浓硫酸处理对海滨木槿10个家系种子萌发的影响   总被引:1,自引:0,他引:1  
以海滨木槿10个家系种子为材料,采用浓硫酸不同时间(0、10、15和20 min)处理,测定种子发芽率、发芽势、发芽指数及萌发过程中相对吸水率、可溶性糖、淀粉和α-淀粉酶含量,研究浓硫酸处理对海滨木槿种子萌发的影响以及不同家系间种子萌发差异.结果表明:浓硫酸处理显著提高了海滨木槿种子的发芽率、发芽指数和发芽势,缩短了发芽时间.浓硫酸处理15 min后,种子发芽率、发芽势和发芽指数分别为95.7%、91.3%和13.28,萌发时间最短,效果最佳.海滨木槿不同家系间种子发芽率、发芽势和发芽指数差异显著,最高发芽指数(15.13)是最低发芽指数(7.15)的2.12倍,最高发芽势(98.0%)是最低发芽势(55.5%)的1.77倍.浓硫酸处理可加快海滨木槿种子萌发代谢生理生化进程.相对吸水率、可溶性糖和α-淀粉酶均呈单峰曲线变化(先升后降),在萌发初期达到最大值;淀粉含量与种子发芽指数呈显著负相关.  相似文献

为了探讨亚热带树木休眠理论以及休眠与树木分布的相互关系,研究了不同种源白花泡桐种子的休眠生理生态学特性。结果表明:白花泡桐种子适宜发芽温度范围在20~30℃,经过低温处理后,适宜发芽温度范围扩大到15~30℃,具有浅低温休眠特性;5℃的低温处理不仅提高了不同种源白花泡桐种子的发芽率和发芽势,而且加快了发芽速度,尤其在15℃发芽温度条件下的促进效果更加明显;40d的5℃低温处理效果最好.20d、60d的低温处理效果相对较差;没有用低温处理的不同种源白花泡桐种子,除了在25℃发芽温度下的发芽势差异不显著外,在不同发芽温度条件下的发芽率与发芽势都存在着显著差异,但在20℃、25℃发芽温度下的平均发芽时间差异不显著。在15℃发芽温度条件下,除了桂林种源以外,没有用低温处理的白花泡桐种子的发芽率与发芽势都随着种源纬度的增加而增加,而在20~30℃的适宜发芽温度范围内随种源纬度的变化趋势不明显;不同种源白花泡桐种子对低温处理反应差异明显。低温处理后的中亚热带以南的低纬度种源白花泡桐种子发芽率、发芽势都较北亚热带的高纬度种源高。低温处理后的中亚热带株州种源和北亚热带南部的宜昌种源白花泡桐种子的平均发芽时间最长,而随着纬度的降低或增加平均发芽时间都变短。不同种源白花泡桐种子的浅休眠特性不同,是对不同种源地气候的一种生理生态适应。  相似文献

为明确激光对番茄耐盐性的影响,用番茄(Lycoperslcon esculentum Mill)种子为实验材料,以不同剂量的He-Ne激光辐照刚萌动的种子,用0.3%的NaCI溶液模拟盐胁迫,观察出苗时间、出苗率,测定相关的生理生化指标.结果表明:激光处理后,番茄种子的发芽率(Gr)和发芽指数(Gi)有不同程度的降低;番茄幼苗游离脯氨酸积累量、根系活力、超氧化物歧化酶(SOD)活性都有不同程度的升高;丙二醛(MDA)含量降低.分析不同处理的各项指标可知,辐照量为23.04 J/cm2是较合适的处理.说明利用一定程度的激光处理可以提高番茄的抗盐性.  相似文献

以哥伦比亚生态型(Columbia-0)的野生型拟南芥为材料,利用He-Ne激光生物辐照仪(632.8nm,5mW/mm2)处理拟南芥种子,统计其发芽势和发芽率,并测定根长及叶绿素、可溶性糖、可溶性蛋白和丙二醛(MDA)的含量,以探讨He-Ne激光对拟南芥种子萌发和幼苗生长的影响。结果显示,短时间的He-Ne激光辐照可促进拟南芥的生长发育,且最佳辐照时间为4min,此时的发芽势、发芽率、根长、叶绿素、可溶性糖及可溶性蛋白的含量均达到最大,而丙二醛(MDA)的含量为最低。当辐照时间大于4min时,促进作用逐渐变小,且随着辐照时间的延长而表现出抑制作用。研究表明,适量的He-Ne激光辐照在一定程度上可以促进拟南芥种子的萌发和幼苗生长以及保护其细胞膜免受伤害。  相似文献

以紫金龙为材料,研究了1%CuSO_4、0.1%KMnSO_4、1%CH_2O、75%C_2H_6O、10%H_2O_2、0.1%HgCl_2、10%NaClO等消毒剂处理对紫金龙种子萌发的影响。结果表明:用10%H_2O_2消毒的紫金龙种子,发芽势和发芽指数最高,且10%H_2O_2消毒对种子发芽率抑制不明显,是较为理想的表面消毒剂。相反,其它消毒处理都不同程度地削弱了种子的发芽率、发芽势和发芽指数,在一定程度上抑制了紫金龙种子的萌发。  相似文献

以哥伦比亚生态型(Columbia-0)拟南芥(Arabidopsis thaliana)为实验材料, 人工模拟UV-B辐照处理拟南芥种子, 统计其发芽势和发芽率, 测定根长、株高、叶绿素、可溶性糖、可溶性蛋白以及丙二醛(MDA)含量, 研究UV-B辐照处理对拟南芥种子萌发和幼苗生长的影响。研究结果表明, 低剂量的UV-B辐照可以促进拟南芥种子萌发和幼苗生长, 并且最佳辐照剂量为1.0 kJ·m–2(P<0.05), 此时的发芽势、发芽率、根长、株高、叶绿素、可溶性糖及可溶性蛋白的含量均达到最大, 而丙二醛的含量变化则不明显(P>0.05)。当辐照剂量大于1.0 kJ·m–2时, 促进作用逐渐变小, 并且随着辐照剂量的增加, 表现出了抑制作用。实验结果表明, 适当剂量的UV-B辐照在一定程度上可以促进拟南芥种子萌发和幼苗生长, 而过高剂量的辐照则对其产生抑制或损伤作用。  相似文献

沙生针茅是重要的荒漠植物,研究其种子萌发特性及其对主要影响因子的响应对于认识其生存适应性以及荒漠植被恢复具有重要意义。鉴于沙生针茅种子常处休眠状态、不易萌发,而现有研究尚未充分揭示各种因素和处理对沙生针茅种子萌发的影响,本论文报道了不同因素、不同处理对沙生针茅种子萌发的影响。结果表明:(1)60℃蒸馏水浸泡种子24 h,种子吸水率、发芽率、发芽势分别达到26%、38%和32%;(2)-20/20℃变温处理96 h,发芽率、发芽势分别达到48%和46%;(3)切除绝大部分胚乳(3/4)后,种子发芽率、发芽势分别可达到58%和54%;(4)用80 mg·L~(-1)赤霉素(GA3)水溶液浸泡种子24 h,发芽率、发芽势分别达到54%和48%;(5)用聚乙二醇(PEG-6000)处理,随浓度增加,种子发芽率、发芽势、发芽指数均呈下降趋势,种子萌发水分胁迫的临界值和极限值分别为15.9%和26.3%;(6)盐胁迫处理下,种子发芽率、发芽势、发芽指数均随盐碱浓度增加呈下降趋势,其中Na2CO3处理下的种子萌发率下降幅度明显大于Na Cl,表明碱性盐胁迫对沙生针茅种子萌发的抑制作用大于中性盐胁迫;(7)采用综合处理方法,种子发芽率、发芽势分别达到78%和76%。  相似文献

以野生乌拉尔甘草种子为材料,研究不同温度、覆土厚度、干旱胁迫、pH值、盐胁迫、干旱和盐双重胁迫下种子发芽与萘乙酸、3-吲哚乙酸、褪黑素浸种对种子发芽特性的影响。结果表明,(1)乌拉尔甘草种子最适发芽温度为20~25℃,发芽率≥90%,发芽时滞显著低于其他温度;(2)乌拉尔甘草种子对pH适应范围较宽,pH值在4~8发芽率较高;(3)覆土厚度在0~0.5 cm时发芽率显著高于其他覆土厚度;(4)盐碱、干旱或盐碱和干旱双重胁迫下乌拉尔甘草种子发芽势和发芽率显著降低;(5)100~300μmol/L褪黑素预处理可显著提高盐胁迫处理下乌拉尔甘草种子的发芽率和发芽势;(6)萘乙酸对乌拉尔甘草种子发芽无促进作用,但当浓度>50 mg/L时,乌拉尔甘草种子发芽势和发芽率显著降低;(7)吲哚乙酸浓度为50 mg/L时,乌拉尔甘草种子发芽势和发芽率最高。综上所述,乌拉尔甘草种子发芽具有一定的抗非生物胁迫能力,且适宜的外源植物激素浓度预处理可促进种子发芽。  相似文献

Background

Seed germination is a complex physiological process during which mobilization of nutrient reserves happens. In different crops, this event might be mediated by different regulatory and metabolic pathways. Proteome profiling has been proved to be an efficient way that can help us to construct these pathways. However, no such studies have been performed in soybean germinating seeds up to date.

Results

Proteome profiling was conducted through one-dimensional gel electrophoresis followed by liquid chromatography and tandem mass spectrometry strategy in the germinating seeds of soybean (glycine max). Comprehensive comparisons were also carried out between rice and soybean germinating seeds. 764 proteins belonging to 14 functional groups were identified and metabolism related proteins were the largest group. Deep analyses of the proteins and pathways showed that lipids were degraded through lipoxygenase dependent pathway and proteins were degraded through both protease and 26S proteosome system, and the lipoxygenase could also help to remove the reactive oxygen species during the rapid mobilization of reserves of soybean germinating seeds. The differences between rice and soybean germinating seeds proteome profiles indicate that each crop species has distinct mechanism for reserves mobilization during germination. Different reserves could be converted into starches before they are totally utilized during the germination in different crops seeds.

Conclusions

This study is the first comprehensive analysis of proteome profile in germinating soybean seeds to date. The data presented in this paper will improve our understanding of the physiological and biochemical status in the imbibed soybean seeds just prior to germination. Comparison of the protein profile with that of germinating rice seeds gives us new insights on mobilization of nutrient reserves during the germination of crops seeds.  相似文献

为探讨卫星搭载处理对甘草种子萌发中生理特性的影响,本文运用卫星搭载技术对甘草种子进行太空诱变处理,考察了卫星搭载后甘草种子(卫星1号,GF)在干旱胁迫条件下,萌发过程中的发芽率、胚根长势、可溶性蛋白变化以及过氧化物酶和过氧化氢酶的活力,并与地面对照组种子(地面组,GN)进行了比较。结果表明,在干旱胁迫条件下(水势-1.0MPa),飞行组种子的发芽率、胚根长势要高于地面对照组,其萌发过程中的过氧化物酶、过氧化氢酶活力也高于地面对照组。卫星搭载处理后,甘草种子的抗旱性能增强。  相似文献

Experiments were conducted to characterise the changes, especially of water status in germinating and non-germinating wheat seeds by nuclear magnetic resonance (NMR) spectroscopy. NMR relaxation time (T2) measurements showed tri-phasic or bi-phasic characteristics during different stages of hydration, depending on the seed's ability to germinate. Component analysis of T2 data revealed the existence of only two components, bound and bulk water, in dry seeds. In contrast, both the germinating and non-germinating wheat seeds had a three-component water proton system (bound, bulk and free water) in phase I of hydration. During the lag phase (phase II) of hydration, bulk water component of non-germinating seeds disappeared completely, resulting in a two component water proton system. Nevertheless, the three component water proton system was observed in the germinating seeds in phase II. Following phase II, rapid hydration (phase III) was observed in germinating seeds only. Water protons were re-organised and there were increases in bulk and free water but decreases in bound water concomitantly. Comparison of the physical state of water in these seeds by NMR spectroscopy with that of tissue leachate conductivity measurement suggests that the seed membrane system was affected more evidently in non-germinating seeds, leading to the disorganised cell structure. The present study provides evidence that the reorganisation of physical state of water in germinating wheat seeds during hydration is essential for its subsequent event of germination.  相似文献

Summary The germination of lettuce seeds is inhibited by the nucleotide base analogue 6-methylpurine. RNA synthesis has been measured during imbibition and germination as 32P-phosphate incorporation into RNA species as fractionated by polyacrylamide gel electrophoresis. Seeds were surface sterilized and imbibed in the presence of various antibiotics. RNA preparations from lettuce seeds were coelectrophoresed with 3H-RNA prepared from bacteria to check for bacterial contamination of the seeds. There is a much higher rate of RNA synthesis in illuminated, germinating seeds as compared to dark, non-germinating seeds. This difference does not develop until after 12 hours of imbibition at 27°, which is the time of onset of germination and radicle growth.This investigation was supported by a contract from the United States Department of Agriculture (No. 616-15-3). Susceptibility to UV irradiation of B. cereus BIS-59 spores undergoing germination at various stages-dormant spores to vegetative cell stage and their ability to recover from radiation damage were studied. For a given dose of radiation, the number of spore photoproducts (SPP) formed in the DNA of dormant spores was about 5-times greater than that of thymine dimers (TT) formed in the DNA of vegetative cells. At intermediate stages of the germination cycle, there was a rapid decline in the UV radiation-induced SPP formed in DNA with a concomitant increase in the UV radiation-induced TT formed in DNA. Bacterial spores undergoing germination (up to 3 hr) in the low nutrient medium (0.3% yeast extract) displayed much higher resistance to UV radiation than those germinating in the rich nutrient medium, even though there was no discernible difference under the two incubation conditions in respect of the extent of germination and the time at which the outgrowth stage appeared (3 hr). This was due to the formation TT in the DNA of spores germinating in the low nutrient as compared to that of spores germinating in the rich-nutrient medium. In UV-irradiated dormant spores, SPP formed in the spore DNA did not disappear even after prolonged incubation in the non-germinating medium. However, when the UV-irradiated dormant spores were germinated in low or rich nutrient medium, a significant proportion of SPP in DNA was eliminated. The dormant spores incubated in either of the germinating media for 15 min and then UV-irradiated were capable of eliminating SPP (presumably by monomerization) even by incubation in a non-germinating medium and in the complete absence of protein synthesis (buffer holding recovery), thereby implying that spore-repair enzymes were activated in response to initial's germination. The acquisition of photo-reactivation ability appeared in spores subjected to germination only in the rich-nutrient medium at the outgrowth stage and required de novo synthesis of the required enzymes.  相似文献

The timing of germination is a key life‐history trait that may strongly influence plant fitness and that sets the stage for selection on traits expressed later in the life cycle. In seasonal environments, the period favourable for germination and the total length of the growing season are limited. The optimal timing of germination may therefore be governed by conflicting selection through survival and fecundity. We conducted a field experiment to examine the effects of timing of germination on survival, fecundity and overall fitness in a natural population of the annual herb Arabidopsis thaliana in north‐central Sweden. Seedlings were transplanted at three different times in late summer and in autumn covering the period of seed germination in the study population. Early germination was associated with low seedling survival, but also with high survival and fecundity among established plants. The advantages of germinating early more than balanced the disadvantage and selection favoured early germination. The results suggest that low survival among early germinating seeds is the main force opposing the evolution of earlier germination and that the optimal timing of germination should vary in space and time as a function of the direction and strength of selection acting during different life‐history stages.  相似文献

为了阐明干旱胁迫与不同储藏时间对杠柳种子萌发的影响机理,在室内采用不同浓度的聚乙二醇(PEG)模拟干旱胁迫条件,对3种不同储藏时间的杠柳种子进行了萌发期对比试验,观测了种子的总萌发率、发芽势、活力指数和胚根的生长情况,分析了抗旱特性,结果表明:不同程度的干旱胁迫与储藏时间对杠柳种子萌发均有一定的影响,当PEG-6000浓度为5%,对储藏1年杠柳种子的萌芽率和发芽势起促进作用,对储藏2年种子萌芽率轻度抑制,对储藏3年种子抑制影响较大,当PEG-6000浓度>5%,3种种子的萌芽率和发芽势均呈下降趋势,但当胁迫浓度增大到25%时,种子基本没有萌发。在不同的水分胁迫处理中,3种种子的活力指数和胚根生长速度一直呈下降趋势,尤其3年种子的下降幅度最大。不同储藏时间杠柳种子对干旱胁迫的适应性有差异,储藏1~2年的杠柳种子在PEG-6000浓度为5%~15%中萌发效果较好,且储藏1年种子好于2年种子,3年种子较差。3种种子的耐旱能力由强到弱依次为:1年种子>2年种子>3年种子。  相似文献

The fruit-bat Rousettus aegyptiacus (Pteropodidae) in Israel consumes a variety of cultivated and wild fruits. The aim of this study was to explore some of its qualities as a dispersal agent for six fruit-bearing plant species. The feeding roosts of the fruit-bat are located an average of 30 m from its feeding trees and thus the bats disperse the seeds away from the shade of the parent canopy. The bat spits out large seeds but may pass some (2%) of the small seeds (<4 mg) through its digestive tract. However, neither the deposited seeds nor the ejected seeds (except in one case) had a significantly higher percentage germinating than intact seeds. Although the fruit-bat did not increase the percentage germinating, seeds of three plant species subject to different feeding behaviors (deposited in feces or spat out as ejecta) had a different temporal pattern of germination from the intact seeds. The combined seed germination distribution generated by these different treatments is more even over time than for each treatment alone. It is sugested that this increases asynchronous germination and therefore enhances plant fitness by spreading the risks encountered during germination, especially in eastern Mediterranean habitats where the pattern of rainfall is unpredictable.  相似文献

Alfalfa sprouts and other seed sprouts have been implicated in numerous outbreaks of salmonellosis. The source of these epidemics appears to have been low-level contamination of seeds by Salmonella bacteria that developed into clinically significant populations during the seed germination process. To test the possibility that Salmonella enterica strains carry host range determinants that allow them to grow on alfalfa, strains isolated from alfalfa or other sources were surveyed for their ability to grow on germinating alfalfa seeds. An S. enterica serovar Cubana strain originally isolated from contaminated alfalfa sprouts multiplied most rapidly during the initial 24 h of the seed germination process. Germinating alfalfa seeds supported the multiplication of S. enterica cells prior to the emergence of the root radicle at 72 h. Thereafter, much lower rates of multiplication were apparent. The ability of S. enterica to grow on germinating alfalfa seeds was independent of the serovar, isolation source, or virulence of the strain. Isolates obtained from alfalfa attained population levels similar to those observed for strains isolated from contaminated meat products or stools. Each of the strains could be detected in the waste irrigation water, with populations being strongly correlated with those detected on the germinating alfalfa seeds. The S. enterica strains were capable of utilizing the waste irrigation water as a sole carbon and nitrogen source. S. enterica strains thus appear to grow saprophytically on soluble organics released from seeds during early phases of germination. The ability to detect S. enterica in the waste irrigation water early in the germination process indicates that this method may be used as a simple way to monitor the contamination of sprouts during commercial operations.  相似文献
