以伴刀豆球蛋白为介质定向固定化脲酶的研究   总被引:1,自引:0,他引:1  
将戊二醛将伴刀豆球蛋白(ConA)和壳聚糖载体交联,然后利用ConA与脲酶糖链的特异性结合作用,实现脲酶的定向固定化.定向固定化的最适条件为戊二醛浓度3.5%、ConA浓度1mg/mL、ConA溶液pH值7.0、脲酶浓度0.4mg/mL.定向固定化脲酶的最适pH 5.0~6.0、最适温度77℃,米氏常数Km11.76mmol/L,与游离酶及非定向固定化脲酶比较,定向固定化脲酶的最适pH向酸性范围发生了偏移并有更宽的pH适用范围,最适温度提高,与底物的亲和力较大,且有较好的操作稳定性.  相似文献

为了提高蒜氨酸酶的稳定性并实现酶的反复利用,研究了影响蒜氨酸酶固定化的因素及固定化蒜氨酸酶的酶学性质。蒜氨酸酶的固定化以壳聚糖微球为载体,戊二醛为交联剂,固定化的最适条件为:戊二醛浓度4%,给酶量20.2U,交联时间2h。固定化蒜氨酸酶的最适pH值7.0,最适温度35℃,米氏常数Km 7.9 mmol/L,操作稳定性比较好,连续使用10次后酶活力损失低于10%。  相似文献

重组大肠杆菌热稳定性过氧化氢酶的纯化及性质研究   总被引:12,自引:0,他引:12  
将产热稳定性过氧化氢酶的重组大肠杆菌培养后菌体破碎得到的粗酶液经热处理、硫酸铵分级沉淀、DEAE\|Sephadex A\|50离子交换层析、HiPrep16/10 Phenyl疏水作用层析、Superdex200 HR 10/30凝胶层析提纯后得到电泳纯的酶,比酶活达到15629U/mg。此酶的最适温度为70℃,最适pH70,在60℃保温60min酶活力基本不变,在pH3~8的范围内比较稳定。此酶的Km和Vmax分别为775mmol/L和278mmol\5min\+\{-1\}·mg-1。1mmol/L的Zn2+、Ba2+、Mn2+可使该酶完全失活,KCN、NaN\-3、Na\-2S\-2O\-4、巯基乙醇对酶活力有抑制作用,50mmol/L的EDTA不影响酶活性。  相似文献

变色栓菌(Trametes versicolor)胞外产酶培养液经硫酸铵沉淀、DEAE-cellulose DE52离子交换柱层析后,获得两个活性组分D1和D2,其中活性组分D2经Phenyl SepharoseTM6Fast Flow疏水层析后,所得样品MnP1经SDS-PAGE检测已达到电泳纯。活性组分D1经Phenyl SepharoseTM6Fast Flow疏水层析、Sephacryl S-200HR凝胶过滤层析后,所得样品MnP2经SDS-PAGE检测已达到电泳纯。两种同工酶MnP1及MnP2,各自的比活力为579.09、425.00U/mg;纯化倍数为17.51、12.85;活力回收率为6.17%、2.47%。由SDS-PAGE法测得MnP1及MnP2的表观分子量分别为46.3kD、43.0kD。两种同工酶催化DMP(2,6-二甲氧基酚)氧化反应的最适pH值及最适反应温度有所不同,最适pH值分别为pH5.8、pH6.2,最适反应温度分别为60℃、65℃。在45℃以下,pH4.0~7.0之间,MnP1及MnP2的稳定性好。DMP为最佳酶促反应底物,以DMP为底物的Km分别为13.43μmol/L、12.45μmol/L。在无Mn2 存在的条件下,酶促反应几乎不发生。EDTA在较高浓度时抑制酶的活性,DTT在所试浓度下都完全抑制酶的活性。  相似文献

高产菊粉酶酵母筛选、发酵和酶学性质研究   总被引:18,自引:0,他引:18  
筛选到1株菊粉酶高产克鲁维酵母菌株,采用酵母高密度细胞发酵方法,最高菊粉酶产量达到288.78u/mL,比80~90年代国际上报道的克鲁维酵母菊粉酶最高产量高6.8倍。该酶的菊粉酶/转化酶活性比为1/24.72;菊糖m=13.3mmol/L,蔗糖Km=62.6mmol/L;最适反应pH值为4.4,但在pH3.8~5.6的范围内均保持了较高的活性,相当于最适pH值下活性的90%;最适反应温度为55℃,在50~575℃范围内能够保持较高活性,50℃下酶的半衰期约为16h;外加Mg2+提高酶活性11.28%。  相似文献

研究了芳腈水合酶催化水合3-氰基吡啶生成尼克酰胺的反应条件及影响因子.酶反应的最适pH为8.0,最适温度为25℃.酶在pH8.5于25℃保温4小时或在25—30℃于pH8.0保温3小时是稳定的.反应液中加入Fe~(3+)(1.5 mmol/L)可使酶活力增加 50%,而加入NH_4~+(300 mmol/L)则使酶活降低了67%.Ag~+和 Hg(2+)”强烈地抑制酶反应活性,在浓度均为 5mmol/L时,抑制率分别为99.7%和100%.NaCN(50 mmol/L)和苯甲腈(100 mmol/L)对酶活性的抑制率分别为78%和85%.该酶作用于 3-氰基吡啶的Km为62.5 mmol/L,V_(max)为85.8 μmol·min~(-1)·mg~(-1).  相似文献

葡萄球菌Staphylococcus hominis来源的N-乙酰神经氨酸裂合酶基因shnal(GenBank Accession No.EFS20452.1)构建至pET-28a质粒并在大肠杆菌中得到表达.通过目的蛋白的纯化和酶学性质研究发现,ShNAL是一个四聚体,裂解方向的最适反应pH为8.0;合成方向的最适反应pH为7.5,最适反应温度为45℃.在45℃下孵育2h对ShNAL的活力基本无影响,高于45℃时,活力迅速下降.该酶在pH 5.0~10.0的环境中比较稳定,4℃下放置24 h酶的残余活力在70%以上.ShNAL对N-乙酰神经氨酸(Neu5Ac)、N-乙酰甘露糖胺(Man)和丙酮酸(Pyr)的Km值分别是(4.0±0.2) mmol/L、(131.7±12.1)mmol/L和(35.14±3.2) mmol/L,kcat/Km值分别为1.9 L/(mmol·s)、0.08 L/(mmol·s)和0.08 L/(mmol·s).  相似文献

以木枣试管苗叶片为材料,以愈创木酚为底物,研究其POD同功酶动力学,结果表明:木枣试管苗叶片POD同功酶酶促反应的最适pH值为5.0,最适温度范围为42~46℃之间,Km值为40.66 mmol/L,Vmax值为673.6U/s.gFW。在H2O2浓度为0.4%的反应体系中,反应3~18 min内测定酶活可获得较好的结果。  相似文献

以琼脂粉为基质制备金属螯合载体,并用于固定重组腈水解酶。研究发现:制备金属螯合载体最合适的金属离子为Zn2+。当Zn2+离子浓度0.3 mol/L、给酶量15.6 mg/g、固定化pH 8.0、固定化温度40℃时,制得的固定化酶活性最高。固定化酶最适反应温度为50℃、最适反应pH为7.0。当扁桃腈浓度为10 mmol/L、反应1 h时,固定化酶最大产率为0.041 mmol/(g·h);在反应12 h时,产物e.e.值可达到99%以上。固定化酶重复使用8次以后,酶活力仍保持在45%。  相似文献

Although the importance of the endosperm as a food store inmany angiosperm seeds is well known, its significance duringearly embryogenesis has been neglected. In many interspecifichybrids, and in some other situations, embryos do not developfully and abort. It has often been stated that this is causedby the endosperm failing to conduct sufficient nutrients tothe embryo, but seldom has it been suggested that the endospermactively controls most of the early stages of morphogenesisof the embryo. Information gleaned from a broad survey of theliterature, combined with additional evidence presented here,obtained fromSolanum incanumand interspecific hybrids, indicatethat the endosperm is dynamic and very active in regulatingearly embryo development. This requires highly integrated geneticcontrol of rapidly changing metabolism in the endosperm. Ininterspecific hybrids, lack of coordination may cause unbalancedproduction of growth regulating substances by the endospermand hence abortion of the embryo, or even unregulated productionof nucleases and proteases resulting firstly in autolysis ofthe endosperm and then digestion of the embryo. The endospermmay thus serve to detect inappropriate hybridization of speciesor ploidy levels and so prevent waste of resources by producingseeds that would result in sterile hybrids or unthrifty subsequentgenerations. For most organisms oxygen is essential fo life. When oxygen levels drop below those required to maintain the minimum physiological oxygen requirement of an organism or tissue it is termed hypoxia. To counter act possible deleterious effects of such a state, an immediate molecular response is initiated causing adaptation responses aimed at cell survival. This response is mediated by the hypoxia-inducible factor-1 (HIF-1), which is a heterodimer consisting of an alpha- and a beta-subunit. HIF-1 alpha protein is stabilized under hypoxic conditions and therefore confers selectivity to this response. Hypoxia is characteristic of tumors, mainly because of impaired blood supply resulting from abnormal growth. Over the past few years enormous progress has been made in the attempt to understand how the activation of the physiological response to hypoxia influences neoplastic growth. In this review some aspects of HIF-1 pathway activation in tumors and the consequences for pathophysiology and treatment of neoplasia are discussed.  相似文献

Studies of human constitution and mental features are traditionally among the main concepts of anthropological research. In this research, we attempted to discover connections between morphological and constitutional features and self-esteem in children and adolescents. 综述了白蚁螱客的主要种类、共生关系及相关机制的研究进展。白蚁螱客中,已报道的动物种类达170种。在与动物的共生关系中存在偏利共生(宾主共栖和异种共栖)、互利共生和无关共生三种;在与微生物的共生关系中,存在与内生菌(原生动物、细菌、真菌和放线菌)和外生菌(蚁巢伞菌等)间的互利关系。指出了白蚁与螱客研究中存在的问题,给出了解决方案,并提出了今后可能的研究热点或方向,为白蚁的综合利用(如纤维素酶)及今后研究物种间的协同进化提供了基础资料。  相似文献
