Characterization of organic matter microstructure dynamics during co-composting of sewage sludge, barks and green waste 总被引:1,自引:0,他引:1
A microstructure characterization study using transmission electron microscopy (TEM) was conducted to specify organic matter dynamics during the co-composting process of sewage sludge, green waste and barks. TEM results showed that ligneous and polyphenolic compounds brought by barks were not biodegraded during composting. Green waste brought more or less biodegraded ligneous constituents and also an active microbial potential. Chloroplasts and sludge flocs appeared to be relevant indicators of green waste and sewage sludge in composted products, as they were still viewable at the end of the process. TEM characterization of the final product highlighted two main fractions of organic matter, one easily available and a more recalcitrant one, and also a remaining microbial activity. Thus microstructure characterization appeared to be an appropriate way of taking the heterogeneity of the organic constituents' size and composition into account when attempting to specify such compost quality parameters as maturity and stability. 相似文献

Humic substances change during the co-composting process of municipal solid wastes and sewage sludge

The change of the degree of stability of compost during the composting process was a kind of guideline for our study. This
stability was estimated by monitoring the chemical fractionation (extraction of humic and fulvic acids, and humin) during
two cycles of composting. Change of humin (H), humic-like acid carbon (CHA) and fulvic-like acid carbon (CFA) fractions during
the composting process of municipal solid wastes were investigated using two windrows W1 (100% of municipal solid wastes)
and W2 (60% of municipal solid wastes and 40% of dried sewage sludge). Humin and fulvic acid fractions in the two windrows
decreased since the start of composting process and tend to stabilize. At the end of composting process, humic acid fraction
is more important in the windrow without sludge (W1) than the one with sludge (W2). The humification indexes used in this
study showed that the humic-like acid carbon fraction production takes place largely during the phase of temperature increase
(thermophilic phase), and it appeared very active in the windrow W2. At the end of composting process, the E4/E6 ratio value indicated that the compost of W1 is more mature than the compost of W2. The humification ratio (HR) allowed a

In order to assess effects of drying method on chemical composition and NIR spectra of pasture silage, 20 samples of silages from permanent pastures of different qualities were subsampled and dried either by forced draught oven at 65°C or freeze-dried. Samples were analysed for crude protein (CP), crude fibre (CF), neutral detergent fibre (NDF), acid detergent fibre (ADF), gross energy (GE) and in vitro digestible organic matter in the dry matter (DOMD) and composition expressed on a dry-matter (DM) basis, either obtained as oven 105°C DM (DM105°) or toluene DM (DMtol). Effects of the drying method on chemical composition were estimated by paired comparisons. Near-infrared reflectance (NIR) spectra were taken and the difference spectra between treatments for each sample were plotted in order to visually inspect effects of treatments, either as log 1/R or transformed by their first derivative. Principal components explaining spectral variability were computed and samples graphically displayed according to the eigenvalues of the first (1 and 2), or second (2 and 3) pairs of principal components. Oven-drying resulted in a reduction in CP (p<0.02) content and an increase in CF (p<0.1), NDF (p<0.001) and ADF (p<0.06) content of silages, when expressed on a DM105° basis. When expressed on a DMtol basis, a reduction in CP (p<0.02) and DOMD (p<0.06) content was observed with oven drying, but fibre fractions were not affected (p>0.1). Effects of the drying treatment were visually apparent on NIR spectra when plotted as log 1/R against wavelengths, mainly as a baseline shift. Difference spectra (OD − FD), both as log 1/R and its first derivative, showed consistent absorption bands, indicative of different molecular responses of OD and FD silages to incident light. Plotting samples according to eigenvalues of the first two or second two principal components suggests that drying methods affect the distribution of samples. This is also indicative of an effect of drying treatment on spectral features and reinforces the general advice that sample processing should be consistent in order to avoid adding errors to NIR analysis. 相似文献

Influence of humic substances on bioavailability of Cu and Zn during sewage sludge composting 总被引:4,自引:0,他引:4
Influence of humic substances (HS) on bioavailability of Cu and Zn was characterized during 120 days co-composting of sewage sludge and maize straw. At the initial stage of composting, Cu and Zn in sewage sludge were released as organic matter was degraded, and water soluble Cu and Zn increased markedly. Water soluble Cu and FA content decreased after 21 days whereas water soluble Zn increased during the whole process. Both HA-Cu and HA-Zn were significantly and positively correlated with HA and H/F, respectively. At the end of composting, the distribution coefficients of HA-Cu and HA-Zn reached 27.50% and 3.33% respectively with HA-Cu/HA-Zn ratio increased from 1.29 to 2.73. The results suggest that Cu combined with HA more strongly than Zn, and composting treatment could decrease bioavailability of Cu markedly. 相似文献

Negative correlations between corn vitreousness and ruminal dry matter and starch degradabilities have been widely reported. To measure corn vitreousness and density more rapidly, Correa et al. [Correa, C.E.S., Shaver, R.D., Pereira, M.N., Lauer, J.G., Kohn, K., 2002. Relationship between corn vitreousness and ruminal in-situ starch degradability. J. Dairy Sci. 85, 3008–3012] initiated the development of near-infrared reflectance spectroscopy (NIRS) calibrations from 47 samples derived from 14 US and five Brazilian commercial hybrids. In this study, we generated more data to add to these NIRS calibrations with the objective of making them more robust. We also evaluated the potential of using Stenvert hardness measurements for NIR calibrations. Thirty-three diverse corn germplasm sources were grown at University of Wisconsin West Madison Research Station. These included a wide range of endosperm characteristics from opaque 2 (o2) types to densely packed flint types, and a number of intermediates. Harvest was at 1/2 milkline and black-layer maturity stages. Dried kernels from middle portions of ears from 12 selected inbreds, four each from low (0–30%), medium (30–70%), and high (70–100%) vitreousness classifications were used to determine vitreousness by manual dissection and density by water displacement using a pycnometer. Hardness was determined on all 33 inbreds on a 20 g sample using a Stenvert micro hammer-cutter mill with 2 mm screen size and 3600 rpm to measure time to collect ground sample to a set receptacle height (T); total column height (CH); and height ratio of coarse to fine (C/F) particles. The NIRS equations were selected on the basis of high R2-values (0.90, 0.92, 0.85, and 0.85) and low SEC (4.85, 0.01, 1.39, and 0.19) and SECV (6.04, 0.02, 1.79, and 0.25), for vitroueness, density, T and CH factors, respectively. Calibrations for vitreousness and density were regarded as the best prediction models compared to stenvert hardness measurements as determined by their RPD values (3.73 and 2.50, respectively). These results show that NIRS can be used as a screening tool in large-scale breeding trials to develop corn hybrids of desired endosperm properties for improved ruminal degradabilities. 相似文献

Using resonance light scattering and UV/vis absorption spectroscopy to study the interaction between gliclazide and bovine serum albumin 下载免费PDF全文
At different temperatures (298, 310 and 318 K), the interaction between gliclazide and bovine serum albumin (BSA) was investigated using fluorescence quenching spectroscopy, resonance light scattering spectroscopy and UV/vis absorption spectroscopy. The first method studied changes in the fluorescence of BSA on addition of gliclazide, and the latter two methods studied the spectral change in gliclazide while BSA was being added. The results indicated that the quenching mechanism between BSA and gliclazide was static. The binding constant (Ka), number of binding sites (n), thermodynamic parameters, binding forces and Hill's coefficient were calculated at three temperatures. Values for the binding constant obtained using resonance light scattering and UV/vis absorption spectroscopy were much greater than those obtained from fluorescence quenching spectroscopy, indicating that methods monitoring gliclazide were more accurate and reasonable. In addition, the results suggest that other residues are involved in the reaction and the mode ‘point to surface’ existed in the interaction between BSA and gliclazide.
