During the auxin-sensitive phase of root initiation, rates of 3-indolyl- [2-14C] acetic acid (IAA) uptake into the 1 cm bases of shoots of the apple rootstock M.9 ( Malus pumila Mill.) 'in vitro' were not significantly affected by the presence of 10−3 M phloroglucinol (PG) using either liquid or agar-solidified media. The use of a liquid medium did however reduce rates of uptake over a 10-day period of auxin application. The distribution of labelled IAA between the 1-cm base and the shoot remainder was not affected by PG.
Exposure of shoots of the difficult-to-root M.9 and the easy-to-root M.26, to 2.8 × 10−5 M IAA containing [2-14C] IAA revealed no positive correlation between the amount of label taken up by the 1-cm base and rooting performance. M.9 bases absorbed almost twice as much label as M.26 after 9 days but had produced only one-third as many roots. Measurements of label distribution between the 1-cm base and the shoot remainder showed that less than 10% of the label moved to the shoot remainder over a 6-day period of auxin application. Dose-response curves of IAA and rooting over the range 1 × 10−5 M and 3 × 10−3 M showed that root number in M.9 was at an optimum at 1 × 10−3 M IAA after 6 days whilst M.26 required only 1 × 10−4 M for a similar response. These data support the hypothesis that differences in rooting of the two rootstocks reflect differences in the endogenous metabolism of exogenous IAA and not differences in its rates of uptake or distribution in the shoots.  相似文献

The behaviour of juvenile Arctic charr, Salvelinus alpinus (L.) , to an abrupt concentration step of L-amino acids, L-alanine and ammonium chloride was studied by fluviarium technique. The emission rates of these substances were studied. Juvenile Arctic charr emit 8.0 × 10−4 mol total ammonia-N kg−1 h−1 and 3.3 × I0−5 mol amino acids kg−1 h−1. In behaviour tests the charr avoided 5.6x 10−6and 5.6 × 10−7 M ammonium chloride. The 17 L-amino acid mixture, ranked as observed in the analysis of emission, was avoided at 4.6 × 10−7 M, while 100 times dilution of this value gave neither avoidance nor attraction. The charr avoided L-alanine tested alone at the concentration of 4.6 × 10 −7 M. Anosmic charr showed neither avoidance nor attraction to the mixture of 17 amino acids tested at 4.6 × 10−7 M. The results indicate that ammonia as well as emitted amino acids are not responsible for the olfactory mediated attraction to conspecific odour shown earlier in Arctic charr. On the contrary, these substances may have a negative effect by reducing the strength of attraction.  相似文献

In white spruce ( Picea glauca ) protoplasts, abscisic acid (ABA) and optically pure ABA analogs induced expression of a reporter gene under regulation of a wheat ABA-responsive promoter. A fusion of a 650 bp promoter fragment from the wheat Em gene promoter and the Escherichia coli uidA sequence encoding β -glucuronidase (GUS) was linked in the plasmid pBM 113Kp. Expression of the Em-uidA fusion varied among 6 white spruce genotypes. Protoplasts from 4-day-old embryogenic suspension cultures gave the highest GUS activity relative 10 other stages in the 7-day growth cycle of suspension cultures. Racemic ABA [R.S-(±)-ABA] induced a significant increase of protoplast GUS activity over background at a concentration of 1 × 10−5 M , but maximum GUS activity was found at 1 × 10−3 M , ABA stereochemistry had a significant effect on gene expression. The natural isomer of ABA [S-(+)-ABA] was an effective inducer at a concentration as low as 1 × 10−7 M , but a concentration of greater than 1 × 10−4 M was required for induction by [R-(—)-ABA]. Moreover, analogs with the same configuration at C-l1 as that of natural ABA were more effective for induction of expression from the Em-uidA . insert at 1 × 10−4 M than were their enamiomers. Plasnud pBI511. carrying the chloramphenicol acety] transferase (CAT) gene driven by the constitutively expressed, tandemly duplicated cauliflower mosaic virus 35S promoter, was co-electroporated with pBM113Kp for monitoring Ihe influence of addition of exogenous ABA or ABA analogs on heterologous gene expression in protoplasts. The apple rootstock A2 can be readily propagated in vitro both in the juvenile and in the adult growth phase. Shoots were produced by meristem tip culture from the apple rootstock A2 in different growth phases. The influence of growth phases and different concentrations of PG and IBA was investigated as to rooting percentage, survival percentage, number of roots per rooted shoot, root length, shoot length and formation of callus. IBA at 15 μ M without PG gave a significantly lower rooting percentage than 5 and 10 μ M IBA. PG together with IBA stimulated rooting, the optimum concentrations of PG being, however, not the same for the different growth phases. For the adult growth phase, 10−4 M PG promoted rooting, whereas 10−3 M PG markedly inhibited rooting. In the juvenile growth phases, both 10−4 and 10−3 M PG stimulated rooting. PG at 10−4 M also increased the number of roots. The longest roots were obtained at 10−3 M PG and 5 μ M IBA. PG at 10−3 M reduced callus formation at all IBA concentrations used. Neither shoot length nor root length influenced the survival percentage.  相似文献

Sea urchin embryos, which were treated with 5 × 10−3 M chloramphenicol for 1 to 4 hr at certain stages before hatching, developed to several types of abnormal embryos. No significant effect on the shape of the embryo was observed when the concentrations of chloramphenicol used in the short-period treatment were lower than 2 × 10−3 M. Embryos up to the 2-cell stage, treated with 5 × 10−3 M chloramphenicol for a short period, became small blastulae filled with mesenchyme-like cells (type A). A similar effect of puromycin (2 μg/ml) was also observed at this stage. When the chloramphenicol treatment (for 1 to 4 hr) was applied at 8 ∽ 32-cell stages, vegetalized larvae were produced (type B). Embryos treated with chloramphenicol at 7 hr after insemination at 20°C, developed to another type of abnormal larva different from the previous types (type C). A concentration of puromycin (2 μg/ml) which inhibited protein synthesis to the same degree as 5 × 10−3 M chloramphenicol, induced only type A. Between these chloramphenicol-sensitive stages, there were chloramphenicol-insensitive stages for forming abnormal embryos.  相似文献

Abstract: The effects of the proadrenomedullin N-terminal 20-amino acid peptide (PAMP) on the nicotinic acetylcholine (ACh) receptor (nAChR)-mediated inward current were investigated in neurons acutely dissociated from the rat locus coeruleus using whole-cell recording under voltage clamp. Nicotine and cytidine mimicked the ACh response, whereas the maximal response to dimethyl-phenylpiperazinium was lower in amplitude compared with that to ACh. Nicotine-induced current ( I nic) was suppressed more effectively by mecamylamine than by hexamethonium. In addition, neither atropine nor α-bungarotoxin affected the I nic. PAMP reversibly and noncompetitively suppressed the peak amplitude of 10−4 M I nic. PAMP concentrations for the threshold, half-maximal inhibition, and maximal inhibition of 10−4 M I nic were 10−8, 2.6 × 10−7, and 10−5 M , respectively. The peak amplitudes of 10−4 M I nic elicited at 2-min intervals showed a gradual decline in the presence of 10−7 M PAMP. This decline in the I nic was independent of the period of PAMP pretreatment. The suppression of I nic by PAMP did not show any voltage dependency at a holding potential ( V H) of <0 mV, although the inhibitory effect was masked by the marked inward rectification of I nic at a V H of 0 mV. These results suggest that PAMP could thus be a unique endogenous peptide that antagonizes the nAChR in the CNS.  相似文献

The promotion of root growth by capillarol [methyl 3-(3-methylbut-2-enoyl)-4-hydroxycinnamate] and related phenolic compounds were studied in relation to structure-activity relationships. Concentrations above 5 × 10−5 M capillarol stimulated the root growth of rice ( Oryza sativa L. cv. Tanginbozu and cv. Nihonbare) seedlings to about 180% of the control value at 5 × 10−4 M . Capillarol had no promotive and hardly any inhibitory effect on the growth of the second leaf sheath. Capillarol promoted the root growth also in seedlings of lettuce ( Lactuca sativa L. cv. Grand Rapids) to ca 150% of the control value at 5 × 10−4 M . The free acid form of capillarol (capillaric acid) was about as effective as capillarol. Para -hydroxy- but not m -methoxy- substituted cinnamic acid, phenylpyruvic acid, phenylacetic acid and amino-hydrocinnamic acid could stimulate root growth, but p -hydroxybenzoic acid was inactive. It is concluded that the important structural requirements for high root growth-promoting activity of phenolic compounds are the hydroxyl group substitution at the C-4 position of the benzene ring, and the propanoic or propenoic acid side chain at the C-1 position. A possible mode of the action of capillarol on root growth-promoting activity is also discussed.  相似文献

The effect of varied supply of P (2.5× 10−5 to 6× 10−4 M) and Zn (0 to 10−6 M) on uptake and concentrations of P and Zn was studied in cotton ( Gossypium hirsutum L. cv. Deltapine 15/21) grown in nutrient solution under controlled environmental conditions. At a given Zn supply, increasing levels of P had no significant effect on the concentrations of total Zn in plants. However, increasing levels of P induced or enhanced visual Zn deficiency symptoms when the Zn concentration in the nutrient solution was low. The concentrations of water-soluble Zn in roots and shoots constituted 60% of the total Zn concentrations for plants grown with low P and 30% for plants grown with high P. The concentration of water-soluble Zn in leaves, but not total Zn, was closely correlated with visual Zn deficiency symptoms, levels of chlorophyll, super oxide dismutase and membrane permeability. The critical deficiency concentration of water-soluble Zn in cotton leaves was in the range of 6 to 7 μg (g dry weight)−1 or about 1.0 μg (g fresh weight)−1. The results show that high P concentrations in plant tissue decrease the physiological availability of Zn. Water-soluble Zn in the tissue appears to be a suitable indicator for Zn nutritional status in general and phosphorus-induced Zn deficiency in particular. Also in field-grown orange trees (Citrus sinensis) visual Zn deficiency symptoms in leaves were closely related to the concentration of water-soluble Zn.  相似文献

Abstract The effect of ketoconazole on growth, sterol composition, in vitro sterol biosynthesis and P450-CO complex formation and its interaction with microsomal P450 was determined. On solid medium and in liquid medium ketoconazole inhibited Aspergillus fumigatus growth completely at 5 × 10−5 M and 50% of the growth at 1.3 × 10−5 M and 2.1 × 10−5 M respectively. A close relationship between accumulation of 14α-methyl sterols (eburicol, obtusifoliol and 14α-methyl fecosterol) and depletion of ergosterol with growth arrest was observed in ketoconazole treated cultures. The half inhibitory concentration for in vitro ergosterol biosynthesis and half saturating concentration for type II binding spectrum of ketoconazole were calculated as 73.8 ± 6.3 nM and 0.13 ± 0.04 μM respectively. CO displacement studies revealed inhibition of CO-P450 complex formation by ketoconazole.  相似文献

Efficiency of imidacloprid and pymetrozine on population growth parameters of cabbage aphid, Brevicoryne brassicae L. (Homoptera: Aphididae) was determined using demographic toxicology by leaf dip method. At first, bioassay tests were performed. The LC50 value and confidence limit for imidacloprid and pymetrozine were 1.61 × 10^-5 mol/L (0.74 × 10^-5-2.66 × 10^-5) and 2.14 × 10^-4 mol/L (1.24 × 10^-4-3.40 × 10^-4), respectively. To evaluate the sublethal effect of two insecticides on population growth parameters of cabbage aphid, LC30 concentrations of imidacloprid and pymetrozine were used at 5 mol/L and 30 mol/L. The experiments were carried out in a incubator at 20±1℃, 60% ± 5% RH and 16:8 (L:D) photoperiod on canola seedlings, Brassica napus L. var. 'PF'. Net fecundity rate decreased in both insecticide-treated populations. Intrinsic rates of increase (rm) were lower in imidacloprid and pymetrozine treatments than in controls. Intrinsic birth rates also decreased in treated populations. There was a relative increase in intrinsic death rates of treated populations. The mean generation times and doubling time were also lower in populations treated with insecticides than in controls. There was a considerable reduction in the average numbers of nymphs reproduced per female as compared with the control. The average longevity of female adults in the control was significantly different from those treated with imidacloprid and pymetrozine. However, there was no significant differences in aphid life-table parameters between the two insecticide-treated populations (P 〉 0.01).  相似文献

Abstract The oxidation of protoporphyrinogen by membranes from Desulfovibrio gigas with nitrite as the electron acceptor proceeds at the ratio of 1 mol protoporphyrin produced for each mol of nitrite reduced. Coupled to the protoporphyrinogen-nitrite reaction is the esterification of ortho-phosphate with a P/2e value of 0.92. Pentachlorophenol, 3 × 10−5 M, and carbonyl cyanide m -chlorophenyl hydrazone, 3 × 10−6 M, serve as uncouplers of phosphorylation while rotenone, 9 × 10−6 M, and hydroxyquinoline- N -oxide, 0.1 × 10−6 M, inhibit electron flow from protoporphyrinogen oxidase to nitrite reductase.  相似文献

Abstract The pathogenic bacterium Helicobacter pylori , which causes active, chronic type B gastritis and peptic ulcer disease, and increases the risk for development of gastric cancer, could tentatively interfere with growth factors and growth factor receptors of importance for the gastroduodenal mucosa, e.g. heparin-binding FGFs (fibroblast growth factors). H. pylori binds FGF with an extremely strong affinity (3.8 × 10−12 M), and also heparan sulfate and heparin with higher affinity ( K d 9 × 10−9 M) than FGFs bind to heparin (10−8–10−9 M). FGF receptors are also dependent on heparin for their activation. Heparan sulfate binding proteins (HSBP) are exposed on and shed from the surface of H. pylori , which often are localised close to the epithelial stem cells in the gastroduodenal glands. H. pylori could thus efficiently interfere with growth factors and growth factor receptors, tentatively resulting in disturbance of the delicate balance that control the renewal, maintenance and repair of the gastroduodenal mucosa. This mode of action has previously not been considered, but may constitute part of its pathogenic mechanism. Such a dynamic mode of action of H. pylori may explain the reason for that infected victims may either suffer from gastrointestinal symptoms or lack clinical evidence of disease or discomfort.  相似文献

The cytokinin-like activity of the growth regulating chemical EDU, N-[2-(2-oxo-1-imidazolidinyl)ethyl]-N'-phenylurea, was determined and compared with the actitivity of kinetin using the tobacco callus bioassay. EDU has a pronounced stimulatory effect on callus growth at concentrations of 5 × 10−4 and 1 × 10−3 M but was 5 000 times less potent than the synthetic cytokinin, kinetin. Senescence regulation and oxidant resistance induced by EDU and kinetin were also studied. EDU retarded the breakdown of chlorophyll, protein and RNA in 03-sensitive tobacco leaf discs during senescence. EDU was much more effective in arresting senescence and in protecting against 03 injury than kinetin. Results indicate the EDU-induced plant tolerance to 03 phytotoxicity may be indirect through enzyme induction regulation.  相似文献

Cultured guinea pig epidermal cells and dermal fibroblasts were chosen as model systems to study possible growth inhibition by cyclic AMP (cAMP)-elevating drugs. The rate of DNA synthesis was used to assay growth rate in control cultures and those treated with agents which increase intracellular cAMP, including dibutyryl cAMP, the phosphodiesterase inhibitors papaverine and theophylline and agents which stimulate adenylate cyclase, iso-proterenol and prostaglandin E2 methyl ester. Treatment for 24 h with dibutyryl cAMP (10−4 to 10−2 M) inhibited cell growth by 50 to 95%, whereas butyrate(10−4M) showed essentially no effect. This inhibition could not be attributed to decreased precursor transport or to drug toxicity. Papaverine (10−6 to 10−4 M) and theophylline (10−4 to 10−3 M) also gave dose-dependent growth inhibition as did isoproterenol and prostaglandinE2methyl ester. Radioautographic analysis of grain density after dibutyryl cAMP treatment and 3H-thymidine incorporation indicated no S-phase inhibition. Cyclic AMP-elevating drugs appear to inhibit growth of guinea-pig epidermal cells and dermal flbroblasts by blocking the cell cycle in G−2, M1, or G. −1  相似文献

The olfactory sensitivity of a herbivore, the grass carp, Ctenopharyngodon idella , to ten amino acids was investigated using electrophysiological recording techniques. Underwater electroolfactogram recordings increased exponentially with logarithmic increases of stimulus concentration from threshold to 1 × 10−3 m. Calculated detection thresholds ranged from 1 × 10−7.15 m to 1 × 10−8.7 M and are similar to those previously reported for carnivorous fish species. The relative stimulatory efficacy (RSE) of the ten amino acids determined at 1 × 10−4 M revealed five groups of stimuli, with cysteine the most stimulatory and proline the least. Measures of amino acid RSE did not differ significantly from those of previously studied carnivores, except for arginine. Grass carp showed strong responses to this compound, whereas it has produced only limited responses in carnivores. Differences in the responsiveness of herbivores and carnivores were not observed, suggesting that feeding niche segregation is probably not facilitated by differential olfactory sensitivities to feeding stimuli.  相似文献

Abstract: We investigated the interaction between Pb2+ and protein kinase C (PKC) in the Pb2+-induced release of norepinephrine (NE) from permeabilized adrenal chromaffin cells. Our analysis of endogenous PKC activity in permeabilized cells suggests that Pb2+ interacts with the adrenal enzyme at multiple sites. Pb2+ activates the enzyme through high-affinity ( K A(Pb) = 2.4 × 10−12 M ) interactions and inhibits the enzyme by competitive and noncompetitive interactions with nanomolar-( K i = 7.1 × 10−9 M ) and micromolar- ( K 'i = 2.8 × 10−7 M ) affinity sites, respectively. Activation of PKC by 12- O -tetradecanoylphorbol 13-acetate (TPA) in Ca2+-deficient, Pb2+-containing medium, enhances the Pb2+-induced NE release from permeabilized chromaffin cells by lowering the concentration of Pb2+ required for half-maximal activation of the secretory response from 7.5 × 10−10 to 5.7 × 10−11 M . The PKC inhibitors staurosporine and pseudosubstrate PKC (19–36) abolish the effect of TPA without affecting the Pb2+-induced secretion in the absence of TPA. These results indicate that (a) Pb2+ is a partial agonist of PKC, capable of both activating and inhibiting the enzyme and (b) synergistic activation of PKC by TPA and Pb2+ results in increased sensitivity of exocytosis to Pb2+ but is not obligatory for Pb2+-triggered secretion.  相似文献

Abstract: The effect of pardaxin, a new excitatory neurotoxin, on neurotransmitter release was tested using purely cholinergic synaptosomes of Torpedo marmorata electric organ. Pardaxin elicited the release of acetylcholine with a biphasic dose dependency. At low concentrations (up to 3 × 10−7 M ), the release was calcium-dependent and synaptosomal structure was well preserved as revealed by electron microscopy and measurements of occluded lactate dehydrogenase activity. At concentrations from 3 × 10−7 M to 10−5 M , the pardaxin-induced release of acetylcholine was independent of extracellular calcium, and occluded synaptosomal lactate dehydrogenase activity was lowered, indicating a synaptosomal membrane perturbation. Electron microscopy of 10−6 M pardaxin-treated synaptosomes revealed nerve terminals depleted of synaptic vesicles and containing cisternae. At higher toxin concentrations ( 10−5 M ), there were striking effects on synaptosomal morphology and occluded lactate dehydrogenase activity, suggesting a membrane lytic effect. We conclude that, at low concentrations, this neurotoxin is a promising tool to investigate calcium-dependent mechanisms of neurotransmitter release in the nervous system.  相似文献

Abstract Batch mating experiments were employed to study the kinetics of the conjugal transfer of a TOL plasmid, using the transconjugant strain Pseudomonas aeruginosa PAO 1162 (TOL) as the plasmid donor and Pseudomonas putida PB 2442 and Pseudomonas aeruginosa PAO 1162N as the plasmid recipients. Transfer rates from PAO 1162 (TOL) to PAO 1162N and PB 2442 measured for exponentially grown PAO 1162 (TOL) were 1.81 × 10−14 (standard error (S.E.) 1.25 × 10−15) ml·cell−1min−1 and 3.32 × 10−13 (S.E. 4.42 × 10−14) ml·cell−1min−1, respectively. The instability of the TOL plasmid in PAO 1162 (TOL) was evaluated under conditions that were non-selective for maintenance of the TOL catabolic functions. The measured rates of instability were 6.7 10−6 to 8.3 10−6 min−1, and the loss of the catabolic functions was mainly caused by structural instability of the plasmid.  相似文献

The permeability of internodal cells of Chara australis R. Brown for polyol molecules was determined by using a turgor balance to measure the increase in the osmotic pressure of an internodal cell incubated in artificial pond water containing one of the polyol compounds tested. The permeabilities for ethylene glycol, glycerol, meso -erythritol, xylitol and mannitol were (4. 39 ± 0. 52) × 10−9, (1. 49 ± 0. 40) × 1010, (4. 92 ± 0. 27) × 10−10 (9. 9 ± 3. 4) × 1011 and (7. 6 ± 4. 8) × 10−12 m s−3, respectively. The permeability for glycerol was slightly smaller than that for meso -erythritol, whose molecular weight is larger than that of glycerol in this homologous series: but the reason for this is not clear.  相似文献

The schooling behaviour of Atlantic mackerel was studied in a large tank at different light intensities in the range 12.6–1.8 × 10−10μEs−1 m−2. Variable light intensity was produced by accurately controlling the current to a green light-emitting diode (LED) 3 m above the experimental tank. Under high light levels (1.8 × 10−6μEs−1 m−2) mackerel always formed a single school, whereas at lower levels (1.8 × 10−8μEs−1 m−2) they swam as individuals. At light levels down to 1.0 × 10−6μEs−1 m−2 the mean nearest neighbour distance in a school remained relatively constant (0.3–0.9 body lengths), and individual mackerel swam along a path which deviated from the position of their nearest neighbours by less than 14°. As light dropped below 1.8 × 10−7μEs−1 m−2, both nearest neighbour distance and heading angle between nearest neighbours increased, with mean values of 1–1.8 body lengths and 23–92°, respectively, at 1.8 × 10−9μEs−1 m−2. The results are discussed in terms of ambient light conditions in the sea.  相似文献
