The effect of an arbuscular mycorrhizal fungus “AMF” (Glomus constrictum Trappe) on growth, pigments, and phosphorous content of marigold (Tagetes erecta) plant grown under different levels of drought stress was investigated. The applied drought stress levels reduced growth vigor (i.e. plant height, shoot dry weight, flower diameter as well as its fresh and dry weights) of mycorrhizal and non-mycorrhizal plant as compared to control plant (non-drought stressed plant). The presence of mycorrhizal fungus, however, stimulated all growth parameters of the treated plant comparing to non-mycorrhizal treated plant. The photosynthetic pigments (carotene in flowers and chlorophylls a and b in leaves) were also stimulated by the mycorrhizal fungi of well-watered as well as of water-stressed plants. The total pigments of mycorrhizal plants grown under well-watered conditions were higher than those of non-mycorrhizal ones by 60%. In most cases, drought-stressed mycorrhizal plants were significantly better than those of the non-mycorrhizal plants. So, the overall results suggest that mycorrhizal fungal colonization affects host plant positively on growth, pigments, and phosphorous content, flower quality and thereby alleviates the stress imposed by water with holding.  相似文献

Auxins (Aux) are primary growth regulators that regulate almost every aspect of growth and development in plants. It plays a vital role in various plant processes besides controlling the key aspects of cell division, cell expansion, and cell differentiation. Considering the significance of Aux, and its potential applications, a study was conducted to observe the impact of indole acetic acid (IAA), a most active and abundant form of Aux on Brassica juncea plants growing under natural environmental conditions. Different concentrations (0, 10−10, 10−8, 10−6 M) of IAA were applied once in a day at 25-day stage of growth for 5 days, consecutively. Various parameters (growth, photosynthetic, biochemical, oxidative biomarkers and nutrient composition) were assessed at different days after sowing (DAS). Scanning electron microscopy (SEM) of leaf stomata, reactive oxygen species (ROS) localization in leaf and roots, and confocal microscopy were also conducted. The results revealed that all the IAA concentrations were effective in growth promotion and ROS reduction, however, the 10−8 M of IAA exhibited the maximum improvement in all the above mentioned parameters as compared to the control.  相似文献

Melatonin was found in the fresh water characeae Chara australis. The concentrations (~4 μg/g of tissue) were similar in photosynthesizing cells, independent of their position on the plant and rhizoids (roots) without chloroplasts. Exogenous melatonin, added at 10 μM to the artificial pond water, increased quantum yield of photochemistry of photosystem II by 34%. The increased efficiency appears to be due to the amount of open reaction centers of photosystem II, rather than increased efficiency of each reaction center. More open reaction centers reflect better functionality of all photosynthetic transport chain constituents. We suggest that melatonin protection against reactive oxygen species covers not only chlorophyll, but also photosynthetic proteins in general.  相似文献

This study characterized the morphological and photosynthetic responses of two wetland plant species when they were subject to 2–6°C fluctuations in growth temperature and ±50% of precipitation, in order to predict the evolution of natural wetlands in Sanjiang Plain of North-eastern China. We investigated the morphological and photosynthetic responses of two dominant and competitive boreal freshwater wetland plants in Northeastern China to manipulation of warming (ambient, +2.0°C, +4.0°C, +6.0°C) and altered precipitation (−50%, ambient, +50%) simultaneously by incubating the plants from seedling to senescence within climate-controlled environmental chambers. Post-harvest, secondary growth of C. angustifolia was observed to explore intergenerational effects. The results indicated that C. angustifolia demonstrated a greater acclimated capacity than G. spiculosa to respond to climate change due to higher resistance to temperature and precipitation manipulations. The accumulated effect on aboveground biomass of post-harvest secondary growth of C. angustifolia was significant. These results explain the expansion of C. angustifolia during last 40 years and indicate the further expansion in natural boreal wetlands under a warmer and wetter future. Stability of the natural surface water table is critical for the conservation and restoration of G. spiculosa populations reacting to encroachment stress from C. angustifolia expansion.  相似文献

Our objective was to evaluate the role of plant growth-promoting bacteria to protect maize (Zea mays L.) plants against salt damage. Bacillus aquimaris DY-3 based on their 16S rDNA sequences, the most tolerant to salinity and the synthesis of indole acetic acid was selected for further studies. Strain was inoculated on maize roots growing in sterilized sand under salt stress conditions (1% NaCl). After one week, plant growth was promoted by bacterial inoculation regardless of salt stress and non-salt stress. Chlorophyll content, leaf relative water content, accumulation of proline, soluble sugar and total phenolic compound, and activities of superoxide dismutase, catalase, peroxidase and ascorbate peroxidase were enhanced, while lipid peroxidation levels and Na+ content were decreased. The results showed that B. aquimaris DY-3 alleviated the salt stress in maize, likely through the integration of the antioxidant enzymes and the non-antioxidant systems that improve the plant response. Hence, the application of indole acetic acid synthesizing plant growth-promoting bacteria may represent an important alternative approach to decrease the impact of salt stress on crops.  相似文献

An efficient callus induction and plant regeneration system has been standardized for an ethnomedicinal plant, Elephantopus scaber Linn. Two explants i. e. seeds and leaf segments were used for callus induction. Murashige and Skoog (MS) medium supplemented with 5.0 μM 2, 4-dichlorophenoxy acetic acid (2, 4-D) and 0.5 μM kinetin (Kn) gave the optimum frequency (89 %) of callus induction from seed explant. The results showed that the highest response in terms of percent callus regenerating (91 %) and number of shoots (56) per culture was recorded on MS medium supplemented with 6.0 μM N6-benzylaminopurine (BA) and 1.5 μM α naphthalene acetic acid (NAA). The best rooting of regenerated shoots was obtained on half strength MS medium supplemented with 6.0 μM indole-3- butyric acid (IBA). On this medium, 100 % of the shoots produced roots with a mean number of 3.2 roots per shoot. The positive role of vesicular arbuscular mycorrhizae (VAM) along with potting mix has been well established in the present study. Of the various potting mix employed for plant acclimatization, the highest response of 100 % plant survival was noticed when autoclaved garden soil, sand (2:1) and VAM was utilized as potting mix. Inter-simple sequence repeats (ISSR) were used to establish the clonal fidelity of regenerated plantlets and the banding profiles from callus derived plants were monomorphic and similar to those of mother plant, thus ascertaining the true-to-type nature of these plants.  相似文献

Cotton (Gossypium hirsutum L.) plants grown under field water deficits exhibited an 80 to 85% reduction in leaf area index, plant height, and dry matter accumulation compared with irrigated controls. Midday photosynthetic rates of dryland plants decreased 2-fold, and canopy temperatures increased to 40°C at 80 days after planting compared with canopy temperatures of 30°C for irrigated plants. Leaves from dryland plants which had exhibited canopy temperatures of 40°C for several weeks accumulated stainable levels of polypeptides with apparent molecular weights of 100, 94, 89, 75, 60, 58, 37, and 21 kilodaltons. These polypeptides did not accumulate in leaves from irrigated plants.

Addition of [35S]methionine to leaves of growth chamber-grown cotton plants and subsequent incubation at 40°C for 3 hours radiolabeled polypeptides with molecular weights similar to those that accumulate in dryland cotton leaves. These data suggest that the proteins which accumulate in water-stressed cotton leaves at elevated temperatures (40°C) are heat shock proteins and that these proteins can accumulate to substantial levels in field-stressed plants.

The members of the Scabiosa genus are one of the traditional medicinal plants used in the treatment of many diseases, in particular the treatment of scabies. In this study, it was aimed to determine antioxidant activities and chemical composition of methanolic extracts of leaves and flowers of Scabiosa columbaria subsp. columbaria var. columbaria. The phenolic contents of both parts of the plant were analyzed by LC–MS/MS. A total of 6 phenolic compounds were determined and chlorogenic acid was the major compound in both flower and leaf parts of the plants, with 5936.052 µg/g and 8021.666 µg/g, respectively. 6 different methods were used to determine the antioxidant activity of the plant parts. Both leaf and flower parts of the plant showed high antioxidant activity in all tested methods and the antioxidant activity values of the leaf part were measured higher than those of the flower part for four tests. The methanol extracts of the plant parts was analyzed with GC–MS and number of the essential oil compounds in the leaf and flower parts were determined as 17 and 13, respectively. Linalool compound was also found to be common in both parts of the plant. The major compounds of the essential oils were identified as 4-Octadecenal (30.01%) in the flower and carvone (35.44%) in the leaf. In addition, terpene derivatives was determined as 90.32% of the highest essential oil group in the leaf, while this value was determined as 1.42% in the flower. For the flower, aromatics were determined as the main component group with 21.31%.  相似文献

Azospirillum brasilense, a nitrogen-fixing bacterium found in the rhizosphere of various grass species, was investigated to establish the effect on plant growth of growth substances produced by the bacteria. Thin-layer chromatography, high-pressure liquid chromatography, and bioassay were used to separate and identify plant growth substances produced by the bacteria in liquid culture. Indole acetic acid and indole lactic acid were produced by A. brasilense from tryptophan. Indole acetic acid production increased with increasing tryptophan concentration from 1 to 100 μg/ml. Indole acetic acid concentration also increased with the age of the culture until bacteria reached the stationary phase. Shaking favored the production of indole acetic acid, especially in a medium containing nitrogen. A small but biologically significant amount of gibberellin was detected in the culture medium. Also at least three cytokinin-like substances, equivalent to about 0.001 μg of kinetin per ml, were present. The morphology of pearl millet roots changed when plants in solution culture were inoculated. The number of lateral roots was increased, and all lateral roots were densely covered with root hairs. Experiments with pure plant hormones showed that gibberellin causes increased production of lateral roots. Cytokinin stimulated root hair formation, but reduced lateral root production and elongation of the main root. Combinations of indole acetic acid, gibberellin, and kinetin produced changes in root morphology of pearl millet similar to those produced by inoculation with A. brasilense.  相似文献

Studies were conducted that supported the hypothesis that the mutation to the psbA plastid gene that confers S-triazine resistance (R) in Brassica napus also results in an altered diurnal pattern of photosynthetic carbon assimilation (A) relative to that of the susceptible (S) wild type, and that these patterns change over the ontogeny of a plant. Photosynthetic photon flux density, under closely controlled environmental conditions, was incrementally increased and decreased on either side of the midday maxima of 1150 to 1300 μmol quanta m−2 s−1. In all experiments, A approximately tracked the increasing and decreasing diurnal light levels. Younger (3- to 4-leaf) R plants had greater photosynthetic rates early and late in the diurnal light period, whereas those of S plants were greater during midday as well as during the photoperiod as a whole. These relative photosynthetic characteristics of R and S plants changed in several ways with ontogeny. As the plants aged during the vegetative phase of development, S plants gradually assimilated more carbon in the early, and then in the late, part of the day. At the end of the vegetative phase of development, R plant carbon assimilation was less relative to S plants at most times of the day, and was never greater. This relationship between the two biotypes dramatically changed with the onset of the reproductive phase (8½ to 9½ leaf) of plant development: R plants assimilated more carbon than S plants during all periods of the diurnal light period with the exception of the late part of the day. In addition to these differences in A, R plant stomatal function differed from that in S plants. R plant leaves were always cooler than S plant leaves under the same environmental and diurnal conditions. Correlated with this difference in leaf temperature were equal or greater total conductances to water vapor and intercellular CO2 partial pressures in R compared to S leaves in most instances. These studies indicate a more complex pattern of photosynthetic carbon assimilation than previously observed. The photosynthetic superiority of one biotype relative to the other was a function of the time of day and the age of the plant. These studies also suggest that R plants may have an adaptive advantage over S plants in certain unfavorable ecological niches independent of the presence of S-triazine herbicides, such as cool, low-light environments early and late in the day, as well as late in the plants' development. This advantage could result in R biotypes appearing in populations of a species in greater numbers than plastidic mutation alone could cause.  相似文献

Experiment was carried out to determine the effect of Sclerotinia sclerotiorum on the disease development, growth, oil yield and biochemical changes in the plants of Mentha arvensis. With the increase in initial inoculum levels of S. sclerotiorum a corresponding decrease in plant fresh and dry weights were recorded. The maximum reduction in the shoot-roots/suckers fresh weight and shoot-roots/suckers dry weights (39.8%, 43.6%, 40.3% and 42.9%), respectively, was observed at the highest initial inoculum level of 12 g fungal mycelium/5 kg soil as compared to uninoculated control. The infection of roots and suckers due to S. sclerotiorum increased with increasing initial inoculum levels. At the lowest initial inoculum (1.0 g mycelium/5 kg soil), infection was observed 18.0% and at the highest (12 g mycelium/5 kg soil), it was 80.2%. Significant (P ⩽ 0.01) reduction in oil yield, total chlorophyll, total phenol and total sugar content of M. arvensis plants was observed at the lowest inoculum level as compared to uninoculated control.  相似文献

Withania somnifera has been an important herb in the Ayurvedic and indigenous medical systems for centuries in India. However, these grow as weeds mostly in the wastelands, which receive contaminated water from municipal and industrial sources. In the present investigation, plants of Withania somnifera were exposed to various concentrations of arsenate (AsV) and arsenite (AsIII) (0, 10, 25, 50, 100 μM) for 10 days and analysed for accumulation of arsenic (As) and physiological and biochemical changes. Plants showed more As accumulation upon exposure to AsIII (320 μg g−1 DW in roots and 161 μg g−1 DW in leaves) than to AsV (173 μg g−1 DW in roots and 100 μg g−1 DW in leaves) after 10 days of treatment. Consequently, AsIII exposure caused more toxicity to plants as compared to that AsV, as evaluated in terms of the level of photosynthetic pigments and oxidative stress parameters (superoxide, hydrogen peroxide and lipid peroxidation), particularly at higher concentrations and on longer durations. Plants could tolerate low concentrations (variable for AsIII and AsV) until longer durations (10 days) and high concentrations for shorter durations (1–5 days) through increase in antioxidant enzymes and by augmented synthesis of thiols. In conclusion, As tolerance potential of Withania plants on one hand advocates its prospective use for remediation under proper supervision and on the other demonstrates possible threat of As entry into humans due to medicinal uses.  相似文献

Opuntia ficus-indica, a Crassulacean acid metabolism plant cultivated for its fruits and cladodes, was used to examine chemical and physiological events accompanying low-temperature acclimation. Changes in osmotic pressure, water content, low molecular weight solutes, and extracellular mucilage were monitored in the photosynthetic chlorenchyma and the water-storage parenchyma when plants maintained at day/night air temperatures of 30/20°C were shifted to 10/0°C. An increase in osmotic pressure of 0.13 megapascal occurred after 13 days at 10/0°C. Synthesis of glucose, fructose, and glycerol accounted for most of the observed increase in osmotic pressure during the low-temperature acclimation. Extracellular mucilage and the relative apoplastic water content increased by 24 and 10%, respectively, during exposure to low temperatures. These increases apparently favor the extracellular nucleation of ice closer to the equilibrium freezing temperature for plants at 10/0°C, which could make the cellular dehydration more gradual and less damaging. Nuclear magnetic resonance studies helped elucidate the cellular processes during ice formation, such as those revealed by changes in the relaxation times of two water fractions in the chlorenchyma. The latter results suggested a restricted mobility of intracellular water and an increased mobility of extracellular water for plants at 10/0°C compared with those at 30/20°C. Increased mobility of extracellular water could facilitate extracellular ice growth and thus delay the potentially lethal intracellular freezing during low-temperature acclimation.  相似文献

The rate of protein degradation in Zea mays leaves has been estimated by using tritiated water and [3H]acetic anhydride as the labeling agents. Both methods circumvent many of the problems usually associated with measuring protein degradation in plants. The half-life of ribulose-1,5-bisphosphate carboxylase protein in second leaves of 13-day-old seedlings under continuous light was found to be 7.8 ± 0.9 days by the tritiated water technique and 6.5 ± 0.8 days by the [3H]acetic anhydride method. The half-lives determined under a 14-hour-light, 10-hour-dark photoperiod are 6.2 ± 0.8 days with tritiated water and 5.4 ± 0.4 days with [3H]acetic anhydride. Whereas the values obtained by the two methods do not differ significantly, the use of either method for the determination of protein half-life can be recommended.  相似文献

Ultraviolet-B radiation (UVBR) affects plants in many important ways, including reduction of growth rate and primary productivity, and changes in ultrastructures. Rice (Oryza sativa) is one of the most cultivated cereals in the world, along with corn and wheat, representing over 50 % of agricultural production. In this study, we examined O. sativa plants exposed to ambient outdoor radiation and laboratory-controlled photosynthetically active radiation (PAR) and PAR + UVBR conditions for 2 h/day during 30 days of cultivation. The samples were studied for morphological and ultrastructural characteristics, and physiological parameters. PAR + UVBR caused changes in the ultrastructure of leaf of O. sativa and leaf morphology (leaf index, leaf area and specific leaf area, trichomes, and papillae), plant biomass (dry and fresh weight), photosynthetic pigments, phenolic compounds, and protein content. As a photoprotective acclimation strategy against PAR + UVBR damage, an increase of 66.24 % in phenolic compounds was observed. Furthermore, PAR + UVBR treatment altering the levels of chlorophylls a and b, and total chlorophyll. In addition, total carotenoid contents decreased after PAR + UVBR treatment. The results strongly suggested that PAR + UVBR negatively affects the ultrastructure, morphology, photosynthetic pigments, and growth rates of leaf of O. sativa and, in the long term, it could affect the viability of this economically important plant.  相似文献

Groundwater is increasingly becoming a permanent and steady water source for the growth and reproduction of desert plant species due to the frequent channel cutoff events in arid inland river basins. Although it is widely acknowledged that the accessibility of groundwater has a significant impact on plant species maintaining their ecological function, little is known about the water use strategies of desert plant species to the groundwater availability in Daryaboyi Oasis, Central Tarim Basin. This study initially determined the desirable and stressing groundwater depths based on ecological and morphological parameters including UAV‐based fractional vegetation cover (FVC) images and plant growth status. Then, leaf δ13C values of small‐ and big‐sized plants were analyzed to reveal the water use strategies of two dominant woody species (Populus euphratica and Tamarix ramosissima) in response to the groundwater depth gradient. The changes in FVC and growth status of plants suggested that the actual groundwater depth should be kept at an appropriate range of about 2.1–4.3 m, and the minimum groundwater depth should be less than 7 m. This will ensure the protection of riparian woody plants at a normal growth state and guarantee the coexistence of both plant types. Under a desirable groundwater condition, water alternation (i.e., flooding and rising groundwater depth) was the main factor influencing the variation of plant water use efficiency. The obtained results indicated that big‐sized plants are more salt‐tolerant than small ones, and T. ramosissima has strong salt palatability than Peuphratica. With increasing groundwater depth, P. euphratica continuously decreases its growth status to maintain hydraulic efficiency in drought condition, while T. ramosissima mainly increases its water use efficiency first and decreases its growth status after then. Besides, in a drought condition, T. ramosissima has strong adaptability than Peuphratica. This study will be informative for ecological restoration and sustainable management of Daryaboyi Oasis and provides reference materials for future research programs.  相似文献

A potential may exist for survival of and resistance development by Escherichia coli O157:H7 in environmental niches of meat plants applying carcass decontamination interventions. This study evaluated (i) survival or growth of acid-adapted and nonadapted E. coli O157:H7 strain ATCC 43895 in acetic acid (pH 3.6 ± 0.1) or in water (pH 7.2 ± 0.2) fresh beef decontamination runoff fluids (washings) stored at 4, 10, 15, or 25°C and (ii) resistance of cells recovered from the washings after 2 or 7 days of storage to a subsequent lactic acid (pH 3.5) stress. Corresponding cultures in sterile saline or in heat-sterilized water washings were used as controls. In acetic acid washings, acid-adapted cultures survived better than nonadapted cultures, with survival being greatest at 4°C and lowest at 25°C. The pathogen survived without growth in water washings at 4 and 10°C, while it grew by 0.8 to 2.7 log cycles at 15 and 25°C, and more in the absence of natural flora. E. coli O157:H7 cells habituated without growth in water washings at 4 or 10°C were the most sensitive to pH 3.5, while cells grown in water washings at 15 or 25°C were relatively the most resistant, irrespective of previous acid adaptation. Resistance to pH 3.5 of E. coli O157:H7 cells habituated in acetic acid washings for 7 days increased in the order 15°C > 10°C > 4°C, while at 25°C cells died off. These results indicate that growth inhibition by storage at low temperatures may be more important than competition by natural flora in inducing acid sensitization of E. coli O157:H7 in fresh meat environments. At ambient temperatures in meat plants, E. coli O157:H7 may grow to restore acid resistance, unless acid interventions are applied to inhibit growth and minimize survival of the pathogen. Acid-habituated E. coli O157:H7 at 10 to 15°C may maintain a higher acid resistance than when acid habituated at 4°C. These responses should be evaluated with fresh meat and may be useful for the optimization of decontamination programs and postdecontamination conditions of meat handling.  相似文献

Ziziphora tenuior L. (Lamiaceae) is an aromatic herb used for its medicinal values against fungi, bacteria. Micropropagation can be used for large-scale multiplication of essential oil producing plants thus avoiding an overexploitation of natural resources. This work aims to develop a reliable protocol for the in vitro propagation of Z. tenuior, and to compare the antioxidant activity between in vitro propagated and wild plants.The explants were sterilized and cultured on MS medium containing different concentrations of growth regulators naphthalene acetic acid (NAA) or indole-3-butyric acid (IBA) with 0.5 mg/L of kinetin (Kin) callus formation was 70.2% after 45 days of incubation in dark on medium supplemented with 1.5 mg/L of NAA. After one month of callus culture on medium supplemented with 2 mg/L BA the shoot number was 5.12 and for the multiplication stage. The shoot number was 4.21 and length was 6.17 cm on medium supplemented with 1 mg/L Kin + 0.1 mg/L NAA.DPPH• reagent was used to test the antioxidant activity. The aqueous and methanol extracts of in vitro plants which were treated with 1.5 and 1 mg/L of kin plus 0.1 mg/L of NAA showed a strong DPPH• scavenging activity where IC50 was 0.307 and 0.369 mg/ml, respectively, while the IC50 of aqueous and methanol extracts of wild plants was 0.516 and 9.229 mg/ml, respectively. Our results suggested that plant growth regulators and in vitro culture conditions increased the antioxidant activity.  相似文献

Elevated CO2 interactions with other factors affects the plant performance. Regarding the differences between cultivars in response to CO2 concentrations, identifying the cultivars that better respond to such conditions would maximize their potential benefits. Increasing the ability of plants to benefit more from elevated CO2 levels alleviates the adverse effects of photoassimilate accumulation on photosynthesis and increases the productivity of plants. Despite its agronomic importance, there is no information about the interactive effects of elevated CO2 concentration and plant growth regulators (PGRs) on potato (Solanum tuberosum L.) plants. Hence, the physiological response and source-sink relationship of potato plants (cvs. Agria and Fontane) to combined application of CO2 levels (400 vs. 800 µmol mol−1) and plant growth regulators (PGR) [6-benzylaminopurine (BAP) + Abscisic acid (ABA)] were evaluated under a controlled environment. The results revealed a variation between the potato cultivars in response to a combination of PGRs and CO2 levels. Cultivars were different in leaf chlorophyll content; Agria had higher chlorophyll a, b, and total chlorophyll content by 23, 43, and 23%, respectively, compared with Fontane. The net photosynthetic rate was doubled at the elevated compared with the ambient CO2. In Agria, the ratio of leaf intercellular to ambient air CO2 concentrations [Ci:Ca] was declined in elevated-CO2-grown plants, which indicated the stomata would become more conservative at higher CO2 levels. On the other hand, the increased Ci:Ca in Fontane showed a stomatal acclimation to higher CO2 concentration. The higher leaf dark respiration of the elevated CO2-grown and BAP + ABA-treated plants was associated with a higher leaf soluble carbohydrates and starch content. Elevated CO2 and BAP + ABA shifted the dry matter partitioning to the belowground more than the above-media organs. The lower leaf soluble carbohydrate content and greater tuber yield in Fontane might indicate a more efficient photoassimilate translocation than Agria. The results highlighted positive synergic effects of the combined BAP + ABA and elevated CO2 on tuber yield and productivity of the potato plants.  相似文献
