  • 1 The genus Castor comprises two species: the Eurasian beaver Castor fiber, and the North American beaver Castor canadensis. Both species suffered from overexploitation, but have seen a revival since the 1920s due to increased protection and reintroduction programmes. Increases in the populations and distributions of species that are able to modify ecosystems have generated much scientific interest. Here we review the available literature concerning the possible ecological impact of beaver species in the Old and New World.
  • 2 Beavers, being ecosystem engineers, are among the few species besides humans that can significantly change the geomorphology, and consequently the hydrological characteristics and biotic properties of the landscape. In so doing, beavers increase heterogeneity, and habitat and species diversity at the landscape scale. Beaver foraging also has a considerable impact on the course of ecological succession, species composition and structure of plant communities, making them a good example of ecologically dominant species (e.g. keystone species).
  • 3 Nevertheless, the strength of beavers’ impact varies from site to site, depending on the geographical location, relief and the impounded habitat type. Consequently, they may not be significant controlling agents of the ecosystem in all parts of their distribution, but have strong interactions only under certain circumstances. We suggest that beavers can create important management opportunities in the Holarctic, and this review will help land managers determine the likely outcome of beaver activity.

蓖麻是一种高蓄能植物和工业原料植物,具有很大的开发利用价值。本文从组织培养和遗传转化两个方面并结合本实验室的工作综述了蓖麻生物工程研究的最新进展。在蓖麻组织培养方面,不同的外植体中以成熟种子的胚轴最适宜,而在不同激素中以TDZ诱导丛生芽的效率最高,并以此为基础建立了蓖麻离体再生体系。在遗传转化方面,不同的转化方法中以农杆菌介导法最适合蓖麻转化。蓖麻胚轴对卡那霉素不敏感,潮霉素是蓖麻转化的适宜筛选剂。文中指出了蓖麻生物工程研究中存在的问题,并对应用生物技术培育蓖麻新品种和促进蓖麻生产的可能性进行了讨论。  相似文献

How arsenic (As) is transported in phloem remains unknown. To help answer this question, we quantified the chemical species of As in phloem and xylem exudates of castor bean (Ricinus communis) exposed to arsenate [As(V)], arsenite [As(III)], monomethylarsonic acid [MMA(V)], or dimethylarsinic acid. In the As(V)- and As(III)-exposed plants, As(V) was the main species in xylem exudate (55%–83%) whereas As(III) predominated in phloem exudate (70%–94%). The ratio of As concentrations in phloem to xylem exudate varied from 0.7 to 3.9. Analyses of phloem exudate using high-resolution inductively coupled plasma-mass spectrometry and accurate mass electrospray mass spectrometry coupled to high-performance liquid chromatography identified high concentrations of reduced and oxidized glutathione and some oxidized phytochelatin, but no As(III)-thiol complexes. It is thought that As(III)-thiol complexes would not be stable in the alkaline conditions of phloem sap. Small concentrations of oxidized glutathione and oxidized phytochelatin were found in xylem exudate, where there was also no evidence of As(III)-thiol complexes. MMA(V) was partially reduced to MMA(III) in roots, but only MMA(V) was found in xylem and phloem exudate. Despite the smallest uptake among the four As species supplied to plants, dimethylarsinic acid was most efficiently transported in both xylem and phloem, and its phloem concentration was 3.2 times that in xylem. Our results show that free inorganic As, mainly As(III), was transported in the phloem of castor bean exposed to either As(V) or As(III), and that methylated As species were more mobile than inorganic As in the phloem.Arsenic (As) is an environmental and food chain contaminant that has attracted much attention in recent years. Soil contamination with As may lead to phytotoxicity and reduced crop yield (Panaullah et al., 2009). Food crops are also an important source of inorganic As, a class-one carcinogen, in human dietary intake, and there is a need to decrease the exposure to this toxin (European Food Safety Authority, 2009). Paddy rice (Oryza sativa) is particularly efficient in As accumulation, which poses a potential risk to the population based on a rice diet (Meharg et al., 2009; Zhao et al., 2010a). Other terrestrial food crops generally do not accumulate as much As as paddy rice; however, where soils are contaminated, relatively high concentrations of As in wheat (Triticum aestivum) grain have been reported (Williams et al., 2007; Zhao et al., 2010b). On the other hand, some fern species in the Pteridaceae family are able to tolerate and hyperaccumulate As in the aboveground part to >1,000 mg kg−1 dry weight (e.g. Ma et al., 2001; Zhao et al., 2002); these plants offer the possibility for remediation of As-contaminated soil or water (Salido et al., 2003; Huang et al., 2004). A better understanding of As uptake and long-distance transport, metabolism, and detoxification is needed for developing strategies for mitigating As contamination, through either decreased As accumulation in food crops or enhanced As accumulation for phytoremediation.The pathways of As uptake by plant roots differ between different As species; arsenate [As(V)] enters plant cells via phosphate transporters, whereas arsenite [As(III)] is taken up via some aquaporins (for review, see Zhao et al., 2009). In rice, a silicic acid efflux protein also mediates As(III) efflux toward stele for xylem loading (Ma et al., 2008). Methylated As species, such as monomethylarsonic acid [MMA(V)] and dimethylarsinic acid [DMA(V)], which may be present in the environment as products of microbial or algal methylation of inorganic As or from past uses of methylated As pesticides, are taken up by rice roots partly through the aquaporin NIP2;1 (for nodulin 26-like intrinsic protein; also named Lsi1; Li et al., 2009). Once inside plant cells, As(V) is reduced to As(III), possibly catalyzed by As(V) reductase(s) such as the plant homologs of the yeast (Saccharomyces cerevisiae) ACR2 (Bleeker et al., 2006; Dhankher et al., 2006; Ellis et al., 2006; Duan et al., 2007). As(III) has a high affinity to thiol (-SH) groups and is detoxified by complexation with thiol-rich phytochelatins (PCs; Pickering et al., 2000; Schmöger et al., 2000; Raab et al., 2005; Bluemlein et al., 2009; Liu et al., 2010). As(III)-PC complexation in roots was found to result in reduced mobility for efflux and for long-distance transport, possibly because the complexes are stored in the vacuoles (Liu et al., 2010). Excess As(III) causes cellular toxicity by binding to the vicinal thiol groups of enzymes, such as the plastidial lipoamide dehydrogenase, which has been shown to be a sensitive target of As toxicity (Chen et al., 2010). The As hyperaccumulating Pteris species differ from nonhyperaccumulating plants because of enhanced As(V) uptake (Wang et al., 2002; Poynton et al., 2004), little As(III)-thiol complexation (Zhao et al., 2003; Raab et al., 2004), and efficient xylem loading of As(III) (Su et al., 2008). Recently, an As(III) efflux transporter, PvACR3, has been found to play an important role in As(III) detoxification by transporting As(III) into vacuoles in Pteris vittata (Indriolo et al., 2010).With the exception of As hyperaccumulators, most plant species have a limited root-to-shoot translocation of As (Zhao et al., 2009). The chemical species of As in xylem exudate have been determined in a number of plant species. As(III) was found to be the predominant species (80%–100%) in the xylem sap of rice, tomato (Solanum lycopersicum), cucumber (Cucumis sativus), and P. vittata even when these plants were fed As(V) (Mihucz et al., 2005; Xu et al., 2007; Ma et al., 2008; Su et al., 2010), suggesting that As(V) is reduced in roots before being loaded into the xylem. In other plant species, such as Brassica juncea (Pickering et al., 2000), wheat, and barley (Hordeum vulgare; Su et al., 2010), As(V) accounted for larger proportions (40%–50%) of the total As in the xylem sap. Studies using HPLC-inductively coupled plasma (ICP)-mass spectrometry (MS) coupled with electrospray (ES)-MS showed no evidence of As(III)-thiol complexation in the xylem sap of sunflower (Helianthus annuus; Raab et al., 2005). When rice plants were exposed to MMA(V) or DMA(V), both As species were found in the xylem sap (Li et al., 2009). Generally, methylated As species are taken up by roots at slower rates than inorganic As, but they are more mobile during the xylem transport from roots to shoots (Marin et al., 1992; Raab et al., 2007; Li et al., 2009).It has been shown that phloem transport contributes substantially to As accumulation in rice grain (Carey et al., 2010). However, little is known about how As is transported in phloem (Zhao et al., 2009). There are no reports on the chemical species of As in phloem exudate. The speciation of As in phloem is important because it dictates how As is loaded in the source tissues and unloaded in the sink tissues, such as grain. Questions with regard to the oxidation state, methylation, and complexation of As in phloem sap remain to be answered. Unlike xylem sap, phloem sap is much more difficult to obtain in sufficient quantities for analysis. In this study, we investigated As speciation in phloem and xylem exudates of castor bean (Ricinus communis), which is widely used as a model plant to investigate phloem transport of solutes (e.g. Hall et al., 1971; Hall and Baker, 1972; Allen and Smith, 1986; Bromilow et al., 1987).  相似文献

Across their range, a large number of biotic and abiotic factors are known to influence the choice of browse plant and the foraging behaviour of the North American beaver (Castor canadensis). We used generalized linear mixed-effects models to investigate sets of variables that may influence the foraging choices of beaver: forage species, distance of forage from water, forage density, and site. Communities across the study sites in central British Columbia, Canada, were dominated by Salix sitchensis, Salix lucida, and Alnus spp. Density had no impact on forage selection, while site, distance from water, and species identity all influenced the foraging decisions of beaver. We postulated that these factors may be ordered hierarchically: large-scale factors, such as site, followed by the medium-scale distance from water, and species of plant at the finest scale. Forage items in some sites had a higher probability of being browsed than in others, while in all sites the probability of being browsed decreased with increasing distance from water. Beaver appeared to be foraging as “picky” generalists; of the 9 plant species examined, 3 species of Salix (S. scouleriana, S. drummondiana and S. sitchensis) were selected by beaver, Salix bebbiana was avoided, and 5 species were neither selected for nor against. Browse selection within the genus Salix implied that beaver were able to differentiate among closely related species. Detailed information on forage selection is a crucial first step in designing and interpreting models that predict large-scale distributional patterns of beaver.  相似文献

The acid lipase of castor endosperm lipid bodies has been studied using colorimetric assay based on the measure of the hydrolytic activity of p-nitrophenyl ester of palmitate and other acyl derivatives. These substrates are compatible with the natural triacylglycerols for the measure of lipolytic activities. The subcellularly-surveyed acid lipolytic activity in the germinated castor bean endospermal tissue was found to be enhanced in the lipid bodies. The lipase, which is partially latent and tightly associated with lipid bodies, is an exceptionally stable enzyme with an optimum activity at pH 4.5 and displays an inverse relationship between its activity and the acyl chain length of its substrate. To facilitate isolation of the acid lipase, a procedure has been developed to solubilise the membrane-bound enzyme in an active form. The detergent-solubilised acid lipase after two chromatographic steps yielded an eight-fold active preparation which after gel permeation resolved as heterogeneous aggregate in excess of 500 kD. Lipase-enriched preparations showed consistent presence of 14 and 60 kD proteins which constituted the most abundant species of the lipid bodies. Although it has not been possible to obtain an active lipase preparation in a state free of either the 14 or 60 kD protein, the lipase activity in the detergent extracts of lipid bodies was immunoprecipitable with antibodies raised against the 60 kD component.  相似文献

Nucleotide variation in an approximately 490 bp fragment of the mitochondrial DNA control region (mtDNA CR) was used to describe the genetic variation and phylogeographical pattern in the Eurasian beaver (Castor fiber) over its entire range. The sampling effort was focused on the relict populations that survived a drastic population bottleneck, caused by overhunting, at the end of the 19th century. A total of 152 individuals grouped into eight populations representing all currently recognized subspecies were studied. Sixteen haplotypes were detected, none of them shared among populations. Intrapopulation sequence variation was very low, most likely a result of the severe bottleneck. Extreme genetic structure could result from human-mediated extinction of intermediate populations, but it could also be an effect of prior substantial structuring of the beaver populations with watersheds of major Eurasian rivers acting as barriers to gene flow. Phylogenetic analysis revealed the presence of two mtDNA lineages: eastern (Poland, Lithuania, Russia and Mongolia) and western (Germany, Norway and France), the former comprising more divergent haplotypes. The low level of sequence divergence of the entire cytochrome b gene among six individuals representing six subspecies suggests differentiation during the last glacial period and existence of multiple glacial refugia. At least two evolutionary significant units (ESU) can be identified, the western and the eastern haplogroup. The individual relict populations should be regarded as management units, the eastern subspecies possibly also as ESUs. Guidelines for future translocations and reintroductions are proposed.  相似文献

Hen and Montagu's Harriers breed in the same cultivated areas of eastern France. We present data from an extensive study conducted in three adjacent areas where 757 nests of the two harriers were monitored between 1993 and 2000, with the aim of comparing the breeding ecology of these two species and to evaluate their possible future trends. Breeding habitat for harriers consisted nearly exclusively of winter cereals, causing great conservation concern in this intensively farmed region. The Hen Harrier was almost absent from two of the study areas. This species showed significantly larger clutch size and higher breeding success than the Montagu's Harrier, and an earlier breeding phenology. It was thus less adversely affected by harvesting activities than Montagu's Harrier. Both species showed a reduced breeding success with increasing laying date. There was a large diet overlap between the two species, possibly leading to competitive interactions. Overall, the Montagu's Harrier should be considered as the more vulnerable of the two species, necessitating conservation measures, such as protection of nests from early harvesting activities. Nevertheless, to maintain both species in agricultural habitats, farming practices that preserve sufficient food should also be promoted.  相似文献

  • 1 The Eurasian beaver Castor fiber suffered a drastic reduction in both geographical range and population size, due to human persecution, until the end of the 19th century. After the adoption of protection measures, natural expansion and reintroductions led to the recovery of this species over much of its European range.
  • 2 We review historical events that led to the recovery of beavers in France, and summarize the status of beavers in various river systems. Beaver establishment in France is a story of overall success: several major river systems are presently occupied, so that the species is no longer at risk in France.
  • 3 However, beaver recolonization took place in parallel with increasing human impacts on the environment. In addition to natural limiting factors, anthropogenic factors impeded beaver settlement in many areas. Today, beavers often occupy suboptimal habitats and, as a consequence, come into conflict with human activities. Effective solutions for preventing beaver damage include the restoration of riparian habitats to discourage crop damage and the provision of physical barriers to protect crops.
  • 4 Beaver populations reintroduced into France all originate from the relict Rhône population. However, in recent years, beavers from populations in neighbouring countries have been expanding into north‐eastern France. Therefore, our review of beaver origin and distribution in these countries may contribute to the development of appropriate national management strategies and towards important decisions, e.g. the decision to try to keep Rhône beavers genetically isolated, or to allow populations to mix.
  • 5 The recently discovered presence of North American beavers Castor canadensis in three countries surrounding France has raised an important issue. This species may out‐compete C. fiber in places where the species come into contact. A programme based on field‐trapping sessions and genetic analyses has recently been initiated in some western countries in order to eradicate this non‐native species.

Microbial mats present in two shallow atolls of French Polynesia were characterized by high amounts of exopolysaccharides associated with cyanobacteria as the predominating species. Cyanobacteria were found in the first centimeters of the gelatinous mats, whereas deeper layers showing the occurrence of the sulfate reducers Desulfovibrio and Desulfobacter species as determined by the presence of specific biomarkers. Exopolysaccharides were extracted from these mats and partially characterized. All fractions contained both neutral sugars and uronic acids with a predominance of the former. The large diversity in monosaccharides can be interpreted as the result of exopolymer biosynthesis by either different or unidentified cyanobacterial species. Sites of restriction endonucleases were mapped on pOAD2, a plasmid harbored in Flavobacterium sp. KI72. The plasmid codes 6-aminohexanoic acid cyclic dimer hydrolase and 6-aminohexanoic acid linear oligomer hydrolase. pOAD2 (molecular weight: 28.8 megadaltons [Mdal]) had 6 HindIII and 5 EcoRI sites, which were located at 0, 8.4, 8.9, 11.1, 19.0 and 25.0 Mdal (for HindIII) and 3.3, 5.4, 20.4, 20.8, 22.6 Mdal (for EcoRI). A mutant which could not grow on 6-aminohexanoic acid cyclic dimer but grew on the linear dimer as the sole carbon and nitrogen source harbored a deletion plasmid pOAD21 derived from pOAD2. By comparing the restriction sites of these two plasmids, the deleted region was localized on which the 6-aminohexanoic acid cyclic dimer hydrolase was coded.  相似文献

Genome sequence analysis of Ricinus communis has indicated the presence of at least 22 putative terpene synthase (TPS) genes, 13 of which appear to encode sesquiterpene synthases (SeTPSs); however, no SeTPS genes have been isolated from this plant to date. cDNAs were recovered for six SeTPS candidates, and these were subjected to characterization in vivo and in vitro. The RcSeTPS candidates were expressed in either Escherichia coli or Saccharomyces cerevisiae strains with engineered sesquiterpene biosynthetic pathways, but only two (RcSeTPS1 and RcSeTPS7) produced detectable levels of product. In order to check whether the engineered microbial hosts were adequately engineered for sesquiterpene production, a selection of SeTPS genes was chosen from other plant species and demonstrated consistently high sesquiterpene titers. Activity could be demonstrated in vitro for two of the RcSeTPS candidates (RcSeTPS5 and RcSeTPS10) that were not observed to be functional in our microbial hosts. RcSeTPS1 produced two products, (−)-α-copaene and (+)-δ-cadinene, while RcSeTPS7 produced a single product, (E, E)-α-farnesene. Both RcSeTPS5 and RcSeTPS10 produced multiple sesquiterpenes.  相似文献

Modification of tryptophan residues in castor bean hemagglutinin (CBH) with N-bromosuccinimide (NBS) was investigated in detail. Tryptophan residues accessible to NBS increased with lowering pH and six tryptophan residues/mol were oxidized at pH 3.0, while two tryptophan residues/mol were oxidized at pH 5.0. From the pH-dependence curve for tryptophan oxidation, we suggest that the extent of modification of tryptophan in CBH is influenced by an ionizable group with pKa = 3.6. The saccharide-binding activity was decreased greatly by modification of tryptophan concomitantly with a loss of fluorescence. A loss of the saccharide-binding activity was found to be principally due to the modification of two tryptophan residues/mol located on the surface of the protein molecule. In the presence of raffinose, two tryptophan residues/mol remained unmodified with retention of fairly high saccharide-binding activity. The results suggest that one tryptophan residue is involved in each saccharide-binding site on each B-chain of CBH.  相似文献

We estimate habitat loss and fragmentation in a hoverfly, Aneriophora aureorufa, used as a representative forest specialist species. This species is a pollinator specialist of two native trees, forming a triad endemic to the South American Temperate Rainforest (SATR). We combine species distribution models with species-specific requirements to estimate the habitat range of A. aureorufa over two non-overlapping time periods (before human settlement to 2000, and from 2000 to 2014). We analyzed the predicted distribution range of A. aureorufa in Chile, quantifying habitat loss in both periods and fragmentation in the latter. In addition, we evaluated the representativeness of the Chilean protected areas system in relation to the current habitat of the species. We found that the total habitat of A. aureorufa decreased by 68.3% compared to historic pre-settlement levels; in the period 2000–2014 the loss was 4.9%. The northern zone was the most affected by habitat loss and fragmentation, with an estimated total loss of 89.9% from the historic period to 2014, with the loss of 238.2 km2 per year between 2000 and 2014. Eighteen percent of the habitat of A. aureorufa occurs within protected areas. We found an overrepresentation in the southern zone (24.79%) and an underrepresentation in the northern zone (3.44%). We propose that forest specialist species of the northern zone of the SATR could be threatened due to the high pressure of habitat loss and the underrepresentation of the Chilean protected areas systems.  相似文献

High-quality data on species distribution and knowledge of species ecology are necessary pre-requisites for their effective conservation. In the European Union, a network of protected areas called the “Natura 2000 network” was set up to conserve the most valuable and threatened species. The Natura 2000 network was supposed to improve rare species conservation, but has it? In this paper, an analysis of data collected from 1 × 1 km grids on the distribution of orchids in East Macedonia (NE Greece) in two consecutive periods of time (before and after 2010), revealed the degree to which a larger data set (including both periods instead of only one) improved our knowledge of the distributions of species and whether the Natura 2000 network can be used as a basis for the conservation of the orchid flora in this area. When the data for the first period and cumulative data for both periods were compared, orchid species richness and density were very similar. However, comparison of the two datasets showed that more orchid taxa were recorded per grid in the first than the second period. However, based on the surveys in the second period, the knowledge on orchid distribution increased significantly. In particular, ten orchid taxa were recorded for the first time in East Macedonia during the second period and significantly more (54.79 % of the total number of species) were recorded. This resulted in a better understanding of their altitudinal requirements. It also confirmed that most of the orchid-rich grid cells in East Macedonia overlap with the Natura 2000 network, which highlights importance of Natura 2000 for orchid conservation.  相似文献

We developed nine new nuclear dinucleotide microsatellite loci for Castor canadensis. All loci were polymorphic, except for one. The number of alleles ranged from two to four and from five to 12 in populations from Arizona and Wisconsin, respectively. Average heterozygosity ranged from 0.13 to 0.86 per locus. Since cross-species amplification in Castor fiber was successful only in four loci, we tested also nine recently published C. canadensis loci in the Eurasian species. Eight of the published loci amplified; however, three were monomorphic. The number of alleles was lower in C. fiber than in C. canadensis at all loci tested.  相似文献

Cooperative oxidation of succinate and exogenous NADH was followed in the mitochondria from five- to six-day-old castor bean (Ricinus communisL.) seedlings. Although succinate was oxidized at a much higher rate than NADH, the former inconsiderably (less than 15%) inhibited the oxidation of the latter substrate in state 4, while, in state 3 (in the presence of ATP), the two substrates did not compete and were jointly oxidized. When two substrates were oxidized by the mitochondria with the alternative CN-resistant oxidase (AO) inhibited with salicylhydroxamic acid, the rate of NADH oxidation in state 4 dropped by over 40% as compared to the initial rate. Meanwhile, the rate of succinate oxidation was not considerably affected by AO inhibition. We believe that one of the AO functions in the mitochondria is to provide for noncompeting oxidation of two (or more) substrates by employing two (or several) dehydrogenases of the respiratory chain.  相似文献

Oviposition behavior in two co-occurring species of predatory ladybirds of aphids were investigated. The two species of beetles often shared the same aphid colonies in bean plants in space and time and showed a similar oviposition pattern in response to aphid abundance. Overlapping oviposition presents potential risks of interspecific predation of eggs in unstable aphid colonies. Eggs in clusters by two ladybird species, Menochilus sexmaculatus and Coccinella transversalis, were defended from reciprocal predation and the defense was found to be chemical. Eggs occurring singly were found to be highly vulnerable to predation. Strength of chemical defense significantly increased from single eggs to eggs in mixed clusters and eggs in pure clusters. Results are not only consistent with the hypothesis that eggs in clusters are better protected from natural enemies but also extend the ambit of the hypothesis that coexisting ladybird predators have evolved to avoid the risks of heterospecific predation of eggs in a patch. Corynebacterial isolates, obtained by selective and non-selective media from conjunctival swabs, were speciated by using two recently developed systems. All 25 strains from 92 individuals, as well as 12 strains from a previous similar study, belonged to lipophilic species. The significance of this unusual finding is discussed.
