Summary Dissociated single cells from chicken retina or tectum kept in rotation-mediated cell culture aggregate, proliferate and establish a certain degree of histotypical cellto-cell relationships (sorting out), but these systems never form highly laminated aggregates (nonstratified R- and T-aggregates). In contrast, a mixture of retinal plus pigment epithelial cells forms highly stratified aggregates (RPE-aggregates, see Vollmer et al. 1984). The present comparative study of stratified and nonstratified aggregates enables us to investigate the process of cell proliferation uncoupled from that of tissue stratification. Here we try to relate these two basic neurogenetic processes with patterns of expression of cholinesterases (AChE, BChE) during formation of both types of aggregates.During early aggregate formation, in both stratified and nonstratified aggregates an increased butyrylcholinesterase activity is observed close to mitotically active cells. Quantitatively both phenomena show their maxima after 2–3 days in culture. In contrast, AChE-expression in all systems increases with incubation time. In nonproliferative areas, in the center of RPE-aggregates, the formation of plexiform layers is characterized initially by weak BChE and then strong AChE-activity. These areas correspond with the inner (IPL) and outer (OPL) plexiform layers of the retina in vivo. Although by sucrose gradient centrifugation we find that the 6S- and the fiber-associated 11S-molecules of AChE are present in all types of aggregates, during the culture period the ratio of 11S/6S-forms increases only in RPE-aggregates, which again indicates the advanced degree of differentiation within these aggregates.It is thus demonstrated that cholinesterases first correlate with neuronal cell proliferation and later with stratification, which indicates functions of both enzymes during both developmental periods.Abbreviations AChE acetylcholinesterase - BChE butyrylcholinesterase - iso-OMPA specific inhibitor of BChE - BW 284C51 specific inhibitor of AChE - IPL inner plexiform layer - OPL outer plexiform layer  相似文献

Man kann daraus schließen, daß diese beiden photomorphogenetischen Reaktionen des Senfkeimlings ebenfalls durch eine von P730 über eine Signalkette ausgelöste Aktivierung von potentiell aktiven Genen veranlaßt werden.
The inhibition of phytochrome-mediated photomorphogenesis (positive photoresponses) by actinomycin D and puromycin in the mustard seedling (Sinapis alba L.)
Summary The many photochrome-mediated photoresponses of a seedling (Sinapis alba L., white seeded mustard) can be divided into 3 categories: positive, negative, and complex photoresponses. Positive photoresponses are those which are characterized by an initiation or a promotion of biosynthetic or growth processes (Mohr, 1966b). Phytochrome-mediated anthocyanin synthesis is the prototype of a positive photoresponse. It has been shown in previous papers (e.g. Lange and Mohr, 1965; Mohr et al., 1965) that positive photoresponses can be specifically inhibited by actinomycin D and puromycin. It has been concluded that in the case of positive photoresponses P730 (the active phytochrome) exerts its function through differential gene activation.—In the present paper it has been demonstrated that phytochrome-mediated positive photoresponses of the mustard seedling like opening of the hypocotylar hook and unfolding of the cotyledons can be inhibited by relatively low doses of actinomycin D and puromycin in very much the same way as anthocyanin synthesis or cotyledon enlargement is inhibited. It has been concluded that in these cases too the action of P730 must be attributed to an activation of potentially active genes in the manner postulated on the basis of the data on anthocyanin synthesis.

As a supporting component of the VALIMARproject, physicochemical investigations wereconducted monthly from 1995 to 1999 at theKrähenbach/Aich stream system (two samplingsites) and at the Körsch stream (sixsampling sites). Several physicochemicalparameters were analysed continuously bydataloggers during the entire sampling period.Moreover, a selection of the most importantmorphological parameters of the stream wererecorded in 1998.The results of these physicochemicalinvestigations demonstrated that theKrähenbach/Aich system could becharacterized as a natural rhithral submontanecarbonate stream system that is welloxygenated. Low temporal variations in pH,conductivity and chloride occurred at allsampling sites in the Krähenbach whichcoincided with the geological conditions atthis test stream. Because of relatively lowconcentrations of organic and inorganiccompounds during the entire study period thesites AB and AC in the Krähenbach/Aichsystem were classified as slightly polluted.The morphological assessment of the streamindicated a moderately affected status atboth of these streams.Physicochemical and morphologicalinvestigations along the Körsch streamsystem (KA to KE including the tributary streamSulzbach) indicated a highly affected rhithralsubmontane carbonate system characterized byhigh water velocity, flow rate and dissolvedoxygen. The Körsch, however, differssignificantly with respect to watertemperature, conductivity, pH, BOD5,nutrients, and chloride concentrations from theKrähenbach. As a result of differentanthropogenic inputs (e.g. several sewageplants, agriculture activities and waste waterdumping) the Körsch sites KA to KD wereclassified as critically to heavilypolluted. The morphological assessmentindicated a clearly affected to damagedstatus. The upstream site KE could becharacterized as slightly polluted and itsmorphological structure as moderatelyaffected.  相似文献

Summary Chorionic villi from first trimester and term human placentas have been incubated in vitro and shown to release the lysosomal enzymes, -hexosaminidase, -glucosidase and -gluctlronidase. There was negligible release of the cytoplasmic enzyme, lactate dehydrogenase, under the same conditions. The first trimester villi released proportionally more of their lysosomal enzyme content than did the term villi. Extracellular levels of -hexosaminidase were raised and those of -glucosidase and, -glucuronidase were lowered when tissue was incubated with 1 M colchicine, suggesting that microtubules are involved in the control of lysosomal enzyme release from placental villi.  相似文献

Summary The pattern of inheritance of several polymorphic restriction sites associated with the -gene cluster, and spanning a region of 52 kb, demonstrates that a determinant for hereditary persistence of fetal hemoglobin (HPFH) segregates independently from the non- globin gene cluster, as we postulated several years ago on purely genetical grounds. This finding provides additional evidence for the existence of diffusible factors affecting -chain expression. Moreover, we have identified a private HinccII polymorphism, in the vicinity of the gene in the family studied.  相似文献

Summary In human placental villi the connective tissue is constructed by mesenchymal cells, small and large reticulum cells and fibroblasts. During early pregnancy mesenchymal cells dominate; starting with the third month of gestation the reticulum cells are in the majority within the terminal villi, the fibroblasts within the stem villi. Ultrastructurally intermediary types of cells can be differentiated. Together with reticular and collagenous fibres the reticulum cells form the basic architecture of the villous stroma during the first 2/3 of gestation: the reticular type of stroma. This consists of a network of cells and fibres with fetal vessels fitted in between. The remaining interspaces form a fluid system of compartments in which Hofbauer cells are suspended. They are called stromal channels. During the last trimester these channels and the Hofbauer cells as well are progressively replaced either by voluminous masses of fibres (fibrous type of stroma, mainly in the stem villi) or by sinusoidal enlargements of fetal capillaries (sinusoidal type of stroma, mainly in the terminal villi).Supported by grants from the Deutsche ForschungsgemeinschaftThe authors are indebted to Mrs. E. Examination of the binding of (+)[3H]SKF 10,047 to rat brain membranes indicated that at a low concentration most of the binding was to the haloperidol-sensitive binding site. Titration curves exhibited a displacement potency order of haloperidol >(+)SKF 10,047 =1,3-diorthotolyl-guanidine(–)SKF 10,047 phencyclidine analogues. The effect of Fab fragments from a monoclonal antibody, OR-689.2.4, directed against the opioid receptor on the binding of(+)[3H]SKF 10,047 to rat brain membranes was examined. The specificity of this antibody for the opioid receptor has been determined by its ability to inhibit the binding of and opioid peptides to rat brain but not the binding of opioid ligands or nonopioid ligands specific for other receptors. The Fab fragments blocked and displaced specifically bound (+)[3H]SKF 10,047 in a titratable manner. Increasing the incubation time of the membranes with the Fab fragments increased the percent inhibition obtained. The Fab fragments acted as noncompetitive inhibitors of (+)[3H]SKF 10,047 binding. A(+)SKF 10,047 binding site in rat brain appears to share a common structural domain with and opioid receptors.  相似文献

Summary The distribution of acid phosphatase, -n-acetylglucosaminidase, -glucuronidase, and acid -galactosidase was studied in mm. extensor digitorum longus, soleus, and diaphragm of rats. Using the technic of semipermeable membranes activities of these enzymes were demonstrated beside cells of the interstitial tissue in muscle fibers themselves as well. Acid phosphatase displayed the highest activity which appeared in many small dots dispersed in the fiber. The activity of acid phosphatase was about 1.2 x higher in the m. soleus than in the m. extensor digitorum longus. In the latter muscle a somewhat higher activity was often found in muscle fibers displaying a higher staining for NADH tetrazolium reductase. The activity of -n-acetylglucosaminidase was slightly lower, that of -glucuronidase very weak but still discernible. The activity of acid -galactosidase was not ascertained in the majority of fibers. The ratio of activities measured in an area of the same size in cells of the interstitial tissue and in muscle fibers amounted in average to 2.6: 1 in the case of acid phosphatase, 2.5:1 in the case of -n-acetylglucosaminidase, 5.7: 1 in the case of -glucuronidase, and 44.3:1 in the case of acid -galactosidase. The importance of the histochemical technic in studies concerned with acid hydrolases in striated muscle fibers in normal and pathological conditions is pointed out.  相似文献

Summary The M. complexus in the chick, commonly called the hatching muscle, undergoes conspicuous growth during the latter stages of embryonic development. Myogenesis of this muscle was compared to that of M. biceps femoris with regard to development of types of muscle fiber and their innervation. In both muscles fibers are of relatively uniform size and show little growth in diameter between 12 days of development and hatching; fibers develop continuously and display a wide range of diameters at all stages.Initial thickenings on the sarcolemma of fibers where axons are closely approximate were first observed at 10 days of development in both muscles. In both muscles fibers are innervated prior to fibers. Terminal axon networks bridge intercellular spaces and contact fibers in different myogenic clusters, fibers that develop on the surface membrane of fibers exhibit focal thickenings of the membrane and some cell projections that are directed toward axon- fiber contacts. These changes occurred only in fibers of M. complexus.At 14 days of embryogenesis, the processes of synaptogenesis and of myelin formation are less advanced in M. biceps femoris than in M. complexus. At this stage a fibers were observed to be innervated in M. complexus, but not yet in M. biceps femoris. The effects of physiological and excessive levels of growth hormone (GH) on reproductive functions are poorly understood, and impairment of fertility is frequently observed in transgenic animals overexpressing GH genes. The present study was undertaken to determine the effects of chronic exposure to heterologous bovine GH (bGH) on the testes and accessory reproductive glands in transgenic mice. Endocrine function of the testes was evaluated by measuring the activities of two steroidogenic enzymes, 5-3-hydroxysteroid dehydrogenase (5-3-HSD) and 17-hydroxysteroid dehydrogenase (17-HSD). The activities of acid phosphatase, alkaline phosphatase and -glucuronidase, important hydrolytic enzymes of lysosomal origin, were measured in testes, seminal vesicles and ventral prostates in normal and transgenic mice. Testicular 5-3-HSD activity was higher in transgenic than in normal mice, while testicular 17-HSD activity in transgenic mice was not altered. Acid phosphatase activity was elevated in both seminal vesicles and ventral prostates of transgenic mice, while alkaline phosphatase activity was increased only in the prostate. The activity of -glucuronidase was elevated in the testes, seminal vesicles and ventral prostate gland of transgenic mice. These results suggest that chronic exposure to bGH is associated with significant stimulation of some hydrolytic enzymes in the testes and in the accessory reproductive glands of transgenic mice.  相似文献

A triple-resonance NMR technique suitable for the determination ofcarbonyl-related couplings in polypeptide systems is introduced. Theapplication of three novel pulse sequences to uniformly13C/15N-enriched proteins yields E.COSY-likemultiplet patterns exhibiting either one of the3J(Ci–1,Hi ), 3J(Ci–1,Ci ) and3J(Ci–1,Ci)coupling constants in the indirectly detected 13Cdimension, depending on the passive spin selected. The experiments aredemonstrated with oxidized flavodoxin from Desulfovibrio vulgaris. On thebasis of the J-values measured and the backbone -angles derived from ahigh-resolution X-ray structure of the protein, the three associated Karplusequations were reparametrized. The root-mean-square differences between theexperimental coupling constants and those predicted by the optimized Karpluscurves are 0.41, 0.33 and 0.32 Hz for3J(Ci–1,Hi ),3J(Ci–1,Ci ) and3J(Ci–1,Ci),respectively. The results are compared with the Karplus parameters previouslypublished for the same couplings.  相似文献

The properties of seven enzymes were studied in extracts from Myxobacter AL-1. The enzymes were isocitrate dehydrogenase (E.C. 1.1.1.42), succinate dehydrogenase (E.C. 1.3.99.1), alkaline phosphatase (E.C. 3.1.3.1), -glucosidase (E.C. 3.2.1.20), -glucosidase (E.C. 3.2.1.21), -galactosidase (E.C. 3.2.1.23), and N-acetyl-glucosaminidase (E.C. 3.2.1.30). Four of these enzymes: isocitrate dehydrogenase, -glucosidase, -glucosidase, and -galactosidase are cytosolic enzymes. Succinate dehydrogenase was found to be located on the cytoplasmic membrane system, whereas alkaline phosphatase and N-acetyl-glucosaminidase were considered as enzymes which bind the outer membranes resp. the cell wall. During the cell cycle, all enzymes have a pattern of discontinuous activity increase. Succinate dehydrogenase and isocitrate dehydrogenase exhibit a stepwise increase of activity, whereas the other enzymes follow the pattern of a peak enzyme.  相似文献

This paper describes and analyzes a type of Toraja (South Sulawesi, Indonesia) suicide in which a person kills him or herself after having been slighted or offended, usually by a close family member. Comparing and contrasting such suicides to similar types found elsewhere in Austronesia-speaking Oceania, the paper argues that self-inflicted deaths of this nature are not so much anomic, as suggested by some analysts, as indignant; they are committed by persons who feel that they have been severely mistreated or abused according to traditional notions of reciprocity, mutual aid, and the dangers of frustrated desire. The paper concludes by suggesting that Durkheim may have underestimated the frequency and importance of indignant suicide in traditional societies and that, conversely, contemporary analysts may be underestimating the importance of traditional norms and values in accounting for the high rates of suicide found in many parts of the Pacific today.  相似文献

The ADAMs (A Disintegrin And Metalloprotease-like) family is a large and rapidly expanding group of metalloproteinases with structural similarity. They are normally characterized by the presence of a proteolytic domain and disintegrin and signalling domains. Although 21 ADAMs proteins have been already cloned to date, in most cases their natural substrates are unknown. The best characterized representative of the mammalian ADAMs family is the TNF- converting enzyme (TACE). TACE is an integral membrane metalloproteinase that causes the secretion of the active form of TNF- from its plasma membrane precursor and thus can be regarded as a membrane protein secretase. Secretion of membrane proteins is a very well documented biological phenomenon and was demonstrated for a diverse range of membrane proteins, two examples being angiotensin converting enzyme (ACE) and Alzheimer's amyloid precursor protein (APP). ACE and APP secretion was shown to possess substantial similarity with the secretion of TNF-. In the present study, we have attempted to demonstrate that a metalloproteinase might be involved in the shedding of another membrane-bound protein – acetylcholinesterase (AChE). Secretion of AChE by human neuroblastoma SH-SY5Y cells was found to be inhibited by a selective hydroxamate metalloproteinase inhibitor batimastat (20 M), and stimulated by carbachol (20 M), which have previously been shown to regulate the activity of APP -secretase in a similar manner. The role of ADAMs proteins in the shedding of molecules from the cell surface is discussed.  相似文献

Summary The morphology of the absorbing cells of the duodenal villi in the mouse, the rat, the hamster and the guinea-pig is described. The polymorphism of the dense bodies is pointed out. The fine localization of acid and alkaline phosphatase is investigated and compared. In all the species, acid phosphatase activity is observed in the dense bodies, Golgi vesicles and rare smooth endoplasmic profiles. Alkaline phosphatase is localized on the microvilli, Golgi apparatus, some smooth endoplasmic cisternae and numerous dense bodies. The presence of an alkaline phosphatase reaction in the dense bodies, probably lysosomes, of the absorbing cells is discussed. Proteinl-isoaspartyl methyltransferase, an enzyme enriched in brain, is implicated in the repair of age-damaged proteins containing atypical, isoaspartyl peptide bonds. We have investigated the kinetics of methylation using a synthetic peptide substrate having the structure Trp-Ala-Gly-Gly-isoAsp-Ala-Ser-Gly-Glu. Double-reciprocal plots of initial velocity versus concentration of S-adenosylmethionine (AdoMet) at different fixed concentrations of peptide gave straight lines converging at a positive 1/v value and a negative 1/AdoMet value. The product S-adenosylhomocysteine (AdoHcy) was a competitive inhibitor towards AdoMet and a linear mixed-type inhibitor towards peptide. These results are consistent with the rapid-equilibrium random sequential bi-bi mechanism previously proposed for the enzyme, but they also reveal the formation of the deadend, enzyme-peptide-AdoHcy, complex. The rate constants were:V max=32–34 nmol/min/mg,K peptide=7.6–9.4 M,K AdoMet=1.9–2.2 M, =0.43–0.53,K AdoHcy=0.08 M, =2.9. The interaction factors and indicate that binding of enzyme to peptide increases its affinity for AdoMet and decreases its affinity for AdoHcy. Methylation was linear with time throughout the transfer of 2 mol of methyl groups/mol of enzyme. Summary The present study compares the distribution of -glucuronidase and succinic dehydrogenase in young and old spinal ganglion cells of rat. In young cells there are indications of cyclic activity of these enzymes, i.e., in some stages there are perinuclear concentrations of the enzymes, at other times -glucuronidase and succinic dehydrogenase are uniformly distributed throughout the cytoplasm. These stages have been discussed with the identical distribution of mitochondria. However, in old spinal ganglion cells both -glucuronidase and succinic dehydrogenase become mainly concentrated in the pigment areas, suggesting thereby their possible role in the production of pigment, through the medium of the mitochondria.  相似文献

Lumbricillus lineatus selectively ingests masses of organic and inorganic interstitial particles from a sand-clay substratum in the upper littoral zone. Particle-masses are ingested, passed along the esophagus and into the anterior intestine where the pH becomes acid. A- and C-esterases, acid -galactosidase, acid phosphatase and -N-acetylglucosaminidase are present in the epithelium, while the rotating food masses are surrounded by a membrane of sulphated, acid glycoprotein. These enzymes, with the exception of acid phosphatase and the addition of aminopeptidase M, are also present in the epithelia of the mid and posterior intestinal regions where the pH is alkaline. The cells in the ventral wall of the mid intestinal region contain high concentrations of alkaline phosphatase, acid -galactosidase and -N-acetylglucosaminidase. The food consists of absorbed organics and bacteria with absorption and intracellular digestion occurring along the intestine, particularly in the mid ventral region.  相似文献

Summary The temporal muscles of the guinea pig show a sexual differentiation reflected in their histochemical enzyme pattern. Using histochemical methods for mitochondrial (SDH, -GPDH), and glycolytic enzymes (phosphorylase, LDH) it could be shown, that in adult animals the male muscle is a white muscle with marked activity of glycolytic enzymes, the female muscle a red muscle displaying high activity of mitochondrial enzymes. This differential enzyme pattern can be converted by the application of testosterone to the female type during the postnatal development. The male sex hormone thus affects the histochemical enzyme pattern of the muscle, converting the red, female into a white, male muscle in the female guinea pig.  相似文献

Summary The histochemical distribution of various enzymes, such as alkaline phosphatase, acid phosphatase, esterase, -glycosidase, aminopeptidase, succinic dehydrogenese and TPN diaphorase, in human submaxillary glands has been determined.Acini and ducts of human submaxillary gland were devoid of alkaline phosphatase activity, but this enzyme was observed in capillaries and somewhat in myoepithelium.Activities of acid phosphatase, esterase, -glucuronidase and -galactosidase were generally observed in the entire cytoplasm of serous acini; but the cytoplasm of mucous acini was either negative or showed only trace amounts.Aminopeptidase reaction of both acini and ducts was generally negative.Succinic dehydrogenase and TPN diaphorase activities were strongly active in intralobuler ducts. Serous acini exhibited less activity with these enzymes; and mucous cells showed still less and were almost negative.
