Adipocyte fatty acid binding protein (aP2) is a key mediator of intracellular transport and metabolism of fatty acids. Its expression during adipocyte differentiation is regulated through the actions of peroxisome proliferator-activated receptor gamma (PPARgamma) and CCAAT/enhancer binding protein alpha (C/EBPalpha). Macrophages also express aP2, and the lack of macrophage aP2 significantly reduces atherosclerotic lesion size in hypercholesterolemic mice. We investigated the regulation of expression of macrophage aP2 and CD36, a fatty acid membrane binding protein and scavenger receptor, in response to the adipogenic agents isobutylmethylxanthine (IBMX), insulin, and dexamethasone, a combination of agents shown to induce fibroblast-to-adipocyte differentiation. Treatment of J774 macrophages with adipogenic agents significantly induced aP2 mRNA expression, while CD36 expression was inhibited. Dexamethasone was essential and sufficient to induce aP2 expression, and insulin had a synergistic effect. However, IBMX antagonized induced-aP2 expression. aP2 protein expression and [14C]oleic acid uptake by macrophages were also increased by dexamethasone. Unlike what occurs in adipocytes, adipogenic agents had mixed effects on the expression of PPARgamma and C/EBPalpha in macrophages. Our data demonstrate differences in the regulation of aP2 in adipocytes and macrophages and show that macrophage aP2 expression by adipogenic agents is independent of the PPARgamma and/or C/EBPalpha signaling pathway.  相似文献

To understand the relationship between intramuscular adipogenesis in the pig and the supply fatty acids, we established a clonal porcine intramuscular preadipocyte (PIP) line from the marbling muscle tissue of female Duroc pig. Confluent PIP cells exhibited a fibroblastic appearance. Their adipogenic ability was investigated using confluent PIP cells after exchanging growth medium for adipogenic medium containing 50 ng/mL insulin, 0.25 microM dexamethasone, 2 mM octanoate, and 200 microM oleate. Appropriate concentrations of octanoate and oleate for the induction of adipogenesis were determined from the ability of cells to accumulate lipid and the toxicity of fatty acids. When cells were cultured in differentiation medium for 8 days, large numbers of lipid droplets were observed in differentiated PIP cells, and their cytosolic TG content increased in a time-dependent manner. While oleate only induced the expression of PPARgamma mRNA, but not that of C/EBPalpha, octanoate significantly induced the expression of both PPARgamma and C/EBPalpha mRNA. Octanoate and oleate accelerated the inducing effect of insulin and dexamethasone on the expression of aP2 mRNA. These results indicate that a combination of octanoate and oleate synergistically induced PIP adipogenesis, and that the stimulation of octanoate was essential to the trigger for the adipogenesis in PIP cells.  相似文献

为研究Wnt/b-catenin信号通路对猪脂肪组织发育的影响, 并探讨其可能的作用机制, 以1~120日龄长白猪脂肪组织为试验材料, 采用SQ RT-PCR法检测Wnt信号通路中相关基因β-catenin、GSK3β、Fz1及主要的脂肪转录因子PPARγ、C/EBPα 和分化早期标志基因LPL mRNA的表达变化; 石蜡切片免疫组化方法定性检测β-catenin在脂肪组织发育中的时序表达变化。SQ RT-PCR结果显示, β-catenin mRNA在猪出生第1天有高水平表达, 随着个体日龄的增长, 其表达量逐渐降低, 60日龄后维持在一个较低水平。GSK3β和Fz1 mRNA的表达量也伴随脂肪组织的发育而逐渐降低。而LPL、PPARγ和C/EBPα的 mRNA表达量随着个体日龄的增长而逐渐升高, 60日龄后仍保持高水平的表达。石蜡切片免疫组化结果显示, 随着脂肪组织的发育, β-catenin蛋白的表达也随之降低, 并且β-catenin蛋白的表达部位逐渐由细胞核和细胞质共表达变为只在细胞质中表达。上述基因表达的时序变化规律提示, β-catenin在维持前体脂肪细胞的未分化状态, 抑制脂肪组织发育中具有重要作用, 其作用机制可能是通过对脂肪细胞转录因子PPARγ、C/EBPα及分化早期标志基因LPL mRNA的调控来进行的。  相似文献

In this study, we show that expression of FoxC2 blocks the capacity of 3T3-L1 preadipocytes to undergo adipogenesis in the presence of dexamethasone, isobutylmethylxanthine, and insulin. This block is characterized by an extensive decrease in the expression of proteins associated with the function of the mature fat cell, most notably C/EBPalpha, adiponectin, perilipin, and the adipose-specific fatty acid-binding protein, FABP4/aP2. Since the expression of these proteins lies downstream of PPARgamma, we overexpressed PPARgamma in Swiss mouse fibroblasts to promote adipocyte differentiation. We show that FoxC2 blocks the ability of PPARgamma to induce adipogenic gene expression in response to exposure of the cells to dexamethasone, isobutylmethylxanthine, insulin, and a PPARgamma ligand. Interestingly, the expression of aP2 escapes the inhibitory action of FoxC2 under conditions that promote maximum PPARgamma activity. In contrast, FoxC2 inhibits the expression of C/EBPalpha, perilipin, and adiponectin even in the presence of potent PPARgamma ligands. Finally, we show that FoxC2 does not affect the ability of PPARgamma to bind to or transactivate from a PPARgamma response element. These data suggest that FoxC2 blocks adipogenesis by inhibiting the capacity of PPARgamma to promote the expression of a subset of adipogenic genes.  相似文献

The aim of present study is to evaluate the effects of Garcinia cambogia on the mRNA levels of the various genes involved in adipogenesis, as well as on body weight gain, visceral fat accumulation, and other biochemical markers of obesity in obesity-prone C57BL/6J mice. Consumption of the Garcinia cambogia extract effectively lowered the body weight gain, visceral fat accumulation, blood and hepatic lipid concentrations, and plasma insulin and leptin levels in a high-fat diet (HFD)-induced obesity mouse model. The Garcinia cambogia extract reversed the HFD-induced changes in the expression pattern of such epididymal adipose tissue genes as adipocyte protein aP2 (aP2), sterol regulatory element-binding factor 1c (SREBP1c), peroxisome proliferator-activated receptor gamma2 (PPARgamma2), and CCAT/enhancer-binding protein alpha (C/EBPalpha). These findings suggest that the Garcinia cambogia extract ameliorated HFD-induced obesity, probably by modulating multiple genes associated with adipogenesis, such as aP2, SREBP1c, PPARgamma2, and C/EBPalpha in the visceral fat tissue of mice.  相似文献

Dexamethasone and transforming growth factor-beta (TGF-beta) show contrary effects on differentiation of adipocytes. Dexamethasone stimulates adipocyte differentiation whereas TGF-beta inhibits it. In the present study, we investigated whether dexamethasone could reverse the TGF-beta-mediated inhibition of preadipocyte differentiation. Primary rat preadipocytes, obtained from Sprague-Dawley rats, were pretreated with dexamethasone in the presence or absence of TGF-beta, prior to the induction of differentiation. Co-treatment of dexamethasone and TGF-beta before inducing differentiation reversed the TGF-beta-mediated inhibition of preadipocyte differentiation. In order to elucidate the mechanism by which dexamethasone reversed the effect of TGF-beta on the inhibition of preadipocyte differentiation, the expression of CCAAT/enhancer binding protein-alpha (C/EBPalpha) and peroxisome proliferator-activated receptor gamma (PPARgamma) was examined. Dexamethasone increased C/EBPalpha and PPARgamma expression in the absence of TGF-beta and also recovered the TGF-beta-mediated suppression of C/EBPalpha expression in preadipocytes. Its effect was sustained in differentiated adipocytes as well. However, those effects were not observed in 3T3-L1 preadipocytes or differentiated adipocytes. These results indicate that dexamethasone reverses the TGF-beta-mediated suppression of adipocyte differentiation by regulating the expression of C/EBPalpha and PPARgamma, which is dependent on the cellular context.  相似文献

The family of mammalian Sirtuin proteins comprises seven members homologous to yeast Sir2. Here we show that SIRT2, a cytoplasmic sirtuin, is the most abundant sirtuin in adipocytes. Sirt2 expression is downregulated during preadipocyte differentiation in 3T3-L1 cells. Overexpression of SIRT2 inhibits differentiation, whereas reducing SIRT2 expression promotes adipogenesis. Both effects are accompanied by corresponding changes in the expression of PPARgamma, C/EBPalpha, and genes marking terminal adipocyte differentiation, including Glut4, aP2, and fatty acid synthase. The mechanism underlying the effects of reduced SIRT2 in 3T3-L1 adipocytes includes increased acetylation of FOXO1, with direct interaction between SIRT2 and FOXO1. This interaction enhances insulin-stimulated phosphorylation of FOXO1, which in turn regulates FOXO1 nuclear and cytosolic localization. Thus, Sirt2 acts as an important regulator of adipocyte differentiation through modulation of FOXO1 acetylation/phosphorylation and activity and may play a role in controlling adipose tissue mass and function.  相似文献
