研究结核杆菌pckA基因编码的磷酸烯醇型丙酮酸羧激酶(PEPCK)诱导机体产生的保护性免疫反应。用敲除pckA基因的牛结核杆菌BCG和野生型BCG分别感染小鼠,取肝、肺、脾进行病理分析,并进行脾细胞培养,检测CD4 、CD4 /CD8 、细胞因子IFNI-γI、L-12和TNF等。用敲除pckA基因的BCG感染的小鼠比野生型BCG感染的小鼠体内产生的结核结节少且不典型,炎性程度低。野生型BCG感染的小鼠脾脏内的CD4 T细胞和CD4 /CD8 、细胞因子IFN-γ、IL-12、TNF均明显高于敲除pckA基因BCG感染的小鼠。pckA基因为结核杆菌生长所必需,其编码产物PEPCK能够刺激机体产生免疫反应,是一种很好的疫苗候选分子。  相似文献

利用Triton X-100对叶绿体膜的作用,可快速地从蚕豆幼叶制备较纯净的细胞核,它具有较高的RNA聚合酶活力。比较了两种分离核的方法,证明利用匀浆法制备的核具有较高的活力。核活力与发育时期有关系,茎端和第1对幼叶的核活力显著高于第2和第3对叶片的核活力。此外,核活力明显地受反应液内锰离子的抑制。  相似文献

目的:为利用原生质体融合技术转移霸王抗旱基因。方法:采用酶解法分离霸王原生质体,比较了霸王子叶和愈伤组织游离原生质体的产量和活力,不同渗透压和起始密度对原生质体分裂频率的影响。结果:愈伤组织游离的原生质体产量和活力均高于子叶,原生质体产率可达2.4×106个/g.FW,活力达89%。采用液体浅层培养,在附加2,4-D(2mg/L)、6-BA(1.0mg/L)、2%蔗糖和甘露醇(0.4mol/L)的DPD培养基中,原生质体分裂频率最高,达68.6%。转移到附加2-iP(3mg/L)、KT(1.0mg/L)、6-BA(1.0mg/L)的分化培养基上,获得2个再生苗。结论:采用酶解法游离霸王愈伤组织,可获得高活力和高分裂频率的霸王原生质体。  相似文献

本文应用本实验室发现的着染HeLa细胞染色体鞘的一例红斑狼疮病人抗血清对分离出的蚕豆染色体及核和蚕豆根尖生长点压片进行了免疫荧光染色,从两种不同制片方法得到的染色体上均染出了清晰的染色体鞘免疫荧光。之后又用此血清对蚕豆三天龄幼苗中(去除子叶、种皮)提取出的总蛋白进行Western印迹分析,获得的带型显示分子量为41K,39K,18K,17K的蛋白质可与抗血清中的抗体特异性结合,另有分子量为87K,38K,35K,34K,32K的蛋白质也可能具有与抗体特异性结合的能力。  相似文献

Summary A monoclonal anti-DNA antibody (HB2) specific for poly dG-poly dC nucleotides was used to stain metaphasic lymphocyte or amniotic cell human chromosomes. HB2 fixation was revealed using either a peroxidase-or a rhodaminelabeled anti-mouse immunoglobulin antiserum. The staining pattern of the chromosomes was dependent on the HB2 concentration: R-banding could be observed at high antibody dilution. Previous trypsinization of metaphasic preparations demonstrated a precise and reproducible typical R-banding independent of the HB2 concentration. This technique appears to be an interesting alternative to other R-banding procedures. The specificity of the antibody allows a better understanding of the biochemical mechanism of R-banding.  相似文献

The work been concerned with a study of the kinetics of reassociation of total DNA and that of the fraction of unique sequences in plants from the Vicia family, i. e. Vicia faba and Vicia sativa. The size of the genome was determined by the kinetics of reassociation of the DNA of the fraction of unique sequences and the amount of DNA per nucleus was determined cytophotometrically. It has been shown that the size of the genome expressed in C(0)t units and the size expressed in gramms are not the same which testifies to the absence of true unique genes in the genome of the species studied. The analysis of the possible methodical errors was carried out. On the basis of the data obtained a suggestion was made of a model of chromosomes organization including 12 units of polynemization for Vicia faba and 4 units for Vicia sativa.  相似文献

The observations have been made on the structures of mitotic prophase nuclei and chromosomes in Vicia faba root meristematic cells. Two methods, i.e., the cell squashing and the nucleus isolation methods, were applied in present study to prepare the specimen of chromosomes and nuclei. Chromosomal fibers 0.3—0.5 μm in diameter were observed in the squashed preparations stained with Giemsa, and in the isolated nucleus preparations treated with 0.05% EDTA followed by Giemsa staining. Using Feulgen reaction, it has been demonstrated that these fibers are nuclear origin containing DNA. The results suggest that this order of chromosomal fiber may be one structural level in the chromosomes in Vicia faba. This conclusion is in support of the view which holds that there exists an intermediate level of structure between the 250–300Å chromatin fiber and the chromosome.  相似文献

In situ hybridization with synthetic plant telomeric sequences resulted in labeling of all broad bean (Vicia faba) chromosomes at their ends only. Telocentric chromosomes derived by fission of the metacentric satellite chromosome of V. faba also showed signals at both of their ends, whereas the ancestral metacentric did not display signals at its primary constriction, the point of fission. As in V. faba, all acrocentric mouse chromosomes were labeled by in situ hybridization with a vertebrate telomeric probe at both ends of each chromatid exclusively. However, different metacentric Robertsonian chromosomes derived by fusion of defined acrocentrics did not show signals at their primary constrictions. The mechanism of Robertsonian rearrangement leading to a pseudoaneuploid increase or decrease in chromosome number therefore cannot consist solely of a simple fission or fusion of chromosomes without a concomitant gain or loss of chromatin material. The additional assumption of a subdetectable deletion of telomeric sequences after fusion and amplification of these sequences following fission is necessary to explain the present observations.  相似文献

The effect of exogenous DNA of syngeneic origin on the course of reparation of meristematic cells ofVicia faba primary roots followed after treatment with monofunctional alkylating agent, ethyl methanesulphonate (EMS), was tested. Time course of alternations in mitotic activity of investigated cellular population and the dynamics of formation of postmetaphase chromosomal aberrations was evaluated. A reparation of damaged cells was significantly supported by syngeneic DNA; its application induced an increased incidence of cellular division already in the early intervals of the repair which was accompanied by concomitant decrease of microscopically detectable rupture in the chromosomes. The study performed on the localization of induced damages occurring in metaphase chromosomes pointed out an increased sensitivity of small chromosomes ofVicia faba to EMS. Similarly, a reparative action of syngeneic DNA was exhibited by significant decrease of aberrations frequency, predominantly in the same chromosomal group. Per cent representation of individual types, not affected by the action of syngeneic DNA, was established by detailed classification of induced aberrations. In both cases, isochromatide breaks were found of greatest predominance.  相似文献

The short-term evolution of micronuclei derived from acentric fragments and whole chromosomes was studied in root tips of Vicia faba. Micronuclei were induced by X-rays (30 cGy and 120 cGy) and colchicine (10(-5) M and 3 X 10(-4) M). Frequencies of chromosome breakage or loss of micronuclei in interphase and mitotic cells were studied. The DNA content of micronuclei in interphase cells was also measured. Micronuclei derived from whole chromosome showed a higher probability to survive and to undergo mitotic condensation in synchrony with main nuclei than micronuclei derived from an acentric fragment. PCC (Premature Chromosome Condensation) was not observed for both types of micronuclei in Vicia faba, in contrast to the ones reported in mammalian cells in culture.  相似文献

During the spring and summer, in 2003-2004, pea viruses were identified in twenty pea fields of Tehran. Some leaf samples were collected randomly from pea fields of Tehran. Samples were tested by Double Antibody Sandwich Enzyme Linked Immunosorbent Assay (DAS-ELISA) technique using polyclonal antiserum of Alfalfa mosaic virus (AMV), AS-0001, DSMZ, Braunschweig, Germany). The samples were extracted in 0.1 M Phosphate buffer pH 7 to 7.5 and inoculated on Chenopodium amaranticolor, Chenopodium quina, Phaseolus valgaris, Vicia faba, Vignia unguiculata. Pea cultivars were infected by AMV, causing mild mosaic, translucent veins and a diffuse green-yellow of tender parts and spots may also was involved necrosis of tissue. Infected plants grow slowly and malformed pods produce fewer ovules. In Chenopodium amranticolor, C. quina chlorotic and necrotic flecks, and Vicia faba systemic mosaic had produced. Phaselous vulgaris and Viginia unguiculata are good assay hosts for strains that produce local lesions after 3-5 days in these plants. Back inoculated on Pisum sativum and Vicia faba and tested with DAS-ELISA that had been confirmed the results. This is the first report of AMV on pea from Iran.  相似文献

Meristematic cells of Vicia faba L. were labeled with rabbit anti-actin antibody and FITC-conjugated goat anti-rabbit lgG antibody and observed with fluorescence microscopy. Both the nuclei and chromosomes sent forth distinctive fluorescence, indicating that actin is present in the nuclei and chromosomes. Sections were reacted with the anti-actin antibody and protein A-colloidal gold and observed with transmission electron microscopy. Gold particles were found over the whole nuclei, and a lot of particles were concentrated in condensed chromatin areas and nucleoli, confirming the observations with the fluorescence microscopy. V. faba nuclei and chromosomes were treated with DNase Ⅰ and 2 mol/L NaC1, and DNA and histone-depleted nuclei and chromosomes were obtained. Indirect immunofluorescence tests showed that the DNA and histone-depleted nuclei and chromosomes reacted positively with the anti-actin antibody. These results demonstrated that actin exists not only in intact nuclei and chromosomes but also in DNA and histone-depleted nuclei and chromosomes of V. faba. In addition, the authors' results indicate that tropomyosin is present in the nuclei and chromosomes of V. faba. Presence of actin in nuclei and chromosomes as well as in DNA and histone-depleted nuclei and chromosomes of higher plants is discussed.  相似文献
