To examine the effects of maternal supranutritional selenium (Se) and nutrient restriction during mid and late gestation on placental characteristics and fetal liver glycogen, ewes received either adequate Se (ASe) or high Se (HSe) prior to breeding. On d 64 of gestation, ASe and HSe ewes remained at 100% of requirements (controls; CON) or were restricted (RES; 60% of requirements). On d 135 of gestation, fetal weight (P  0.08) was greatest in both HSe and CON ewes. Placentome number, mass, and caruncular and cotyledonary weight were not different (P  0.17) among treatments. Fetal mass:placental mass ratio was less (P = 0.06) in RES compared to CON ewes. Compared to ASe, HSe exhibited increased (P  0.08) cellular proliferation and DNA concentration and decreased (P = 0.07) cellular size in cotyledonary tissue. Nutritional restriction decreased (P  0.08) cotyledonary protein concentration and cellular size. VEGF receptor 1 (Flt) mRNA in cotyledonary tissue was greater in HSe compared with ASe ewes (P = 0.06) and in RES compared with CON ewes (P = 0.08). There was no effect of diet on caruncular growth variables (P  0.13) or on placental vascularity (P  0.11). Progesterone was greater (P  0.08) in ASe–RES ewes compared to all groups at d 90 and ASe–CON and HSe–CON at d 104. Although fetal glucose and cortisol concentrations were not affected by diet, fetal liver glycogen was greater (P = 0.04) in ASe–RES compared to ASe–CON and HSe–RES ewes with HSe–CON being intermediate. Both Se and nutritional plane may impact placental function and fetal growth, as fetal weight and liver glycogen are altered despite similar placental vascularity measurements.  相似文献

The objective of this study was to investigate the effects of selenium (Se) and alpha-tocopherol supplementation on uterine involution and ovarian function in dairy heifers fed a prepartum diet containing low concentrations of Se and alpha-tocopherol. Twenty-four pregnant Friesian heifers were randomly allocated to one of four experimental groups in a 2 x 2 design balanced for age and body weight. Prepartum treatments consisted of supplementation with either 2 intraruminal Se pellets or 3600 mg of alpha-tocopherol p.o. 4 times per wk, or both. Control animals received no supplementation. For 8 wk before calving, the heifers were fed exclusively on pasture hay which contained less than 10 microg/kg of Se and 19 mg/kg of alpha-tocopherol. After calving, the heifers grazed perennial ryegrass and white clover pasture. Concentrations of Se and alpha-tocopherol in serum for the prepartum heifers of the control group were 10 ng/ml and 1.3 microg/ml, respectively, indicating deficiencies of these nutrients. Treatment with Se and alpha-tocopherol increased prepartum serum concentrations of Se and alpha-tocopherol to 74 ng/ml and 5 microg/ml, respectively (P < 0.001). However, treatment with Se, alpha-tocopherol, or both, failed to enhance uterine involution, hasten resumption of postpartum ovarian activity or reduce the incidence of clinical postpartum abnormalities. These findings suggest that postpartum reproductive dysfunction is not a primary feature of moderate Se or vitamin E deficiency of cattle at pasture.  相似文献

Lactating Holstein cows (n = 135) were randomly assigned to one of the two sources of supplemental selenium (Se), sodium selenite (SS) or Se yeast (SY), fed at 0.3 mg/kg diet dry matter from 25 d before calving to 70 d in milk (DIM), in diets not suboptimal in basal Se concentrations. Cows were evaluated for health daily in the first 10 DIM, and uterine cytology of the previously gravid uterine horn was assessed at 30 DIM. The Ovsynch protocol was initiated at 42 DIM; ovarian responses to hormonal treatments were evaluated by ultrasonography. The uteri of cows were flushed 6 d after timed AI for collection of embryos and oocytes. Plasma concentrations of Se and progesterone were measured throughout the postpartum period and during the reproductive protocol, respectively, and plasma glutathione peroxidase activity was determined 6 d after AI. Concentrations of Se in pre- and postpartum diets ranged from 0.43 to 0.56 mg/kg of dry matter. Incidence of retained placenta, fever, ketosis, mastitis, acute puerperal metritis, clinical endometritis, and subclinical endometritis were not significantly different between treatments. There were no differences between groups in concentrations of Se and progesterone or glutathione peroxidase activity in plasma. Treatment did not influence ovarian responses to the synchronization protocol, fertilization rate, number of blastomeres and live blastomeres, or proportions of grades 1 and 2, degenerated, and degenerated-unfertilized embryos/oocytes. Odds of subclinical endometritis on Day 30 postpartum more than doubled in cows with fever of unknown origin or acute puerperal metritis in the first 10 DIM. Fertilization rate tended to be reduced in cows with subclinical endometritis. In summary, replacing SS with an organic source of Se in diets not suboptimal in basal Se concentrations did not improve Se status, uterine health, fertilization, or embryo quality in early lactation dairy cows.  相似文献

The timing of dietary fat intake may modify breast cancer risk. In addition, n-3 fatty acids reduce, and n-6 fatty acids increase, the risk of breast cancer and a maternal high n-6 fat diet results in a greater risk of breast cancer in the female offspring. We hypothesized that the timing of n-3 fatty acid-enriched fish oil supplementation would be important for reducing the risk of breast cancer. Female rats were fed to a high n-6 fat diet containing 20% of the sunflower oil by weight during pregnancy and lactation, and the female offspring were exposed to fish oil by oral gavage either during the perinatal period via maternal intake or during puberty or adulthood. Exposure during the perinatal period to a maternal high n-6 fat diet with fish oil supplementation significantly reduced the incidence of carcinogen-induced mammary tumors in the female offspring compared to a maternal high n-6 fat diet with no fish oil supplementation or fish oil supplementation later in life (P=.0228 by Cox proportional hazards model). We found that a maternal high n-6 fat diet during pregnancy is more important in increasing the risk of mammary tumors in the female offspring than a maternal high n-6 fat diet during lactation. This study suggests that fish oil supplementation during the perinatal period decreases the effect of a maternal high n-6 fat diet on subsequent carcinogen-induced mammary tumor risk, whereas fish oil supplementation during puberty or adulthood does not.  相似文献

The effect of maternal dietary selenium (Se) and gestation on the concentrations of Se and zinc (Zn) in the porcine fetus were determined. Mature gilts were randomly assigned to treatments of either adequate (0.39 ppm Se) or low (0.05 ppm Se) dietary Se. Gilts were bred and fetuses were collected throughout gestation. Concentrations of Se in maternal whole blood and liver decreased during gestation in sows fed the low-Se diet compared to sows fed the Se-supplemented diet. Maternal intake of Se did not affect the concentration of Se in the whole fetus; however, the concentration of Se in fetal liver was decreased in fetuses of sows fed the low-Se diet. Although fetal liver Se decreased in both treatments as gestation progressed, the decrease was greater in liver of fetuses from sows fed the low-Se diet. Dietary Se did not affect concentrations of Zn in maternal whole blood or liver or in the whole fetus and fetal liver. The concentration of Se in fetal liver was lower but the concentration of Zn was greater than in maternal liver when sows were fed the adequate Se diet. These results indicate that maternal intake of Se affects fetal liver Se and newborn piglets have lower liver Se concentrations compared to their dams, regardless of the Se intake of sows during gestation. Thus, the piglet is more susceptible Se deficiency than the sow.  相似文献

The working hypothesis was that restricted protein content in diets of prepubertal ewe lambs would delay time of onset of puberty to a greater extent than restricted energy content. Restriction of dietary protein (metabolizable protein, MP) or energy (ME) intake on age and weight at puberty was studied in ewe lambs in two experiments. In Experiment 1, 85 crossbred ewe lambs were assigned by weight to one of five dietary treatment groups: control (CTL; 18 mg ME day−1 and 173 g MP day−1); energy restricted (ER; 10.2 mg ME day−1 and 96 g MP day−1); protein restricted (PR; 11 mg ME day−1 and 66 g MP day−1); ER for 9 weeks then switched to CTL (ER/CTL); PR for 9 weeks then switched to CTL (PR/CTL). Ewe lambs were weighed biweekly, bled weekly, and serum was assayed for progesterone to determine time of puberty. Average age at puberty differed among lambs fed the various diets (P < 0.01). Ewe lambs in the CTL group were younger and weighed more at puberty. Lambs fed diets restricted in protein or energy content were similar in age at puberty. Dietary switching, ER/CTL and PR/CTL, affected body weight and gain (P < 0.05), but did not result in changed age at puberty compared with lambs fed ER or PR diets. In Experiment 2, two breed types (maternal type and meat type) were used to compare responses of dietary protein restriction to dietary energy restriction. In Experiment 2, lambs (n = 60) were assigned to either the CTL, PR or ER diets. Lambs in the CTL group gained more weight than lambs fed the PR diet and lambs fed the PR diet gained more weight than lambs fed the ER diet (P < 0.05). Diet affected age (P < 0.001), but not weight at puberty (P = 0.13). Dietary energy and protein restriction influenced age at puberty in ewe lambs with energy restriction having a greater influence on delaying onset of puberty than protein restriction. Realimentation did not overcome effects of previous feed restriction by inducing an earlier age at puberty than ewe lambs fed the ER or PR diet throughout the experimental period. Body weight gain prepuberty is more important than type of dietary restriction (protein or energy) with regard to impacting age at puberty. We, therefore, reject the working hypothesis that dietary protein restriction prepuberty would have a greater impact in delaying onset of puberty than dietary energy restriction.  相似文献

Octylphenol (OP) is one of a number of compounds found in the environment that has estrogen-mimicking actions in vivo. Our objective was to determine if maternal exposure to octylphenol during fetal and/or postnatal life would affect the onset of puberty, endocrine status, and subsequent ovarian follicular dynamics of ewe lambs. Lambs were born in March to ewes that received twice weekly s.c. injections of octylphenol (1000 micro g/kg/day) from Day 70 of gestation to weaning (n = 6); Day 70 of gestation to birth (n = 3); birth to weaning (n = 5; gestation = 145 days); or corn oil from Day 70 of gestation to weaning (control; n = 5). Blood samples were collected twice weekly to determine progesterone and FSH concentrations from 20 wk of age throughout the first breeding season. Onset of puberty and interestrous intervals were determined from 20 wk of age by twice daily observation for estrus in the presence of a vasectomized ram. During January the ovaries of each lamb were examined using transrectal ultrasonography from the day of estrus for 15 days. Blood samples were collected every 8 h to examine FSH concentrations and every 2 h to detect the preovulatory gonadotropin surge throughout this estrous cycle. The onset of puberty and first progesterone rise was advanced and the FSH preovulatory surge was elevated for longer in the OP-treated lambs compared with the control lambs (P < 0.05). Interestrous intervals, FSH profiles, and ovarian follicular dynamics were not affected (P > 0.05) by exposure to octylphenol. In conclusion, octylphenol exposure advanced the onset of puberty but it did not disrupt FSH concentrations or the dynamics of ovarian follicular growth.  相似文献

The effect of undernutrition on ovarian and uterine venous progesterone concentrations and endometrial progesterone content on Days 5 and 10 of the estrous cycle were studied. Forty ewes were synchronized using progestagen pessaries. At pessary withdrawal, the ewes were fed diets to provide either 1.5 or 0.5 times the daily maintenance requirement (Group H, n = 20 and Group L, n = 20, respectively). Ewes fed the low nutrition diet (Group L) had higher mean peripheral progesterone concentrations than those fed the high plane diet (Group H; P < 0.05) but lower endometrial progesterone content on Day 5 (P < 0.05). Neither ovarian nor uterine venous levels were affected by nutrition on either Day 5 or 10. Progesterone concentrations in blood samples collected ipsilateral to ovaries bearing a corpus luteum (CL) were higher than in the contralateral samples (P < 0.001). It is concluded that undernutrition can produce a reduction of endometrial content of progesterone the first week after mating. Since no differences in ovarian venous concentrations were observed, it remains to be shown whether this variation is due to other variables, such as the population of endometrial progesterone receptors or other nonhormonal factors.  相似文献

We have examined the effects of dietary selenium (Se) supplementation on larval growth and immunocompetence of the lepidopteran pest, the cabbage looper, Trichoplusia ni. Supplementation of the diet of T. ni larvae with 10–20 ppm Se resulted in a 1 day delay in pupation. The effects of the addition and/or removal of dietary Se on total Se bioaccumulation and sequestration were determined by neutron activation analysis of pupae. Early penultimate instar larvae moved from selenium containing diet to basal diet lost total pupal Se content down to the level of those fed basal diet. Conversely, larvae moved from basal diet to diet containing additional Se rapidly attained pupal Se levels comparable to larvae fed Se throughout larval development. Therefore, dietary Se is rapidly accumulated or lost during larval development, but significant amounts are sequestered from diet into pupae. Larvae were reared on diet supplemented with 5 or 10 ppm Se until the onset of the penultimate instar then infected per os with increasing concentrations of the fatal baculovirus Autographa californica multiple nucleopolyhedrovirus (AcMNPV). Larvae fed Se in the penultimate and ultimate instars were more resistant to viral infection than larvae not fed Se in the final instars. This study indicates that dietary Se levels rapidly impact Se assimilation and sequestration and that tissue Se levels are an important factor in resistance to AcMNPV infection in larval T. ni.  相似文献

The study was conducted to investigate the effects of dietary maternal selenomethionine or sodium selenite supplementation on performance and selenium status of broiler breeders and their next generation. Two hundred and forty 39-week-old Lingnan yellow broiler breeders were allocated randomly into two treatments, each of which included three replicates of 40 birds. Pretreatment period was 2 weeks, and the experiment lasted 8 weeks. The groups were fed the same basal diet supplemented with 0.30 mg selenium/kg of sodium selenite or selenomethionine. After incubation, 180 chicks from the same parental treatment group were randomly divided into three replicates, with 60 birds per replicate. All the offspring were fed the same diet containing 0.04 mg selenium/kg, and the experiment also lasted 8 weeks. Birth rate was greater (p < 0.05) in hens fed with selenomethionine than that in hens fed with sodium selenite. The selenium concentration in serum, liver, kidney, and breast muscle of broiler breeders, selenium deposition in the yolk, and albumen and tissues' (liver, kidney, breast muscle) selenium concentrations of 1-day-old chicks were significantly (p < 0.01) increased by maternal selenomethionine supplementation compared with maternal sodium selenite supplementation. The antioxidant status of 1-day-old chicks was greatly improved by maternal selenomethionine intake in comparison with maternal sodium selenite intake and was evidenced by the increased glutathione peroxidase activity in breast muscle (p < 0.05), superoxide dismutase activity in breast muscle and kidney (p < 0.05), glutathione concentration in kidney (p < 0.01), total antioxidant capability in breast muscle and liver (p < 0.05), and decreased malondialdehyde concentration in liver and pancreas (p < 0.05) of 1-day-old chicks. Feed utilization was better (p < 0.05), and mortality was lower (p < 0.05) in the progeny from hens fed with selenomethionine throughout the 8-week growing period compared with those from hens fed with sodium selenite. In summary, we concluded that maternal selenomethionine supplementation increased birth rate and Se deposition in serum and tissues of broiler breeders as well as in egg yolk and egg albumen more than maternal sodium selenite supplementation. Furthermore, maternal selenomethionine intake was also superior to maternal sodium selenite intake in improving the tissues Se deposition and antioxidant status of 1-day-old chicks and increasing the performance of the progeny during 8 weeks of post-hatch life.  相似文献

The overall goal was to describe the importance of fibroblast growth factors (FGFs) during development of the bovine embryo. An inhibitor of FGF receptor kinase activity (SU5402) was used to examine whether FGF signaling is required for embryo development. Addition of 20 μM SU5402 on Day 0 (Day of IVF) reduced (P = 0.04) the percentage of oocytes becoming blastocysts on Day 7 compared to controls (5.9 ± 2.1 vs 16.9 ± 2.4; average ± SEM). Also, Day-8 blastocysts placed into individual culture drops of medium containing SU5402 tended to have decreased (P = 0.08) blastomere numbers at Day 11 (211.1 ± 27.5 vs 297.8 ± 25.0). A second series of studies determined if supplemental FGF2 enhances development in vitro. There was no effect of FGF2 on cleavage or blastocyst development rates when 5 or 100 ng/mL FGF2 was provided immediately after fertilization. Also, FGF2 supplementation beginning on Day 5 post-fertilization did not significantly affect blastocyst rates or the number of trophoblast and inner cell mass cells. However, addition of 500 ng/mL FGF2 at both Day 0 and Day 4 increased (P = 0.03) the percentage of oocytes that became blastocysts on Day 7 compared with controls (27.4 ± 1.3 vs 19.7 ± 1.3). In a final study, the thermal-protective ability of FGF2 was examined by adding FGF2 1 h before exposing Day 5 embryos to heat shock. Addition of FGF2 did not significantly influence embryo thermal-tolerance. In conclusion, FGF receptor activation was important for optimal blastocyst formation and FGF2 supplementation increased bovine blastocyst formation when provided at high concentrations.  相似文献

Selenium (Se), selenoprotein N (SelN) and selenoprotein W (SelW) play a crucial role in muscle disorders. Se status highly regulates selenoprotein mRNA levels. However, few attempts have been performed on the effect of dietary Se supplementation on muscle SelN and SelW mRNA levels in birds. To investigate the effects of Se on the regulation of SelN and SelW mRNA levels in muscle tissues, one-day-old male chickens were fed either a commercial diet or a Se-supplemented diet containing 1.0, 2.0, 3.0 or 5.0 mg/kg sodium selenite for 90 days. Muscle tissues (breast, flight, thigh, shank and cardiac muscles) were collected and examined for Se content and mRNA levels of SelN and SelW. Moreover, Selenophosphate synthetase-1 (SPS-1) and selenocysteine-synthase (SecS) mRNA levels were analyzed. Significant increases in SelN mRNA levels were obtained in breast, thigh and shank muscles treated with Se, with maximal effects at 3.0 mg Se/kg diet, but 2.0 mg Se/kg diet resulted in peak levels of Sel N mRNA in flight muscles. Changes in SelW mRNA abundance in thigh and shank muscles increased in response to Se supply. After reaching a maximal level, higher Se supplementation led to a reduction in both SelN and SelW mRNAs. However, SelN and SelW mRNA levels displayed a different expression pattern in different skeletal and cardiac muscles. Thus, it suggested that skeletal and cardiac muscles SelN and SelW mRNA levels were highly regulated by Se supplementation and different muscle tissues showed differential sensitivity. Moreover, Se supplementation also regulated the levels of SPS1 and SecS mRNAs. The mRNA levels of SPS1 and SecS were enhanced in the Se supplemented groups. These data indicate that Se regulates the expression of SelN and SelW gene and affect the mRNA levels of SecS and SPS1.  相似文献

To investigate the role of selenium (Se) in the developing porcine fetus, prepubertal gilts (n=42) were randomly assigned to either Se-adequate (0.39 ppm Se) or Se-deficient (0.05 ppm Se) gestation diets 6 wk prior to breeding. Maternal and fetal liver was collected at d 30, 45, 70, 90, and 114 of pregnancy. Concentrations of Se in maternal liver decreased during gestation in gilts fed the low-Se diet. The activity of cellular glutathione peroxidase (GPx) was decreased at d 30 and 45 of gestation in liver of gilts fed the low-Se diet. Concentrations of malondialdehyde (MDA) and hydrogen peroxide (H2O2) were greater in liver homogenates from gilts fed the low-Se diet. Within the fetuses, liver Se decreased in those fetuses of gilts fed the low-Se diet. Although the activity of GPx in fetal liver was not affected by the maternal diet, concentrations of H2O2 and MDA in fetal liver were greater in fetuses from gilts fed the low-Se diet. Maternal liver GPx activity was approx 12-fold greater than fetal liver GPx activity regardless of dietary treatment. These results indicate that maternal dietary Se intake affects fetal liver Se concentration and feeding a low-Se diet during gestation increases oxidative stress to the fetus, as measured by fetal liver H2O2 and MDA.  相似文献

In the present report, we determined the impact of dietary selenium (Se) deficiency and supplementation on the expression of two ER-resident selenoproteins i.e., Selenok and Selenom in the ovaries of aging mice. The mRNA expression of Selenok and Selenom (RT-qPCR) was significantly higher in the ovaries of mice fed diets supplemented with inorganic (ISe-S: 0.33 mg Se/kg) and organic (OSe-S: 0.33 mg Se/kg) Se compared to those fed a Se-deficient (Se-D: 0.08 mg Se/kg) diet and both Se-adequate (ISe-A: 0.15 mg Se/kg and OSe-A: 0.15 mg Se/kg) diets. Similarly, the protein signals of SELENOK (immunofluorescence assay) were also significantly higher in the Se-supplemented groups compared to those fed Se-D and Se-adequate (ISe-A and OSe-A) diets. Meanwhile, the rate of in vitro-produced blastocysts developing from MII oocytes was also evaluated and it was revealed that this rate was significantly higher in the Se-supplemented mice compared to those fed a Se-D diet. Altogether, the dietary Se supplementation increased the expression of Selenok (also its protein expression) and Selenom in the ovaries of aging mice, potentially contributing to an improved developmental potential of in vitro-matured M II oocytes.  相似文献

Selenium (Se), an essential micronutrient, is believed to enhance neutrophil functions. This study aimed to compare the effects of supplemented organic (Sel-Plex®) and inorganic (sodium selenite) Se on neutrophil functions in high-producing dairy cows, during the periparturient period. Twenty-five Holstein cows were randomly allocated to five dietary treatments as follows: control diet (basal diet without Se supplementation), IN 0.3 (basal diet supplemented with inorganic Se at 0.3 mg/kg dry matter (DM)), IN 0.5 (inorganic Se at 0.5 mg/kg DM), OR 0.3 (organic Se at 0.3 mg/kg DM) and OR 0.5 (organic Se at 0.5 mg/kg DM). Some evaluated parameters included neutrophil functions and plasma Se concentrations in cows and plasma Se concentrations in calves. Neutrophil phagocytosis did not significantly differ among the five groups. However, organic Se supplementation significantly increased (P < 0.01) the respiratory burst of neutrophils when compared to cows fed IN 0.3 and the control diet. In comparison to inorganic Se, neutrophil apoptosis was decreased (P < 0.01) when cows were fed organic Se or the control diets. These effects of organic Se on respiratory burst activities and apoptosis of neutrophils were in a dose-dependent manner. Supplementation with l-arginine can increase uterine arterial blood flow and vascular perfusion of the preovulatory follicle in mares. Increased vascular perfusion of the preovulatory follicle has been correlated with successful pregnancy in mares. The objective of this study was to determine if supplemental l-arginine would increase ovarian arterial blood flow, vascular perfusion of the preovulatory follicle, and embryo recovery rates in mares. Mares were blocked by age and breed and assigned at random within block to l-arginine supplementation or control groups. Mares were fed l-arginine beginning 17 days before and through the duration of the study. Transrectal Doppler ultrasonography was used to measure ovarian arterial blood flow and vascular perfusion of the preovulatory follicle daily when it reached 35 mm and subsequent CL on Days 2, 4, and 6. Mares, on achieving a follicle of 35 mm or more were bred via artificial insemination and an embryo collection was attempted 7 days after ovulation. Treatment did not affect interovulatory interval (arginine-treated, 18.1 ± 2.6 days; control, 20.7 ± 2.3 days) or embryo recovery rate (arginine-treated, 54%; control, 48%). Mares treated with l-arginine had a larger follicle for the 10 days preceding ovulation than control mares (30.4 ± 1.2 and 26.3 ± 1.3 mm, respectively; P < 0.05) and vascular perfusion of the dominant follicle tended (P = 0.10) to be greater for the 4 days before ovulation. No differences were observed between groups in diameter or vascular perfusion of the CL. Resistance indices, normalized to ovulation, were not significantly different between groups during the follicular or luteal phase. Oral l-arginine supplementation increased the size and tended to increase perfusion of the follicle 1, but had no effect on luteal perfusion or embryo recovery rates in mares.  相似文献

Fifty-four Landrace × Yorkshire gilts (59.0 ± 4.2 kg and 147 ± 3 d old) were used to examine the effects of dietary energy source (starch or mixed fat) at high [112.5% of energy requirements recommended by NRC (1998)], normal (100%), and low (87.5%) energy feeding levels on ovarian follicular development and oocyte maturation. Forty-seven estrus gilts were slaughtered at Day 19 after the second estrus; oocytes were recovered from follicles >4 mm in diameter, and matured in vitro for 44 h. Gilts fed high-energy diets had more follicles >4 mm (mean, 25.8 vs. 19.1, P < 0.05) and more oocytes that reached metaphase II (80.3 vs. 64.0%, P < 0.05) than those fed the low-energy diet. Furthermore, gilts fed starch-rich diets had enhanced oocyte nuclear maturation relative to those fed fat-rich diets (75.4 vs. 68.0%, P < 0.05). Compared to the lower-energy feeding groups, high-energy feeding groups had higher (P < 0.05) blood concentrations of postprandial insulin (1562.4 vs. 990.0 ng/4 h), IGF-I (321.2 vs. 256.9 ng/mL), and LH pulses (2.7 vs. 1.4 pulses/6 h). Follicular fluid concentrations of IGF-I (198.5 vs. 143.1 ng/mL) and estradiol (152.6 vs. 124.8 ng/mL) were higher (P < 0.05) in the high-energy group than in the normal group. Compared with gilts fed the high-energy diet supplemented with fat, gilts fed the high-energy diet supplemented with starch had a tendency (P < 0.10) towards increased IGF-I concentration in both blood and follicular fluid, and improved oocyte nuclear maturation during culture in vitro. We inferred that starch-rich, high-energy diets during rearing may improve ovarian follicular development and oocyte maturation in replacement gilts.  相似文献

We estimated the nutritional availability of selenium (Se) in Se-enriched Kaiware radish sprouts (SeRS) by the tissue Se deposition and glutathione peroxidase (GPX) activity of rats administered the sprouts, and examined the effect of SeRS on the formation of aberrant crypt foci (ACF) in the colon of mice administered 1,2-dimethylhydrazine (DMH) to evaluate anti-tumor activity. Male weanling Wistar rats were divided into seven groups and fed a Se-deficient basal diet or the basal diet supplemented with 0.05, 0.10, or 0.15 μg/g of Se as sodium selenite or SeRS for 28 d. Supplementation with Se dose-dependently increased serum and liver Se concentrations and GPX activities, and the selenite-supplemented groups showed a higher increase than the SeRS-supplemented groups. The nutritional availability of Se in SeRS was estimated to be 33 or 64% by slope ratio analysis. Male 4-week-old A/J mice were divided into seven groups and fed a low Se basal diet or the basal diet supplemented with selenite, SeRS, or selenite + non-Se-enriched radish sprouts (NonSeRS) at a level of 0.1 or 2.0 μg Se/g for 9 weeks. After 1 week of feeding, all mice were given six subcutaneous injections of DMH (20 mg/kg) at 1-week intervals. The average number of ACF formed in the colon of mice fed the basal diet was 4.3. At a supplementation level of 0.1 μg Se/g, only SeRS significantly inhibited ACF formation. At a supplementation level of 2.0 μg Se/g, both selenite and SeRS significantly inhibited ACF formation. The addition of NonSeRS to the selenite-supplemented diets tended to inhibit ACF formation, but this was not statistically significant. These results indicate that SeRS shows lower nutritional availability but higher anti-tumor activity than selenite.  相似文献

This study aims to determine the effects of maternal protein and energy malnutrition during lactation on the linear growth, body weight and onset of puberty of the female offspring. At parturition, dams were randomly assigned to the following groups: (C) control group, with free access to a standard laboratory diet containing 23% protein; (PR) protein-restricted group, with free access to an isoenergy and protein-restricted diet containing 8% protein; and (ER) energy-restricted group, receiving standard laboratory diet in restricted quantities. After weaning, the female pups had free access to standard laboratory diet. From day 30 onwards, the pups were inspected daily for vaginal opening. Cyclic stages of the ovaries were studied by daily vaginal smears after vaginal opening until day 40 when all animals were sacrificed with pentobarbital. From day 4 after birth until day 40, body weight and linear growth in the PR and ER rats were significantly lower than in controls (p < 0.001). In spite of the significant (p<0.05) delayed in the vaginal opening in PR and ER rats, the first estrous cycle occurred at the same time of vaginal opening in all groups. The PR and ER rats exhibited a lower uterine (PR = 42%, ER = 40%, p < 0.001) and ovarian (PR = 26%, ER=19%, p < 0.05) absolute weight and uterus relative weight (PR = 27%, ER = 22%, p < 0.05). Our data showed that maternal protein and energy malnutrition during lactation leads to growth retardation and delayed on the onset of puberty in female pups, with vaginal opening and estrous cycle occurring at the same time.  相似文献

The objective of the study was to establish an effective ovarian superstimulatory protocol and subsequently obtain oocytes from bison by transvaginal ultrasound-guided follicular aspiration. Two experiments involving 22 wood bison were done during the breeding season (September to December). In experiment 1, the bison were given a luteolytic dose of prostaglandin (Day 0) and underwent follicular ablation (Day 8) to induce ovarian synchrony. Synchronized bison were then assigned randomly to two groups (n = 11 per group) and given either 200 mg FSH diluted in saline sc, or 200 mg FSH diluted in a proprietary slow-release formulation (SRF) im on Days 9 and 11. Prostaglandin was given to both groups on Day 11 followed by 25 mg LH on Day 13. Oocytes were collected by transvaginal ultrasound-guided aspiration of follicles ≥5 mm on Day 14. In experiment 2, bison were synchronized as in experiment 1 and assigned randomly to one of two groups (n = 11 per group) and given either a single dose of 2500 IU eCG im on Day 9, or 200 mg FSH sc on Days 9 and 11. Prostaglandin was given to both groups on Day 11, and LH (25 mg) was given on Day 13. Oocyte collection was done as described in experiment 1. Cumulus-oocyte-complexes (COC) were classified according to morphologic characteristics. In experiment 1, more follicles ≥5 mm were detected on Day 14 in bison treated with FSH versus eCG (12.2 ± 1.73 vs. 5.8 ± 0.52; P < 0.05), and more COC were collected from FSH-treated animals (7.2 ± 1.41 vs. 3.4 ± 0.62; P < 0.05). In experiment 2, the FSH-saline and FSH-SRF groups had a similar number (mean value ± standard error of the mean) of follicles ≥5 mm on Day 14 (12.4 ± 1.49 vs. 13.8 ± 1.24, respectively) and a similar number of COC were collected (6.5 ± 1.13 vs. 6.3 ± 0.96, respectively). The proportion of COC collected per follicle aspirated and the percentage of compact, expanded, and denuded oocytes did not differ between groups in either experiment 1 or 2. In summary, a two-dose regimen of FSH diluted in saline and given sc or in a SRF and given im induced a similar ovarian response in wood bison, whereas a single dose of eCG resulted in a significantly lower ovarian response. Overall, COC were collected from 55% of follicles after transvaginal, ultrasound-guided needle aspiration in wood bison.  相似文献
