The cotyledons and embryo axes of seeds of Phaseolus vulgaris L. cv. Pinto contained 16% of the total calcium in the seed. The remaining 84% was in the testas. There was no evidence that calcium in testas was used in seedling growth or that calcium was leached from seedlings during growth.  相似文献

The efficiency of calcium in enhancing the tolerance of beans(Phaseolus vulgaris L.) to sodium salinization was studied inpot experiments in both cool and hot seasons. In the cool seasonincreased yields of dry matter, roots, nodules and pods werepositively correlated with increased Ca application and negativelycorrelated with the Na contents of irrigation water and planttissue. The Na levels both in roots and tops declined significantlyas increased amounts of calcium were added. Ca in the rangeof 2.0 to 8.0 mmol/1 caused competitive inhibition of Na uptakeand Na translocation. At Ca levels between 0 to 2.0 mM onlyNa translocation to tops was markedly inhibited. In warm seasonsCa had no beneficial effects on bean yields at any level ofNa. On the contrary, high rates of Ca application resulted ina higher death rate.  相似文献

During the early stages of germination and vegetative development,cotyledons of intact bean (Phaseolus vulgaris L.) seedlingsshowed active ABA catabolism causing a low endogenous ABA content.At the end of the substrate mobilizing phase, when the cotyledonsbecame senescent, a drastic increase of the endogenous ABA contentlinked with a decrease of the ABA catabolic activity was observed.This indicates that a close connection exists between senescenceand endogenous ABA content and metabolism in bean cotyledons. Removal of the apical growth region induced in the cotyledonsactivation of the ABA catabolism and the endogenous ABA concentrationdecreased below the detection limit (0.1 ng/g fr wt) at theonset of the outgrowth of the axillary buds. From then, apicaldominance was restored and the cotyledons returned to the senescentstate, which was correlated with a drastic increase of theirendogenous ABA content. 1 Bevoegd verklaard navorser N. In the last few years, much attention has been given to the role of proteins that accumulate during water deficit. In this work, we analyzed the electrophoretic patterns of basic protein extracts, enriched for a number of cell-wall proteins, from bean (Phaseolus vulgaris L.) seedlings and 21-d-old plants subjected to water deficit. Three major basic proteins accumulated in bean seedlings exposed to low water potentials, with apparent molecular masses of 36, 33, and 22 kD, which we refer to as p36, p33, and p22, respectively. Leaves and roots of 21-d-old plants grown under low-water-availability conditions accumulated only p36 and p33 proteins. In 21-d-old plants subjected to a fast rate of water loss, both p33 and p36 accumulated to approximately the same levels, whereas if the plants were subjected to a gradual loss of water, p33 accumulated to higher levels. Both p36 and p33 were glycosylated and were found in the cell-wall fraction. In contrast, p22 was not glycosylated and was found in the soluble fraction. The accumulation of these proteins was also induced by abscisic acid (0.1-1.0 mM) treatment but not by wounding or by jasmonate treatment.  相似文献

Two bean (Phaseolus vulgaris L.) cultivars differing in growth responses to zinc were examined for differences in uptake and subcellular localization of 65Zn during a 15-day growth period. The zinc-sensitive cultivar Sanilac showed initially a much higher rate of absorption, which declined after 24 hours. The zinc-tolerant cultivar Saginaw showed a slow but steady rate of absorption for 10 days. In roots as well as in stem callus tissues of both cultivars, three-fourths of the absorbed 65Zn was localized in the “cytoplasmic” supernatant fractions (containing ribosomes and vacuolar sap). Very little (less than 7%) 65Zn was localized in the cell wall fraction. There was a much greater proportion of the absorbed 65Zn localized in root mitochondria and nuclei of the zinc-sensitive Sanilac than in the zinc-tolerant Saginaw. Stem callus tissues, however, did not show such cultivar differences in zinc accumulation at the sub-cellular level.  相似文献

Alteration in KNO3 and KCl levels of culture solutions resultedin the production of a series of Phaseolus vulgaris seedlingscontaining a range of chloride levels in the primary leaves;such treatments did not affect leaf potassium content. Irrespectiveof the leaf chloride content the stomata responded to increasedlight intensity by a decrease in stomatal resistance. The stomataof seedlings grown in culture solutions containing KCI offeredhigher diffusive resistances than those of seedlings not receivingthis treatment. This increase in stomatal resistance was foundover the whole range of elevated leaf chloride contents produced.Presence of chloride in the leaf tissue did not, however, affectstomatal responses to fluctuations in atmospheric CO2 concentration.  相似文献

Abstract

Callus production and plant regeneration from different explants of Phaseolus vulgaris L. cv. Giza are reported. Calli cultures were induced from leaf, hypocotyl, embryo and root explants. Rapid growth of callus was achieved by leaf explants cultured on MS salts, B5 vitamins and supplemented with 2,4— dichlorophenoxyacetic acid (2, 4—D)+0.5 mg/l kinetin (kin). Addition of casein hydrolysate at 2 g/l to maintenance medium enhanced callus growth and hindered the early appearance of necrotic parts. This report also provides a detailed method for production of multiple shoots directly from the wounded edges of immature cotyledon explant via organogenesis on 1 mg/l benzyladenine (BA) or indirectly on 0.5 mg/l naphthaleneacetic acid (NAA)+2 mg/l BA. The regeneration of bean plants through the two ways described here (direct or indirect) may be of use in genetic improvement of bean.  相似文献

Young plants of Phaseolus vulgaris were grown in nutrient solutions at different levels of calcium concentration. When the calcium concentration was low more palisade parenchyma and less extended bundle sheath was formed at the adaxial side of minor veins of the leaves as compared to leaves of plants grown with higher calcium supply. The number of calcium oxalate crystals in the bundle sheath extensions was positively correlated to the amount of calcium fed to the plants. The ion induces additional cell divisions in the bundle sheath extensions. A high supply of calcium leads to the formation of a second type of crystal in the bundle sheath.  相似文献

Patterns of 14C-photosynthate translocation in bean (Phaseolusvulgaris L.) seedlings have been investigated in relation tovascular-bundle continuity between exporting and importing organsby use of radioassay and tissue-clearing techniques. Assimilatefrom the primary leaves reaches the first trifoliate leaf byan indirect route. There is no direct vascular connection betweenthe primary leaves and distal tissues. Bundles of the primaryleaf petiole connect with an anastomosis at the node, and allbundles which originate from this structure descend the stem.Assimilate from primary leaves moves down the stem, and is thentransferred to an ascending pathway, the bundles of which traversethe anastomosis at the second node. The lateral leaflets ofthe first trifoliate leaf are served differentially by primaryleaves with respect to assimilate supply. Differences are relatedto position, and may be accounted for by differences in vascularcontinuity.  相似文献

Although determinations of the ABA content of lateral buds ofPhaseolus vulgaris revealed no difference between decapitatedand intact control plants in the first 12 h following decapitation,a relative decrease in the ABA content of lateral buds of decapitatedplants was detectable 24 h following decapitation. Shoot decapitationwas also observed to result in a decrease in the ABA contentof stem tissue. The application of IAA to the stem of decapitatedplants prevented these changes and increased the ABA contentof stem tissue relative to that of intact plants. The levelsof IAA and ABA were also determined in the stem tissue fromthe nodes of intact bean plants. The possible interdependenceof these two plant hormones was further investigated by a studyof [2–14ClABA metabolism. The major cytokinins in stems of decapitated, disbudded bean plants have been identified by enzymic degradation, Sephadex LH20 and reversed phase high performance liquid chromatography, and by combined gas chromatography-mass spectrometry as 6-(4-hydroxy-3-methylbut-trans-2-enylamino)-9--D-ribofuranosylpurine (zeatin riboside), 6-(4-hydroxy-3-methylbutylamino)-9--D-ribofuranosylpurine (dihydrozeatin riboside), and the 5-phosphates of these compounds (zeatin ribotide and dihydrozeatin ribotide). Minor cytokinins in this tissue were tentatively identified as dihydrozeatin-O--D-glucoside and zeatin ribotide-O--D-glucoside. [8-14C-]Dihydrozeatin appeared to be rapidly metabolized to dihydrozeatin ribotide when supplied to segments of stems from decapitated plants. Leaf area expansion, dry weight, and water relations of Phaseolus vulgaris L. and P. acutifolius Gray were compared during a drying cycle in the greenhouse to understand the characteristics which contribute to the superior drought tolerance of P. acutifolius. Stomates of P. acutifolius closed at a much higher water potential than those of P. vulgaris, delaying dehydration of leaf tissue. P. acutifolius had a more deeply penetrating root system, which also contributes to its drought tolerance. Root-shoot ratios did not differ between the two species either under well watered or water stressed conditions. Leaf osmotic potential was also similar in the two species, with no apparent osmotic adjustment during water stress. These results indicate that P. acutifolius postpones dehydration and suggest that sensitive stomates and a deeply penetrating root system are characteristics which, if incorporated into cultivated beans, might increase their drought tolerance.  相似文献

Phytochrome was partially purified from etiolated seedlings of Avena sativa L. Several properties of the red-absorbing (PR) and far-red absorbing (PFR) forms of the pigment were compared. The 2 forms could not be shown to differ with respect to their sedimentation velocity in sucrose density gradients, elution volume from Sephadex G-200 columns, binding properties on calcium phosphate, or electrophoretic mobility. PFR, however, was more labile than PR during precipitation with 50% ammonium sulfate. Sephadex G-200 elution diagrams obtained with fresh phytochrome preparations revealed 2 components of different molecular weights, 1 roughly 180,000, and 1 roughly 80,000. Native phytochrome had an absorption spectrum in vivo showing an absorption maximum for PR of 667 nm. Both the large and small forms of phytochrome mentioned above can be maintained with an absorption maximum for PR of 667 nm. However, allowing them to remain for several hours as PFR, even at 4°, shifted this peak to 660 nm. The protein conformational change during phytochrome transformation may be quite small, though the various comparative techniques used do not strictly rule out a fairly large one. The need for maintaining the pigment as PR during all steps of purification, but particularly during ammonium sulfate precipitation is underscored.  相似文献

Plastids prepared from dark-grown spruce seedlings showed a negligible activity of photosystem II, and no fluorescence variation was observed during actinic illumination. The photosystem II reaction centre, however, was present in primary thylakoids. Exposure of such seedlings to continuous light induced the development of photosystem II activity via three stages (rapid, lagged and gradual), and the variable fluorescence appeared. The rapid development of photosystem II may be attributed to the activation of the oxygen-evolving system, possibly the manganese-catalyzing site, and the lagged and gradual developments may be closely related to the formation of thylakoid membranes and their assembly to grana.  相似文献

Primary leaves of one-week-old seedlings of dwarf beans effectivelyinhibit stem elongation either in natural daylight or in continuousdarkness. Stem elongation is promoted by exogenous gibberellicacid and kinetin and inhibited by indol-3-ylacetic acid (IAA)or abscisic acid, and it is suggested that IAA is the inhibitorysubstance emanating from the primary leaf blades which affectsthe growth of the stem.  相似文献

The ability of aseptically cultured Macrozamia communis seedlingsto form apogeotropic root nodules in light and darkness wasexamined. In darkness, apogeotropic root nodules developed atthe junction of the primary-root and the cotyledonary node.More acropetally formed lateral-roots were not nodular and weretypical ageotropic or plagiotropic secondary-roots. When thesecultures were exposed to light, all of the roots became typical,apogeotropic cycad root nodules. Light stimulated dichotomousbranching of nodules previously formed in the dark. Nodulesexposed to light were more papillose than those maintained indarkness. Light also inhibited primary and secondary-root elongation. The distribution of ureide-N was studied throughout vegetativeand reproductive growth of non-nodulated Phaseolus vulgarisL. (bushbean) grown in nitrate nutrient solution. Largest increasesin ureide-N per plant were correlated with flowering and earlypod formation and with seed filling. Highest amounts of ureidesper organ were measured in stems and axillary trifoliates. Highestconcentrations (µmol ureide-N g–1 fr. wt.) weremeasured in young developing organs and stems. Seeds did notaccumulate ureides until the ureide content of pods had reacheda maximum. Results obtained using the inhibitor of xanthine oxidase, allopurinol,are consistent with the origin of ureides via purine degradationbut alternative pathways cannot be discounted. Leaves and stems were shown to have the ability to degrade allantoatevia an enzymic process.  相似文献
