We observed sexual reproduction in a clonal culture of Stephanodiscus niagarae Ehrenb. and used light and scanning electron microscopy to absent flagellated male cells, auxospore growth, initial valve structure and production, and subsequent daughter cell division. Free auxospores were spherical and nonsiliceous throughout growth, producing hemispherical initial valves devoid of spines and with nonfasciculate striae. Pregametangial cells averaged 43% of the diameter of the daughter cell population and were 1/9 the biovolume of initial, cells. This paper is the first confirmed report of sexual reproduction in S. niagarae, although it appears that specimens of Actinocyclus niagarae H. L. Smith, described from Lake Erie in 1878, are actually initial valves of S. niagarae.  相似文献

Ice diatom assemblages in the Canadian Arctic are dominated by the Naviculaceae, a family which includes the genus Stenoneis Cleve. A taxonomic history of S. inconspicua var. baculus (Cleve) Cleve (including type material of Navicula baculus Cleve) is presented with morphological data including the first SEM views of the genus. Stenoneis inconspicua var. baculus was compared with S. obtuserostrata (Hustedt) comb. nov. (basionym: Navicula obtuserostrata Hustedt). In the SEM, external, short, flattened siliceous ridges were revealed beside the proximal parts of the raphe branches and central pores. This may be a significant taxonomic feature of the genus. Valves of S. inconspicua var. baculus ranged from 40–103 μm in length, 7–11 μm in width and had 17–20 striae in 10 μm, whereas valves of S. obtuserostrata were shorter (27–53 μm), narrower (6–7.5 μm) and more finely striated (24–30 in 10 μm). Sequence variation of ribosomal DNA internal transcribed spacers (ITS) among populations, species, and genera of the diatom genus Stephanodiscus was investigated. ITS 1 and ITS 2, including the 5.8S gene, were sequenced from geographically distant and nearby populations of S. niagarae Ehrenberg. In addition, repeats from S. hantzschii Grunow and Cyclotella meneghiniana Kützing were sequenced to determine the taxonomic range over which the ITS region could be used for diatom systematics. The morphologically distinct S. yellowstonensis Theriot & Stoermer, thought to have evolved from S. niagarae in Yellowstone Lake between 12,000 and 8000 years ago, also was sequenced to assess its relationship to nearby S. niagarae populations. The organization and relative sizes of ITS 1 and ITS 2 in Stephanodiscus species were similar to those reported for other eukaryotes. In general, ITS 2 was slightly larger and more variable than ITS 1. Cladistic analysis of ITS sequences did not resolve relationships of nearby S. niagarae and S. yellowstonensis populations. However, central North American S. niagarae populations were in a clade supported by two nucleotide changes. For Cyclotella, much of the ITS region was not alignable with that for Stephanodiscus species; therefore, generic-level comparison within the Thalassiosiraceae may not be possible. The variation (95–96% similarity) between S. hantzschii and other Stephanodiscus species suggests that interspecific relationships could be assessed with ITS sequences. Although S. yellowstonensis is morphologically distinct from S. niagarae, no autapomorphic nucleotide sites were identified. Two S. niagarae populations (Heart and Lewis Lakes), however, did possess autapomorphic ITS sites.  相似文献

中国淡水硅藻新记录——偏肿内丝藻兴凯变种   总被引:1,自引:0,他引:1  
于2016年5月对浙江省仙居国家公园进行藻类多样性调查时,发现了中国淡水硅藻的1个新记录种——偏肿内丝藻兴凯变种[Encyonema ventricosum var.hankensis(Skvortzov) RodionovaPomazkina]。该文采用光镜和扫描电镜对偏肿内丝藻兴凯变种浙江种群的形态特征进行了观察和描述,并将其与该种的其他地理种群及内丝藻属其他相似物种的形态特征进行比较,同时讨论了该物种的生境特征。结果表明:(1)偏肿内丝藻兴凯变种壳面均具明显背腹之分,背缘明显拱起,腹缘轻微波曲;端部呈头状至喙状。(2)偏肿内丝藻兴凯变种浙江地理种群在背部中央区域具一孤点,而其兴凯种群对孤点这一特征未有描述。(3)偏肿内丝藻兴凯变种在俄罗斯的兴凯湖和贝加尔湖等地有分布,该种在中国是首次报道。  相似文献

Ten clonal isolates of Thalassiosira tumida (Janisch) Hasle were grown in duplicate semi-continuous batch cultures at 116 and 11.6 μE·m−2·s−1 acclimated cells were harvested during exponential growth and cleaned for examination by light microscopy (LM) and scanning electron microscopy (SEM). Number of strutted processes surrounding the central annulus (SP) and average number of satellite pores per process (AVSAT) were counted using SEM on 20 valves from each culture grown in high light, for a total of 400 valves examined; number of marginal labiate processes (LP) and overall diameter (DIAM) were measured using LM on 20 valves from each culture grown in both high and low light for a total of 800 valves examined. Univariate analysis of variance showed that bottle effects resulting from microenvironmental differences between replicates were a small but significant source of variation in DIAM, LP, and SP but not AVSAT. Significant differences among clones were observed for all characters. Decreased irradiance resulted in a significant decrease in valve diameter but no significant effect on LP; no light × clone interaction was observed. Significant covariance between characters among clones was also observed; since valve diameter is known to decrease during asexual growth, the correlation coefficients for SP, AVSAT, and LP with DIAM were used to correct the data for this source of nongenetic differences between clones. Analysis of the size-corrected data showed that the proportion of total phenotypic variance in SP, LP, and AVSAT caused by genetic differences among clones was 0.14, 0.14, and 0.30, respectively. This indicates that the majority of total phenotypic variance was due to environmental or developmental causes, but that sufficient genetic variability exists to support rapid phenotypic evolution in SP, LP, and AVSAT under continued directional selection. Finally, the results of the genetic analysis revealed a high (0.82) genetic correlation between SP and LP.  相似文献

Teratological forms of Stephanodiscus niagarae Ehrenb. and S. parvus Stoermer & Håkansson were observed during a study of diatoms preserved in a radiometrically dated core from Hamilton Harbour (Lake Ontario, Canada). Morphological features and ultrastructures of both species were studied under the light and scanning electron microscope. The valve structure of abnormal forms of S. niagarae appears to be weakly silicified, especially in the central area. The shape of satellite pores are very irregular in comparison with the round shape of the normal specimen. The central fultoportulae are characterized as small tubes extending out of the external valve. Two types of abnormal frustules are present in the population of S. parvus studied. In one type valves are underdeveloped and only the siliceous layer and ribs are present, and in the other type the valves are strongly silicified and the areolae are almost completely occluded.The teratological forms of both species appeared in the core sediments after 1911, and both became the dominant components of diatom assemblages after 1970. Their occurrence and increased abundance coincides with records of heavy metal pollution in the harbour.  相似文献

The eutrophic, freshwater diatom species Stephanodiscus binderanus (Kütz.) Willi Krieg. has long been considered a nuisance exotic alga introduced from Eurasia to the Great Lakes in North America in the early to mid‐20th century. However, our paleolimnological data from Lake Simcoe, Ontario, provide unequivocal evidence that this taxon has been present in the Great Lakes region since at least the late 17th century. Subfossil diatom valves were identified and enumerated at high resolution in 210Pb‐dated sediment cores from four sites across the lake. The taxonomic identification of S. binderanus was confirmed using SEM. The historical presence of this species in Lake Simcoe indicates somewhat naturally productive conditions and also refutes the idea that S. binderanus is a nonindigenous species to North America. This study underscores the caution that should be applied to questions of diatom (and protistan) distributions in time and space. Clearly, the absence of evidence is not evidence of absence.  相似文献

The possible effect of different salinities on the valve morphology of the marine centric diatom Thalassiosira decipiens was investigated. Cultures were grown in media of salinities ranging from 2 to 35%. for 40 days. Only the 15, 20, 25, and 30%. cultures had sufficient growth for statistical analysis of population differences. Areola shape, pattern, and density changed significantly with decreasing salinity. Valves with areola patterns skewed off-centre were more frequent in the lower salinities. Valve diameter also showed a significant decrease. Valves having reduced or missing fultoportulae and rimoportulae were common in the 15 and 20%. cultures.  相似文献

Three fragilarioid diatom taxa were studied in detail at the light microscopy and scanning electron microscopy levels from samples collected from Lake Hovsgol, Mongolia. Two of the taxa are new to science, described here as Staurosirella minuta Morales et M. B. Edlund and Pseudostaurosira tenuis Morales et M. B. Edlund, and may be endemic to Lake Hovsgol. The third taxon has been identified as Fragilaria polonica Witak et Lange‐Bertalot and it is transferred to the genus Pseudostaurosira (Grunow) D. M. Williams et Round as Pseudostaurosira polonica (Witak et Lange‐Bertalot) Morales et M. B. Edlund comb. nov. based on the ultrastructural features of its valves. The relationship of the above taxa to others reported in the literature is included herein, and the nomenclatural transfer, Pseudostaurosira naveana (Le Cohu) Morales et M. B. We discovered two new taxa of Achnanthidium and Encyonema occurring with high abundance in the upstream of the Yom River, which is one of four major rivers in Northern Thailand. Achnanthidium pseudoconspicuum var. yomensis var. nov. closely resembles Achnanthidium pseudoconspicuum var. pseudoconspicuum in scanning electron microscopy (SEM), but has clearly different valve morphometry under the light microscopy (LM). Achnanthidium pseudoconspicuum var. yomensis is also similar to A. japonicum in valve morphology under LM, but differs to this species based on SEM observation. Encyonema yuwadeeanum sp. nov. shows similarities to Encyonema leei and Encyonema subkukenanum, but differs in valve shape and/or polar fissure shape. We discuss the possible valve ontogenetic relationship between Achnanthidium and Cymbellales based on the detailed areolae structures of these two new taxa.  相似文献

Diatom studies carried out in Central and Eastern Poland have shown a diversified composition and considerable changes of relative frequency of dominant diatoms in profiles of lake sediments of the Mazovian and Ferdynandovian interglacials. Cyclotella spp. are dominant in a large part of the sediment profiles at Adamówka, awki-7 and Wola Grzymalina-59 and, at Krpiec and Podlodów, Stephanodiscus is also common while at Biaa Podlaska and Falcice, Aulacoseira is most abundant. Cyclotella comta var. lichvinensis and C. vorticosa are the typical diatoms in the Mazovian Interglacial. In the sediments representing the Ferdynandovian Interglacial, the characteristic diatoms are Cyclotella cf. reczickiae and the Stephanodiscus rotula/niagarae complex.  相似文献

The external morphology of female heads of three populations of each of two cytological races of Meloidogyne hapla (race A-meiotic, race B-mitotic) and single populations of M. arenaria, M. incognita, and M. javanica was compared by light (LM) and scanning electron microscopy (SEM). Perineal patterns of all nine populations were observed with a LM and then examined with a SEM. In addition, female stylets of each population were excised, viewed with a SEM, and compared with observations made with a LM. Head morphology of the females, including shape of medial and lateral lips, expression of sensilla, and head annulation, was distinct for each species, each race of M. hapla, and each population of M. hapla race A. The morphology of a given perineal pattern appeared similar with the SEM and the LM. The SEM emphasized surface details, whereas the LM revealed subcuticular structure as well. Stylet morphology was unique for each species but similar in all populations of M. hapla. There were differences between species in the shape of the cone, shaft, and knobs and in the distance of the dorsal esophageal gland orifice from the stylet knob base. Several of the morphological characters first detected in the SEM were seen subsequently with the LM and are helpful in species identification.  相似文献

Are size and arrangement of valve mantle areolae in Aulacoseira Thwaites adapted to light intensity? To test one criterion demonstrating an adaptation, heritability experiments were run on isolates of Aulacoseira subarctica (Müller) Haworth. Several clones of A. subarctica were isolated from Yellowstone Lake (Wyoming, USA), Lewis Lake (Wyoming), and East Rosebud Lake (Montana, USA). Two to four clones from each lake were grown in batch cultures under three irradiance levels: 2, 11.4, and 115 μmol photons·m?2·s?1. Five randomly chosen valves for each of two replicates of each clone were examined using a scanning electron microscope for a total of 300 valves. Size measurements were taken for each valve examined, and images of mantle areolae were captured on film at a magnification of 20,000×. Each image was digitized, and quantitative morphometric areolar characters were measured. A quantitative genetic analysis was performed within each light environment for the mean area of the external opening of mantle areolae, the mean distance between areolae within pervalvar striae, and the mean distance between pervalvar striae. Resulting estimates of heritability from among‐lake and within‐lake analyses indicate that all three mantle areolar characters could presently respond to selection and thus have potentially done so in the past.  相似文献

Three Cocconeis Ehrenberg species, epiphytic on the leaves of Posidonia oceanica (L.) Delile, were studied by light and electron microscopy. A new species, C. pseudonotata, is described which is closely related to C. notata Petit and C. diruptoides Hustedt. Gross morphology and the presence of a sigmoid raphe and sternum are the characters shared by the three species, whereas the central areas of sternum and raphe-sternum valves, and the stria and areola densities are the main distinctive characters for the species when observed by light microscopy. Scanning and transmission electron microscopic observations revealed that the fine structure of areolae must be also considered in order to identify the species. Comparison among the three species has been carried out by observations on material from our samples, as well as on the type material of C. diruptoides and cleaned samples from the Hustedt collection. The study also provides new information on the geographical distribution of these species.  相似文献

Fossil sequences provide observations of phenotypes within a lineage over time and represent essential data for increasing our understanding of phyletic evolution beyond microevolutionary timescales. I investigate if fossil time series of the diatom Stephanodiscus niagarae/yellowstonensis follow evolutionary dynamics compatible with hypotheses for how the adaptive landscape changes when a population enters a new environment. The lineage—which has a remarkably detailed stratigraphic record—invaded Yellowstone Lake immediately after recession of ice from the basin 14,000 years ago. Several phyletic models portraying different types of evolutionary dynamics—both compatible and not compatible with changes in the adaptive landscape following ecological opportunity—were fitted to the fossil times-series of S. niagarae/yellowstonensis. Different models best describe the three analyzed traits. Two of the models (a new model of decelerated evolution and an Ornstein–Uhlenbeck model) capture trait dynamics compatible with an event of ecological opportunity, whereas the third model (random walk) does not. Entering a new environment may accordingly affect trait dynamics for thousands of years, but the effects can vary across phenotypes. However, tests of model adequacy reveal shortcomings in all three models explaining the trait dynamics, suggesting model development is needed to more fully understand the phyletic evolution in S. niagarae/yellowstonensis.  相似文献

Diatom sediment records of large lakes can be used to decipher the history of ancient phytoplankton. The upper layer of the sediment is an important area of remineralization of the sedimenting phytoplankton biomass. It hosts a bacterial community different from those of both the water column and deeper sediment layers. In this work, we analyzed the structure and diversity of the communities of Bacteria and Archaea in the surface sediment core containing valves of diatoms, the major producers in Lake Baikal. Pyrosequencing of the bacterial V3–V4 region of the 16 S ribosomal RNA (rRNA) and archaeal V1–V3 16 S rRNA gene regions yielded 29,168 and 36,997 reads, respectively. In total, we have identified 33 bacterial phyla; uncultured Actinobacteria were the most abundant in the upper layers, while lower sediment was dominated by Firmicutes and Alphaproteobacteria. The composition of the archaeal community changed with depth, but was generally dominated by Crenarchaeota from the classes Marine Group I and Miscellaneous Crenarchaeotic Group, as well as Euryarchaeota from the class Thermoplasmata. These dominant bacterial and archaeal taxa are presumed to participate in the destruction of buried organic matter, which eventually leads to degradation of the diatom valves.  相似文献
