Because of their wide geographic distribution, position in the trophic chain, rapid turnover, huge biomass and role in ocean biogeochemical cycles, copepods are regarded as important marine test species. Tigriopus brevicornis Müller, 1776 is a shallow water benthic marine species (Crustacea, Copepoda Harpacticoida). The toxicity of essential and non-essential metals (Ag, Cd, Cu, Hg, Ni and Zn) to Tigriopus brevicornis was determined by 96-h LC(50) testing. Comparative crustacean 96-h LC(50) data in the literature show that Tigriopus brevicornis is a sensitive species suggesting that copepods are good indicators of minimal lethal concentrations of metals. Groups of 1000 to 1500 adult copepods were exposed for 1 to 14 days to metals at concentrations in water, 3 for each metal, considered realistic in comparison with those encountered in polluted environments and far below lethal concentrations, in order to avoid protein metabolism disturbance. The response of Tigriopus brevicornis in terms of MT induction has been examined in specimens exposed to metals. The induction of these proteins and their implication in detoxificatory mechanisms and trophic transfer are discussed.  相似文献

We synthesized a cDNA library from the intertidal copepod Tigriopus japonicus, converted it to phagemids and sequenced expressed sequence tags (ESTs). Of these, Tigriopus translationally controlled tumor protein/histamine releasing factor (TCTP/HRF) was further characterized. The Tigriopus TCTP/HRF gene encoded 172 amino acid residues and showed high similarity to Drosophila but moderate similarity to other annelids (e.g. Brugia, Wuchereria and C. elegans). The Tigriopus TCTP/HRF gene appeared in the same clade as the annelids. Here, we describe the analysis of the Tigriopus TCTP/HRF gene.  相似文献

Differences between the muscle mercury contents in fish from lakes Gusinoye and Karasinoye and the Selenga River may indicate an inconsiderable income of mercury in aquatic ecosystems with atmospheric precipitation, along with a more intensive migration of mercury with the riverine flux. The minimal concentration of mercury (less than 0.15 mg/kg, dry weight) has been registered in the muscles of Amur sleeper from Lake Karasinoye and of Baikal omul from Lake Baikal, while the maximal concentration (1.0–2.3 mg/kg, dry weight) has been revealed in the fish caught in the Selenga River delta (in predatory pike, Amur catfish, and perch, as well as in omnivorous roach and ide). The muscle metal content in dace decreased along the downstream direction.  相似文献

Volatile, organic solvent soluble mercury has been found in leaves and seeds of several angiosperms. Leaves of garlic vine (Pseudocallyma alliacium), avocado (Persea americana) and haole-koa (Leucaena glauca) release mercury in volatile form rapidly at room temperature. In garlic vine, the most active release is temperature dependent, but does not parallel the vaporpressure temperature relationship for mercury. Mercury can be trapped in nitric-perchloric acid digestion fluid, or n-hexane, but is lost from the hexane unless the acid mixture is present. Seeds of haole-koa also contain extractable mercury but volatility declines in the series n-hexane (90%) > methanol (50%) > water (10%). This suggests that reduced volatility may accompany solvolysis in the more polar media. Gas chromatographic analysis shows that the volatile compound is not dimethyl mercury.  相似文献

Histone genes are one of the most widely studied multigene families in eucaryotes. Over 200 histone genes have been sequenced, primarily in vertebrates, echinoderms, fungi and plants. We present here the structure and genomic orientation of an H3-H4 histone gene pair from the marine copepod, Tigriopus californicus. These histone gene sequences are the first to be determined for the class Crustacea and among the first to be determined for protostomes. The H4 and H3 genes in Tigriopus are shown to be adjacent, to have opposite polarity, and to contain a 26 bp region of dyad symmetry centrally located within the spacer region between the two genes. A similarly located dyad element has been found in yeast which contributes to the coordinated cell cycle control of the adjacent histone genes. The Tigriopus H3-H4 histone gene pair is clustered with one H2A and two H2B histone genes on a 15 kb genomic Bam H1 fragment. The H4 gene sequence predicts an H4 protein with an unusual serine to threonine substitution at the amino terminal residue. The H3 gene sequence predicts an H3 protein which is identical to the vertebrate H3.2 histone.  相似文献

In the modern world, mercury has become an extremely dangerous pollutant due to intensive human activity. Currently, sources of mercury are wastes from chemical industries, as well as mines, oil combustion products, and household waste. Phytoextraction of heavy metals from soil is considered one of the most promising and cost-effective technologies. The efficiency of this process can be increased by introducing various amendments. The use of additives in phytoextraction can enhance the absorption of heavy metals and increase their concentration in various parts of the plant. This article presents the results of a study of various chelating agents for effective phytoextraction of mercury with white clover (Trifolium repens L.) and watercress (Lepidium sativum). In the present study, the monoethanolamine salt of dithiodiacetic acid (MEDBA) was used. The optimal concentration of MEDBA on watercress and creeping clover has been determined for highly efficient phytoextraction of mercury. Research has been carried out with a complex of exogenous growth regulators (GA / IAA / Fe-EDDHA). The results showed that the use of phytohormones and plant growth regulators led to a synergistic effect in combination with thiosulfate, but a pronounced inhibitory effect was observed with the use of MEDBA.  相似文献

The effect of temperature on inorganic mercury toxicity was investigated using kidney tissue culture systems. The relative susceptibility of rabbit (homeothermic) kidney to mercury intoxication was compared to that of Coho salmon (poikilothermic) kidney to mercury intoxication was compared to that of Coho salmon (poikilothermic) kidney over temperature ranges consistent with the habitat of each of the two species. It was demonstrated that susceptibility to mercury toxicity is species dependent; that is, the rabbit kidney cells tolerated higher mercury concentrations in the medium than did the fish-derived cells. Within a given species, susceptibility to mercury toxicity was temperature dependent. Decreasing the temperature increased the toxicity of mercury to cultures of rabbit kidney cells, whereas decreasing temperatures decreased the effect of mercury toxicity on the salmon kidney cells. As a consequence, fish taken from arctic waters are liable to be more toxic when introduced into mammalian food chains. Albumin was shown to act as a protective agent in vitro against inorganic mercury toxicity.  相似文献

The presence of Chlorella cells in a medium containing HgCl2 causes a rapid decrease of the mercury content in the algal suspension. The rate of decrease depends on the inoculum concentration. The presence of Hg in the medium induces a lag in the growth, whose length depends on the initial concentration of Hg and of the inoculum. Binding and/or uptake of mercury by the cells is not dependent on temperature. The mercury content per cell declines somewhat at the time at which the culture resumes growth at a rate similar to that of the controls without mercury.  相似文献

The extent to which acclimation and genetic adaptation might buffer natural populations against climate change is largely unknown. Most models predicting biological responses to environmental change assume that species' climatic envelopes are homogeneous both in space and time. Although recent discussions have questioned this assumption, few empirical studies have characterized intraspecific patterns of genetic variation in traits directly related to environmental tolerance limits. We test the extent of such variation in the broadly distributed tidepool copepod Tigriopus californicus using laboratory rearing and selection experiments to quantify thermal tolerance and scope for adaptation in eight populations spanning more than 17° of latitude. Tigriopus californicus exhibit striking local adaptation to temperature, with less than 1 per cent of the total quantitative variance for thermal tolerance partitioned within populations. Moreover, heat-tolerant phenotypes observed in low-latitude populations cannot be achieved in high-latitude populations, either through acclimation or 10 generations of strong selection. Finally, in four populations there was no increase in thermal tolerance between generations 5 and 10 of selection, suggesting that standing variation had already been depleted. Thus, plasticity and adaptation appear to have limited capacity to buffer these isolated populations against further increases in temperature. Our results suggest that models assuming a uniform climatic envelope may greatly underestimate extinction risk in species with strong local adaptation.  相似文献

The usefulness of reductive electrochemical detection at mercury drop electrodes has been determined for platinum complexes separated by solvent-generated anion-exchange high-performance liquid chromatography. Both current-sampled dropping mercury and hanging mercury drop electrodes (DME and HMDE) provide significant advantages over UV absorbance and off-line non-flame atomic absorption detection. The effects of chromatographic and polarographic parameters on analytical system performance have been investigated. By raising the detector cell temperature, the detector response to cis-dichlorodiammineplatinum(II) (DDP) can be shifted anodically to 0.0 V vs. Ag/AgCl, thereby increasing detector selectivity for this compound. The noise-limited minimum detectable quantities of DDP with DME and HMDE are 1.8 ng and 70 pg injected, respectively. DDP can be determined in untreated urine at levels below 100 ng/ml.  相似文献

Summary Rice soils with different cropping age ranging from 0 to 600 years on Saga polder lands have been sampled and the total mercury has been determined with a high sensitivity mercury analyzer. Organomercurial application had the effect of raising soil mercury levels above the natural background. The mercury was generally confined to the surface soils where the content varied from 333 ppb to 86 ppb with an average of 214 ppb. The mercury content diminished quickly with depth, whereas the trend was less marked with decreasing cropping age (the younger soils). The natural background level of the soils in this area is approximately 79 ppb. Industrial and sewage outfalls from another area may be a significant factor with regard to the establishment of an elevated mercury level in the sediments available as a foundation material for future poldering works. Plant analysis indicates that the rice takes up small amounts of mercury regardless of the concentration in the soil. The average mercury content of unhulled rice was 11 ppb and of straw 31 ppb, suggestive of its minor role in the food chain. Calculation showed that crop removal is a fruitless source of the mercury decline in the soil. The discussion of mercury behavior in soil including the fate of applied PMA suggests that mercuric sulfide formation is probably more important than other factors in regulating mercury retention in rice soils. re]19751027  相似文献

Uptake and cellular distribution of mercury203 from dilute mercuric acetate or phenylmercuric acetate solutions by excised pea roots (Pisum sativum) have been investigated. The time course of uptake showed that the amount of mercury uptake was increased with the time of incubation, and was similar for inorganic mercury or phenylmercuric acetate. The trend of mercury203 incorporation into cellular components from mercuric acetate and phenylmercuric acetate differed greatly as the time of incubation increased. The concentrations of mercuric acetate and phenylmercuric acetate solutions or the temperature of incubation also affected the mercury203 uptake as well as its cellular distribution. Longer time of exposure or higher concentration resulted in a greater mercury incorporation into mitochondrial fraction from phenylmercuric acetate than from inorganic mercury. This difference in intracellular distribution may be responsible for the degree of toxicity between inorganic mercury and phenylmercuric acetate in biological systems.  相似文献

Volatilization of mercury under acidic conditions from soil polluted with mercuric chloride (1.5 mg Hg/kg soil) was studied with resting cells of a mercury-resistant strain, Acidithiobacillus ferrooxidans SUG 2-2. When resting cells of SUG 2-2 (0.01 mg of protein) were incubated for 10 d at 30 degrees C in 20 ml of 1.6 mM sulfuric acid (pH 2.5) with ferrous sulfate (3%) and mercury-polluted soil (1 g), which contained 7.5 nmol of Hg, approximately 4.1 nmol of mercury was volatilized, indicating that 54% of the total mercury in the soil was volatilized. The amount of mercury volatilized from the soil was dependent on the concentration of Fe2+ added to the medium. When elemental sulfur, sodium tetrathionate, and pyrite were used as an electron donor for the mercury reduction, 16, 2.4 and 0.84%, respectively, of the total mercury added to the solution were volatilized. The optimum pH and temperature for mercury volatilization were 2.5 and 30 degrees C. Approximately 92% of the total mercury in a salt solution (pH 2.5) with resting cells of SUG 2-2 (0.01 mg of protein), ferrous sulfate (3%) and mercury-polluted soil (1 g) was volatilized by further addition of both resting cells and Fe2+ and by incubating for 30 d at 30 degrees C.  相似文献

The long-term performance of bioreactors retaining mercury from contaminated industrial wastewater was analyzed at the laboratory scale, and its response to mechanical perturbations (gas bubbles and shaking) as well as to physical (increased temperature and hydraulic load) and chemical stresses (increased mercury concentration) likely to occur during on site operation was studied. Two packed-bed bioreactors with 80-cm(3) lava chips as biofilm carrier were inoculated with nine Hg(II)-resistant natural isolates of alpha- and gamma-proteobacteria. Chloralkali wastewater containing ionic mercury (3.0 to 9.7 mg/L Hg(2+)), amended with sucrose and yeast extract, flowed through the bioreactors at 160 mL/h. During the 16-month investigation the bioreactors showed no sign of depleted performance in terms of mercury-retaining capacity. After 16 months, both bioreactors still retained 96% of the mercury load. The performance of the bioreactors was sensitive to mechanical perturbations (e.g., sheer forces of gas bubbles). Shifts to higher Hg(2+) inflow concentrations initially decreased the mercury retention efficacy slightly. However, the bioreactors could adapt to Hg(2+) concentrations of up to 7.6 mg/L within several days. Old biofilms were less affected than the younger ones. The performance of the bioreactors was not affected by an increase in temperature up to 41 degrees C and an increased volumetric load (up to 240 mL/h). The bioreactors regained activity spontaneously after the stress had stopped. Recovery could be accelerated by increased nutrient concentration, although this may lead to blocking of the packed bed.  相似文献

The study concerns the Tigriopus fulvus specimens fatty acid composition, sampled in Ligurian coast rockpools from September 1980 to June 1981. By the study of frequency distribution it was observed that the fatty acid found in greater quantity was the palmitic one, followed by the palmitoleic, the oleic and stearic acids. Short and long chain fatty acids are less frequent. Investigation about each fatty acid concentration shows that the seasonal trends between males and females are different and the trend of some fatty acids (e.g. linolenic and stearic acid) is characteristic.  相似文献

The sex-determining mechanism has important demographic and genetic consequences by virtue of its effect on the population sex ratio. Here we investigate the effect of temperature dependent sex determination (TSD) on the primary sex ratio of the harpacticoid copepod, Tigriopus californicus . At the two experimental temperatures (15° and 22°C) used in this study, the primary sex ratio is almost always biased in favour of males. Higher temperatures induce masculinization and the change in sex ratio is not caused by differential mortality of the sexes. The mean level of TSD in the population is small (proportion of males increases by ~5% between 15° and 22°C) because only one-third of the families actually exhibit a significant sex-ratio response while the rest of the population is insensitive to temperature. A comparison of the primary sex ratio and the level of TSD between two locations reveals few differences among populations. Finally, individuals still exhibited TSD after having been maintained under constant temperature conditions in the lab for several generations. In addition the proportion of temperature-sensitive individuals remained unchanged. This suggests that the observed level of TSD is not an artefact of testing field-captured individuals in a novel laboratory environment.
