Experiments were performed in vivo and in vitro to determine the effects of enalaprilat, a specific inhibitor of angiotensin-converting enzyme, on various aspects of the decidual cell reaction in rats. Ovariectomized, adult female rats were sensitized for the decidual cell reaction with steroid treatments. For in vivo experiments, intrauterine infusions of enalaprilat alone, and in combination with angiotensin II and prostaglandin E2 (PGE2), were initiated on the day of uterine sensitivity. Enalaprilat inhibited the increases in uterine PG concentrations, endometrial vascular permeability, alkaline phosphatase activity and uterine weight that occurred sequentially following infusion of vehicle. Concurrent infusion of angiotensin II did not reverse any of these inhibitory effects; PGE2 infusion partially, but not completely, reversed the inhibition of increase in uterine weight, although it did not alter the inhibition of endometrial vascular permeability. For in vitro experiments, endometrial stromal cells were obtained from uteri on the day of sensitivity and cultured for up to 3 days in the presence of enalaprilat and angiotensin II. Enalaprilat inhibited in a dose-dependent manner the increases in stromal cell alkaline phosphatase activity and media PGE concentration that occurred in the control cultures; these effects were fully reversed by concurrent treatment with angiotensin II. The inhibition of stromal alkaline phosphatase activity was also reversed by PGE2; conversely, the ability of angiotensin II to reverse the effect of enalaprilat was lost in the presence of indomethacin. These studies provide evidence of a requirement for angiotensin II during the decidual cell reaction in rats and suggest that it acts, at least in part, through a PG-dependent mechanism.  相似文献

Avidin is used in conjunction with polyethylene glycol to coprecipitate biotin-labeled carcinoembryonic antigen (CEA) antibody and the CEA-antibody complex together with biotin-labeled normal serum proteins. Among 15 strains of methanogens, one plasmid, pMP1, was identified in the new coccoid isolate PL-12/M. It could not be detected in the cleared lysate, but it was detected in the viscous pellet. The plasmid had a molecular weight of ca. 4.6 x 10(6). A restriction enzyme cleavage map of the cloned plasmid was derived.  相似文献

We present a model for diffusion in a molecularly crowded environment. The model consists of random barriers in a percolation network. Random walks in the presence of slowly moving barriers show normal diffusion for long times but anomalous diffusion at intermediate times. The effective exponents for square distance vs time usually are below one at these intermediate times, but they can also be larger than one for high barrier concentrations. Thus, we observe sub- and superdiffusion in a crowded environment.  相似文献

I am so very honored to receive the Women in Cell Biology Sandra K. Masur Senior Leadership Award from the American Society for Cell Biology (ASCB), particularly because many of the previous awardees have served as mentors and sources of inspiration throughout my own career. I also thank the ASCB for always striving to be maximally inclusive, in terms of both the scientists it supports and its broad vision of what constitutes cell biology. As a graduate student I gave one of my first talks at an ASCB meeting, and I am proud to have been an ASCB member for almost 30 years. In this essay, I describe my own career to illustrate the support that I believe is needed to achieve a career in science.

Obesity has been associated with an increased risk of osteoarthritis (OA). However, the mechanism by which obesity contributes to OA remains uncertain. Adiponectin, an adipocyte-derived hormone, has shown anti-diabetic and anti-atherogenic properties. In the present study, we aimed to investigate the potential role of adiponectin in OA disease. We demonstrated that adiponectin was present in OA synovial fluid (SF) and its expression level was almost 100-fold decrease compared with that in OA plasma. FPLC and ELISA studies revealed the distribution and abundance of the adiponectin complexes in plasma and SF from patients with OA. The percentage of high molecular weight (HMW) per total adiponectin in OA SF was lower than in OA plasma, while that of the hexamer form was similar and the trimer form was higher. The expression levels of adiponectin receptors AdipoR1 and AdipoR2 were examined in human OA tissues by RT-PCR. AdipoR1 was abundantly expressed in cartilage, bone and synovial tissues, whereas AdipoR2 was rarely detected. Finally, the effects of adiponectin on primary chondrocyte functions were studied by using antibody-based protein array and RT-PCR. The patterns of mRNA expression and protein production strongly indicate that adiponectin is involved in the modulation of cartilage destruction in chondrocytes by up-regulating TIMP-2 and down-regulating IL-1beta-induced MMP-13. Together these findings clearly indicate that the adiponectin may act as a protective role in the progression of OA, and this also provide new thinking on the relationship between obesity and OA.  相似文献

Summary Indirect gradient analysis methods require an explicit vegetation model which must be based on direct gradient analysis studies. Various vegetation models are reviewed. Field evidence for the models is discussed. Experimental studies of species response to environmental gradients are reviewed and discussed. Three types of gradient are recognized as important for development of models: indirect environmental gradients where the environmental factor has no direct physiological influence on plant growth e.g. elevation; direct environmental gradients where the factor has a direct physiological effect on growth but is not an essential resource, e.g. pH; resource gradients where the factor is an essential resource for plant growth. The behaviour of the ecological carrying capacity and the role of competition along such gradients are shown to be important for developing vegetation models.  相似文献

Schooling as a strategy for taxis in a noisy environment   总被引:8,自引:0,他引:8  
Many aquatic animals face a fundamental problem during foraging and migratory movements: while their resources commonly vary at large spatial scales, they can only sample and assess their environment at relatively small, local spatial scales. Thus, they are unable to choose movement directions by directly sampling distant parts of their environment. A common strategy to overcome this problem is taxis, a behaviour in which an animal performs a biased random walk by changing direction more rapidly when local conditions are getting worse. Such an animal spends more time moving in right directions than wrong ones, and eventually gets to a favourable area. Taxis is inefficient, however, when environmental gradients are weak or overlain by noisy small-scale fluctuations. In this paper, I show that schooling behaviour can improve the ability of animals performing taxis to climb gradients, even under conditions when asocial taxis would be ineffective. Schooling is a social behaviour incorporating tendencies to remain close to and align with fellow members of a group. It enhances taxis because the alignment tendency produces tight angular distributions within groups, and dampens the stochastic effects of individual sampling errors. As a result, more school members orient up-gradient than in the comparable asocial case. However, overly strong schooling behaviour makes the school slow in responding to changing gradient directions. This trade-off suggests an optimal level of schooling behaviour for given spatio-temporal scales of environmental variations. Social taxis may enhance the selective value of schooling in pelagic grazers such as herrings, anchovies and Antarctic krill. Furthermore, the degree of aggregation in a population of schooling animals may affect directly the rate and direction of migration and foraging movements.  相似文献

In this paper, we develop a conceptual model for autocatalytic reactions in which the autocatalyst undergoes a mutation process in a completely stirred tank reactor. Three cases with different mutation coefficient, !, and mutation efficiency, #, have been considered. The analysis of these cases shows the importance of the two parameters and the complexity of the resulting bifurcation diagrams. Co-existence of a mushroom and an isola have been determined for the first case. The isola portion is a low main product yield solution and is to be avoided. A sub-region of 7 co-existing steady states, with three stable static branches, has been determined for the second case. Chaos developing through period doubling and type 1 intermittency has been shown to occur for a fourth case.  相似文献

We study the optimal conservation effort for a population in a fluctuating environment. The survivorship of a population is affected by unpredictable environmental fluctuation (noise) and can be improved by conservation effort accompanied by a cost. The optimal effort level is the one that minimizes the total cost, defined as the weighted sum of the population extinction risk and the economic cost of conservation effort. The optimal effort depends on the variance and the probability distribution of the noise, the relative importance of the population's survival vs. the economic cost, the effectiveness of conservation effort, and the time scope over which we optimize. The analysis of dynamic programming illustrates that the choice of extinction risk function greatly affects the optimal effort level. The conservation effort level that is the best solution of a multiple-year optimization may be higher than that for the corresponding single-year optimization, if the population is relatively safe. However, the conservation level for the multiple-year optimization becomes lower than for the single-year optimization if the population is endangered. In a similar manner, the optimal conservation effort level for the problem with a short time scope is either higher or lower than that for the problem with a long time scope, depending on the extinction risk of the population. Next, for each parameter of the model, we define five different sensitivities of extinction probability or of the total cost. We then study the mean increase in the total cost caused by the uncertainty of parameters. To achieve the best conservation result, we need to invest the limited research effort to the parameter with the largest effect to the optimal effort level, rather than to those with large impacts on the extinction probability or on the total cost. The recommended policy should depend critically on the choice of the criterion to optimize, which shows the importance of theoretical study of the relationship in performing proper decision making in conservation practice.  相似文献

As demonstrated in our earlier studies, the differentiationof apogamous sporophytes on the secondary protonema of the mossPhyscomitrium pyriforme Brid. requires an exogenous supply ofsucrose in the medium. In the present work, similar differentiationwas observed in the leaf cells of aged gametophytes. The experimentsindicate an accumulation in the leaves of a sporophytic factorwhich initiates a de novo differentiation of sporophytes fromleaf cells without the intervention of sexual reproduction.In the absence of sucrose, the factor for apogamy was not present.Highlight intensity (5000–6000 lx) also inhibited itsproduction. There was no evidence that its presence interferedwith or inhibited production of gameto-phores. Growth regulatorssuch as IAA and kinetin altered only the effectiveness of thissporophytic factor, demonstrating that it was endogenous. Sporogenesisin the apogamous sporophytes took place without orthodox meiosis. Results obtained by using different exogenous environments forthe in vitro differentiation of callus into gametophytes orsporophytes are also reported. These support our contentionthat there is an accumulation of a sporophytic factor in thegametophytic callus cells, which is diluted during the processof differentiation. The morpho-regulatory influence of lightin the differentiation of apical cells with three cutting facesfrom unorganized callus is also considered.  相似文献

The stable isotope ratio and seasonal changes in diet of Alluaud's haplo Astatoreochromis alluaudi, a cichlid fish with massive pharyngeal jaws well known for its ability to process hard‐bodied prey, are described. The diet of A. alluaudi was quantified in Lake Saka, Uganda, over a period of 30 months. Variation in physico‐chemical variables (mean monthly rainfall, water temperature, turbidity and dissolved oxygen), as well as potential competitor density and food abundance, was measured throughout the second half of the study (14 months). Stomach contents and isotope analysis revealed a diet comprised mainly of fishes and insects, with a low contribution of molluscs (0–33%) in any given month. No correlation was detected between diet and either macroinvertebrate abundance or competitor abundance. The running average rainfall was positively related to the percentage of fish consumed per month. Although A. alluaudi exhibits an apparent molluscivorous trophic morphology in Lake Saka, molluscs did not appear to compose a major portion of its diet. Gradients of rainfall seemed to be the most important environmental predictor of diet choice in Lake Saka. These results are discussed with reference to Liem's Paradox that apparently morphologically specialized fishes often function as generalist feeders in the wild.  相似文献

The control and management of non-indigenous plant species (NIS) can be conceptually divided into three phases: inventory/survey, monitoring and management. Here we focus on phase one, determining which species are present and where they are located within the environment. Sampling for NIS is inherently time-consuming and thus costly. Many management areas are large and therefore can only be surveyed (partial observation of the total area by sampling) and not inventoried (total observation of area). Survey data should reflect the spatial distribution of the target species populations over the landscape. Such data can then be used in combination with environmental data, to create probability maps of target species occurrence for the entire area of interest. We used a GIS model to evaluate seven different survey methods for consistency and reliability of intersecting NIS species’ patches and producing samples which reflect the spatial distribution of the population, and which can be performed in a cost and time-efficient manner. The GIS model was developed to create NIS populations which were then sampled using the different survey methods, and the results recorded. To improve the applicability of the model, four patch sizes and levels of occurrence were used, along with random and weighted distribution patterns in relation to patch proximity to roads and trails. Grid and random points, and targeted (stratified continuous) transects (starting on a road or trail (rights of way (RoW)) and finishing 2 km from any RoW) methods provided the most consistent samples of the population. Logistically, point methods required an unrealistic distance and time commitment in comparison with transect methods. The importance of collecting information on the size of NIS patches was demonstrated as more small patches were intersected than larger ones when the area infested was held constant. Thus, if frequency of patches is used to explain the results of a survey then comparisons between species and methods are difficult to interpret thus leading to erroneous conclusions. However, use of percentage of area infested estimates provides for easier comparison between species and sample methods. The targeted transect method provided the most reliable, efficient and consistent sample with the expected spatial distribution.  相似文献
