Summary Six monosomic addition lines were produced in which different Dasypyrum villosum (L.) Candargy chromosomes were added to the chromosome complement of Triticum durum Desf. cv. Creso. Each added alien chromosome was found to have a specific effect on plant morphology and fertility. Transmission rate varied widely (from 7.5 to 27.7%) among the six univalent chromosomes. Different monotelosomic addition plants derived by a relatively high frequency of chromosome misdivision were isolated. The addition lines should be useful for studying Dasypyrum chromosome homoeology and the introduction of alien variation into durum and common wheats.Research supported by a grant from the Italian Research Council for Finalized Project IPRA. We have characterized biochemical effects of Idh GB1 in Drosophila melanogaster. This is a null-activity allele for NADP+-dependent isocitrate dehydrogenase (NADP-IDH) isolated from a natural population. The homozygous mutant strain has 5% of the NADP-IDH specific activity found in controls and less than 24% of the immunologically cross-reacting material (CRM). This mutation maps to 27.2 on the third chromosome, to the right of h. The biochemical phenotype of this mutant strain includes a coordinate reduction in malic enzyme (ME) specific activity and CRM and an increase in specific activity for the pentose-phosphate shunt enzymes, 6-phosphogluconate dehydrogenase and glucose-6-phosphate dehydrogenase. The K m values for purified NADP-IDH are not different from those found for the purified control enzyme for NADP+ or isocitrate. It is suggested that this allele may represent a cis-acting control mutation for one of at least two loci involved in the production of NADP-IDH in D. melanogaster.Research supported by an Alberta Heritage Foundation for Medical Research Establishment Grant to MMB and a Natural Sciences and Engineering Research Council Operating Grant to JHW.  相似文献

Summary C-banding patterns of T. ovatum (Ae. ovata) and four T. aestivum cv Poros-T. ovatum chromosome addition lines are presented, and the added chromosomes of T. ovatum have been identified. Furthermore, nucleolar activity and powdery mildew resistance were analyzed in the Poros-ovatum addition lines and compared to that of T. ovatum and T. aestivum cv Poros. The addition lines II, III and IV and Poros were highly susceptible to powdery mildew isolates nos. 8 and 9, whereas the addition lines VI1 and VI2 showed high resistance. Even for an Ml-k virulent isolate, these two lines were highly resistant. By combining the cytological results and those of the powdery mildew analysis, the added chromosomes of T. ovatum can be excluded from responsibility for the high powdery mildew resistance of the addition lines VI1 and VI2. The same is true for a modified chromosome 6B, which is present in the Poros-ovataum addition lines II, III and VI. The high variation in C-banding pattern observed in the A-, B- and D-genome complement of the addition lines is believed to be the result of crossing different lines of T. aestivum instead of Poros alone. Thus, we cannot trace the powdery mildew resistance back to a specific chromosome.  相似文献

Summary The locus, I2, which in tomato confers resistance against Fusarium oxysporum f. sp. lycopersici race 2, was introgressed into Lycopersicon esculentum from the wild species L. pimpinellifolium (P.I. 126915). We searched for restriction fragment length polymorphisms (RFLPs) between nearly isogenic lines (NILs) in clones that map to the region introgressed from the wild species. Since I2 maps to chromosome 11, we used DNA clones from this chromosome as hybridization probes to Southern blots containing bound DNA of the NILs digested with 23 restriction enzymes. Of the 14 chromosome 11 clones, 9 exhibited polymorphism. These clones were further hybridized to verification filters that contained DNA from resistant and susceptible L. esculentum varieties digested with the enzymes that gave the polymorphism. One clone, TG105, was found to be associated with I2; 19 susceptible lines showed a different RFLP with this probe than 16 resistant lines, including the original L. pimpinellifolium accession used as a source for the resistance gene. These results together with our mapping analysis indicate that TG105 is closely linked to the resistance gene.  相似文献

Summary An attempt to produce a set of addition lines of Aegilops sharonensis to the wheat variety Chinese Spring produced only one addition line. This was due to preferential transmission of one chromosome from Ae. sharonensis. This chromosome was studied in detail by established cytological methods of chromosome observation and by the newer techniques of C-banding and in situ hybridization of a cloned DNA sequence. The chromosome was found to be partially homologous to an Ae. sharonensis chromosome of similar behaviour in another wheat addition line. The incomplete homology of the two Ae. sharonensis chromosomes was due to the presence of a translocated segment of a wheat chromosome. — Substitution lines of the Ae. sharonensis chromosome for wheat homoeologous group 4 were produced and the Ae. sharonensis chromosome thereby designated 4 S l .  相似文献

Four bread wheat (Triticum aestivum L.) cultivars, Aobakomugi, Chinese Spring, Norin 61 and Shinchunaga, were pollinated with five barley lines/cultivars consisting of three cultivated barley (Hordeum vulgare L.) lines, Betzes, Kinai 5 and OHL089, and two wild barley (Hordeum spontaneum C. Koch) lines, OUH602 and OUH324. Crossability, expressed as the percentage of embryo formation, varied from 0 to 55.4% among the cross combinations. The two wild barley lines generally had a higher crossability than the previously reported best pollinator, Betzes, and some Japanese wheat cultivars were better as the female parent than Chinese Spring. Ninety four hybrid plants were obtained from 250 embryos cultured, and their somatic chromosome numbers ranged from 21 to 36. Eighteen plants were mosaic in chromosome number. Twenty one-chromosome plants appeared most frequently (45.7%) followed by 28-chromosome plants (14.9%). C-banding analysis revealed that elimination of barley chromosomes was mainly responsible for the occurrence of aneuploid plants. In hypoploids derived from Betzes-crosses, chromosome 5 was preferentially eliminated as previously reported, while in hypoploids derived from OUH602-crosses, chromosome 4 was preferentially eliminated. The wild barley line OUH602 may be a useful parent for producing a new wheat-barley addition set because of its high crossability with wheat and a different pattern of chromosome elimination.  相似文献

Near-isogenic lines (NILs) for the leaf rust resistance gene Lr9 were screened for polymorphisms at the molecular level. RAPD (random amplified polymorphic DNA) primers as well as RFLP (restriction fragment length polymorphism) markers were used. Out of 395 RAPD primers tested, three showed polymorphisms between NILs, i.e., an additional band was found in resistant lines. One of these polymorphic bands was cloned and sequenced. Specific primers were synthesized, and after amplification only resistant lines showed an amplified product. Thus, these primers define a sequence-tagged site that is specific for the translocated fragment carrying the Lr9 gene. A cross between a resistant NIL and the spelt (Triticum spelta) variety Oberkulmer was made, and F2 plants were analyzed for genetic linkage. All three polymorphisms detected by the PCR (polymerase chain reaction) and one RFLP marker (cMWG684) showed complete linkage to the Lr9 gene in 156 and 133 plants analyzed, respectively. A second RFLP marker (PSR546) was closely linked (8±2.4 cM) to the Lr9 gene and the other four DNA markers. As this marker maps to the distal part of the long arm of chromosome 6B of wheat, Lr9 and the other DNA markers also map to the distal region of 6BL. All three PCR markers detected the Lr9 gene in independently derived breeding lines and varieties, thus proving their general applicability in wheat breeding programs.  相似文献

In Drosophila melanogaster lines LA (low activity), HA (high activity), andOregon-R (wild type), the effect of genetic background on endoreplication in giant chromosomes of salivary glands, fecundity, and expression of mutation eywas studied. The degree of chromosome polyteny and the number of adult flies in saturated lines ey L, ey H, and ey Or were significantly higher than in the original lines. The degree of chromosome polyteny was correlated with fecundity. The expressivity of the ey character was shown to be far lower in lines ey L, ey H, and ey Or than in the original eyelessline. In the saturated lines, the eye facets were reduced to a similar degree. All the lines studied displayed clear-cut sexual distinctions in this parameter. In the ey L line, the coefficients of variation for the degree of chromosome polyteny, fecundity, and expressivity of the ey mutation were much lower than in lines eyeless, ey LAandey Or.  相似文献

C-banding polymorphism was analyzed in 14 accessions of Triticum searsii from Israel, and a generalized idiogram of the species was established. One accession was homozygous for whole arm translocations T1SsS·4SsS and T1SsL·4SsL. C-banding analysis was also used to identify 7 T. aestivum cv Chinese Spring-T. searsii disomic chromosome addition lines, 14 ditelosomic chromosome addition lines, 21 disomic whole chromosome, and 31 ditelosomic chromosome substitution lines. The identity of these lines was further confirmed by meiotic pairing analysis. Sporophytic and gametophytic compensation tests were used to determine the homoeologous relationships of the T. searsii chromosomes. The results show that the T. searsii chromosomes do not compensate well for their wheat homoeologues. The C-banding patterns of T. searsii chromosomes are distinct from those of other S-genome species and from the B-genome chromosomes of wheat, indicating that T. searsii is not a direct B-genome donor species of T. turgidum and T. aestivum.Contribution No. 95-72-J from the Kansas Agricultural Experiment Station, Kansas State University, Manhattan, Kansas, USA  相似文献

We report a new enzyme xylose dehydrogenase, the structural locus for which is on chromosome 7 of the mouse, closely linked to Tam-1. Three alleles have been detected in both laboratory strains and wild populations. Two of these determine proteins differing in electrophoretic mobility and the third is a null. Summary Analysis of intergeneric substitution lines in hexaploid wheats by a two-step electrophoretic method of protein separation revealed that low-molecular-weight (LMW) subunits of glutelin in Triticum longissimum, T. Umbelullatum, Elytrigia elongata (2 x) were controlled by chromosomes/chromosome arms 1S l , 1U, and 1ES, respectively. A LMW glutelin band in Secale montanum was detected but its chromosomal location could not be determined. Genes controlling gliadins and HMW subunits of glutelin were also located on chromosome 1S l in T. longissimum.The term glutelin refers to the polymeric prolamins of cereals, e.g., glutenins in wheat, HMW, and 75-k secalins in rye  相似文献

Summary Three lines derived from wheat (6x) x Agropyron elongatum (10x) that are resistant to wheat streak mosaic virus (WSMV) were analyzed by chromosome pairing, banding, and in situ hybridization. Line CI15321 was identified as a disomic substitution line where wheat chromosome 1D is replaced by Ag. elongatum chromosome 1Ae-1. Line 87-94-1 is a wheat-Ag. elongatum ditelosomic addition 1Ae-1L. Line CI15322 contains an Ag. elongatum chromosome, 1Ae-2, that substitutes for chromosome 1D. The short arm of 1Ae-2 paired with the short arm of 1Ae-1 at metaphase I (MI) in 82% of the pollen mother cells (PMCs). However, the long arms of these two chromosomes did not pair with each other. In CI15322, the long arm of chromosome 4D has an Agropyron chromosome segment which was derived from the distal part of 1Ae-1L. This translocation chromosome is designated as T4DS·4DL-1L. T4DS·4DL-1Ae-1L has a 0.73 m distal part of the long arm of 4D replaced by a 1.31 m distal segment from 1Ae-1L. Summary Stable wheat-Aegilops introgression lines with 42 chromosomes (H-93), derived by repeated selfing from a cross (Triticum turgidum x Aegilops ventricosa) x T. aestivum, have been characterized using the following DNA probes and isozyme markers: (1) single or low-copy DNA fragments from Ae. ventricosa; (2) known cDNA probes corresponding to 1-thionin, monomeric -amylase inhibitor, the CM3 subunit of tetrameric -amylase inhibitor, and sucrose synthase from wheat; (3) anonymous cDNA probes from wheat that have been mapped by Sharp et al. (1989); (4) isozyme markers corresponding to aconitase, shikimate dehydrogenase, adenylate kinase, and endopeptidase. Meiotic metaphases of appropriate hybrids involving selected H-93 lines have been investigated by the Giemsa C-banding technique. The substitution of whole chromosomes [(5A) 5Mv; (4D) 4Mv; (5D) 5Mv; (7D) 7Mv] and chromosomal segments (1Mv; 3Mv; 5Mv; 7Mv) from the Mv genome of Aegilops ventricosa has been demonstrated. The distribution of selected markers among putative wheat-Ae. ventricosa addition lines has also been investigated. The 7Mv addition has been characterized for the first time, while the identity of the previously reported 5Mv and 6Mv additions has been confirmed.  相似文献

Summary The chromosomal locations of two enzymecoding genes were investigated using allele dosage effects in two Allium cepa alien addition lines possessing different A. fistulosum chromosomes as the trisomes. Adh-1 has been assigned to the A. fistulosum sub-telocentric chromosome 5, while Pgm-1 is on the A. fistulosum submetacentric chromosome 4. We have developed a protocol for the in vitro propagation of the genus Clivia. Shoots were regenerated when fragments of the peduncle-pedicel junction (PP junction) from young inflorescences were used as explants. The optimal media for PP junction were Murashige and Skoog (MS)-based medium containing 10 M of 6-benzyladenine (BA) and 10 M of 2,4-dichlorophenoxyacetic acid (2,4-D) or MS supplemented with 5 M BA, 10 M -naphthaleneacetic acid (NAA), 250 mg l-1 glutamine and 500 mg l–1 casein hydrolysate and their usage depended on the breeding lines. Multiplication from initiations and in vitro seedlings was the best when the explants were cut longitudinally through the meristem and placed on MS plus 44 M BA. Plantlets were transferred on to hormone -free MS medium with charcoal for rooting.  相似文献

Summary A 5BS/5RmS translocation chromosome spontaneously recovered from a Chinese Spring — Secale montanum wheat-rye telocentric 5RmS addition line has been identified and cytologically studied using C-banding in somatic and meiotic cells. Analysis of the translocated chromosome showed that a terminal segment of the short arm of 5B had been replaced by a short terminal region of chromosome arm 5RmS. The translocation led to the deletion of the genetic system promoting pairing located in 5BS, which is slightly compensated for when doses of 5RmS are increased, indicating homoeology to wheat chromosome 5BS. The -amylase phenotype in 5B/5Rm translocated material was studied and found to be identical to that of ditelocentric line 5BL of Chinese Spring. An effect on the -amylase activity was detected as a result of the removal of the terminal region of 5BS, perhaps as a consequence of variation in dormancy period duration.  相似文献

The patterns of variation of chromosome numbers in theCentrospermae suggest a common ancestry of centrospermous anthocyanin and betalain families. Phylogenetic divergence in the order may have originated with progenitors similar to extantMolluginaceae, Aizoaceae orPhytolaccaceae taxa with x = 9. Evolutionary radiation and advancement in several lines then seems to have been paralleled by trends for increasing chromosome base numbers through dysploidy and polyploidy, e.g. towardsCaryophyllaceae, Portulacaceae-Basellaceae, Hectorellaceae, Cactaceae, Didieraceae, Nyctaginaceae andChenopodiaceae-Amaranthaceae. Summary C-banding patterns were analysed in 19 different accessions of Aegilops caudata (= Ae. markgrafii, = Triticum dichasians) (2n = 14, genomically CC) from Turkey, Greece and the USSR, and a generalized C-banded karyotype was established. Chromosome specific C-bands are present in all C-genome chromosomes, allowing the identification of each of the seven chromosome pairs. While only minor variations in the C-banding pattern was observed within the accessions, a large amount of polymorphic variation was found between different accessions. C-banding analysis was carried out to identify Ae. caudata chromosomes in the amphiploid Triticum aestivum cv Alcedo — Ae. caudata and in six derived chromosome addition lines. The results show that the amphiploid carries the complete Ae. Caudate chromosome complement and that the addition lines I, II, III, IV, V and VIII carry the Ae. caudata chromosome pairs B, C, D, F, E and G, respectively. One of the two SAT chromosome pairs (A) is missing from the set. C-banding patterns of the added Ae. caudata chromosomes are identical to those present in the ancestor species, indicating that these chromosomes are not structurally rearranged. The results are discussed with respect to the homoeologous relationships of the Ae. caudata chromosomes.  相似文献

The tobacco (Nicotiana tabacum L.) breeding line NC 152 is a doubled haploid that possesses an addition chromosome from N. africana [Merxm. and Buttler]. A gene on this chromosome confers potyvirus resistance (PotyR). Our objective was to use the addition chromosome as a base on which to construct a designer chromosome containing a foreign gene linkage package. A mutant dhfr gene conferring resistance to methotrexate (Mtx) was inserted into NC 152-haploid (n = 25) leaf tissue via Agrobacterium tumefaciens-mediated transformation. After chromosome doubling, 135 NC 152dhfr transformants (2n = 50) were pollinated with the potyvirus-susceptible (PotyS) cultivar McNair 944 (2n = 48). Linkage analysis was performed in the BC1 generation. Two transformants, NC 152dhfr-996 and NC 152dhfr-1517 exhibited complete linkage between Mtx resistance (MtxR) and PotyR. Segregants from these two transformants which were MtxR and PotyR possessed 49 chromosomes, while Mtx sensitive (MtxS) and PotyS progeny possessed 48 chromosomes. Eighty percent of the NC 152dhfr transformants transmitted the dhfr gene as one locus. Other foreign genes can be directed to the addition chromosome through transformation followed by selection for single loci with linkage to PotyR or MtxR. The integrity of both the foreign-gene linkage package and the rest of the genome will be maintained because recombination between the N. africiana and the N. tabacum chromosomes has not been observed.  相似文献
