Membrane bioreactor for drinking water denitrification   总被引:1,自引:0,他引:1  
The aim of this study is to evaluate the performance of a membrane bioreactor with cell recycle to be used for drinking water denitrification, when operated with a high nitrate load (up to 7.68?kgNO3 ?/m3?day) and low hydraulic retention time (down to 0.625?h). Nitrate and nitrite were always completely removed for all the operational conditions used. The effluent's nitrite concentration kept below 0.1?mg NO2 ?/l with exception of a short period, during the reactor start-up, when it accumulates. The performance of the membrane bioreactor was also evaluated using a groundwater containing 148?mg NO3 ?/l. Nitrate and nitrite concentration in the effluent were below the recommended values for drinking water when the reactor was controlled at pH 7.0. The membrane flux decreases during operation as a consequence of membrane fouling. The flux decrease was more severe during operation with synthetic medium than with contaminated groundwater due to the existence of molecular complexes in the synthetic broth. A backshock technique was used to reduce the surface fouling of the membrane. Combining this technique with the use of a reserve asymmetric structured membrane it was found that the membrane flux remains nearly unchanged.  相似文献

Biotreatment of petroleum-contaminated water in a fluidized-bed bioreactor (FBB) is a relatively new and promising technology, which efficiency is strongly affected by the biocatalyst used. Our laboratory experiments involved biotreatment of the water contaminated with a synthetic petroleum mixture consisting of aliphatic and polyaromatic hydrocarbons (PAHs) using a continuous column bioreactor with recycle. Different type biocatalysts were tested, including Rhodococcus bacteria immobilized in hydrophobized carriers such as sawdust, poly(vinyl alcohol) cryogel (cryoPVA) and poly(acrylamide) cryogel (cryoPAAG). Biocatalyst abilities to oxidize petroleum hydrocarbons were evaluated using the Columbus Micro-Oxymax® respirometer. The hydrophobized sawdust-supported biocatalyst demonstrated substantially higher metabolic activity than C12-cryoPAAG-based biocatalyst due to larger number of immobilized Rhodococcus cells and, therefore, had benefits for application in FBBs. The obtained results showed that designed FBB process is successful, providing 70–100% removal of n-alkanes (C10–C19) and 66–70% removal of 2–3-ring PAHs from contaminated water after 2–3 weeks.  相似文献

Soil water resource, together with the surface and sub-surface water resource, is essential to the regional water balance and world water cycle. A total of 90 soil samples were collected from 30 different soil profiles of dry fields throughout Chongqing, China randomly to show how soil could be a crucial part of water resources by discussing their five types of calculated soil water reservoir capacities, namely the total soil water reservoir capacity (mm) (TC), soil water storage capacity (mm) (SC), unavailable soil water reservoir capacity (mm) (UC), available soil water reservoir capacity (mm) (AC), and soil dead water storage capacity (mm) (DC) in certain layer, respectively. Overall, the total soil water reservoir capacity in 0–40 cm was about 209 mm, of which 70 mm belonged to available soil water reservoir capacity. Not all the five types of soil water reservoir capacities had significant correlations between each other. Soil structure, especially the size and quantity of soil pore was mainly determined by soil particle composition (clay, silt, and sand content). The more sand and less clay led to the more soil macropores, which provided room for soil water. Thus, clay, silt, and sand content jointly produced profound influence on soil water reservoir capacities. Nevertheless, specific water capacity and topographic factors displayed weak correlations to soil water reservoir capacities, which required further research works. Ultimately, the better regression models were achieved by multiple regression analysis coupled with “merged groups PCA” than by multiple regression analysis with “all variables PCA”. UC, SC, TC and DC could be well simulated (mostly R2 > 0.70; P < 0.05) through normal multiple regression analysis using original variables as well as multiple regression analysis with “merged groups PCA”. Only regression models of TC and DC were highly significant (mostly R2 > 0.70; P < 0.05) through “all variables PCA” method. And there were poor coefficients of determination (R2) for AC (mostly R2 < 0.40; P < 0.05) by all the three regression methods.  相似文献

We investigated the diversity and distribution of fungi in nine different sites inside 30 residential dishwashers. In total, 503 fungal strains were isolated, which belong to 10 genera and 84 species. Irrespective of the sampled site, 83% of the dishwashers were positive for fungi. The most frequent opportunistic pathogenic species were Exophiala dermatitidis, Candida parapsilosis sensu stricto, Exophiala phaeomuriformis, Fusarium dimerum, and the Saprochaete/Magnusiomyces clade. The black yeast E. dermatitidis was detected in 47% of the dishwashers, primarily at the dishwasher rubber seals, at up to 106 CFU/cm2; the other fungi detected were in the range of 102 to 105 CFU/cm2. The other most heavily contaminated dishwasher sites were side nozzles, doors and drains. Only F. dimerum was isolated from washed dishes, while dishwasher waste water contained E. dermatitidis, Exophiala oligosperma and Sarocladium killiense. Plumbing systems supplying water to household appliances represent the most probable route for contamination of dishwashers, as the fungi that represented the core dishwasher mycobiota were also detected in the tap water. Hot aerosols from dishwashers contained the human opportunistic yeast C. parapsilosis, Rhodotorula mucilaginosa and E. dermatitidis (as well as common air-borne genera such as Aspergillus, Penicillium, Trichoderma and Cladosporium). Comparison of fungal contamination of kitchens without and with dishwashers revealed that virtually all were contaminated with fungi. In both cases, the most contaminated sites were the kitchen drain and the dish drying rack. The most important difference was higher prevalence of black yeasts (E. dermatitidis in particular) in kitchens with dishwashers. In kitchens without dishwashers, C. parapsilosis strongly prevailed with negligible occurrence of E. dermatitidis. F. dimerum was isolated only from kitchens with dishwashers, while Saprochaete/Magnusiomyces isolates were only found within dishwashers. We conclude that dishwashers represent a reservoir of enriched opportunistic pathogenic species that can spread from the dishwasher into the indoor biome.  相似文献

Unoperated, sham-operated (SO) and allatectomized (CA) adult, male Blaberus discoidalis lost ca. 300 mg of live weight (LW) during the first 10 days after ecdysis. This decrease resulted from losses in both dry weight (DW) and water. An additional 100 mg LW loss was found in allatectomized-cardiacectomized (CA+CC) animals as a result of increased water loss.The LWs of operated and unoperated controls remained nearly constant between 10 and 30 days of age as DW increased and water decreased in comparable amounts. However, the LW of 30-day-old CA+CC animals was 100 mg greater than the controls due to increased water retention.Compared to controls, food consumption and faecal production were reduced by 50 per cent in CA and CA+CCanimals, but the percentage of food digested was unchanged. Efficiency of converting food to DW in CA+CCanimals was twice that of SO and CA animals and four times that of the unoperated controls.These results indicate that extirpation of the retrocerebral complex decreases food intake but not digestion. Nutrient storage or conversion to body matter is apparently enhanced by neuroendocrine deficiency. Therefore, a gross nutritional deficiency is unlikely in CA+CCanimals.The LW changes occurring after neuroendocrine deficiency were related to changes in water content. DW changes were comparable in all control and experimental groups. The results indicate both diuretic and antidiuretic effects by the neuroendocrine system depending on the age of the animals.  相似文献

The distribution of culturable hyperthermophiles was studied in relation to environmental conditions in the Kubiki oil reservoir in Japan, where the temperature was between 50 and 58°C. Dominant hyperthermophilic cocci and rods were isolated and shown to belong to the genera Thermococcus and Thermotoga, respectively, by 16S rDNA analyses. Using the most-probable-number method, we found that hyperthermophilic cocci were widely distributed in several unconnected fault blocks in the Kubiki oil reservoir. In 1996 to 1997, their populations in the production waters from oil wells were 9.2 × 103 to 4.6 × 104 cells/ml, or 10 to 42% of total cocci. On the other hand, hyperthermophilic rods were found in only one fault block of the reservoir with populations less than 10 cells/ml. Dominant Thermococcus and Thermotoga spp. grew at reservoir temperatures and utilized amino acids and sugars, respectively, as sole carbon sources. While organic carbon was plentiful in the environment, these hyperthermophiles were unable to grow in the formation water due to lack of essential nutrients. Concentrations of some organic and inorganic substances differed among fault blocks, indicating that the movement of formation water between fault blocks was restricted. This finding suggests that the supply of nutrients via fluid current is limited in this subterranean environment and that the organisms are starved in the oil reservoir. Under starved conditions at 50°C, culturable cells of Thermococcus sp. remained around the initial cell density for about 200 days, while those of Thermotoga sp. decreased exponentially to 0.01% of the initial cell density after incubation for the same period. The difference in survivability between these two hyperthermophiles seems to reflect their populations in the fault blocks. These results indicate that hyperthermophilic cocci and rods adapt to the subterranean environment of the Kubiki oil reservoir by developing an ability to survive under starved conditions.  相似文献

Hourly drift samples were collected once each month over a 24 hour period for 10 months at the mouth of a channel returning heated coolant water from a steam electric generating station to a 330 ha reservoir. Most benthic macroinvertebrates existed at lower densities on the bottom of the channel than in the reservoir. No organisms were found in the channel during summer months when water temperatures reached 42.2°C. The most abundant organisms drifting from the channel were Chaoborus larvae which probably were drawn from the reservoir, through the condensors of the power plant, and down the effluent channel. The maixmum drift rate for Chaoborus larvae occured in August (8.91 × 104 mg/day) while the minimum rate occurred in December (3.92 × 102 mg/day). Chaoborus larvae exhibited a diurnal periodicity in drift corresponding to their nightly emergence from bottom sediments in the reservoir.  相似文献

In Morocco, storage reservoirs are particular systems of water supply in rural areas. These reservoirs are fed with rainwater and/or directly from the river, which are very contaminated by several pathogenic bacteria. They are used without any treatment as a drinking water by the surrounding population. In this context, the aim of this study is to evaluate the impact of consuming contaminated water stored in reservoirs on health status for six rural communities located in Assif El Mal, Southern East of Marrakech. This was investigated using a classical methodology based on population survey and by molecular approach using PCR–DGGE technique to determine the intestinal bacterial diversity of consumers. The survey showed that, the residents of the studied area suffered from numerous health problems (diarrheal diseases, vomiting or hepatitis A) due to the lack of waste management infrastructures. The consumer’s stool analysis by molecular approach revealed that numbers of Escherichia coli, Aeromonas hydrophila and Clostridia, were significantly higher in the diarrheal feces. In addition, PCR–DGGE study of the prevalence and distribution of bacteria causing human diseases, confirmed that, there is a relationship between water bacterial contaminations of storage reservoirs and microbial disease related health status. Therefore, water reservoir consumption is assumed to be the mean way of exposure for this population.It’s clear that this approach gives a very helpful tool to confirm without any doubt the relationship between water bacterial contamination and health status.  相似文献

Although the occurrence of intracellular glasses in seeds and pollen has been established, physical properties such as rotational correlation times and viscosity have not been studied extensively. Using electron paramagnetic resonance spectroscopy, we examined changes in the molecular mobility of the hydrophilic nitroxide spin probe 3-carboxy-proxyl during melting of intracellular glasses in axes of pea (Pisum sativum L.) seeds and cattail (Typha latifolia L.) pollen. The rotational correlation time of the spin probe in intracellular glasses of both organisms was approximately 10−3 s. Using the distance between the outer extrema of the electron paramagnetic resonance spectrum (2Azz) as a measure of molecular mobility, we found a sharp increase in mobility at a definite temperature during heating. This temperature increased with decreasing water content of the samples. Differential scanning calorimetry data on these samples indicated that this sharp increase corresponded to melting of the glassy matrix. Molecular mobility was found to be inversely correlated with storage stability. With decreasing water content, the molecular mobility reached a minimum, and increased again at very low water content. Minimum mobility and maximum storage stability occurred at a similar water content. This correlation suggests that storage stability might be at least partially controlled by molecular mobility. At low temperatures, when storage longevity cannot be determined on a realistic time scale, 2Azz measurements can provide an estimate of the optimum storage conditions.  相似文献

Fresh produce is known to carry nonpathogenic epiphytic microorganisms. During agricultural production and harvesting, leafy greens can become contaminated with antibiotic-resistant pathogens or commensals from animal and human sources. As lettuce does not undergo any inactivation or preservation treatment during processing, consumers may be exposed directly to all of the (resistant) bacteria present. In this study, we investigated whether lettuce or its production environment (irrigation water, soil) is able to act as a vector or reservoir of antimicrobial-resistant Escherichia coli. Over a 1-year period, eight lettuce farms were visited multiple times and 738 samples, including lettuce seedlings (leaves and soil), soil, irrigation water, and lettuce leaves were collected. From these samples, 473 isolates of Escherichia coli were obtained and tested for resistance to 14 antimicrobials. Fifty-four isolates (11.4%) were resistant to one or more antimicrobials. The highest resistance rate was observed for ampicillin (7%), followed by cephalothin, amoxicillin-clavulanic acid, tetracycline, trimethoprim, and streptomycin, with resistance rates between 4.4 and 3.6%. No resistance to amikacin, ciprofloxacin, gentamicin, or kanamycin was observed. One isolate was resistant to cefotaxime. Among the multiresistant isolates (n = 37), ampicillin and cephalothin showed the highest resistance rates, at 76 and 52%, respectively. E. coli isolates from lettuce showed higher resistance rates than E. coli isolates obtained from soil or irrigation water samples. When the presence of resistance in E. coli isolates from lettuce production sites and their resistance patterns were compared with the profiles of animal-derived E. coli strains, they were found to be the most comparable with what is found in the cattle reservoir. This may suggest that cattle are a potential reservoir of antimicrobial-resistant E. coli strains in plant primary production.  相似文献

Three different types of compost, PM-5 (poultry manure compost), 338 (dairy cattle manure compost), and NVIRO-4 (alkaline-pH-stabilized dairy cattle manure compost), and irrigation water were inoculated with an avirulent strain of Salmonella enterica serovar Typhimurium at 107 CFU g−1 and 105 CFU ml−1, respectively, to determine the persistence of salmonellae in soils containing these composts, in irrigation water, and also on carrots and radishes grown in these contaminated soils. A split-plot block design plan was used for each crop, with five treatments (one without compost, three with each of the three composts, and one without compost but with contaminated water applied) and five replicates for a total of 25 plots for each crop, with each plot measuring 1.8 × 4.6 m. Salmonellae persisted for an extended period of time, with the bacteria surviving in soil samples for 203 to 231 days, and were detected after seeds were sown for 84 and 203 days on radishes and carrots, respectively. Salmonella survival was greatest in soil amended with poultry compost and least in soil containing alkaline-pH-stabilized dairy cattle manure compost. Survival profiles of Salmonella on vegetables and soil samples contaminated by irrigation water were similar to those observed when contamination occurred through compost. Hence, both contaminated manure compost and irrigation water can play an important role in contaminating soil and root vegetables with salmonellae for several months.  相似文献

The aim of the present study was to focus on the impact of two different methods and the effects of cryoprotectants on the survival of a probiotic bacterium, Streptococcus phocae PI80, during storage. For the protection of freeze dried cells, the optimal storage conditions were determined with a high survival rate. After the freeze drying process, all cryoprotectants exhibited a protective effect on cell viability at all storage temperatures. High relative cell viability was observed when cells were incubated at ?20°C, which was optimum for the protection of S. phocae PI80. Trehalose was the most promising cryoprotectant at all temperatures during the storage period of bacterial cells. The combination of trehalose + skim milk showed more than 85% survivability compared to other combinations at ?20°C for 60 days. In addition, encapsulation of probiotic cells into alginate-chitosan gel capsules showed better survival of S. phocae cells (5.468 ± 0.15 LogCFU/mL) with high bacteriocin activity at ?20°C for six months. The cell-loaded microcapsules remained stable when treated with simulated gastric and intestinal fluids. After 6 h in vivo treatment, the capsules were found to be broken, releasing the probiotic cells directly into the intestinal system of rats. Therefore, microencapsulation was found to be the most efficient technique, which not only protected the cells for a longer time but also released the cells into the in vivo intestinal system.  相似文献

The filamentous nitrogen-fixing cyanobacterium, Anabaena sp. strain PCC 7120 was found to tolerate very high doses of 60Co-gamma radiation or prolonged desiccation. Post-stress, cells remained intact and revived all the vital functions. A remarkable capacity to repair highly disintegrated genome and recycle the damaged proteome appeared to underlie such high radioresistance and desiccation tolerance. The close similarity observed between the cellular response to irradiation or desiccation stress lends strong support to the notion that tolerance to these stresses may involve similar mechanisms.  相似文献

Disturbances, such as rainfall and hydrological management of Vouglans reservoir, seem to influence the establishment of the ecological system inside this man-made lake. In this paper, the main effect of these disturbances are changes in the zmix:zeu ratio, with a shift in the composition and phytoplankton dynamics, and consequently in the Shannon's diversity. The seasonal changes found in the surface layers of Vouglans reservoir were typical for mesotrophic and temperate lakes, but some perturbations induced the ‘reversion’ to an earlier stage in the expected successional pattern during the stratification period. This early-state displayed an intensive proliferation of the filamentous ulotrichal Planctonema lauterbornii, which remained dominant in terms of abundance from the middle to the end of the stratification. The beginning of this period was characterized by the highest value of Shannon diversity index, followed by its marked decrease, whilst zmix:zeu remained around 1. Despite the environmental variations which induced the reversion in this expected pattern, we found some sort of equilibrium, that is, stability in the assemblage simply expressed as a ‘no-change situation’, although not in the sense of equilibrium inside a competitive system. Moreover, peculiar freshwater red tides occurred in this reservoir involving Rhodomonas lacustris and Peridiniopsis durandi, whose dynamical events were also influenced by external forces.  相似文献

Aims

To determine the fate of Escherichia coli on vegetables that were processed through commercial wash treatments and stored under simulated retail conditions at 4°C or wholesale at fluctuating ambient temperatures (0–25°C, dependent on season).

Methods and Results

Bovine slurry that was naturally contaminated with E. coli O145 was applied without dilution or diluted 1:10 using borehole water to growing potatoes, leeks or carrots. Manure was applied 1 week prior to harvest to simulate a near‐harvest contamination event by manure deposition or an application of contaminated water to simulate a flooding event or irrigation from a contaminated water source. At harvest, crops were contaminated at up to 2 log cfu g?1. Washing transferred E. coli into the water of a flotation tank used for potato washing and did not completely remove all traces of contamination from the crop. Manure‐contaminated potatoes were observed to contain 0·72 cfu E. coli O145 g?1 after processing and retail storage. Manure‐contaminated leeks harboured 0·73–1·55 cfu E. coli O145 g?1 after washing and storage. There was no cross‐contamination when leeks were spray washed. Washing in an abrasive drum resulted in less than perfect decontamination for manure‐contaminated carrots. There were five post‐distribution isolations from carrots irrigated with contaminated water 24 h prior to harvest.

Conclusions

Standard commercial washing and distribution conditions may be insufficient to reliably control human pathogenic E. coli on fresh produce.

Significance and Impact

Previous speculation that the cause of a UK foodborne disease outbreak was soil from imperfectly cleaned vegetables is plausible.  相似文献

The Macau storage reservoir (MSR) has experienced algal blooms in recent years, with high levels of Cylindrospermopsis and Microcystis and detectable concentrations of cyanotoxins. To analyze the cyanotoxin-producing genotypes and relate the corresponding cyanotoxins to the water quality parameters, a quantitative real-time polymerase chain reaction was developed and applied to the water samples in three locations of MSR. Cylindrospermopsin polyketide synthetase (pks) gene and a series of microcystin synthetase (mcy) genes were used for identifying and quantifying cylindrospermopsin- and microcystin-producing genes, and the corresponding water parameters were measured accordingly. Our results showed that high concentrations of cylindrospermopsin and low concentrations of microcystin were measured during the study period. There was a strong correlation between the pks gene numbers and cylindrospermopsin concentrations (R 2 = 0.95), while weak correlations were obtained between the mcy genes numbers and microcystin concentrations. Furthermore, the pks gene numbers were strongly related to Cylindrospermopsis (R 2 = 0.88), cyanobacterial cell numbers (R 2 = 0.96), total algae numbers (R 2 = 0.95), and chlorophyll-a concentrations (R 2 = 0.83), consistent with the dominant species of Cylindrospermopsis among the cyanobacteria existing in MSR. NH4–N (R 2 = 0.68) and pH (R 2 = 0.89) were the water quality parameters most highly correlated with the pks gene numbers. These results contribute to monitoring for potential cyanotoxins in raw water.  相似文献

The balance equations to a continuous plug flow recycle reactor performing a protease-mediated autocatalytic reaction were analytically developed under the assumption of a Michaelis-Menten type of mechanism. The necessary condition for the recycle ratio that leads to the minimum reactor volume was set up and the resulting expression was numerically solved for the optimum ratio of recycle, R opt, as a function of the desired conversion of reactant (or pro-enzyme) into product (or enzyme), X PE, for selected values of the relevant dimensionless parameters, i.e. the ratio of total concentrations of enzyme and product moieties, γ, and the ratio of the Michaelis-Menten parameter to the total concentration of product moiety, K m *. It was numerically proven that variation of K m * by five orders of magnitude centered around unity did not affect R opt to a significant extent, whereas an empirical expression for R opt versus γ and X PE could be found and successfully fitted to the results of several numerical runs.  相似文献

The time course of the concentration of radiocalcium was studied in the serum, skeleton, pelt, muscles, and pooled internal organs of 10-day-old rats. Within 10 hours of injection, the specific activity of the tissue groups exceeded the specific activity of the serum and remained above it during the period studied (120 hours). Chemical and autoradiographic analyses showed how rapidly most of the injected Ca45 found its way into the skeleton. A model was constructed with the assumption that the skeleton constitutes an essentially irreversible reservoir for the tracer in a multicompartment system in which the blood is the central or feeding compartment. The rate of transfer of the tracer from the soft tissue compartments to the serum was calculated from the equation See PDF for Equation in which C = concentration in serum (expressed as a series of exponential terms) C' = concentration in a given soft tissue Substitution in the integrated form of this equation yielded equations which had the major properties of the empirical equations fitted to the experimental points. As a microbial-environmental-control-type deodorizing system, we have developed a compact biotrickling filter system for small-scale livestock farms. The performance of the compact co-current biotrickling filter operated at high recycle liquid flow rates was systematically examined. In particular, we studied improvements in the nitrification ability of the system due to the resultant enhancement of absorption and dissolution of NH3 and absorption of O2 with the high flow rates of recycle liquid flowing downward co-currently with gas flow. At the empty bed residence time of 50 s, almost complete removal of NH3 was obtained with recycle liquid flow rates of 103 and 205 L m−3 day−1 for 20 days while the inlet NH3 concentration was increased from 200 to 500 ppm. With a recycle liquid flow rate of 411 L m−3 day−1 the removal efficiency remained above 95% for 57 days while the inlet NH3 concentration was increased from 200 to 700 ppm. The biodegradation kinetics for NH3 removal was successfully analyzed using the Haldane substrate inhibition kinetics. The present data and kinetic analyses showed that the substrate inhibition was suppressed and the biodegradation of ammonia in the compact biotrickling filter could be improved by the high recycle liquid flow rate.  相似文献
