Mohapatra, S. S., Poole, R. J. and Dhindsa, R. S. 1987. Coldacclimation, freezing resistance and protein synthesis in alfalfa(Medicago sativa L. cv. Saranac).—J. exp. Bot. 38: 1697–1703. Changes in freezing resistance (percent survival at —10°C), pattern of protein synthesis and translatable mRNApopulation during cold acclimation of alfalfa (Medicago sativaL. cv. Saranac) have been examined. Two days of cold acclimationat 4 °C increased freezing resistance from about 6% to 40%,protein content by 200% and total RNA content by 100%. Acclimationfor longer periods did not cause further increases in freezingresistance, protein content or RNA content. Examination of proteinchanges by sodium dodecyl sulphate-polyacrylamide gel electrophoresis(SDS-PAGE) coupled with protein staining, and by fluorographyof in vivo labelled proteins separated by SDS-PAGE, showed thatseveral proteins are increasingly or newly synthesized duringcold acclimation. Analysis of in vitro translation productsby SDS-PAGE and fluorography shows changes in the populationof translatable mRNAs. It is concluded that in this varietyof alfalfa cold acclimation for only 2 d is sufficient to confermaximum freezing resistance, and that changes in proteins duringcold acclimation are regulated most probably at the transcnptionallevel. Larvae (72 hr old) of P. lividus and A. lixula grazed on varioussuspensions of natural particulate matter with a size rangeof 2 to 30 microns, and on two species of algae (Phaeodactylumtricor-nutum and Nitzschia sp.) — Larvae graze most in the size range where the particleconcentration is highest. — If larvae deplete certain size categories of particlesthey then graze other size ranges in which the concentrationis still high. — The grazing rate of the two species varied between 988and 91.949 µm3 per pluteus per hour. — For A. lixula larvae the grazing rate increases withincreasing temperature to a maximum at 22°C.  相似文献

Vertical distributions and diel migrations of the main speciesof micronekton, four euphausiids, one mysid, one decapod andthree fishes, were described in detail in the 0–1000 mwater column on a fixed station in the Northwestern MediterraneanSea. The euphausiids Euphausia krohni and Thysanopoda aequalis,the decapod Gennadas elegans and, to a lesser extent, the fishArgyropelecus hemigymnus were shown to perform clear diel verticalmigrations. Results of horizontal hauls at a given depth aroundsunrise and sunset showed a marked diurnal symmetry of the migratorycycles, particularly for E.krohni, T.aequalis and G.elegans.The behaviour of the euphausiid Nematoscelis megalops was morecomplex: it presented a repetitive bimodal day distributionand only part of its population migrated. As very weak or non-migratorswe found the euphausiid Stylocheiron longicorne and the bathypelagicmysid Eucopia unguiculata, for which migration concerned onlysome of the older individuals. The fishes Cyclothone braueriand Cyclothone pygmaea appeared to be non-migrants. As depthincreased, C.braueri was replaced by C.pygmaea, with maximumconcentrations at 350–550 and 550–700 m depth, respectively.  相似文献

Excised winter buds of very hardy fir supercooled to —30or — 35?C, though primordial shoots excised from thesewinter buds (freezing point: about —5.5?C) supercooledonly to —12 to — 14?C. Also, excised primordialshoots did not tolerate freezing, but were rather resistantto desiccation. Differential thermal analysis (DTA) of primordialshoots revealed that the capability of supercooling increasedwith decreasing water content and that no exotherm could bedetected in the primordial shoots with a water content belowabout 20%. When excised whole buds were cooled very slowly,the exotherm temperature shifted markedly to a lower value andthe exotherm became much smaller. Also, masses of needle icewere observed, mainly beneath the crown of the primordial shoot.From these results, it may be concluded that most of the waterin primordial shoots gradually migrates out through the crownand freezes as the temperature decreases (extraorgan freezing),which enables primordial shoots to survive at very low temperatures.Winter buds of Abies balsamea held at — 20?C for 30 daysand then slowly cooled down to —50 or —60?C remainedalive. Thus, there seems to be no low temperature limit to thisfrost avoidance mechanism, if the primordial shoots can resistintensive freeze-dehydration. Two major ethanolamine phosphate-substituted inositol phosphosphingolipidshave been identified in the unsaponifiable acidic lipid fractionsof Tritrichomonas foetus and Trichomonas vaginalis. The compoundswere radiolabelled and purified by high-performance thin-layerchromatography followed by high-performance liquid chromatography.The structures were determined by a combination of tandem massspectrometry (MS/MS) and nuclear magnetic resonance (NMR) experiments,and gas—liquid chromatography of components obtained bydegradation and derivatization. Inositol in the T.foetus componentwas 1-linked to the phosphosphingolipid, had the phospho-ethanolaminegroup at the 3-position and a fucosyl residue at the 4-position.The T.vaginalis component lacked the fucosyl moiety. Both organismsalso produced inositol phosphosphingolipids having the samelong-chain base (sphingosine or dihydrosphingosine) and thesame fatty acyl distribution as the inositol diphosphate compounds.These glycosphingolipids may represent metabolic intermediatesfor new types of membrane anchors for surface glycopeptidesor glycolipids that mediate the host—parasite relationshipof these trichomonads. The distribution and abundance of gymnosome gastropods in theArgentine Sea and Brazil—Malvinas Confluence during 1978–1979and 1988 were studied. The collections analyzed included 768quantitative samples obtained between 48°W and the coast,and from 35°S to 55°S. Two species were found. Spongiobranchaeaaustralis was the most frequent and abundant (up to 730 per1000 m3); its presence in the area was associated with the coreof the Malvinas Current. Clione antarctica was less abundant(maximum abundance: 230 per 1000 m3) and was also associatedwith the Malvinas Current. The geographic ranges of both speciesin the area are wider than previously described. Since the rangeof S. australis in the area extends far from the range of itsprey Clio, it is not clear whether S. australis can feed onthe thecosomatous pteropod Limacina (and not only on Clio, asdescribed in the bibliography) or it starves in that area. Duringthe 1978–1979 annual cycle, the abundance of both speciesfollowed neither the abundance patterns of their prey nor ofthe total zooplankton, and differed from each other. The residencetime of swarms of both gymnosomes were shorter than one month.As a general pattern, the aggregates are rapidly transportednorthward by the Malvinas Current and also penetrate the outershelf water, but they remain there only during a short periodand cannot preclude the final expatriation. Published data on the biomass and specific respiration ratesof mesozooplankton in the oceans across all latitudes were combinedto assess their community respiration on a global basis. Mesozooplanktonbiomass was higher in boreal/anti-boreal and polar waters, intermediatein equatorial waters and lowest in the subtropical gyres. Specificrespiration rates were the highest in equatorial waters anddecreased rapidly poleward. Global community respiration ofmesozooplankton in the upper 200 m of the oceans integratedover all latitudes was 10.4 ± 3.7 (SE) Gt C year–1(n = 838). Below the epipelagic zone, mesozooplankton respirationliving in the mesopelagic (200–1000 m) and bathypelagic(below 1000 m) zones was estimated as 2.2 ± 0.4 (n =57) and 0.40 ± 0.2 (n = 12) Gt C year–1, respectively.Thus, global depth-integrated mesozooplankton respiration was13.0 ± 4.2 Gt C year–1 (17–32% of globalprimary production), which is 3–8-fold higher than thevalues assigned to mesozooplankton respiration in recent estimatesof total respiration in the ocean. Thus, it appears that mesozooplanktonrepresent a major, but neglected component of the carbon cyclein the ocean.  相似文献

NG, a strain of cultured tobacco cells of Nicotiana glutinosahad high growth rates and carboxylate contents (C—A) of100 to 130 meq/100 g of dry cells on media containing 42 meqNO3/liter as the sole N source. (C—A) is the amount ofinorganic cations minus inorganic anions in meq per 100 g ofdry cells. NG, cultured on media containing NH4 10+NO3 42 in meq/liter,had lower growth rates and lower (C—A) values as comparedwith NG on media containing NO3 as the sole N source. NG, cultured on media containing NH4 30+NO3 42 in meq/liter,had high growth rates and (A—C) values of 22 to 53 meq/100gof dry cells. In this case, the (A—C) content may correspondto organic cations, basic organic N compounds such as free asprotein-bound basic amino acids. The easily absorbed Cl mayhave been required maintain good growth conditions such as ionicbalance and a favorable pH in the cells. Thus cultured cells of Nicotiana glutinosa may have physiologicaladaptability against variations in a relatively wide range of|C—A| contents [|C—A| being the absolute valuesof (C—A)]. The human red blood cell sialoglycoprotein, glycophorin A (GpA),contains a ‘mucin-like’ extensively O-glycosylatedextracellular domain which carries the MN blood group antigens.We have revised the sites of O-glyccsylation in the extracellulardomain of GpA by automated solid-phase Edman degradation, whichallowed positive identification and quantitation of O-glycosylatedSer and Thr residues, as well as the single N-glycosylationsite. One N-linked and 16 O-linked sites were identified. Carbohydratewas absent on Ser 1, Ser14, Ser15, Ser23, Thr28 and Thr58 inGpA. We propose that the glycosyltransferases present in erythrocytesrecognize specific flanking sequences around potential O-glycosylationsites. All 16 O-glycosylation sites are explained on the basisof four motifs. Three motifs are associated with Thr-glycosylation:Xaa—Pro—Xaa—Xaa where at least one Xaa = Thr;Thr—Xaa—Xaa—Xaa where at least one Xaa = Thr;Xaa—Xaa—Thr—Xaa where at least one X = Argor Lys. When tea plants were shaded with black lawn cloth for severaldays in the field, the accumulations of (—)-epicatechin,(—)-epicatechin-3-gallate, (—)-epigallocatechinand (—)-epigallocatechin-3-gallate decreased in newlydeveloping tea shoots. Bittium reticulatum and Barleeia unifasciata are two of themost abundant microgastropods inhabiting the alga Gelidium latifoliumin the North of Spain. Bittium reticulatum produces one cohortannually, born in August, and living over two years. Barleeiaunifasciata shows two population groups, each one formed bytwo cohorts. There are important differences in density andgrowth rate between the two groups. The two species are separatedspatially on Gelidium. Bittium reticulatum prefers to live atthe base of the plant and Barleeia unifasciata prefers the upperpart. There are different demographic patterns in each population.This fact supports the concept of distribution in compartmentsrelative to intertidal systems. SYNOPSIS. Development is not a gradual but a saltatory process.A combination of qualitative changes in form and function—thresholds—createsboundaries between a succession of quantitative intervals—steps.Thresholds can be modified by an altered time of appearanceof structures and functions (heterochrony), especially duringearly ontogeny, to form an operational basis for the prolongationof juvenile characters and adaptability into later ontogeny.Whereas such prolongation enables juvenilization in phylogeny,analogous principles may operate on a much shorter time scaleto produce the r-selection-like altricial and the K-selection-likeprecocial trends in ontogeny. The inherited capacity to adjustconstantly to the environment (heterochrony) selects for structural,biochemical and behavioral improvements. The tendency is towardthe precocial but the way back to altricial forms is left open.The heterochronous adaptations in early ontogeny can "reverse"the vulnerable specialization, should the environment becomeless stable and/or the community less competitive. Juvenilization,capable of turning gerontomorphosis into paedomorphosis in evolution,and heterochronous shifts of character anlagen, capable of turninga precocial trend into an altricial trend in ontogeny are bothpart of the same biological process which operates during earlyontogeny.  相似文献

Tropical high Andes plants may be exposed to sub-zero temperaturesany night of the year. These plants have to rely on mechanismswhich protect them from these environmental conditions but atthe same time allow their growth and development. Supercoolinghas been found to be the principal avoidance mechanism in leavesof the caulescent giant rosette genus Espeletia in the Andes.We report here the differences in supercooling capacity andcold injury in several Espeletia schultzii populations growingalong an altitudinal gradient. The relationships between supercooling,water potential and leaf anatomy were also investigated. Thesupercooling capacity increased and injury temperature decreasedfrom lower to higher elevation populations. These changes maybe explained in terms of physiological, morphological and anatomicalcharacteristics of the leaves. The herbivorous and carnivorous feeding biology of Tropocyclopsprasinus mexicanus, an especially small cyclopoid copepod, wasstudied under in situ food conditions in three different lakesand under experimentally augmented food supplies. The mass-specificfood uptake is compared to that of two larger species—Cyclopskolensis and Cyclops vicinus. Under in situ food conditions,T.p.mexicanus depended to a larger extent on algae than invertebrateprey and showed lower mass-specific ingestion rates than thetwo larger species. Daily mass-specific uptake rates for algaeranged between 10 and 24% of its body mass versus 0.7–7%for invertebrate prey. The larger species C.vicinus and C.kolensisingested a total dry mass equivalent to 106% or up to 143% oftheir body mass with algae contributing 66 and 81%. However,under enriched food availability, T.p.mexicanus is able to ingesta biomass equivalent to its body mass, with an algae (54%) andprey (40%) portion similar to that of the larger species. Bodysize appears to be an important factor for the relative importanceof algal versus invertebrate prey for cyclopoid copepods.  相似文献

Floral initiation and development of four contrasting whiteclover (Trifolium repens) populations was examined after differentinduction treatments (16 h, 5 ?C and 8 h, 5 ?C. The number of reproductive stolons and of reproductive budsper stolon was increased after cold induction. Varietal differencesin response to daylength were large; some varieties respondingbetter to a long day cold period, others to a short day coldperiod while one variety required no induction at all. Whetherthe daylength effect was due to photoperiod, irradiance or totheir interaction was not known. The induction periods had a subsequent effect upon pedunclelength, floret and ovule number. Short days and chilling reducedpeduncle length but increased ovule number, whereas long daysand chilling tended to increase floret number. Nectar concentrationwas highest after short day induction. Soil water resulting from episodic growing season rainfall evaporatesrapidly in semi-arid regions. Plants may mnot benefit from suchwater additions if near-surface roots are unable to resume wateruptake rapidly following periods of soil water deficit. Ourobjectives were to develop a means of quantifying root uptakeresponses in the upper soil layer following rewetting aftersoil water deficit, and to evaluate the existence of genotypicdifferences among four diverse barley (Hordeum vulgare L.) genotypesin this regard. Plants were grown in replicate soil columnshaving hydraulically isolated surface and subsoil layers, andinstrumented with time-domain reflectometry (TDR) waveguides.The upper 0.05 m soil layer was allowed to dry to —1.8to —3.0 MPa for 10-14 d, during which time subsoil wetnesswas maintained at about —0.6 to —0.7 MPa. The time-courseof soil water uptake was monitored at 0.5 h intervals followingrewetting of the surface layer. Substantial water uptake began1 d after rewetting following 10 d, and 2-3 d after rewettingfollowing 14 d of water deficit. Rate of water uptake was morerapid in response to a second rewetting 5-7 d later. Consistentgenotypic responses in terms of cumulative water uptake on awhole plant and leaf area-specific basis were observed duringeach trial. A comparison between bat-pollinated plant assemblages at twosites in different altitudinal ranges covered by the Atlanticrainforest in southeastern Brazil is presented. The lowlands(5–90 m) harbour ten plant species in seven families,pollinated mostly by three glossophagine bat species. The highlands(1540–1600 m) harbour seven plant species in five families,pollinated by a single species of glossophagine. Bromeliadsaccount for about 30% of the bat-pollinated species at eachassemblage. Tube and brush shapes prevail in the flowers atboth sites. Corolla lengths averaged 26.8 and 37.2 mm, sugarconcentrations in nectar averaged 15.0 and 18.1%, and nectarvolumes averaged 150.8 and 167.0 µl in the lowland andhighland assemblages, respectively. The flowers are pollinatedmostly by glossophagine bats during hovering visits, and theplants are visited in the trap-line foraging pattern. The plantsbloom annually and both assemblages show a staggered continualflowering pattern. Flowering seasonality was found at both sites,with flowering clustering in the drier season at the lowlandsite and in the wetter season at the highland site. The ratiobetween bat-pollinated and hummingbird-pollinated species is0.24 at the lowland site and 0.23 at the highland one. Seeds of the Smooth-leafed Elm (Uimus carpinifolia) and of thetropical forest tree Terb (Terminalia brassii) were stored hermeticallyand sampled at intervals for periods of up to two years. Bothspecies possess ‘orthodox’ seed (increasing longevityis observed as either moisture content or temperature are reduced)within the temperature ranges from — 13 to 52°C (Elm)and from —4 to 42°C (Terb) and within the moisturecontent ranges from 3 to 19 per cent (Elm) and from 5 to 14per cent (Terb) on a fresh weight basis. Elm seed stored at—75°C showed the expected relationship between longevityand moisture content, but did not differ significantly in longevityfrom seed kept at — 13°C when moisture contents wereheld constant. Probit analysis of the relationship between germinationpercentage and time was performed for each storage environment,yielding a slope from which the standard deviation of the distributionof seed deaths over time () was calculated. Standard deviationvalues were used in turn to determine the values of constantsin a viability equation which had previously been applied toseed of barley, chickpea, cowpea and soybean. The equation,which gave a good fit to the results obtained, can be used topredict viability for seed in storage over a wide range of environmentalconditions. Some limitations to the applicability of the viability equationwere defined. At 22 per cent and higher moisture contents Elmseed survived longer than predicted. Furthermore, all Elm andTerb seed was killed quickly on placing in —75°C at22 and 20 per cent moisture content respectively, but high viabilitywas retained for several days at 19 and 17 per cent respectively.Practical implications of the results are discussed. A population of Pseudotachea litturata (Pfeiffer, 1851) fromTarifa (Càdiz, Spain) has been studied. The morphologicalresults are compared with those from P. splendida, Iberus gualtierianus,I. alonensis, I. marmoralus, I. guiraoanus and four speciesof the genus Cepaea using, as an exploratory method, the Wagnerparsimony procedure and 18 characters of the shell, genitalsystem and karyotype have been analysed. According to this methodit seems that the taxonomical position of P. litturata in thegenus Pseudotachea is confirmed, and agrees with the phylogeneticalrelationships in this group of species. The genus Cepaea seemsto be well established, although two species groups can be distinguished:C. nemoralis—C. hortensis and C. syluatica—C. vindobonensis.These differ mainly in chromosome number, diverticulum lengthand degree of shell polymorphism. Primary production, and bacterial production as measured byincorporation of [3H-methyl]thymidineinto ice cold TCA insolublematerial were investigated during 1984 in Lake Kvernavatnet,west Norway. Primary production averaged 222 mg C m–2day–1 and bacterial production averaged 163 mg C m–2day–1. The bacterial production in the euphotic pelagiczonecontributed -60% of the total pelagic bacterial production.The zooplankton was dominated byDaphnia longispina. From growthexperiments with animals fed only natural food in coarse filteredlake water, the population daily growth increments were calculated.The average production of D.longispina was 151 mg C m–2day–1 during the period investigated. The estimated primaryproduction was too low to sustain both the bacterial productionand the zooplankton food requirements. These results imply thatthe carbon cycle of the lake is dependent on the supply of allochtonousmaterial, or that the current methods for measuring productionrates in aquatic environments are systematical erratic.  相似文献
