Forked spleenwort, Asplenium septentrionale (L.) Hoffm., is mainly a petrophilous fern species of European mountains and very rare in Estonia, a country with flat topography and few rocky habitats. The single extant population is very small, occupies a restricted area, and is threatened seasonally by adverse human activity. An introduction project of the species was prepared according to the A. septentrionale Management Plan. The goal of the project is to introduce new populations of the species in new protected sites from ex situ propagated young sporophytes. The aim of this study was to obtain detailed data on the species ecology and provide information for selecting suitable introduction sites. We carried out a factorial pot experiment on light availability, soil pH and soil biota gradient in order to ascertain optimal growth and development conditions for A. septentrionale. Soil and light conditions must be considered the most important factors affecting species growth and reproduction, particularly under conditions of largely fluctuating water regime. Experimental plants grown in abiotic conditions closely emulating the natural habitat were the most successful and may, therefore, represent the optimal establishment potential. The survival of plants was greater, and plants grew larger with a higher number of longer leaves in acidic soil (pH 4.9) than in neutral soil. Plants performed best under high, approximately 75% total available illumination. Deep shade (90%) and well as full daylight had strong negative effect on the plants. No positive effect of “home-soil” biota was detected.  相似文献

Background and Aims

Human-mediated environmental change is increasing selection pressure for the capacity in plants to colonize new areas. Habitat fragmentation combined with climate change, in general, forces species to colonize areas over longer distances. Mating systems and genetic load are important determinants of the establishment and long-term survival of new populations. Here, the mating system of Asplenium scolopendrium, a diploid homosporous fern species, is examined in relation to colonization processes.

Methods

A common environment experiment was conducted with 13 pairs of sporophytes, each from a different site. Together they constitute at least nine distinct genotypes, representing an estimated approx. 95 % of the non-private intraspecific genetic variation in Europe. Sporophyte production was recorded for gametophytes derived from each parent sporophyte. Gametophytes were grown in vitro in three different ways: (I) in isolation, (II) with a gametophyte from a different sporophyte within the same site or (III) with a partner from a different site.

Key Results

Sporophyte production was highest in among-site crosses (III), intermediate in within-site crosses (II) and was lowest in isolated gametophytes (I), strongly indicating inbreeding depression. However, intragametophytic selfing was observed in most of the genotypes tested (eight out of nine).

Conclusions

The results imply a mixed mating system in A. scolopendrium, with outcrossing when possible and occasional selfing when needed. Occasional intragametophytic selfing facilitates the successful colonization of new sites from a single spore. The resulting sporophyte, which will be completely homozygous, will shed large amounts of spores over time. Each year this creates a bed of gametophytes in the vicinity of the parent. Any unrelated spore which arrives is then selectively favoured to reproduce and contribute its genes to the new population. Thus, while selfing facilitates initial colonization success, inbreeding depression promotes genetically diverse populations through outcrossing. The results provide further evidence against the overly simple dichotomous distinction of fern species as either selfing or outcrossing.  相似文献

广泛的杂交和多倍化使得铁角蕨属(Asplenium)下存在着许多分类困难的物种复合体, 针对这些类群进行整合分类学的研究, 有助于我们更加全面和深入地理解物种的界限以及形成机制。线裂铁角蕨复合体(Asplenium coenobiale complex)是铁角蕨属下一个形态多样性较高的类群, 由于缺乏全面取样和系统研究, 该复合体的物种划分长期存在争议。本研究选取线裂铁角蕨复合体中形态变异和地理分布具有代表性的个体, 通过孢粉学研究确定该类群的生殖特性, 运用流式细胞分析获取倍性信息, 同时结合叶绿体和核基因组片段系统发生分析的证据, 对该类群的系统演化关系和起源方式进行了探讨。结果表明: (1)虽然部分孢子囊败育的情况在线裂铁角蕨复合体中十分普遍, 但正常孢子囊内形成的64个孢子说明该类群植物仍能进行正常的有性生殖; (2)该复合体中存在着倍性变异, 其中多角铁角蕨(A. cornutissimum)是二倍体, 而其他成员均为四倍体; (3)依据母系遗传的叶绿体序列所构建的系统发生关系将该类群划为4个分支, 与基于核基因序列构建的系统树存在冲突, 这暗示杂交可能在该复合体的形成过程中起到了重要的推动作用。综上所述, 我们建议将线裂铁角蕨复合体划分为4个物种, 即同源四倍体新种马关铁角蕨(A. maguanense sp. nov.), 二倍体多角铁角蕨, 以及两个由同一对亲本正反交产生的异源四倍体线裂铁角蕨(A. coenobiale)和叶基宽铁角蕨(A. pulcherrimum)。  相似文献

Background and Aims

Asexual reproduction is a prominent evolutionary process within land plant lineages and especially in ferns. Up to 10 % of the approx. 10 000 fern species are assumed to be obligate asexuals. In the Asplenium monanthes species complex, previous studies identified two triploid, apomictic species. The purpose of this study was to elucidate the phylogenetic relationships in the A. monanthes complex and to investigate the occurrence and evolution of apomixis within this group.

Methods

DNA sequences of three plastid markers and one nuclear single copy gene were used for phylogenetic analyses. Reproductive modes were assessed by examining gametophytic and sporophyte development, while polyploidy was inferred from spore measurements.

Key Results

Asplenium monanthes and A. resiliens are confirmed to be apomictic. Asplenium palmeri, A. hallbergii and specimens that are morphologically similar to A. heterochroum are also found to be apomictic. Apomixis is confined to two main clades of taxa related to A. monanthes and A. resiliens, respectively, and is associated with reticulate evolution. Two apomictic A. monanthes lineages, and two putative diploid sexual progenitor species are identified in the A. monanthes clade.

Conclusions

Multiple origins of apomixis are inferred, in both alloploid and autoploid forms, within the A. resiliens and A. monanthes clades.  相似文献

Plant cell division requires the dynamic organisation of several microtubule arrays. The mechanisms of regulation of the above arrays are under rigorous research. Among several factors that are involved in plant microtubule dynamics, the Targeting Protein for Xklp2 (TPX2) has been found to play a role in spindle organisation, in combination with Aurora kinases, in dividing cells of angiosperms. Microtubule organisation in dividing cells of ferns exhibits certain peculiarities. Accordingly, the presence and distribution of a TPX2 homologue might be helpful in understanding the patterns and regulatory mechanisms of microtubule arrays in this plant group. In this study, a putative TPX2 homologue was identified using Western blotting in the fern Asplenium nidus. It was found, using immunostaining and CLSM, that it is co‐localised with perinuclear preprophase microtubules and the prophase spindle, and follows the microtubule pattern during metaphase/anaphase and telophase. During cytokinesis, while in angiosperms TPX2 is degraded, in A. nidus the TPX2 signal persists, co‐localising with the phragmoplast. In early post‐cytokinetic cells, a TPX2 signal is present on the nuclear surface facing the daughter cell wall and, thereafter it is co‐localised with the fern‐specific microtubule aggregation that lines the new wall, which is possibly involved in cortical microtubule assembly.  相似文献

In the Southern Hemisphere there has been little phylogeographical investigation of forest refugia sites during the last glacial. Hooker's spleenwort, Asplenium hookerianum, is a fern that is found throughout New Zealand. It is strongly associated with forest and is a proxy for the survival of woody vegetation during the last glacial maximum. DNA sequence data from the chloroplast trnL-trnF locus were obtained from 242 samples, including c. 10 individuals from each of 21 focal populations. Most populations contained multiple, and in many cases unique, haplotypes, including those neighbouring formerly glaciated areas, while the predominant inference from nested clade analysis was restricted gene flow with isolation by distance. These results suggest that A. hookerianum survived the last glacial maximum in widespread populations of sufficient size to retain the observed phylogeography, and therefore that the sheltering woody vegetation must have been similarly abundant. This is consistent with palynological interpretations for the survival in New Zealand of thermophilous forest species at considerably smaller distances from the ice sheets than recorded for the Northern Hemisphere. Eastern and central North Island populations of A. hookerianum were characterized by a different subset of haplotypes to populations from the remainder of the country. A similar east-west phylogeographical pattern has been detected in a diverse array of taxa, and has previously been attributed to recurrent vulcanism in the central North Island.  相似文献

Asplenium trichomanes L. subsp. trichomanes spermatozoids are spirals of about five turns. Keels link the elements of the microtubular ribbon with the plates of the lamellar layer (LL) which are uninterrupted, parallel and curved with an inner angle of about 150°. Electron-opaque filaments connect the microtubules of the multilayered structure (MLS) and the osmiophilic crest, the LL and the MLS-associated mitochondrion and the latter and the plasmalemma. The nucleus occupies the 2.5–3 posterior turns and has an inner honeycomb-shaped chromatin mass and an outer highly condensed chromatin mass with randomly scattered electron-transparent areas. The basal bodies of the ca. 50 flagella are bounded by a reticulum of granular material which forms a plug inside their proximal region; the proximal region of the flagellum has a 9 + 0 pattern. Background and Aims

The pattern of callose deposition was followed in developing stomata of the fern Asplenium nidus to investigate the role of this polysaccharide in guard cell (GC) wall differentiation and stomatal pore formation.

Methods

Callose was localized by aniline blue staining and immunolabelling using an antibody against (1 → 3)-β-d-glucan. The study was carried out in stomata of untreated material as well as of material treated with: (1) 2-deoxy-d-glucose (2-DDG) or tunicamycin, which inhibit callose synthesis; (2) coumarin or 2,6-dichlorobenzonitrile (dichlobenil), which block cellulose synthesis; (3) cyclopiazonic acid (CPA), which disturbs cytoplasmic Ca2+ homeostasis; and (d) cytochalasin B or oryzalin, which disintegrate actin filaments and microtubules, respectively.

Results

In post-cytokinetic stomata significant amounts of callose persisted in the nascent ventral wall. Callose then began degrading from the mid-region of the ventral wall towards its periphery, a process which kept pace with the formation of an ‘internal stomatal pore’ by local separation of the partner plasmalemmata. In differentiating GCs, callose was consistently localized in the developing cell-wall thickenings. In 2-DDG-, tunicamycin- and CPA-affected stomata, callose deposition and internal stomatal pore formation were inhibited. The affected ventral walls and GC wall thickenings contained membranous elements. Stomata recovering from the above treatments formed a stomatal pore by a mechanism different from that in untreated stomata. After coumarin or dichlobenil treatment, callose was retained in the nascent ventral wall for longer than in control stomata, while internal stomatal pore formation was blocked. Actin filament disintegration inhibited internal stomatal pore formation, without any effect on callose deposition.

Conclusions

In A. nidus stomata the time and pattern of callose deposition and degradation play an essential role in internal stomatal pore formation, and callose participates in deposition of the local GC wall thickenings.  相似文献

Abstract The birds nest fern, Asplenium nidus, contributes greatly to the epiphytic biomass in the canopies of both south‐east Asia and tropical north Queensland rainforests. It is generally believed that their abundance and their capacity to store water is an important feature for habitat fragmentation in the canopy. We investigated the microclimate of A. nidus and the effects of severe drought periods on the A. nidus population over a 20‐year period, hypothesizing that water availability is the most important factor controlling the population under drought conditions. One of two neighbouring A. nidus plants of the same size and age was irrigated artificially before, during and after a significant dry period in 2000. By monitoring the microclimate within and around both ferns we were able to estimate that four continuous weeks of rainless weather completely dried out the accumulated humus of the non‐irrigated A. nidus fern. Prolonged dry periods were shown to kill the roots of A. nidus, which attach the fern to the bark and eventually the affected A. nidus on verticals stems fell to the ground. Periods longer than 8 weeks may even kill adult plants sitting in more protected branch forks. Analysis of the whole A. nidus population within the 1‐ha Canopy Crane plot and the determination of the morphological age of all plants enabled an evaluation of the historical development of the population. The oldest plant originated in 1985, just 1 year after the longest recorded drought for the site. We suspect that the 1984 drought killed every A. nidus plant within the study plot. Years with low recruitment coincide with years with long drought periods.  相似文献

Background and Aims

Successful establishment of newly formed polyploid species depends on several interlinked genetic and ecological factors. These include genetic diversity within and among individuals, chromosome behaviour and fertility, novel phenotypes resulting from novel genomic make-up and expression, intercytotypic and interspecific competition, and adaptation to distinct habitats. The allotetraploid rock fern Asplenium majoricum is known from one small population in Valencia, Spain, and several larger populations on the Balearic island of Majorca. In Valencia, it occurs sympatrically with its diploid parents, A. fontanum subsp. fontanum and A. petrarchae subsp. bivalens, and their diploid hybrid A. × protomajoricum. This highly unusual situation allowed the study of polyploid genetic diversity and its relationship to the formation and establishment of nascent polyploid lineages.

Methods

Genetic variation for isozyme and chloroplast DNA markers was determined for A. majoricum and A. × protomajoricum sampled thoroughly from known sites in Majorca and Valencia. Results were compared with variation determined previously for the diploid parent taxa.

Key Results

A highly dynamic system with recurring diploid hybrid and allotetraploid formation was discovered. High diversity in the small Valencian A. majoricum population indicates multiple de novo origins from diverse parental genotypes, but most of these lineages become extinct without becoming established. The populations on Majorca most probably represent colonization(s) from Valencia rather than an in situ origin. Low genetic diversity suggests that this colonization may have occurred only once.

Conclusions

There is a striking contrast in success of establishment of the Majorcan and Valencian populations of A. majoricum. Chance founding of populations in a habitat where neither A. fontanum subsp. fontanum nor A. petrarchae subsp. bivalens occurs appears to have been a key factor enabling the establishment of A. majoricum on Majorca. Successful establishment of this polyploid is probably dependent on geographic isolation from diploid progenitor competition.  相似文献

Little is known about the mode of transmission of chloroplasts in ferns, despite the importance of such knowledge for molecular phylogenetic and biosystematic studies. Andersson-Kötto (1930, 1931) inferred from crossing experiments that chloroplasts are inherited biparentally in Asplenium scolopendrium L. Here we present evidence from artificial hybridisation that demonstrates maternal inheritance of chloroplast DNA in the genus Asplenium (Aspleniaceae, Pteridophyta) using length variation in a non-coding spacer (trnLUAA-trnFGAA) in the chloroplast DNA.  相似文献

Maguire primrose (Primula cusickiana var. maguirei) is a threatened, narrow endemic found only along a 20‐km section within a single canyon in northern Utah, USA. Here we describe the breeding strategy of this perennial, distylous, primarily cliff‐dwelling species to test for reproductive constraints on the plants. Intermorph outcrossing treatments (i.e. between pin and thrum flowers) were the most successful hand pollinations performed, but those were usually inferior to natural pollination. Intramorph out‐crossing also was somewhat successful, but was consistently inferior to intermorph outcrossing. Seed‐set resulting from autogamy and geitonogamy treatments was least successful. Morph‐specific differences in seed set were observed for our hand pollinations, but not in natural pollination conditions. Our results suggest that pollinators play a crucial role by providing the pollen transfer necessary for maximal seed set in this rare and narrowly distributed distylous plant.  相似文献

Conservation policies and environmental impact assessments commonly target threatened species and habitats. Nevertheless, macroecological research provides reasons why also common species should be considered. We investigate the consequences of focussing solely on legally protected species and habitats in a spatial conservation planning context using a comprehensive, benthic marine data set from the northern Baltic Sea. Using spatial prioritization and surrogacy analysis, we show that the common approach in conservation planning, where legally listed threatened species and habitats are the focus of conservation efforts, could lead to poor outcomes for common species (and therefore biodiversity as a whole), allowing them to decline in the future. If conservation efforts were aimed solely at threatened species, common species would experience a loss of 62% coverage. In contrast, if conservation plans were based only on common species, threatened species would suffer a loss of 1%. Threatened species are rare and their ecological niches distinct, making them poor surrogates for biodiversity. The best results are achieved by unified planning for all species and habitats. The minimal step towards acknowledging common species in conservation planning would be the inclusion of the richness of common species, complemented by information on indicator species or species of high importance for ecosystem functioning. The trade-off between planning for rare and common species should be evaluated, to minimize losses to biodiversity.  相似文献

The presence of researchers, ecotourists or rangers inside protected areas is generally assumed to provide a protective effect for wildlife populations, mainly by reducing poaching pressure. However, this assumption has rarely been empirically tested. Here, we evaluate and quantify the conservation benefits of the presence of a long-term research area in Taï National Park, Côte d''Ivoire. A wildlife survey following 225 km of line transects revealed considerably higher primate and duiker encounter rates within the research area when compared with adjacent areas. This positive effect was particularly pronounced for threatened and over-harvested species, such as the endangered red colobus monkey (Procolobus badius). This pattern was clearly mirrored by a reversed gradient in signs of poaching, which decreased towards and inside the research area, a trend that was also supported with park-wide data. This study demonstrates that even relatively simple evidence-based analytical approaches can bridge the gap between conservation theory and practice. In addition, it emphasizes the value of establishing long-term research sites as an integral part of protected area management.  相似文献

Background and Aims

Climate change is expected to alter the geographic range of many plant species dramatically. Predicting this response will be critical to managing the conservation of plant resources and the effects of invasive species. The aim of this study was to predict the response of temperate homosporous ferns to climate change.

Methods

Genetic diversity and changes in distribution range were inferred for the diploid rock fern Asplenium fontanum along a South–North transect, extending from its putative last glacial maximum (LGM) refugia in southern France towards southern Germany and eastern-central France. This study reconciles observations from distribution models and phylogeographic analyses derived from plastid and nuclear diversity.

Key Results

Genetic diversity distribution and niche modelling propose that genetic diversity accumulates in the LGM climate refugium in southern France with the formation of a diversity gradient reflecting a slow, post-LGM range expansion towards the current distribution range. Evidence supports the fern''s preference for outcrossing, contradicting the expectation that homosporous ferns would populate new sites by single-spore colonization. Prediction of climate and distribution range change suggests that a dramatic loss of range and genetic diversity in this fern is possible. The observed migration is best described by the phalanx expansion model.

Conclusions

The results suggest that homosporous ferns reproducing preferentially by outcrossing accumulate genetic diversity primarily in LGM climate refugia and may be threatened if these areas disappear due to global climate change.  相似文献
