The present study outlines a rapid and sensitive on-the-baseline conditional emotional response (CER) procedure. Using rats as the experimental subject, the method detects delay conditioning, incubation, extinction and spontaneous recovery. In addition, the method detects conditional responding using electric shock ranging from 0.23 to 0.50 mA as the unconditional stimulus. Because of its speed and sensitivity, the method shelters the subject from unnecessary long-term deprivation and pain.  相似文献

Neurotransmitter turnover of biogenic monoamines (dopamine, norepinephrine, and serotonin) and amino acids (glutamate, aspartate, and gamma-aminobutyric acid) was evaluated in rats exposed to the conditioned emotional response (CER) paradigm in the absence (total suppression) or presence of acute 5 mg/kg i.p. diazepam (which reversed suppression and restored normal responding). Based on previous studies of CER, with controls for shock and stimulus histories, the results with respect to the anxiolytic could be divided into several categories: changes in turnover which are associated only with the CER behavior; changes associated only with the drug, diazepam; changes which augmented the effects of the behavior; or changes which were the reverse of those associated with the behavior. Due to the multitude and complexity of the results, not all observations have clear explanations at this time. However, for the CER behavior per se, it is apparent that a combination of neurotransmitters, including some implications about acetylcholine, act in concert to bring about the behavioral suppression. The action of diazepam is more complex, involving the full spectrum of neurotransmitters to bring about its direct and indirect effects.In honor of Distinguished Professor Morris Herman Aprison  相似文献

Packet theory is based on the assumption that the momentary probability of producing a bout or packet of responding is controlled by the conditional expected time function. Bouts of head entry responses of rats into a food cup appear to have the same characteristics across a range of conditions. The conditional expected time function is the mean expected time remaining until the next food delivery as a function of time since an event such as food or stimulus onset. The conditional expected time function encodes mean interval duration as well as the distribution form so that both the mean response rate and form of responding in time can be predicted. Simulations of Packet theory produced accurate quantitative predictions of: (1) the effect of reinforcement density (mean food-food interval) and distribution form on responding; (2) scalar variance in fixed interval responding; (3) CS-US and intertrial interval effects on the strength of conditioning; and (4) the effect of the ratio of cycle:trial time on the strength of conditioning.  相似文献

Rate expectancy theory (RET) predicts that in Pavlovian procedures conditioned responding will be directly related to the ratio of time spent in the experimental context (C) relative to the trial time (T) or duration of the conditioned stimulus (CS). This prediction was discussed in the context of three experiments. The first and third experiments involved sexual conditioning in quail [Learn Motiv 31 (2000) 211; J Exp Psychol Anim Behav Processes 27 (2001) 269]; the second experiment involved conditioning rats with food as the unconditioned stimulus (US) [Learn Motiv 28(1997) 465]. In each experiment, one type of conditioned response was directly related to the C/T ratio (as predicted by RET) but another conditioned response was inversely related. In addition, the conditioned behavior that occurred with low C/T ratios was controlled by contextual cues rather than the putative CS. The implications of these findings for possible boundary conditions of RET were discussed. The discussion revealed serious shortcomings in the characterization of context conditioning by RET.  相似文献

A fast inferential, multivariable adaptive optimization algorithm based on a fast responding off-gas data, the carbon dioxide evolution rate (CER), has been developed and applied to a continuous baker's yeast culture to maximize the cellular productivity in simulation and experimental studies. In the simulation study the process was optimized based on CER measurements using readily available steady-state data on the ratio between the cellular productivity and the CER. It was shown that the algorithm is two to three times faster than the algorithm based on cell mass concentration measurements. In the experimental study the CER was maximized without any information on the relationship between the cellular productivity and the CER. It took about 40 h for the process to converge, while about 80 h was required when the optimization was based on cell mass measurements. The attained steady state was found to be different but fairly close to that obtained with cell measurements. Briefly discussed is a switching to the cell-mass-based algorithm at the final stage of the optimization to overcome a potential inaccuracy.  相似文献

The lipid lamellae in the stratum corneum (SC) play a key role in the barrier function of the skin. The major lipids are ceramides (CER), cholesterol (CHOL) and free fatty acids (FFA). In pig SC at least six subclasses of ceramides (referred to as CER 1, 2-6) are present. Recently it was shown that in mixtures of isolated pig SC ceramides (referred to as CER(1-6)) and CHOL two lamellar phases are formed, which mimic SC lipid organisation very closely [J.A. Bouwstra et al., 1996, J. Lipid Res. 37, 999-1011] [1]. Since the CER composition in SC originating from different sources/donors often varies, information on the effect of variations in CER composition on the SC lipid organisation is important. The results of the present study with mixtures of CHOL including two different CER mixtures that lack CER 6 (CER(1-5) mixtures) revealed that at an equimolar molar ratio their lipid organisation was similar to that of the equimolar CHOL:CER(1-6) and CHOL:CER(1,2) mixtures, described previously. These observations suggest that at an equimolar CHOL:CER ratio the lipid organisation is remarkably insensitive toward a change in the CER composition. Similar observations have been made with equimolar CHOL:CER:FFA mixtures. The situation is different when the CHOL:CER molar ratio varies. While in the CHOL:CER(1-6) mixture the lamellar organisation hardly changed with varying molar ratio from 0.4 to 2, the lamellar organisation in the CHOL:CER(1-5) mixtures appeared to be more sensitive to a change in the relative CHOL content, especially concerning the changes in the periodicities of the lamellar phases. In summary, these findings clearly indicate that at an equimolar CHOL:CER molar ratio the lamellar organisation is least sensitive to a variation in CER composition, while at a reduced CHOL:CER molar ratio the CER composition plays a more prominent role in the lamellar phases. This observation may have an implication for the in vivo situation when both the CER composition and the CHOL:CER molar ratio change simultaneously.  相似文献

Suppression of operant behavior by exposure to pain reactions of conspecifics was examined with pigeons. Three groups of pigeons were trained on a VI schedule, and were then exposed to the pain reactions of an adjoining bird to electric shocks. Although every subject showed suppression of responding, the suppression decreased with repeated exposures. Following this assessment, a conditioning group received conditioned suppression training in which the pain reaction of the adjoining bird was the CS and an electric shock was the US; a shock exposure group received the electric shock without any explicit CS; and, a no-shock group did not receive any shock. After these treatments, every group was exposed to the pain reactions of the adjoining bird (test 1). The conditioning group and the shock exposure group showed clear suppression in responding, but the no-shock group did not.The no-shock group then received the shock exposure treatment and the conditioned suppression training succesively, and the shock exposure group received the conditioned suppression training. Results of tests with the pain reaction of the adjoining bird supported the results of the test 1, however, suppression caused by the shock exposure was not so clear in the no-shock group.The present results demonstrated that conspecific behavior can become a CS by conditioned suppression training, and, the behavior to an aversive stimulus can acquire aversive properties for other conspecifics when they have shared the exposure to the same aversive stimulus.  相似文献

Bradycardic reflexes in man are both of scientific and clinical interest. Using the methods of experimental psychophysiology, control over relevant independent variables permits the study of fine-grained temporal physiologic response topographies, and of psychological factors that may modify the reflex. In addition, information can also be sought through interdisciplinary collaborations with experimental physiologists in order to shed light on the mechanism of the reflexes. These general features of the approach are illustrated by presenting data on two bradycardic reflex preparations: the laboratory dive analog, and the 90-degree negative tilt. The dive-analog studies have shown that a) the dive-reflex proper is a late-occurring bradycardia accompanied by a late-occurring vasoconstriction; and b) for the elicitation of this reflex, both breath-holding and face immersion are necessary. In addition, the physiologic manipulation of temperature affects the reflex in an inverse way over the range of 10 degrees to 40 degrees C, while the sense of control (a psychological variable) attenuates the reflex. The negative-tilt preparation produces a bradycardic response that is ideal as a Pavlovian unconditional response. Some Pavlovian conditioning arrangements, especially an "imaginational" form, do produce significant conditional bradycardic responding, and this has both potential clinical (e.g., biofeedback-related) and theoretical (e.g., S-R vs. S-S accounts of Pavlovian conditioning) applications. The paper ends with a comment on the cognitive paradigm shift in psychology. Although this shift is of importance, it is suggested that it is also important to "remember the response."  相似文献

A conceptually simple two‐dimensional conditional reference curve is described. The curve gives a decision basis for determining whether a bivariate response from an individual is “normal” or “abnormal” when taking into account that a third (conditioning) variable may influence the bivariate response. The reference curve is not only characterized analytically but also by geometric properties that are easily communicated to medical doctors – the users of such curves. The reference curve estimator is completely non‐parametric, so no distributional assumptions are needed about the two‐dimensional response. An example that will serve to motivate and illustrate the reference is the study of the height/weight distribution of 7–8‐year‐old Danish school girls born in 1930, 1950, or 1970.  相似文献

The mechanism of allosuppression has been investigated. As previously described, negative allogeneic effects result when T cells recognize MHC-encoded alloantigens on responding lymphocytes, preventing the generation of a secondary anti-PFC response in vitro. Several experiments suggested that this suppression was not due to the generation of cytotoxic effectors. First, effective suppression occurred only when T cells, either unprimed or alloantigen activated, were added during the first 24 hr of the responding culture. Even previously generated cytotoxic effectors were relatively ineffective when added late in the response. Furthermore, no detectable cytotoxic effectors were found in suppressed cultures. The major target of suppression was the responding B cell. Only B cells carrying alloantigens (thus recognized by the T cells) were suppressed; bystander B cells were little affected. Thus allosuppression appears to involve the recognition by T cells of alloantigens on responding B cells and direct suppression of some early event in the development of these B cells into PFC. The responses of primed B cells were found to be preferentially sensitive to the suppressive effects of allo- T cells, whereas the response of unprimed B cells was influenced preferentially by the helper effects of alloantigen-activated T cells. It is possible that the state of differentiation of the B cell may determine the outcome of the interaction with regulatory T cells.  相似文献

The lipids in the uppermost layer of the skin, the stratum corneum (SC), play an important role in the skin barrier function. The three main subclasses in the SC lipid matrix are ceramides (CER), cholesterol, and free fatty acids. In inflammatory skin diseases, such as atopic dermatitis and psoriasis, the SC lipid composition is modulated compared to the composition in healthy SC. One of the main alterations is the molar ratio between the concentration of CER N-(tetracosanoyl)-sphingosine (CER NS) and CER N-(tetracosanoyl)-phytosphingosine (CER NP), which correlated with an impaired skin barrier function. In the present study, we investigated the impact of varying the CER NS:CER NP ratios on the lipid organization, lipid arrangement, and barrier functionality in SC lipid model systems. The results indicate that a higher CER NS:CER NP ratio as observed in diseased skin did not alter the lipid organization or lipid arrangement in the long periodicity phase encountered in SC. The trans-epidermal water loss, an indication of the barrier functionality, was significantly higher for the CER NS:CER NP 2:1 model (mimicking the ratio in inflammatory skin diseases) compared to the CER NS:CER NP 1:2 ratio (in healthy skin). These findings provide a more detailed insight into the lipid organization in both healthy and diseased skin and suggest that in vivo the molar ratio between CER NS:CER NP contributes to barrier impairment as well but might not be the main factor.  相似文献

A new Pavlovian conditioning preparation was developed using the nictitating membrane of the restrained pigeon. Either visual or auditory stimuli served as conditioned stimuli (CSs) with an unconditioned stimulus (US) of a puff of air to the cornea. Movement of the nictitating membrane constituted the conditioned and unconditioned responses (CR and UR). Conditioning was studied with the Kamin blocking procedure. In agreement with findings from other conditioning preparations, responding to the redundant stimulus was attenuated relative to a stimulus that received the same number of CS-US pairings in a compound-conditioning procedure. Although response attenuation occurred, substantial individual variation was observed within the blocking procedure, a finding with some precedent in the experimental literature. Theoretical analysis and neural-network simulations indicate that inter-subject variation in response attenuation may result from differences in the extent to which contextual stimuli contribute to the functional CS.  相似文献

This paper reviews a variety of studies designed to examine the effects of extinction upon control by specific stimulus-outcome (S-O) associations in Pavlovian conditioning. Studies conducted with rats in a magazine approach conditioning paradigm have shown that control by specific S-O associations is normally unaffected by extinction treatments, although other aspects of conditioned responding seem affected in a more enduring way. However, recent work suggests that extinction can undermine control by such associations if it is administered after the conditioned stimulus is weakly encoded. The results from these studies suggest that it may be important to consider multiple response systems in assessing the impact of extinction. Studies conducted with the flavor preference learning paradigm in rats also show that specific S-O associations can be undermined by procedures that involve presenting a flavor cue in the absence of its associated nutrient. These findings provide no support for the view that flavor preference learning necessarily entails some unique learning process that differs from more conventional processes. As in other situations, some of these effects likely involve a masking process, but the extent to which masking or true associative weakening occurs in extinction more generally is a topic that is not well understood. Finally, we present some data to suggest that extinction also involves conditional "occasion-setting" control by contextual cues. Special procedures are recommended in assessing such learning when the goal is to distinguish this form of learning from other more conventional mechanisms of extinction.  相似文献

Exposure of human keratinocytes to UVA radiation induced an increase in ceramide (CER) intracellular content, with a dose-dependent effect within the range of 4-9 J/cm(2). The production of CER reached a maximum 2 h after UVA irradiation. The increase of CER was proportional to the intracellular content of reactive oxygen species, was prevented by the antioxidant vitamin E, and enhanced by the prooxidant buthionine-sulfoximine, suggesting the involvement of an oxidative stress. UVA decreased both neutral and acid sphingomyelinase activities measured in vitro. A direct cleavage of sphingomyelin to CER by UVA, recently described, was not observed under our experimental conditions. We also show that, downstream of CER, UVA activated the Ser/Thr kinases ERK, JNK, and p38. Since ceramide has been shown to play a role in stress kinase activation, our results provide a possible mechanism for UVA-induced activation of stress kinases via ceramide formation. However, the actual mechanisms whereby CER is produced in cultured cells under UVA exposure remain to be specified.  相似文献

Nonaversive unconditional stimuli (USs) are seldom used in human classic conditioning of autonomic responses. One major objection to their use is that they produce deficits in electrodermal (ED) second- and third-interval response conditioning. However, a nonaversive reaction time (RT) task that includes feedback of success has been shown to be an effective US while avoiding this disadvantage (Lipp and Vaitl 1988). The present study compared this new RT task (RT-new) with a traditional RT task (RT-old) and with a standard aversive US (shock) in differential classic conditioning of ED, heart rate (HR), and digital pulse volume (DPV) responses. Eight-second-delay differential conditioning was applied in three groups of 12 subjects each. Simple geometric features (square, cross) displayed on a television screen served as conditional stimuli (CS+ and CS-). In acquisition, there were no statistically significant differences among the groups; differential conditioning did occur in HR, first- and second-interval ED responses, and first-interval DPV responses. Separate analyses within each group, however, revealed that there was no second-interval ED conditioning in the RT-old group. During extinction, neither DPV nor second-interval ED conditioning could be obtained, whereas HR and first-interval ED conditioning occurred in each group. In third-interval omission ED responses, RT-old and shock groups exhibited extinction, while response differentiation was maintained in the RT-new group throughout extinction. The RT task including feedback proved to be as reliable a US as a standard aversive US, whereas application of a traditional RT task again yielded some weaknesses in second-interval ED conditioning.  相似文献

Characterization of overall ceramide species in human stratum corneum   总被引:1,自引:0,他引:1  
Ceramides (CERs) in human stratum corneum (SC) play physicochemical roles in determining barrier and water-holding functions of the skin, and specific species might be closely related to the regulation of keratinization, together with other CER-related lipids. Structures of those diverse CER species, however, have not been comprehensively revealed. The aim of this study was to characterize overall CER species in the SC. First, we constructed 3D multi-mass chromatograms of the overall CER species, based on normal-phase liquid chromatography (NPLC) connected to electrospray ionization-mass spectrometry (ESI-MS) using a gradient elution system and a postcolumn addition of a volatile salt-containing polar solvent. The CERs targeted from the 3D chromatograms were structurally analyzed using NPLC-ESI-tandem mass spectrometry (MS/MS), which resulted in the identification of 342 CER species in the inner forearm SC. This led to the discovery of a new CER class consisting of alpha-hydroxy fatty acid and dihydrosphingosine moieties, in addition to the 10 classes generally known. The results also revealed that those CERs contain long-chain (more than C(18))-containing sphingoids and a great number of isobaric species. These novel results will contribute not only to physiochemical research on CERs in the SC but also to lipidomics approaches to CERs in the skin.  相似文献

Suppression of dengue and malaria through releases of genetically engineered mosquitoes might soon become feasible. Aedes aegypti mosquitoes carrying a conditionally lethal transgene have recently been used to suppress local vector populations in small-scale field releases. Prior to releases of transgenic insects on a wider scale, however, most regulatory authorities will require additional evidence that suppression will be effective in natural heterogeneous habitats. We use a spatially explicit stochastic model of an Ae. aegypti population in Iquitos, Peru, along with an uncertainty analysis of its predictions, to quantitatively assess the outcome of varied operational approaches for releases of transgenic strains with conditional death of females. We show that population elimination might be an unrealistic objective in heterogeneous populations. We demonstrate that substantial suppression can nonetheless be achieved if releases are deployed in a uniform spatial pattern using strains combining multiple lethal elements, illustrating the importance of detailed spatial models for guiding genetic mosquito control strategies.  相似文献

Ten rats were deprived of water and trained to lick a tube for saccharin reinforcement. In each of the two sessions that followed, the rats received six contiguous pairings of a 30-second illumination of the houselight and a 0.75 second, 0.10 mA electric shock while licking. No sign of conditioning was observed during the first experimental session, but profound conditioning was observed on the first and subsequent trials of the second conditioning session. No comparable change in the rate of licking was observed in groups of rats that received only presentations of the visual stimulus, only presentations of the electric shock, or random presentation of the visual stimulus and electric shock during the first conditioning session. These data establish that the incubation of conditional suppression is an associative phenomenon.  相似文献

Plant viruses encode RNA silencing suppressors (VSRs) to counteract the antiviral RNA silencing response. Based on in-vitro studies, several VSRs were proposed to suppress silencing through direct binding of short-interfering RNAs (siRNAs). Because their expression also frequently hinders endogenous miRNA-mediated regulation and stabilizes labile miRNA* strands, VSRs have been assumed to prevent both siRNA and miRNA loading into their common effector protein, AGO1, through sequestration of small RNA (sRNA) duplexes in vivo. These assumptions, however, have not been formally tested experimentally. Here, we present a systematic in planta analysis comparing the effects of four distinct VSRs in Arabidopsis. While all of the VSRs tested compromised loading of siRNAs into AGO1, only P19 was found to concurrently prevent miRNA loading, consistent with a VSR strategy primarily based on sRNA sequestration. By contrast, we provide multiple lines of evidence that the action of the other VSRs tested is unlikely to entail siRNA sequestration, indicating that in-vitro binding assays and in-vivo miRNA* stabilization are not reliable indicator of VSR action. The contrasted effects of VSRs on siRNA versus miRNA loading into AGO1 also imply the existence of two distinct pools of cellular AGO1 that are specifically loaded by each class of sRNAs. These findings have important implications for our current understanding of RNA silencing and of its suppression in plants.  相似文献
