Aims: Bacillus thuringiensis is toxic to many insects including Coleopteran pests. However, there is no report that B. thuringiensis is toxic to the adults of long‐horned beetle, Batocera horsfieldi, a pest of poplar trees. This work aims to select a B. thuringiensis strain toxic towards the adults of Asian long‐horned beetle. Methods and Results: A total of 504 B. thuringiensis strains were tested for the insecticidal activity to B. horsfieldi adults by artificial feeding. A strain of ZQ‐89 was found with a high toxicity to B. horsfieldi adults. The rectified lethal rate of ZQ‐89 to beetle was 55·33%. Additionally, the body weight and egg‐hatching rate of beetle, respectively, decreased by 2·22 and 19·62% after being fed with ZQ‐89. Further investigation found that the pure parasporal crystal had high toxicity to beetle adults. The ZQ‐89 crystal protein was purified and analysed by peptide‐mapping fingerprint and found it was highly homologous to Cry1Ac protein. The crystal protein gene was cloned and named cry1Ac89. The cry1Ac89 gene and its promoter were inserted into the plasmid pHT304 and then transformed into B. thuringiensis acrystalliferous strain BMB171. SDS‐PAGE analysis showed the BMB171‐Cry1Ac89 recombinant strain successfully expresses a 133‐kDa recombinant crystal protein with highly lethal activity to B. horsfieldi adults. Conclusions: The strain of ZQ‐89 is highly toxic to the adults of long‐horned beetle, and the crystal protein mainly contributes to the antipest role of this strain. The cry1Ac89 gene is a good candidate to be used for making transgenic trees or develop environment‐friendly bioinsecticides against long‐horned beetles such as B. horsfieldi in the future. Significance and Impact of the Study: It is the first report of a B. thuringiensis strain toxic to the adults of Asian long‐horned beetle, and the Cry1Ac protein is also firstly reported to be toxic to Coleopteran pests. 相似文献

In the recent past research has been mainly focused on the expression of cry1 genes of Bacillus thuringiensis (Bt) to engineer lepidopteran insect resistance in plants. Search for structurally different toxins is necessary for the management of resistance development in insects. The intact cry2Aa operon (3.95 kb) of a new isolate of Bt, 47-8, was subcloned into a Bt shuttle vector, pHT3101 (6.7 kb). Recombinant pHT3101 containing the cry2Aa operon of Bt strain 47-8 was named as pTN2Aa and used to transform acrystalliferous Bt strain 4Q7 by electroporation. Phase contrast microscopic observation revealed the presence of crystalline inclusions in the transformants of Bt strain 4Q7 harbouring pTN2Aa. SDS–PAGE of a spore–crystal mixture prepared from transformants of acrystalliferous Bt strain 4Q7 harbouring pTN2Aa showed a single band of about 65 kDa alone confirming the expression of the cloned cry2Aa. Bioassay with Helicoverpa armigera showed 71.4% mortality caused by the proteins encoded by the newly cloned cry2Aa gene (at the concentration of 2.3 g/l) on the seventh day and all the survivors that escaped from Cry2Aa toxicity showed severe (81–99%) inhibition in larval growth. 相似文献

Hybrid poplars (Populus tremula ×Populus tremuloides) have been genetically engineered viaAgrobacterium tumefaciens, to express a syntheticcry3Aa gene derived from the native Bacillusthuringiensis subsp. tenebrionis cry3Aa gene.The presence and the expression of the transgene have been verified in fourtransgenic poplar lines, using Southern, northern and western analyses. Thetransgenic poplar's toxicity towards the phytophagous beetleChrysomela tremulae (Coleoptera, Chrysomelidae) has beenassessed on six month-old greenhouse-grown selected plants in laboratoryconditions. Laboratory experiments consisted of feeding tests of fresh detachedleaves on C. tremulae at all developmental stages. Ourresults indicate that the transgenic poplar leaves, expressing a Cry3Aa proteinamount in a range of 0.05–0.0025% of total soluble protein, weredefinitely deleterious for C. tremulae, regardless of thedevelopmental stage. 相似文献

Bacillus thuringiensis insecticidal proteins toxic action relies on the interaction with receptor molecules on insect midgut target cells. Here, we describe an ADAM metalloprotease as a novel type of B. thuringiensis toxin receptor on the basis of the following data: (i) by ligand blot and N-terminal analysis, we detected a Colorado potato beetle Cry3Aa toxin binding molecule that shares homology with an ADAM10 metalloprotease; (ii) Colorado potato beetle brush border membrane vesicles display ADAM activity since it cleaves an ADAM fluorogenic substrate; (iii) Cry3Aa acts as a competitor of the cleavage of the ADAM fluorogenic substrate; (iv) Cry3Aa sequence contains the recognition motif R(345)FQPGYYGND(354) present in ADAM10 substrates. Accordingly, a peptide representative of the recognition motif localized within loop 1 of Cry3Aa domain II (Ac-F(341)HTRFQPGYYGNDSFN(358)-NH(2)) effectively prevented Cry3Aa proteolytic processing and nearly abolished pore formation, evidencing the functional significance of the Cry3Aa-ADAM interaction in relation to this toxin mode of action. 相似文献

通过对晶体蛋白N-末端氨基酸测序,设计简并探针,从对根结线虫高毒力苏云金芽胞杆菌YBT-1518菌株中克隆到1个含有杀线虫晶体蛋白基因的片段。序列测定表明该序列含有两个ORF(orf1和orf2),其中orf1与基因cry6Aa1同源性为98%,已在GenBank上登录(Acc.NO.AF499736),并被命名为cry6Aa2。将克隆的该片段克隆到穿梭载体pHT304上,并转化苏云金芽胞杆菌无晶体突变株BMB171,重组菌株可形成米粒状伴胞晶体。生物测定表明,表达的毒素蛋白对北方根结线虫的LC50为9.47μg/mL,毒力与出发菌株(10.74μg/mL)相当。 相似文献

Bt菌Ly30株是我国自行分离的对多种害虫具有高毒力的苏云金芽孢杆菌,经CAPS(cleaved amplified polymorphic sequences)系统鉴定,它含有cry1Aa基因。以全长基因PCR产物的粘端定向克隆的方法, 设计一对特异引物,分别引入NcoⅠ和BamHⅠ/NcoⅠ酶切位点。以Ly30质粒DNA为模板扩增cry1Aa全长基因,与表达载体Pkk233-2相应酶切产物连接,转化大肠杆菌,获得含有cry1Aa基因重组质粒pKKLy1Aa。完成了该基因的亚克隆和序列测定,结果表明,该基因的编码区为3 531 bp,编码蛋白分子量为133.2kD,含1.176个氨基酸,等电点Pi为4.99。该基因序列已在GenBank中登记注册,登录号为AF384211,并被国际Bt杀虫晶体蛋白基因命名委员会正式命名为cry1Aa12。对重组菌KKLy1Aa进行诱导表达研究。在0.6 mmol/L IPTG、37℃、8 h培养条件下,该基因获得高效表达,SDS-PAGE电泳检测到明显的133.2 kD蛋白带。室内生测结果表明,Cry1Aa蛋白对不同的小菜蛾品系均有较高的杀虫活性,其LC50值分别为0.203 μg/mL和0.554 μg/mL。 相似文献

N 乙酰高丝氨酸内酯 (N acyl homoserinelactones,AHLs) ,是一类数量感知 (Quorum sensing)系统中的信号分子 ,它参与诱导调控许多植物病原菌致病基因的表达。苏云金芽胞杆菌的AiiA蛋白能降解这类AHLs分子 ,进而可减弱病原菌致病基因表达产生的病害。苏云金芽胞杆菌杀虫晶体蛋白基因cry3Aa的启动子是一种不依赖芽胞形成的启动子 ,它相对于其它cry类基因的启动子有启动基因转录时间早 ,转录时间长的优点。通过重叠延伸PCR ,用杀虫晶体蛋白基因cry3Aa启动子替换编码AiiA蛋白的基因aiiA自身的启动子 ,构建了融合基因pro3A aiiA。将融合基因装入穿梭载体pHT3 0 4的BamHI SphI位点 ,得到重组质粒pBMB686并转化苏云金芽胞杆菌无晶体突变株BMB171,重组菌株BMB686的AiiA蛋白表达量在各个生长时期均高于对照菌株 ,对AHLs分子的降解活性和对胡萝卜软腐欧文氏菌感染马铃薯产生病害的抑制能力也明显优于对照菌株 相似文献

苏云金芽胞杆菌幕虫亚种的伴胞晶体在预芽胞外壁内侧形成,呈现晶体芽胞粘连的现象。根据已发表的cry26Aa1和cry28Aa1基因序列设计引物,从苏云金芽胞杆菌幕虫亚种T02中扩增得到cry26Aa和cry28Aa基因,通过穿梭载体将这两个基因分别和同时转化到苏云金芽胞杆菌无晶体突变株BMB171后,透射电镜下可在芽胞外壁内侧和外侧同时观察到伴胞晶体,而单独表达时可在芽胞外壁外侧观察到伴胞晶体。结果表明,伴胞晶体在芽胞外壁内侧表达不单独依赖于启动子的时空调控,可能还受到晶体蛋白相互作用的影响。 相似文献

Using the shuttle vector pBU4, the mosquitocidal toxin gene mtx1 from Bacillus sphaericus strain SSII-1 was introduced into an acrystalliferous strain of B. thuringiensis both individually and in combination with the accessory protein gene p20 and the cytolytic protein gene cyt1Aa from B. thuringiensis subsp. israelensis. Bioassay results indicated that the recombinants B-pMT4(Mtx1) and B-pMT9(Mtx1), both individually containing mtx1, had moderate toxicities to binary toxin susceptible and binary toxin resistant Culex quinquefasciatus larvae during the vegetative growth stage, but that their toxicities declined rapidly during the sporulation phase. The LC50 values were 2.5 and 4.8 mg/ml respectively, against 3-4 instar susceptible and resistant larvae for the final sporulated cultures of recombinants B-pMT9(Mtx1), and little toxicity was detected for B-pMT4(Mtx1). Meanwhile, the recombinant B-pMPX2(Mtx1+Cyt1Aa) expressing Mtx1, P20 alone, and Cyt1Aa in combination had stable toxicities during both the vegetative phase and the sporulation phase, with a LC50 ranging from 0.45-0.58 mg/ml. Furthermore, expression of Cyt1Aa appeared to enhance the activity of Mtx1 to target mosquito larvae, suggesting a synergism between Cyt1Aa and Mtx1 toxins. 相似文献

The inheritance and expression patterns of the cry1Ab gene were studied in the progenies derived from different Bt (Bacillus thuringiensis) transgenic japonica rice lines under field conditions. Both Mendelian and distorted segregation ratios were observed in some selfed and crossed
F2 populations. Crosses between japonica intra-subspecies had no significant effect on the segregation ratios of the cry1Ab gene, but crossing between japonica and indica inter-subspecies led to distorted segregation of the cry1Ab gene in the F2 population. Field-release experiments indicated that the cry1Ab gene was stably transmitted in an intact manner via successive sexual generations, and the concentration of the Cry1Ab protein
was kept quantitatively stable up to the R6 generation. The cry1Ab gene, driven by the maize ubiquitin promoter, displayed certain kinds of spatial and temporal expression patterns under field conditions. The content of the Cry1Ab
protein varied in different tissues of the main stems, the primary tillers and the secondary tillers. Higher levels of the
Cry1Ab protein were found in the stems, leaves and leaf sheaths than in the roots, while the lowest level was detected in
grains at the maturation stage. The content of the Cry1Ab protein in the leaves peaked at the booting stage and was lowest
at the heading stage. Furthermore, the Cry1Ab content of cry1Ab expression in different tissues of transgenic rice varied individually with temperature.

A new gene, cry11Bb2 from a field isolate of Bacillus thuringiensis, was cloned for expression in Escherichia coli. The encoded protein, with a deduced molecular mass of 89.5 kDa, exhibits 97 and 79% identities with the overlap regions of Cry11Bb1 from B. thuringiensis ssp. medellin and Cry11Ba1 from ssp. jegathesan, respectively. It is however longer than Cry11Bb1 by 42 amino acids in its carboxy-terminus, of which 32 comprise 2 tandem repeats additional to the 5 existing in the latter polypeptide. Possible roles for this recurrent motif among Cry toxins and their accessory proteins, and for their encoding genes are proposed. 相似文献

Bacillus thuringiensis (Bt) is the major source for transfer of genes to impart insect resistance in transgenic plants. Cry2A proteins of Bt are promising candidates for management of resistance development in insects due to their difference from the currently used Cry1A proteins, in structure and insecticidal mechanism. Two insecticidal crystal protein genes of Bt, viz. cry2Aa and cry2Ab were cloned from new isolates of Bt, 22-4 and 22-11, respectively. Expression of both the genes was studied in an acrystalliferous strain of Bt (4Q7) by fusing the cry2Aa and cry2Ab genes downstream of cry2Aa promoter and orf1 + orf2 sequences. Western blot analysis revealed a low level expression of the cloned cry2Aa and cry2Ab genes in the recombinant Bt strains. High-level expression of cry2Aa and cry2Ab genes was achieved in the recombinant E. coli by cloning the cry2A genes under the control of the T7 promoter. 相似文献

利用重叠PCR的方法,通过两次PCR扩增,分别获得cry2A10操纵元的orf1、orf1 orf2与cry2Ab5基因的融合片段。融合片段经BamHⅠ和EcoRⅠ双酶切与pHT315连接,分别构建了基因融合片段的原核表达载体pFU(orf1 2Ab)和pFU(orf1 orf2 2Ab),电转化Bt无晶体突变株4Q7后,扫描电镜下可观察到典型的方形晶体,通过SDS-PAGE可检测到60kD大小的蛋白表达带。结果表明,cry2Ab5可在cry2a0的启动子帮助下有效转录和表达,并在orf2产物帮助下形成蛋白晶体。 相似文献

protein of the expected size of 20 kDa, but a 10-amino acid peptide. A second, highly conserved start codon is located 25
bp downstream from the first one and corresponds to an open reading frame of the same size in all known orf1-related sequences. Expression of lacZ gene fusions created at the level of the first ATG, second ATG, and stop codon of the NRD-12 orf1 sequence showed that orf1 is translated from the second ATG. The expected protein is 19 kDa in size. The expression starts at t2, which is in agreement
with the presence of a BtI promoter in the cry2Aa1 operon.

Helix 3 of the Cry1Aa toxin from Bacillus thuringiensis possesses eight charged amino acids. These residues, with the exception of those involved in intramolecular salt bridges (E90, R93, E112, and R115), were mutated individually either to a neutral or to an oppositely charged amino acid. The mutated genes were expressed, and the resultant, trypsin-activated toxins were assessed for their toxicity to Manduca sexta larvae and their ability to permeabilize M. sexta larval midgut brush border membrane vesicles to KCl, sucrose, raffinose, potassium gluconate, and N-methyl-D-glucamine hydrochloride with a light-scattering assay based on osmotic swelling. Most mutants were considerably less toxic than Cry1Aa. Replacing either E101, E116, E118, or D120 by cysteine, glutamine, or lysine residues had only minor effects on the properties of the pores formed by the modified toxins. However, half of these mutants (E101C, E101Q, E101K, E116K, E118C, and D120K) had a significantly slower rate of pore formation than Cry1Aa. Mutations at R99 (R99C, R99E, and R99Y) resulted in an almost complete loss of pore-forming ability. These results are consistent with a model in which alpha-helix 3 plays an important role in the mechanism of pore formation without being directly involved in determining the properties of the pores. 相似文献

The spotted asparagus beetle, Crioceris quatuordecimpunctata (Coleoptera: Chrysomelidae), is one of the most devastating pests of asparagus in China. Sprayed synthetic pesticides have been used to control C. quatuordecimpunctata damage, but they pose problems because of residues and harm to natural enemies. Neither the microbial coleopteran-specific toxin from Bacillus thuringiensistenebrionis, Cry3Aa, nor the fungal pathogen Beauveria bassiana have sufficient activity to effectively control C. quatuordecimpunctata damage to asparagus. However, second instar C. quatuordecimpunctata larvae exposed to a sublethal dose of Cry3Aa toxin demonstrated significantly higher larval mortality when exposed to B. bassiana. Our results suggest that a combination of Cry3Aa and B. bassiana may be effective in reducing damage by C. quatuordecimpunctata larvae to asparagus. 相似文献

A cry1Ab-type gene was cloned from a new isolate of Bacillus thuringiensis by PCR. When restriction pattern was compared with that of known genes it was found to have additional restriction site for ClaI. Nucleotide sequencing and homology search revealed that the toxin shared 95% homology with the known Cry1Ab proteins as compared to more than 98% homology among the other reported Cry1Ab toxins. The gene encoded a sequence of 1,177 amino acids compared to 1,155 amino acids encoded by all the other 16 cry1Ab genes reported so far. An additional stretch of 22 amino acids after the amino acid G793 in the new toxin sequence showed 100% homology with several other cry genes within cry1 family. Homology search indicated that the new cry1Ab-type gene might have resulted by nucleotide rearrangement between cry1Ab and cry1Aa/cry1Ac genes. 相似文献

Vegetative insecticidal protein (Vip) is a newly discovered family of toxin protein isolated from Bacillus thuringiensis (Bt). An 88.5-kDa Vip3Aa protein was secreted by a local strain of the bacterium during the vegetative growth phase. The full length of the coding region ‘2.3 kbp’ of the vip3Aa gene was isolated from plasmid DNA, cloned in pGEM-T vector and finally cloned in pQE-30 expression vector. Nucleotide sequence revealed 98% homology with that of the previously isolated genes. Expression of the vip3Aa in Escherichia coli was carried out and the expressed protein was detected in the concentrated supernatant, not in the pellet. This indicated that vip3Aa is secreted into the culture medium. Expressed protein was purified, blotted, and assayed against the cotton leaf worm Spodoptera littoralis. The LC50 was found to be 142.4 µ/mL while the LC50 was 90 ppm for the wild strain. These results suggest the use of either the isolated Bt strains or the expressed vip3Aa in an integrated pest management program against lepidopteran insect pests. 相似文献

We characterised a novel holotype cry gene (cry1Hc1) harboured in BN23-5 Bt strain which was isolated from soil samples in Sichuan, China. In this study, the full length of the cry1Hc1 gene was cloned from the strain. The cry1Hc1 gene was inserted into a shuttle vector (pSTK) and expressed in an acrystalliferous mutant Bacillus thuringiensis HD73?. In this transformant, cry1Hc1 was expressed and diamond-shaped parasporal crystals were formed. The resulting insecticidal activity showed that the Cry1Hc1 protein exhibited high larvicidal activity against larvae of Plutella xyllostella and Ostrinia furnacalis with lethal concentration 50 of 55.21 and 118.44?μg/ml, respectively. Sequence analysis of the gene was also performed; the Cry1Hc1 protein retained eight conserved regions commonly found in the existing Cry proteins. 相似文献
