Leaves of Vitis vinifera L., nearly fully expanded, imported only trace amounts of 14C following assimilation of 14CO2 by a lower leaf on the same shoot, but benzyladenine (BA) application at 4.4 × 10−3m caused a marked increase in the movement of 14C into these leaves. Older leaves near the shoot base were less responsive; BA treatment alone had little effect on import of labeled assimilates from adjacent leaves but when the BA-treated leaves were darkened there was an increased import of labeled materials. When these 2 treatments were combined and applied to leaves on shoots with ringed bases, relatively high levels of radioactivity were detected in the BA-treated leaves but under these conditions darkening, without the application of BA, also resulted in an increased import of 14C. Accumulation of imported 14C was found to be restricted to the area of the leaf blade treated with BA. Separation of labeled compounds in ethanol extracts of treated leaves showed a lower percentage of radioactivity present in the sugar fraction from BA-treated leaves and an increased percentage present in the amino acid fraction.  相似文献

3,4-Dichloropropionanilide-14C (propanil) labeled in either the C-1 or C-3 carbon atoms of the propionic acid moiety was applied to the roots of pea (Pisum sativum L.) and rice (Oryza sativa L.) plants in nutrient solution (0.1 mm-0.28 mm). Radioactivity was detected throughout the treated plants, but the greatest labeling was found in the roots. None of the products that contained aniline were radioactive, suggesting that the plants split the propionic acid moiety from propanil. The fate of the propionate moiety of propanil was determined by recovery of 14CO2 from plants exposed to propanil-14C. The time-course of the 14CO2 production demonstrated that the intact propionic acid was cleaved from the propanil and subsequently catabolized by the β-oxidation catabolic sequence. The appearance of radioactivity in the shoots was attributed to the incorporation of products of propionate metabolism. Both the susceptible pea plants and the tolerant rice plants converted a high percentage of the administered propanil-14C to 14CO2.  相似文献

The content of 14C in the products of photosynthesis of the source leaf and xylem sap was investigated in kidney bean (Phaseolus vulgaris L.) plants during the stage of mass tillering. 14C partition was measured a day after two-minute photoassimilation of 14CO2 by an individual mature leaf located in the middle part of the shoot. The source-sink relations were disturbed by the excision of all mature leaves (except the source leaf) or all growing axillary shoots. The leaves or growing axillary shoots were excised 15 min after leaf feeding with 14C2. A day later, in plants with excised growing axillary shoots, the content of 14C in the source leaf was by 18% higher and in those with removed leaves by 15% lower than in control plants. The next day after the excision of growing axillary shoots, radioactivity of the xylem sap increased; after defoliation, both the volume of the xylem sap and its specific radioactivity decreased. In the xylem sap of defoliated plants, the proportion of 14C in malate decreased more than six times, whereas the proportion of 14C in amino acids somewhat increased (1.5 times). In two days, the volume of the xylem sap exuded by treated plants became the same as in control plants and its radioactivity decreased almost by an order of magnitude but essentially did not differ in the both types of treatment. White mustard (Sinapis alba L.) plants, susceptible to 2,4-D, and scentless mayweed (Tripleurospermum maritimum L.) plants, resistant to 2,4-D, markedly differ in the distribution and metabolism of 2,4-D. When 2,4-D-14C was applied onto the leaf, radioactivity was found in mustard after 8 days in the entire plant, whereas in scentless mayweed radioactivity occurred mainly in the leaf onto which it was applied and in another one or two leaves, with the roots showing only traces of radioactivity. The two plant species differed both in the character and in the number and amount of metabolites detected in aqueous and ether fractions. The metabolite with Rf 0.63 -0.65, soluble in ethylether (39% of the total radioactivity applied), and that with Rf 0.35 -0.36 (23% of the total radioactivity) were present exclusively in the roots of scentless mayweed plants. The Rf values established indicate that the former metabolite may be a conjugate with the amino acids alanine and valine and the latter a hydroxylated derivate of 2,4-D. In the shoots of both plants, 2,4-D-14C was metabolized to identical metabolites with Rf 0.59 -0.61, which occurred in mustard plants only in trace amounts. Free 2,4-D-14C occurred in the shoots of both plants and in considerable amounts also in mustard roots; it could not be demonstrated in the roots of scentless mayweed. The two species did not differ in the uptake rate or in the amount of absorbed 2,4-D-14C.  相似文献

The effect of exogenous growth regulators on movement of assimilates into flowers and young fruits of `Black Corinth' grapes was studied. Clusters were treated with growth regulator and after 0.5 hr to 5 days the leaves above the clusters were exposed to 14CO2. Control shoots received 14CO2 but no growth regulator. At harvest, counting and radioautographic techniques were used to ascertain amount and distribution of activity in clusters. Clusters were dipped in 4-CPA (4-chlorophenoxyacetic acid), GA3 (gibberellic acid), or BA (benzyladenine). All berries were heavier than controls within 3 days. Total counts in the fruits were increased by 4-CPA, and the distribution of radioactivity among the sugar, organic acid, and amino acid fractions was usually altered by all treatments. In a time series experiment, within 6 hr after treatment of fruits with GA3 there was almost an 8-fold increase in total counts relative to the control. After 12 hr there was about a 9-fold and 6-fold increase in counts in tartaric and malic acids, respectively, and in γ-aminobutyric acid, pipecolic acid, and valine increases of 56, 150, and 330%. Radioactivity in fructose was increased 70% in gibberellin-treated clusters over the controls. After 96 hr there were only about 1000 cmp per g fr wt in controls, but there were about 31,000 cpm counts in treated clusters. Treatment of clusters with gibberellin attracted less assimilates into the fruits when shoots had also been sprayed with gibberellin. Dipping portions of clusters in gibberellin increased the movement of 14C assimilates into the treated portions. Hormonal control of mobilization is discussed.  相似文献

Sucrose-14C was fed to the leaf blades of squash plants (Cucurbita pepo L. var. melopepo torticallis Bailey) for 30 or 60 minutes. Petioles of treated plants were cut into sections and extracted. The majority of the 14C within the petiole was in sucrose rather than stachyose, the sugar that is transported by the squash plants when 14CO2 is supplied. This indicates that the phloem loading system of squash plants is not the system that specifies which sugar is transported.  相似文献

Like other halophytic chenopods, sugar beet (Beta vulgaris L.) can accumulate high betaine levels in shoots and roots. N,N,N-trimethylglycine impedes sucrose crystallization and so lowers beet quality. The objective of this research was to examine the genetic variability and physiological significance of betaine accumulation in sugar beet and its relatives. Three cultivated genotypes of B. vulgaris and two genotypes of the wild progenitor B. maritima L. were grown with and without gradual salinization (final NaCl concentration = 150 millimolar). At 6 weeks old, all five genotypes had moderately high betaine levels in shoots and roots when unsalinized (averages for all genotypes: shoots = 108 micromoles per gram dry weight; roots = 99 micromoles per gram dry weight). Salinization raised betaine levels of shoots and roots 2- to 3-fold, but did not greatly depress shoot or root growth. The genotype WB-167—an annual B. maritima type—always had approximately 40% lower betaine levels in roots than the other four genotypes, although the betaine levels in the shoots were not atypically low.

The site and pathway of betaine synthesis were investigated in young, salinized sugar beet plants by: (a) supplying 1 micromole [14C]ethanolamine to young leaf blades or to the taproot sink of intact plants; (b) supplying tracer [14C]formate to discs of leaf, hypocotyl, and taproot tissues in darkness. Conversion of both 14C precursors to betaine was active only in leaf tissue. Very little 14C appeared in the phospholipid phosphatidylcholine before betaine was heavily labeled; this was in marked contrast to the labeling patterns in salinized barley. Phosphorylcholine was a prominent early 14C metabolite of both [14C]ethanolamine and [14C]formate in all tissues of sugar beet. Betaine translocation was examined in young plants of sugar beet and WB-167 by applying tracer [methyl-14C]betaine to a young expanded leaf and determining the distribution of 14C after 3 days. In all cases, extensive 14C translocation to young leaves and taproot sink occurred; neither in the fed leaf nor in sink organs were any 14C metabolites of betaine detected.

Nodulated and denodulated roots of adzuki bean (Vigna angularis), soybean (Glycine max), and alfalfa (Medicago sativa) were exposed to 14CO2 to investigate the contribution of nodule CO2 fixation to assimilation and transport of fixed nitrogen. The distribution of radioactivity in xylem sap and partitioning of carbon fixed by nodules to the whole plant were measured. Radioactivity in the xylem sap of nodulated soybean and adzuki bean was located primarily (70 to 87%) in the acid fraction while the basic (amino acid) fraction contained 10 to 22%. In contrast, radioactivity in the xylem sap of nodulated alfalfa was primarily in amino acids with about 20% in organic acids. Total ureide concentration was 8.1, 4.7, and 0.0 micromoles per milliliter xylem sap for soybean, adzuki bean, and alfalfa, respectively. While the major nitrogen transport products in soybeans and adzuki beans are ureides, this class of metabolites contained less than 20% of the total radioactivity. When nodules of plants were removed, radioactivity in xylem sap decreased by 90% or more. Pulse-chase experiments indicated that CO2 fixed by nodules was rapidly transported to shoots and incorporated into acid stable constituents. The data are consistent with a role for nodule CO2 fixation providing carbon for the assimilation and transport of fixed nitrogen in amide-based legumes. In contrast, CO2 fixation by nodules of ureide transporting legumes appears to contribute little to assimilation and transport of fixed nitrogen.  相似文献

The release of apical dominance by the physical destruction in situ of the apical meristem and associated leaf primordia (decapitation) promoted the growth of tillers in non-herbicide-treated wild oat plants, as indicated by increased tiller lengths and fresh weights. At 96 h after [14C] herbicide treatment following decapitation, the absorption of [14C]imazamethabenz and total translocation of radioactivity were respectively increased by 28% and 49%. By 96 h after [14C]imazamethabenz application, the radioactivity detected in the roots of decapitated plants was 45% higher than that in the roots of nondecapitated plants while the radioactivity in tillers of decapitated plants was 2.6-fold that in tillers of intact plants. Decapitation together with foliar spraying of imazamethabenz at 200 g ha–1 further reduced tiller fresh weight, greatly decreased the total tiller number, and thereafter significantly increased overall phytotoxicity by 32% as measured by total shoot fresh weight. The results of this study support the hypothesis that main shoot apical dominance limits translocation of applied imazamethabenz to lateral shoots, rendering tillers less susceptible to growth inhibition by the herbicide.  相似文献

The influence of stage of development (preflowering versus flowering) in nodulated and nonnodulated soybeans (Glycine max [L.] Merr. cv. Wells) on partitioning of 14C into assimilates following exposure of a soybean leaf to 14CO2 by both steady-state and pulse-labeling techniques was studied. Blades on the second fully expanded leaf from the stem apex were exposed to 14CO2. Radioactive assimilates were extracted from source leaf blades, petioles, and stems (both the path up and path down from source leaf), were separated into neutral (sugars), basic (amino acids), and acidic (organic acids, sugar phosphates) fractions by ion exchange chromatography. The basic fraction was further resolved using thin layer chromatography and the percentage of radioactivity recovered in each amino acid was determined.  相似文献

The effects of kinetin on starch and sugar levels and on 14CO2 and 32P-orthophosphate labeling patterns of floated Chinese cabbage (Brassica pekinensis) leaf discs were investigated. Kinetin caused gross starch degradation. Neutral sugars were depressed by 30 to 40% in leaf tissue treated with kinetin for 24 hours. 14CO2 labeling of leaf discs pretreated with kinetin for 24 hours showed increased radioactivity in chloroform-soluble material and most sugar phosphates, and a 35 to 40% decrease in radioactivity in the neutral sugars, glucose, sucrose, and fructose. Incorporation into ATP was increased by 40% by kinetin. 32P-Orthophosphate uptake was inhibited 30% by kinetin. When corrected for uptake, kinetin stimulated incorporation into chloroform-soluble material but had little effect on other cell fractions. These results indicate that kinetin mobilizes starch reserves and increases the flow of sugars required for the synthesis of lipids and structural materials in floated discs.  相似文献

The proportion of the total plant radioactivity present in shoots at the end of a 24-hour exposure of the roots to 0.5 milligram per liter 14C-3-amino-2,5-dichlorobenzoic acid (14C-amiben) ranged from 1.4 to 74.3% in 13 species. When roots of 10-day-old wheat (Triticum aestivum L. em. Thell., Triumph) and 13-day-old barnyard grass (Echinochloa crusgalli L. Beauv.) plants were treated with 0.5 milligram per liter 14C-amiben for 12 or 24 hours, barnyard grass shoots contained at least eight times more of the total plant radioactivity than did wheat shoots. In similar experiments with 14C-2-chloro-4-(ethylamino)-6-(isopropylamine)-s-triazine (14C-atrazine), there were no differences in translocation between these two species.  相似文献

The effect of gibberellic acid (GA3) on sucrose export from source leaves was studied in broad bean (Vicia faba L.) plants trimmed of all but one source and one sink leaf. GA3 (10 micromolar) applied to the source leaf, enhanced export of [14C]sucrose (generated by 14CO2 fixation) to the root and to the sink leaf. Enhanced export was observed with GA treatments as short as 35 minutes. When GA3 was applied 24 hours prior to the 14CO2 pulse, the enhancement of sucrose transport toward the root was abolished but transport toward the upper sink leaf was unchanged. The enhanced sucrose export was not due to increased photosynthetic rate or to changes in the starch/sucrose ratio within the source leaf; rather, GA3 increased the proportion of sucrose exported. After a 10-min exposure to [14C]GA3, radioactivity was found only in the source leaf. Following a 2 hour exposure to [14C]GA3, radioactivity was distributed along the entire stem and was present in both the roots and sink leaf. Extraction and partitioning of GA metabolites by thin layer chromatography indicated that there was a decline in [14C]GA3 in the lower stem and root, but not in the upper stem. This pattern of metabolism is consistent with the disappearance of the GA3 effect in the lower stem with time after treatment. We conclude that in the short term, GA3 enhances assimilate export from source leaves by increasing phloem loading. In the long term (24 hours), the effect of GA3 is outside the source leaf. GA3 accumulates in the apical region resulting in enhanced growth and thus greater sink strength. Conversely, GA3 is rapidly metabolized in the lower stem thus attenuating any GA effect.  相似文献

Summary Young trees of Larix decidua, in their 4th and 5th year of development, were permitted to photoassimilate a pulse of 14CO2 at different times throughout the growing season. After chase periods between 1 h and 7 days, the distribution of 14C in these plants was determined. CO2 fixation followed a maximum curve with highest rates of photosynthesis of 123 ± 4 mol CO2·h-1·mg chl-1 in June. Translocation of 14C assimilate was observed throughout the growing season. The main quantity of fixed 14C was always retained in the fed leaves. Radiocarbon moved basipetally into the roots at all times, particularly in spring and late summer. Sprouting young shoots and leaves at the stem apex attracted assimilate in spring. Incorporation of 14C into soluble low-molecular-weight substances prevailed; less radioactivity was incorporated into insoluble polymeric compounds. Distribution of 14C among the sugar, amino acid and organic acid fraction was determined. Labelled free sugars were analysed.  相似文献

Transport and metabolism of [2,3-14C] 1-aminocyclopropane-1-carboxylic acid (ACC) from roots to shoots in 4-day-old sunflower (Helianthus annuus L.) seedlings were studied. [14C]ACC was detected in, and 14C2H4 was evolved from, shoots 0.5 hours after [14C]ACC was supplied to roots. Ethylene emanation from the shoots returned to normal levels after 6 hours. The roots showed a similar pattern, although at 24 hours ethylene emanation was still slightly higher than in those plants that did not receive ACC. [14C]N-malonyl-ACC (MACC) was detected in both tissues at all times sampled. [14C]MACC levels surpassed [14C]ACC levels in the shoot at 2 hours, whereas [14C]MACC levels in the root remained below [14C]ACC levels until 6 hours, after which they were higher. Thin-layer chromatography analysis identified [14C] ACC in 1-hour shoot extracts, and [14C]MACC was identified in root tissues at 1 and 12 hours after treatment. [14C]ACC and [14C] MACC in the xylem sap of treated seedlings were identified by thin-layer chromatography. Xylem transport of [14C]ACC in treated seedlings, and transport of ACC in untreated seedlings, was confirmed by gas chromatography-mass spectrometry. Some evidence for the presence of [14C]MACC in xylem sap in [14C]ACC-treated seedlings is presented. A substantial amount of radioactivity in both ACC and MACC fractions was detected leaking from the roots over 24 hours. A second radiolabeled volatile compound was trapped in a CO2-trapping solution but not in mercuric perchlorate. Levels of this compound were highest after the peak of ACC levels and before peak MACC levels in both tissues, suggesting that an alternate pathway of ACC metabolism was operating in this system.  相似文献

The vasculature of the dorsal suture of cowpea (Vigna unguiculata [L.] Walp) fruits bled a sugar-rich exudate when punctured with a fine needle previously cooled in liquid N2. Bleeding continued for many days at rates equivalent to 10% of the estimated current sugar intake of the fruit. A phloem origin for the exudate was suggested from its high levels (0.4-0.8 millimoles per milliliter) of sugar (98% of this as sucrose) and its high K+ content and high ratio of Mg2+ to Ca2+. Fruit cryopuncture sap became labeled with 14C following feeding of [14C]urea to leaves or adjacent walls of the fruit, of 14CO2 to the pod gas space, and of [14C] asparagine or [14C]allantoin to leaflets or cut shoots through the xylem. Rates of translocation of 14C-assimilates from a fed leaf to the puncture site on a subtended fruit were 21 to 38 centimeters per hour. Analysis of 14C distribution in phloem sap suggested that [14C]allantoin was metabolized to a greater extent in its passage to the fruit than was [14C] asparagine. Amino acid:ureide:nitrate ratios (nitrogen weight basis) of NO3-fed, non-nodulated plants were 20:2:78 in root bleeding xylem sap versus 90:10:0.1 for fruit phloem sap, suggesting that the shoot utilized NO3-nitrogen to synthesize amino acids prior to phloem transfer of nitrogen to the fruit. Feeding of 15NO3 to roots substantiated this conclusion. The amino acid:ureide ratio (nitrogen weight basis) of root xylem sap of symbiotic plants was 23:77 versus 89:11 for corresponding fruit phloem sap indicating intense metabolic transfer of ureide-nitrogen to amino acids by vegetative parts of the plant.  相似文献

With Chlorella ellipsoidea cells, the effect of oxygen was investigated on the products of enhanced dark 14CO2 fixation immediately following preillumination in the absence of CO2. When the reaction mixture was made aerobic by bubbling air (CO2-free) throughout preillumination and the following dark 14CO2 fixation periods, the initial fixation product was mainly 3-phosphoglyceric acid. When nitrogen gas had been used instead of air, only about one-half of the total radioactivity in the initial fixation products was in 3-phosphoglyceric acid and the rest in aspartic, phosphoenolpyruvic, and malic acids. The percentage distribution of radioactivity incorporated in these initial products rapidly decreased during the rest of the dark period. Concurrent with the decrease in the initial 14CO2 fixation products, some increase was observed in the radioactivities of the sugar phosphates. The maximal radioactivity incorporated in sugar mono- and diphosphates accounted for only 10% of total 14C, under either the aerobic or anaerobic conditions. Under anaerobic conditions most of the 14C incorporated was eventually transferred to alanine, whereas the main end products under aerobic conditions were aspartate and glutamate. The pattern of 14CO2 fixation products was unaffected by the atmospheric condition during the period of preillumination. The preferential flow of the fixed carbon atom to alanine or aspartate depended on the presence or absence of oxygen during the period of dark CO2 fixation.  相似文献

Transfer of the nitrogen and carbon of allantoin to amino acids and protein of leaflets, stems and petioles, apices, peduncles, pods, and seeds of detached shoots of nodulated cowpea (Vigna unguiculata L. Walp. cv. Caloona) plants was demonstrated following supply of [2-14C], [1,3-15N]allantoin in the transpiration stream. Throughout vegetative and reproductive growth all plant organs showed significant ureolytic activity and readily metabolized [2-14C]allantoin to 14CO2. A metabolic pathway for ureide nitrogen utilization via allantoic acid, urea, and ammonia was indicated. Levels of ureolytic activity in extracts from leaves and roots of nodulated cowpea were consistently maintained at higher levels than in non-nodulated, NO3 grown plants.

[14C]Ureides were recovered in extracts of aphids (Aphis craccivora and Macrosiphum euphorbieae) feeding at different sites on cowpea plants supplied with [2-14C]allantoin through the transpiration stream or to the upper surface of single leaflets. The data indicated that the ureides were effectively transferred from xylem or leaf mesophyll to phloem, and then translocated in phloem to fruits, apices, and roots.

Summary Two weeks after anthesis, when the grain is filling rapidly, the rate of photosynthesis by flag leaves of wheat cv. Gabo was between 20 and 30 mg CO2 dm-2 leaf surface hour-1 under the conditions used. About 45% of flag-leaf assimilates were translocated to the ear, and only about 12% to the roots and young shoots.On removing the ear, net photosynthesis by the flag leaves was reduced by about 50% within 3–15 hours, and there was a marked reduction in the outflow of 14C-labelled assimilates from the flag leaves.Subsequent darkening of all other leaves on plants without ears led to recovery of flag-leaf photosynthesis, as measured by gas analysis and 14CO2 fixation, and to increased translocation of assimilates to the roots and young shoots. Minor changes in the rates of dark respiration accompanied these major, reversible changes in photosynthetic rate.After more than a week in continuous, high-intensity light, the rate of photosynthesis by flag leaves of intact plants had fallen considerably, but could be restored again by a period in darkness, or by inhibiting photosynthesis in the ears by spraying them with DCMU. The inhibition of ear photosynthesis increased translocation of labelled assimilates from the flag leaf to the ears, without affecting leaf sugar levels.The application of TIBA to the culm below the ear inhibited auxin movement throught the culm, but had no influence on flag-leaf photosynthesis.These results suggest that, at least in this system, photosynthesis by the flag leaf is regulated directly by the demand for assimilates from the flag leaf and not indirectly through action in the leaf of auxins produced by the sink organs.  相似文献

After a 5-second exposure of illuminated bermudagrass (Cynodon dactylon L. var. `Coastal') leaves to 14CO2, 84% of the incorporated 14C was recovered as aspartate and malate. After transfer from 14CO2-air to 12CO2-air under continuous illumination, total radioactivity decreased in aspartate, increased in 3-phosphoglyceric acid and alanine, and remained relatively constant in malate. Carbon atom 1 of alanine was labeled predominantly, which was interpreted to indicate that alanine was derived from 3-phosphoglyceric acid. The activity of phosphoenolpyruvate carboxylase, alkaline pyrophosphatase, adenylate kinase, pyruvate-phosphate dikinase, and malic enzyme in bermudagrass leaf extracts was distinctly higher than those in fescue (Festuca arundinacea Schreb.), a reductive pentose phosphate cycle plant. Assays of malic enzyme activity indicated that the decarboxylation of malate was favored. Both malic enzyme and NADP+-specific malic dehydrogenase activity were low in bermudagrass compared to sugarcane (Saccharum officinarum L.). The activities of NAD+-specific malic dehydrogenase and acidic pyrophosphatase in leaf extracts were similar among the plant species examined, irrespective of the predominant cycle of photosynthesis. Ribulose-1, 5-diphosphate carboxylase in C4-dicarboxylic acid cycle plant leaf extracts was about 60%, on a chlorophyll basis, of that in reductive pentose phosphate cycle plants.  相似文献
