Prostaglandin F (PGF) has been shown to be an effective stimulant of hepatic bile flow producing a specific chloride rich bile. Subsequent evaluation by radioimmunoassay has shown that prostaglandin F compounds are present in relatively large amounts in canine hepatic bile. This study evaluates the effect of PGF administration and of prostaglandin synthetase inhibition by aspirin and indomethacin on bile flow and radioimmunoassayable prostaglandin F (iPGF) secretion. Chronic, canine bile fistula preparations were utilized and the enterohepatic circulation was maintained by intravenous bile salts. Bile volume and composition were evaluated by standard techniques as well as bile PGF concentration by radioimmunoassay during bile salt infusion and during bile salt and PGF, aspirin and indomethacin infusion in varying doses. Both aspirin and PGF were potent stimulatns of hepatic bile flow with aspirin producing a chloride rich bile similar to that produced by PGF. PGF produced dose related increases in bile iPGF concentration and output indicating that as the systemic concentration increases during infusion of PGF the lipid appears in bile. Aspirin in the highest dose administered, decreased iPGF concentration in bile while output was unchanged. Indomethacin was ineffectual in consistently altering bile flow or iPGF secretion. This study demonstrates that iPGF is present in canine bile, that its concentration can be altered by prostaglandin infusion while prostaglandin synthetase inhibition has minimal effects on bile iPGF secretion.  相似文献

The synthetic prostaglandin analogs 16, 16-dimethyl PGF and 16, 16-dimethyl PGE2 were administered to dogs with chronic biliary and gastric fistulas. The effects of 16, 16 diMePGF and 16, 16 diMePGE2 were evaluated on bile flow and composition and bile adenosine 3′, 5′ monophosphate (cyclic AMP) secretion. 16, 16 diMePGF in doses of 0.125 and 0.25 μg-kg-min significantly increased hepatic bile flow. The choleresis was characterized by increased cloride and bicarbonate secretion. Measurement by radioimmunoassay of bile cyclic AMP concentration demonstrated no evident increase in bile cyclic AMP secretion associated with the choleresis produced by 16, 16 diMePGF. The administration of 16, 16 diMePGE2 in a dose range 0.01 to 1.0 μg-kg-min did not significantly alter bile flow rates or composition. Bile erythritol-14C clearance, a measure of canalicular bile flow, was significantly increased by PGF but not by 16, 16-dimethyl PGF, suggesting that the mechanism of action of PGF in stimulating hepatic bile flow may be different from that involved in 16, 16-dimethyl PGF choleresis. The results of this study indicate that the synthetic PGF analog produces a choleretic response not mediated by adenylate cyclase and associated with increased chloride and bicarbonate secretion.  相似文献

Radioimmunoassay of 5α,7α-dihydroxy-11-keto-tetranorprosta-1,16-dioic acid, main urinary metabolite of prostaglandin F2α (PGF2α), was performed using an antiserum produced in the rabbit.The antibody in 100 μ1 of 1,600-fold diluted antiserum binds with 60 picograms of metabolite.The main urinary metabolite level fell when flufenamic acid, a prostaglandin synthetase inhibitor, was given to rats. Thw radioimmunological (RIA) determination of prostaglandin (PG) E2 and of PGF in urine humans and rats is described in detail. After extraction and chromatography PGE2 was determined by using a PGE specific antibody or by using either PGB or PGF specific antibodies after the respective conversion procedures. The three different RIA procedures were compared to each other. PGF was determined by a specific antibody to PGF. Basal excretion of PGE2 and of PGF in healthy women on free diet was 9.3 ng/hour ± 0.96 and 18.3 ng/hour ± 2.5 respectively. Furosemide increased the excretion of PGE2 and of PGF in humans significantly, while PG-excretion rates decreased on indomethacin. In rat urine PGE2 and PGE increased markedly from 46.2 pg/min ± 9.3 and 27 ± 3.4 to 253.8 ± 43.3 and 108 ± 12.6 pg/min (per one kidney) in the anesthetized-laparotomized animal. This increase was abolished after giving two different PG synthetase inhibitors.  相似文献

Dose response relationships for pregnancy termination in hamsters following administration of prostaglandin F (PGF by three subcutaneous methods were determined in 526 hamsters. The median effective dose (ED50) for PGF given as a single subcutaneous injection in 500 μl of saline was 22.2 μg. Administration of the prostaglandin with an Alzet® osmotic minipump (subcutaneous insertion for 24 hours) required 1.35 times more PGF (ED50 = 30.0 μg). The least effective method of prenancy termination in the hamster involved administration of PGF by a single subcutaneous injection in 20.4 μl of saline (the same volume delivered by the minipump in 24 hours); the ED50 for this method of administration was 41.3 μg of PGF.  相似文献

In a number of assay ssytems, some 17-phenyl-trinor prostaglandins were similar in activity and potency to the corresponding parent prostaglandin. In others, the 17-phenyl analogs appeared several times more potent. In the hamster antifertility assay, which is considered to measure luteolytic activity, 17-phenyl-18,19,20-trinor prostaglandin F was about 90-times PGF in potency.Rat blood pressure responses to 17-phenyl analogs were significant. The 17-phenyl-trinor PGF pressor potency was 5 times that of PGF. The 17-phenyl-trinor PGE2 blood pressure response was atypical since a pressor rebound phenomenon followed the expected depressor response. Lastly, 17-phenyl-trinor PGF was more potent than PGF in synchronizing the estrous cycle in beef cows.  相似文献

Using specific radioimmunoassay procedures we measured prostaglandin F2α (PGF2α) and 13, 14-dihydro-15-keto prostaglandin F2α (PGF2α metabolite) in 12 patients with carcinoid tumors. Although PGF2α and PGF2α metabolite were each modestly elevated in 17% of the patients the magnitude of the elevation did not correlate with the symptoms of the carcinoid syndrome. The 24 hour urinary 5-hydroxyindoleacetic acid excretion showed a good correlation with carcinoid symptoms while the serum serotonin concentration showed a fair correlation with carcinoid symptoms. We conclude that serum elevation of PGF2α is not a frequent occurrence in patients with the carcinoid syndrome.  相似文献

Lipid mediators variedly affect adipocyte differentiation. Anandamide stimulates adipogenesis via CB1 receptors and peroxisome proliferator-activated receptor γ. Anandamide may be converted by PTGS2 (COX2) and prostaglandin F synthases, such as prostamide/prostaglandin F synthase, to prostaglandin F ethanolamide (PGFEA), of which bimatoprost is a potent synthetic analog. PGFEA/bimatoprost act via prostaglandin FFP receptor/FP alt4 splicing variant heterodimers. We investigated whether prostamide signaling occurs in preadipocytes and controls adipogenesis. Exposure of mouse 3T3-L1 or human preadipocytes to PGFEA/bimatoprost during early differentiation inhibits adipogenesis. PGFEA is produced from anandamide in preadipocytes and much less so in differentiating adipocytes, which express much less PTGS2, FP, and its alt4 splicing variant. Selective antagonism of PGFEA receptors counteracts prostamide effects on adipogenesis, as does inhibition of ERK1/2 phosphorylation. Selective inhibition of PGFEA versus prostaglandin F biosynthesis accelerates adipogenesis. PGFEA levels are reduced in the white adipose tissue of high fat diet-fed mice where there is a high requirement for new adipocytes. Prostamides also inhibit zebrafish larval adipogenesis in vivo. We propose that prostamide signaling in preadipocytes is a novel anandamide-derived antiadipogenic mechanism.  相似文献

Radioimmunoassays for measuring prostaglandin F (PGF) and 5α, 7α-dihydroxy-11-keto tetranorprosta-1,16-dioic acid, PGF-main urinary metabolite (PGF-MUM), with 125I-tyrosine methylester amide (TMA) of PGF and PGF-MUM were developed.Antibody to PGF was produced in rabbits immunized with conjugates of PGF coupled to bovine serum albumine. Antibody to PGF-MUM was also produced in rabbits immunized with conjugates of PGF-MUM coupled to bovine serum albumin.PGF-125I-TMA had an affinity to antiserum to PGF. We have investigated in vitro prostaglandin synthesis by human isolated glomeruli and papillary homogenates and compared the results with those obtained in parallel studies using rat material. Prostaglandins were measured by two methods, namely radiometric high performance liquid chromatography after incubation with 14C arachidonic acid and radioimmunoassay. The relative abundance of various prostaglandins synthesized by glomeruli was different in man (6 keto PGF > TXB2 > PGF > PGE2) and in the rat (PGE2 TXB2 > 6 keto PGF1α). Unidentified peaks eluting between 6 keto PGF and TXB2 were observed only in rat glomeruli. These peaks were suppressed by indomethacin. Direct radioimmunoassay of prostaglandins in the incubation medium of human glomeruli confirmed the predominance of 6 keto PGF synthesis and showed its stimulation by arachidonic acid, its progressive decrease with time and its linear relationship with glomerular protein at low concentrations. On the contrary, the profile of prostaglandin synthesis by the papilla was similar in man and in the rat, PGE2 and PGF being the major products in both species. However, related to one mg of protein, papillary synthesis of these two prostaglandins was greater in the rat. These results show that PGI2 is the major prostaglandin synthesized in human glomeruli and suggest a role for this prostaglandin in glomerular physiology in man.  相似文献

The cerebral cortical action of prostaglandin F (PGF) has been determined by recording the effects of intracarotid injections of PGF on cerebral evoked potentials. PGF differentially reduced cortical evoked potentials. The cortical action of PGF appeared to be qualitatively identical with that of norepinephrine (NE) but weaker. A protection of the cortex from the inhibitory action of NE by a preceding dose of PGF was demonstrated. The actions of both PGF and NE appear to be on the same or related postsynaptic receptors. The actions described were at doses that did not reduce oxygen availability. PGF may act as a modulator of adrenergic transmission in the cortex. The intracellular recording in the companion paper supplies the further critical evidence that PGF has a synaptic inhibitory action.  相似文献

The effect of prostaglandin PGF on the hCG stimulated and basal progesterone production by human corpora lutea was examined . hCG (40 i.u./ml) stimulated progesterone formation in corpora lutea of early (days 16–19 of a normal 28 day cycle), mid (days 20–22) and late (days 23–27) luteal phases. This stimulation was inhibited by PGF (10 μg/ml) in corpora lutea of mid and late luteal phases. PGF alone did not show a consistent effect on basal progesterone production. The inhibition of hCG stimulated progesterone production by PGF at times corresponding to luteolysis indicates a role for that prostaglandin in the process of luteolysis in the human corpus luteum.  相似文献

In humans eicosapentaenoic acid can be converted to 3-series prostaglandins (PGF, PGI3, and PGE3). Whether 3-series prostaglandins can protect the gastric mucosa from injury as effectively as their 2-series analogs is unknown. Therefore, we compared the protective effects of PGF and PGF against gross and microscopic gastric mucosal injury in rats. Animals received a subcutaneous injection of either PGF or PGF in doses raning from 0 (vehicle) to 16.8 μmol/kg and 30 min later they received intragastric administration of 1 ml of absolute ethanol. Whether mucosal injury was assessed 60 min or 5 min after ethanol, PGF was significantly less protective against ethanol-induced damage than PGF. These findings indicate that the presence of a third double bond in the prostaglandin F molecule between carbons 17 and 18 markedly reduces the protective effects of this prostaglandin on the gastric mucosa.  相似文献

The synthesis of prostaglandins by rheumatoid synovial tissue in organ culture was studied utilizing radioimmunoassay, with antisera to PGB1, PGF and PGF. It was established that PGE2 and PGF were the major prostaglandins formed by analyses of culture media with the two antisera to PGF, before and after alkali treatment. Indomethacin at 5 μg/ml suppressed prostaglandin synthesis, usually to <1% of control cultures. Colchicine, 0.1 μg/ml resulted in marked stimulation of prostaglandin synthesis, in some cases over 10 fold. It is suggested, because of the colchicine effect, that the state of the microtubules may regulate the rate of prostaglandin biosynthesis. It is possible that prostaglandin E2 produced by rheumatoid synovia may contribute to the pathogenesis of the inflammatory reaction and lead to destruction of juxta-articular bone in rheumatoid arthritis.  相似文献

The estimation of prostaglandin (PG) E2 and of PGF by radioimmunoassay is described in detail. PGE2 was measured after conversion to either PGB2 or PGF and the results compared to bioassay. The methods were used to follow the excretion of PGE2 and PGF after salt loading in rabbits. Potential interactions between PGD2 and PGF in the mesenteric and renal vascular beds were investigated in the anesthetized dog. Regional blood flows were measured with electromagnetic flow probes. PGD2, PGF and Norepinephrine (NE) were injected as a bolus directly into the appropriate artery, and responses to these agents were obtained before, during and after infusion of either PGD2 or PGF into the left ventricle. In each case, the infused prostaglandin caused vascular effects of its own. Left ventricular infusion of PGD2 reduced responses to local injections of PGD2 in the intestine, and a similar effect was observed for PGF, suggesting significant receptor or receptor-like interactions for each of the prostanoids. However, systemic infusion of prostaglandin F (20–100 ng/kg/min) had no effect on renal or mesenteric vascular responses to local injection of prostaglandin D2. Similarly, PGD2 administration (100 ng/kg/min) did not affect responses to PGF in the intestine. The present results therefore suggest that these prostaglandins, i.e., D2 and F, act through separate receptors in the mesenteric and renal vascular beds. In addition, increased prostaglandin F levels produced by infusion of F reduced mesenteric but not renal blood flow, suggesting that redistribution of cardiac output might participate in side effects often observed with clinical use of this prostaglandin, such as nausea and abdominal pain.  相似文献

The effect of prostaglandin F (PGF) on the sperm output of six boars was investigated in two studies. Although PGF did not significantly affect sperm numbers in the ejaculate, a significantly longer (P < 0.05) ejaculation of the sperm rich fraction occurred following injection of PGF. In the second study it was found that PGF produced a 49% increase (P < 0.05) in the number of sperm in the sperm rich fraction of the ejaculate. The implications of these results on artificial breeding are discussed.  相似文献

The airway and lung dynamics of prostaglandin F (PGF) and three of its metabolites were examined in the spontaneously-ventilated, pentobarbital anesthetized dog. Changes in expiratory flow rate, tidal volume, respiration rate, lung resistance and dynamic lung compliance were evaluated and compared quantitatively. In a dose range of 0.3–3.0 μg/kg i.v., PGF and its 13,14-dihydro metabolite were found to be exceptionally potent agents. This metabolite was approximately twice as potent as PGF on most parameters studied. Two other metabolites, 15-keto-PGF and 15-keto-13,14-dihydro-PGF, were only slightly effective, even in a dose range of 1.0–30.0 μg/kg i.v. These latter two metabolites produced dose-response curves with significantly shallower slopes than PGF and were shown to be at least thirty-five times less potent than the parent compound. Therefore, oxidation of PGF at the carbon-15 position by 15-hydroxy prostaglandin dehydrogenase appears to produce compounds with minimal bronchopulmonary activity.  相似文献

The effect of prostaglandin F on ovulation and fertilization was studied in rabbits. The number of ovulation was not affected by subcutaneous injection of PGF but the recovery of ova was significantly decreased when PGF was given either at 12 or 16 h after HCG injection and autopsied 24 h latter. The results suggest that exogenous PGF accelerates ovum transport and expels the eggs prematurely from the female tract and does not impair ovulation or the fertilization processes when given to rabbit at 1 mg/kg B.W.  相似文献
