Dairy manure with a total solids content of 77.2g TS/l was separated by means of screening and coagulation-flocculation treatments, using CaO as coagulant and a cationic polyacrylamide as flocculant, obtaining liquid and solid fractions. The solid fraction separated contained 33.4% of the initial total mass of dairy manure plus chemical solutions, containing also 75.2% of the TS, 80.4% of the VS, 58.5% of the total Kjeldahl nitrogen (TKN) and 87.4% of the total phosphorus (P(T)) present in the initial dairy manure. 83.7% of the liquid fraction chemical oxygen demand (COD) was anaerobically biodegradable (COD(BD)). Methane production for the separated liquid fraction was 0.604l CH4 NCTP/g VS added, being 0.307 and 0.371l CH4 NCTP/g VS added for dairy manure and screened dairy manure, respectively. The characteristics of this liquid fraction would allow its treatment in high loading anaerobic reactors having shorter hydraulic retention times, smaller reactor size and a higher methane volumetric production rate than conventional anaerobic reactors treating either manure or screened manure.  相似文献

A number of researchers have verified the inhibitory effects of elevated H2 concentrations on various anaerobic fermentation processes. The objective of this work was to investigate the potential for using hydrogen gas production to predict upsets in anaerobic digesters operating on dairy cattle manure. In an ammonia nitrogen overload experiment, urea was added to the experimental digesters to obtain increased ammonia concentrations (600, 1,500, or 3,000 mg N/l). An increase in urea concentration resulted in an initial cessation of H2 production followed by an increase in H2 formation. Additions of 600, 1,500, or 3,000 mg N/l initially resulted in the reduction of biogas H2 concentrations. After 24 h, the H2 concentration increased in the 600 and 1,500 mg N/l digesters, but production remained inhibited in the 3,000 mg N/l digesters. Both methane and total biogas production decreased following urea addition. Volatile solids reduction also decreased during these periods. The digester effluent pH and alkalinity increased due to the increased NH4 formed with added urea. Based on these results, changes in H2 concentration could be a useful parameter for monitoring changes due to increased NH3 in dairy cattle manure anaerobic digesters.  相似文献

While the use of anaerobic digestion to generate methane as a source of bioenergy is increasing worldwide, our knowledge of the microbial communities that perform biomethanation is very limited. Using next-generation sequencing, bacterial population profiles were determined in three full-scale mesophilic anaerobic digesters operated on dairy farms in the state of Vermont (USA). To our knowledge, this is the first report of a metagenomic analysis on the bacterial population of anaerobic digesters using dairy manure as their main substrate. A total of 20,366 non-chimeric sequence reads, covering the V1-V2 hypervariable regions of the bacterial 16S rRNA gene, were assigned to 2,176 operational taxonomic units (OTUs) at a genetic distance cutoff value of 5 %. Based on their limited sequence identity to validly characterized species, the majority of OTUs identified in our study likely represented novel bacterial species. Using a naïve Bayesian classifier, 1,624 anaerobic digester OTUs could be assigned to 16 bacterial phyla, while 552 OTUs could not be classified and may belong to novel bacterial taxonomic groups that have yet to be described. Firmicutes, Bacteroidetes, and Chloroflexi were the most highly represented bacteria overall, with Bacteroidetes and Chloroflexi showing the least and the most variation in abundance between digesters, respectively. All digesters shared 132 OTUs, which as a “core” group represented 65.4 to 70.6 % of sequences in individual digesters. Our results show that bacterial populations from microbial communities of anaerobic manure digesters can display high levels of diversity despite sharing a common core substrate.  相似文献

The objectives of the study were to measure the levels of manure nutrients retained in psychrophilic anaerobic sequencing batch reactors (PASBRs) digesting swine manure, and to determine the distribution of nutrients in the sludge and supernatant zones of settled bioreactor effluent. Anaerobic digestion reduced the total solids (TS) concentration and the soluble chemical oxygen demand (SCOD) of manure by 71.4% and 79.9%, respectively. The nitrogen, potassium, and sodium fed with the manure to the PASBRs were recovered in the effluent. The bioreactors retained on average 25.5% of the P, 8.7% of the Ca, 41.5% of the Cu, 18.4% of the Zn, and 67.7% of the S fed to the PASBRs. The natural settling of bioreactor effluent allowed further nutrient separation. The supernatant fraction, which represented 71.4% of effluent volume, contained 61.8% of the total N, 67.1% of the NH4-N, and 73.3% of the Na. The settled sludge fraction, which represented 28.6% of the volume, contained 57.6% of the solids, 62.3% of the P, 71.6% of the Ca, 89.6% of the Mg, 76.1% of the Al, 90.0% of the Cu, 74.2% of the Zn, and 52.2% of the S. The N/P ratio was increased from 3.9 in the raw manure to 5.2 in the bioreactor effluent and 9.2 in the supernatant fraction of the settled effluent. The PASBR technology will then substantially decrease the manure management costs of swine operations producing excess phosphorus, by reducing the volume of manure to export outside the farm. The separation of nutrients will also allow land spreading strategies that increase the agronomic value of manure by matching more closely the crop nutrient requirements.  相似文献

污泥厌氧消化是在消化污泥微生物组的协调下将剩余污泥中有机物转化为甲烷的微生物过程。与传统厌氧消化过程不同,污泥厌氧消化系统的进料底物为含有大量微生物细胞及胞外多聚物等复杂大分子有机物的剩余污泥。因此,厌氧消化污泥微生物组的种群组成、功能及种群间互作关系等异常复杂,使厌氧消化污泥微生物组分析成为难点问题。但近年来高通量测序技术及生物信息学分析方法的快速发展为消化污泥微生物组研究提供了契机,并迅速推动了该研究领域的发展。本文从4个方面梳理、总结厌氧消化污泥微生物组的研究及应用现状:剩余活性污泥结构、组成及其厌氧消化;基于16SrRNA基因序列测序的微生物组研究;基于宏基因组及宏转录组分析的微生物组研究;厌氧消化污泥微生物组研究案例分析。最后我们提出了厌氧消化污泥微生物组研究亟待解决的关键科学问题。  相似文献

We determined the effect of different mixing intensities on the performance, methanogenic population dynamics, and juxtaposition of syntrophic microbes in anaerobic digesters treating cow manure from a dairy farm. Computer automated radioactive particle tracking in conjunction with computational fluid dynamics was performed to quantify the shear levels locally. Four continuously stirred anaerobic digesters were operated at different mixing intensities of 1,500, 500, 250, and 50 revolutions per min (RPM) over a 260-day period at a temperature of 34 +/- 1 degrees C. Animal manure at a volatile solids (VS) concentration of 50 g/L was fed into the digesters daily at five different organic loading rates between 0.6 and 3.5 g VS/L day. The different mixing intensities had no effect on the biogas production rates and yields at steady-state conditions. A methane yield of 0.241 +/- 0.007 L CH(4)/g VS fed was obtained by pooling the data of all four digesters during steady-state periods. However, digester performance was affected negatively by mixing intensity during startup of the digesters, with lower biogas production rates and higher volatile fatty acids concentrations observed for the 1,500-RPM digester. Despite similar methane production yields and rates, the acetoclastic methanogenic populations were different for the high- and low-intensity mixed digesters with Methanosarcina spp. and Methanosaeta concilii as the predominant methanogens, respectively. For all four digesters, epifluorescence microscopy revealed decreasing microbial floc sizes beginning at week 4 and continuing through week 26 after which no microbial flocs remained. This decrease in size, and subsequent loss of microbial flocs did not, however, produce any long-term upsets in digester performance.  相似文献

Guinea pig is one of the most common livestock in rural communities of the Andes. The aim of this research was to study the anaerobic digestion of guinea pig manure in low-cost unheated tubular digesters at high altitude. To this end, the performance of two pilot digesters was monitored during 7 months; and two greenhouse designs were compared. In the dome roof digester the temperature and biogas production were significantly higher than in the shed roof digester. However, the biogas production rate was low (0.04 m(biogas)(3)m(digester)(-3) d(-1)), which is attributed to the low organic loading rate (0.6 kg(VS)m(digester)(-3)d(-1)) and temperature (23°C) of the system, among other factors. In a preliminary fertilization study, the potato yield per hectare was increased by 100% using the effluent as biofertilizer. Improving manure management techniques, increasing the organic loading rate and co digesting other substrates may be considered to enhance the process.  相似文献

In the conventional treatment of sewage, the solids are normally disposed of by anaerobic digestion. This leaves a considerable amount of plant nutrients, such as nitrogen and phosphate, as soluble compounds which will eventually find their way into the plant's final effluent, since the supernatant fluid from the digesters is normally returned to the raw sewage. In a recent investigation, we found that, if the sludges were treated by an aerobic process, a significant portion of the carbonaceous matter was oxidized to carbon dioxide and water, and the rest was assimilated into microbial protoplasm. This process tied up the available nitrogen and phosphorus so that practically none remained dissolved in the suspending liquid. The accumulated solids, consisting mostly of microbial cells, were separated very easily from the liquid, leaving a slightly colored supernatant fluid that was water-clear, free from plant nutrients, and very low in BOD and COD. The overall process was accomplished in a detention time not exceeding 20 days, in contrast to anaerobic digestion which requires from 50 to 70 days.  相似文献

The objective of this study is to investigate the responses of methanogen populations to poultry waste addition by comparing the archaeal microbial populations in continuous anaerobic digesters with or without the addition of poultry waste as a co-substrate. Poultry waste was characterized as an organic/nitrogen-rich substrate for anaerobic digestion. Supplementing dilute dairy waste with poultry waste for anaerobic co-digestion to increase organic loading rate by 50% resulted in improved biogas production. Elevated ammonia derived from poultry waste did not lead to process inhibition at the organic loadings tested, demonstrating the feasibility of the anaerobic co-digestion of dairy and poultry wastes for improved treatment efficiency. The stability of the anaerobic co-digestion process was linked to the robust archaeal microbial community, which remained mostly unchanged in community structure following increases in organic loading and ammonia levels. Surprisingly, Crenarchaeota archaeal populations, instead of the Euryarchaeota methanogens, dominated the archaeal communities in the anaerobic digesters. The ecological functions of these abundant non-methanogen archaeal populations in anaerobic digestion remain to be identified.  相似文献

Even though ammonia is an essential nutrient for bacterial growth, it may inhibit methanogenesis during anaerobic digestion process if it is available at high concentrations. Therefore, ammonia is regarded as a potential inhibitor during anaerobic digestion, particularly when dealing with complex type of substrates such as manure or the organic fraction of municipal solid waste (OFMSW). Ammonia is produced through biological degradation of nitrogenous matter. Ammonium ion (NH4+) and free ammonia (NH3) are the two principal forms of inorganic ammonia nitrogen. Both forms can directly and indirectly cause inhibition in an anaerobic digestion system. Particularly, free ammonia (FAN) is a powerful inhibitor in an anaerobic digester above threshold concentrations. Process inhibition is related to the particular characteristics of the substrate to be anaerobically digested, pH, process temperature (mesophilic or thermophilic), type of the seed sludge (inoculum), the reactor configuration and to the concentrations of ammonium and ammonia. In this paper, ammonia inhibition in anaerobic digestion systems and the recovery efforts after inhibition are discussed. Furthermore, the impacts of ammonia inhibition on the microbial population available in anaerobic digesters, namely bacteria and Archaea, are also evaluated in detail.  相似文献

Anaerobic digesters have been responsible for the removal of large fraction of organic matter (mineralization of waste sludge) in conventional aerobic sewage treatment plants since the early years of domestic sewage treatment (DST). Attention on the anaerobic technology for improving the sustainability of sewage treatment has been paid mainly after the energy crisis in the 1970s. The successful use of anaerobic reactors (especially up-flow anaerobic sludge blanket (UASB) reactors) for the treatment of raw domestic sewage in tropical and sub-tropical regions (where ambient temperatures are not restrictive for anaerobic digestion) opened the opportunity to substitute the aerobic processes for the anaerobic technology in removal of the influent organic matter. Despite the success, effluents from anaerobic reactors treating domestic sewage require post-treatment in order to achieve the emission standards prevailing in most countries. Initially, the composition of this effluent rich in reduced compounds has required the adoption of post-treatment (mainly aerobic) systems able to remove the undesirable constituents. Currently, however, a wealth of information obtained on biological and physical-chemical processes related to the recovery or removal of nitrogen, phosphorus and sulfur compounds creates the opportunity for new treatment systems. The design of DST plant with the anaerobic reactor as core unit coupled to the pre- and post-treatment systems in order to promote the recovery of resources and the polishing of effluent quality can improve the sustainability of treatment systems. This paper presents a broader view on the possible applications of anaerobic treatment systems not only for organic matter removal but also for resources recovery aiming at the improvement of the sustainability of DST.  相似文献

Summary A novel anaerobic hybrid reactor (AHR) configuration incorporating floating support media for biomass immobilization and biogas recirculation for enhanced mixing was used for anaerobic digestion of dairy manure. No pretreatment or solid liquid separation was applied. The reactor was operated at high influent volatile solids (VS) and organic loading rates (OLR) of up to 9.87% and 7.30 g VS/l day, respectively. After 149 days of continuous operation the results revealed that a high amount (38.1 g VSS) of biomass was able to attach itself to the support medium being used. The investigated AHR configuration achieved COD, BOD, TS, and VS removal efficiencies of 48–63, 64–78, 55–65, and 59–68%, respectively, at a hydraulic retention time (HRT) of 15 days. The corresponding average methane production value obtained in this study was 0.191 l/g VS added.  相似文献

Anaerobic digestion (AD) is an attractive microbiological technology for both waste treatment and energy production. Syntrophic acetogenic bacteria are an important guild because they are essential for maintaining efficient and stable AD operation. However, this guild is poorly understood due to difficulties to culture them. In this study, we developed specific PCR assays targeting the propionate-CoA transferase genes (pct) to investigate their diversity and distribution in several mesophilic anaerobic digesters and a bench-scale temperature-phased AD (TPAD) system. Phylogenetic analysis of sequenced pct amplicons revealed the occurrence of Syntrophobacter fumaroxidans and six other clusters of putative pct genes. Principal coordinate analysis (PCoA) showed that pct diversity and abundance were largely correlated to the feedstocks of the digesters, while little difference was seen between the granular and the liquid fractions of each digester or between the two digesters of the TPAD system. Cluster-specific qPCR analysis revealed major impact of feedstocks and fractions on the abundance of pct genes. Readily fermentable substrates such as sugar- or starch-rich feedstocks selected for pct genes (Cluster I) related to Syntrophobacter, while manure feedstock selected for pct clusters related to pct of Clostridium spp. These results suggest that propionate metabolism can be affected by feedstocks and partition differently between solid and liquid phases in digesters. The PCR assays developed in this study may serve as a tool to investigate propionate-oxidizing bacteria in anaerobic digesters and other anaerobic environments.  相似文献

A method for long chain fatty acids (LCFA) extraction, identification and further quantification by gas chromatography was developed and its application to liquid and solid samples collected from anaerobic digesters was demonstrated. After validation, the usefulness of this method was demonstrated in a cow manure digester receiving pulses of an industrial effluent containing high lipid content. From the LCFA analysis data it was showed that the conversion of oleic acid, the main LCFA fed to the reactor, by the adapted biomass became faster and more effective along the successive pulses. Conversely, the accumulation of palmitic acid in the solid phase suggests that degradation of this LCFA, under these conditions, is less effective.  相似文献

Controlling the input of nitrogen (N) and phosphorus (P) from dairies and other livestock operations into the surrounding air- and water-sheds poses both technical and economic challenges to the agricultural community. The purpose of this paper is to assess the economics of algal turf scrubber treatment technology at the farm-scale for a hypothetical 1000-cow dairy. Costs were developed for farms with and without anaerobic pretreatment. The majority of capital costs were due to land preparation, installation of liner material, and engineering fees. The majority of operational costs were due to energy requirements for biomass drying, pumping water, and repayment of capital investment. On farms using anaerobic pretreatment, waste heat from burning of biogas could be used to offset the energy requirements of biomass drying. In addition, biogas combustion exhaust gas could then be recycled back to the algal system to supply dissolved inorganic carbon for optimal algal production and pH control. Under the best case (algal system coupled with anaerobic digestion pretreatment), the yearly operational costs per cow, per kg N, per kg P, and per kg of dried biomass were $454, $6.20, $31.10, and $0.70, respectively. Without anaerobic digestion pretreatment, the yearly operational costs were 36% higher, amounting to $631 per cow, $8.70 per kg N, $43.20 per kg P, and $0.97 per kg of dried biomass. For perspective, a recent survey of 36 Maryland dairy farms found long-term annual profits of about $500 per cow. As no market currently exists for manure grown algal biomass, our cost analysis does not include any value of the biomass generated during manure treatment. In addition, there are a variety of potential uses for the algal biomass from manure treatment that could defray treatment costs. Future opportunities for dairies to participate in nutrient trading approaches to watershed nutrient management may also become important.  相似文献

Mixed plug‐flow loop reactor (MPFLR) has been widely adopted by the US dairy farms to convert cattle manure to biogas. However, the microbiome in MPFLR digesters remains unexplored. In this study, the microbiome in a MPFLR digester operated on a mega‐dairy farm was examined thrice over a 2 month period. Within 23 days of retention time, 55–70% of total manure solid was digested. Except for a few minor volatile fatty acids (VFAs), total VFA concentration and pH remained similar along the course of the digester and over time. Metagenomic analysis showed that although with some temporal variations, the bacterial community was rather stable spatially in the digester. The methanogenic community was also stable both spatially and temporally in the digester. Among methanogens, genus Methanosaeta dominated in the digester. Quantitative polymerase chain reaction (qPCR) analysis and metagenomic analysis yielded different relative abundance of individual genera of methanogens, especially for Methanobacterium, which was predominant based on qPCR analysis but undetectable by metagenomics. Collectively, the results showed that only small microbial and chemical gradients existed within the digester, and the digestion process occurred similarly throughout the MPFLR digester. The findings of this study may help improve the operation and design of this type of manure digesters.  相似文献

Seven low-cost digesters in Costa Rica were studied to determine the potential of these systems to treat animal wastewater and produce renewable energy. The effluent water has a significantly lower oxygen demand (COD decreased from 2968 mg/L to 472 mg/L) and higher dissolved nutrient concentration (NH4-N increased by 78.3% to 82.2 mg/L) than the influent water, which increases the usefulness of the effluent as an organic fertilizer and decreases its organic loading on surface waters. On average, methane constituted 66% of the produced biogas, which is consistent with industrial digesters. Through principle component analysis, COD, turbidity, NH4-N, TKN, and pH were determined to be the most useful parameters to characterize wastewater. The results suggest that the systems have the ability to withstand fluctuations in the influent water quality. This study revealed that small-scale agricultural digesters can produce methane at concentrations useful for cooking, while improving the quality of the livestock wastewater.  相似文献

Anaerobic digestion of dairy cow manure (CM), the organic fraction of municipal solid waste (OFMSW), and cotton gin waste (CGW) was investigated with a two-phase pilot-scale anaerobic digestion (AD) system. The OFMSW and CM were digested as single wastes and as combined wastes. The single waste digestion of CM resulted in 62m(3) methane/ton of CM on dry weight basis. The single waste digestion of OFMSW produced 37m(3) methane/ton of dry waste. Co-digestion of OFMSW and CM resulted in 172m(3) methane/ton of dry waste. Co-digestion of CGW and CM produced 87m(3) methane/ton of dry waste. Comparing the single waste digestions with co-digestion of combined wastes, it was shown that co-digestion resulted in higher methane gas yields. In addition, co-digestion of OFMSW and CM promotes synergistic effects resulting in higher mass conversion and lower weight and volume of digested residual.  相似文献

Aims: This study was performed to investigate the microbiological contamination of digestate product (DP) obtained from the anaerobic co‐digestion of bovine manure and agricultural by‐products. Methods and results: Microbiological analyses were performed on bovine manure, fresh DP, liquid and solid fractions and stored liquid fraction of DP. A statistically significant reduction of faecal bacterial indicator was found after anaerobic digestion except for enterococci. After liquid/solid DP separation, bacteria tend to be concentrated in the solid fraction. Storage does not seem to influence the indicator parameters, except for enterococci. Escherichia coli O157:H7 and Yersinia were not found in any samples analysed. Salmonella was rarely detected in DP samples and its derivates, while Listeria monocytogenes was encountered in many samples. Conclusions: The results obtained indicate that the hygienic quality of DP is for almost all microbiological parameters better than that of the bovine manure (range of reduction 1.6–3.1 log10) and suggest the need to identify specific pathogen indicators related to the hygienic characteristics of DPs. Significance and Impact of the Study: This study highlights that the anaerobic co‐digestion of bovine manure and agricultural by‐products in a field‐scale biogas plant does not increase human health risk with respect to the use of animal manure for agricultural fertilization.  相似文献

The performance of a 10 L sequencing batch reactor (SBR) treating slaughterhouse wastewater was examined at ambient temperature. The influent wastewater comprised 4672+/-952 mg chemical oxygen demand (COD)/L, 356+/-46 mg total nitrogen (TN)/L and 29+/-10 mg total phosphorus (TP)/L. The duration of a complete cycle was 8 h and comprised four phases: fill (7 min), react (393 min), settle (30 min) and draw/idle (50 min). During the react phase, the reactor was intermittently aerated with an air supply of 0.8L/min four times at 50-min intervals, 50 min each time. At an influent organic loading rate of 1.2g COD/(Ld), average effluent concentrations of COD, TN and TP were 150 mg/L, 15 mg/L and 0.8 mg/L, respectively. This represented COD, TN and TP removals of 96%, 96% and 99%, respectively. Phase studies show that biological phosphorus uptake occurred in the first aeration period and nitrogen removal took place in the following reaction time by means of partial nitrification and denitrification. The nitrogen balance analysis indicates that denitrification and biomass synthesis contributed to 66% and 34% of TN removed, respectively.  相似文献
