The in vitro germination of chrysanthemum pollen is promotedby adding floral organs, excepting anther, to a basal mediumconsisting of sucrose and boric acid. Some other plant organs,such as young fruit of tomato and onion bulb, are also effective.In Chrysanthemum morifolium, the percentage of pollen germinationin the presence of such plant tissues is two to three timesas high as in the control (10 %). In Ch. leucanthemum, it ishigher than 50 %, in contrast to the control in which no germinationis noted. This promotion of germination may be due to a substance extractablefrom the tissues with water, ether or methanol.   1. Some recent works on the formation of oxalic acid by variousfungi are critically considered.
  2. The present work deals withthe role of oxalic acid in the metabolismof Aspergillus niger.
  3. When glucose solutions were supplied to preformed mats ofthefungus oxalic acid accumulated, attaining an equilibriumlevelwhich was not exceeded despite the presence of a considerableconcentration of glucose.
  4. When the glucose supplies were depletedthe oxalic acid concentrationfell steeply to a low level.
  5. Theconcentration of oxalic acid was dependent on the glucoseconcentration.In three separate series of experiments it wasshown that theoxalic acid concentration diminished with increasingglucoseconcentration.
  6. Similar results were obtained when the cultureswere rearedfrom spores on culture solutions with the normalamounts ofnutrient salts but different glucose concentrations.
  7. In all cases the CO2 output increased with the glucose concentration.
  8. When cultures were supplied with glucose+oxalic acid, theconcentrationof the latter fell steeply to the equilibriumlevel attainedon glucose only. In a culture receiving glucose+oxalicacid,with the oxalic acid concentration somewhat below thenormalequilibrium concentration, the formation of oxalic acidfromthe glucose ceased as soon as the equilibrium level hadbeenattained.
  9. When 1 per cent. oxalic acid only was suppliedto the fungusthe concentration gradually diminished to a lowlevel. When3 per cent. oxalic acid was supplied the rate ofacid utilizationsoon fell to low value.
  10. In several experimentsit was shown that the rate of CO2 outputwas higher from culturessupplied with glucose+excess oxalicacid than from culturessupplied with glucose only.
  11. The rate of oxalic acid carbonloss was always below that ofthe CO2 carbon output both incultures supplied with oxalicacid only and in cultures receivingglucose+oxalic acid.
  12. The cultures were incapable of utilizingneutral sodium oxalateand the presence of this substance hadno effecft on the ofCO2 output.
  13. The results indicate thatthe utilization of oxalic acid isassociated with the liberationof at least an equivalent amountof CO2.
  14. It is suggested thatthe utilization of oxalic acid is promotedby the presence ofglucose, thus accounting for the lower oxalicacid concentrationsand higher rates of CO2 output of cultureswith higher glucoseconcentrations.

The Effects of Gibberellins on the Growth of Excised Tomato Roots   总被引:3,自引:0,他引:3  
  1. At appropriate concentrations both gibberellic acid (GA) and1-naphthalene-acetic acid (NAA) enhance the main axis growthof excised tomato roots grown in culture media containing sucroseat concentrations below 1 per cent. Lateral root extension growthis enhanced by GA at all sucrose concentrations tested; onlyat the lower sucrose concentrations is this effect observedwith NAA. Both GA and NAA increase the number of emergent lateralroots and this effect is most marked in media of low sucrosecontent. Both GA and NAA at higher concentrations inhibit rootgrowth but NAA exhibits its full range of growth effects overa much narrower concentration range than GA.
  2. GA, like NAA,speeds up the loss of meristematic activity whichoccurs whenindividual meristems are repeatedly subculturedin media containing1 per cent, or higher concentrations ofsucrose.
  3. The promotionof main axis growth by both GA and NAA involvesenhanced cellelongation and cell division. At a moderatelyinhibitory concentrationGA reduces both cell elongation andcell division; this is notthe case with NAA.
  4. Gibberellins A1, A2, and A4 resemble GA(gibberellin A3) intheir growth effects. Allogibberic acidlike G A promotes lateralroot extension growth but causes markedinhibition of root growthat a much lower concentration thanGA.

  1. The induction of an IAA-destroying enzyme in Arthrobacter sp.that can utilize IAA as its sole source of carbon and nitrogenwas investigated.
  2. 1. The enzyme was most effectively inducedby 10–3 to2x10–3 M IAA, at pH 6.5.
  3. 2. All testedIAA analogs were unable to induce the enzyme.Analogs otherthan indole-3-lactic acid were rather inhibitoryon the inductionwith IAA.
  4. 3. The induction period was shortened with the ageof culturein both polypeptone and acetate media.
  5. 4. Pretreatmentof the bacterium with IAA caused a shorteningof the inductionperiod.
  6. 5. The induction was inhibited by various antibiotics,aminoacid analogs and nucleobase analogs.

  1. An apparatus is described by means of which the absorptionofions from a complete nutrient solution of constant compositionby excised root systems of plants, grown under known nutrientdeficiencies, may be measured in standard conditions of aerationand temperature. Results of some prelimi nary experiments aredescribed.
  2. It was found that the roots readily absorbed theelement inwhich they were deficient, but tended to lose thoseelementswhich were already present in normal amounts.
  3. Therewas almost invariably a loss in fresh weight of the rootsafterthe absorption period and also a loss in dry weight. Thislossappears to be complex and is partly attributable to lossofrespiratory material.
  4. The addition of 2 per cent. sucroseto the solution from whichthe root systems of phosphorus-deficientbarley plants wereabsorbing increased the nitrogen and phosphoruscontents ofthe roots and maintained the potassium content,while in absenceof sucrose only the phosphorus content increased,but this increasewas significantly less than in the presenceof sucrose.
  5. It was shown that roots excised from plants growingin soilwere capable of absorbing phosphorus or nitrogen—elementsin which they were apparently deficient.
  6. The interpretationof data obtained from excised roots is discussed,and it isconcluded that excised roots from plants grown incomplete nutrientare not likely to behave in the same way,as regards absorption,as corresponding roots of intact plants,but that roots grownunder conditions of deficiency will behaverather similarlywhether excised or intact. This fact providesa potential methodfor diagnosing and evaluating nutrient deficiencies.
  7. The low-saltcondition of roots postulated by Hoagland and Broyeris notnecessarily the primary requisite for rapid absorptionof aparticular ion. It is rather that the roots should be deficientin that ion. The roots could be high in other salts.

Summary Water-homogenized stigma pellets of pearl millet and precipitates resulting from dialysis of their salt extracts were observed to: (1) chemotropically attract pearl millet pollen tubes on a sucrose-containing pollen germination and growth medium, (2) have acid invertase activity as assayed by the arsenomolybdate method, (3) hydrolyze sucrose in the pollen germination and growth medium to glucose as assayed by coupled glucose oxidation with Nitro Blue Tetrazolium, and (4) lose chemotropic and invertase activities upon heat treatment. The results indicate that the in vitro chemotropic attraction of pearl millet pollen tubes to water-homogenized stigma pellets is a response to glucose produced by homogenate-pellet-bound invertase hydrolyzing the sucrose present in the pollen germination and growth medium. Yeast and tomato invertases used as controls verified this conclusion. Water extracts of whole stigmas contained water-soluble acid invertase. The effect of auxin on the molecular weight (Mw) distributionof cell wall xyloglucans was investigated by gel permeationchromatography using coleoptile segments of Avena sativa L.cv. Victory, and the following results were obtained.
  1. The water-insoluble hemicellulose (HC-A) mainly consisted ofxyloglucans. Iodine staining method revealed that relativelylarge amounts of xyloglucans were present in the water-solublehemicellulose (HC-B) and water-soluble polysaccharide (WS) fractions.
  2. IAA did not cause remarkable changes in xyloglucan contentsin the hemicellulose, but significantly increased the xyloglucancontent in the WS fraction.
  3. IAA substantially decreased theweight-average Mw of HC-A. Thiseffect became apparent within30 min of the incubation period,and was not affected by the0.15 M mannitol or 2% sucrose applied.Hydrogen ions also causeda decrease in the weight-average Mwof HC-A; its effect beingreversible.
  4. Neither IAA nor hydrogen ions caused any remarkablechangesin the weightaverage Mw of water-soluble xyloglucansin theHC-B.

(南京农业大学园艺学院,南京210095)摘要:研究培养基成分、pH值和培养方式对梅花花粉离体萌发和花粉管生长的影响。结果表明:不同品种梅花花粉离体萌发的最适培养基为ME3+200g.L-1PEG4000(pH5.0),品种‘淡丰后’、‘久观绿萼’、‘喧妍宫粉’和‘月光玉蝶’最高萌发率可分别达到58.6%、60.6%、85.6%和50.7%。PEG4000能显著促进梅花花粉萌发,在培养基各成分中作用最大,不可替代。低浓度(50g.L-1)蔗糖对梅花品种花粉萌发作用不显著,而高浓度(≥100g.L-1)蔗糖明显抑制花粉萌发和花粉管生长。固体和液体培养对梅花花粉离体萌发的影响差异不显著。  相似文献

Summary The hydrodynamics of mature pollen rehydration in Nicotiana tabacum was used to study reversible inhibition of pollen germination in vitro. Tobacco pollen was incubated for various times in media containing calcium, potassium and magnesium salts, boric acid, and exhibiting different osmotic pressures as a function of sucrose concentration. Total inhibition of germination with complete viability preservation was achieved for 56 h by keeping the grains in medium with 80% sucrose, since typical percentages of germination and pollen tube lengths were recovered after this treatment. These effects were considered as consequences of natural osmoregulation of rehydration/germination in mature pollen. The possibility of applying these findings to the incubation of pollen with Agrobacterium tumefaciens to develop a pollen transformation method is discussed.  相似文献

In the fern Pteris vittata, low-energy blue-light-inducedinhibition of phytochrome-dependent spore germination and darkrecovery from this inhibition were repeatedly observed severaltimes at intervals of 3 days at 26. The same amount of incidentenergy of blue light was required for inhibition in each successivetreatment.
  2. 2. The recovery from blue-light-induced inhibitionof germinationwas markedly accelerated by continuous illuminationwith redlight, and this red light effect was not affected bythe presenceof CMU.
  3. 3. The recovery process was not influencedby a single exposureto redlight, but was definitely promotedby brief red irradiationsgiven intermittently, at least 2 times,at equal intervals duringthe first 8 hr after blue light treatment.The effect of intermittentlygiven red light was annulled wheneach red exposure was followedby brief far-red irradiation.These facts suggest that phytochromemay be involved in therestoration of the ability of sporesto germinate (in responseto red light) which had been lostby blue irradiation.

The exine surrounding the colpal furrows in the pollen grainsof Linum grandiflorum is rapidly degraded following the twocompatible pollinations between ‘pin’ and ‘thrum’types of flower and also following the incompatible pollinationbetween ‘thrum’ and ‘thrum’. In allthese pollinations the pollen germinates normally. Exine isnot degraded in the incompatible pollination between ‘pin’and ‘pin’. In this pollination the pollen does notswell or germinate. The exine degradation is restricted to thecolpal regions. Hydration of the pollen grain by buffer solutionwithout contact with the stigma also stimulates the degradationof the exine. The erosion of the exine reveals a fibrous, PAS-positivematrix which follows the original contours of the exine. Vesicles are discharged into the intinc following pollination;these appear to contribute their contents to the intine-likewall which eventually invests the pollen tube. The observations are interpreted by assuming:
  1. 1. an enzyme situatedwithin the pollen and localized underthe colpi; this enzymeis active after hydration;
  2. 2. the degradation is a means bywhich colpal furrows are enlargedto permit the emergence ofthe pollen tube;
  3. 3. the degradation process both by bufferand on the stigmais purely the consequence of hydration andis only secondarilyrelated to incompatibility through the ‘pin’by‘pin’ pollination not permitting hydration ofthepollen.

麻疯树因其种子含油率较高,种子油提炼的生物柴油可部分替代汽油,而成为一种极具潜能的能源作物,但由于产量低,麻疯树在热带、亚热带的发展受到极大限制。杂交育种是提高产量的重要手段,杂交亲本花粉生活力的高低直接影响到育种的成效。因此,寻求麻疯树离体花粉萌发的最适培养基配方,探明花粉萌发培养基中各主要培养基成分间的交互作用对生产上麻疯树杂交结实率和种子产量的提高具有重要意义。该研究以麻疯树开花初期雄花上花药刚散粉时的成熟花粉粒为材料,采用Box-Behnken设计(Box-Behnken design,BBD)的响应面法,对麻疯树花粉离体萌发培养基中各主要培养基成分的浓度配比及各主要培养基成分的交互作用进行了研究。以花粉萌发率为响应指标,建立了4种营养成分(蔗糖、硼酸、硝酸钙、硝酸钾)与花粉萌发率的响应面模型,并对各主要培养基成分的浓度配比进行了优化。通过R软件进行响应面分析的结果表明:4因素对花粉萌发率的影响顺序为蔗糖硼酸硝酸钙硝酸钾;蔗糖与硼酸、蔗糖与硝酸钙、蔗糖与硝酸钾之间的交互作用显著。响应面建模优化后的最佳培养基为13.77%蔗糖+32.14 mg·L~(-1)硼酸+22.21 mg·L~(-1)硝酸钙+19.95 mg·L~(-1)硝酸钾+200 mg·L~(-1)硫酸镁,在此条件下的理论萌发率为99.73%。采用此培养基成分配比得到麻疯树花粉离体试验萌发率为98.97%,与理论响应值相吻合,同时也表明利用BBD设计的响应面模型进行麻疯树花粉离体萌发培养条件优化方法的有效性。  相似文献

山玉兰(Magnolia delavayi)是木兰科木兰属的常绿乔木或大型灌木,是重要的园林造景、庭院绿化素材,也是重要的育种资源。山玉兰花粉的研究对其杂交育种的成败具有重要影响,但目前尚未见其花粉活力的相关报道。该研究以新鲜的山玉兰花粉为对象,采用悬滴培养法分析了温度、光照以及培养液的蔗糖和硼酸浓度对山玉兰花粉萌发的影响。结果表明:(1)山玉兰花粉萌发时,最适宜的温度为27℃。(2)光暗条件下,山玉兰花粉以浓度为5%的蔗糖培养效果最佳,其萌发率在16%以上;而硼酸浓度则以0.001%的培养效果最佳。(3)蔗糖与硼酸共同作用可有效促进花粉萌发和花粉管生长。其中,在光照条件下,以5.0%蔗糖+0.001%硼酸为最适宜的培养液,花粉萌发率达41.27%,花粉管长达281.49μm;而在黑暗条件下,则以5.0%蔗糖+0.01%硼酸为最适宜的培养液,花粉萌发率达45.71%,花粉管长达254.00μm。该研究结果为进一步开展人工辅助授粉、发掘山玉兰的种质资源工作奠定了基础。  相似文献

  1. From a strain of Acetobacter suboxydans, a glucose and a lacticenzyme were obtained in cell-free states. The lactic enzymeshows as strong activity as the glucose enzyme but is more stablethan the latter toward various purification procedures: bothare sensitive to high temperature treatment. Activities of thetwo enzymes and the MICHAELIS constants of the glucose enzymewere determined under both aerobic and anaerobic conditions.
  2. Carbon monoxide inhibits the oxygen-uptake in both glucoseandlactate oxidation. WARBURG's distribution constant for lactateoxidation is 6.7. These results suggest the participation ofan heme enzyme in the oxidation system.
  3. Effects of copperreagents, narcotics and PCMB were also examined.
  4. The dehydrogenaseactivities (reduction of dye) of the enzymesare more sensitiveto high temperature than the correspondingactivities in oxygen-uptake.
  5. By combining a dehydrogenase preparation which has lost itsoxygen-absorbing activity through acetone treatment, with aheated extract, a partial recovery of oxygen-uptake can be realizedin lactate oxidation.
  6. L-Cysteine is utilized as hydrogen donorby the bacterium. Thisoxidative reaction, unlike the oxidationof lactate, is notinhibited by surface active reagents.

  1. The sugars which induced gigantism of Chlorella cells wereglucose,fructose, galactose, mannose, xylose and arabinose.These sugarswere utilized as respiratory substrates by thealgal cells.
  2. The cellular division of Chlorella was stimulatedby glucoseand galactose, but suppressed by fructose, mannose,xylose andarabinose, while all these sugars evoked gigantism.No correlationwas found between cellular division and gigantism,
  3. The photosynthetic activity of giant Chlorella varied withthesorts of sugars added. It was decreased by glucose, fructoseand mannose, but was unaffected by other sugars such as galactose,xylose and arabinose.
  4. The respiratory activity of giant Chlorellacells as much higherthan that of control cells.
  5. The amountsof protein-N and dry weight per unit volume of giantChlorellawere much less than those of control cells.

Mode of action of FC was compared with that of auxin in differentexperimental systems and the following results were obtained.
  1. FC, as well as auxin, primarily induced elongation of the epidermisof pea epicotyl segments, but it also promoted elongation ofthe inner tissue, as judged by its action in split stem tests,elongation of hollow-cylinder segments and elongation of unpeeledand peeled segments.
  2. FC decreased the minimum stress relaxationtime (T0) and increasedthe extensibility (mm/gr) of the epidermalcell wall of peaepicotyl segments, as did auxin.
  3. FC failedto induce expansion growth of Jerusalem artichoketuber sliceswhen given alone or in combination with kinetinor gibberellicacid.
  4. FC at concentrations lower than 10–6 M, when givenwithauxin at concentrations lower than 0.03 mg/liter, promotedelongationof Avena coleoptile segments in an additive manner,to achievethe maximum elongation at higher concentrations.
  5. An antiauxin, 2,4,6-trichlorophenoxyacetic acid, inhibitedtheelongation of Avena coleoptile segments due to auxin butnotthat due to FC.
  6. Nojirimycin, an inhibitor of ß-glycosidases,inhibitedelongation of pea internode segments due not onlyto auxin butalso to FC.
  7. At concentrations more than 10–5MFC promoted root elongationof intact lettuce seedlings, whichwas inhibited by exogenousauxin.
From these results it is concluded that FC and auxin have acommon mechanism, which may involve hydrogen ion extrusion,leading to cell wall loosening and thus cell elongation. Thisgrowth is limited to the extent that the cells are capable ofelongating in response to hydrogen ions. Otherwise there isa definite difference in the mode of actions between FC andauxin, including the nature of cellular receptors for thesetwo compounds. (Received August 29, 1974; )  相似文献

  1. Several factors affecting sporulation of a wild yeast, Hansenulasaturnus, especially carbon sources and the carbon-nitrogenratio of sporulation medium were studied.
  2. The sporulationis stimulated at a certain definite C/N ratioof glucose medium.
  3. Several carbon sources such as ethanol, acetate, lactate,glycerol,succinate, glucose, gluconate and citrate are utilizedby theorganism both for growth and sporulation.
  4. The numberof spores in an ascus depends on the C/N ratio ofthe medium.An increase in the ratio stimulates the yield of2-and 3-sporedasci, especially of the former. One-spored ascibecome abundantas this ratio decreases.
  5. Lysine promotes sporulation in anacetate medium, and its presencein a large amount in glucosemedium also stimulates sporulation,while a small amount isinhibitory. When lysine was employedas the sole nitrogen source,most of the asci were 1-spored.
  6. It is discussed that sporulationof yeast is induced by a balanceof metabolism, rather thanby one definite "sporulation substrate".

  1. As previously demonstrated, normal cells of Chlorella protothecoidesare bleached with degeneration of chloroplasts when they areincubated, under aerobic conditions—either in the lightor in darkness—, in a glucose-containing medium withoutadded nitrogen source ("glucose-bleaching"). It was found inthe present study that under the atmosphere of N2, neither bleachingnor growth of algal cells occurs in the dark, while in the lighta significant growth of cells takes place with formation ofa certain amount of chlorophyll.
  2. Studies on the effects ofvarious inhibitors (ammonium ion,DNP, CMU, -hydroxysulphonates,arsenate, cyanide, azide, andantimycin A) under different conditionsshowed that oxidativephosphorylation is a necessary processfor the occurrence ofthe glucosebleaching as well as the assimilationof glucose(cellular growth). Under light-anaerobic conditionsin the presenceof glucose, assimilation of glucose (cellulargrowth) takesplace being supported by photophosphorylation,but no bleachingoccurs.
  3. When the algal cells in the courseof bleaching were transferredto the glucose-free mineral medium,the cell growth ceased immediatelybut the cell bleaching proceededfor several hours before itscessation. The respiratory activity,which was high in the glucose-containingmedium, became loweron transferring the algal cells into theglucose-free medium.The lowered level of respiration was maintained,for more than8 hr after the transfer of cells to the glucose-freemedium.
  4. When the cells in the course of bleaching were placed underthe atmosphere of N2, the cell bleaching ceased almost instantaneously.
  5. Based on these observations and other inhibition experiments,it was inferred that a certain intermediate(s) produced by theaerobic respiration of glucose is closely associated with theoccurrence of cell bleaching, and that an O2-requiring stepmay be involved in the process of chlorophyll degradation.
