All but one of the specific pathogens of Tribolium castaneum were found in samples of insects from British food stores. The commonest pathogen, Nosema whitei, occurred in an average of 10.1% of the insects in 27% of the samples. At the time of sampling, diseases were probably not limiting appreciably the numbers of insects in stored food. Disease is not likely to curb infestations before the level of economic importance is reached. However, diseases may be of some economic importance by reducing or eliminating sources of infestation in undisturbed residues of food.  相似文献

Female reproduction includes maturation of oocytes and the synthesis of yolk proteins (vitellogenin, Vg) in the fat body and their deposition into the oocytes. Our recent studies showed that juvenile hormone (JH) regulates Vg synthesis and 20-hydroxyecdysone (20E) regulates oocyte maturation in the red flour beetle (Tribolium castaneum). Here, we report on the role of nutritional signaling on vitellogenesis and oogenesis. Comparison of gene expression between fed and starved beetles by microarray analysis showed the up-regulation of genes involved in energy homeostasis and down-regulation of genes involved in egg production in the starved beetles. The RNA interference (RNAi) aided knock-down in the expression of genes involved in insulin and TOR signaling pathways showed that both these signaling pathways play key roles in Vg synthesis and oocyte maturation. Starvation of female beetles resulted in a block in Vg synthesis but not in the progression of primary oocyte development to the resting stage. Feeding after starvation induced Vg synthesis and the progression of primary oocytes from the resting stage to the mature stage. However, in the beetles where JH or 20E synthesis or action was blocked by RNAi, both Vg synthesis and oocyte maturation were affected suggesting that both these hormones (JH and 20E) and nutritional signaling and their cross-talk regulate vitellogenesis and oogenesis.  相似文献

Production of secretion in the accessory reproductive glands of male Melanoplus sanguinipes has been examined by electrophoresis and radiolabelling. The secretion of each group of tubules (long hyaline glands, white glands, short hyaline glands, and seminal vesicles) can be resolved into more than 20 protein bands and includes several glycoproteins and, in the long hyaline and white glands only, lipoproteins. Each group of tubules has a characteristic pattern of synthesis and accumulation of proteins; that is, specific proteins appear in the secretion at particular times during sexual maturation. In allatectomized insects, the long hyaline glands accumulate very little secretion; the white glands and short hyaline glands accumulate about one-third the normal amount; and accumulation in the seminal vesicles is not affected by the operation. Allatectomy exerts its effect by inhibiting the synthesis of particular proteins. The observations are discussed in terms of juvenile hormone-specific protein synthesis in the accessory reproductive glands.  相似文献

Juvenile hormone (JH) involvement in male reproduction is poorly understood. In Drosophila melanogaster adults, JH deficiency has been shown to result in lowered protein synthesis in male accessory glands. To probe additional roles, we have examined males homozygous for a null allele of Methoprene-tolerant (Met). This gene is involved in the action of JH, possibly at the JH receptor level, and Met27 null mutants reflect a diminution of JH action. Met27 males were found to have reduced protein accumulation in male accessory glands and to court and mate wild-type females much less avidly than do either Met+ or Met27; Met+ transgenic males. Exposure of Met27 males to methoprene partially rescued the courtship deficiency. However, sperm transfer as reflected by fertility of Met27 fathers was found to be similar to that of Met+. Taken together with previous work examining the JH-deficient mutant apterous, these results corroborate JH involvement in protein synthesis in the male accessory glands and suggest a role for JH in promoting male mating behavior in these flies.  相似文献

To elucidate the endocrine regulation of vitellogenin (Vg) synthesis in the red flour beetle, Tribolium castaneum, the titers of juvenile hormone (JH) and ecdysteroids in the whole body of female beetles were measured and compared with Vg mRNA levels. Juvenile hormone levels remained high while the ecdysteroid levels declined steadily during 1–5 days post adult emergence (PAE). The Vg mRNA levels began to increase by the end of 3rd day PAE and peaked by the 4th–5th day PAE. Gene expression profiling by microarray and quantitative real-time PCR analyses of RNA isolated from 1 to 5 days PAE beetles revealed that the genes coding for proteins involved in JH biosynthesis and action, but not those involved in 20-hydroxyecdysone (20E) biosynthesis and action had similar expression patterns as the genes coding for Vg. RNA interference (RNAi)-aided knock-down in the expression of these genes showed that both JH and 20E were required for Vg gene expression. However, Vg mRNA was induced by the application of JH III but not by the injection of 20E into the previtellogenic females. These data suggest that JH is required for Vg synthesis in the fat body and 20E influences Vg synthesis through its action on oocyte maturation.  相似文献

Summary The accessory reproductive glands of Leptinotarsa decemlineata males are simple epithelial tubular glands. They are surrounded by a network of slow acting muscle cells. All the gland cells have a very extensive granular endoplasmic reticulum, a prominent Golgi apparatus, but few secretory granules. The secretory product in the lumen, which is a neutral mucopolysaccharide-protein complex, appears as a granular material in which fibrillar structures occur. Neither in the gland cells nor in the lumen were regional differences observed. After allatectomy the gland cells degenerate in a remarkable way. A zone containing microvillous structures appears between the outer and inner cells. Antenna structure varies widely among insects, in contrast to the well-conserved structure of legs. The adult capitate antenna of the red flour beetle, Tribolium castaneum, is composed of eleven articles, organized into four distinct morphological regions (scape, pedicel, funicle and club). Here, we report the use of RNA interference to examine the functions of 21 genes during antenna metamorphosis in T. castaneum. Genes with conserved functions relative to the developmental model species Drosophila melanogaster include Distal-less and EGF signaling (antennal growth), spineless (determination of antennal identity) and the Notch signaling pathway (antennal growth, joint formation, and sensory bristle development). However, the functions of many genes differed from those predicted from the Drosophila model. In addition to a conserved gap phenotype, depletion of dachshund transformed funicle articles toward club-like identity. Depletion of Distal-less or homothorax did not cause antenna-to-leg transformation. Lim1 was required only for development of the scape-pedicle joint. Depletion of odd-skipped-related genes led to the loss of the entire funicle, while spalt, rotund, spineless, and dachshund affected smaller regions. Growth and joint formation were linked developmentally in the funicle, but not in the club. Joint formation within the club required bric-a-brac, aristaless, apterous, and pdm. Gene functions are discussed in terms of a model of antenna development in T. castaneum. This model provides a contrast to knowledge of antenna development in D. melanogaster, insight into the likely ancestral mode of antenna development, and a framework for considering diverse antenna morphologies.  相似文献

The dramatic transformation from a larva to an adult must be accompanied by a coordinated activity of genes and hormones that enable an orchestrated transformation from larval to pupal/adult tissues. The maintenance of larval appendages and their subsequent transformation to appendages in holometabolous insects remains elusive at the developmental genetic level. Here the role of a key appendage patterning gene Distal-less (Dll) was examined in mid- to late-larval stages of the flour beetle, Tribolium castaneum. During late larval development, Dll was expressed in appendages in a similar manner as previously reported for the tobacco hornworm, Manduca sexta. Removal of this late Dll expression resulted in disruption of adult appendage patterning. Intriguingly, earlier removal resulted in dramatic loss of structural integrity and identity of larval appendages. A large amount of variability in appendage morphology was observed following Dll dsRNA injection, unlike larvae injected with dachshund dsRNA. These Dll dsRNA-injected larvae underwent numerous supernumerary molts, which could be terminated with injection of either JH methyltransferase or Methoprene-tolerant dsRNA. Apparently, the partial dedifferentiation of the appendages in these larvae acts to maintain high JH and, hence, prevents metamorphosis.  相似文献

We report on juvenile hormone (JH) biosynthesis in vitro by male accessory glands (MAGs) in the longhorned beetle, Aprionona germari, accompanied by the transfer of JH from males to females during copulation. JH was extracted from the MAGs and separated by reversed‐phase high‐performance liquid chromatography. JH III was identified as the major JH by gas chromatography–mass spectrometry. A radiochemical assay and a non‐radioactive method were used to measure the in vitro rate of JH biosynthesis by the MAGs. After 4 h of incubation with 3H‐methionine in the medium, the radioactivity in the MAGs substantially increased. In a separate assay, incubation of the MAGs with non‐radioactive methionine for 4 h resulted in a 39% increase in JH III. Seven‐day‐old males were injected with medium 199 containing 3H–methionine and 24 h later they were mated with virgin females. Hemolymph and the MAGs were collected from the mated males and hemolymph, ovaries and eggs were collected from the mated females for assaying radioactive JH. The radioactivity incorporated into JH in the MAGs was transferred to the females during copulation and later transferred into their eggs. Assayed 1 h after copulation, JH III level in the MAGs decreased 42% and the content of JH III in the male hemolymph did not change, whereas the content of JH III in the female hemolymph and ovaries both increased. © 2010 Wiley Periodicals, Inc.  相似文献

The ultrastructural differentiation of the secretory cells and the nature of secretory proteins in the male accessory gland of Drosophila funebris have been studied by electron-microscopic and immunological methods. (1) In the pupae at 112 days before eclosion, secretory products can be detected in the lumen, even though most glandular cells are at the initial phase of differentiation. At the time of eclosion both main and secondary cells are fully differentiated, but the whole set of five immunologically active proteins are detectable only on the second to third day of adult life. (2) The secondary cells contain giant protein granules, the so-called filamentous bodies, which become partially fused and the filaments assume a twisted form. Randomly dispersed filaments and closely packed filament bundles are also visible in the gland lumen. Antigenic labelling of ultrathin sections and immunoreplica electrophoresis yielded no evidence for the microtubular nature of these filaments. The secretion stored in the lumen contains in addition a large quantity of flocculent proteins which have their origin in the main cells. (3) During the period of high secretory activity in the 7-day-old male flies no vacuolization and disintegration of either the main or secondary cells have been observed. We conclude that both types of cells have the merocrine secretory mechanism. (4) Ultrastructural alterations in the glandular cells confirmed our previous observation that copulation stimulates RNA and protein synthesis.  相似文献

This study clarifies the role of female-controlled processes contributing to sperm storage in the red flour beetle, Tribolium castaneum. Evidence presented indicates that sperm motility is not affected by extreme hypoxia produced by anesthetization of the female with either carbon dioxide or nitrogen. Sperm location and motility in low-oxygen environments did not differ from that of sperm in reproductive tracts immersed in fully aerated saline. Sperm motility was unaffected by exposure to potassium cyanide, an aerobic respiratory system poison, but was inhibited by exposure to iodoacetic acid, a glycolysis poison. Based on the retention of sperm motility under extreme hypoxia, female control over sperm storage was then examined. Both anesthetized females and dead females had fewer stored sperm after mating than unanesthetized control females. These results suggest that female T. castaneum play an active role in moving sperm from the site of deposition into storage.  相似文献

Protein production during sexual maturation or after allatectomy (followed by juvenile hormone replacement therapy) has been examined in the long hyaline glands, short hyaline glands, white glands, and seminal vesicles, which make up the accessory gland complex of male Melanoplus sanguinipes. During maturation, the amount of protein in the long hyaline glands increases about 14-fold, and in each of the other components between 5- and 6-fold. Most protein accumulates between days 3 and 5, and this is reflected in high levels of incorporation of radiolabelled leucine in this period.

The components show differential sensitivity to the effects of allatectomy. After this operation, the protein content of, and incorporation of radiolabel into, the long hyaline glands remain near the day 0 level. In the white glands and short hyaline glands, allatectomy also has a marked, though less severe, effect on protein synthesis and accumulation. The seminal vesicles are least affected by allatectomy and continue to accumulate protein (though more slowly) to about 60% of the normal level by day 10. Juvenile hormone compounds applied topically to allatectomized insects on day 2 restored the ability of the gland components to accumulate proteins, though to differing degrees. JHI is the most effective compound, stimulating synthesis and accumulation of protein to near normal levels by day 10, whereas application of JHIII or Stauffer's synthetic JH led to only partial restoration of protein synthesis in the glands.  相似文献

Our recent studies identified juvenile hormone (JH) and nutrition as the two key signals that regulate vitellogenin (Vg) gene expression in the red flour beetle, Tribolium castaneum. Juvenile hormone regulation of Vg synthesis has been known for a long time in several insects, but the mechanism of JH action is not known. Experiments were conducted to determine the mechanism of action of these two signals in regulation of Vg gene expression. Injection of bovine insulin or FOXO double-stranded RNA into the previtellogenic, starved, or JH-deficient female adults increased Vg mRNA and protein levels, thereby implicating the pivotal role for insulin-like peptide signaling in the regulation of Vg gene expression and possible cross-talk between JH and insulin-like peptide signaling pathways. Reduction in JH synthesis or its action by RNAi-mediated silencing of genes coding for acid methyltransferase or methoprene-tolerant decreased expression of genes coding for insulin-like peptides (ILPs) and influenced FOXO subcellular localization, resulting in the down-regulation of Vg gene expression. Furthermore, JH application to previtellogenic female beetles induced the expression of genes coding for ILP2 and ILP3, and induced Vg gene expression. FOXO protein expressed in baculovirus system binds to FOXO response element present in the Vg gene promoter. These data suggest that JH functions through insulin-like peptide signaling pathway to regulate Vg gene expression.  相似文献
