The yield (gramme bioproduct gramme–1 sugar consumed)of plant cells increases on transition from heterotrophic tophotoheterotrophic nutrition and when specific growth rate increases.There are several advantages in using gas analysis to estimateyield over other methods involving wet chemistry. However, applyinggas analysis to reactions involving plant cells is not withoutdifficulty, in particular that of measuring the small differencein partial pressure of carbon dioxide or oxygen between thegas inlet and outlet of the reactor. The experience of the workreported here is that difficulties such as this can be overcomeby using accurate gas analysers and by carefully calibratingthem. In this paper the theory of material balancing is applied tothe growth of higher plant cells (Ocimum basilicum L. sweetbasil) in culture. Three equations are derived relating yieldeither to the rate of carbon dioxide produced, the rate of oxygenconsumed, or their ratio. The validity of these equations andtheir underlying assumptions are tested by comparing yieldsin continuous cultures based on wet chemistry to yields basedon gas analysis. Generally similar yield values were obtainedand these varied between 0.65 and 0.96. Making further assumptions, the analysis was extended to estimatephotosynthetic efficiency in photoheterotrophic conditions.Photosynthetic efficency varied from 0% to 5% and increasedin the following circumstances: on transition from phosphateto fructose limitation and when fructose concentration in thefresh medium was decreased, when specific growth rate was increaseduntil a critical value of 0.64 of the maximum specific growthrate was reached. Although absolute values of photosyntheticefficiency were not verified, the relative changes agreed withother measurements of photosynthetic development (i.e. specificproduction rate of chlorophyll and actual photosynthesis rateunit–1 dry biomass). The lacustrine blue-green alga (= Cyanobacterium) Oscillatoriarubescens D.C. was cultured in chemostats with temperature andlight intensity as the only limiting factors. The experimentswere carried out under combinations of three temperatures 10,20 and 30°C and three irradiances 6, 18 and 30 µEm–2 s–1. Dilution rate, photosynthesis rate anda large number of abiotic and cellular factors were regularlymeasured to determine steady-state periods and the level ofthe associated variables. Various mathematical models were fittedwith series of data by a non-linear regression method derivedfrom Marquardt's method. Four models for calculation of specificgrowth rate and photosynthesis rate are presented. The lastone computes growth rate from calculated photosynthesis: µ= µmax pmaxI/[KpKi + I(Kp + Pmax)], with Pmax =100 + 257Tand K1 = 300/chl a. Values of adjusted parameters are discussed.One conclusion confirms that values of maximal growth ratesand half-saturation constants frequently considered in the literatureas absolute species characteristics should always be consideredas relative to associated prevalent growth conditions.  相似文献

Reddy, A. R. and Das, V. S. R. 1987. Modulation of sucrose contentby fructose 2,6-bisphosphate during photosynthesis in rice leavesgrowing at different light intensities.—J. exp. Bot. 38:828–833. The relationship between the rate of CO2 fixation and sucroseconcentration in the leaves of rice (Oryza sativa L.) grownat different light intensities was investigated. Maximum sucrosecontent coincided with maximum rates of CO2 fixation, achievedat a photon flux density of 1600 µmol m–2 s–1.The levels of sucrose and fructose 2,6-bisphosphate were alsocompared in the leaves under different light intensities. Fructose2,6-Msphosphate accumulated during growth at low light. Theactivity of fructose-6-phosphate 2-kinase was high in the leavesgrown at low light while that of fructose-2,6-bisphosphatasewas low. The activities of phosphoglucose isomerase and phospho-glucomutasewere slightly increased by growth at low light The activitiesof UDP glucose pyrophosphorylase were adversely affected invitro with increased concentrations of fructose 2,6-bisphosphatewhile those of sucrose phosphate synthase were moderately affected.Phosphoglucose isomerase and phosphoglucomutase were activatedby fructose 2,6-bisphosphate (8-0 mmol m–3) by 12-15%.The results suggested that low light intensities during growthresult in an accumulation of fructose 2,6-bisphosphate whichmodulates the key enzymes of sucrose biosynthesis thus regulatingcarbon flow under conditions of limited photosynthesis. The rate of net photosynthesis (P) of whole plant stands oftomato (Lycopersicon esculentum Mill.), cucumber (Cucumis sativusL.) and sweet pepper (Capsicum annuum L.) was measured in sixlong-term experiments in large greenhouses under normal operatingconditions and CO2-concentrations between 200 and 1200 µmolmol-1. The objective was to quantify the responses to lightand carbon dioxide and to obtain data sets for testing simulationmodels. The method of measuring canopy photosynthesis involvedan accurate estimation of the greenhouse CO2 balance, usingnitrous oxide (N2O) as tracer gas to determine, on-line, theexchange rate between greenhouse and outside air. The estimatedrelative error in the observed P was about ± 10%, exceptthat higher relative errors could occur under particular conditions. A regression equation relating P to the photosynthetically activeradiation, the CO2 concentration and the leaf area index explained83-91% of the variance. The main canopy photosynthesis characteristicscalculated with the fitted regression equations were: canopyPmax 5-9 g m-2 h-1 CO2 uptake; ratio Pmax/LAI 1·5-3 gm-2 h-1; light compensation point 32-86 µmol s-1 m-2;light use efficiency (quantum yield) at low light 0·06-0·10µmol µmol-1 and CO2 compensation point 18-54 µmolmol-1. Rates of net photosynthesis of the flag leaves of 15 genotypesof wheat and related species were measured throughout theirlife, using intact leaves on plants grown in the field. At thestage when rates were maximal, they were in general highestfor the diploid species, intermediate for the tetraploidspeciesand lowest for Triticum aestivum (means of 38, 32 and 28 mgCO2 dm–2 h–1 respectively). Rates were stronglynegatively correlated with leaf area, leaf width and the meanplan area per mesophyll cell and positvely correlated with stomatalfrequency and number of veins per mm of leaf width. The differencesamong species in these attributes were mainly related to ploidylevel. It was not possible to determine the relative importanceof each anatomical feature, though the changes in stomatal frequencyhad only slight effects on stomatal conductance and the observeddifferences in rates of photosynthesis were much greater thanwould be expected from those in stomatal conductance alone. There was genetic variation in rates of light dependent oxygenevolution of isolated protoplasts and intact chloroplasts butno difference attributable to ploidy. The mean rate, 91 µmolO2 mg–1 chlorophyll h–1, equivalent to 3.9 mg CO2mg-1chlorophyll h-1 was considerably less than the rate of photosynthesisin comparable intact leaves, which was 7.2 mg CO2 mg–1chlorophyll h–1. The total above-ground dry matter yields were least for thewild diploids T. urartu and T. thauodar and the wild tetraploidT. dicoccoides, but the other wild diploids produced as muchdry matter as the hexaploids. The prospects of exploiting differences in photosynthetic ratein the breeding of higher yielding varieties are discussed. The effects of phthalate esters on chlorophyll a2 fluorescencein radish plants (Raphanus sativus L. cv. Cherry Belle) wereexamined Fluorescence yield was increased in those plants exposedto an aerial concentration of 120 ng dm–3 dibutyl phthaiatc(DBP) at a rate of 3.0 dm3 min–1 for 13 d. Comparisonof fluorescence enhancement ratios and Fred/Fox, suggests thatDBP inhibits photosynthesis in radish plants at a site afterQA. Both DBP and diisobutyl phthalate (DIBP) strongly inhibiteduncoupled (PS2+PS1) electron transport rates in thylakoids isolatedfrom spinach. At a chlorophyll concentration of 10 µgcm–3 the concentrations of DBP and DIBP exhibiting 50%inhibition were 44 mmol m–3 and 42 mmol m–3 respectively.Basal electron transport rates were also inhibited, with 87mmol m–3 of DBP or DIBP producing 50% inhibition. Measurementof photosystcm 1 activity suggested that the main site of actionof these phthalates was localized at a site near the reducingside of photosystem 2. Seasonal changes in the photosynthesis and primary productionof Microcystis aeruginosa Kütz. were investigated in LakeKasumigaura during 1981–1982. Microcystis always showeda light-saturated photosynthesis-light curve. Both Pmax andthe initial slope of the photosynthesis-light curve of Microcystisin early summer were very high, so it was concluded that Microcystisutilized both low and high light intensities efficiently. ThePmax of Microcystis was found to be a function of the watertemperature except in August and September. The linear regressionon the temperature-Pmax relationship discontinued at 11°C,where the Pmax value dropped; Microcystis did not photosynthesizebelow 4°C. The initial slope of the curve was also descendingbelow 11°C. It is suggested that Microcystis changes itsphysiological properties below 11°C. The highest value ofgross production calculated for M. aeruginosa was 5.4 gC m–2d–1 in July; the annual gross production was estimatedto be 300 gC m–2year–1 (i.e., 40% of the total primaryproduction in this lake).  相似文献

Barley (Hordeum vulgare L. cv. Digger) was grown for 22 d inenclosed chambers with a CO2 enrichment of 35, 155, 400 or 675µmol CO2 mol1. CO2 enrichment increased photosyntheticcapacity in the plants grown at either of the two highest levelsof pCO2. A CO2 enrichment of 675µmol CO2 caused a significantincrement of shoot dry weight, whereas no changes were observedin fresh weight, chlorophyll or protein levels. At a light intensityof 860µmol m–2s–1 CO2 enrichment caused photosyntheticcapacity to increase by 250%, whereas no effect was observedat 80 µmol m–2 s–1. Over time, photosynthesisdecreased by 70% independent of CO2. A time-dependent increasein the level of extractable fructose was observed whereas totalextractable carbohydrate only changed slightly. The contribution of HCO3to photosynthetic O2 evolutionin the seagrass Zostera muelleri Irmisch ex Aschers. increasedwith increasing salinity of the bathing seawater when the inorganiccarbon concentration was kept constant. K1/2 (seawater salts)for HCO3 -dependent photosynthesis was 66% of seawatersalinity. Both short- and long-term pretreatment at low salinitiesstimulated photosynthesis in full strength seawater. Twentyfour hours pre-incubation of seagrass plants in 3·0 molm–3 NaHCO3 resulted in increased photosynthesis at allsalinities, apparently due to stimulation of HCO3 use(K1/2 (seawater salts) = 26%). Vmax (HCO3) was not affectedby low salinity pretreatment. The kinetics of HCO3 stimulationby the major seawater cations was investigated. Ca2+ was themost effective cation with the highest Vmax (HCO3) andwith K1/2(Ca2+) = 14 mol m–3. Mg2+ was also very effectiveat less than 50 mol m–3 but higher concentrations wereinhibitory. This inhibition cannot be accounted for solely byprecipitation of MgCO3. Na+ and K+ were both capable of stimulatingHCO3 use. Stimulation was in two distinct parts. Up to500 mol m–3, both citrate and chloride salts gave similarresults (K1/2(Na+) 81 mol m–3, Vmax(HCO3) 0·26µmol O2 mg–1 chl min–1), but use of citratesalts above 500 mol m–2 caused a second stimulation ofHCO3 use (K1/2(Na+) 830 mol m–3, Vmax(HCO3)0·68 µmol O2 mg–1 chl min–1). Vmax(HCO3)for the second-phase Na+ or K+ stimulation was of the same orderas for Ca2+-stimulated HCO3 use. To further characterizesalt-dependent HCO3 use, the sensitivity of photosynthesisto Tris and TES buffers was investigated. The effects of Trisappear to be due to the action of Tris+ causing stimulationof HCO3 -dependent photosynthesis in the absence of salt,but inhibition of HCO3 use in saline media. TES has noeffect on photosynthesis. External carbonic anhydrase, althoughimplicated in salt-dependent HCO3 use in Z. muelleri,could not be detected in whole leaves. Species-specific differences in the assimilation of atmosphericCO2 depends upon differences in the capacities for the biochemicalreactions that regulate the gas-exchange process. Quantifyingthese differences for more than a few species, however, hasproven difficult. Therefore, to understand better how speciesdiffer in their capacity for CO2 assimilation, a widely usedmodel, capable of partitioning limitations to the activity ofribulose-1,5-bisphosphate carboxylase-oxygenase, to the rateof ribulose 1,5-bisphosphate regeneration via electron transport,and to the rate of triose phosphate utilization was used toanalyse 164 previously published A/Ci, curves for 109 C3 plantspecies. Based on this analysis, the maximum rate of carboxylation,Vcmax, ranged from 6µmol m–2 s–1 for the coniferousspecies Picea abies to 194µmol m–2 s–1 forthe agricultural species Beta vulgaris, and averaged 64µmolm–2 s–1 across all species. The maximum rate ofelectron transport, Jmax, ranged from 17µmol m–2s–1 again for Picea abies to 372µmol m–2 s–1for the desert annual Malvastrum rotundifolium, and averaged134µmol m–2 s–1 across all species. A strongpositive correlation between Vcmax and Jmax indicated that theassimilation of CO2 was regulated in a co-ordinated manner bythese two component processes. Of the A/Ci curves analysed,23 showed either an insensitivity or reversed-sensitivity toincreasing CO2 concentration, indicating that CO2 assimilationwas limited by the utilization of triose phosphates. The rateof triose phosphate utilization ranged from 4·9 µmolm–2 s–1 for the tropical perennial Tabebuia roseato 20·1 µmol m–2 s–1 for the weedyannual Xanthium strumarium, and averaged 10·1 µmolm–2 s–1 across all species. Despite what at first glance would appear to be a wide rangeof estimates for the biochemical capacities that regulate CO2assimilation, separating these species-specific results intothose of broad plant categories revealed that Vcmax and Jmaxwere in general higher for herbaceous annuals than they werefor woody perennials. For annuals, Vcmax and Jmax averaged 75and 154 µmol m–2 s–1, while for perennialsthese same two parameters averaged only 44 and 97 µmolm2 s–1, respectively. Although these differencesbetween groups may be coincidental, such an observation pointsto differences between annuals and perennials in either theavailability or allocation of resources to the gas-exchangeprocess. Negative linear relations were detected (P < 0·005)between the rate of progress from sowing to panicle initiationand CO2 concentration (210-720 µmol CO2 mol-1 air) fortwo genotypes of sorghum [Sorghum bicolor (L.) Moench]. Relationsbetween CO2 concentration and the rate of progress from sowingto first flowering were also negative in soyabean [Glycine max(L.) Merrill] (P < 0·025), but positive in cowpea[Vigna unguiculata (L.) Walp.] (P < 0·025), albeitthat in both grain legumes sensitivity was much less than insorghum. Thus CO2 elevation does not delay flowering in allshort-day species. The considerable effect of CO2 concentrationon times to panicle initiation resulted in large differencesamong the sorghum plants at this developmental stage; with increasein CO2 concentration, plants were taller with slightly moreleaves and more pronounced apical extension. At the same timeafter sowing however, sorghum plants were heavier (P < 0·05)at 210 than at 360 µmol CO2 mol-1 air. In contrast, relationsbetween the dry masses of the soyabean and cowpea plants andCO2 concentration were positive and curvilinear (P < 0·05).It is suggested that the impact of global environmental changecould be severe for sorghum production in the semi-arid tropics.Copyright1995, 1999 Academic Press Sorghum bicolor (L.) Sodium chloride, at a concentration of 88 mol m-3in half strengthHoagland nutrient solution, increased dry weight per unit areaofXanthium strumarium L. leaves by 19%, and chlorophyll by 45%compared to plants grown without added NaCl at ambient (350µmol mol-1) CO2concentration. Photosynthesis, per unitleaf area, was almost unaffected. Even so, over a 4-week period,growth (dry weight increment) was reduced in the salt treatmentby 50%. This could be ascribed to a large reduction in leafarea (>60%) and to an approx. 20% increase in the rate ofdark respiration (Rd). Raising ambient [CO2] from zero to 2000 µmol mol-1decreasedRd in both control and salinized plants (by 20% at 1000, andby 50% at 2000 µmol mol-1CO2concentration) compared toRd in the absence of ambient CO2. High night-time [CO2] hadno significant effect on growth of non-salinized plants, irrespectiveof day-time ambient [CO2]. Growth reduction caused by salt wasreduced from 51% in plants grown in 350 µmol mol-1throughoutthe day, to 31% in those grown continuously in 900 µmolmol-1[CO2]. The effect of [CO2] at night on salinized plants depended onthe daytime CO2concentration. Under 350 µmol mol-1day-time[CO2], 900 µmol mol-1at night reduced growth over a 4-weekperiod by 9% (P <0.05) and 1700 µmol mol-1reduced itby 14% (P <0.01). However, under 900 µmol mol-1day-time[CO2], 900vs . 350 µmol mol-1[CO2] at night increasedgrowth by 17% (P <0.01). It is concluded that there is both a functional and an otiose(functionless) component to Rd, which is increased by salt.Under conditions of low photosynthesis (such as here, in thelow day-time [CO2] regime) the otiose component is small andhigh night-time [CO2] partly suppresses functional Rd, therebyreducing salt tolerance. In plants growing under conditionswhich stimulate photosynthesis (e.g. with increased daytime[CO2]), elevated [CO2] at night suppresses mainly the otiosecomponent of respiration, thus increasing growth. Consequently,in regions of adequate water and sunlight, the predicted furtherelevation of the world atmospheric [CO2] may increase plantsalinity tolerance. Primary production was measured for 7 years, using the in situ14C-method in hypertrophic Hartbeespoort Dam, South Africa,to examine the influence of light and water temperature on theupper limit of Microcystis aeruginosa production. Water temperaturesvaried from 11 to >25°C and chlorophyll concentrationsreached 6500 mg m–3. The maximum volumetric rate of production(Amax) was 12->8800 mg C m–3 h–1 with areal productions(A) of 69->3300 mg C m–2 h–1 for euphotic zonedepths of <0.5–8.4 m. The intrinsic parameters of phytoplanktonproduction (, Amax/B, Ik) indicated that the phytoplankton populationwas adapted to high light levels. Both Amax/B and Ik were correlatedwith temperature. Under optimal conditions, , the theoreticalupper limit of A, was calculated to be 2.8 g Cm–2 h–1,while the measured rate was 2.5 g Cm–2 h–1. Measuredareal rates exceeding were overestimated due to methodologicalproblems when working with Microcystis scums. Light and watertemperature interacted to yield high production rates: watertemperature through its direct effect on photosynthetic ratesand indirectly in the formation of diurnal mixed layers; lightindirectly through water temperature and directly through itsattenuation and induction of light-adapted physiology in Microcystis.  相似文献

The cell quotas of microcystin (Qmcyst), protein (Qprot), chlorophylla (Qchloro) and carbohydrate (Qcarbo), as well as the net productionrates of these parameters, were determined during the exponentialand stationary phases in nine batch cultures of Microcystisaeruginosa (CYA 228) at light regimes from 33 to 53 µmolphotons m–2 s–1. The following results were obtained.(i) A parallel pattern was found in the changes of Qmcyst, Qprot,Qchloro and Qcarbo during the entire growth cycle and significantcorrelations were recorded between Qmcyst and Qprot, Qchloroand Qcarbo. (ii) The net microcystin production rate (µmcyst)was positively correlated with the specific cell division rate(µc), the chlorophyll production rate (µchloro)and the protein production rate. (iii) A significant inverselinear relationship was found between µc and Qmcyst, i.e.cultures with a positive µc had a Qmcyst between 110 and400 fg microcystin cell–1, while declining cultures hadQmcyst values >400 fg microcystin cell–1. Maximum variationin Qmcyst within cultures was 3.5-fold. Collectively, the resultsshow that cells produced microcystin at rates approximatingthose needed to replace losses to daughter cells during divisionand that microcystin was produced in a similar way to proteinand chlorophyll, indicating a constitutive microcystin production.  相似文献

A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

Phytoplankton photosynthesis parameters in central Canadian lakes   总被引:1,自引:0,他引:1  
Values for two biological parameters — (i) PBm, the lightsaturated rate of photosynthesis per unit of chlorophyll and(ii) , the slope of the light limited part of the photosynthesisversus light curve per unit of chlorophyll — must be knownin order to be able to estimate rates of phytoplankton primaryproduction from chlorophyll data. These parameters were measuredfor periods of up to 9 years in central Canadian lakes locatedin temperate, subarctic and arctic climatic zones. Regardlessof their geographic locations, lakes where integral photosynthesiswas nutrient limited had lower values of these parameters thandid lakes where integral photosynthesis was light limited. Temperatureset an upper limit to the variability of PBm but was not a goodpredictor of its actual value. Year-to-year variability of photosynthesisparameters in the most intensively studied group of lakes waslarge: annual means varied by a factor of three over a 9-yearperiod. Until the sources and extent of this variability areknown, accurate production estimates can be obtained from chlorophylldata only if PBm and are measured in each water body and inevery year. Implications for estimating primary production fromremotely-sensed chlorophyll data are discussed.  相似文献

The relationships between photosynthesis, dry matter accumulationand translocation have been studied during the development ofthe first true leaf of cucumber. The leaf was grown in an irradianceof 50W m–2 photosynthetically active radiation for 10h–1 at 20 C and 2 g m–3 CO2. The maximum rate of net photosynthesis, on a leaf area basis,occured at full expansion. Photochemical efficiency, based onincident radiation, also increased up to this stage and wasrelated to the concentration of chlorophyll in the leaf. Darkrespiration and the light compensation point fell over the wholeperiod of leaf expansion. A carbon budget analysis showed that the rate of carbon accumulationin the leaf reached a peak at 70 percent expansion. The leafchanged from a net importer to a net exporter of carbon whenit was about 30 percent expanded. The rate of export increasedwith leaf expansion (and with net photosynthesis) and was twiceas high in the day an in the night at full expansion. At fullleaf expansion there was a reduction in the amount of starchlost overnight, and the carbon exported amounted to 80 per centof the daily net carbon fixed. Barley (Hordeum vulgare L., cvs Golf, Mette, and Laevigatum)was grown under nitrogen limitation in solution culture untilnear maturity. Three different nitrogen addition regimes wereused: in the ‘HN’ culture the relative rate of nitrate-Naddition (RA) was 0·08 d–1 until day 48 and thendecreased stepwise to, finally, 0·005 d–1 duringgrain-filling; the ‘LN’ culture received 45% ofthe nitrogen added in HN; the ‘CN’ culture was maintainedat RA 0·0375 d–1 throughout. Kinetics of net nitrateuptake were measured during ontogeny at 30 to 150 mmol m–3external nitrate. Vmax (which is argued to reflect the maximuminflux rate in these plants) declined with age in both HN andLN cultures. A pronounced transient drop was observed just beforeanthesis, which correlated in time with a peak in root nitrateconcentration. Similar, but less pronounced, trends were observedin CN. The relative Vmax (unit nitrogen taken up per unit nitrogenin plants and day) in all three cultures declined from 1·3–2·3d–1 during vegetative growth to 0·1–0·7d–1 during generative growth. These values are in HN andLN cultures 15- to more than 100-fold in excess of the demandset by growth rates throughout ontogeny. Predicted balancingnitrate concentrations (defined as the nitrate concentrationrequired to support the observed rate of growth) were below6·0 mmol m–3 in HN and LN cultures before anthesisand then decreased during ontogeny. In CN cultures the balancingnitrate concentration increased during grain-filling. Apartfrom the transient decline during anthesis, most of the effectof ageing on relative Vmax can be explained in terms of reducedcontribution of roots to total biomass (R:T). The loss in uptakeper unit root weight is largely compensated for by the declinewith time in average tissue nitrogen concentrations. The quantitativerelationships between relative Vmax and R:T in ageing plantsare similar to those observed for vegetative plants culturedat different RAs. The data support the contention that the capacity for nitrateacquisition in N-limited plants is under general growth control,rather than controlled by specific regulation of the biochemicalpathway of nitrate assimilation. Mistletoes usually have slower rates of photosynthesis thantheir hosts. This study examines CO2assimilation, chlorophyllfluorescence and the chlorophyll content of temperate host–parasitepairs (nine hosts parasitized by Ileostylus micranthus and Carpodetusserratus parasitized by Tupeia antarctica). The hosts of I.micranthus had higher mean annual CO2assimilation (3.59 ±0.41 µmol m-2 s-1) than I. micranthus(2.42 ± 0.20µmol m-2 s-1), and C. serratus(2.41 ± 0.43 µmolm-2 s-1) showed higher CO2assimilation than T. antarctica(0.67± 0.64 µmol m-2 s-1). Hosts saturated at significantlyhigher electron transport rates (ETR) and light levels thanmistletoes. The positive relationship between CO2assimilationand electron transport suggests that the lower CO2assimilationrates in mistletoes are a consequence of lower electron transportrates. When photosynthetic rates, ETR and chlorophyll a /b ratioswere adjusted for photosynthetically active radiation, hostsdid not have significantly higher CO2assimilation (3.21 ±0.37 µmol m-2 s-1) than mistletoes (2.54 ± 0.41µmol m-2 s-1), but still had significantly higher ETRand chlorophyll a / b ratios. The electron transport rates,saturating light and chlorophyll a / b ratios of sun leavesfrom mistletoes were similar to host shade leaves. These responsesindicate that in comparison with their hosts, mistletoe leaveshave the photosynthetic characteristics of the leaves of shadeplants. Tomato plants were grown in solution culture in a controlledenvironment at 20 ?C with a 12 h photoperiod of 400 µmolquanta m–2 s–1 PAR with either normal ambient CO2,approximately 340 vpm, or with 1000 vpm CO2. The short- andlong-term effects of CO2 enrichment on photosynthesis were determinedtogether with the levels of ribulose-1, 5-bisphosphate carboxylase(RuBPco) E.C. 4.1.1.39 [EC] protein and activity throughout leafdevelopment of the unshaded 5th leaf above the cotyledons. Thehigh CO2 concentration during growth did not appreciably affectthe rate of leaf expansion or final leaf area but did increasethe fresh weight per unit area of leaf. With short-term CO2enrichment, i.e. only during the photosynthesis measurements,the light-saturated photosynthetic rate (Pmax) of young leavesdid not increase while those reaching full expansion more thandoubled their net rate of CO2 fixation. However, with longerterm CO2 enrichment, i.e. growing the crop in high CO2, theplants did not maintain this photosynthetic gain. While theCO2 concentration during growth did not affect the peak in Pmaxmeasured in 300 vpm CO2 or Pmax in 1000 vpm CO2, RuBPco proteinor its activity, the subsequent ontogenetic decline in theseparameters was greatly accelerated by the high CO2 treatment.Compared with plants grown in normal ambient CO2 the high CO2grown leaves, when almost fully expanded, contained only approximatelyhalf as much RuBPco protein and Pmax in 300 vpm CO2 and Pmaxin1000 vpm CO2 were similarly reduced. The loss of RuBPco proteinmay be a major factor associated with the accelerated fall inPmax since it was close to that predicted from the amount andkinetics of RuBPco assuming RuBP saturation. In the oldest leavesexamined grown in high CO2 additional factors may be limitingphotosynthesis since RuBPco kinetics marginally overestimatedPmax in 300 vpm CO2 and the initial slope of photosynthesisin response to intercellular CO2 was also less than expectedfrom the extractable RuBPco.
