The ultrastructural features of embryos were studied from mature dry and soaked seeds of the parasitic angiospermCuscuta japonica. Outer tangential walls in the protoderm cells were thickened and covered by a thin cuticle layer. These walls could play important roles in preventing water loss from theCuscuta seedling surfaces after germination and in strengthening the surfaces against various environmental stresses. In the protoderm cells of soaked embryos, lipid materials were released into the thick outer walls through the fusion of lipid bodies with the plasma membrane. In the dry embryos were stored a large number of protein bodies with globoid crystals and lipid bodies. Numerous lipid bodies also were aligned under the plasma membrane. In both dry and soaked embryos, protein bodies were digested and transformed into small vacuoles. The degraded reserves of the lipid and protein bodies could then be mobilized to nourish subsequent germination and seedling growth. Proplastids in the soaked embryo cells contained a few thylakoids and electron-dense plastoglobuli, and crystallized phytoferritin. The phytoferritin, an iron-protein complex, would also be utilized in chloroplast development for autotrophic seedling growth.  相似文献

Summary The pollination biology of a group of European orchids (Dactylorhiza, Ophrys, Orchis, Platanthera, Goodyera, andSerapias species) are investigated, and their anthecological characteristics considered in relation to natural levels of reproductive success. Pollen ovule (P O) ratios of the European orchids surveyed range from 10 1 (Goodyera repens) to 24 1 (Platanthera chlorantha). Average pollen-load ovule ratios are consistently lower than P O ratios. Naturally occurring pollen loads range from 1 massula to >1 pollinium. Even the smallest pollen load is sufficient to stimulate embryogenesis in experimentally pollinatedDactylorhiza purpurella flowers, although more seeds are set with larger loads. Pollen tubes grow rapidly through the stylar canal and into the top of the ovary within 2 or 3 days of pollination, and grow down either side of the 3 parietal ridges in the ovary. Fertilisation occurs throughout the length of the ovary but its distribution is non-random, especially when pollen loads are limiting, with more seeds being set at the stylar end. All species ofDactylorhiza, Ophrys, andOrchis studied are highly self-compatible. In the absence of pollination,Ophrys andOrchis flowers remain open and fertile for at least 3 weeks. Pollinated flowers remain receptive to further pollinations for at least 8 days. Some fruits can even be obtained on selfing 20-day-old unpollinatedOrchis morio flowers. Excised pollinia retain germinability for a long time, up to 51 days inDactylorhiza purpurella. The arrival of pollen on the stigma hastens floral senescence, but post-pollination changes are relatively slow when compared with those reported for tropical orchid species. It is concluded that characteristics of the pollination biology of European orchids act to maximise reproductive success by (1) prolonging the opportunity for effective pollen deposition both pre- and post-pollination, (2) increasing the likelihood of widespread dispersal, (3) reducing pollen wastage, and (4) increasing seed quality by promoting some pollen competition. As most European orchids are xenogamous and require pollen to arrive on the stigma before seed can be set, reproductive maximisation is of particular adaptive advantage because many of them are infrequently visited by insects so that the probability of successful pollination can be very low.Abbreviations P O pollen ovule ratio - PL O pollen-load ovule ratio - FCR fluorochromatic reaction test  相似文献

 The fatty acid compositions of seed oils from 34 conifer species, mainly Pinaceae and secondarily Cupressaceae, have been determined by gas-liquid chromatography of the methyl esters. As noted in earlier studies, these oils were characterized by the presence of several Δ5-olefinic acids, i.e., 5,9-18:2, 5,9,12-18:3, 5,9,12,15-18:4, 5,11-20:2, 5,11,14-20:3, and 5,11,14,17-20:4 acids, in addition to the more common saturated, oleic, linoleic and α-linolenic acids. Based on these fatty acid compositions, and on those established in earlier systematic studies (totalling 82 species), we established a chemotaxonomic grouping of the main conifer families, i.e., of the Pinaceae, Taxodiaceae, Cupressaceae, and Taxaceae. This was achieved using multivariate analyses (principal component analysis and discriminant analysis). The fatty acids that discriminate best in this classification are the 5,11,14,17-20:4, 9,12,15-18:3 and 5,9,12-18:3 acids. Moreover, it was possible to differentiate between several genera of the Pinaceae: Pinus (including Tsuga and Pseudotsuga), Abies, Cedrus, and Picea plus Larix, represented quite distinct groups. Other fatty acids such as oleic, linoleic, and 5,9-18:2 acids were also important for this purpose. The fatty acid compositions, and particularly the Δ5-olefinic acid contents of conifer seed oils, may thus be applied to the chemosystematic distinction among conifer families as well as genera of the Pinaceae. Abstract. Phylogenetic relationships amongst Megastigmus species (Chalcidoidea: Torymidae) associated with conifer seeds were inferred from DNA sequence data. Twenty‐nine species of seed chalcids were analysed using two different genes, cytochrome b (mitochondrial DNA) and the D2 domain of the 28S ribosomal DNA. Maximum‐parsimony and maximum‐likelihood analyses showed that taxa formed two monophyletic groups, one clade comprising all species associated with Cupressaceae and Taxodiaceae hosts with the exception of Chamaecyparis, and the other clade composed of species associated with Pinaceae. Species infesting Cupressaceae and Taxodiaceae seemed to be specialized to particular host genera or even to be species specific, which was consistent with a taxonomic radiation following initial host adaptation. By contrast, Megastigmus species associated with Pinaceae appeared capable of shifting onto different congeneric species or even onto a new host genus, with their evolution apparently less constrained by plant association. We hypothesized that the Megastigmus group associated with Pinaceae may have a much higher invasive potential than that related to Cupressaceae. The study also confirmed the presence of invasive Nearctic species in the Palaearctic, and demonstrated the existence of a cryptic species complex.  相似文献

Radicle of matureEchinocactus platyacanthus embryo is approximately 320 m long and represents less then 1/7 of the embryonal axis length. The radicle-hypocotyl boundary can be distinguished according to the striking difference in the size and shape of cells in protoderm and procambium, as well as discontinuity and different number of the cell files in the ground meristem. The root cap is small, consists of 4 layers of cells covering the apex of the radicle. The upper limit of the root cap is approximately 100 m closer towards the radicle tip than the radicle-hypocotyl boundary. Ultrastructure of radicle cells showed numerous lipid bodies as is typical for other oily seeds. Protein bodies of variable structure were also present together with other cell structures. Striking differences in protein body structure were found when protoderm and ground meristem were compared. Several small globoid crystals were present in each protein body of the protoderm, while protein bodies in the radicle ground meristem mostly contained one large globoid crystal. X-ray microanalysis revealed presence of P, K and Mg in all analyzed globoid crystals. A 5.5 kb Eco RI fragment containing a vicilin gene was selected from a Pisum sativum genomic library, and the protein-coding region and adjacent 5 and 3 regions were sequenced. A DNA construction comprising this 5.5 kb fragment together with a gene for neomycin phosphotransferase II was stably introduced into tobacco using an Agrobacterium tumefaciens binary vector, and the fidelity of expression of the pea vicilin gene in its new host was studied. The seeds of eight transgenic tobacco plants showed a sixteen-fold range in the level of accumulated pea vicilin. The level of accumulation of vicilin protein and mRNA correlated with the number of integrated copies of the vicilin gene. Pea vicilin was confined to the seeds of transgenic tobacco. Using immunogold labelling, vicilin was detected in protein bodies of eight out of ten embryos (axes plus cotyledons) and, at a much lower level, in two out of eleven endosperms. Pea vicilin was synthesized early in tobacco seed development; some molecules were cleaved as is the case in pea seeds, yielding a major parental component of M r50000 together with a range of smaller polypeptides.  相似文献

Background

Plant and animal ferritins stem from a common ancestor, but plant ferritins exhibit various features that are different from those of animal ferritins. Phytoferritin is observed in plastids (e.g., chloroplasts in leaves, amyloplasts in tubers and seeds), whereas animal ferritin is largely found in the cytoplasm. The main difference in structure between plant and animal ferritins is the two specific domains (TP and EP) at the N-terminal sequence of phytoferritin, which endow phytoferritin with specific iron chemistry. As a member of the nonheme iron group of dietary iron sources, phytoferritin consists of 24 subunits that assemble into a spherical shell storing up to ∼ 2000 Fe3 + in the form of an iron oxyhydroxide-phosphate mineral. This feature is distinct from small molecule nonheme iron existing in cereals, which has poor bioavailability.

Scope of review

This review focuses on the relationship between structure and function of phytoferritin and the recent progress in the use of phytoferritin as iron supplement.

Major conclusions

Phytoferritin, especially from legume seeds, represents a novel alternative dietary iron source.

General significance

An understanding of the chemistry and biology of phytoferritin, its interaction with iron, and its stability against gastric digestion is beneficial to design diets that will be used for treatment of global iron deficiency.  相似文献

Drought-tolerant and drought-susceptible genotypes of sorghum (Sorghum bicolor L. Monech) were analyzed by the energy-dispersive X-ray fluorescence (EDXRF) technique to study the correlation of trace elements with drought-tolerance capacities. Samples prepared from mature seeds, young seedlings, and old plants were analyzed using a 109Cd radioisotope source and a Si(Li) semiconductor detector of resolution 170 eV for 5.9-keV MnK{ie255-1} X-rays. Elements such as K, Fe, Cu, Zn, Rb and Sr and Y were found to be present in varying concentrations in different samples. The trace element profile studied in the seeds of 11 genotypes and in seedlings (young and old) of 4 sorghum genotypes showed considerable variation. The genotype Arfa Gadamak (AG) showed a distinct presence of a high level of Zn in its young seedling. It was observed that in most of the genotypes (seeds), K and Fe concentrations were more in the tolerant genotype as compared to the susceptible type. The concentration of Fe decreased with maturity in the tolerant group and it increased with maturity in the susceptible group.  相似文献

Summary Crystalline and paracrystalline arrays of electron-opaque granules have been found in plastids of the cambial zone and its immediate derivatives in crack willow (Salix fragilis L.). These granules have a diameter of 55 to 70 Å and, when in crystalline arrangement, show a centre to centre spacing of 100 Å with adjacent, slightly curved, or linear rows running parallel. The 70 Å particles have a substructure of four to six subunits 15 Å in diameter. These units are arranged around an electron-translucent core 20 Å diameter. It is suggested that this complex is phytoferritin. It is assumed that the electron-translucent area around the opaque granules represents the proteinaceous shell characteristic of both plant and animal ferritin as described by other authors. The phytoferritin is commonly found spread in a thin, regular, array over the surface of plastoglobuli in the plastids.It is further suggested that the phytoferritin is an iron-protein complex which allows the plant to store iron in non-toxic form. This theory would be in accord with the presence of phytoferritin in plastids which appear to be morphologically mature but which, on account of their position within the stem, would not be expected to be photosynthetically very active.  相似文献

Different populations of metal-loaded and uncontaminated Porcelio scaber Latreille were studied. Combined light and electron microscopial methods as well as X-ray microanalysis were applied for localization and characterization of intracellular sites of metal deposition within the small cells of hepatopancreas. By means of cytochemistry and X-ray microanalysis it was shown that membrane-limited vesicles are important sites of deposition for lead, copper, zinc and probably smaller amounts of cadmium. The vesicles also contained phosphorus. They are identical with the reported cuprosomes and belong to the lysosomal system. Summary Using energy-dispersive X-ray microanalysis, the concentrations of ions, especially potassium and chlorine, were determined in different tissues of primary and tertiary pulvini of Mimosa pudica. It was shown that stimulating the leaf was followed by ion displacements which were most striking in the outer extensor cells, resulting in turgor loss. Since Ca concentration remains relatively constant in cell walls of collapsed cells, the changes of K concentration are best described by the K:Ca ratio. After stimulation the K:Ca ratio dropped in the outer extensor of the primary pulvinus from 775.3 to 2.37 in the cytoplasm, and from 542.2 to 9.25 in the cell wall. Changes in chlorine content were less striking in the primary pulvinus. The KCl ratios in some cases were lower than 1.0, which indicates that Cl content can increase, while K content is diminished. In the non-stimulated tertiary pulvini the outer extensor cells show high concentrations of Cl, but much lower Cl concentrations were found after stimulation. In contrast to the primary pulvinus the K content of the tertiary pulvini is very low. In the vascular tissues of both primary and tertiary pulvini stimulation is followed by a release of K and Cl out of the sieve element cytoplasm into the apoplast. K then appears accumulated in the cell walls of the collenchymatous tissue. These displacements lead to the assumption that the collenchymatous apoplast temporarily functions as a reservoir for K and to a lesser extent for Cl. With regard to the mechanism of leaf movement after stimulation, the accumulation of ions in the apoplast seems to be initiated by the decrease of water potential triggered by an apoplastic accumulation of unloaded sucrose (Fromm and Eschrich 1988a). The geographical variation in the composition of triacylglycerols in seeds of Scots pine (Pinus sylvestris L.) and Norway spruce [Picea abies (L.) Karst.], grown in Finland, was analyzed. The total lipid content of pine seeds was slightly higher in the northernmost provenance (68 °50N), whereas the lipid content of spruce seeds was not affected systematically by the geographical origin of the seeds. The species studied differed in the proportions of fatty acids in their triacylglycerols, though the three most abundant components were the same, i.e. oleic acid (181 n9), linoleic acid (182n-6) and 5,9,12-octadecatrienoic acid (183 5c9c12c). These fatty acids corresponded to more than 80 mol% of the total fatty acids. According to mass spectrometric analyses, the triacylglycerols of both spruce and pine seeds consisted of the same molecular species with 52–56 acyl carbons, but in different proportions. Molecules with 54 acyl carbons represented approximately 75% of the pine and 85% of the spruce triacylglycerols, with the most abundant molecular species being 545, 546 and 547. Some minor differences in the fatty acid composition of triacylglycerols of pine seeds from different seed collecting areas were found: the proportion of linoleic acid slightly increased whereas that of 5,11,14-eicosatrienoic acid decreased towards the northern origins. Similar differences were not found in the proportions of fatty acids in spruce seed lots. Furthermore, the proportions of triacylglycerols in both pine and spruce seeds from northern and southern collecting areas were not significantly different. The higher content of total lipids in spruce seeds compared with pine seeds may be due to the structure of the seed coat, and the lipophilic layers inside it, acting as a barrier to imbibition.  相似文献

The stability of bimanual performance of the frequency ratios 38 and 58 was examined from the perspective of the sine circle map and the associated Farey mode-locking hierarchy. By gradually increasing movement frequency, abrupt transitions from the initial frequency ratios to other frequency ratios were induced. In general, transitions occurred to frequency ratios that were near the initial frequency ratio but lower in the Farey ordering and, hence, of higher stability in the sine circle map. A fair percentage of these transitions were to unimodularly related ratios. The transition routes from 38 and 58 remained largely unaffected by extensive practice of the lower-order ratios 25 and 35. Collectively, these results suggest that (i) bimanual tapping occurs in a domain in which frequency-locked states either overlap or are located sufficiently close to each other to make stochastic switching possible (coupling parameter K > 1 or close to 1); (ii) the overall stability of these frequency-locked states decreases as movement frequency increases (due to a decrease in K) and, consequently, (iii) the probability of transitions to nearby frequency ratios increases as movement frequency increases, due to the differential stability of the frequency locks.  相似文献

Competition experiments betweenPhaeodactylum tricornutum andSkeletonema costatum showed that even at temperatures higher than 10°C (i.c. 14°C), the development ofSkeletonema can be favoured by adjusting nutrient levels and nutrient ratios. Low NSi ratios were found to favourSkeletonema. Additionally, high NP ratios further enhanced the ability ofSkeletonema to dominate the cuftures. Contrary to some statements in literature, it seems that high concentrations of silicates are more important for the dominance ofSkeletonema costatum in large-scale cultures than just low temperatures. This finding is important with regard to stimulating the blooming ofSkeletonema costatum in natural phytoplankton populations as food for bivalve molluscs.  相似文献

Soybean seeds, Glycine max L. Merrill, were produced by plants treated from anthesis to seed maturity with 59Fe supplied as ferric ethylenediaminedi (o-hydroxyphenylacetate). Seed coats accounted for 7.4% of dry seed weight and had Fe concentrations 5 times greater than the embryos. After germinating 2 days, cotyledons contained 69.6% and radicles 5.0% of original seed Fe. Fractions of seed Fe unavailable to seedlings were: 19.8% in seed coats, 1.7% in germination paper, 0.1% in the water under germinating seeds, and 3.8% unaccounted for. Every 3 days seedlings received nutrient solution without Fe or with 10 μm ferric ethylenediaminedi (o-hydroxyphenylacetate) and developed as deficient Fe or normal Fe plants. The deficient Fe cotyledons on day 18 retained 13% of the labeled Fe originally present. Cotyledons of normal Fe plants retained 50 to 70% of their original Fe. Moreover, cotyledons of the normal Fe plants accumulated externally supplied Fe and finally contained twice the quantity of Fe originally present. Stem exudate collected above cotyledons of deficient Fe plants contained 5.3 μm59Fe. Electrophoresis of exudate showed that most of the 59Fe migrated anodically as a single band and was in the position of ferric citrate.  相似文献

The effect of carbonnitrogenphosphorus (CNP) ratio of organic substrates on the regeneration of ammonium and phosphate was investigated by growing natural assemblages of freshwater bacteria in mineral media supplemented with the simple organic C, N, and P sources (glucose, asparagine, and sodium glycerophosphate, respectively) to give 25 different substrate CNP ratios. Both ammonium and phosphate were regenerated when CN and NP atomic ratios of organic substrates were 101 and 161, respectively. Only ammonium was regenerated when CN and NP ratios were 101 and 10–201, respectively. On the other hand, neither ammonium nor phosphate was regenerated when CN and NP ratios were 151 and 51, respectively. In no case was phosphate alone regenerated. As bacteria were able to alter widely the CNP ratio of their biomass, the growth yield of bacteria appeared primarily dependent on the substrate carbon concentration, irrespective of a wide variation in the substrate CNP ratio.  相似文献

Summary Filaments of the desiccation-tolerant cyanobacteriumNostoc commune are embedded within, and distributed throughout, a dense glycan sheath. Analysis of the glycan of field materials and of pure cultures ofN. commune DRH 1 through light and electron microscopy, immunogold labelling and staining with dyes, revealed changes in the pattern of differentiation in glycan micro-structure, as well as localized shifts in pH, upon rehydration of desiccated field material. A Ca/Si rich external (pellicular) layer of the glycan acts as a physical barrier to epiphytic bacteria on the surface ofN. commune colonies. A purified fraction (>12 kDa) of an aqueous extract of the glycan from desiccated field material contained glucose, N-acetylglucosamine, glucosamine, mannose, and galactosamine with ratios of 3.11.410.10.06, respectively. Lipid soluble extracts ofN. commune contained trehalose and sucrose and the levels of both became undetectable following cell rehydration. Intracellular cyanobacterial trehalase was identified using immunoblotting and its synthesis was detected upon rehydration of desiccated field cultures. Elemental analysis of glycan extracts showed a flux in the concentrations of salts in the glycan matrix following rehydration of desiccated colonies. Water-stress proteins (Wsp; most abundant proteins in glycan), a water soluble UV-A/B-absorbing pigment, the lipid-soluble UV-protective pigment scytonemin (in both its oxidized and reduced forms), as well as two unidentified cyanobacterial glycoproteins (75 kDa and 110 kDa), were found within the glycan matrix. An unidentified 68 kDa protein, the second most abundant protein in aqueous extracts of the glycan, was isolated and its N-terminal sequence was determined as AFIFGTISPNNLSGTSGNSGIVGSA. Gene bank searches with this sequence identified significant homologies (35–45%) with various carbohydrate-modifying enzymes. Summary Sex ratios from 62 single-pair matings of normal broodstock O. aureus were highly heterogeneous with an overall deficit of males (41.4%). Peaks in the sex ratio frequency distribution occurred at 11, 35 and 13 (malefemale). Hybridisation of O. aureus with O. mossambicus, O. spilums and O. niloticus produced highly variable sex ratios, suggesting a complexity of hybrid sex determination. Few valid inferences could be made regarding intraspecific sex determination from these hybrid data. Sex ratios from progeny testing of sex-reversed males (13) and most sex-reversed females (10) provide evidence for female heterogamety in O. aureus. Several aberrant ratios observed suggest Mendelian inheritance of an autosomal recessive gene (F,f), epistatic to the major sex-determining gene (W,Z). Sex ratios of triploids and gynogens support the hypothesis of recombination between the centromere and the major sex-determining locus. Progeny testing of a female mitogyne demonstrated the viability of a novel WW superfemale, which gave only female offspring. Not all data could be explained by a two-factor model of sex determination. Further exceptional sex ratios may be accounted for by rare autosomal or environmental sex-modifying factors. It is concluded that O. aureus has a multifactorial mechanism of sex determination with the underlying primary mechanism of female heterogamety.  相似文献

采用X-射线能谱仪测定非糯与糯性等品种小麦籽粒不同部位的矿质元素组成(H和He元素除外)和含量的结果表明:小麦籽粒中除含有大量C、O外,皮层富含K、P、se,其次是Cl、Si、S、Mg和Ca等;糊粉层富含P、K和Mg,其次是Si、Se、S、Ca、Cl和Fe等;胚乳层中相应的矿质元素含量比皮层和糊粉层低。不同品种籽粒各部位的矿质元素含量存在遗传性差异。据此认为籽粒磨成粉时应减少糊粉层的损失,以提高面粉的矿质价值。  相似文献
