Coccidioidomycosis is a systemic disease caused by the dimorphic fungus Coccidioides, endemic in parts of the Southwestern USA and Central and South America. Two species, Coccidioides immitis and Coccidioides posadasii, were differentiated. Primary cutaneous coccidioidomycosis (PCC) has been reported rarely. An unusual case of PCC characterized by a persistent solitary lesion diagnosed in Italy in an immunocompetent Italian nun living in Argentina is described. The isolate was identified by sequence analysis as C. posadasii. Antibody screening was negative. A total of 39 cases of PCC have been reported in the literature. Infections occurred as a consequence of traumatic implantation in a natural setting in endemic areas or of accidental inoculation in laboratory workers. Importance of accurate investigation of travel history and of occupational hazards to laboratory workers is outlined.  相似文献

Work was carried out on pea (Pisum sativum L.) seedling roots to assess the attachment of the nitrogen-fixing symbiotic bacteria Rhizobium leguminosarum bv. vicea (Rlv) and the bacterial phytopathogens—specific Pseudomonas syringae pv. pisi (Psp) and nonspecific Clavibacter michiganensis ssp. sepedonicus (Cms). Different root zones were examined: (I) the meristem, 2 mm from the root tip; (II) the root hair-free zone, 27 mm; (III) the zone of root hair anlages, 712 mm; (IV) the young root hair zone, 1217 mm; and (V) the zone of root hair that completed the growth, 1722 mm. It was found earlier that the zones differed in their susceptibility to Rlv. In the present work, reactions of particular components of the adenylate cyclase signaling system (ACSS) were estimated, i.e., concentration of cAMP and activities of transmembrane adenylate cyclase (tAC) and soluble adenylate cyclase (sAC) in these zones after different times post inoculation (5, 15, 120, and 360 min). It was revealed that the degree of activation of particular components of ACSS did not depend on the sorption rate of differently specialized bacteria. Upon contact with Rlv, the character of changes in tAC and sAC activities was almost the same in different root zones and resembled the dynamics of the cAMP content. Inoculation with Psp changed the cAMP level similarly to that with Rlv, but the dynamics of tAC and sAC was opposite to each other in most cases. Inoculation with Cms, in spite of the absence of its attachment, elevated the cAMP content and activated tAC and sAC. It is suggested that the above-mentioned changes in ACSS is associated with exometabolites of Rlv, Psp, and Cms, which activate the PAMP-induced immunity of the pea seedling cells. The uniform dynamics of cAMP in different root zones upon the exposure to Rlv and Psp seems to reflect the specific reaction and, presumably, fulfills different functions—regulatory with Rlv and defensive with Psp. Upon short-term contact with Cms, the cAMP dynamics in the same root zones displayed a nonspecific character that might be related to the rate of adsorption of exopolysaccharides by the root hair. The systemic response of ACSS was observed in the hypocotyls of the seedlings exposed to any of the three organisms.  相似文献

Paracoccidioidomycosis ceti is a cutaneous disease of cetaceans caused by uncultivated Paracoccidioides brasiliensis or Paracoccidioides spp. Serological cross-reactions between paracoccidioidomycosis ceti and paracoccidioidomycosis, paracoccidioidomycosis and histoplasmosis, and paracoccidioidomycosis and coccidioidomycosis have been reported before. The present study aimed to detect immunohistochemical cross-reaction between antibodies to Paracoccidioides sp. and Histoplasma capsulatum, and vice versa. Thirty murine sera, obtained from experimental infections of 6 isolates of H. capsulatum, were reacted with paraffin-embedded yeast-form cells of Paracoccidioides sp. derived from a case of paracoccidioidomycosis ceti in Japan. The murine sera were also reacted with human isolates of H. capsulatum yeast cells, with P. brasiliensis yeast cells, and with fungal cells of Coccidioides posadasii. Three dolphins’ sera from cases of paracoccidioidomycosis ceti, two human sera from patients with paracoccidioidomycosis, and a serum from a healthy person with a history of coccidioidomycosis were used in order to determine that the tested fungal cells reacted properly. Sera derived from mice infected with an isolate of H. capsulatum reacted positively against yeast cells of Paracoccidioides sp., yeast cells of P. brasiliensis, and fungal cells of C. posadasii, while those derived from other strains were negative. The present study recorded for the first time the cross-reaction between the yeast cells of H. capsulatum and antibodies against Paracoccidioides spp., the yeast cells of Paracoccidioides sp. and antibodies against H. capsulatum, the yeast cells of Paracoccidioides sp. and antibodies against Coccidioides sp., and fungal cells of C. posadasii and antibodies against Paracoccidioides spp.  相似文献

Aspergillus spp. are emerging causative agents of non-dermatophyte mould onychomycosis (NDMO). New Aspergillus spp. have recently been described to cause nail infections. The following criteria are required to diagnose onychomycosis due to Aspergillus spp.: (1) positive direct microscopy and (2) repeated culture or molecular detection of Aspergillus spp., provided no dermatophyte was isolated. A review of 42 epidemiological studies showed that onychomycosis due to Aspergillus spp. varies between < 1 and 35% of all cases of onychomycosis in the general population and higher among diabetic populations accounting for up to 71% and the elderly; it is very uncommon among children and adolescence. Aspergillus spp. constitutes 7.7–100% of the proportion of NDMO. The toenails are involved 25 times more frequently than fingernails. A. flavus, A. terreus and A. niger are the most common aetiologic species; other rare and emerging species described include A. tubingensis, A. sydowii, A. alliaceus, A. candidus, A. versicolor, A. unguis, A. persii, A. sclerotiorum, A. uvarum, A. melleus, A. tamarii and A. nomius. The clinical presentation of onychomycosis due to Aspergillus spp. is non-specific but commonly distal–lateral pattern of onychomycosis. A negative culture with a positive KOH may point to a NDM including Aspergillus spp., as the causative agent of onychomycosis. Treatment consists of systemic therapy with terbinafine or itraconazole.  相似文献

Coccidioidomycosis is a fungal disease caused by Coccidioides immitis or Coccidioides posadasii. These fungi are endemic in the southern USA and northern Mexico. Immunocompromised patients are susceptible to develop severe forms of this fungal infection. Cytokines play an important role in controlling the fungal infection, but little is known about the predominant immunological environment in human lung tissue from fatal cases. Our aim was to analyze the pro-inflammatory and anti-inflammatory cytokines and monocyte/macrophages markers (CD14 and CD206) in the granulomas of six fatal cases of coccidioidomycosis. Cytokines and surface markers were higher in coccidioidomycosis cases when compared to control (P?<?0.05). CD14 positive cells were increased inside the coccidioidal granuloma when compared to the outside (P?<?0.05). No differences were found in the number of CD206+ cells inside the granuloma when compared to the outer population (P?>?0.05). Interestingly, an analysis of stain intensity signals showed an increased signaling of CD14, CD206, IL-10 and TNFα inside the granuloma when compared to the outside (P?<?0.05). iNOS and IL-12 gene expression were not detected in coccidioidomycosis cases, while IL-10, IL-6 and TGFβ gene expression were detected, but the differences when compared to healthy lungs were not significant (P?>?0.05). TNFα gene expression was lower in coccidioidomycosis cases when compared to healthy lung (P?=?0.05). In conclusion, pro- and anti-inflammatory responses co-exist inside of the granulomas of fatal cases of coccidioidomycosis and the absent of iNOS and IL-12 gene expression may be related with patient’s outcome.  相似文献

Staphylococcus aureus is responsible for significant and increasing number of hospital-and community-acquired infections worldwide. A pool of pathogenesis factors helps the bacterium to cause the range of mild to severe infections leading the high mortality and morbidity. Staphylococcus aureus and Candida albicans can be co-isolated from all human mucosal sites and are responsible for diverse infections. Vaccine design for related polymicrobial infections should consider the consortia of microorganisms responsible for the disease. In this study we considered biofilm mode of growth and polymicrobial nature of the infections caused by S. aureus. In the first phase of study the prediction of putative antigenic targets of S. aureus and C. albicans was conducted based on data mining and bioinformatic characterization of their proteins. Various properties of proteins were evaluated such as subcellular localization, hydrophilicity, repeat containing modules, beta turns, surface accessibility and number of antigenic determinants. The second phase includes various immunoinformatics analyses on six proteins include ALS, ClfA, FtmB, SdrE, Spa and Bap leading to design a novel sub-unit hexavalent vaccine. Several potential T cell and B-cell epitopes are present in our vaccine. Also the vaccine is expected to strongly induce IFN-gamma production. The amino acid sequence introduced here is expected to enhance cell-mediated and humoral responses against S. aureus biofilm-related infections to clear biofilm communities of S. aureus and intracellular colonies of pathogen as well as planktonic cells and thus reduces colonization and persistence.  相似文献

Bacterial–bacteriophage interactions are a well-studied and ecologically-important aspect of microbiology. Many commercial fermentation processes are susceptible to bacteriophage infections due to the use of high-density, clonal cell populations. Lytic infections of bacterial cells in these fermentations are especially problematic due to their negative impacts on product quality, asset utilization, and fouling of downstream equipment. Here, we report the isolation and characterization of a novel lytic bacteriophage, referred to as bacteriophage DTL that is capable of rapid lytic infections of an Escherichia coli K12 strain used for commercial production of 1,3-propanediol (PDO). The bacteriophage genome was sequenced and annotated, which identified 67 potential open-reading frames (ORF). The tail fiber ORF, the largest in the genome, was most closely related to bacteriophage RTP, a T1-like bacteriophage reported from a commercial E. coli fermentation process in Germany. To eliminate virulence, both a fully functional Streptococcus thermophilus CRISPR3 plasmid and a customized S. thermophilus CRISPR3 plasmid with disabled spacer acquisition elements and seven spacers targeting the bacteriophage DTL genome were constructed. Both plasmids were separately integrated into a PDO production strain, which was subsequently infected with bacteriophage DTL. The native S. thermophilus CRISPR3 operon was shown to decrease phage susceptibility by approximately 96%, while the customized CRISPR3 operon provided complete resistance to bacteriophage DTL. The results indicate that the heterologous bacteriophage-resistance system described herein is useful in eliminating lytic infections of bacteriophage DTL, which was prevalent in environment surrounding the manufacturing facility.  相似文献

Cystic fibrosis (CF) is a genetic inherited disease due to mutations in the gene cystic fibrosis transmembrane conductance regulator (CFTR). Because of the huge diversity of CFTR mutations, the CF phenotypes are highly heterogeneous, varying from typical to mild form of CF, also called atypical CF. These atypical features are more frequently diagnosed at adolescence or adulthood, and among clinical signs and symptoms leading to suspect a mild form of CF, colonization or infection of the respiratory tract due to well-known CF pathogens should be a warning signal. Exophiala dermatitidis is a melanized dimorphic fungus commonly detected in respiratory specimens from CF patients, but only very rarely from respiratory specimens from non-CF patients. We described here two cases of chronic colonization of the airways by E. dermatitidis, with recurrent pneumonia and hemoptysis in one patient, which led clinicians to diagnose mild forms of CF in these elderly patients who were 68- and 87-year-old. These cases of late CF diagnosis suggest that airway colonization or respiratory infections due to E. dermatitidis in patients with bronchiectasis should led to search for a mild form of CF, regardless of the age and associated symptoms. On a broader level, in patients with chronic respiratory disease and recurrent pulmonary infections, an allergic bronchopulmonary mycosis or an airway colonization by CF-related fungi like E. dermatitidis or some Aspergillus, Scedosporium or Rasamsonia species, should be considered as potential markers of atypical CF and should led clinicians to conduct investigations for CF diagnosis.  相似文献

Diversity of Ukrainian winter common wheat varieties was studied with respect to the storage protein loci Gli-A1, Gli-B1, Gli-D1, Glu-A1, Glu-B1, Glu-D1, Gli-A3, Gli-B5, and Gli-A6 (362 varieties) and markers for the Lr34/Yr18/Pm38/Sr57/Bdv1 gene conferring moderate resistance to a number of biotrophic pathogens, the Tsn1 gene for sensitivity to the toxins A of the necrotrophic fungi Pyrenophora tritici-repentis and Stagonospora nodorum, the Tsc2 gene for sensitivity to the toxin B of P. tritici-repentis, and the TDF_076_2D gene for moderate resistance to Fusarium head blight (181 varieties). Significant differences in frequencies of alleles at these marker loci between groups of varieties developed in different soil and climatic zones were revealed. The retention of a set of predominant alleles in groups of varieties of a certain zone in different periods of breeding was confirmed. At the same time, the appearance of new allele associations in the groups of varieties of the Steppe (in particular Gli-A1g and Glu-B1al) and the Central Forest-Steppe (1AL/1RS and Glu-B1d) in the last two decades has been noted. Nonrandom associations between alleles of disease resistance genes as well as alleles of disease resistance genes and storage protein alleles were revealed.  相似文献

The Early Triassic is a critical interval for the study of recovery from the terminal Permian mass extinction, as there are small-scale extinction events, which may have contributed to the delayed recovery. The systematic measuring and sampling of a 12-m-thick section at the Mokrice locality in eastern Slovenia has resulted in the recovery of a conodont fauna from the Olenekian beds. Four conodont zones have been recognized. These zones are in ascending order as follows: the Hadrodontina aequabilis Zone, Platyvillosus corniger Zone, Platyvillosus regularis Zone, and Triassospathodus hungaricus Zone. These conodont zones confirm the proposed conodont biozonation sequence in western Slovenia and have correlation value especially for the western marginal Tethys. Multielement conodont apparatuses of Triassospathodus hungaricus and Platyvillosus regularis have been reconstructed based on conodont elements that were recently obtained from the Slovenian sections. Although the S2element was not found, the apparatus indicates that the conodont species “Spathognathodushungaricus should be assigned to the genus Triassospathodus.  相似文献

Background

Tsetse flies are vectors of African trypanosomes, protozoan parasites that cause sleeping sickness (or human African trypanosomosis) in humans and nagana (or animal African trypanosomosis) in livestock. In addition to trypanosomes, four symbiotic bacteria Wigglesworthia glossinidia, Sodalis glossinidius, Wolbachia, Spiroplasma and one pathogen, the salivary gland hypertrophy virus (SGHV), have been reported in different tsetse species. We evaluated the prevalence and coinfection dynamics between Wolbachia, trypanosomes, and SGHV in four tsetse species (Glossina palpalis gambiensis, G. tachinoides, G. morsitans submorsitans, and G. medicorum) that were collected between 2008 and 2015 from 46 geographical locations in West Africa, i.e. Burkina Faso, Mali, Ghana, Guinea, and Senegal.

Results

The results indicated an overall low prevalence of SGHV and Wolbachia and a high prevalence of trypanosomes in the sampled wild tsetse populations. The prevalence of all three infections varied among tsetse species and sample origin. The highest trypanosome prevalence was found in Glossina tachinoides (61.1%) from Ghana and in Glossina palpalis gambiensis (43.7%) from Senegal. The trypanosome prevalence in the four species from Burkina Faso was lower, i.e. 39.6% in Glossina medicorum, 18.08%; in Glossina morsitans submorsitans, 16.8%; in Glossina tachinoides and 10.5% in Glossina palpalis gambiensis. The trypanosome prevalence in Glossina palpalis gambiensis was lowest in Mali (6.9%) and Guinea (2.2%). The prevalence of SGHV and Wolbachia was very low irrespective of location or tsetse species with an average of 1.7% for SGHV and 1.0% for Wolbachia. In some cases, mixed infections with different trypanosome species were detected. The highest prevalence of coinfection was Trypanosoma vivax and other Trypanosoma species (9.5%) followed by coinfection of T. congolense with other trypanosomes (7.5%). The prevalence of coinfection of T. vivax and T. congolense was (1.0%) and no mixed infection of trypanosomes, SGHV and Wolbachia was detected.

Conclusion

The results indicated a high rate of trypanosome infection in tsetse wild populations in West African countries but lower infection rate of both Wolbachia and SGHV. Double or triple mixed trypanosome infections were found. In addition, mixed trypanosome and SGHV infections existed however no mixed infections of trypanosome and/or SGHV with Wolbachia were found.

Fermentation is one of the oldest ways of processing food. Some fermented food is produced industrially, but can also be produced in an artisanal way by certain ethnic groups, called traditional fermented foods. In Mexico, there are a variety of traditional fermented beverages which are produced in an artisanal way. They include those made with maize (atole agrio, pozol, and tesgüino), fruit (tepache and colonche), and obtained by plant fermentation (pulque, tuba, and taberna). These beverages have been used since ancient times for religious and medicinal purposes. The medicinal effect may be due to fermented microorganisms. The presence of beneficial microorganisms known as probiotics provides beneficial effects to consumer health, improving the balance of intestinal host, and reducing the risk of gastrointestinal diseases, mainly. Most probiotics belong to the genus Lactobacillus, but Bifidobacterium, Bacillus, and yeast are also found. Therefore, it is important that the microbiological diversity of the beverages is studied and documented. This review includes information on the microbial diversity and probiotic potential of the most important traditional fermented beverages from Mexico.  相似文献

The array of outer membrane proteins (OMPs) found in Helicobacter pylori provides a crucial component for persistent colonization within the gastric niche. Not only does H. pylori harbor a wide number of OMPs, but these OMPs often vary across strains; this likely contributes to immune evasion, adaptation during long term colonization, and potentially differential disease progression. Previous work from our group described OMP differences among the Bab family (babA, babB, and babC) and Hom family (homA and homB) from 80 American H. pylori clinical isolates (AH) and 80 South Korean H. pylori clinical isolates (KH). In the current study, we expanded our investigation to include the less well characterized Hom family member, HomC.  相似文献

Neoscytalidium dimidiatum is an opportunistic fungus causing cutaneous infections mostly, which are difficult to treat due to antifungal resistance. In Malaysia, N. dimidiatum is associated with skin and nail infections, especially in the elderly. These infections may be mistaken for dermatophyte infections due to similar clinical appearance. In this study, Neoscytalidium isolates from cutaneous specimens, identified using morphological and molecular methods (28 Neoscytalidium dimidiatum and 1 Neoscytalidium sp.), were evaluated for susceptibility towards antifungal agents using the CLSI broth microdilution (M38-A2) and Etest methods. Amphotericin B, voriconazole, miconazole and clotrimazole showed high in vitro activity against all isolates with MIC ranging from 0.0313 to 1 µg/mL. Susceptibility towards fluconazole and itraconazole was noted in up to 10% of isolates, while ketoconazole was inactive against all isolates. Clinical breakpoints for antifungal drugs are not yet available for most filamentous fungi, including Neoscytalidium species. However, the results indicate that clinical isolates of N. dimidiatum in Malaysia were sensitive towards miconazole, clotrimazole, voriconazole and amphotericin B, in vitro.  相似文献

Hemolytic factor production by pathogenic Candida species is considered an important attribute in promoting survival within the mammal host through the ability to assimilate iron from the hemoglobin-heme group. Hemolytic capability has been evaluated for Candida species based on hemolysis zones on plate assay, analysis of hemolytic activity in liquid culture medium, and hemolysis from cell-free culture broth. The production of hemolytic factor is variable among Candida species, where C. parapsilosis is the less hemolytic species. In general, no intraspecies differences in beta-hemolytic activities are found among isolates belonging to C. albicans, C. glabrata, C. krusei, C. tropicalis, and C. parapsilosis. The production of hemolytic factor by Candida species is affected by several factors such as glucose supplementation in the culture medium, blood source, presence of erythrocytes and hemoglobin, and presence of electrolytes. On the basis of existing achievements, more researches are still needed in order to extend our knowledge about the biochemical nature of hemolytic molecules produced by distinct Candida species, the mechanism of hemolysis, and the molecular basis of the hemolytic factor expression.  相似文献

Abstract

Mangrove forests have high net primary productivity (NPP), and it is well known that these trees allocate high amounts of biomass to their root systems. In particular, fine root production (FRP) comprises a large component of the NPP. However, information on root production remains scarce. We studied FRP in three zones (Avicennia, Rhizophora, and Xylocarpus) of a mangrove forest in eastern Thailand using ingrowth cores (0–30 cm of soil depth). The root biomass and necromass were periodically harvested from the cores and weighed during the one-year study. The FRP was determined by summation of the fine root biomass (FRB) and root necromass. The results showed that the FRB clearly increased in the wet and cool dry seasons. Magnitude of FRB in the Rhizophora and Xylocarpus zones was 1171.07 and 764.23 g/m2/30 cm, respectively. The lowest FRB (292.74 g/m2/30 cm) was recorded in the Avicennia zone locating on the river edge where there is a greater frequency of inundation than the other zones. Root necromass was high in the Rhizophora and Xylocarpus zones, and accumulated noticeably when soil temperatures rapidly declined during the middle of the wet season to cool dry season. However, root necromass in the Avicennia zone varied within a small range. We attributed the small accumulation of root necromass in the Avicennia zone to the relative high soil temperature that likely caused a high root decomposition rate. The average FRP (3.403–4.079 ton/ha/year) accounted for 74.4, 81.5, and 92.4 % of the total root production in the Avicennia, Rhizophora, and Xylocarpus zone, respectively. The root production and causative factors (i.e., soil temperature and inundation regime) are discussed in relation to the carbon cycle of a mangrove forest.

Bulrushes of the genus Bolboschoenus are robust, ecologically important sedges occurring in wetlands, including intertidal marshes and mudflats. Despite their importance and multiple serious threats to their habitats, estuarine Bolboschoenus species remain poorly known. We conducted herbarium and field research in order to document historic and current geographic distributions, characterize the habitats, and assess the conservation status of Bolboschoenus species in the Hudson River Estuary, New York, U.S.A. Three species of Bolboschoenus grow in intertidal zones in the Hudson Estuary. Bolboschoenus fluviatilis occurs in the northern, upstream, and freshwater portion of the estuary with multi-year mean surface salinities of 0.078–2.0 ppt. Bolboschoenus robustus occupies the southernmost, downstream, and brackish to saline portion of the estuary with salinities of 4.9–16 ppt. Bolboschoenus novae-angliae occurs in the slightly to strongly brackish region between the other two species with salinities of 1.8–8.0 ppt. The geographic ranges of B. fluviatilis and B. robustus do not overlap, but B. novae-angliae has short zones of sympatry with each of the other two species. Syntopy of B. novae-angliae with each of the other two species is rare. In the Hudson Estuary, B. fluviatilis is secure, but B. novae-angliae and B. robustus are critically imperiled. Threats to future survival of Bolboschoenus species in the Hudson Estuary include competition from invasive plant species (especially Phragmites australis), eutrophication resulting from excess nutrient pollution, and habitat destruction. Our data and analyses provide critical new information for management of existing environmental problems and planned habitat restoration efforts in the Hudson River Estuary.  相似文献

Trichophyton rubrum is a common human dermatophyte that is the causative agent of 80–93% of fungal infections of the skin and nails. While dermatophyte infections in healthy people are easily treatable with over-the-counter medications, such infections pose a higher risk for patients with compromised immune function and impaired regenerative potential. The efficacy of boric acid (BA) for the treatment of vaginal yeast infections prompted an investigation of the effect of BA on growth and morphology of T. rubrum. This is of particular interest since BA facilitates wound healing, raising the possibility that treating athlete’s foot with BA, either alone or in combination with other antifungal drugs, would combine the benefits of antimicrobial activity and tissue regeneration to accelerate healing of infected skin. The data presented here show that BA represses T. rubrum growth at a concentration reported to be beneficial for host tissue regeneration. Oxygen exposure increases BA toxicity, and mycelia growing under BA stress avoid colonizing the surface of the growth surface, which leads to a suppression of aerial mycelium growth and surface conidia formation. BA penetrates into solid agar matrices, but the relative lack of oxygen below the substrate surface limits the effectiveness of BA in suppressing growth of embedded T. rubrum cells.  相似文献

The subgenus Pocusogetus Rtt. of the genus Otiorhynchus Germ. is revised. The subgenus includes O. rosti Strl., O. shapovalovi Davidian et Yunakov, O. obsulcatus Strl., O. fischtensis Rtt., and O. gusakovi sp. n. closely related to O. fischtensis (both from Mt. Fisht, the Western Caucasus). O. fischtensis is transferred from the subgenus Vicoranius Rtt., its lectotype is designated. A key to species of Pocusogetus is given. The systematic position of the subgenera Pocusogetus and Vicoranius in the genus Otiorhynchus is discussed. New data on the geographical distribution and ecology of the little-known species of the subgenera Obvoderus Rtt., Pseudoprovadilus Magnano, and Clypeorhynchus Yunakov et Arzanov are given. Some features of ecological differentiation between Otiorhynchus species in the alpine and subalpine zones of the Caucasus are discussed.  相似文献
