Experiments on frogs (Rana ridibunda) showed that, unlike in the whole nerve, prolonged after-depolarization and the associated phase of summation are absent in the single node of Ranvier of isolated nerve fibers. If, however, special measures are taken to prevent the stretching of the Ranvier node of single fibers, prolonged after-depolarization can be found in them. It is concluded that the absence of prolonged after-depolarization in single Ranvier nodes of nerve fibers isolated without additional precautions is the result of injury to the excitable membrane or to disturbance of the integrity of the ionic barrier surrounding it as a result of stretching of the fibers during dissection.  相似文献

Diabetes-induced damage to peripheral nerve culminates in development of peripheral diabetic neuropathy (PDN), one of the most devastating complications of diabetes mellitus and a leading cause of foot amputation. The pathogenesis of PDN occurs as a consequence of complex interactions among multiple hyperglycemia-initiated mechanisms, impaired insulin signaling, inflammation, hypertension, and disturbances of fatty acid and lipid metabolism. This review describes experimental new findings in animal and cell culture models as well as clinical data suggesting the importance of 1) previously established hyperglycemia-initiated mechanisms such as increased aldose reductase activity, non-enzymatic glycation/glycooxidation, activation of protein kinase C, 2) oxidative–nitrosative stress and poly(ADP-ribose) polymerase activation; 3) mitogen-activated protein kinase and cyclooxygenase-2 activation, impaired Ca++ homeostasis and signaling, and several other mechanisms, in PDN.  相似文献

Summary The histochemical and cytochemical distribution of acetylcholinesterase activity in the anterior and posterior spinal nerve roots and ganglia of the rat was demonstrated by the Karnovsky method using acetyl and butyrylthiocholine as substrates and eserine and DFP as inhibitors. Light and electron microscopic examination of transverse frozen sections enabled the simultaneous visualization of end product in relationship to the various fiber components of each nerve root. While the enzymatic activity of the anterior roots was consistantly observed in the large extrafusal and small intrafusal motor fibers a relatively greater amount of precipitate occurred in aggregates of myelinated and unmyelinated fibers believed to represent preganglionic sympathetic nerves. In contrast, no significant enzymatic activity could be demonstrated in the myelinated nerve fibers of the posterior root. In the sensory sytem, the limited enzymatic precipitate was largely restricted to the unmyelinated afferent fibers and to their small cell bodies in the dorsal root ganglia. The ultrastructural distribution of enzymatic activity was located in the granular endoplasmic reticulum and perinuclear spaces of the ganglion cells. Within peripheral nerves this end product occurred between the apposing axonal and Schwann cell membranes and along the membranous aspect of occasional axoplasmic vesicles of both myelinated and unmyelinated nerve fibers.This study was supported by grants NB 04161-04 and NB 04161-05 of the National Institute of Neurological Diseases and Blindness. — The author would like to thank MissMaria C. la Valle for her skillful technical assistance.  相似文献

P311 accelerates nerve regeneration of the axotomized facial nerve   总被引:9,自引:0,他引:9  
In axotomized adult neurons, a process of axonal regrowth and re-establishment of the neuronal function has to be activated. Developmentally regulated factors may be reactivated during neuronal regeneration. Here we identify a gene, previously designated P311, that is up-regulated in the axotomized facial motoneurons. Ectopically expressed P311 localizes in the cytoplasm and the nucleus. Over-expression of P311 induces p21(WAF1/Cip1) expression, leading PC12 cells to differentiate and to have neuron-like morphologies. Adenovirus-mediated P311 gene transfer promotes neurite outgrowth of postnatal dorsal root ganglion neurons and embryonic hippocampal neurons in vitro. This effect is abolished by the activation of Rho kinase. P311 also facilitates nerve regeneration following facial nerve injury in vivo. Our data provide evidence that genes involved in the differentiation process contribute to the regeneration of injured mature neurons, and may provide a practical molecular target.  相似文献

The circumoral nerve ring and the radial nerve cords (RNCs) of Eupentacta fraudatrix and Pseudocnus lubricus (Holothuroidea) were examined as an example of holothurian nervous tissue. The RNC is composed of outer ectoneural and inner hyponeural layers, which are interconnected with one another via short neural bridges. The circumoral nerve ring is purely ectoneural. Both ectoneural and hyponeural components are epithelial tubes with a thick neuroepithelium at one side. A thin ciliated non-neuronal epithelium complements the neuroepithelium to form a tube, thereby enclosing the epineural and hyponeural canals. The whole of the ectoneural and hyponeural subsystems is separated from the surrounding tissue by a continuous basal lamina. The nerve ring and the ectoneural and hyponeural parts of the radial nerves are all neuroepithelia composed of supporting cells and neurons. Supporting cells are interpreted as being glial cells. Based on ultrastructural characters, three types of neurons can be distinguished: (1) putative primary sensory neurons, whose cilium protrudes into the epineural or hyponeural canal; (2) non-ciliated neurons with swollen rough endoplasmic reticulum cisternae; (3) monociliated neurons that are embedded in the trunk of nerve fibers. Different types of synapses occur in the neuropile area. They meet all morphological criteria of classical chemical synapses. Vacuolated cells occur in the neuroepithelium of E. fraudatrix, but are absent in P. lubricus; their function is unknown. The cells of the non-neuronal epithelia that overlie the ectoneural and hyponeural canals are hypothesized to belong to the same cell type as the supporting cells of the neuroepithelium.  相似文献

Pituitary adenylate cyclase-activating polypeptide (PACAP) has been reported as a strong neurotrophic factor in the various sites of nervous system. The facial nerve injury is one of the common problems in patients at the Otolaryngology since the nerve damage could occur easily due to the anatomical characteristics. Once it happens, the regeneration is little observed and functional recovery is poor. Thus, we investigated that PACAP might have some influence for regeneration after the facial nerve transaction in the guinea pig. PACAP treatment accelerated time for the appearance of compound muscle action potentials (CMAP) after the nerve transaction (first appeared at 1 versus 2 weeks in control) and shortened the latency at 4 weeks. The number of myelinated fibers increased at 4 weeks. Histochemical demonstration of GAP-43, a growth cone protein, was observed at the injury area at 2-4 days. PACAP increased the level of glial cell line-derived neurotrophic factor (GDNF), a neurotrophin, in facial target muscles at 1 day-4 weeks. These data indicated that PACAP promotes the regeneration factors and increases the possibility of functional recovery following the facial nerve injury.  相似文献

An aberrant cutaneous nerve of the thigh arising from the peroneal portion of the human sciatic nerve or common peroneal nerve was observed in 9 cases (4.6% of sides). After giving a branch to the short head of the biceps femoris muscle and a branch to the knee joint, this cutaneous nerve reaches the subcutaneous tissue by passing between the short head of the biceps femoris and the vastus lateralis or by piercing through the biceps femoris. The authors presume that the cutaneous nerve shows the presence of the potential cutaneous nerve routes from the common peroneal nerve to the skin of the lateral aspect of the thigh.  相似文献

The work has been performed on Wistar rats and non-inbred animals. Their ischiatic nerves have been dissected at the femoral superior third under nembutal narcosis. The end of the sectioned nerve are connected by a fragment of an aorta from rats of the same age. The state of nervous elements and dermal epithelium of the hind extremity sole in the animals is studied by means of general histological and neurohistological techniques. Mitotic activity of cells in the plantar epidermis, thickness as a whole and its separate layers are estimated, keratinization coefficient and correlation of thickness of separate sheaths in the whole layer are calculated. Use of the arterial vessels for connecting the end of the cut ischiatic nerve, trophic ulcers, that usually take place after the nerve section, do not develop. At early stages after the operation mitotic activity in the epidermis decreases by 70%, and the layer thickness--by 40%. Restoration of both indices proceeds slowly. As soon as the regenerating nerve fibers reach the distal part of the ischiatic nerve, the state of the epidermis improves: the mitotic activity differs from the normal by 20-30%, and thickness of the epithelium--by 28-30%. Coordination of thickness of separate layers in the epidermis is not nearly disturbed. It remains in the same state up to complete restoration of receptory structures in the rat plantar skin (during 9-9.5 months after the operation).  相似文献

Summary The mean peak CV's of two electrophysiologically defined groups of fibres in the intestinal nerve of the chicken have been determined.One group of fibres is constituted by the processes of enteric cholinergic neurones which project along the side branches of the intestinal nerve and synapse within the nerve trunk. These preganglionic fibres have a mean peak CV (at 40 °C) of 0.31 m·s–1.The other group is made up of fibres of postganglionic neurones which project orally along the nerve trunk. The results suggest that some postganglionic neurones project only as far as the next ganglion whilst others project beyond the next two ganglia for distances greater than 5 mm. The postganglionic fibres have a mean peak CV (at 40 °C) of 0.71 m·s–1.These figures demonstrate that both pre- and postganglionic fibres are unmyelinated. The distribution of myelinated and nonmyelinated nerve fibers of the saphenous nerve of cats in the ventral and dorsal roots of the spinal cord was investigated by methods improving the signal—noise ratio in records of evoked responses from the nerve. The fibers of this nerve enter the spinal cord through roots of segments L4–6. Nerve fibers with conduction velocities of between 80 and 0.38 m/sec were distributed in the dorsal roots of these segments. Four groups of nerve fibers with conduction velocities of 80–60, 40–30, 12.0–3.0, and 1.1–0.51 m/sec, possibly afferent in nature, were found in the ventral roots. The conditions of origin and detection of low-amplitude potentials in the roots of the spinal cord and the probable functional role of the nerve fibers in the ventral roots are discussed.Research Institute of Applied Mathematics and Cybernetics, N.
