The yield and quality of leafy vegetables can be compromised by reduced water availability. Glutamic acid is involved in different biological processes and among them it plays an important role in chlorophyll and proline biosynthesis. The aim of this work was to evaluate the possible efficacy of glutamic acid in counteracting water stress in romaine lettuce. Lettuce plants were grown in pots filled with substrate and subjected to water deprivation. A glutamic acid solution (1.9 mM) was applied as foliar treatment, both in stressed and non-stressed plants. The effect of the treatment was evaluated at different time points during the experiment in order to evaluate changes at a molecular, physiological, biochemical and agronomic level. Yield was reduced by 35% in stressed plants, while no significant changes in quality parameters were observed, except for nitrate content, which increased under water stress. At a molecular level, the expression of genes encoding for ROS scavenging enzymes was monitored but, apparently, glutamic acid did not significantly prevent the water stress response. Slightly positive effects deriving from glutamic acid application were found for nitrate and proline contents, suggesting that a possible mode of action of glutamic acid would involve a role for these molecules. Further studies are required, also on other crop species, for confirming these results. Different concentrations and application modes should be also tested.  相似文献

Selenium is considered an essential element for animals. Despite that it has not been demonstrated to be essential for higher plants, it has been attributed with a protective role against reactive oxygen species in plants subjected to stress. In this study, lettuce plants ( Lactuca sativa cv. Philipus) received different application rates (5, 10, 20, 40, 60, 80 and 120 μM) of selenite or selenate, with the aim of testing the effect of Se on the production and detoxification of H2O2 in non-stressed plants. The results indicate that the form selenate is less toxic than selenite; that is, the plants tolerated and responded positively to this element, and even increasing in growth up to a rate of 40 μM for the form selenate. On the contrary, the application of selenite triggered a higher foliar concentration of H2O2 and a higher induction of lipid peroxidation [malondialdehyde content and lipoxygenase activity] in comparison to that observed after the selenate application. Also, the plants treated with selenate induced higher increases in enzymes that detoxify H2O2, especially ascorbate peroxidase and glutathione (GSH) peroxidase, as well as an increase in the foliar concentration of antioxidant compounds such as ascorbate and GSH. These data indicate that an application of selenate at low rates can be used to prevent the induction in plants of the antioxidant system, thereby improving stress resistance.  相似文献

Gaseous hydrogen cyanide stimulated subsequent lettuce seed ( Lactuca sativa L. cv. Grand Rapids) germination in darkness when applied for 1 or 22 h. Optimum concentrations were 5 × 10-5 M and 10-6 M, respectively. However, seeds did not germinate in the presence of HCN even at 10-8 M. The effects of unsaturating red light (R) and HCN (1 μM) showed a slight synergism. On the other hand, there was no difference between the effects of the sequences HCN – R and R – HCN. Stimulation of lettuce germination by an HCN pulse was practically not affected by far-red illumination, independently of the sequence of treatments. It was concluded that the primary stimulatory effect of HCN is of a regulatory character. Cyanide controls a regulatory point different from that affected by activated phytochrome.  相似文献

Summary Amino acids antioxidant capacity has been investigated and compared with the chain-breaking antioxidant activity of known compounds as ascorbic acid and Trolox. Basic and acidic amino acids and most of neutral ones showed no antioxidant capacity. On the contrary, tryptophan, tyrosine, cysteine and homocysteine showed antioxidant ability at concentrations which are within the usually reported physiological ranges.These findings are discussed in connection with the antioxidant capacity ascribed to plasma proteins, as human serum albumin.  相似文献

Ascorbic acid (AsA) also known as vitamin C is considered as an essential micronutrient in the diet of humans. The human body is unable to synthesize AsA, thus solely dependent on exogenous sources to accomplish the nutritional requirement. AsA plays a crucial role in different physiological aspects of human health like bone formation, iron absorption, maintenance and development of connective tissues, conversion of cholesterol to bile acid and production of serotonin. It carries antioxidant properties and is involved in curing various clinical disorders such as scurvy, viral infection, neurodegenerative diseases, cardiovascular diseases, anemia, and diabetes. It also plays a significant role in COVID-19 prevention and recovery by improving the oxygen index and enhancing the production of natural killer cells and T-lymphocytes. In plants, AsA plays important role in floral induction, seed germination, senescence, ROS regulation and photosynthesis. AsA is an essential counterpart of the antioxidant system and helps to defend the plants against abiotic and biotic stresses. Surprisingly, the deficiencies of AsA are spreading in both developed and developing countries. The amount of AsA in the major food crops such as wheat, rice, maize, and other raw natural plant foods is inadequate to fulfill its dietary requirements. Hence, the biofortification of AsA in staple crops would be feasible and cost-effective means of delivering AsA to populations that may have limited access to diverse diets and other interventions. In this review, we endeavor to provide information on the role of AsA in plants and human health, and also perused various biotechnological and agronomical approaches for elevating AsA content in food crops.  相似文献

In vivo total antioxidant capacity: comparison of different analytical methods   总被引:12,自引:0,他引:12  
Several methods have been developed to measure the total antioxidant capacity of a biological sample. The use of peroxyl or hydroxyl radicals as pro-oxidants in the oxygen radical absorbance capacity (ORAC) assay makes it different and unique from the assays that involve oxidants that are not necessarily pro-oxidants. An improvement in quantitation is achieved in the ORAC assay by taking the reaction between substrate and free radicals to completion and using an area-under-curve technique for quantitation compared to the assays that measure a lag phase. The interpretation of the changes in plasma or serum antioxidant capacity becomes complicated by the different methods used in detecting these changes. The interpretation also depends upon the conditions under which the antioxidant capacity is determined because the measurement reflects outcomes in a dynamic system. An increased antioxidant capacity in plasma or serum may not necessarily be a desirable condition if it reflects a response to increased oxidative stress. Similarly, a decrease in plasma or serum antioxidant capacity may not necessarily be an undesirable condition if the measurement reflects decreased production of reactive species. Because of these complications, no single measurement of antioxidant status is going to be sufficient, but a "battery" of measurements, many of which will be described in Forum articles, will be necessary to adequately assess oxidative stress in biological systems.  相似文献

Growth substances, α-naphthaleneacetic (NAA) and kinetin, had an important role in the regulation of lateral root (LR) formation in lettuce ( Lactuca sativa L. cv. Grand Rapids) seedling roots. NAA (10-5 M ) was a potent stimulator of LR initiation and caused a 600% increase in the number of lateral root primordia (LRP) compared to untreated roots. NAA was required for only the first 20 h of the 72 h treatment period for maximum stimulation of LRP initiation. Kinetin (2 × 10-5 M ) effectively prevented the spontaneous formation of LRP and inhibited the NAA-stimulated production of LRP. Kinetin inhibition was maximal during the first 20 h of NAA treatment and this effect was not overcome by subsequent supply of NAA. Also, lettuce roots were most sensitive to kinetin at 20 h of NAA treatment, when the first signs of cell division were observed in the pericycle.  相似文献

BackgroundKIO3 and KI are the most common salt iodization agents. Coincidentally, iodine exists naturally in high-iodine drinking water in the form of iodide (I) or iodate (IO3). As an oxidizing substance, IO3 should be reduced to I before it can be effectively used by the thyroid. However, there is a lack of systematic studies on the metabolic process of high dose KIO3 in vivo.MethodsThe iodine metabolism processes in the thyroid and serum of rats after high KIO3 intake were determined using high-performance liquid chromatography-inductively coupled plasma-mass spectrometry (HPLC/ICP–MS) and arsenic cerium catalytic spectrophotometry. The changes of redox activity in the serum, thyroid, liver, and kidneys were observed by detecting total antioxidative activity (TAA).ResultsHigh doses of IO3 were completely reduced to I in vivo within 0.5 h. The level of organic bound iodine in the serum was stable, while the organic bound iodine in the thyroid increased to a plateau after intake of high-dose KIO3. The levels of total iodine and I in serum and thyroid increased quickly, then all decreased after reaching the maximum absorption peak, and I had two absorption peaks in serum. The thyroid blocking dose of I was 0.5 mg/kg in rat. Additionally, high KIO3 intake did not influence the TAA in serum and other tissues.ConclusionThe body is able to reduce and utilize high doses of KIO3 ingested through the digestive tract. The metabolism of high KIO3 in vivo is characterized by two absorption process of I in serum and the thyroid blocking effect. Moreover, a single intake of high-dose KIO3 does not affect TAA in vivo. The results suggest that such excess IO3 may have be reduced in the digestive tract before I enters the blood.  相似文献

Peroxynitrite and hypochlorite are oxidants relevant in many pathological situations. We propose a simple spectrophotometric assay to determine antioxidant capacity against hypochlorite and peroxynitrite based on protection against Pyrogallol Red decolorization. The assay can be performed on a microplate and requires minute amounts of material. Standard antioxidants show different reactivities for both oxidants. Antioxidant capacity of blood plasma (anticoagulated with EDTA) of healthy persons was found to be 559 ± 49 μmol/l and 11.6 ± 1.2 mmol/l of ascorbic acid equivalents for peroxynitrite and hypochlorite, respectively.  相似文献

 The influence of culture chamber capacity, medium volume and culture density on the growth yields of lettuce (Lactuca sativa L.) and spearmint (Mentha spicata L.) shoots were determined in an environment containing either 350 or 10,000 μmol mol–1 CO2 after 8 weeks of incubation. High positive correlations occurred between the culture vessel capacity and spearmint fresh weight, leaf number, root number, and shoot number. Similarly, high positive correlations occurred between culture vessel capacity and lettuce fresh weight, leaf number, and root number. Higher fresh weights, leaf numbers, and root numbers were obtained from lettuce and spearmint shoots when cultured in 1-quart Mason jars containing 100- or 150-ml aliquots of medium compared to jars containing 25- or 50-ml aliquots of medium within an environment containing either 350 or 10,000 μmol mol–1 CO2. High culture density decreased growth yields, and this phenomenon could only be slightly off-set by the employment of an elevated CO2 environment or larger culture vessels. A new disease of lettuce has been observed in the surroundings of Bahía Blanca, Argentina. The symptoms include dwarfing, general chlorosis, wilting, root rot, and leaf blight, sometimes followed by plant death. Pythium tracheiphilum Matta was found to be consistently associated with those symptoms. The morphological and morphometric characteristics of this oomycete are described. Isolates obtained from field-infected lettuce plants were inoculated to lettuce and caused similar symptoms as those found in natural infections. Seedling emergence was also severely affected following experimental inoculation. Koch’s Postulates were fulfilled by recovering the fungus from inoculated plants. The new disease shows a high destructive potential but currently has a limited prevalence and incidence.  相似文献

Abstract

The ‘Total antioxidant capacity’ (TAC) is a parameter frequently used for characterization of food products and of the antioxidant status of the body. This mini-review shows shortcomings of TAC assays and points of concern that should be considered when performing and interpreting results of such assays. The term TAC is not optimal since the assay measures only part of antioxidant capacity, usually excluding enzymatic activities. Antioxidant and oxidant-regenerating enzymes in blood cells and the blood vessel wall have a profound impact on the antioxidant properties of blood plasma, which is not reflected in the in vitro assays of isolated plasma. The term ‘Non-enzymatic antioxidant capacity’ (NEAC) is suggested as more relevant than TAC. NEAC is estimated by various methods, which yield different values and results obtained using different methods do not always show satisfactory correlation. One reason for the discrepancy of results is the use of different oxidants in NEAC assays. The use of hydroxyl radical as the oxidant is not recommended in view of the high and non-specific reactivity of this species.  相似文献

Lettuce growth under unstressed conditions was compared to growth under four limiting conditions, i.e. no phosphorus fertilization (0_P), no nitrogen fertilization (0_N), low light (LR) and water stress (WR) over two different growing periods. We investigated the adaptive changes in terms of the morphological and physiological leaf traits, identifying stress‐specific and ‘stable’ indicators suitable for use in breeding programmes. The plants subjected to the WR treatments had lower leaf expansion and specific leaf area (SLA), as well as lower soil–plant analysis development (SPAD) values, stomatal conductance (POR), water index (WI) and leaf temperature (TIR) compared with plants in the unstressed CONTROL. Low light increased the leaf area (LA), SLA and leaf mass ratio (LMR). The 0_N treatment induced a general reduction in the normalised difference vegetation index (NDVI) values, as well as strong changes in LMR and SLA. In general, 0_P induced less pronounced effects than the other treatments. Principal component analysis indicated that the stable and suitable selection indicators of adaptive changes for low nitrogen and low light conditions were LA, SLA, leaf area per unit total plant mass (LAR), LMR, SPAD and POR, while SPAD, POR, TIR and WI were suitable indicators for drought.  相似文献

The aim of this work was to compare the antimicrobial activity against Paenibacillus larvae and the antioxidant capacity of two Laurus nobilis L. extracts obtained by different extraction methods. The hydroalcoholic extract was moreover added as supplementary diet to bees in field conditions to test behavioural effects and colony strength. Both laurel extracts were subjected to different phytochemical analysis to identify their bioactive compounds. Antimicrobial activity was analyzed by the minimal inhibitory concentration (MIC) determination by means the agar dilution method. The hydroalcoholic extract (HE) was able to inhibit the bacterial growth of all P. larvae strains, with 580 µg/mL mean value. This better antibacterial activity in relation to the essential oil (EO) could be explained by the presence of some phenolic compounds, such as flavonoids, evidenced by characteristic bands resulting from the Fourier Transform Infrared Spectroscopy (FTIR) analysis. Antioxidant activities of the extracts were evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging ability and ferric reducing antioxidant power (FRAP) assays. The HE showed the highest antioxidant activity as measured by DPPH, with IC50 values of 257 ± 12 μg/mL. The FRAP assay method showed that the HE was 3-fold more effective reducing agent than the EO. When the bee colonies were supplied with laurel HE in sugar paste an improvement in their general condition was noticed, although neither the hygienic behavior nor the proportions of the breeding cells varied statistically due to the treatment. In conclusion, the inhibition power against P. larvae attributable to the phenolic compounds, the antioxidant capacity of the HE, and the non-lethal effects on adult honey bees on field trials suggest the HE of laurel as a promising substance for control American foulbrood disease.  相似文献

Nasturtium officinale R. Br. (Brassicaceae) has been used as a home remedy by the people of south eastern (SE) region of Iran as a medicinal plant. This therapeutical application has been attributed to Nasturtium officinale (N. officinale) antioxidant capacity which is mostly tested by means of cell-free assays: 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP). In addition, the antioxidant effect of N. officinale extract has been investigated in hypercholesterolaemic rats in vivo. The results revealed that the extract has notable scavenging activity against DPPH radicals as well as potent reducing power in FRAP assay. Intragastric administration of N. officinale (500 mg/kg body weight per day) to groups of hypercholesterolaemic rats for 30 days lowered their blood total cholesterol (TC), triglyceride (TG), and low density lipoprotein cholesterol (LDL-C) levels by 37, 44 and 48%, respectively. However, the blood high density lipoprotein cholesterol (HDL-C) levels in the same treated rats increased by 16%. To evaluate the mechanism(s) of action, we studied the antioxidative potential of N. officinale extract in terms of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione reductase (GR) activities and also the level of reduced glutathione (GSH) in the liver tissues. In addition, hepatic tissue malondialdehyde level (MDA, an index of lipid peroxidation) was also determined. Under hypercholesterolaemic condition, hepatic MDA was increased. Moreover, our data indicated GSH depletion along with significant reduction in the activities of CAT and SOD in rats fed high-fat diet rats. On the other hand, significant elevation in the activities of GPx and GR were seen in the same group of rats. Treatment of hypercholesterolaemic rats with N. officinale extract significantly increased the GSH level along with enhanced CAT and SOD activities in liver tissues. Furthermore, N. officinale extract significantly decreased hepatic MDA as well as GPx and GR activities in plant-treated rats. Based on our data, it can be concluded that N. officinale has a high hypolipidaemic activity and this may be attributed to its antioxidative potential.  相似文献

The genetics of partial resistance (PR) of lettuce to the leaf aphid species Macrosiphum euphorbiae and Uroleucon sonchi was studied in F1 and F2 generations of six crosses between four cultivars. Partial resistance inherited mainly additively. With M. euphorbiae the estimates of broad-sense F2 heritability were low and non-significant in crosses between parents of the same resistance level (S x S and PR x PR), and high (0.69-0.76) in crosses between parents of different resistance level (PR x S). The level of resistance in lettuce cultivars ‘Charan’ and ‘Marbello’ led to a marked reduction in percentage of plants with colonies of M. euphorbiae in a field experiment. With U. sonchi the highest estimates of F2 heritability were found in ‘Charan x ‘Avoncrisp’ (0.51) and ‘Charan’ x ‘Chou de Naples' (0.48). In the cross ‘Charan’ x ‘Marbello’ (PR x PR) no significant segregation for resistance to M. euphorbiae was found, indicating that these cultivars may have the same genetic constitution for resistance to M. euphorbiae.‘Charan’ has additional genes for resistance to U. sonchi, that are not, or only weakly linked to the gene(s) for resistance to M. euphorbiae. The heritability estimates for the partial resistance to M. euphorbiae and U. sonchi indicate that it is possible to effectively select for these traits in plant breeding programmes.  相似文献

Few methods for assessing total antioxidant capacity (TAC) include both the percentage of inhibition and the length of inhibition in the measurement. Available methods require above ambient constant temperature incubation, reaction preheating, and/or separate assays for testing hydrophilic and hydrophobic samples. We describe a high-throughput method, antioxidant inhibition of oxygen radicals (AIOR), that overcomes these difficulties. AIOR uses peroxyl radicals to trigger a decrease in fluorescence of the indicator molecule, uroporphyrin I, which is delayed by the presence of antioxidants. The area under the curve is measured by a fluorescence spectrophotometer in a 96-well microplate format, and TAC results are expressed as millimole/liter Trolox equivalents. AIOR is performed at ambient temperature and is applicable to samples in either aqueous or common organic solvents. The reaction between uroporphyrin I and the peroxyl radicals generated from 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) was found to be of first-order kinetics with a mean rate constant (k) of 0.0254. Applications to measure antioxidant capacity are demonstrated on individual chemicals and biological samples. The method has good linearity, within- and between-assay precision, and recovery.  相似文献
