The contribution of attached and free-floating bacteria to the bacterial numbers and heterotrophic uptake in 44 diverse aquatic environments was studied. A factor analysis reduced the variability of the raw data base to three major factors explaining 53.6% of total variance. These factors were (i) salinity, (ii) heterotrophic uptake, and (iii) particulate load. A cluster analysis categorized the 44 habitats into five distinct environmental types based on these three factors. There was no significant pattern in the distribution of attached versus free-floating bacteria when assessed by epifluorescent microscopy. However the contribution of attached bacteria to the uptake of an amino acids mix was reduced in marine waters. Heavy particulate loads resulted in an increased percentage uptake of amino acids and glucose from the attached bacteria. Uptake response was found to be substrate specific especially in oliogotrophic freshwater. Amino acid uptake was more associated with the attached fraction, whereas glucose uptake was mediated more by the free-floating fraction.  相似文献

Concentrations and glucose mineralization potentials ( V max) of attached and free bacteria in waters from the Humber estuary were determined. The waters had high concentrations of suspended solids. Most bacteria were attached to suspended solids and V max of attached bacteria was greater than that of free bacteria. These observations suggest that breakdown of organic matter is carried out mainly by attached bacteria. Examination of samples from other estuaries which contained less suspended solids than the Humber suggested that in these estuaries free bacteria might be more abundant than attached bacteria.  相似文献

为更好地防治鲍鱼病害的发生和流行,对分离自广东汕尾健生鲍鱼养殖场九孔鲍养殖环境及其肠道中不同细菌菌群的耐药性进行了研究。结果表明,四环素、青霉素G、卡那霉素、丁胺卡那霉素和新生霉素均对绝大多数异养菌和弧菌菌株都不敏感或无作用;相反,氟哌酸、红霉素、氯霉素以及环丙沙星等则均对它们比较敏感;复方新诺明、链霉素和多粘霉素B对弧菌菌株均有作用,而且多粘霉素B也对水体中的异养菌群相当有效。  相似文献

1. After observing that juvenile roach fed intensively on cyanobacteria and that cyanobacteria were densely colonized by heterotrophic bacteria, we tested whether the bacteria are used by underyearling roach and the extent to which they contribute to the energy requirements of the fish.
2. We radiolabelled attached bacteria in a natural cyanobacterial suspension, fed the fish with these particles, and estimated their assimilation by roach. Biomass of attached bacteria on cyanobacteria increased with the proportion of the cyanobacterium Microcystis in total cyanobacteria. Biomass-specific thymidine incorporation of attached bacteria was higher than that of free bacteria.
3. In feeding experiments, we detected assimilation of bacterial biomass into muscle tissue of underyearling roach. Fish consumed Microcystis to a lesser extent compared with Aphanizomenon but assimilation of attached bacteria was higher when roach fed on Microcystis because of the higher biomass of epibacteria on this cyanobacterium. However, biomass of attached bacteria was too low to be an important food source for underyearling roach.
4. We conclude that assimilation of epibacteria from cyanobacteria cannot explain the success of roach in eutrophic lakes.  相似文献

The relationship between the physiological group of gelatinolytic bacteria and the abundance of heterotrophic bacteria in freshwater ecosystems was described, based on analysis of 1082 different freshwater samples collected in Croatia. Percentages of gelatinolytic bacteria among the population of heterotrophic bacteria showed a significant negative correlation with the abundance of heterotrophic bacteria. The relation between the physiological group (gelatinolytic bacteria) and heterotrophic bacteria can be considered to be an indicator of the pollution degree of freshwaters. A high relative content of gelatinolytic bacteria (> 76%) always indicates the colony-forming units of heterotrophic bacteria < 1000/mL, which corresponds to the high water quality; gelatinolytic bacteria < 11% indicate polluted waters. Isolated strains of aerobically grown gelatinolytic bacteria were Gram-negative rod-shaped or Gram-positive endospore-forming rod-shaped cells.  相似文献

In order to understand the large‐scale distribution of microbial populations simultaneously and their relationship with environmental parameters, flow cytometry was used to analyse samples collected from 46 stations in the coastal waters of Qingdao in spring, 2007. The distribution of virus was significantly and positively correlated with heterotrophic bacteria. Two groups of picophytoplankton (Synechococcus and picoeukaryotes) were detected; however, Prochlorococcus was not found. Picoeukaryotes and nanophytoplankton were abundant in the near‐shore waters, whereas Synechococcus was abundant in the off‐shore areas. No variation was found in vertical distribution of virus, heterotrophic bacteria, Synechococcus and nanophytoplankton abundances, except picoeukaryotes abundance in the bottom layer was dramatically lower than that in the upper layers. Correlation analyses indicated that the relationship between abiotic variables and heterotrophic bacteria, pico‐ and nanophytoplankton was closer than that between abiotic variables and virioplankton. Temperature and nutrients were the synchronous factors controlling the growth of heterotrophic bacteria, pico‐ and nanophytoplankton in the coastal waters of Qingdao in spring. The results suggested that synergistic and antagonistic effects existed among microbial groups.  相似文献

Abstract The distribution of heterotrophic bacteria in polluted coastal and unpolluted pelagic seawaters was studied using a 14C-MPN method with either five of seven kinds of 14C-organic compounds as substrates. The total number of heterotrophic bacteria in pelagic waters ranged from 9.2 × 103 to 5.4. ¢ 104 cell/ml and more than 85% of the heterotrophic bacteria were represented by obligate oligotrophs. In coastal waters, the number of heterotrophs was one order of magnitude higher (av. 3.5 ¢ 105 cells/ml), and eutrophic and facultatively oligotrophic bacteria were predominant. Oligotrophs in pelagic waters had a high specificity for the utilization of amino acids, especially glycine, and acetate-utilizing bacteria were scarce. The in situ maximum uptake rates of glutamate and glycine were much higher than those of glycolate and acetate. Acetate uptake rates were extremely low or not detectable in pelagic waters. The specificity of uptake kinetics is assumed to depend on the existence of obligate oligotrophs as dominant bacteria in pelagic seawater.  相似文献

We examined the abundance and heterotrophic uptake of bacteria attached to particulate matter suspended in five coastal ponds and two marshes near Woods Hole, Mass. Although the number of particle-bound bacteria was low (<10%), these bacteria incorporated a large proportion (>40%) of [14C]glucose and [14C]glutamate in selected aquatic systems. The uptake per cell was significantly higher for epibacteria than for unattached bacteria in all systems. Two groups of the aquatic environments sampled were statistically different in the contribution made by particle-bound bacteria to total bacterial abundance and to total assimilation of [14C]glucose and [14C]glutamate. Particle-bound bacteria were more important in those waters with a high particle concentration and not flushed regularly by tides than in waters with a low particle concentration and flushed regularly.  相似文献

The abundance and heterotrophic activity of attached and free-living bacteria were examined seasonally in coastal water. Heterotrophic activity was determined by the uptake of [14C]glucose. The density of attached bacteria was always minor, not showing a seasonal variation, whereas the free-living bacteria were more numerous and showed a marked seasonal variation, their density being higher under warmer conditions. The contribution of the attached bacteria to the total assimilation of [14C]glucose (from 10 to 38%) was lower than that of the free-living bacteria, neither of them showing a seasonal variation. On a cellular basis, attached bacteria were more active, since they assimilated more [14C]glucose and showed, under warmer conditions, a higher cellular volume (0.102 versus 0.047 μm3). We consider that the factors responsible for these observations were the amount and quality of the particulate material, the different availability of organic matter for the two types of bacteria, and in a fundamental way, the variation in water temperature.  相似文献

Urease, a nickel-dependent metalloenzyme, is synthesized by plants, some bacteria, and fungi. It catalyzes the hydrolysis of urea into ammonia and carbon dioxide. Although the amino acid sequences of plant and bacterial ureases are closely related, some biological activities differ significantly. Plant ureases but not bacterial ureases possess insecticidal properties independent of its ureolytic activity. To date, the structural information is available only for bacterial ureases although the jack bean urease (Canavalia ensiformis; JBU), the best-studied plant urease, was the first enzyme to be crystallized in 1926. To better understand the biological properties of plant ureases including the mechanism of insecticidal activity, we initiated the structural studies on some of them. Here, we report the crystal structure of JBU, the first plant urease structure, at 2.05 Å resolution. The active-site architecture of JBU is similar to that of bacterial ureases containing a bi-nickel center. JBU has a bound phosphate and covalently modified residue (Cys592) by β-mercaptoethanol at its active site, and the concomitant binding of multiple inhibitors (phosphate and β-mercaptoethanol) is not observed so far in bacterial ureases. By correlating the structural information of JBU with the available biophysical and biochemical data on insecticidal properties of plant ureases, we hypothesize that the amphipathic β-hairpin located in the entomotoxic peptide region of plant ureases might form a membrane insertion β-barrel as found in β-pore-forming toxins.  相似文献

A 6-m-deep lake has been sampled to measure the temporal and depth-wise distribution of heterotrophic bacteria and biological activity in the water. Surface, mid-depth and bottom waters were analysed at monthly intervals for a period of one year. The coefficient of heterotrophic activity, alkaline phosphatase activity and biological oxygen demand are used as an index of biological activity. The bacterial community was at maximum during spring, coinciding with high values of biological activity. Highest biological activity was observed in the bottom waters. Dissolved organic carbon showed a significant positive correlation with most of the biological activity parameters. This suggests that biological activity, as measured by the coefficient of heterotrophic activity, was more closely related to the concentration of substrates than to population density of heterotrophic bacteria.  相似文献

Cryptoperidiniopsoids are an unclassified group of delicately thecate heterotrophic dinoflagellates known to be common in eastern U.S. estuarine waters. Over the past 10 years cryptoperidiniopsoids were isolated from different geographical regions and cultured with cryptophyte algal prey. In the seven clonal isolates examined, reproduction was strongly linked to the availability of prey cells. The dinoflagellates phagocytized the contents of prey cells through a tube‐like peduncle, similarly as close relatives of Pfiesteria spp. and several other heterotrophic species. Cell division occurred while encysted, most commonly yielding two biflagellated offspring. Abundant fusing gametes, phagotrophic planozygotes, and cysts with a pronounced nuclear cyclosis characterized persistent sexuality. Cysts with nuclear cyclosis produced two flagellated offspring cells. The resistance of reproductive cysts to antimicrobial treatments was examined, and a simple high‐yield technique was developed for population synchronization while ridding the dinoflagellates of most contaminating vacuolar prey DNA and external contaminants. The DNA content and population DNA profiles of synchronously excysted cryptoperidiniopsoids from different isolates were measured using flow cytometry and were related to the life history of these and other dinoflagellates. Cryptophyte‐fed cultures with versus without extracellular bacteria were compared, and bacteria apparently promoted cryptoperidiniopsoid feeding and growth. Externally bacteria‐free dinoflagellates were cultured in media enriched with dissolved organic nutrients, and nutritional benefit may have occurred in some treatments. The potential for mixotrophic nutrition from maintenance of cryptophyte chloroplasts was examined using flow cytometrically sorted cells, but evidence of kleptoplastidy was not found in these isolates under the conditions imposed.  相似文献

Nitrate addition to oil field waters stops the biogenic formation of sulfide because the activities of nitrate-reducing bacteria (NRB) suppress the activities of sulfate-reducing bacteria (SRB). In general, there are two types of NRB — the heterotrophic NRB and the chemolithotrophic NRB. Within the latter group are the nitrate-reducing, sulfide-oxidizing bacteria (NR-SOB). To date, no study has specifically addressed the roles of these different NRB in controlling sulfide concentrations in oil field produced waters. This study used different culture media to selectively enumerate heterotrophic NRB and NR-SOB by most probable number (MPN) methods. Produced waters from three sulfide-containing western Canadian oil fields were amended with nitrate as an electron acceptor, but no exogenous electron donor was added to the serum bottle microcosms. Changes in the chemical and microbiological characteristics of the produced waters were monitored during incubation at 21°C. In less than 4 days, the sulfide was removed from the waters from two of the oil fields (designated P and C), whereas nearly 27 days were required for sulfide removal from the water from the third oil field (designated N). Nitrate addition stimulated large increases in the number of the heterotrophic NRB and NR-SOB in the waters from oil fields P and C, but only the NR-SOB were stimulated in the water from oil field N. These data suggest that stimulation of the heterotrophic NRB is required for rapid removal of sulfide from oil field-produced waters. The actual extracellular release of organic matter by algae was determined in water samples in which the heterotrophic activity of bacteria was inhibited by gentamycin. Gentamycin rapidly and efficiently inhibited the activity of aquatic bacteria without affecting phytoplankton metabolism. Aquatic bacteria utilized the products of algal extracellular release. The amount of algal photosynthetic products metabolized by bacteria can be taken as a measure of their heterotrophic activity in waters.  相似文献

The major objective of these studies was to assess the extent of the flow of released dissolved organic carbon (RDOC) from phytoplankton to heterotrophic bacteria. The second aim was to test Nalewajko and Schindler's (1976) hypothesis that the percentage of extracellular release (PER) of RDOC by phytoplankton is higher in oligotrophic than eutrophic waters. The studies on the relationship between the bacterial assimilation of RDOC and productivity of phytoplankton in the lakes of different trophy have shown a direct correlation. It was observed that higher utilization rate of phytoplankton RDOC occurred in lakes with higher primary production. The inverse relationship between productivity of lakes, i.e. trophy of waters, and amount of RDOC or PER is closely dependent on the heterotrophic activity of bacteria in waters.  相似文献

It is now possible to obtain accurate total counts of the bacteria of natural waters with the use of acridine orange staining and epifluorescence microscopy. This approach can be coupled to highly sensitive measurements of heterotrophic activity using radioisotopes. To accomplish this, three variations of a “specific activity index” are suggested, based on different approaches to measuring heterotrophic activity with radiolabeled organic solutes. The denominator of each index is the direct count of bacteria from a given natural sample. Three numerators are presented, each of which has been shown to vary directly with heterotrophic bacterial activity: Vmax, turnover rate, and direct uptake (at high substrate concentrations). Each approach is illustrated with data from estuarine and coastal waters of northeastern Massachusetts. The data show major differences in specific activity that accompany such habitat differences as distances within or offshore from an estuary and vertical location in the water column. These and other data suggest that specific activity is a valid indicator of the physiological state and metabolic role of the bacteria. Some evidence is presented in support of the hypothesis that the natural bacteria are adapted to conditions of nutrient starvation by becoming “dormant,” existing for an unknown period of time in a reversible physiological state that reflects the availability of organic nutrients.  相似文献

Free bacterial populations were separated from an intact planktonic community in water of a eutrophic reservoir in Japan by filtration through Whatman GF/ C glass fiber filters (mean porosity 1.2 µm). Urea decomposition by the free bacterial populations and the intact planktonic community was determined in six different months.The separated free bacteria apparently did not take part in urea-decomposition in waters of the reservoir through the year: the number of free heterotrophic bacteria increased during the urea-decomposition experiments, however, the concentration of urea did not decrease. Whereas, in five cases out of six, urea was decomposed by the intact planktonic community. Probably, phytoplankton were responsible for most of the urea-decomposition. On the assumption that the decomposition of urea obeyed first-order kinetics, rate constants were calculated to be 0.00–0.63 day–1 with a mean value of 0.21 day–1.  相似文献

The heterotrophic bacterial community of oceanic aggregates which mediates particle solubilization, displays features (low carbon demand and low turnover) that are difficult to reconcile with the observed high enzyme activities and cell numbers. Hypotheses are proposed to explain this discrepancy. Furthermore, production of both free and attached bacteria may have been underestimated by neglecting the ultramicrobacteria (UMB). Production of UMB may represent up to 28% of the total bacterial production.  相似文献

Microbial ureases hydrolyze urea to ammonia and carbon dioxide. Urease activity of an infectious microorganism can contribute to the development of urinary stones, pyelonephritis, gastric ulceration, and other diseases. In contrast to these harmful effects, urease activity of ruminal and gastrointestinal microorganisms can benefit both the microbe and host by recycling (thereby conserving) urea nitrogen. Microbial ureases also play an important role in utilization of environmental nitrogenous compounds and urea-based fertilizers. Urease is a high-molecular-weight, multimeric, nickel-containing enzyme. Its cytoplasmic location requires that urea enter the cell for utilization, and in some species energy-dependent urea uptake systems have been detected. Eucaryotic microorganisms possess a homopolymeric urease, analogous to the well-studied plant enzyme composed of six identical subunits. Gram-positive bacteria may also possess homopolymeric ureases, but the evidence for this is not conclusive. In contrast, ureases from gram-negative bacteria studied thus far clearly possess three distinct subunits with Mrs of 65,000 to 73,000 (alpha), 10,000 to 12,000 (beta), and 8,000 to 10,000 (gamma). Tightly bound nickel is present in all ureases and appears to participate in catalysis. Urease genes have been cloned from several species, and nickel-containing recombinant ureases have been characterized. Three structural genes are transcribed on a single messenger ribonucleic acid and translated in the order gamma, beta, and then alpha. In addition to these genes, several other peptides are encoded in the urease operon of some species. The roles for these other genes are not firmly established, but may involve regulation, urea transport, nickel transport, or nickel processing.  相似文献

The hot-spring waters of numerous hot springs at the Kamchatka Peninsula, Russia differ in their chemical characteristics and elemental composition. Total bacterial abundance (TBA) as well as enzymatically active and respiring bacteria was enumerated in water samples collected from the Nalychevskie, Oksinskie, Apapelskie, and Dachnye hot springs. 5-Carboxyfluorescein diacetate activity was detected in all water samples and comprised 29-65% of the TBA as determined by 4',6-diamidino-2-phenylindol staining. The respiratory activity of iron-oxidizing bacteria was assayed by 5-cyano-2,3-ditolyltetrazolium chloride reduction. Respiring cells accounted for 9-14% of the TBA, indicating a positive correlation with the number of iron-oxidizing bacteria from the hot-spring biomats. Enumeration of heterotrophic bacteria revealed a high-density bacterial population only in the water of the Apapelskie hot spring, which has a temperature of 36 degrees C. Therefore, it appears that heterotrophic and iron-oxidizing bacteria inhabiting the hot-spring waters are essential for the geochemical processes occurring in hydrothermal systems.  相似文献
