The distribution of IAA-2-14C or IAA-5-3H applied to the apexin the upper and lower (with respect to gravity) halves of geotropicallystimulated stems of cucumber and pea was examined and the resultsobtained are as follows: 1. A larger amount of IAA-2-14C or IAA-5-3H was detected inthe lower than upper half of cucumber hypocotyls with 3-hr geostimulation. 2. A larger amount of IAA-2-14C was distributed in the lowerthan upper epidermis of pea epicotyls with 1-hr geostimulation. 3. Freezing autoradiography revealed that IAA-2-14C was concentratedin the vascular bundles and epidermis of cucumber hypocotyls,the distribution being affected by geostimulation only in thelatter. 4. Application of 1% TIBA in lanolin inhibited the distributionof IAA-2-14C in the lower epidermis of pea epicotyls, causingsuppression of geotropic curvature. 5. From these results, we concluded that IAA which accumulatedin the lower epidermis of the stem upon geostimulation causedthe negative geotropic curvature of the stem. Abstract A detailed study of the changes in growth rate on the two sides of a shoot prior to, and following geostimulation has been undertaken. Data from etiolated Zea seedlings, and cucumber hypocotyls, and from light grown sunflower hypocotyls are presented. In all cases, the differential growth which brings about geocurvature begins simultaneously along the length of the organ. This is contrary to previous reports in the literature which suggested that curvature begins first at the tip of an organ and progresses basipetally. The feature which is common to all species investigated is that growth of the upper side of the shoot ceases following geostimulation. In some cases there is also a marked acceleration in growth rate on the lower side of the shoot (sunflower) but other species show no such acceleration of growth (cucumber). It has been assumed for many years that the major factor causing upward curvature was an acceleration of growth on the lower side of the organ. The data presented here show that the cessation of growth on the upper side is a major, and in some cases the only factor bringing about geocurvature. The data are discussed in relation to the mechanisms which might control geotropic curvature.  相似文献

Apical root segments of Zea mays L. cv. Orla 264 undergo some geotropic curvature in complete darkness but the curvature increases considerably if prior to geostimulation the segments are given a light pretreatment. If the light treatment is follwed by a dark treatment before the root is geostimulated the light-induced response is not changed by dark periods up to 2 h but declines with longer ones, and disappears completely after 5 h of darkness.  相似文献

Analysis of growth during geotropic curvature in seedling hypocotyls   总被引:4,自引:4,他引:0  
Abstract. The patterns of growth in organs curving under the influence of gravity were analysed by time-lapse photography of cress and cucumber hypocotyls which were delimited into 1 mm zones by ion-exchange beads. Geotropic curvature resulted from changes in growth rate on both sides of the organ. Growth inhibition of varying degrees of intensity occurred in all the previously growing zones of the upper (concave) side. An absolute reduction in length due to compression frequently occurred in some zones. Also, in both species growth stimulation was observed on the lower (convex) side. The disparity in growth rate between the upper and lower surfaces varied with time, being more apparent in the subapical region in the first hour of curvature. A later promotion of growth rate on the lower surface subsequently increased the curvature of the more basal zones. Autotropic straightening occurred as a consequence of growth changes, both inhibitory and stimulatory, in the apical zones. These events indicate a polarity of response in which apical zones have precedence over basal zones.  相似文献

Submerged hypocotyl sections from Helianthus have been used to test the effect of neutral buffers on shoot geotropism. When hypocotyls have been abraded, it is found that increasing the molarity (0.25 to 20 mm) of pH 6.8 K-phosphate buffer, as well as other buffering systems, results in a strong inhibition of geotropic curvature. Buffer strength has no such effect on the curvature of nonabraded segments. One possible explanation for these data is that asymmetric shoot growth following geostimulation may require the establishment of a proton gradient across the cell walls of the shoot. When neutral buffers have access to the wall space (i.e. in abraded segments), they may prevent the establishment of such a gradient.  相似文献

The aim of this study was to examine correlations between vegetational zones and thermal climate on the basis of analysis of the thermal distribution of main-component trees in each zone. The whole and main thermal distribution ranges for 73 tree species were obtained from their distributions along the warmth index (WI) gradicnt. Cluster analysis using the WI values at the upper and lower limits of the main distribution ranges as parameters gave seven species groups. The geographical distributions of the species in each group were described. Moreover, the frequency distribution curves of the seven species groups were determined along the WI grandient, and four borderlines (lines I, II, III and IV) were found among them. The WI values at the lines I, II and IV were 50, 90, 175°C month, respectively, and corresponded to boundaries between the subarctic and cool-temperate zones (I), the cool-and warm-temperate zones (II) and the warmtemperated and subtropical/tropical zones (IV), respectively. Line III corresponded to a WI of 135°C month. The WI values at the forest limit and at the lower limit of the subarctic forest were estimated, and their averages were 15.2 and 49.6°C month, respectively. In conclusion, thermal climatic zones in China could be divided by the following WI values: alpine (arctic) zone, WI<15; subarctic zone, 15–50; cool-temperate zone, 50–90; warm-temperate zone, 90–175; subtropical/tropical zone >175°C month.  相似文献

The transport of14C-IAA and14C-ABA applied exogenously to root cap toward the elongation zone was investigated in gravi- and light-stimulated primary roots ofZea mays L. cv. Golden Cross Bantam 70. No significant difference of either IAA or ABA in radioactivities was observed between upper and lower halves of elongation zones during the latent period (0–60 min after the stimulation) of gravitropic response. When quantitative analysis of endogenous IAA and ABA by an internal standard method was carried out 60 min after gravi- and/or light-stimulation, no asymmetric redistribution of either IAA or ABA was observed between upper and lower halves of elongation zones. Light irradiation increased by 20% the contents of ABA in elongation zones. These results suggest that although both IAA and ABA are basipetally transportable and can transmit their information to the elongation zone during a latent period we cannot explain the gravitropic curvature by their redistributions between the two (upper and lower) halves of primary roots ofZea. On the basis of results from the present work and previous papers, the distribution of IAA and ABA in gravistimulatedZea roots is discussed. The recovery from “lodging,” or bending over, by shoots of 42-day-old Avena sativa plants is controlled primarily by a negatively geotropic differential growth of the lower halves of the p-1 node-pulvinus and the base of the p-1 internode, relative to the upper halves. Although geostimulation causes a significant reduction in p-1 internode length, dry matter accumulation in the p-1 node-pulvinus is increased, apparently at the expense of the sheath. Recovery to an angle of 30° is associated with changes in endogenous gibberellin-like substances (GAs), and in differential metabolism of applied [3H]GA4 (1.4 Curie per millimole). Although geostimulation depressed total GAs (relative to upright plant parts) to 0.40 and 0.13 for node-pulvini and sheaths, respectively, it increased them 2-fold for internodes. Within the plant part geostimulation increased GAs (relative to upper halves) 29- and 7-fold in lower halves of node-pulvini and internodes, respectively, but reduced GAs to 0.3 in lower halves of sheaths. At age 42 days a GA4/7-like (nonpolar) substance predominates, with lesser amounts of a GA3-like (polar) substance. Native GAs of Avena include GA3, GA4, and GA7. Geostimulation enhanced the ratio of nonpolar to polar GAs for both halves of internodes, but tended to depress it for sheaths and nodepulvini.  相似文献

The genetically inactive stage 6 oocyte nuclei ofRana temporariacontain certain nuclear bodies that label with nucleolus-specific and coiled body (CB)-specific antibodies. We designate them multicomponent bodies (MCBs) to reflect their mixed composition. Morphologically, each MCB contains five distinct zones: zone I composed of electron-dense fibrils similar to the dense fibrillar component (DFC) of the typical eukaryotic nucleoli; zone II resembled the fibrillar material of the inactive agranular nucleoli of stage 6 oocytes; zone III consisted of fine filamentous material corresponding to the fibrillar center (FC) of lower electron density seen in the typical nucleoli; and zones IV and V contained packed coiled threads typical of CBs. Of these, zone IV was seen in the interior of MCBs and contained tightly packed coiled threads (20 nm thick), while zone V occurred at the periphery and consisted of similar threads but loosely packed and electron dense. The material of both zones IV and V resembled that of CBs. To determine the composition of these zones, we extracted oocytes with a buffer that removes chromatin and most of the soluble proteins and processed them for immunogold labeling with a variety of antibodies. Anti-p80 coilin antibody predominantly labeled zone IV and, to a lesser extent, zone V. Anti-snRNP antibody also showed a similar labeling pattern. Anti-fibrillarin antibody predominantly labeled zone I and to a lesser extent zones IV and V. Anti-B23 antibody labeled all zones. These observations suggest that MCBs contain both nucleolar and CB material. We postulate that MCBs represent storage structures which provide material needed for the early stage of embryogenesis. The demonstration of MCBs further supports the close interrelationship between nucleoli and CBs.  相似文献

Transverse deep zones are often included in treatment wetlands due to an expectation that they can improve contaminant removal by these natural systems. This paper explores the hydraulic behavior of deep zones to provide a better understanding of their potential benefit. First, we use a bench-scale model to show that recirculation within a deep zone is likely under field conditions. Recirculation is believed to provide a mechanism for mixing. We then use a mathematical model to explore the effect on the outlet concentration of replacing a section of a wetland marsh with a deep zone that provides mixing. We observe that even when deep zones have a lower volumetric removal rate than the marsh areas they replace, the presence of a deep zone may decrease the outlet concentration and hence improve the performance of the wetland segment.  相似文献

We used a video digitizer system to measure surface extension and curvature in gravistimulated primary roots of maize (Zea mays L.). Downward curvature began about 25 +/- 7 min after gravistimulation and resulted from a combination of enhanced growth along the upper surface and reduced growth along the lower surface relative to growth in vertically oriented controls. The roots curved at a rate of 1.4 +/- 0.5 degrees min-1 but the pattern of curvature varied somewhat. In about 35% of the samples the roots curved steadily downward and the rate of curvature slowed as the root neared 90 degrees. A final angle of about 90 degrees was reached 110 +/- 35 min after the start of gravistimulation. In about 65% of the samples there was a period of backward curvature (partial reversal of curvature) during the response. In some cases (about 15% of those showing a period of reverse bending) this period of backward curvature occurred before the root reached 90 degrees. Following transient backward curvature, downward curvature resumed and the root approached a final angle of about 90 degrees. In about 65% of the roots showing a period of reverse curvature, the roots curved steadily past the vertical, reaching maximum curvature about 205 +/- 65 min after gravistimulation. The direction of curvature then reversed back toward the vertical. After one or two oscillations about the vertical the roots obtained a vertical orientation and the distribution of growth within the root tip became the same as that prior to gravistimulation. The period of transient backward curvature coincided with and was evidently caused by enhancement of growth along the concave and inhibition of growth along the convex side of the curve, a pattern opposite to that prevailing in the earlier stages of downward curvature. There were periods during the gravitropic response when the normally unimodal growth-rate distribution within the elongation zone became bimodal with two peaks of rapid elongation separated by a region of reduced elongation rate. This occurred at different times on the convex and concave sides of the graviresponding root. During the period of steady downward curvature the elongation zone along the convex side extended farther toward the tip than in the vertical control. During the period of reduced rate of curvature, the zone of elongation extended farther toward the tip along the concave side of the root. The data show that the gravitropic response pattern varies with time and involves changes in localized elongation rates as well as changes in the length and position of the elongation zone. Models of root gravitropic curvature based on simple unimodal inhibition of growth along the lower side cannot account for these complex growth patterns.  相似文献

We find a differential distribution of dictyosomes in the avascular tip cells of the oat (Avena sativa) coleoptile upon geostimulation. A differential activation (increased vesicle production) of the dictyosomes with respect to gravity also occurs, but only in the tip cells of the lower tissues. Similar differences in distribution and activation of dictyosomes occur also in cells subjacent to the avascular tip (1st and 5th millimeter from the apex) of both the upper and lower half-tissues. When only the outer epidermal cells below the apex are considered, the differential distribution and activation of dictyosomes occur only in the lower outer epidermis. The changes in distribution of dictyosomes begin at 6 minutes, or sooner, from the start of geostimulation, before the onset of geotropism. The kinetics of amyloplast sedimentation and Golgi movement do not appear to differ in the cells of the avascular tip. We suggest that the Golgi participates in, and possibly initiates, the differential elongation of cells of geotropically stimulated coleoptiles.  相似文献

Abstract. Ethylene production during geostimulation was examined in peduncles of dandelion ( Taraxacum officinale ). Significant amounts of ethylene were evolved from geostimulated peduncles compared with vertical controls only after geostimulation of 2 h or longer and the extra ethylene arose mainly from the lower portion of geostimulated peduncles. Reduction of the ethylene produced from geostimulated peduncles by pretreatment with ethylene-synthesis inhibitors did not affect or enhanced subsequent geobending. It is proposed that ethylene may control the later stages of geobending and influence the straightening process of autotropism.  相似文献

Experiments have been performed to investigate the nitrogen removal performance in a novel combined biofilm reactor using synthetic wastewater. In the reactor, one cubic box was separated by two baffles into three zones: aerobic zone, buffering zone and anoxic zone. Nitrification and denitrification were supposed to be mainly accomplished in the aerobic and anoxic zones, respectively. When the influent total nitrogen (TN) and organic carbon loadings were averaged at 0.093 and 0.40 kg/m3/d, 84% TN removal efficiency was achieved by adjusting the aeration rate and the configuration of the reactor. Continuous experimental results demonstrated that NH3-N removal efficiency increased by adjusting the clapboards of the reactor at a certain aeration rate. Energy produced by aeration was used for liquid recycle, so TN could be more efficiently removed at lower cost in this reactor.  相似文献

We examined cell length, mitosis, and root meristem “cuticle” in different tissues of geostimulated, red light-exposed primary roots of corn (Zea Mays, Wisconsin hybrid 64A × 22R). The examination was done at 15-minute intervals for a period of 240 minutes. Differences in cell elongation between the upper and lower sides were most prominent between 1.5 and 2.5 mm from the root meristem; the outer cortex had the greatest elongation growth, and the upper cells showed a significant increase in length compared to the lower. A differential mitosis was also found, with the lower tissue being greater. We infer that the mitotic activity is indicative of cell division, and this division occurs strictly in the first 1.5 mm of the root meristem. The combined effect of differential cell elongation and cell division results in the localization of the geotropic curvature in the 1.5- to 2.5-mm region from the root meristem. Mitosis that occurs primarily in the cortex and stele were asynchronous; the peak of cortical division preceded that of the stele. Both peaks occurred before the peak of geotropism. A densely stained layer separates the cap from the root meristem. This layer is thinner at the apex of the root meristem. The area of the thin region increased with time and peaked at 180 minutes after geostimulation, which was coincidental with the peak of the geotropic response.  相似文献

Fatty acid metabolism was studied in periportal and perivenous hepatocytes isolated by the method of Chen & Katz [Biochem. J. (1988) 255, 99-104]. The rate of fatty acid synthesis and the activity of acetyl-CoA carboxylase were markedly enhanced in perivenous hepatocytes as compared with periportal cells. However, the response of these two parameters to short-term modulation by cellular effectors such as the hormones insulin and glucagon, the phorbol ester 4 beta-phorbol 12 beta-myristate 13 alpha-acetate and the xenobiotics ethanol and acetaldehyde was similar in the two zones of the liver. In addition, perivenous hepatocytes showed a higher capacity of esterification of exogenous fatty acids into both cellular and very-low-density-lipoprotein lipids. Nevertheless, no difference between the two cell sub-populations seemed to exist in relation to the secretion of very-low-density lipoproteins. On the other hand, the rate of fatty acid oxidation was increased in periportal cells. This could be accounted for by a higher activity of carnitine palmitoyltransferase I and a lower sensitivity of this enzyme to inhibition by malonyl-CoA in the periportal zone. No differences were observed between periportal and perivenous hepatocytes in relation to the short-term response of fatty acid oxidation and carnitine palmitoyltransferase I activity to the cellular modulators mentioned above. In conclusion, our results show that: (i) lipogenesis is achieved at higher rates in the perivenous zone of the liver, whereas the fatty-acid-oxidative process occurs with a certain preference in the periportal area of this organ; (ii) the short-term response of the different fatty-acid-metabolizing pathways to cellular effectors is quantitatively similar in the two zones of the liver.  相似文献

We investigated the biological response of chondrocytes isolated from different zones of articular cartilage and their cellular behaviors in poly (ethylene glycol)-based (PEG) hydrogels containing exogenous type I collagen, hyaluronic acid (HA), or chondroitin sulfate (CS). The cellular morphology was strongly dependent on the extracellular matrix component of hydrogels. Additionally, the exogenous extracellular microenvironment affected matrix production and cartilage specific gene expression of chondrocytes from different zones. CS-based hydrogels showed the strongest response in terms of gene expression and matrix accumulation for both superficial and deep zone chondrocytes, but HA and type I collagen-based hydrogels demonstrated zonal-dependent cellular responses.  相似文献

Summary Bush regeneration programmes aim to restore native plant communities by interventions designed to assist natural recovery, supplemented by planting or direct seeding where necessary. The present study compared ground-dwelling ant communities and rates of seed removal by ants in weed-infested, regenerating and undamaged urban bushland in northern Sydney. Three replicate dry sclerophyll sites on sandstone were studied, each containing zones of weed-infested, regenerating and undamaged bushland. Canopy cover, midstorey cover, ground cover and litter depth were measured at each site. Each zone type showed distinctive vegetation and cover characteristics, particularly at the ground level. Ground foraging ants were sampled with pitfall traps, sorted to morphospecies and placed in functional groups. There were no significant differences in ant abundance, number of genera or number of morphospecies between site types. Ant diversity and equitability was highest in the undamaged zones, followed by regenerating zones, then weedy zones. There was a significant difference between zone types in the proportion of ant species in different functional groups. Weedy zones were dominated by Opportunist species whereas regenerating zones had a high proportion of Dominant Dolichoderinae. At undamaged zones, the distribution of ants across functional groups was more even. Differences in the ant communities between the three zone types were reflected by differences in interactions between ants and seeds. Seed removal rates of both native and exotic seeds were significantly lower at weedy zones than at undamaged zones, with the regenerating zones having intermediate rates. Sites subject to regeneration 4–10 years previously were intermediate between undamaged and weedy zones for ground cover, litter depth, ant species diversity and equitability, proportion of Opportunist ant species, and seed removal rates. Thus, the regeneration techniques used have been at least partially successful in restoring some aspects of the invertebrate fauna and ecological interactions typical of undamaged dry sclerophyll bushland.  相似文献

Paneth cells are one of the histologic components of intestinal metaplasia of the stomach, as are mucin-producing goblet cells. With the aid of an image quantifier, the distribution of Paneth cells histochemically labeled with acid fuchsin was analyzed for a gastrectomy specimen containing an adenocarcinoma of the intestinal type; the topographic distribution of goblet cells histochemically labeled with Alcian blue (pH 2.5) was also analyzed. The specimen was cut into 63 blocks (0.5 X 4.0 cm) in four zones; antrum (zone I), intermediate region (zone II) and fundus (zones III and IV). Paneth cells were found only in sections containing mucin-producing goblet cells. Paneth cells were found in 12.5% of the 16 sections from the antral zone I containing Alcian blue-positive goblet cells. The rates were 44.4% for the intermediate zone II and 55.5% for the distal fundic zone III. The total area occupied by Paneth cells was significantly lower in the gastric mucosa as compared to the duodenal mucosa. The "Paneth cell index" (total Paneth cell area/total goblet cell area) was highest in the duodenum, followed by the distal fundic zone III. This method of quantitating Paneth cell metaplasia of the stomach will be used to investigate the topographic distribution of those cells in populations with low and high incidences of intestinal metaplasia.  相似文献

This retrospective study was undertaken to investigate the soft-tissue response rate to the skeletal and soft-tissue alterations following a rhinoplasty. Ninety-eight patients, 80 females and 18 males, with a mean follow-up of 13 months, were included in this study. The tracings of the outline of preoperative cephaloxerograms and life-size photographs were superimposed on the postoperative ones, and the differences were measured and confirmed with measurements of intraoperative resected segments. The soft-tissue response to skeletal alterations was measured in seven different zones. Zone 1 (nasion) and zone 7 (nasal spine area) had the lowest mean response rate of approximately 25 percent. Zone 2 (proximal bridge) and zone 3 (midbridge) had a 60 percent response rate. Zone 4 (supratip area) had a 43 percent response, zone 5 had a 41 percent response, and zone 6 had a 40 percent response rate. There were statistically significant differences among the response rates of thick, medium, and thin noses. Age was an important factor in zones 1, 4, 5, 6, and 7. The patient's sex did not influence the soft-tissue response rate to skeletal alterations. The soft-tissue response in relation to the alar base narrowing was about 52 percent. This study reveals a predictable soft-tissue response to skeletal alterations on all zones except zone 7 (nasal spine area).  相似文献
