The determination of the members of Charales (Charophyceae) is not easy. The main reason of this is a wide range of variation in vegetative characters within most of the species. Many features such as the extent of branchlet cortication, the degree of incrustation, length and number of bract cells, spine cells, stipulodes, diameter of the main axis and oospore size and color do not have universal taxonomic significance. In similar, many Nitella species characters such as: general habitus, branchlets and presence of the top segment (mucro), are helpful in determination, however they are often not developed. In this case, the features of oospore wall ornamentation revealed by light and scanning microscopy can be important taxonomic characters of the Charales. The present study shows interspecific variations of the oospore in two Nitella species from Poland which can be helpful in correct determination of both species. Some of the oospore features like number of striae and fossa width can not be used, as they appeared statistically unimportant, and cannot be used in distinguishing between both species. The presented results are the first detailed report of oospore wall ornamentation pattern from Poland for the species studied. Oospore features are presented in detail on the background of some additional tips and among them particularly: length and width of the oospore, ISI idex, width of fossa, width of striae and number of striae.  相似文献

The ultrastructural features of oospore wall ornamentation patterns revealed by scanning electron microscopy are important taxonomic characters of the Charales. The present study shows inter-and intrapecific variations in 19 species of the genera Chara, Lamprohamnium, Nitella and Tolypella. This is the first time that the oospore wall ornamentation of Swedish Charales has been documented in detail. In the studied Chara species the ornamentation within species was variable, and partly overlapping between species, but only between closely related species. In contrast, the Nitella species showed distinct differences in ornamentation patterns between species, although the same patterns can be found in different species. This study presents for the first time SEM images of the species Chara rudis (pustular ornamentation), Nitella opaca (pitted ornamentation) and Nitella wahlbergiana (anastomosing network ornamentation). The ornamentation pattern in the Nordic species TV. wahlbergiana , supports its separation from Nitella mucronata which has a reticulate ornamentation. The relationship between length and width of the oospores is also of taxonomic significance. Nitella and Tolypella oospores are roundish, whereas those of Chara and Lamprothamnium are elongate. The extent to which environmental and genetic factors can affect oospore size and shape within a species are still unknown, but in the present study both ornamentation pattern and size provided evidence for the distinction between Chara globularis and Chara aspera. Some further taxonomic problems in the Charales are discussed in the light of the results of this study.  相似文献

Biometric measurements were performed on oospores of Chara tomentosa from Core 63-J from Lake Zeribar in Iran. The germinability of Chara vulgaris oospores collected from the sediments of four Ontario lakes varies considerably, ranging in germination percentage from 7% to 54%. Chemical analysis of the interstitial water of the sediments indicated that oospores with low germination occurred in lakes which have high acid volatile sulfides (H2S, FeS, HS) and high soluble Fe2+. The inhibitory effects of sediment on oospore germination were demonstrated by transplant experiments, and suggested that sulfide was the toxic agent. Exposure of high-germinating sedimentary oospores to free sulfide concentrations greater than 2.0 mM caused a greater than 30% reduction in oospore germination. The presence of sulfide in sediments was shown to result from sulfate reduction by bacteria in sediment pore water of those lakes where oospore viability was lowest. Differences in oospore germination percentage appear, therefore, to be due to the toxicity of H2S produced in the sediment, either by a direct effect on the oospore, or on the parent plants.  相似文献

Characteristics of the oospores have been used to delimit sections and, in some cases, species in the genus Tolypella A. Braun. To test the utility of oospore characters for identifying North American species of Tolypella, we investigated oospores from field‐collected and herbarium specimens. Oospore dimensions (length, width, and length to width ratio) and morphology (color, ridge number and shape, wall ornamentation, and basal impression number) were measured. Oospore dimensions were statistically analyzed and oospore morphology was studied with light and scanning electron microscopy. Statistical analyses showed significant differences in length, width, and length to width ratios among most Tolypella species and populations but there was considerable overlap, which suggested that species identification based on oospore measurements alone is not wholly reliable. The ultrastructural features of oospore wall ornamentation in the genus Tolypella were examined using scanning electron microscopy (SEM). The taxonomic relationships among several species were discussed on the basis of oospore ultrastructure and measurements. In the case of T. glomerata and T. nidifica, our results support the status of separate species. Close relationships and transitional forms may exist between T. nidifica and T. normaniana, and not only in oospore wall ornamentation. Oospores of T. hispanica exhibited the same distinct type of reticulate oospore wall as previously reported, but our results do not support the recognition of T. hispanica as a separate species. Ultrastructure of the oospore walls of T. prolifera and T. intricata was almost identical, suggesting that these species are closely related. We therefore reject previous suggestions that morphological characteristics of oospores as observed in SEM are sufficient for identification of individual species. Although significant differences were found among oospores in individual species of Tolypella, large variation among populations, and among individuals belonging to the same population, caused substantial overlap among species.  相似文献

Results from the present study suggest that Chara braunii encompasses a complex of geographical races or incipient species that although morphologically similar, are isolated to varying degrees ranging from partial interfertility to complete reproductive incompatibility. Most of the 6 clones studied were interfertile to the extent that fertilization was followed by the development of black oospores, but subsequent germination tests disclosed that many of these either failed to germinate or gave rise to offspring that were partially or completely sterile. No support was encountered for the suggestion that C. braunii is of recent development or that it has been distributed rapidly throughout the world.  相似文献

The paper deals with cytotaxonomic studies of three forms ofChara braunii-f.coromandelina (n = 14), f.schweinitzii (n = 14) and f.kurzii (n = 28) from West Bengal (India). An improved technique involving the use of a pretreating agent permitted a detailed analysis of the karyotypes of the three taxa studied. It has been concluded that structural alterations of chromosomes and polyploidy are associated with evolution of phenotypic differences in theChara braunii complex. Oospores of Peronospora manshurica, the causal agent of soybean downy mildew, were stained by a variety of techniques. TTC (tetrazolium chloride) and NBT (nitroblue tetrazolium chloride) primarily stained oospores which were cytologically abnormal and appeared degenerating. Cytological normal oospores were not stained by these compounds presumably because the dyes were excluded from the oospore cytoplasm by the oospore wall or the plasmalemma. Strong autofluorescence of dead/degenerating oospores in the FDA test (fluorescein diacetate) made scoring of the oospore viability by this technique unreliable. Phloxine B was found in a consistent way to stain the degenerating oospores and a small proportion of the oospores which by light microscopic, observations could not be scored cytologically abnormal. Control experiments with live and dead cells of yeast (Saccharomyces cerevisiae) confirm that phloxine B is excluded from live cells and dead cells become stained. The presumed mode of action is that the semipermeability of the plasma membrane of live cells excludes the stain. The phloxine B test described here appears a promising technique for the determination of oospore viability of P. manshurica.  相似文献

Dimensions of oospores such as length, width, fossa-breadth, length-to-width ratio and number of striae were considered for morphological identification of the members of Charales. Oospores were collected from natural stands of three species, viz. Chara braunii, C. corallina and C. wallichii in ponds in West Bengal (India). Oospore dimensions (length, width and fossa-breadth) correlated well: though the fossa breadth–length relation of C. braunii showed lower correlation coefficient (r2 = 0.076, p < 0.001), all other five breadth or width–length relationships showed r2 ≥ 0.23, p < 0.001. These bivariate patterns were significantly different among the species (one-way ANCOVA of length–width ratio to fossa-breadth: adjusted means and slopes both p < 0.001), C. braunii and C. corallina has similar length to width ratios, C. wallichii and C. corallina were similar in width, whereas the species differed in breadth and length in the order C. braunii, C. wallichii, and C. corallina.  相似文献

The influence of nitrogen (N) deficiency on the cell growth and intracellular lipid production of the alga Botryococcus braunii UTEX 572 was investigated. Biomass concentration and lipid content of B. braunii cultivated in modified Chu-13 medium containing 0.04, 0.37, and 3.66 mM nitrate were 0.23–0.38 g L−1 and 36–63% of dry cell weight, respectively. The specific growth rate of B. braunii reached a constant of 0.185 day−1 during cultivation with an initial nitrate feed of 3.66 mM. The maximum lipid content of B. braunii was 63% with 0.04 mM nitrate. However, the maximum lipid productivity of 0.019 g L−1 day−1 was achieved with 0.37 mM nitrate. The level of oleic acid, an important component of biodiesel, was higher at 86% of the total fatty acids under N-limited conditions (0.04 mM nitrate) compared to 69% under N-sufficient conditions (3.66 mM nitrate). Furthermore, expression of the stearoyl-ACP desaturase gene (sad) encoding a stearoyl-ACP desaturase involved in the synthesis of oleic acid was 2.6-fold higher under N-limited conditions than under N-sufficient conditions.  相似文献

When dark-grown mature oospores of Phytophthora cactorum were activated to germinate by exposure to 5 uW cm-2nm-1 of fluorescent light at 20–22°C in the presence of certain flavin inhibitors such as KI, salicylhydroxamic acid and phenylaceric acid at 40. 1. and 0.1 mM respectively, photoactivation and hence subsequent germination of oospores were inhibited without appreciable irreversible effect on oospore viability. Likewise, when applied during the light period, NaN3 and KCN at 1 mM reduced photoactivation but had a minimal effect on dark reactions. Diphenylamine, an inhibitor of certain carotenoids, had no effect on photoactivation of oospores. The data suggest that the photoreceptor pigment for activation of oospore germination is a flavin.  相似文献

High moisture content of the host tissue ( 88%) and low ambient r.h. (50-54%) favoured oospore formation under controlled environments. It took 14–16 days for oospores to develop; thereafter the number of oospores increased with time and decreased with moisture content of host tissue. High ambient r.h. (> 80%) did not favour oospore formation under field or controlled conditions. Oospore formation was detected in inoculated plants grown in the field when the ambient r.h. declined to 74% and moisture content of host tissue decreased to 83.7–85.6%. It took 8 days (cv. Kufri Chandramukhi) to 13 days (cv. Kufri Jyoti and Kufri Badshah) for oospores to develop. Cultivars also differed in their response to oospore production, cv. Kufri Chandramukhi being more responsive (4800 oospores g−1 f wt) than cv. Kufri Jyoti and Kufri Badshah (1320 and 390 oospores g−1 f wt respectively). Oospores produced in vitro remained viable when buried in soil in the temperate highlands of Himachal Pradesh and sub-tropical plains of Uttar Pradesh, India for more than 150 days, i.e. beginning of the next crop season. The oospores germinated and initiated late blight infection at the base of the stems after 21–30 days of incubation of the potato plants raised in oospore-infested soil. It took 2 days for newly formed oospores to germinate and this delay time increased to 75–77 days after 180-days burial. It took 15 days for their germination (47%) in soil extract as compared to 50 days in sterilised distilled water.  相似文献

A new species of the genus Glyptothorax, Glyptothorax obliquimaculatus sp. nov. is described from the Xiaohei River, a tributary of the Nanting River, Salween drainage, in southwestern Yunnan province, China. This new species can be distinguished from its congeners by the following combination of characteristics: unculiferous ridges of the thoracic adhesive apparatus extending anteriorly onto the gular region; body with irregular dark blotches scattered along lateral surface (blotches mostly oblique); skin smooth on head and body; dorsal spine smooth without serrations on its posterior margin; lips smooth; posterior margin of pectoral spine with 7–8 serrations; dorsal-fin base 11.0–13.2% SL; pectoral-fin length 15.6–19.6% SL; depth of caudal peduncle 8.6–9.8% SL; head width 19.1–24.0% SL; nasal barbel length 23.3–33.3% HL.  相似文献

Many of the genes that control photosynthesis are carried in the chloroplast. These genes differ among species. However, evidence has yet to be reported revealing the involvement of organelle genes in the initial stages of plant speciation. To elucidate the molecular basis of aquatic plant speciation, we focused on the unique plant species Chara braunii C. C. Gmel. that inhabits both shallow and deep freshwater habitats and exhibits habitat‐based dimorphism of chloroplast DNA (cpDNA). Here, we examined the “shallow” and “deep” subpopulations of C. braunii using two nuclear DNA (nDNA) markers and cpDNA. Genetic differentiation between the two subpopulations was measured in both nDNA and cpDNA regions, although phylogenetic analyses suggested nuclear gene flow between subpopulations. Neutrality tests based on Tajima’s D demonstrated diversifying selection acting on organelle DNA regions. Furthermore, both “shallow” and “deep” haplotypes of cpDNA detected in cultures originating from bottom soils of three deep environments suggested that migration of oospores (dormant zygotes) between the two habitats occurs irrespective of the complete habitat‐based dimorphism of cpDNA from field‐collected vegetative thalli. Therefore, the two subpopulations are highly selected by their different aquatic habitats and show prezygotic isolation, which represents an initial process of speciation affected by ecologically based divergent selection of organelle genes.  相似文献

Complex and defined media have been previously proposed for production of oospores of Pythium oligandrum , a fungal mycoparasite of several disease-causing fungi. However, oospore production in synthetic media requires long periods of incubation and yields lower oospore numbers than in complex media. Moreover, although complex media produce high oospore yields, these yields are not reproducible because of the variability in the composition of complex nutrient sources. In the present study, the average composition of molasses reported in the literature was utilized as a base to develop a new defined medium for P. oligandrum oospore production. Forty-two substrates defined in nine stock solutions: carbohydrates, vitamins, sterols, non nitrogenous acids, amino acids, minerals, nucleic acid bases, CaCl 2 and MgSO 4 were tested at two levels (present or absent) in two fractional factorial designs. Each of these nine variables had a significant main effect on oospore production. Furthermore, the effect of each variable, except for vitamins, depended on the level of each other variable, expressed by two-variable interactions. The maximal predicted oospore production was calculated from the polynomial regressions associated with the two fractional factorial designs. Oospore productions were 1.3 and 2 ×10 6 oospores mL -1 from the first and second designs. In order to optimize the oospore production, the two levels of each variable were modified and all variables were experimented in a selection of a complete factorial design (Plackett and Burman design). A fitted first-order polynomial regression equation provided the combination of levels of variables for optimal oospore production. A defined medium, based on this combination of levels, was used for P. oligandrum growth. The optimized oospore production after 7 days growth was 4 ×106 oospores mL -1 as predicted by the polynomial regression. Oospore yields, biomass produced from oospores and oospore freeze-drying tolerance were similar when P. oligandrum was previously grown in molasses or in the new defined medium.  相似文献
