Summary Electrophysiological methods were used to examine the effectiveness of food odors in stimulating different olfactory receptor types inPeriplaneta.The sense cells occur in certain fixed combinations in particular sensilla, thus defining a physiological sensillum type.The substances used as stimuli were chopped samples of foods of plant and animal origin.Each of the receptor types investigated responded to several kinds of food. No receptor type was observed to respond specifically to one kind only.Because the food odors always contain compounds included in the response spectra of several cell types, it must be assumed that inPeriplaneta the recognition of food odors depends not on the detection of key odors but on the complicated patterns of excitation in the receptors of different types that arise from differences in the combination of unspecific odor components.The components of an odor are not present in constant quantities over a period of time. Accordingly, the measured response profiles of the receptors vary. However, these variations are never so great that a substance strongly effective on one trial is ineffective on another.  相似文献

Chloroplast photorelocation movement, well-characterized light-induced response found in various plant species from alga to higher plants, is an important phenomenon for plants to increase photosynthesis efficiency and avoid photodamage. The signal for chloroplast accumulation movement connecting the blue light receptor, phototropin, and chloroplasts remains to be identified, although the photoreceptors and the mechanism of movement via chloroplast actin filaments have now been revealed in land plants. The characteristics of the signal have been found; the speed of signal transfer is about 1 µm min?1 and that the signal for the accumulation response has a longer life and is transferred a longer distance than that of the avoidance response. Here, to collect the clues of the unknown signal substances, we studied the effect of temperature on the speed of signal transmission using the fern Adiantum capillus-veneris and found the possibility that the mechanism of signal transfer was not dependent on the simple diffusion of a substance; thus, some chemical reaction must also be involved. We also found new insights of signaling substances, such that microtubules are not involved in the signal transmission, and that the signal could even be transmitted through the narrow space between chloroplasts and the plasma membrane.  相似文献

Summary Electrophysiological investigation has identified a cold receptor on the antenna of Periplaneta americana. This receptor responds neither to variations in light intensity, to mechanical stimuli ranging from sound to touch, nor to any chemical stimulus yet tested. The afferent discharge rate is fairly steady as long as the temperature is constant (almost always below 25 impulses/sec when the temperature varies less than 0.03° C during several seconds). When the temperature is rapidly lowered to a new level, the impulse frequency rises quickly to a peak whose height shows dependence on the extent of cooling (delta-temperature). During the course of the next few seconds the frequency falls below 25 impulses/sec. Rapid rewarming to the original temperature has the opposite effect. Impulse frequency drops sharply at first and then rises during the following seconds to the original level.
Zusammenfassung Durch elektrophysiologische Untersuchungen wurde ein Kälterezeptor auf der Antenne von Periplaneta americana gefunden. Er spricht weder auf Änderung der Lichtintensität, auf Schall, Berührung, Luftströme von 2,5 m/sec Geschwindigkeit, noch auf irgendeinen bisher geprüften chemischen Reizstoff an. Seine stationäre Entladungsfrequenz (nur selten über 25 Hz) bleibt bei konstanter Temperatur (Änderung unter 0,03° C über mehrere Sekunden) ziemlich konstant. Bei plötzlicher Abkühlung auf eine zweite Temperaturebene steigt die Impulsfrequenz schnell an und erreicht meistens innerhalb 50 msec ein Maximum, dessen Höhe eine Abhängigkeit von der Differenz der Ausgangs- und End-temperatur zeigt. Nach dem Maximum nimmt die Frequenz rapide ab, bis sie schließlich wieder konstant bleibt. Bei schneller Erwärmung auf die Ausgangstemperatur setzen die Impulse kurz aus, erreichen aber nach einigen Sekunden ihren ersten Erequenzwert wieder.

Olfactory receptor neurons (ORNs) respond to odorants with characteristic patterns of action potentials that are relevant for odor coding. Prolonged odorant exposures revealed three populations of dissociated toad ORNs, which were mimicked by depolarizing currents: tonic (TN, displaying sustained firing, 49% of 102 cells), phasic (PN, exhibiting brief action potential trains, 36%) and intermediate neurons (IN, generating trains longer than PN, 15%). We studied the biophysical properties underlying the differences between TNs and PNs, the most extreme cases among ORNs. TNs and PNs possessed similar membrane capacitances (approximately 4 pF), but they differed in resting potential (-82 versus -64 mV), input resistance (4.2 versus 2.9 G(Omega)) and unspecific current, I(u) (TNs: 0 < I(u) 1 pA/pF). Firing behavior did not correlate with differences in voltage-gated conductances. We developed a mathematical model that accurately simulates tonic and phasic patterns. Whole cell recordings from rat ORNs in fragments (approximately 4 mm(2)) of olfactory epithelium showed that such a tissue normally contains tonic and phasic receptor neurons, suggesting that this feature is common across a wide range of vertebrates. Our findings show that the individual passive electrical properties can govern the firing patterns of ORNs.  相似文献

We have documented odor responses of all morphological classes of sensilla on the surface of theDrosophila antenna: sensilla basiconica, sensilla trichodea, and sensilla coeloconica. Both subtypes of s. basiconica, large and small, respond to odors. S. trichodea fall into different functional types. Type 1 appears narrowly tuned, as it responded only tocis-vaccenyl acetate, believed to be a pheromone. Type 2 responded totrans-2-hexenal and 4-methyl cyclohexanol. These two types of s. trichodea are differentially distributed on the antennal surface, and have dramatically different frequencies of spontaneous action potentials. Likewise, there are multiple types of s. coeloconica. One type is broadly tuned, responding most strongly to a test stimulus of butyric acid, but also to a variety of other odors; it is restricted to the dorso-medial portion of the third antennal segment. A second type gave detectable responses only totrans-2-hexenal. These results demonstrate that all classes of sensilla are olfactory, and they reveal the organizational complexity of theDrosophila olfactory system.  相似文献

Important differences in binding characteristics between agonists and antagonists of the beta-adrenergic receptor have been described. However, these observations have been complicated since most available antagonists are much more lipophilic than agonists. In order to separate out those binding characteristics of agonist vs. antagonist from those characteristics of lipophilic vs. hydrophilic ligands, we have studied competition of the hydrophilic ligands isoproterenol (agonist) and CGP-12177 (antagonist) with [125I]iodopindolol binding in intact human lymphocytes. Analyzing competition curves from assays performed at 13 degrees C, 25 degrees C and 37 degrees C we demonstrated that at lower temperatures there was a decrease in IC50 for isoproterenol but not for CGP-12177. Using cells preincubated with isoproterenol then extensively washed, competition curves with both isoproterenol and CGP-12177 were biphasic, and characterized by the appearance of a population of receptors with a low affinity for both hydrophilic ligands. Furthermore, at lower temperatures the biphasic nature of these curves was accentuated and was characterized by a 6-fold and 40-fold increase in the apparent KD of a population of low affinity sites for isoproterenol and CGP-12177, respectively.  相似文献

Modulation of baroreceptor reflex (BRR) by endogenous substance P (SP) in the brain was investigated in rats anesthetized with pentobarbital sodium. Intracerebroventricular administration of the undecapeptide (15 or 30 nmol) and its antagonist, (D-Pro2, D-Trp7,9)-SP (30 or 60 nmol) or SP antiserum (1:20), respectively, promoted a significant increase and decrease in the sensitivity of BRR response. Prolonging the endogenous activity of SP with the aminopeptidase blocker, bestatin (200 nmol) or with the endopeptidase-24.11 inhibitor, phosphoramidon (200 nmol) significantly augmented the same reflex. Combining the undecapeptide with either peptidase blocker, moreover, promoted additional potentiation of the BRR response. On the other hand, simultaneous administration of bestatin and (D-Pro2, D-Trp7,9)-SP produced a reduction of the augmented effect of bestatin on the sensitivity of BRR response. Bilateral microinjection of SP (600 pmol) or an antiserum against SP (1:20) into the nucleus tractus solitarius (NTS) elicited respectively an enhancement of and reduction in the BRR response. These data suggest that neurons that contain SP may participate in central cardiovascular control by tonically enhancing the sensitivity of the BRR response, possibly via an action on the NTS.  相似文献

The ligand binding and G-protein coupling of the bovine hippocampal 5-HT1A receptor as a function of temperature was monitored. There is an almost complete and irreversible loss in agonist binding at 50°C. However, the antagonist binding is reduced only by 50%, and this could be reversed if the temperature is lowered to 25°C. Interestingly, the agonist binding of the 5-HT1A receptor in membranes exposed to 50°C is inhibited to a much lesser extent by GTP-γ-S, a non-hydrolysable analogue of GTP, indicating uncoupling of the 5-HT1A receptor to G-proteins at 50°C. We propose that high temperature selectively and irreversibly inactivates G-proteins thereby affecting G-protein-receptor interaction and agonist binding of the 5-HT1A receptor.  相似文献

Chicken prolactin receptor (cPRLR) deciphered from the cDNA sequence showed a unique double antenna structure in its extracellular domain. The predicted cPRLR preprotein was composed of 831 amino acids and contained a signal peptide and a transmembrane region. The extracellular domain comprised 438 residues, and was divided into two tandemly repeated, highly homologous units, each of which corresponded to the extracellular domains of mammalian prolactin receptors. Both extracellular units of cPRLR possessed two structural features characteristic of the ligand binding units of cytokine/prolactin receptor family, namely two pairs of cysteine residues and a WSXWS motif. These findings strongly suggest that cPRLR contains two repeated ligand binding units, that is a double antenna structure. The cPRLR gene is expressed in a wide range of tissues of laying hen.  相似文献

This study was conducted to address the effect of different constant temperatures (15, 20, 25, 30 and 35 °C) on the functional response of the parasitoid wasps Diglyphus isaea Walker and Hemiptarsenus zilahisebessi Erdös to different densities of Liriomyza sativae Blanchard (2, 4, 8, 16, 32 and 64 larvae) under laboratory conditions. The results revealed the Type II functional response for both parasitoids at different temperatures. The highest searching efficiency for D. isaea and H. zilahisebessi occurred at 25 °C (0.926 ± 0.211 h−1) and 30 °C (1.012 ± 0.241 h−1), respectively. In addition, the shortest handling time for D. isaea and H. zilahisebessi were observed at 25 °C (0.063 ± 0.008 h) and 30 °C (0.058 ± 0.008 h), respectively. These results demonstrated that H. zilahisebessi is more efficient at higher temperatures than D. isaea. Both parasitoids had higher parasitism performance when temperature increased (4.67 parasitized hosts/day at 15 °C vs 15.87 parasitized hosts/day at 25 °C for D. isaea and 3.89 parasitized hosts/day at 15 °C vs 17.24 parasitized hosts/day at 30 °C for H. zilahisebessi). In addition, a quadratic regression was found between handling time and temperature as well as between number of the parasitized larvae and temperature in D. isaea and H. zilahisebessi at different densities of L. sativae. This study provided a preliminary information on the parasitic behavior of these parasitoids and that D. isaea and H. zilahisebessi can be used properly beside other non-chemical approaches to manage L. sativae damage at a temperature range of 25–30 °C, respectively.  相似文献

Environmental variables like temperature are important factors that affect the efficiency of biological control agents in greenhouse crops. This study examined the effect of temperature on the functional response of an acarophagous thrips Scolothrips longicornis Priesner to different densities of two spotted spider mite, Tetranychus urticae Koch in laboratory conditions. Five constant temperatures (15, 20, 26, 30, and 35 °C) and seven prey densities (2, 4, 8, 16, 32, 64, and 128) were used in the experiments. At each temperature, 2 to 128 adult of T. urticaue were exposed to a female S. longicornis for a 24 h period. The results indicated that the predatory thrips exhibited type II functional response to different densities of T. urticae at all examined temperatures. Temperature had significant effect on the predation capacity of the adult thrips over the range of 15–35 °C. Instantaneous attack rate (a) of the predator increased linearly with increasing temperature from 15 to 35 °C. At 35 °C, the highest instantaneous attack rate was estimated to be 0.0579 day? 1 using the Holling model and 0.1480 day? 1 using Rogers model. The estimated value of handling time (Th) using both Holling and Rogers models decreased linearly with increasing temperature from 15 to 35 °C. Scolothrips longicornis achieved higher predation at higher temperature (16.1 preys/day at 35 °C). This observation suggested that S. longicornis may be more effective for biological control of two spotted spider mite in warmer conditions.  相似文献

Dimethylsulfoxide (DMSO) is known to protect isolated enzymes during freezing while destabilizing proteins at high temperatures. This apparent paradox is the subject of a review by Arakawa et al. ((1990) Cryobiology 27, 401-415), who present evidence for a temperature-dependent, hydrophobic interaction between DMSO and non-polar moieties of proteins. The present study investigates the interaction of DMSO with phospholipid bilayers. Phospholipid vesicles containing carboxyfluorescein were exposed to several concentrations of DMSO at various temperatures. Leakage rates increased with DMSO concentration and temperature. This effect was not reduced in the presence of solutes that have been shown to neutralize DMSO toxicity in tissues. The increased leakage rates correlate well with the increased partitioning of DMSO from water to octanol at higher temperatures. Additionally, reductions in the CH2 vibrations of the bilayer are also shown to depend on DMSO concentration and temperature. A similar reduction in CH2 vibrations was observed in solutions of octanol and DMSO, suggesting that this effect is not mediated through an interaction with water. Furthermore, investigation of sulfoxide vibrations indicate that DMSO is not hydrogen bonded to the alcohol moiety of octanol, and therefore the interaction between DMSO and octanol is most likely due to a hydrophobic association. These results are consistent with a destabilization of phospholipid membranes at higher temperatures due to a hydrophobic association between DMSO and the bilayer.  相似文献

Cell culture on gels made of poly(ethylene oxide) and poly(propylene oxide) (Pluronic), which has a lower critical solution temperature around 30 degrees C, could be performed for 48 h. However, the Pluronic gels were highly hydrophilic and tended to dissolve in the culture medium. We achieved temperature-dependent detachment of cells from Pluronic gels containing or lacking extracellular matrix (ECM) by cooling the gels to 4 degrees C. Using normal human umbilical vein endothelial cells (HUVECs) grown on and released from Pluronic gels lacking ECM, we further found that the expression ratio of the surface markers CD34 and CD105 was twofold higher than for cells grown on polystyrene and removed with trypsin. In addition, the expression ratios for CD34 and CD105 on HUVECs cultivated on the Pluronic gels containing higher concentrations of ECM were lower, which may be due to ECM coating of the cell surface and, thus, interference with antibody binding. In summary, temperature-dependent detachment of cells from Pluronic gels allows the isolation of cells under mild conditions. This can be a powerful tool for surface marker analysis by flow cytometry.  相似文献
