Most of the nonstarch lipids in brown rice (Oryza sativa) of three rices differing in amylose content were contributed by bran, germ, polish and subaleurone layer. Nonstarch lipids consisted of 82–91% neutral lipids (of which 73–82% were triglycerides), 7–10% phospholipids and 2–8% glycolipids. Linoleic, oleic and palmitic acids were the major fatty acids. Nonwaxy (24 and 29% amylose) milled rice had proportionally more starch lipids and less nonstarch lipids than waxy (2% amylose) milled rice. Starch lipids were mainly lysophosphatidyl choline, lysophosphatidyl ethanolamine and free fatty acids. The major fatty acids were palmitic and linoleic acids, followed by oleic acid. 相似文献

A procedure was developed for preparing lipid- and phenol-free prolamin directly from IR480-5-9 milled rice (Oryza sativa L.).The preparation consisted mainly of one protein band on analytical and SDS-polyacrylamide disc gel electrophoresis with subunit MW of 17000 and a minor fraction with subunit MW 23000. The prolamin eluted as a single peak on SDS-Sephadex G-75 gel filtration and on DEAE-cellulose column chromatography. Prolamin was poor in lysine, histidine, cystine, and methionine but rich in glutamic acid, tyrosine and proline. In dehulled developing grain of two different rices, changes in the aminogram of the prolamin fraction coincided with the start of endosperm protein body synthesis and the appearance from 7 days after flowering of a second prolamin subunit with MW 23 000. 相似文献

Aminograms and SDS-polyacrylamide electrophoresis of milled rice glutelin of 12 Oryza sativa samples showed similar composition and ratio of 1 : 1 : 1 for subunits with MW 38 000:25 000: 16 000, indicating little possibility of finding variants of rice glutelins. Fractionation of S-cyanoethyl glutelin of 3 rices on polyacrylamide-agarose gels gave MW subunits differing in amino acid analysis of which the subunits with MW > 38 000 had the highest lysine content. Of the solubility fractions of endosperm glutelin, the fraction extracted by 0.5 M NaCl-0.6 % β-mercapto-ethanol-0.5% SDS was closest to glutelin in properties. In the developing grain of two varieties, appearance of protein bodies and rapid synthesis of glutelin from 7 days after flowering onward coincided with a drop in lysine content and appearance of MW 38 000 and 25 000 of crude glutelin. The MW 38 000 subunit is thus unique to endo-sperm glutelin. 相似文献

Five rices (Oryza sativa L.) differing in final grain size were studied at the midmilky stage to determine if any factor could be identified which might limit rate of starch accumulation. Only UDP glucose pyrophosphorylase activity increased with increasing grain size. Detached rice panicles incubated in liquid medium containing 1% sucrose and 0.1% glutamine, in addition to minerals and vitamins, produced grains similar to those on intact plants. Sucrose level (0–1.5%) in the medium determined the extent of dry matter and starch accumulation and influenced physiological development of the ripening grains. Chemical and enzymic composition of the grain were similar to previously reported levels in grains of intact panicles analysed at regular intervals after anthesis. Addition of 3-P glycerate or K+ to the medium did not improve dry matter accumulation in the developing grain. 相似文献

The moisture, lipids and fatty acid composition of developing winged bean (Psophocarpus tetragonolobus) seeds were studied. The moisture content decreased steadily as the seeds matured. The lipid content increased gradually and reached a maximum ca 6 weeks after flowering (WAF). In the early stage (2 WAF) of the developing seeds there were more polar lipids (glycolipids and phospholipids) than neutral lipids but, as the seeds developed, neutral lipids gradually accumulated while the polar lipids decreased until 6 WAF. Thereafter, both the neutral lipid and polar lipid levels remained little changed. The amounts of palmitic and stearic acids decreased, but the level of behenic acid increased as the seeds matured. On the other hand, the oleic acid content increased while that of linolenic acid decreased rapidly as the seeds matured. The concentration of linoleic acid, however, fluctuated during the development of the seeds. 相似文献

The lipid composition of tomato fruit and its mitochondrial fraction were examined at various stages of fruit ripeness. Phosphatidyl choline, phosphatidyl ethanolamine, monogalactosyl diglyceride, digalactosyl diglyceride and phosphatidyl inositol were found to be the major lipids of tomato pericarp at all stages of ripeness. Mitochondrial lipids resembled those of the parent tissue except for the absence of monogalactosyl diglyceride and a greater percentage of diphosphatidyl glycerol and phosphatidic acid. Changes in the lipid-protein ratio of mitochondria were noted with ripening. 相似文献

Salt-soluble proteins, albumin and globulin, were prepared from dehulled rice (Oryza sativa L., line IR1541-76-3) during grain development. Albumin and globulin progressively increased during grain development up to about 12 days after flowering (DAF) and then decreased slightly during grain desiccation. Free amino N was maximum at 10 DAF. Total protein and glutelin-prolamin (by difference) continued to increase up to 20 DAF. Aminogram of total protein and globulin showed a progressive decrease in lysine and threonine among the essential amino acids. Albumin showed a similar trend except for the lesser change in lysine content. Disc gel electrophoresis showed a maximum of four major and six minor protein bands for albumin and only one major and three minor bands for globulin. Sodium dodecyl sulfate-gel electrophoresis revealed three major polypeptide subunits for albumin with MW of 11 000, 8 500 and 16 000, and two for globulin with MW of 20 000 and 12 000. 相似文献

The lipids of the brown alga Fucus serratus were isolated, identified and quantified. The major acyl lipids were the three glycosylglycerides, diacylgalactosylglycerol, diacyldigalactosylglycerol and diacylsulphoquinovosylglycerol. These represent over 70% of the total acyl lipids. The fatty acid compositions of the major lipids were examined and most showed rather distinctive fatty acid contents. For example, diacylgalactosylglycerol was enriched in n-3 polyunsaturated fatty acids while phosphatidylcholine and phosphatidylethanolamine had very high levels of arachidonate. Phosphatidylglycerol contained the unusual trans-Δ3-hexadecenoic acid. The labelling of lipids and fatty acids from [14C]acetate was examined and the distribution of label between individual components as a function of the incubation period and in algae collected at different times of the year is reported. Algae collected in the winter incorporated much more radioactivity into non-esterified fatty acids when compared to algae collected in the summer. All algae could label myristate, palmitate, stearate and oleate at high rates. Longer incubation times allowed the labelling of polyunsaturated fatty acids such as linoleic acid. 相似文献

We report here a study of the incorporation and metabolism of various long chain fatty acids in SK-Hep-1 cultured hepatoma cells. Medium supplementation with radiolabelled palmitic, stearic, linoleic, -linolenic and eicosa-8, 11,14-trienoic acids (1 µM, 24 H) resulted in an active uptake of each of these precursors by the cultures. Subsequent analysis of the cellular lipids indicated that they exhibit almost all the enzymic activities of polyunsaturated fatty acid metabolism that are characteristic of normal hepatic cells. With respect to the desaturation capacities of this cell line, although -linolenic acid reacted more extensively than did linoleic acid and the conversion of 8,11,14-eicosatrienoic acid by the 5 specific enzyme was more avid than had been previously seen in normal rat or human liver: the saturated fatty acids constituted relatively poor substrates, being preferentially chain-elongated rather than (mono) desaturated at the 9 position. Analysis of the fatty acid profiles of total cellular lipids and of various lipid subclasses, however, revealed a relative paucity of essential fatty acids when compared with the abundance of endogenous monoenoic acids (particularly oleic). Of the total cellular fatty acids, 58% were present in the form of phospholipids; with 33% of the remaining 42% (i.e., the neutral lipids) being associated with triacylglycerol fraction. Within the total lipids, phosphatidyl-choline and phosphatidyl-ethanolamine were the major sites for the incorporation of all metabolic products derived from the incubated radiolabelled 16- and 18-carbon fatty acid precursors, whereas the phosphatidyl-inositol fraction was the predominat recipient of nascent arachidonic acid when the eicosatrienoate was the substrate. The express purpose of this investigation was to characterize the biochemical routes involved in the anabolism of various essential fatty acids in the human hepatocyte, through the use of cultured human hepatoma cells as an experimental model system. In view of the similarities between certain aspects of the polyunsaturated fatty acid metabolism of these cells and the corresponding properties of other mammalian hepatic or liver-derived tissues, the data presented here would thus constitute a significant beginning alone those lines. Moreover, considering the extreme difficulty in obtaining for such investigation relevant tissue samples from normal human sources, we regard these results — and the availability for use of this particular human hepatoma cell line — as important new developments in the effort to characterize a useful experimental model both for gaining immediate information and for designing future experiments. 相似文献

Phosphoglycerol, triacylglycerol, diacylglycerol, and free fatty acid content was studied in eggs of the codling moth Cydia pomonella at the white, red ring, and black head developmental stages. The composition of total phosphoglycerols and of the three classes of neutral lipids was also analyzed. The highest total lipid content was found in eggs at the white stage, the amount decreasing during development mainly as a result of a diminution in the quantity of phosphoglycerols, which account for approximately 50% of total content at all stages of egg development. The amount of triacylglycerols and free fatty acids changes significantly during development, whereas only minor changes were found in diacyglycerol levels. The total phosphoglycerol acyl composition of eggs at the white and red ring stages is similar, whereas differences are evident at the black head stage of development. Triacylglycerols and free fatty acids are enriched in saturated fatty acids in all analyzed stages. The acyl profile of diacylglycerols is different at each stage. The unsaturation index decreases in diacylglycerols and free fatty acids as a function of egg development. The results of the present paper suggest that triacylglycerols may constitute an important source of energy during the final period of egg development while phosphoglycerols may function as fuel during the beginning. Phosphoglycerols could be precursors for the triacylglycerol biosynthesis that takes place between white and red ring stages. 相似文献

Lipids from cultured cells, leaves and seeds of two varieties each of soybean (Glycine max) and oil seed rape (Brassica napus) were separated into neutral lipids, glycolipids and phospholipids and their fatty acids were analysed. Usually, the fatty acid composition differed between corresponding fractions from cultured cells, leaves and seeds. Differences were least marked in (i) the phospholipids from cultured cells and leaves of soybean and (ii) the neutral lipids from cultured cells and seeds of rape. In the cultured cells, the fatty acid composition of the phospholipids differed from that of the glycolipids and neutral lipids, and fatty acids of chain length greater than C18 comprised a large proportion of the fatty acids of the glycolipids. 相似文献

“Crude” glutelin was prepared from milled rice (Oryza sativa) flour by sequential extraction of the albumin-globulin fraction with 0.5 M NaCl and prolamin with 70% ethanol-0.6% β-mercaptoethanol. The solvent, 0.5% sodium dodecyl sulphate (SDS)-0.6% β-mercaptoethanol, extracted 91% of the endosperm glutelin without gelatinizing starch granules, whereas chaotropic solvents such as urea and guanidine caused extensive gelatinization. The S-cyanoethyl glutelin (Ce-glutelin) prepared by SDS extraction of the “crude” glutelin (9.5% protein) of IR480-5-9 rice gave three major subunits with MW 38000, 25000 and 16000 in the ratio 2:1:1 as determined by SDS polyacrylamide gel electrophoresis. A similar preparation from “crude” glutelin of a lower protein containing rice had the corresponding subunits in the ratio of 16:3:1. The MW 38000 subunit was unique to glutelin and was not present in C3-albumin-globulin or prolamin; the subunits were only partially purified by SDS Sephadex G-150 gel-filtration. The C3-glutelin was also prepared from a crude glutelin-prolamin preparation from IR480-5-9 by NaOH extractions followed by precipitation at pH 10 and ethanol extraction of the precipitate (C3-glutelin). This preparation had the same three major subunits and in the same ratio as C3-glutelin prepared by the SDS method. The subunits of the former preparation were separated by carboxymethyl Sephadex C-50 chromatography; the MW 38000 subunit eluted between pH 6.2–8.5, the MW 25000 in an impure state at pH values above 9, and the MW 16000 subunit was eluted at pH 8.6—9.2. Amino acid composition of the Ce-glutelin preparations were similar to each other. The MW 38000 and 16000 subunits had lower lysine contents than whole C3-glutelin, whereas the MW 25000 subunit had a higher lysine content. 相似文献

Agmatine deiminase activity in rice embryos increased gradually upto 24 hr during germination and then decreased. Gibberellic acid and kinetin inhibited the activity when added to the germination medium. The enzyme was purified 717 fold with specific activity 788.5 nkat/mg protein and yield 8.8%. The Mr of the native enzyme was 18.3 x 104 and the enzyme was a dimer of two identical subunits. The pH and temperature optimum of the enzyme were 6.0 and 28° respectively. The enzyme followed typical Michaelis-Menten kinetics with a Km value of 1.5 x 10?2 M. The enzyme activity was inhibited by various divalent cations and spermidine and spermine, but putrescine showed no effect. 相似文献

Multiple forms of peroxidase from ten-day-old Dee-Geo-Woo-Gen (DGWG) rice seedlings were isolated by ammonium sulphate precipitation, polyacrylamide gel electrophoresis (PAGE) and DEAE-cellulose chromatography. Cellular and chloroplast lipids of the leaves of Mimosa pudica have been analysed. Qualitatively the total lipid composition of this plant is similar to that reported for the photosynthetic tissues of other plants. Chloroplast lipids show some resemblance to those of algae. The cerebroside fraction of both leaves and chloroplasts contains a polyunsaturated fatty acid (20:4ω3) and a long chain sphingosine base whose Rf value coincides with that from ox brain cerebroside and not with that of phytosphingosine from spinach. 相似文献

The effects of cadmium (10?6, 10?5, 10?4, and 10?3 M) on the growth and development of stem apical meristem and fatty acid (FA) composition of particular lipid fractions (neutral lipids, glyco-, and phospholipids) were investigated in the shoots of Karelian birch (Betula pendula var. carelica (Mercklin) Hämet-Ahti) produced in vitro. Low cadmium concentration (10?6 M) slightly stimulated shoot morphogenesis. However, at the concentration of 10?4 M, shoot growth and development was terminated, and the concentration of 10?3 M turned out to be critical (the meristem died within the first 5 days). The presence of Cd in the medium considerably affected FA composition in the shoots. The most pronounced changes occurred in the fraction of glycolipids, and the extent of adverse influence significantly depended on metal concentration in the medium. At the Cd concentration of 10?4 M, the content of di-and trienoic FAs decreased by 3–4 times, whereas the level of monoenoic FAs rose by 5 times. The revealed differences in the FA composition of lipids in birch were considered as an indicator of adverse effect of cadmium on the structure and functions of chloroplasts and therefore on photosynthesis. 相似文献

Neutral lipids of ambrosial fungi, nutritionally important to development of Xyleborus ferrugineus beetle, were analyzed for fatty acid composition. Predominant components were 16:0, 18:0, 18:1 and 18:2 in all three mutualistic fungi, Fusarium solani, Cephalosporium sp. and Graphium sp. The level of lysyl- and prolyl-tRNA in various stages of the maturing wheat grain was measured by the aminoacylation procedure. The levels of these tRNAs changed only slightly during the maturation period. Several species of lysyl- and prolyl-tRNA were obtained from different parts of the developing grain by fractionation on benzoylated-DEAE cellulose (BD-cellulose). The embryo contained three discrete species of prolyl and at least three species of lysyl isoacceptor tRNA throughout development, whilst the tRNA obtained from the endosperm gave more complicated elution profiles on chromatography on BD-cellulose. Small changes were noted in the levels of aminoacylation of individual isoacceptor tRNA species for lysine or proline during seed maturation. However, these were insufficient to account for the changing pattern of lysine and proline in the storage protein during the development of the endosperm. 相似文献

Forty-five 1,3,5-triazine-2,6-dione derivatives were synthesized and their plant growth-promoting activities examined by the rice (Oryza sativa) seedling test in the presence or absence of gibberellic acid (GA). For high activity in promoting the growth of rice seedlings and acting as active GA-synergists, a para-substituted or a 2,4-disubstituted phenyl group, a hydrogen atom and an alkoxy group were required in the 1-, 3- and 4-positions of the 1,3,5-triazine-2,6-dione molecule. 4-Ethoxy-1-(p-tolyl)-s-triazine-2,6(1H, 3H)-dione [TA], one of the most potent triazinones, synergized the effect of GA on the shoot elongation of different varieties of rice including normal type, dwarf mutants and chlorophyll-mutants. TA synergistically increased the growth-promoting activity of GA by both a simultaneous treatment at the same sites and separate treatments at separate sites of rice seedlings. 相似文献
