The seeds of Sophora alopecuroides L, contained 12.8 per cent of endosperm, from which the water-soluble polysaccharide gum was isolated and purified by fractionation with alcohol three times. Hydrolysis of the polysacchatides with sulphuric acid produced galactose and mannose in the molar ratio of 1:2.37, as determined by the phenolsulphuric acid method and by paper chromatographic method. The total sugar content reached 67-70%. The polysaccharides, after complete hydrolysis, reduction with NaBH4 and acetylation, ridded acetates. An analysis on them by gas-chromatography showed that they contained galactose and mannose in the molar ratio of 1:1.83, and the total sugar content reached 56%. In order to identify the linkages of the polysaccharides isolated from the seeds of Sophora alopecuroides, they were methylated by using the methods of Hokomori, Conval and Albersheim. The fully methylated polysaccharides were hydrolyzed and then reduced with NaBH4, followed by acetylation. Finally they yielded acetates. An analysis on IR data, the results of acid hydrolysis, periodate oxidation and Smith type degradation, as well as the acetates by GC, GC-MS and 1HNMR showed that the linkages of Sophora alopecuroides. gum may be, The purified polysaccharides seem to be homogenous after ultracentrifugation analysis. The sedimentation coefficient of the gum obtained from repeated purification with alcohol was 5.31, its intrinsic viscosity was 6.25 dl/g and its molecular weight was estimated to be 635000. By the light-scattering method, however, its molecular weight was estimated to be 715000. The crude polysacchafides contained 44 per cent of the water-soluble polysaccharides.  相似文献

In this study, three chemically sulfated polysaccharides (SPAPs) were derived from one water-soluble polysaccharide (PAP) of Polyporus albicans mycelia by chlorosulfonic acid-pyridine method. The effects of polysaccharides on the immune function were examined after the mice were intragastrical administrated with polysaccharides at three doses of 100, 200, and 300 mg/kg body weight for 7 days. The results showed that both the lymphocytes proliferation and macrophage function were significantly enhanced by SPAP in all groups along with the increase of the substitution degree and dose (P<0.01). It indicated that SPAP could be a potential immunostimulants used in the food and pharmaceutical industry.  相似文献

The water-soluble polysaccharides of brown algae attract the increasing attention of researchers as an important class of polymeric materials of biotechnological interest. The sole source for production of these polysaccharides has been large brown seaweeds such as members of Laminariales and Fucales. A new source of water-soluble polysaccharides is suggested here: it is a filamentous brown alga Streblonema sp., which can be cultivated under controlled conditions in photobioreactors that allow obtaining algal biomass with reproducible content and quality of polysaccharides. The accumulation of water-soluble polysaccharides can be stimulated by macronutrient limitation. In response to nitrogen deficiency, Streblonema sp. accumulated water-soluble polysaccharides (WSPs) rich in laminaran. WSP accumulation started after 3–4 days following nitrate depletion and reached a plateau at around day 7. Polysaccharide accumulation was related to cellular nitrogen content. The critical internal N level that triggered the onset of polysaccharide accumulation was 2.3% dry weight (DW); at a cellular N concentration less than 1.4% DW, the polysaccharide synthesis stopped. Upon nitrate re-supply, mobilization of WSP occurred after 3 days. These results suggest that a two-stage cultivation process could be used to obtain large algal biomass with high water-soluble polysaccharide production: a first cultivation stage using nitrate-supplemented medium to accumulate algal biomass followed by a second cultivation stage in a nitrate-free medium for 3 to 7 days to enhance polysaccharide content in the alga.  相似文献

采用超声提取法,对新疆紫草固体培养毛状根、液体培养毛状根以及2~3年野生根中的总糖和多糖进行了提取测定.结果表明:毛状根中水溶性总糖含量为25.573%,是野生根总糖含量的3.4倍;固体培养的毛状根多糖含量最高,达4.289%,是液体培养毛状根的3.37倍,是野生根的4倍.稳定性试验和重复性试验表明,该研究结果可靠性强,重复性好且稳定.研究发现,新疆紫草毛状根中水溶性总糖和多糖的含量高于野生根.  相似文献

Three water-soluble polysaccharides (CTPS-1A, CTPS-2B, and CTPS-3A) were obtained from roots of Cudrania tricuspidata (Carr.) Bur. in this study. The homogeneity of polysaccharides was determined, and the average molecular weight, ultraviolet, infrared, monosaccharide composition, and methylation analyses were carried out. Immunomodulatory activity assays in vitro showed that the three polysaccharides could directly stimulate the proliferation of mouse splenocytes alone or combining with concanavalin A or lipopolysaccharide. Furthermore, their stimulating activities were higher than that of the widely clinically used lentinan at optimal concentrations. CTPS-1A and CTPS-2B also enhanced the pinocytic activity of mouse peritoneal macrophages.  相似文献

Three polysaccharides (water-soluble (WSP), alkali-soluble (ASP) and insoluble (IMP)) from boat-fruited sterculia seeds were obtained using different extraction methods. Moisture, ash, protein and total carbohydrate content of WSP, ASP and IMP were analyzed. WSP was rich in glucose, rhamnose, arabinose and galactose while small amount of xylose was also detected. The monosaccharide composition as well its relative content for WSP and ASP were similar. The intrinsic viscosity results demonstrated that ASP had much lower intrinsic viscosity than WSP, indicating partial polysaccharides were degraded into low molecular weight polymers during alkaline extraction. The acute anti-inflammatory bioactive results of polysaccharides indicated that WSP demonstrated an inhibitive effect toward acute inflammation.  相似文献

The fucose-containing polysaccharides of the brown alga Dictyota dichotoma were extracted with either trichloroacetic acid or HCl to give both water-soluble and water-insoluble materials. The latter had a high proportion (16 to 11%) of protein, and although all the sugars found in the water-soluble extracts were present, the major sugar in these water-insoluble polysaccharides was glucose. The water-soluble material extracted with HCl was a protein-free sulphated heteropolysaccharide. Complete removal of a glucan from the water-soluble extract was achieved by fractional precipitation with ethanol. The recovered glucan-free sulphated polysaccharide, which was rich in glucuronic acid, galactose, fucose and sulphate, showed high anticoagulating activity.  相似文献

Phellinus linteus is a well-known oriental medicinal fungus that has various biological activities such as stimulation of humoral and cell mediated immunity, anti-mutagenicity activity, and anti-cancer activity. The process of isolating and purifying a water-soluble glycan from P. linteus was achieved by hot water extraction, filtration, solvent precipitation, dialysis, and freeze-drying. Acidic fractions of the polysaccharide were separated from crude polysaccharides by DEAE-cellulose anion exchange chromatography at 0.4 M NaCl. The molecular weight of the proteo-heteroglycan after Sepharose CL-4B gel filtration chromatography was about 150,000. The acidic proteo-heteroglycan consisted of 72.2% polysaccharide and 22.3% protein. The sugar of the proteo-heteroglycan was composed of mannose, galactose, glucose, arabinose, and xylose. The amino acid pattern showed that the fractions contained large amounts of aspartic acid, glutamic acid, alanine, glycine, and serine. The fractions for both alpha-glycan at 860 cm(-1) and beta-glycan at 910 cm(-1) had the characteristics of IR spectrum absorption as compared to those for beta-glucan derived from Lentinus edodes. A 13C and 1H NMR spectroscopy showed that the acidic proteo-heteroglycan was a noble biomolecule mixed both alpha- and beta-linkages, and a (1,6) branched type (1,3) glycan.  相似文献

Crude, pharmaceutically useful water-soluble polysaccharides have been isolated from durian rinds (Durio zibethinus) by hot water extraction followed by ethanol precipitation. The polysaccharides were fractionated by anion exchange chromatography and size exclusion chromatography. Characterisation of the sub-fractions by methanolysis, methylation analysis and NMR spectroscopy revealed that the principal components were pectic polysaccharides with starch as a contaminant. Physical features namely molecular weight (Mw) and intrinsic viscosity of the main fractions were investigated by size exclusion chromatography coupled to multi angle laser light scattering (SEC/MALLS) and capillary viscometer, respectively. The main fractions were subjected to the complement-fixation assay and the relationship of the chemical features of the polysaccharide fractions with their activity was also considered.  相似文献

A method for the routine radiochemical determination of carboxylic acid groups in polysaccharides is described. The procedure is based on sodium borotritide reduction of the product of the reaction between water-soluble carbodiimide derivatives and carboxyl groups on the polysaccharide. A reliable and efficient method for isolating Annona squamosa L. genomic DNA, free from polyphenols and polysaccharides has been developed. Different methods involving use of CTAB and SDS with or without modifications were used. A CTAB based extraction method which uses diatomite to remove polyphenols and polysaccharides proved to be the best. This method allowed recovery of good quality DNA in sufficient quantity suitable for complete digestion by restriction endonucleases and amplifiable in polymerase chain reaction as compared to other methods.  相似文献

The binding of rare earth elements (REEs) with water-soluble polysaccharides of nondeproteinization and deproteinization in the leaves of the fernDicranopteris dichotoma was studied by molecular activation analysis (MAA). Two cold-water-soluble polysaccharides (extracted by 75% ethanol and 90% ethanol, respectively) and one hot-water-soluble polysaccharide (extracted by 90% ethanol) were separated using biochemical separation techniques. The eight rare earth elements (La, Ce, Nd, Sm, Eu, Tb, Yb, and Lu) in these polysaccharides were determined by instrumental neutron activation analysis. Our new results showed that the REEs were bound firmly with the water-soluble polysaccharides in the plant, regardless of whether nondeproteinization or deproteinization was used. The molecular-weight (MW) measurement demonstrated that REEs were mainly bound with low-MW (10,000–20,000) polysaccharides.  相似文献

多型炭角菌在4%葡萄糖,1%蛋白胨,250mL三角瓶装培养液80mL,pH6.5,25℃的培养条件下生长最好,发酵终点为168小时。正交试验得到K+, Mg2+,Ca2+, Zn2+, Fe2+, Mn2+和Cu2+等金属离子的最适浓度分别为0.15%, 0.03%, 0.05%, 0.0015%, 0.001%, 0.00005%和 0.0002%。对培养得到的菌丝体进行多糖提取,得到2种粗多糖:X-1(水溶性多糖)和X-2(碱溶性多糖);从发酵液提取的粗多糖为X-3。经Sephadex G-100和 Sephadex G-150凝胶柱层析,收集、合并的单一峰位部分经聚丙烯酰胺凝胶电泳及纸层析均呈现为单一斑点。多糖酸水解后均能与茚三酮显色,蛋白质组成分析表明其分子中富含16种氨基酸。  相似文献

It was established that the water-soluble polysaccharides of bamboo shoot other than starch consisted of a water-soluble xylan, an arabinogalactan and an α-glucan by isolation and fractionation of the water-soluble polysaccharides extracted with dimethyl sulfoxide from a preparative meal of bamboo shoot. Summary The polymers contained within pellets rich in either dictyosomes or the endoplasmic reticulum have been investigated. These were detected by the incorporation of radioactivity from D-[U-14C]glucose into the sugars in the intact root. Three types of material were obtained 1) water-soluble 2) soluble in chloroform 3) insoluble. The relative proportions of each of these 3 fractions were different in the 2 membrane preparations. The dictyosome pellet contained a high proportion of water-soluble material, some of the water-soluble polysaccharides contained in both membrane preparations had a molecular weight greater than 40,000. These polymers resembled the pectins and xylans deposited in the cell wall. Some of the insoluble material could be rendered soluble by incubation with proteolytic enzymes and all of this from the dictyosome pellet had a molecular weight greater than 4,000 whereas most of that obtained from the endoplasmic reticulum had a molecular weight less than 4,000 and it did not contain fucose.Abbreviations RER rough endoplasmic reticulum - GA golgi apparatus  相似文献

Pectins were extracted either by water from extruded lemon fibre or by hot acid from the raw lemon fibre. The amount of water-soluble polysaccharides from lemon plant cell walls was greatly increased after extrusion-cooking. The pectins obtained by extraction with water from the extruded fibre and the pectins extracted from the raw material by hot acid were studied. The water-soluble pectins obtained after extrusion-cooking had the distinctive feature of being very highly (92%) methylated; they were also particularly rich in arabinose side-chains. High molecular weight material coming from the “hairy” regions was isolated after digestion by an endo-polygalacturonase. Methylation analysis revealed the presence in both pectins of fairly branched (1 → 5)-linked arabinans and arabinogalactans of type I and II side-chains.  相似文献

The changes in the molecular weight distribution of water-soluble hemicelluloses and xyloglucan during hypocotyl growth of intact seedlings of Pinus pinaster Aiton were investigated. The mass-average molecular weight of total polysaccharides of the hemicellulose fraction soluble in 4% KOH dramatically increased during hypocotyl growth while xyloglucan slightly decreased. These phenomena were due to an increase in the degree of polymerization of an arabinogalactan and a slight depolymer-ization in the xyloglucan present in this fraction. In the hemicellulose fraction soluble in 24% KOH, xyloglucan increased its degree of polymerization from day 7 to 10 after which it decreased slightly. The xyloglucan of the hemicellulose fraction soluble in 4% KOH may thus be involved in cell wall loosening which makes cell wall expansion possible during hypocotyl growth.  相似文献

Carbohydrate Accumulation in Siberian Spruce Needles of Various Ages   总被引:1,自引:0,他引:1  
The content of mono- and oligosaccharides, hemicelluloses, and water-soluble polysaccharides was studied in Siberian spruce (Picea obovata Ledeb.) needles of various ages. Accumulation of carbohydrates during the growth period and the ratio of their individual fractions were determined by the proportions between the growth rate and assimilate export in developing needles. Throughout the growth period, the content of monosaccharides in needles of all ages was much higher than that of oligosaccharides. Unlike the period of active growth, the dormancy period was characterized by a wider diversity of oligosaccharides and their rather high content. Hemicelluloses that accumulated during the previous growth season and water-soluble polysaccharides of 2- and 3-year-old needles were involved into metabolism at the time of active growth of current-year needles. Analysis of carbohydrate accumulation in 1- to 9-year-old needles showed that the monosaccharide concentration was higher in needles of the first two years. The content of hemicelluloses A and B and water-soluble polysaccharides was higher in older needles. During the growth period, the assimilates of 2- and 3-year-old needles were partly used for the formation of young needles. The increase in relative content of polysaccharides during needle senescence reflects the involvement of photosynthetic products into the secondary pool of carbon metabolites.  相似文献

The polysaccharides from the fruits of Lycium barbarum have received considerable attention in previous publication, but the polysaccharides from the leaves were rarely reported. In the present work, four water-soluble polysaccharide fractions: LBP-I, LBP-II, LBP-III and LBP-IV isolated from L. barbarum leaves were purified through DEAE-Sephadex A-25. LBP-II and LBP-IV respectively showed one symmetrical peak on HPGPC with average molecular weight of 9.39×10(4)Da and 4.18×10(5)Da. UV and IR analysis of the two fractions showed the characteristics of acidic polysaccharides combined with polypeptides or proteins. GC analysis showed LBP-IV was mainly composed of rhamnose, arabinose, xylose, glucose and galactose with molar ratio of 1.61:3.82:3.44:7.54:1.00, and the uronic acid content was 47.68% (w/w) determined by sulfuric acid-carbazole method. (1)H and (13)C NMR spectra of LBP-IV also showed the presence of carboxyl carbon and five anomeric carbons, and suggested there may be both α- and β-anomeric configurations in this fraction. Moreover, splenocyte proliferation activity assay showed that LBP-IV significantly enhanced the proliferation of splenocyte stimulated by ConA or LPS, indicating the fraction has the beneficial effect on immunostimulating activity.  相似文献
