While 2-aminoanthracene and 2-aminofluorene are converted to frameshift mutagens by microsomal preparations from rat livers, the microsomes from the intestinal mucosa of the same animals, under the experimental conditions used herein, either have little such activity or lack it altogether. Cell-free extracts of the colon anaerobe Bacteroidesfragilis may exhibit such activity to varying degrees depending upon the conditions of incubation. However mixtures consisting of cell-free extracts from B. fragilis and microsomes from intestinal mucosa demonstrate significant- more than additive- activity in converting these chemicals to mutagens.  相似文献

Treatment of whole cell suspensions of Eubacterium aerofaciens and Bacteroides fragilis with lysozyme resulted in a marked increase (>100-fold) in the rates of biotransformation of cholate to 7-ketodeoxycholate (7-KD) in the former but only a 2-fold increase in the latter bacterium. In B. fragilis the total activity of both NAD-dependent 7-α-hydroxysteroid dehydrogenase (7-α-OHSDH) and bile salt hydrolase (BSH) increase markedly during the stationary growth phase. Both enzymes were found in the spheroplast lysate and the Triton-soluble washed membrane fractions but only BSH was found in the spheroplast medium.  相似文献

Bacteroides fragilis. an obligate anaerobe, synthesizes an azide-inhibitable iron-containing superoxide dismutase when grown in complex medium. Cells grown anaerobically in complex media containing dcsferrioxamine (DesferalTM, Ciba-Geigy) and graded concentrations of Mn synthesize the azide-resistant manganese-containing SOD. The fraction of MnSOD activity in dialyzed cell extracts increased prograsively as the Mn concentration in the medium increased. The fraction of MnSOD activity also increased in extracts of cells grown in the medium with I mM Mn but with graded concentrations of desferrioxamine (0–10 micromolar). The SOD activity in the cells grown under the various conditions varied but not in a causal relationship with either Mn or desferrioxamine concentration. Electrophoresis reveaicd that the SOD activity in cells grown in the absence or presence of I mM Mn migrated with the same relative mobility and exhibited identical activity patterns when examined separately or as a mixture. These data are consistent with substitution of Mn for Fe in the B. fragilis apoprotein under anaerobic conditions and support the model of a single protein binding either Fe or Mn.  相似文献

A DNA-dependent ATPase (molecular weight 68000) has been purified from extracts of B. subtilis. The enzyme shows specificity for single-stranded DNA and for hydrolysis of ATP (Km 0.4 mM). Similarities with the rep gene product from E.coli are discussed.  相似文献

Superoxide dismutase and catalase were not detected in M. pneumoniae and several other species of Mycoplasma some of which consume oxygen and secrete H2O2. M. pneumoniae in suspension formed O2? in the presence of NADH and flavins and extracts of M. pneumoniae formed O2? in the presence of either NADH or NADPH. The lack of superoxide dismutase in M. pneumoniae could not be attributed to superoxide dismutase in the complex medium in which the organisms were grown because organisms grown in medium in which the superoxide dismutase had been inactivated by heat still contained undetectable amounts. Mycoplasmas appear to be an exception to the rule that organisms which consume O2 synthesize superoxide dismutase.  相似文献

In skeletal muscle of animals with the phosphorylase b kinase deficiency gene there is < 1% of the normal activity to convert phosphorylase b to a in the presence of Ca++, Mg++, and ATP (1). Correspondingly, there is < 1% of the normal activity to phosphorylate phosphorylase b. Nevertheless, under the same conditions, these extracts catalyze the phosphorylation of troponin at a rate 57% of normal. Phosphorylase b converting activity can be sedimented from skeletal muscle of control mice by centrifugation. This fraction isolated from I strain skeletal muscle extracts phosphorylates troponin at a rate 29–39% of the control. EGTA1 (15 mM) inhibits troponin phosphorylation by 50–60% in this fraction from both strains. The EGTA inhibition is reversed by 15 mM Ca++. Thus the phosphorylase b kinase in skeletal muscle of animals with the phosphorylase b kinase deficiency gene can phosphorylate troponin B, although it shows little or no activity with phosphorylase as a substrate. This observation is consistent with the normal muscle contractility of I strain animals.  相似文献

An endonuclease activity has been identified in extracts of Bacillus subtilis. This activity is stimulated by Mn++ or Ca++ ions but not by Mg++ ions. The enzyme catalyzes the breakdown of native DNA of high molecular weight to fragments of molecular weights ranging from 3 × 106 to 20 × 106. A variety of DNA's from sources such as B. subtilis, Salmonella and T7 phage are attacked. About 61% of the activity of the cells is released into the medium during protoplast formation under conditions where 98% of the glucose 6-P dehydrogenase activity is retained by the cells.  相似文献

Neurosporacrassa was surveyed for cyclic AMP-dependent protein kinase activity. Two peaks (I and II) of protein kinase activity were demonstrated by DEAE-cellulose chromatography of wild type Neurospora extracts. Peak I was stimulated by cyclic AMP, eluted below 60 mM NaCl and had high activity using histone H2B as substrate. Peak II eluted at 200–250 mM NaCl; its activity was not cyclic AMP stimulated and was highest with dephosphorylated casein as a substrate. Cyclic AMP binding to a protein associated with the protein kinase is specifically inhibited by certain cyclic AMP analogs.  相似文献

Two media supporting the growth of several established lepidopteran cell lines in monolayer and suspension culture are described. The medium designated BML-TC10 was developed specifically as an inexpensive medium for production of cells of Spodoptera frugiperda and the homologous nuclear polyhedrosis virus (NPV) of this species. Simultaneously, a second medium was formulated in which the amino acid requirements were provided by enzymatic protein hydrolysates, one of which was termed BML-TC7A. Several cell lines could be adapted easily to this medium. BML-TC10 supported growth of S. frugiperda cells and production of the NPV's of S. frugiperda and Autographa californica. BML-TC7A supported the growth of cells of S. frugiperda. Carpocapsa pomonella, Heliothis zea, and Trichoplusia ni. Cells of the latter produced the polyhedra of T. ni and A. californica NPV's in this medium.  相似文献

Na+-dependent transport of methyl-β-D-thiogalactopyranoside (TMG) mediated by the melibiose transport system was investigated in Escherichia coli mutants lacking the lactose transport system. When an inwardly-directed electro-chemical potential difference of Na+ was imposed across the membrane of energy depleted cells, transient uptake of TMG was observed. Addition of TMG to cell suspensions under anaerobic conditions caused a transient acidification of the medium. This acidification was observed only in the presence of Na+ or Li+. These results support the idea that TMG is taken up by a mechanism of Na+-TMG co-transport via the melibiose transport system in Escherichia coli.  相似文献

Mouse pituitary tumor cells grown in tissue culture release endorphins spontaneously to the culture medium. Depolarization of these cells by incubation with high K+ concentration (56 mM) increased 2–3 folds the release of endorphins. The K+ evoked release was Ca++ dependent by that: a, removal of Ca++ ions inhibited 90% of K+ stimulated release. b, ethyleneglycol-bis (β-aminoethyl ether) N,N′-tetraacetic acid (EGTA) inhibited release of endorphins in the presence of high K+ and Ca++. It is suggested that dual regulatory system inhibit and/or stimulate in-vivo release of endorphins from the pituitary glands.  相似文献

O6-Methyl[8-3H]deoxyguanosine in a synthetic DNA polymer, poly(dC, dG, m6dG), is demethylated by cell-free extracts of EscherichiacoliBr adapted by exposure to N-methyl-N′-nitro-N-nitrosoguanidine, as shown by the appearance of 3H-labeled deoxyguanosine in hydrolysates of the recovered DNA. The demethylating activity could not be detected in extracts of nonadapted E. coli. These results provide direct evidence that a previously described inducible repair activity in E. coli acts by demethylating O6-methylguanine at the DNA level.  相似文献

We determine the kinetic parameters V and KT of lactose transport in Escherichia coli cells as a function of the electrical potential difference (Δψ) at pH 7.3 and ΔpH = 0. We report that transport occurs simultaneously via two components: a component which exhibits a high KT (larger than 10 mM) and whose contribution is independent of Δψ, a component which exhibits a low KT independent of Δψ (0.5 mM) but whose V increases drastically with increasing Δψ. We associate these components of lactose transport with facilitated diffusion and active transport, respectively. We analyze the dependence upon Δψ of KT and V of the active transport component in terms of a mathematical kinetic model developed by Geck and Heinz (Geck, P. and Heinz, E. (1976) Biochim. Biophys. Acta 443, 49–63). We show that within the framework of this model, the analysis of our data indicates that active transport of lactose takes place with a H+/lactose stoichiometry greater than 1, and that the lac carrier in the absence of bound solutes (lactose and proton(s)) is electrically neutral. On the other hand, our data relative to facilitated diffusion tend to indicate that lactose transport via this mechanism is accompanied by a H+/lactose stoichiometry smaller than that of active transport. We discuss various implications which result from the existence of H+/lactose stoichiometry different for active transport and facilitated diffusion.  相似文献

The transport of purine derivatives into vacuoles isolated from Saccharomyces cerevisiae was studied. Vacuoles which conserved their ability to take up purine compounds were prepared by a modification of the method of polybase-induced lysis of spheroplasts.Guanosine > inosine = hypoxanthine > adenosine were taken up with decreasing initial velocities, respectively; adenine was not transported.Guanosine and adenosine transporting systems were saturable, with apparent Km values 0.63 mM and 0.15 mM respectively, while uptake rates of inosine and of hypoxanthine were linear functions of their concentrations.Adenosine transport in vacuoles appeared strongly dependent on the growth phase of the cell culture.The system transporting adenosine was further characterized by its pH dependency optimum of 7.1 and its sensitivity to inhibition by S-adenosyl-l-methionine.In the absence of adenosine in the external medium, [14C]adenosine did not flow out from preloaded vacuoles. However, in the presence of external adenosine, a very rapid efflux of radioactivity was observed, indicating an exchange mechanism for the observed adenosine transport in the vacuoles.In isolated vacuoles the only purine derivative accumulated was found to be S-adenosyl-l-homocysteine.  相似文献

Controlled, prospective studies were performed to compare detection of cell culture mycoplasmas by ratio of uptake of tritiated uridine (UdR) to tritiated uracil (U) and by microbiological culture. Culture was by standard agar and broth inoculation with aerobic and anaerobic incubation; immunofluorescent staining of indicator cell cultures was used to detect M. hyorhinis. The ratio of uptake of UdR to U (UdRU) and interpretation of test results were by standard published methods and performed in triplicate. 115 cell cultures were simultaneously assayed by the two techniques. 84 cultures (73.1%) yielded agreement between the 2 methods; 2 cultures (1.7%) yielded conflicting results, and 29 cultures (25.2%) yielded UdRU results in the questionable range. Conflicting results consisted of two negative UdRU tests in mouse cell cultures infected with M. orale. In separate studies, 3T-6 cultures freshly infected with M. orale yielded negative UdRU results 3 days after infection, questionable results after 10 days and a positive UdRU 17 days after infection. UdRU detected infection in fibroblast, epithelial, and lymphocyte cell cultures. Highest UdRU ratios were detected in human skin fibroblasts at early population doubling levels (PDLs), 4064 in one culture at PDL4. UdRU was determined for IMR-90, a human diploid fibroblast at 12 different PDLs using the same lot of media. UdRU gradually decreased throughout the life of the culture, from 2 125 at PDL6 to 340 at PDL36. Cultures in phase III and others exhibiting poor growth frequently yielded questionable or false-positive UdRU results and were not included in tabulations of these results. UdRU determined in endothelial cell cultures decreased as population density increased. In a representative experiment performed over a 4-day period, the UdRU values were 1 808, 955 and 356 when the number of endothelial cells in culture were 5.3 × 105, 6.6 × 105 and 1.1 × 106, respectively.  相似文献

The primary culture of kidney cells from vitamin D deficient chicks is described. After four days in culture the cells reach confluency and retain their ability to metabolize 25-hydroxyvitamin D3 to 1,25-dihydroxyvitamin D3. Addition of one unit of bovine parathyroid hormone to the culture medium for 48 hours prior to assay had no effect on the cells' ability to produce 1,25-dihydroxy vitamin D3, whereas after 24 hours in the presence of 5×10?8M 1,25-dihydroxyvitamin D3 the cells produced not this metabolite, but 24,25-dihydroxyvitamin D3. This cell culture system will allow the investigation of the regulation of renal 25-hydroxyvitamin D3 metabolism under controlled in vitro conditions.  相似文献

A general method has been developed for determining the rate of entry of lactose into cells of Escherichia coli that contain β-galactosidase. Lactose entry is measured by either the glucose or galactose released after lactose hydrolysis. Since lactose is hydrolyzed by β-galactosidase as soon as it enters the cell, this assay measures the activity of the lactose transport system with respect to the translocation step. Using assays of glucose release, lactose entry was studied in strain GN2, which does not phosphorylate glucose. Lactose entry was stimulated 3-fold when cells were also presented with readily metabolizable substrates. Entry of o-nitrophenyl-β-d-galactopyranoside (ONPG) was only slightly elevated (1.5-fold) under the same conditions. The effects of arsenate treatment and anaerobiosis suggest that lactose entry may be limited by the need for reextrusion of protons which enter during H+/sugar cotransport. Entry of o-nitrophenyl-β-d-galactopyranoside is less dependent on the need for proton reextrusion, probably because the stoichiometry of H+/substrate cotransport is greater for lactose than for ONPG.  相似文献

Antranilate N-acetlytransferase, which is a constitutive enzyme, is responsible for the formation of N-acetylanthranilic acid which accumulated int he culture medium of certain mutants of Aerobacter aerogenes. It has been shown to be dissimilar to serine O-acetyltrasferase and not to be involved in the acetylation of a variety of aliphatic compounds. Aniline and m-aminobenzoic acid are, however, readily acetylated, the Km for the latter compound being the same as that for anthranilic acid, 13 mM. p-Aminobenzoic acid is only slowly acetylated and salicylic acid only acted as an inhibitor of the reaction. N-[3H]Acetyl[1,7-14C2]anthranili acid was prepared but could not be shown to be deacylated for further metabolized when administered to any whole cell, cell extract or toluene-lysed cell preparation.  相似文献

Three phosphorylated dinucleosides designated HS1, HS2, and HS3, isolated from the water-mould Achlya, were shown to significantly inhibit ribonucleotide reductase activity from Achlya. All three compounds decreased CDP reduction in fungal extracts by 50% at concentrations of 0.1mM. At the same concentration HS3 also inhibited partially purified CDP reductase from Chinese hamster ovary cells by at least 80% but showed only 10% inhibition with enzyme from E.coli. ADP reductase activity from Achlya was inhibited 50% by both HS1 and HS3 at 0.1mM. HS2 however, showed no inhibitory effect on purine reduction. The levels of ribonucleotide reductase during the asexual growth cycle of Achlya correlated with thymidine uptake into DNA and with the synthesis of HS compounds.  相似文献

The sole addition of a high, unphysiological, concentration of nicotinamide (25 mM) to a cell culture medium was found to maintain the cytochrome P 450 concentration of rat hepatocytes cultured for 24 hours at 71% of the level found in intact liver, whilst hepatocytes cultured without nicotinamide contained only 20% of their initial cytochrome P 450. Furthermore the P 450 concentration of hepatocytes cultured for 24 hours in the presence of 25 mM nicotinamide could be increased to the same level as found in intact rat liver by the inclusion of 1 mM nicotinamide into the medium used for cell isolation. Although the mechanism of action of nicotinamide is unknown this simple system for the maintenance of cytochrome P 450 in hepatocyte culture could provide the opportunity to study, under defined conditions in vitro, the factors that regulate cytochrome P 450 and hence determine hepatotoxicity and hepatocarcinogenesis.  相似文献
