The efficiency of turnip mosaic virus (TuMV) acqusition fromSinapis alba L. without sinigrin andBrassica nigra (L.) Koch with a high content of sinigrin by non-fastedMyzus persicae (Sulz.) andBrevicoryne brassicae (L.) was investigated in order to determine the effect of the phagostimulatory glucoside sinigrin on virus transmission. There was a significantly higher acquisition of TuMV fromS. alba byM. persicae than fromB. nigra. Conversely,B. brassicae acquired the virus more efficiently fromB. nigra than fromS. alba. The efficiency of transmission was positively correlated with the response of both aphid species to sinigrin. Saturation ofS. alba leaves with a 1 % solution of sinigrin did not significantly affect the acquisition of TuMV, in neitherM. persicae norB. brassicae, whether the aphids were fasted or not.  相似文献

Abstract

The study investigates the presence and quantity of antimicrobial sinigrin glucosinolates in tomato leaves after spraying them with moringa (Moringa oleifera) leaf extract (MLAE). Moringa concentrates (0.5, 0.75, 1.00 and 1.5?kg?L?1 (w v?1)) were prepared. Distilled water was the control. Sampled tomato leaves were air-dried, freeze-dried and extracted firstly using pure methanol in a hot water bath and then pellet re-extracted using 5?mL of hot aqueous methanol (70% v v?1). An ion exchange column, and sulphatase was used to achieve glucosiodesulphonation. High performance liquid chromatography (HPLC) was employed in the identification and quantitative analysis of the sinigrin glucosinolates. Tomato (Solanum lycopersicum) leaves treated with MLAE revealed highly significant (p?<?.001) content of sinigrin glucosinolates. The sinigrin standard and the desulphated sinigrin glucosinolates had a 7?s retention time difference; 5?kg?L?1 (w v?1) resulted in a superior amount of sinigrin in tomato leaves as compared to all the other MLAE concentrations. The study reveals that spraying MLAE on putatively diseased tomato leaves donates specific quantifiable glucosinolates like sinigrin, which may be involved in defense against tomato diseases and, hence, recommends use of 5?kg?L?1 (w v?1) for the highest sinigrin defense tag.  相似文献

Garlic mustard’s (Alliaria petiolata, Brassicaceae) invasive success is attributed in part to its release of allyl isothiocyanate (AITC) into the soil. AITC can disrupt beneficial arbuscular mycorrhizal fungi (AMF) associated with native plant roots, which limits their soil resource uptake. However, AITC and its precursor, sinigrin, have never been detected in garlic mustard-invaded forest soils. Here, we use high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC–MS) to assess the concentration and bioactivity of these putative allelochemicals in paired forest plots uninvaded or invaded by garlic mustard. Our methods detected AITC and sinigrin only where garlic mustard was present and our recovery of AITC/sinigrin coincided with adult senescence. A bioassay of in situ fungal hyphae abundance revealed significantly reduced hyphal abundance in the presence of garlic mustard relative to uninvaded soils. Finally, the lowest concentration of AITC measured in the field (~0.001 mM) is highly inhibitory to the spore germination of a forest AMF species, Glomus clarum. Together, our data provide the first direct evidence of garlic mustard-produced sinigrin and AITC in forest soils and demonstrate that even low levels of these chemicals have the potential to significantly suppress AMF growth and spore germination, strengthening their status as allelopathic novel weapons.  相似文献

Liver cancer is one of the leading causes of cancer death worldwide. A very high incidence of new liver cancer cases is diagnosed every year, and metastasis has been found to correlate to poor prognoses in humans. Better treatments for liver cancer are thus clearly needed. Sinigrin is one of the major ingredients present in Brassica nigra, which has been used in combination with other herbs for treatment of various diseases. The anti-proliferative activities of sinigrin were studied in a model of carcinogen-induced hepatotoxicity in rats. Rats were orally administered with sinigrin on a daily basis for three months before sacrifice. Sinigrin was found to significantly inhibit the proliferation of liver tumor cells; the number of surface tumors in the rat liver was dramatically reduced. Sinigrin induced apoptosis of liver cancer cells through up-regulation of p53 and down-regulation of Bcl-2 family members and caspases. Our findings indicated that the liver functions were gradually restored after treatment with sinigrin and that the agent did not cause liver toxicity. Cell cycle analysis indicated that sinigrin caused cell cycle arrest in G0/G1 phase. The results suggest that sinigrin exerts important anti-proliferative activities in carcinogen-induced hepatocarcinogenesis in rats, and highlight the potential of sinigrin as an anti-cancer agent for liver cancer.  相似文献

* Plant defense traits often show high levels of genetic variation, despite clear impacts on plant fitness. This variation may be partly maintained by trade-offs in the defense against multiple herbivore species, for example between generalists and coevolved specialists. Despite a long-standing discussion in the literature on the subject, no study to date has specifically manipulated specialist and generalist herbivores independently of one another to determine whether the two guilds exert opposing selection pressures on specific defensive traits. * In two separate experiments, the dominant specialist and generalist herbivores of Brassica nigra were independently manipulated to test whether the composition of the herbivore community altered the direction of selection on a major defensive trait of the plant, sinigrin concentration. * It was found that generalist damage was negatively correlated but specialist loads were positively correlated with increasing sinigrin concentrations; and sinigrin concentration was favored when specialists were removed, disfavored (past an intermediate point) when generalists were removed and selectively neutral when plants faced both generalists and specialists.  相似文献

Among the different types of methionine-derived aliphatic glucosinolates (GS), sinigrin (2-propenyl), the final product in 3C GS biosynthetic pathway is considered very important as it has many pharmacological and therapeutic properties. In Brassica species, the candidate gene regulating synthesis of 3C GS remains ambiguous. Earlier reports of GSL-PRO, an ortholog of Arabidopsis thaliana gene At1g18500 as a probable candidate gene responsible for 3C GS biosynthesis in B. napus and B. oleracea could not be validated in B. juncea through genetic analysis. In this communication, we report the isolation and characterization of the gene CYP79F1, an ortholog of A. thaliana gene At1g16410 that is involved in the first step of core GS biosynthesis. The gene CYP79F1 in B. juncea showed presence-absence polymorphism between lines Varuna that synthesizes sinigrin and Heera virtually free from sinigrin. Using this presence-absence polymorphism, CYP79F1 was mapped to the previously mapped 3C GS QTL region (J16Gsl4) in the LG B4 of B. juncea. In Heera, the gene was observed to be truncated due to an insertion of a ~4.7 kb TE like element leading to the loss of function of the gene. Functional validation of the gene was carried out through both genetic and transgenic approaches. An F2 population segregating only for the gene CYP79F1 and the sinigrin phenotype showed perfect co-segregation. Finally, genetic transformation of a B. juncea line (QTL-NIL J16Gsl4) having high seed GS but lacking sinigrin with the wild type CYP79F1 showed the synthesis of sinigrin validating the role of CYP79F1 in regulating the synthesis of 3C GS in B. juncea.  相似文献

Polyclonal rat antibodies were raised to a bovine serum albumin-sinigrin conjugate and used to immunolocalize sinigrin (2-propenylglucosinolate) in imbibed seeds and developing seedlings of Brassica juncea. (L.) Czern. Sinigrin was localized to protein bodies in aleurone-like cells but shown to be absent from myrosin cells. Double labelling techniques were used to co-localize both myrosinase (β-thioglucoside glucohydrolase, EC 3.2.3.1) and sinigrin. Myrosin grains were labelled only with the anti-myrosinase antibody, but aleurone cells were labelled with both anti-myrosinase and anti-sinigrin antibodies. High-performance liquid chromatographic analysis of conventionally fixed and dehydrated seed tissues (4 h post imbibition in water), indicated a high proportion of sinigrin was retained in fixed tissues. Over a time course of 100 h, protein bodies within aleurone-like cells degraded, fused to form the cell vacuole and lost all myrosinase labelling but retained residual sinigrin labelling. The degradation of protein bodies corresponded to a decrease in retention of sinigrin in the fixed tissues. Oviposition by the diamondback moth, Plutella xylostella (Lepidoptera: Plutellidae), on substrates treated with host stimuli (cabbage homogenate or sinigrin) and/or waxes (paraffin or a mixture of 10 single chain n-alkanes) was quantified using continuous observations and endpoint bioassays. Paraffin or an n-alkane mixture applied over cabbage homogenate or sinigrin caused an increase in oviposition compared to that on any single stimulus in choice tests. Sinigrin alone at 10–5 M to 10–2 M is an ovipositional stimulant; addition of alkane over sinigrin made all sinigrin concentrations (10–6 M to 10–2 M) significantly more stimulatory than controls. Waxes alone do not stimulate oviposition. In choice tests, insect movement between sinigrin/alkane treatment combinations was random, however, once encountered, visit duration was significantly longer on sinigrin with alkane than on sites treated with either stimulus alone. Given the ubiquity of waxes on plant surfaces and the interaction between waxes and host-specific chemical stimuli, waxes should be included when considering factors that significantly influence herbivore host acceptance.  相似文献

The cabbage aphid, Brevicoryne brassicae, has developed a chemical defence system that exploits and mimics that of its host plants, involving sequestration of the major plant secondary metabolites (glucosinolates). Like its host plants, the aphid produces a myrosinase (beta-thioglucoside glucohydrolase) to catalyse the hydrolysis of glucosinolates, yielding biologically active products. Here, we demonstrate that aphid myrosinase expression in head/thoracic muscle starts during embryonic development and protein levels continue to accumulate after the nymphs are born. However, aphids are entirely dependent on the host plant for the glucosinolate substrate, which they store in the haemolymph. Uptake of a glucosinolate (sinigrin) was investigated when aphids fed on plants or an in vitro system and followed a different developmental pattern in winged and wingless aphid morphs. In nymphs of the wingless aphid morph, glucosinolate level continued to increase throughout the development to the adult stage, but the quantity in nymphs of the winged form peaked before eclosion (at day 7) and subsequently declined. Winged aphids excreted significantly higher amounts of glucosinolate in the honeydew when compared with wingless aphids, suggesting regulated transport across the gut. The higher level of sinigrin in wingless aphids had a significant negative impact on survival of a ladybird predator. Larvae of Adalia bipunctata were unable to survive when fed adult wingless aphids from a 1% sinigrin diet, but survived successfully when fed aphids from a glucosinolate-free diet (wingless or winged), or winged aphids from 1% sinigrin. The apparent lack of an effective chemical defence system in adult winged aphids possibly reflects their energetic investment in flight as an alternative predator avoidance mechanism.  相似文献

Abstract. The behavioural facilitation hypothesis, tested in the present study, suggests that evolution of host-plant shifts by phytophagous insects is based on the preadaptation of insects to the chemistry of potentially novel plant hosts. Thus, closely-related insects should have similar sensitivities to compounds that are shared by different host plants. The chemoreception is investigated for four phytophagous flies, Delia radicum, Delia floralis, Delia antiqua and Delia platura (Diptera, Calyptratae: Anthomyiidae), belonging to the same genus but developing mainly on different plant families, with particular secondary plant compound profiles. In addition, the carrot fly, Psila rosae, an acalyptrate Diptera, is included as an unrelated species that is associated with completely different host plants. For the comparison, the known oviposition stimulants of the cabbage root fly (glucobrassicin, sinalbin, sinigrin and a thia-triaza-fluorene compound; CIF-1) present on the cabbage leaf surface were chosen. Responses from prothoracic tarsal sensilla are recorded to contact stimulation in a dose-dependent manner. Among the different flies tested, only D. radicum responds to all the compounds. By contrast, D. floralis is only sensitive to CIF-1, and not specifically on the C5 sensillum, a finding that is in conflict with previously published results. This discrepancy is possibly an indication of the variability among flies originating from different cultures or habitats. With the exception of sinigrin at high concentration, the various compounds tested do not stimulate D. antiqua or D. platura. However, the carrot fly appears to be completely insensitive to sinigrin even at the highest tested concentration of 10−1 M. The responses of the contact-chemoreceptor neurones to the selected compounds therefore provide little evidence of common sensitivities that would explain host shift in Delia species and specialization at the physiological level. The wide divergence within closely-related species and rearing cultures appears to indicate that the sensitivity and distribution of sensory receptor neurones is very variable on an evolutionary scale.  相似文献

Zusammenfassung Bei einigen polyphagen und oligophagen Blattlausarten wurde die Nahrungsaufnahme an einer 20% igen wäßrigen Saccharose-Lösung ohne bzw. mit Zusatz von 0,1% Sinigrin mit Hilfe der Tracer-Methode verglichen. Die Anwesenheit von Sinigrin förderte die Nahrungsaufnahme bei Brevicoryne brassicae, Myzus persicae und Megoura viciae, sie hemmte sie stark bei Rhopalosiphum padi und schwächte sie bei Aphis fabae und Acyrhosiphon pisum. Aulacorthum circumflexum reagierte in einigen Versuchen positiv, in anderen aber negativ auf das Glycosid. Bei der Mehrzahl der untersuchten Arten entspricht demnach Annahme bzw. Ablehnung von Cruciferen als Wirte der Reaktion auf Sinigrin. Im Gegensatz dazu stehen die Befunde bei M. viciae, die keine Cruciferen besiedelt, an Lösungen mit Sinigrin jedoch höhere Werte erreichte, so daß vermutlich andere physikalische oder chemische Eigenschaften von Cruciferen deren Ansiedlung verhindern.
Effect of sinigrin on sucrose uptake by some polyphagous and oligophagous aphids (Aphididae)
Summary The effect of 0.1% sinigrin on sucrose uptake (20% aqueous solution) by some polyphagous and oligophagous aphid species was tested using the tracer method. In the presence of sinigrin the sucrose ingestion of Brevicoryne brassicae, Myzus persicae and Megoura viciae was promoted. With Rhopalosiphum padi there was a strong, and with Aphis fabae and Acyrthosiphon pisum only a slight inhibition of sucrose uptake. Aulacorthum circumflexum reacted positively in some experiments, but negatively in others. Thus the acceptance or refusal of cruciferous plants as a host by the majority of the species examined corresponded to their reaction to sinigrin. Contrasting results were obtained with M. viciae. Although cruciferous plants are non hosts for this species, it ingested more sucrose when sinigrin was present. Therefore other physical or chemical properties must be considered for the refusal of cruciferous plants as a host.

The NADPH-dependent conversion of amino acids to their aldoximes is an initial step in glucosinolate biosynthesis. A number of microsomal aldoxime-forming monooxygenase activities were detected in leaves from a variety of glucosi-nolate-containing species, whereas barley, bean and tobacco leaves did not contain any such activities. The substrates for these monooxygenases in each species largely correlated with the spectrum of glucosinolates found in that species. No activity was detected that metabolized homomethionine (supposed precursor of 2-propenylglucosinolate [sinigrin]), even in species where sinigrin was the major glucosinolate. In Sinapis species containing hydroxybenzylglucosinolate (sinalbin), activity with L-Tyr was detected, whereas Brassica species containing sinalbin had no such activity. However, these Brassicas did contain an L-Phe monooxygenase activity. Partial characterization of the monooxygenases indicated that in Brassica species, Nasturtium officinalis and Raphanus sativus these resembled the flavin-linked monooxygenases previously found in oilseed rape (Brassica napus) and Chinese cabbage (Brassica campestris). The L-Tyr-dependent activity in Sinapis species, and the L-Phe-dependent activity in Tropacolum majus, had characteristics of cytochrome P450-type enzymes. No similarity was found with any other known amino acid metabolizing enzymes (including decarboxylases, amino acid oxidases and diamine/polyamine oxidases).  相似文献

Two fungal strains, Aspergillus clavatus II-9 and Fusarium oxysporum @ 149, proved to be capable of degrading sinigrin and sinalbin. During the degradation of sinigrin by whole cells of the Aspergillus strain, allylcyanide accumulated in the liquid incubation mixture. After a maximum concentration had been reached, the concentration of allylcyanide decreased as a result of its instability in the medium used. Incubation of cell-free extracts with sinigrin resulted in accumulation of glucose and allylisothiocyanate, suggesting that myrosinase is involved. Experiments with intact cells and cell-free extracts indicate the formation of an as yet unknown intermediate. When sinigrin was degraded by the Aspergillus strain in mustard seed meal under solid-state fermentation (SSF) conditions, no accumulation of allylcyanide or allylisothiocyanate was measured. Degradation of sinigrin by F. oxysporum @ 149 did not result in accumulation of intermediates, neither in liquid incubation mixtures nor in mustard seed meal under SSF conditions. Sinigrin was not degraded during incubation with cell-free extracts of F. oxysporum @ 149. Degradation of sinalbin by A. clavatus and F. oxysporum was measured during fermentation of yellow mustard seed meal under SSF conditions. Both fungi are useful for laboratory-scale SSF of mustard seed meal, thus opening new perspectives for a cost effective detoxification process for raw feed materials. Correspondence to: J. P. A series of trioxanes featuring sulfide, sulfone, and sulfonamide substituents in diverse positions has been prepared. Structure-activity relationship (SAR) generalizations highlight two major factors controlling the antimalarial potency of these new chemical entities: (1) the proximity of the sulfur-containing substituent to the crucial peroxide bond and (2) the oxidation state of the sulfur-containing substituent. Members of the Brassicaceae are known for their contents of nutrients and health‐promoting phytochemicals, including glucosinolates. The concentrations of these chemopreventive compounds (glucosinolate‐degradation products, the bioactive isothiocyanates) may be modified under salinity. In this work, the effect of the aliphatic glucosinolate sinigrin (2‐propenyl‐glucosinolate) on plant water balance, involving aquaporins, was explored under salt stress. For this purpose, water uptake and its transport through the plasma membrane were determined in plants after NaCl addition, when sinigrin was also supplied. We found higher hydraulic conductance (L0) and water permeability (Pf) and increased abundance of PIP2 aquaporins after the direct administration of sinigrin, showing the ability of the roots to promote cellular water transport across the plasma membrane in spite of the stress conditions imposed. The higher content of the allyl‐isothiocyanate and the absence of sinigrin in the plant tissues suggest that the isothiocyanate is related to water balance; in fact, a direct effect of this nitro‐sulphate compound on water uptake is proposed. This work provides the first evidence that the addition of a glucosinolate can regulate aquaporins and water transport: this effect and the mechanism(s) involved merit further investigation.  相似文献

Introduction – Sinigrin, a major glucosinolate present in Indian mustard (Brassica juncea L.) seeds as the precursor of the anticancer compound allyl isothiocyanate, shows a wide range of biological activities. It's necessary to optimize the extraction methods and conditions, in order to improve the extraction productivity and save raw material. Objective – To systemically investigate and optimize the most important factors affected the productivity of sinigrin in the process of extraction using response surface methodology. Methodology – The ranges of three main factors including the ethanol concentration, extraction time and extraction temperature were selected by the one‐factor‐at‐a‐time method. The conditions of ultrasonic‐stimulated extraction of sinigrin from defatted Indian mustard seed powder were optimized by Box‐Behnken design to obtain the maximum productivity. Result – The predicted productivity (3.81%) was obtained using 57% ethanol concentration at 81°C for 60 min, with the coefficient of the model R2 > 0.96 (n = 17). The actual productivity (3.84 ± 0.02%) of sinigrin under the optimized condition was increased by 70.67% compared with the result of conventional extraction. Meanwhile, HPLC, UV and IR were applied to examine if there is a difference between the ultrasonic‐stimulated solvent extraction and conventional extraction, and the improvement of productivity of sinigrin depended on the destruction of cell wall caused by the elimination of outer pectinous material was explained by SEM and composition content analysis. Conclusion – The ultrasonic‐stimulated solvent extraction was suggested to be a promising method to improve the productivity of sinigrin. And the results demonstrated that sinigrin productivity may be related to pectinous materials existed in the seeds. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Capparis ovata var. palaestina Zoh., C. spinosa var. aegyptia Boiss. and C. spinosa var. deserti Zoh., were investigated for glucosinolates. Glucoiberin, glucocapparin, sinigrin, glucocleomin, glucocapangulin, glucobrassicin and neoglucobrassicin, in addition to two others, were isolated. Four of these viz. glucoiberin, sinigrin, glucobrassicin and neoglucobrassicin were detected for the first time in Capparis species. Comparative chromatographic analyses of the glucosinolates of the plants examined revealed qualitative differences.  相似文献

1. Plant secondary metabolites can govern prey–predator interactions by altering the diet breadth of predators and sometimes provide an ecological refuge to prey. Brassicaceae plants and their specialist pests can be used as a model system for understanding the role of chemically mediated effects restricting the diet breadth of natural enemies, and consequently the occurrence of enemy‐free space for the specialist pest. 2. The objective of the present study was to test the performance of the generalist predator Episyrphus balteatus De Geer (Diptera: Syrphidae) fed on the specialist herbivore Brevicoryne brassicae L.(Homoptera: Aphididae), reared on two different brassica species: black mustard (Brassica nigra), a wild species with high levels of sinigrin; and canola (Brassica napus), a cultivated species without sinigrin. 3. The preference and performance of the predator and the performance of the prey were measured. Sinigrin was quantified by high‐performance liquid chromatography in both leaf samples and aphids reared on the two host plants. 4. The cabbage aphid performed better on canola than on black mustard. The performance of the predator on this aphid when reared on canola was clearly better than when reared on black mustard. Females had a higher overall preference for cabbage aphids reared on canola than on black mustard. 5. The ability of aphids reared on plants with high glucosinolate content to reduce the performance of their generalist predators indicates that the presence of B. nigra may provide enemy‐free space for the cabbage aphid from its predator, a concept that has useful application in the context of biological control for agricultural systems.  相似文献

In Brassica napus, glucosinolates are transported from all parts of the plant into the embryo during seed development. In this study we describe the uptake of the alkenyl glucosinolate sinigrin by microspore derived embryos from high and low glucosinolate genotypes. Microspore derived embryos develop completely isolated from maternal tissues unlike zygotic embryos, which contains glucosinolates transported into the embryo synthesised in the vegetative tissues. The sinigrin in the culture medium was almost completely absorbed by the embryos after three days of culture. The embryos of high and low glucosinolate genotypes were equally capable of absorbing sinigrin from the medium. A significant increase in different alkenyl glucosinolates following feeding of sinigrin suggests induction of biosynthetic enzymes in the embryos. Following excess feeding of sinigrin, we found a strong uptake against a concentration gradient and stable accumulation by the embryos. The glucosinolate was detected in single dissected cotyledons by a photometric test and by HPLC. This test could potentially be useful for screening mutants defective in glucosinolate uptake into the embryo.  相似文献
