Five species belonging to the family Prymnesiaceae (one Prymnesium and four Chrysochromulina) have been identified in cultures obtained from water collected in the Bay of Banyuls‐sur‐Mer (Mediterranean Sea, France) using LM, SEM, and TEM. Two are described as new species, Chrysochromulina lanceolata sp. nov. and C. pseudolanceolata sp. nov. Both species are large and lanceolate with an acute posterior and two anterior arms. They are easily detectable with LM but difficult to distinguish to species level with live cells, without experience. EM reveals two completely different scale patterns in the two species. Cells of C. lanceolata are 21–38 μm long, 7–12 μm wide, and 3–7 μm thick. They possess two subequal flagella (30–51 and 29–44 μm), and the haptonema is shorter than the flagella (23–37 μm). The cell body is covered by plate and spine scales. Cells of C. pseudolanceolata sp. nov. are slightly smaller (15–18 × 6–8 μm) with more rounded extremities, two subequal flagella (19–26 and 17–24 μm), and the haptonema is longer than the flagella (about 35 μm). Three types of plate scales are observed in this species. Other findings are C. alifera Parke et Manton and C. throndsenii Eikrem (a new record for the Mediterranean Sea). Prymnesium faveolatum Fresnel, a new toxic species recently described, is illustrated with both LM and SEM.  相似文献

A phylogeny of 21 haptophyte algae was inferred by maximum parsimony, neighbor-joining, and maximum likelihood analyses of sequences of the plastid-encoded gene, rbcL. Sequence variation in the spacer region of the RUBISCO operon was also investigated. In all the rbcL trees constructed, the haptophytes form two distinct clades: one includes the Pavlovales and the other includes the Prymnesiales, Coccosphaerales, and Isochrysidales (all sensu Parke and Green 1976 . This relationship coincides with the recent taxonomic treatment splitting the division into two subclasses, the Prymnesidae and Pavlovidae ( Cavalier-Smith 1989 ) or the Prymnesiophycidae and the Pavlovophycidae using botanical suffixes ( Jordan and Green 1994 ), or into two classes, the Patelliferea and the Pavlovea ( Cavalier-Smith 1993 ). In the Prymnesiophycidae, all the coccolithophorids examined are placed in a single clade, which suggests a single origin of the coccolithophorids and the ability of coccolith formation in the haptophytes. The genus Chrysochromulina is polyphyletic. Species of Chrysochromulina with a very long haptonema and a compressed cell body (typical of species including the type C. parva Lackey) form a clade, including Imantonia, that is often classified in the Isochrysidales in the neighbor-joining tree, whereas some species possessing a nontypical cell body and cell covering form a clade with Prymnesium and Platychrysis in all trees. It is suggested that loss of the haptonema in Imantonia and the reduction in Prymnesium and Platychrysis occurred secondarily and independently in two different lineages. Within the coccolithophorids, four clades are recognized: Pleurochrysis, Calyptrosphaera-Cruciplacolithus-Calcidiscus-Umbilicosphaera, Helicosphaera, and Emiliania-Gephyrocapsa. A non-coccolith-bearing haptophyte, Isochrysis, is an ingroup of the Emiliania-Gephyrocapsa clade, suggesting its secondary loss of the ability to form a coccolith. Sequence comparison of the spacer region of RUBISCO operon supports most results obtained in the analysis of rbcL sequences. Monophyly of the Prymnesiales sensu Parke and Green is still unclear because of low (<50%) bootstrap support for this group.  相似文献

Based on material collected from Cape Town, a new sand-dwelling, marine species of Prymnesium is described. Using light and electron microscopy, Prymnesium nemamethecum sp. nov. has been found to resemble other species of the genus in size, organelle arrangement, and swimming behavior. It differs from other described species in that it has three types of scales, one of which is confined to the region of appendage insertion and forms a sheath of simple plate scales over the haptonema. In addition, the scales constituting the proximal body scale layer(s) are unusual because they are not simple plate scales but are specifically ornamented.  相似文献

S. sphagnicola resembles other species of Synura previously described by electron microscopy in most features of structure but differs in possessing pyrenoids and up to five cylindrical stacks of smooth cisternae which occur between the pyrenoids and leucosin vesicles. Each stack is surrounded by a tubular cisterna which bears ribosomes on its distal face but there are no clear permanent connections between this and the chloroplast ER. Other features apparently unique to this species previously known from light microscopy are described. These include the axial position of the chloroplasts; the peripheral position of the leucosin vesicles; and the loose attachment of the scales. The structure of the body scales is described for the first time from sections. The flagellar scales are formed in the swollen edges of the Golgi cisternae and appear to pass to the cell surface in large vesicles.  相似文献

The haptophytes Prymnesium parvum Carter and Prymnesium patelliferum Green, Hibberd, and Pienaar are two closely related species, which can only be distinguished by minor differences in the morphology of their organic body scales. The two Prymnesium species are reported to coexist at several locations, including the Sands-fjord system in southwestern Norway. Comparisons of physiology and toxicity within the two species have failed to reveal differences that can add to the small morphological distinctions used to separate them. To investigate the genetic relationship between the two species, we compared the sequence of the first internal transcribed spacer region (ITS1)and length variation in one intron separating calmodulin genes for four P. parvum strains and eight P. patelliferum strains. Both the ITS1 sequence and the banding patterns obtained by PCR amplification of one intron in the calmodulin genes indicated that the Prymnesium isolates are related by their geographic origin instead of 4 their species affiliation. The results indicate that P. parvum and P. patelliferum are so closely related that they could be considered one species. Alternatively, we discuss the possibility that the two species might be joined in a heteromorphic haploid-diploid life cycle, as is now widely reported for other haptophycean algae.  相似文献

The fine structure of the type species of the genusCyanothece Komárek 1976,C. aeruginosa, is described and compared with the main cytological characteristics of morphologically related members of the generaCyanobium, Cyanobacterium andSynechococcus. Several morphological features, such as cell walls with thick outer layers containing a special type of vesicles, position of thylakoids, keritomy (net-like appearance of protoplast caused by arrangement of thylakoids, net-like nucleoids and/or by tendency to form intrathylakoidal spaces) and a special structure of mucilaginous envelopes were found to be characteristic of this genus, supporting its separate position among coccal cyanoprokaryotes (cyanobacteria, cyanophytes). The taxonomic significance of ultrastructural features in all mentioned genera is discussed.  相似文献

Summary Sea urchin embryos, 8-cell stage to pluteus stage, fixed in osmium tetroxide and embedded in Epon 812 were observed by electron microscopy. At no point in the development were syncytial junctions between the embryonic cells found. During the cleavage stages the membrane contact was closer than in later stages. In early blastula stages intercellular clefts appeared which in the gastrula stage demarcate every cell. At the same time a ringshaped desmosome structure develops at the outer cell surface. In the pluteus stage a closer cell contact is re-established. With proceeding embryogenesis endoplasmic membranes will attach to the cell membrane. These membrane structures may even be of nuclear origin. Gradually, long protrusions, vesicles and lamellae begin to be formed from the nuclear membrane. The commencement of this nuclear activity coincides in time with the formation of nucleoli. Summary The Structure of the hypothalamic neurosecretory cells during their activity in continuous light and dark conditions and depending on the seasonal alteration has been investigated in Zoarces viviparus L. in the level of light and electron microscopy.The depletion of AF- or AT-stainable material, of the elementary neurosecretory granules and the disappearance of smooth-surfaced vesicles occur during April and June. The accumulation of the stanable material as well as the elementary neurosecretory granules and the smooth-surfaced vesicles has been observed in the cell body in late September.An excess increase in number of the smooth-surfaced vesicles of dark-induced animals and light-induced animals kept in a black stained tank is apparent. On the other hand, the disappearance of the elementary granules and the smooth-surfaced vesicles and an enlargement in the reletive nuclear surface of the neurosecretory cells of light-induced animals which were kept in a gray stained tank in April is also evident.Taking into consideration the responsiveness of both the elementary neurosecretory granules and the smooth-surfaced vesicles in relation to the external environment as well as their topographical arrangement in the cell body the possible differences in their origin and function are discussed.Numerous studies indicate that the physical environment is in part responsible for the functional properties of the hypothalamic neurosecretory centers of various vertebrates. In the level of light microscope, it has been repeatedly shown that the neurosecretory cells are activated in the animals subjected to continuous illumination, and in those subjected to long daily photo period (Oksche et al., 1958; Fiske and Greep, 1959; Öztan and Gorbman, 1960; Satyanesan, 1965). The enlargement of the nuclei and the amount of Gomori — or aldehyde fuchsin — stainable material, as well as the enzymatic activity, were used as the criteria of cellular activity in the histological preparation. In the level of electronmicroscope it has been shown that Gomori positive material (Bargmann, 1949) is represented as aggregated elementary neurosecretory granules in the ultrathin sections of neurohypophysis (Bargmann et al,. 1957). Although the fine structure of the hypothalamic neurosecretory cells of various fishes and higher vertebrates have been studied in normal and experimental conditions (Palay, 1960; Lederis, 1962, 1964; Follenius et Porte, 1962; Follenius, 1963; Murakami, 1961–1964; Gansler, 1965; Holmes, 1965) the type of granules and their function still remains as an open question (see Bargmann, 1963; Knowles, 1965).This work was aided by a grant from Nato and the Deutsche Forschungsgemeinschaft.This paper was written as a tribute in honor of the 60th birthday of Prof. Dr. Berta Scharrer.  相似文献

Electron microscopy of dried preparations of Arctic nanoplankton has permitted a second species of the recently erected genus Pappomonas Manton et Oates to be described and named. P. virgulosa sp. nov. is characterised by coccolith appendages composed of a central shaft ending in a cluster of four finger-like rods. There are also calcified plates of characteristic construction, without appendages. Detailed observations on the coccoliths, especially on the shapes and arrangement of component calcite crystallites and the presence of unmineralised components, have permitted further comparisons to be made with equivalent structures in Papposphaera lepida Tangen, recently investigated in the southern hemisphere, as well as with Pappomonas flabellifera Manton et Oates. As a working hypothesis, an interpretation of coccolith morphology in all three taxa has been formulated in terms of known structural components of uncalcified scales in Chrysochromulina spp. The need for fuller information on certain details, notably the coccolith bases in Pappomonas spp. is stressed.  相似文献

A mass development of Chrysochromulina parva Lackey — 614 000 cells per ml, associated with fish mortality, is reported from a small Danish lake. None of the analyses performed showed any reason for the fish kill. Even oxygen conditions were favourable. Thus toxins from the Chrysochromulina might be responsible for the fish kill. If true, this would be the first record of toxicity in a freshwater Chrysochromulina.  相似文献

The tribePlucheeae (Benth.)A. Anderb., has been analysed cladistically by means of a computerized parsimony program (Hennig 86), using theArctotideae as outgroup. The results of the analysis are presented in a consensus tree and one cladogram. Four major monophyletic subgroups can be recognized: TheColeocoma group (3 genera), thePterocaulon group (3 genera), theLaggera group (6 genera), and thePluchea group (12 genera). All recognized genera are described and most genera are supplied with taxonomical notes including comments on their taxonomic status. Genera such asBlumea, Pluchea, andEpaltes are demonstrated to be unnatural assemblages.Monarrhenus andTessaria are both closely related to thePluchea complex. The old generic nameLitogyne Harv. has been taken up for one species ofEpaltes, the genusRhodogeron is reduced to a synonym ofSachsia, and the following new combinations are made;Litogyne gariepina (DC.)A. Summary The aim of this study was to search for uncharacterized components of the plant cytoskeleton using monoclonal antibodies raised against spermatozoids of the fernPteridium (Marc et al. 1988). The cellular distribution of crossreacting immunoreactive material during the division cycle in wheat root tip cells was determined by immunofluorescence microscopy and compared to the fluorescence pattern obtained with antitubulin. Five antibodies are of special interest. Pas1D3 and Pas5F4 detect a diffuse cytoplasmic material, which, during mitosis, follows the distribution of microtubules (MTs) at the nuclear surface and in the preprophase band (PPB), spindle and phragmoplast. The immunoreactive material codistributes specifically with MT arrays of the mitotic apparatus and does not associate with interphase cortical MTs. Pas5D8 is relevant to the PPB and spatial control of cytokinesis. It binds in a thin layer at the cytoplasmic surface throughout the cell cycle, except when its coverage is transiently interrupted by an exclusion zone at the PPB site and later at the same site when the phragmoplast fuses with the parental cell wall.Pas2G6 reacts with a component of basal bodies and the flagellar band in thePteridium spermatozoid and recognizes irregularly shaped cytoplasmic vesicles in wheat cells. During interphase these particles form a cortical network.Pas6D7 binds to dictyosomes and dictyosome vesicles. Summary The synaptic organization in the lateral geniculate nucleus of the monkey has been studied by electron microscopy.The axon terminals in the lateral geniculate nucleus can be identified by the synaptic vesicles that they contain and by the specialized contacts that they make with adjacent neural processes. Two types of axon terminal have been recognized. The first type is relatively large (from 3–20 ) and contains relatively pale mitochondria, a great many vesicles and, in normal material, a small bundle of neurofilaments. These terminals have been called LP terminals. The second type is smaller (1–3 ), contains darker mitochondria, synaptic vesicles, and no neurofilaments. These have been called SD terminals.Both types of terminal make specialized axo-somatic and axo-dendritic synaptic contacts, but the axo-somatic contacts are relatively rare. In addition the LP terminals frequently make specialized contacts with the SD terminals, that is, axo-axonal contacts, and at these contacts the asymmetry of the membranes is such that the LP terminal must be regarded as pre-synaptic to the SD terminal.The majority of the synaptic contacts are identical to those that have been described previously (Gray, 1959 and 1963a) but, in addition, a new type of contact has been found. This is characterized by neurofilaments that lie close to the post-synaptic membrane, and by an irregular post-synaptic thickening. Such filamentous contacts have been found only where an LP terminal contacts a dendrite or a soma.The degeneration that follows removal of one eye demonstrates that the LP terminals are terminals of optic nerve fibres. The origin of the SD terminals is not known.The glial cells often form thin lamellae around the neural processes and tend to isolate synaptic complexes. Summary Phloridzin-insensitive, Na+-independentd-glucose uptake into isolated small intestinal epithelial cells was shown to be only partially inhibited by trypsin treatment (maximum 20%). In contrast, chymotrypsin almost completely abolished hexose transport. Basolateral membrane vesicles prepared from rat small intestine by a Percoll® gradient procedure showed almost identical susceptibility to treatment by these proteolytic enzymes, indicating that the vesicles are predominantly oriented outside-out. These vesicles with a known orientation were employed to investigate the kinetics of transport in both directions across the membrane. Uptake data (i.e. movement into the cell) showed aK t of 48mm and aV max of 1.14 nmol glucose/mg membrane protein/sec. Efflux data (exit from the cell) showed a lowerK t of 23mm and aV max of 0.20 nmol glucose/mg protein/sec.d-glucose uptake into these vesicles was found to be sodium independent and could be inhibited by cytochalasin B. TheK t for cytochalasin B as an inhibitor of glucose transport was 0.11 m and theK D for binding to the carrier was 0.08 m.d-glucose-sensitive binding of cytochalasin B to the membrane preparation was maximized withl- andd-glucose concentrations of 1.25m. Scatchard plots of the binding data indicated that these membranes have a binding site density of 8.3 pmol/mg membrane protein. These results indicate that the Na+-independent glucose transporter in the intestinal basolateral membrane is functionally and chemically asymmetric. There is an outward-facing chymotrypsin-sensitive site, and theK t for efflux from the cell is smaller than that for entry. These characteristics would tend to favor movement of glucose from the cell towards the bloodstream.  相似文献

Both scanning electron and light microscopy were used to examine the epidermal structure of scales taken from several ontogenetic stages of Xenosaurus grandis and Shinisaurus crocodilurus. In addition, scales from all xenosaurid species were examined by scanning electron microscopy to determine scale surface variation among genera, species, and subspecies. A varied and phylogenetically informative morphology characterizes the scale surfaces of xenosaurid lizards. Scale surface morphology is conservative among the species and subspecies of Xenosaurus, but is more variable between the two xenosaurid genera. Their scale surfaces are characterized by folds in the oberhautchen, beta, mesos, and alpha epidermal layers, forming polygonal ridges of a type previously described for the Iguania. The three species of Xenosaurus possess lenticular scale organs, whereas Shinisaurus has scale organs with spikes (bristles). The spikes of Shinisaurus are formed by the beta and oberhautchen layers, with the alpha layer forming a dome-shaped cap over a dermal papilla. Shinisaurus crocodilurus exhibits a dramatic ontogenetic change in scale surface morphology, that is here reported for the first time in any lizard. © 1993 Wiley-Liss, Inc.  相似文献

Out of the eight enzymes involved in the biosynthesis of the isoquinoline alkaloid berberine, at least, two enzymes, berberine bridge enzyme and (S)-tetrahydroprotoberberine oxidase, are exclusively located in a vesicle with a specific gravity of =1.14 g·cm–3 as shown by direct enzymatic assay as well as immunoelectrophoresis. Electronmicroscopic examination of the enzyme-containing particulate preparation from Berberis wilsoniae var. subcaulialata cultured cells demonstrated that it is composed mainly of membranous vesicles. The protein composition of this preparation reveals the presence of only about 20 separable proteins, of which two major ones are berberine bridge enzyme and (S)-tetrahydroprotoberberine oxidase. Incubation of these vesicles with the substrate (S)-reticuline in the presence and absence of S-adenosyl-l-methionine leads to the formation of a red product which was identified as dehydroscoulerine. If the cytoplasmic enzyme S-adenosyl-l-methionine:(S)-scoulerine-9-O-methyltransferase is added to the vesicle preparation in the presence of (S)-reticuline and S-adenosyl-l-methionine, not dehydroscoulerine but columbamine, the immediate precursor of berberine is formed. Some of the quaternary alkaloids are located inside the vesicles; fusion of these vesicles leads to vacuoles containing the quaternary alkaloids. Aim A fraction of vesicles in cells treated with hypertonic solution exhibit multiple dense cores and this is enhanced by treatment with l-3,4–dihydroxyphenylalanine (l-DOPA). These cells were examined to determine if the multicore vesicles are the product of endocytosis or homotypic fusion. Methods Electron microscopy was used to determine the number of multicore vesicles and amperometry was used to examine if the multicore vesicles are a competent fraction of the readily releasable pool. Results In this study, we observed that a substantial portion (15.3%) of large dense core vesicles in PC12 cells contained multiple cores in hypertonic saline loaded with l-DOPA, and amperometric measurements show a bimodal distribution of quantal sizes in treated cells. Conclusions The results suggest that the multicored vesicles are formed from homotypic fusion of LCDVs prior to exocytosis.  相似文献

Summary Electron microscopy of the synaptic morphology of synapses in the cerebral ganglion of the adult ascidian (sea squirt) Ciona intestinalis reveals that the synapses are restricted to the central neuropil of the ganglion. Many of the synapses show a polarity of structure such that pre and post synaptic parts can be identified. The vesicles in the presynaptic bag are of two main diameters 80 and 30 nm respectively. The large vesicles have electron dense contents that vary both in their capacity and dimensions.The pre and postsynaptic membranes are more electron dense than the surrounding membranes, but they are only slightly thicker. Both the pre and post synaptic membranes have electron dense dots some 10 nm in diameter associated with their cytoplasmic surfaces. Sometimes the presynaptic membrane has larger peg-like projections between the vesicles. Associated with the post synaptic membrane are tubules some 10 nm in diameter. These tubules may be the dots cut obliquely.The synaptic cleft material is more electron dense than the surrounding intercellular material, and in it there is a dense line made up of granules about 3–5 nm in diameter. This dense line is usually mid way between the pre and post synaptic membranes, but may be nearer the postsynaptic membrane.No tight junctions between adjacent nerve process profiles have been observed.I wish to thank Professors J. The thallus ofThorea ramosissima was studied electron microscopically. The cells of the medulla, the cortex and the assimilatory hairs differ not only in size and number of plastids and their equipment with thylakoids but also in cell wall structure, the number of mitochondria and the activity of the Golgi apparatus, with dictyosomes transforming complete cisternae into Golgi vesicles with mucilaginous contents in the outer region of the cortex. The pit connections have plugs with a distinct plate—like (not dome-like) outer cap layer. BecauseT. riekei was reported to have dome-like outer cap layers and because this character was the main reason to place theThoreaceae into theBatrachospermales (Pueschel & Cole 1982),T. riekei was reinvestigated, too. A distinct outer cap could not be detected. The reliability of pit plug structure as a taxonomic character and the taxonomic position ofThorea is discussed.  相似文献
