results suggest that COI can be a useful barcode marker for mushrooms when cDNA analysis is adopted, permitting identifications in cases where ITS cannot be recovered or where it offers higher resolution when fresh tissue is. The suitability of this approach remains to be confirmed for other mushrooms.  相似文献

DNA barcode (mitochondrial COI) sequences are provided for species identification of aphids from the Korean Peninsula. Most (98%) of the 154 species had distinct COI sequences (average 0.05% intraspecific pairwise divergence) relative to the degree of sequence divergence among species (average value 5.84%). For species in common with other regions, barcodes for Korean samples fell near or within known levels of variation. Based on these results, we conclude that DNA barcodes can provide an effective tool for identifying aphid species in such applications as pest management, monitoring and plant quarantine.  相似文献

The taxonomic status of an alga with complanate thalli, occurring in central Chile and belonging to the genus Scytosiphon, was elucidated. Morphological and molecular features demonstrated that, in addition to the known and widespread constricted S. lomentaria (Lyngb.) Link that occurs along the Chilean coast, there is a Scytosiphon with complanate thalli that occurs only in central Chile—S. gracilis Kogame. Morphological analyses of this previously unreported complanate Scytosiphon showed thalli without constrictions, coherent plurilocular sporangia without ascocysts, and phaeophycean hairs arising from cortical cells. Furthermore, sequences of the internal transcribed spacer (ITS1 and ITS2) regions of the complanate Scytosiphon were 99.8% and 100% identical to those detected in S. gracilis from Korea. ITS‐based comparative analyses showed that complanate Scytosiphon grouped in a different clade than S. lomentaria and S. tenellus Kogame from various parts of the world, including Chilean species. Moreover, molecular analyses suggest the occurrence of two distinct ITS types of S. lomentaria in northern Chile, corresponding to the Korean and Greek types. On the other hand, biochemical analyses of copper‐induced antioxidant responses in S. gracilis and S. lomentaria showed an identical increase in antioxidant enzyme activities. These results suggest that copper tolerance might be a constitutive trait in these species of Scytosiphon.  相似文献

The slug Geomalacus maculosus is a prominent member of the Lusitanian fauna. As its global distribution is restricted to western Ireland and northern Iberia, it is protected under EU legislation. Nothing is known about the genetic variability and population structure of this species, so, with a special view to shedding light on the origin of the Irish G. maculosus, tissue samples from 78 specimens were collected from 13 locations within Ireland and ten locations within Iberia and partial sequences of the mitochondrial 16S rRNA and cytochrome oxidase subunit 1 (COI) and from the nuclear internal transcribed spacer 1 region (ITS‐1) were compared. The genetic diversity of the Irish G. maculosus was found to be greatly reduced compared with the Iberian populations, with only one (16S rRNA) and two (COI) mitochondrial haplotypes identified respectively. No private Irish ITS‐1 haplotype was found. Based on the COI sequences, the Irish specimens clustered closest to Spanish specimens from Northern Asturias and Cantabria, and the bGMYC analysis identified five further Iberian clades that were highly genetically differentiated suggesting long‐term allopatric divergence.  相似文献

Molecular diagnosis is highly valuable for the species identification of microscopic mites. Here, we collected some economically important mites of the superfamily Acaroidea from various stored products in Korea. Those nucleotide sequences of ribosomal second internal transcribed spacer (ITS2) and the mitochondrial cytochrome oxidase subunit I (COI) regions were determined by PCR using universial primers. In nucleotide sequence comparison at GenBank database, seven species including Rhizoglyphus robini, R. echinopus, Sancassania sp. Acarus siro, Tyrophagus putrescentiae, T. similis in Acaridae and Suidasia medanensis in Suidasiidae were identified. Particularly, COI sequences of R. robini, R. echinopus, Sancassania sp. and T. similis were firstly determined. Our results suggest that the phylogenetic relationships inferred from the ITS2 region, rather than COI region, were similar to those derived based on their morphological classification. Our study provides molecular information for the identification and phylogenetic relationship of acaroid mites in Korea.  相似文献

Mites are one of the serious pests of turfgrass. Our survey of turfgrass fields from 2013 to 2015 in Korea showed that the occurrence of leaf chlorotic symptom has gradually extended to at least 60% of the examined golf courses. We identified the zoysiae mite Aceria zoysiae in most damaged zoysiagrasses. Artificial infestation of A. zoysiae into zoysiagrasses in pots resulted in symptoms of chlorosis and marginal rolling of the leaves within 3 weeks. We firstly determined the nucleotide sequence of mitochondrial cytochrome c oxidase subunit I (COI) and internal transcribed spacer 2 (ITS2) of the nuclear ribosomal DNA region of A. zoysiae. The variations in COI and ITS2 between A. zoysiae and other species of the genus were 20.9%–43.0% and 7.5%–67.3%, respectively, suggesting significant genetic divergence within the genus. Our study provides valuable information for the rapid diagnosis and infestation monitoring of A. zoysiae in turfgrass fields.  相似文献

The water deer (Hydropotes inermis) is one of the rare small ruminants in the world. There are two distinct subspecies in Far East Asia: one (H. i. inermis) lives in China, and the other (H. i. argyropus) lives in Korea. Unlike the Chinese subspecies, the diet of the Korean subspecies in the wild has rarely been studied. Here, we analyzed the diets of the Korean water deer by using 202 bp rbcL sequences of plants found in fecal samples. Both similarity (%) and phylogeny (NJ tree) of the sequences were considered for the sequence assignment. These methods successfully assigned each sequence to only one family of one order without exception. We found that the Korean water deer fed on a total of 18 orders and 24 families of plants in the summer season. It fed mainly on Asteraceae family (28.4%) followed by Fagaceae (15.9%) and Polygonaceae (11.5%). There was a significant difference in diet composition between lowland area and mountainous area deer (P < 0.001). The Korean water deer on Daebu Island are presumably browsers feeding mainly on forbs or woody plants.  相似文献

Leafminer flies, especially, Liriomyza huidobrensis, Liriomyza sativae and Liriomyza trifolii, are quarantine species in many countries. Their morphological similarity makes identification difficult. To develop a rapid, reliable, sensitive and simple molecular identification method using multiplex PCR, we newly sequenced the mitochondrial cytochrome oxidase I (COI) genes of Liriomyza bryoniae, Liriomyza chinensis, L. huidobrensis, L. sativae, L. trifolii, Chromatomyia horticola and four parasitoid species. We aligned them with all the COI sequences of the leafminer flies found in the international DNA nucleotide sequence databases (DDBJ/EMBL/GenBank). We then designed species‐specific primers to allow us to differentiate between L. bryoniae, L. chinensis, L. huidobrensis, L. sativae, and L. trifolii.  相似文献

The giant jellyfish Nemopilema nomurai is a scyphozoan species well-known in East Asian Marginal Seas for its damage to fisheries. The genetic diversity and population structure of N. nomurai, collected from five geographic regions in Chinese coastal seas, were examined based on mitochondrial cytochrome c oxidase subunit I (COI) gene and nuclear internal transcribed spacer one (ITS1) sequences. A total of 26 and five unique haplotypes were recovered from the COI and ITS1 genes, respectively. The overall genetic diversity of N. nomurai calculated by the COI and ITS1 sequences was low (haplotype diversity 0.727% and 0.108%, nucleotide diversity 0.212% and 0.039%). The median-joining network analysis revealed a star-like haplotype network. The hierarchical Analysis of Molecular Variance (AMOVA) of COI haplotypes showed that N. nomurai populations form a single population, with a low FST (0.0149, p?=?0.1036). The dispersal ability, together with the biological characteristics, could be important factors for the lack of a geographically structured pattern in N. nomurai in Chinese coastal waters.  相似文献

 Although Korean Adonis has been traditionally recognized as Adonis amurensis Regel and Radde with various infraspecific taxa described, its taxonomic identity is still in dispute. We investigated the genetic variation in 60 individuals from 12 populations in Korea to elucidate the taxonomic identity of the Korean Adonis complex. Random amplified polymorphic DNA (RAPD) analysis revealed that Korean Adonis comprises three species: A. amurensis, A. pseudoamurensis, and A. multiflora. Adonis amurensis is distributed in northern central inland regions of Korea and A. pseudoamurensis is found in southern parts of the Korean peninsula. Adonis multiflora grows only on Cheju Island, which is the southernmost part of Korea. Phylogenetic analysis of nuclear ribosomal internal transcribed spacer (ITS) sequences partially supported the presence of three Adonis taxa in Korea as detected by RAPD analysis. The Adonis population on Jangbong Island located in the West Sea, which was referred to as A. pseudoamurensis on the basis of morphological examination, was separated from the other populations of A. pseudoamurensis. Otherwise, the molecular evidence is well congruent with the recent morphological study that proposes that Korean Adonis consists of these three species. Despite the medical importance of anopheline mosquitoes as vectors of Korean vivax malaria, differentiation between Korean anopheline mosquitoes by traditional morphological taxonomic criteria is difficult. An. yatsushiroensis is the second most common Anopheles mosquito species in Korea and a possible vector of Korean vivax malaria together with An. sinensis, the predominant anopheline species. Recently, An. yatsushiroensis has been declared a synonym of An. pullus, based on comparisons of egg morphology and adult progeny, although they differ in ecology and morphology. To verify the species status of these two ambiguous forms, we established isofemale lines of Korean An. pullus and An. yatsushiroensis (An. pullus form yatsushiroensis) mosquitoes and investigated their genetic relationship by metaphase karyotype analysis, comparing the DNA sequences of rDNA internal transcribed spacer 2 (ITS2) and mitochondrial cytochrome c oxidase subunits I (COI) and II (COII), and by hybridization experiments. Two isofemale lines had differently shaped X and Y chromosomes. However reciprocal crosses between them yielded viable progeny with completely synaptic salivary gland polytene chromosomes. DNA analyses also strongly supported their conspecificity. The two strains also showed great sequence similarity in the ITS2, COI and COII regions (variation rate = 0.0 to 0.8%). Based on these findings, we conclude that the two forms, though differing distinctly in morphological, cytological and ecological traits, remain interfertile.  相似文献

The length–weight relationships (LWRs) for four freshwater fish species in catchment area of Geoje‐island, South Korea, were analyzed. Fish samples were collected quarterly using scoop‐nets (5 mm mesh size), casting nets (7 mm mesh size) from 2016 to 2017.
