The aim of this study was to determine whether the olfactorysystem is responsible for the discriminability of the stereoisomersof nicotine. The EOG was recorded after stimulation with differentconcentrations of undistilled S(–)-, distilled S(–)-and distilled R(–)-nicotine separately in three groupsof frogs (Xenopus laevis). The responses to all types of nicotineused in the experiments increased with increasing stimulus concentration.The responses to undistilled S(–)-nicotine were significantlylower compared to responses to distilled S(–)- and R(+)-nicotine,whereas no significant differences could be found when the purifiedstereoisomers of nicotine [distilled S(–)-nicotine, distilledR(+)-nicotine] were compared. Control measurements of time courseand peak concentration employing a UV-detection method demonstratedthat the differences between distilled and undistilled S(–)-nicotinecould not be explained by different nicotine concentrations. The fact that no differences between the pure nicotine stereoisomerscould be found for all concentrations used in our experimentsand that experiments in humans revealed similar detection thresholdsfor both stereoisomers points to a similar receptor affinityof R(+)- and S (–)-nicotine within the olfactory system.At this point, it cannot be determined whether the observeddifferences in the perception of nicotine enantiomers in humansare due to differences in quality coding by stereospecific receptorson the olfactory sensory cells or by specific receptors on thetrigeminal nerve endings. Sorghum plants were grown in the laboratory with the root systemof each plant split between two pots. Three split pot treatmentswere established: (– –) treatment, where both halvesof the root were free from Striga; (– +) treatment, wherethe soil in one half of the pot had been inoculated with Strigaseed; (+ +) treatment, where the soil in both halves of thepot had been inoculated with Striga. Seed, stem and leaf weight were reduced by 82, 60 and 26 percent respectively in (+ +) plants compared to (– –)plants. Partially infected plants (– +) behaved similarlyto (+ +) plants. Rates of light saturating carbon dioxide fixation in (+ +) and(– +) plants were only 60 per cent of those measured in(– –) plants. This reduction was independent ofchanges in stomatal conductance. The effects of Striga on the growth and photosynthesis of sorghumappear to be independent of the degree of parasitism to whichthe host is subjected. The difference in production betweeninfected and uninfected plants was greater than could be accountedfor in term of competition with the parasite for resources,and Striga appears to have a pathological effect on the host. The olfactory discrimination process of male cabbage loopermoths, Trichoplusa ni (Hübner), was assessed by measuringtheir response to one of two emission sources within a windtunnel. The males discriminated between (1) Z7–12:Ac concentrations;(2) Z7–12:Ac alone and the volatile emission from excisedfemale sex pheromone glands; and (3) Z7–12:Ac and theemission from a mixture of six synthetic pheromone componentsthat mimics the volatile emissions of a female gland. Althoughmales could discriminate between a freshly excised female sexpheromone gland and 7.4x 10–11 M Z7–12:Ac, theycould not discriminate between a gland and 78.5x10–11M Z7–12:Ac. Males also could not discriminate betweenthe mixture of six volatile compounds and 28.7x10–11 Mof Z7–12:Ac. The data show that male cabbage looper mothshave difficulty discriminating between Z7–12:Ac aloneand in mixtures with other female-emitted volatile compounds.  相似文献

Enantiomers of the earthy smelling odorant ‘geosmin’(t-1, 10-dimethyl-t-9-decalol) were tested on 52 human subjectsfor odor threshold ratio (+)/(–) variability. Thresholdconcentrations of (+) and (–) and their (+)/(–)ratios appeared to be monomodally distributed. For most subjects,the ratio varied within a predictable narrow range comparableto that found previously for carvone enantiomers and contraryto the large variability found for -ionone enantiomers (Polaket al., 1989). The impact of a cyclopoid copepod population on the protozoacommunity (two ciliate categories and Cryptomonas) was assessedweekly during the spring cohort of Cyclops vicinus (one monthduration) in hypereutrophic Lake Søbygård by insitu gradient experiments with manipulation of ambient zooplanktonabundance. As C.vicinus always made up >92% of the zooplanktonbiomass, the response of protozoa is assumed to be a resultof predation by the copepod. Significant effects of copepodbiomass on protozoa net population growth rates were obtainedin the four experiments. Copepod clearance rates were significantlyhigher on oligotrichs than on prostomatids and Cryptomonas butdeclined for all three protozoa categories during the firstthree weeks of the copepod cohort, probably because of the changein developmental instar composition of the copepod population.Grazing impact on protozoa at ambient copepod abundance wasconsiderable (range, 0.05–0.87 day–1) and could,together with the estimated reproductive potential of protozoans(range, –0.20–0.87 day–1), account for thedecline in abundance and biomass of protozoa during the cohortdevelopment. Carbon flow from the protozoa to C.vicinus (range,2.8–23.5 µg C l–1 day–1) documents thepresence of a trophic link between protozoa and the spring cohortof C.vicinus in Lake Søbygård.  相似文献

The light-catalysed isomerization of (+)-abscisic acid (ABA)to its trans isomer during isolation from leaves was monitoredby the addition of (±)-[2-14C]ABA to the extraction medium.(+)Trans-abscisic acid (t-ABA) was found to occur naturallyin rose (Rosa arvensis) leaves at 20µg/kg fresh weight;(+)-ABA was present at 594µg/kg. (±)-[2-14D]Trans-abscisicacid was not isomerized enzymically to ABA in tomato shoots. (±)-Abscisic acid was converted by tomato shoots to awater-soluble neutral product, ‘Metabolite B’, whichwas identified as abscisyl-ß-D-glucopyranoside. When(±)-[2-14C]trans-abscisic acid in an equimolar mixturewith (±)-[2-14C}ABA was fed to tomato shoots it was convertedto its glucose ester 10 times faster than was ABA. Trans-abscisyl-ß-D-glucopyrano8ide only was formedfrom (±)-[2-14C]t-ABA in experiments lasting up to 30h. Glucosyl abscisate was formed slowly from ABA and the freeacid fraction contained an excess of the unnatural (–).ABAas did the ABA released from abscisyl-ß-D-glucopyranosideby alkaline hydrolysis. The (+).ABA appeared to be the solesource of the acidic ‘Metabolite C" previously noted. The concentrations of endogenous (+)-, (+)-[2-14C]-, and (–)-[2-14C]ABAremaining as free acid, and also in the hydrolysate of abscisyl-ß-D-glucopyranoside,were measured by the ORD, UV absorption, and scintillation spectrometryof highly purified extracts of ABA from tomato shoots whichhad been supplied with racemic [2-l4C]ABA.  相似文献

Regulation of Branching in Decussate Species with Unequal Lateral Buds   总被引:1,自引:0,他引:1  
In the decussate plants Alternanthera philoxeroides and Hygrophilasp. the opposite axillary bud primordia are of unequal sizefrom the time of their inception; the larger or + buds lie alongone helix and the smaller or – buds along another (helicoidalsystem). In decapitated plants of Alternanthera both buds grewout, but unequally; if the node was vertically split growthof the two shoots was more equal, and if the + buds were excisedgrowth of the – shoots approximately equalled that ofcontrol + shoots. In decapitated shoots of Hygrophila grownin sterile culture only one bud, the + or larger one, grew outat each of the upper nodes. In excised cultured nodes, also,only the + bud grew out; but if the nodes were split longitudinallyboth buds grew out, initially rather unequally. These experimentssupport the view that the regulation of branching in these specieshas two components, apical dominance and the dominance of thelarger (+) bud over the smaller (–) bud at the same node.The restriction of growth potentiality imposed on the –bud is not permanent but can be modified. Further correlativeeffects on bud outgrowth include those of the subtending leavesand of buds at other nodes.  相似文献

The number of mating pairs, the size of the mating partners,and the distribution of individuals of Vivi-parus ater on agrid in Lake Zürich were recorded during one breeding seasonin 1990. There was positive assortative mating with respectto shell size. The proportion of copulating individuals rangedfrom 1% to 6% (average 3%) of the active population at any onetime. Individual snails copulated 60 times on average from Apriluntil November. Snails were abundant and copulated in shallowwater close to the shore in Spring. They moved towards deeperareas in Autumn. V. ater copulated on all substrates at anydepth (1–9 m)of the grid. The spatial distribution ofcopulations throughout the summer reflected the pattern of snailabundance. Two series of experiments were conducted to examine the utilizationof exogenous sperm and their degree of depletion in three hermaphroditicsea hares, Aplysia kurodai Baba, 1937, A. Juliana Quoy &Gaunard, 1832, and A. parvula Guilding in MOrch, 1863. The firstseries of experiments was designed to determine whether onemating is enough for female partners to receive sufficient spermto fertilize at least one egg mass. For this purpose, isolatedindividuals were mated as females once with conspecific individualsand the viability of their subsequent eggs was examined. Whensuccessfully inseminated, individuals of all the three speciesreceived enough sperm to fertilize at least one egg mass, andthey continued to lay 3.6–8.5 viable egg masses. However,10–36% of the matings by these sea hares did not resultin any viable egg production by the female partner, indicatingthat these matings did not involve sperm transfer Such matingswithout sperm transfer tended to be shorter than matings withsuccessful sperm transfer The second experimental series wasconducted to examine whether sperm depletion actually occursor not in field-caught adults. For this purpose, adult individualscaught in the field were allowed to lay eggs under isolatedconditions without mating All the individuals of A. kurodaiand A. juliana stored enough sperm to fertilize at least oneegg mass. Carotenogenic mutants ofPhycomyces, which accumulate excess β-carotene or its intermediates, always failed in zygospore development. No improvement occurred when such mutants were mated together with a helper wild type of the same mating type against the wild type of the opposite mating type. Addition of excess synthesized pheromone, trisporin B, also failed to improve the zygospore development, though the mating response was significantly activated in the early stages and abundant zygophores were formed. Exceptional acceleration of the zygospore development under these experimental conditions occurred in a regulatory albino mutant (carA), which does not accumulate excess intermediate carotenoids. Chemically- or genetically-induced ovarproduction of β-carotene or lycopene also inhibited the zygospore development. These results imply that the zygospore development ofPhycomyces is maximal when the intracellular amount of β-carotene is optimal (=wild type), and that pheromones act mainly in the early stages of mating, while other factors such as the cell-to-cell recognition system may also be involved in the later stages. Intracellular accumulation of excess β-carotene or its intermediates probably disturb such later-stage factors.  相似文献

The life cycles, biomass and secondary production of three sympatricfreshwater basommatophoran snails, Lymnaea palustris (MÜller),Physa fontinalis Linnaeus and Anisus rotundatus (Poiret) werestudied during two years in a freshwater ditch. L. palustrisexhibited an iteroparous life-cycle whereas the two other speciespresented a semelparous life-cycle, adults died just after oviposition.L. palustris secondary production (dry weight) value was higher(P = 11 298.4 mg 0.1 m–1 yr–1) than those of P.fontinalis (P = 846.3 mg 0.1 m–2 yr–1) and A. rotundatus(P = 1192 mg 0.1 m–2 yr–1). Canid social groups are typically thought to consist of extendedfamilies, that is, a dominant breeding pair and related nonbreedingsubordinates, that principally obtain indirect fitness benefitsfrom helping to raise the offspring of the dominant pair. Consequently,the monogamous pair has been viewed as the basic fundamentalunit of canid social organization. However, there have beenfew genetic studies that have tested this assumption. We analyzedthe parentage of red foxes (Vulpes vulpes) in a high-density(19.6–27.7 adult foxes/km2) population in Bristol, UK,to determine (1) whether groups typically produced a singlelitter of cubs annually and (2) whether male and female foxesexhibited monogamous mating strategies. Social monogamy (theproduction of one litter in a social group) was observed orassumed in 54% of breeding attempts (N = 13 group-years). However,polyandrous and polygynous patterns of mating were common. Multiplepaternity was confirmed in 38% of litters (N = 16) containingoffspring with resolved maternity and paternity (N = 30 cubs);when including cubs with unresolved paternity (N = 20), multiple-paternitymay have occurred in 69% of litters. Litters were sired by anaverage of 1.6 identified males (range = 1–4); when includingcubs with unresolved paternity, litters may have been siredby up to seven males. Only 20% (6/30) of cubs with resolvedmaternity and paternity were sired by males within the socialgroup. Within groups, dominant females did not breed with subordinatemales; dominant males did breed with subordinate females. Dominantand subordinate females both produced cubs with dominant andsubordinate males from other social groups. Mean adult relatednessin groups typically ranged from 0.15–0.35, indicativeof second-order rather than first-order relatives.  相似文献

Barley (Hordeum vulgare L., cvs Golf, Mette, and Laevigatum)was grown under nitrogen limitation in solution culture untilnear maturity. Three different nitrogen addition regimes wereused: in the ‘HN’ culture the relative rate of nitrate-Naddition (RA) was 0·08 d–1 until day 48 and thendecreased stepwise to, finally, 0·005 d–1 duringgrain-filling; the ‘LN’ culture received 45% ofthe nitrogen added in HN; the ‘CN’ culture was maintainedat RA 0·0375 d–1 throughout. Kinetics of net nitrateuptake were measured during ontogeny at 30 to 150 mmol m–3external nitrate. Vmax (which is argued to reflect the maximuminflux rate in these plants) declined with age in both HN andLN cultures. A pronounced transient drop was observed just beforeanthesis, which correlated in time with a peak in root nitrateconcentration. Similar, but less pronounced, trends were observedin CN. The relative Vmax (unit nitrogen taken up per unit nitrogenin plants and day) in all three cultures declined from 1·3–2·3d–1 during vegetative growth to 0·1–0·7d–1 during generative growth. These values are in HN andLN cultures 15- to more than 100-fold in excess of the demandset by growth rates throughout ontogeny. Predicted balancingnitrate concentrations (defined as the nitrate concentrationrequired to support the observed rate of growth) were below6·0 mmol m–3 in HN and LN cultures before anthesisand then decreased during ontogeny. In CN cultures the balancingnitrate concentration increased during grain-filling. Apartfrom the transient decline during anthesis, most of the effectof ageing on relative Vmax can be explained in terms of reducedcontribution of roots to total biomass (R:T). The loss in uptakeper unit root weight is largely compensated for by the declinewith time in average tissue nitrogen concentrations. The quantitativerelationships between relative Vmax and R:T in ageing plantsare similar to those observed for vegetative plants culturedat different RAs. The data support the contention that the capacity for nitrateacquisition in N-limited plants is under general growth control,rather than controlled by specific regulation of the biochemicalpathway of nitrate assimilation. Aquatic snails from south western Zimbabwe belonging to theBulinus trunscatus/tropicus complex vary widely in shell formsuggesting that more than one taxon could be present. This possibilitywas investigated by making observations on snail samples from13 populations from the Plumtree area, in respect of allozymevariation (5 polymorphic loci), shell morphology (9 variables),copulatory organ and chromosome number. Comparative data wereobtained from snails from north western Zimbabwe identifieddefinitely as B. tropicus. Analysis of the genetic structurerevealed a high degree of polymorphism (P) ranging from 0.29–0.80among populations from Plumtree and expected heterozygosity(He) from 0.02–0.22. No enzymatic diagnostic loci werefound which could differentiate the different morphs or populationsand discriminant function analysis on the morphological datashowed an overlap of morphs among populations. Snails analyzedfor chromosome number were all diploid (2n = 36). Snails exposedto Schistosoma haematobium mira-cidia were all refractory. Thisinformation supports the view of a single species, B. tropicus,which is differentiated due to migration barriers and whereenvironmental variables might be implicated in the morphometricdivergence. The ingestion rates of the copepod, Diacyclops thomasi, on thesoft-bodied rotifer, Synchaeta pectinata, increased 10-fold(0.07–0.77 Synchaeta h–1) over the range 50–250prey l–1. The saturating functional response curve appearedsigmoid but was statistically indistinguishable from a parabola.The response curve was more linear and 10 times lower over thesame range of density when Diacyclops was offered Kerarellacochleans, a species having a stiffened lorica. Diacyclops maximizedits ingestion rate on Synchaeta as a function of the availablegut space. Predation effort, measured as clearance rates, waslinked tentatively to changes in swimming speed of Diacyclopsand was a function of hunger level. Diacyclops, which were starvedfor varying periods of time, increased their ingestion rateson Synchaeta up to a maximum (-3.0 h–1) after 7–10h of food deprivation. The gut passage time of Diacyclops wasestimated to be 7–8 h. Therefore, ingestion rates (andclearance rates) appeared to be strongly correlated to the volumeof food in the gut.  相似文献

The capacity of adult newt (Triturus viridescens) spleen cellsto secrete antibody at 4 C allows simultaneous visualizationand quantification of non-secretory (S) and secretory(S+) rosette forming cells (RFC). Visualization of mammalianS+ RFC requires 37 C, a temperature at which S RFC appearto be fragile. RFC can be distinguished as S or S+ dueto whether one or more layers of erythrocytes adhere to thesurface of sensitized spleen cells. Different doses of horseerythrocytes (HRBC) affect newt S RFC and S+ RFC differentially.By varying the time between injections of different concentrationsof chicken erythrocytes (CRBC, the "carrier") and a constantdosage of CRBC-TNP (trinitrophenyl, the hapten) it is possibleto measure "helper" activity that correlates with the populationof S RFC and is both dose and time dependent. By varyingassay time after "helper" activity has been maximized, one candetermine the cytodynamics of anti-TNP antibody producing cell(APC) activity. For the first time these morphologically separableRFC can be related to their suspected physiologic behavior.A shift from S RFC to S+ RFC takes place during the immuneresponse. That similar dose-dependent response curves can beshown in adultRana pipiens suggests that the newt responsesrepresent a fundamental vertebrate pattern.  相似文献

The time course of xylem differentiation was determined in explantsof lettuce pith parenchyma (Lactuca sativa L. cv. Romana) culturedon Murashige and Skoog (1962) medium using different concentrationsof auxin (IAA) and one cytokinin (zeatin or kinetin). Increasinglevels of auxin from I mg 1–1 to 15 mg 1–1 in thepresence of a constant level of a cytokinin (zeatin or kinetin)yielded up to 10 mg 1–1 IAA, an increase in the numberof tracheary element formations. Cytokinin concentrations aboveand below o.1 mg 1–1 interacting with an optimal xylogenicamount of auxin inhibited xylogenesis. The IAA (10 mg 1–1)-zeatin(0.1 mg 1–1) treatment produced the greatest number oftracheids, while kinetin compared to zeatin did not producesuch an effect. The different effectiveness of zeatin and kinetinin inducing tracheary element formations was not due to a differentcapacity of the two cytokinins to stimulate cell division butit seems likely that zeatin, because of interaction with IAA,is more active than kinetin in the determination of the dividingcells in a specific type of cytodifferentiation. The IAA (10mg 1–1)-zeatin (0.1 mg 1–1) treatment produced about6.9 per cent tracheids with respect to cell division while IAA(10 mg 1–1)-kinetin (0.1 mg 1–1) produced 4.2 percent. These results are discussed with reference to the problemsof hormonal control of xylem differentiation.  相似文献

The effect of three gibberellin synergists on lettuce hypocotylelongation was studied. Dihydroconiferyl alcohol isolated fromlettuce plants and (–)-(6S, 1'S)-pestalotin isolated fromPestalotia cryptomeriaecola enhanced the promoting effect ofgibberellic acid on hypocotyl elongation of lettuce seedlingswith and without the cotyledons. On the other hand, TA, a triazinonecompound, did not enhance the gibberellin effect. The actionof (–)-(6S, 1'S)-pestalotin was strongly inhibited bycompetitive inhibitors of dihydroconiferyl alcohol such as caffeic,ferulic and trans-cinnamic acids. Of the two stereoisomers ofpestalotin, (+)-(6R, 1'R)-pestalotin enhanced the gibberellineffect but (+)-(6R, 1'S)-epipestalotin did not. (+)-(6R, 1'S)-Epipestalotinstrongly inhibited the action of (–)-(6S, 1'S)-pestalotinand dihydroconiferyl alcohol. TA did not affect the action ofdihydroconiferyl alcohol. Stress-relaxation analysis of the mechanical properties of thelettuce hypocotyl cell wall demonstrated that gibberellic acidcaused cell wall loosening and dihydroconiferyl alcohol andpestalotin did not influence this gibberellin effect. The siphons and mantle edge of Donax serra and D. sordidus possesstwo types of ciliated sensory receptor. Type 1 has > 7 ciliawith an exposed length of 0.7–2.4 µm. Type II hasfewer cilia (2–5) which are 2–6 µm long. Athird type (Type III) described from the tips of the tentaclesof the siphon and mantle edge of D. sordidus, possesses twotufts of cilia which are 12 µm long. All three receptortypes appear to be primary receptors. Estimates of abundanceshow that receptors are most numerous on the tips of the siphontentacles (9.75 x 103. mm–2), and it is suggested thatthese receptors function as chemoreceptors.
