Hydrocarbons present on the cuticle surface of adult face flies, Musca autumnalis, were identified by GC-MS and quantified by GC. Hydrocarbons consisted of n-, monomethyl, and dimethyl alkanes ranging in chain length from 23–29 carbons. Also present were monounsaturated alkenes with chain lengths of 23, 25, 27, and 29 carbons. Wild-caught flies were extracted and hydrocarbon profiles determined for both diapausing and reproductive adult males and females. Few qualitative differences were found between males and females in the hydrocarbon profile. Differences in percent composition were found between diapausing and reproductive flies in monounsaturated alkenes, 4 and 45%, n-alkanes, 24 and 37%, monomethylalkanes, 57 and 15%, and dimethylalkanes, 15 and 2%, respectively, for females. A small difference was found in the total amount of hydrocarbon present, 7.3 ± 0.6 and 9.7 ± 1.1 μg/fly, between diapausing and reproductive female flies, respectively. Adult males also exhibited a similar change in hydrocarbon profile and amount between diapausing and reproductive flies. A laboratory strain of face flies originating in Minnesota was also analyzed, and again similar differences were observed in both male and female flies that were kept under a reproductive or diapause condition. The polyphagous shot hole borer (PSHB), Euwallacea sp., was first detected in 2003 in Los Angeles County, California, USA. Recently, this invasive species has become a major pest of many hardwood trees in urban and wildland forests throughout southern California. PSHB is nearly identical in morphology and life history to the tea shot hole borer (TSHB), Euwallacea fornicatus, an invasive pest of hardwoods in Florida, USA and many other parts of the world. However, molecular studies have suggested that the taxa are different species. We conducted morphometric and chemical analyses of the phenotypes of Euwallacea sp. collected in southern California (Los Angeles County) and E. fornicatus collected in Florida (Miami‐Dade County). Our analyses indicated that PSHB has 3 larval instars. The third larval instar was separated from the first 2 instars by head capsule width with 0 probability of misclassification. The body length, head width, and pronotal width of PSHB adult males were significantly less than those of females. Head width and pronotal width of female PSHB were significantly less than those of female TSHB. In contrast, body length, and ratio of body length to pronotal width of female PSHB were significantly greater than those of female TSHB. However, females of these 2 species could not be separated completely by these 4 measurements because of the overlapping ranges. Cuticular hydrocarbons detected in both species were exclusively alkanes (i.e., n‐alkanes, monomethylalkanes, dimethylalkanes, and trimethylalkanes). Cuticular hydrocarbon profiles of PSHB males and females were similar, but they both differed from that of TSHB females. Cuticular hydrocarbons of PSHB were predominantly internally branched dimethylalkanes with backbones of 31 and 33 carbons, whereas cuticular hydrocarbons of TSHB females were dominated by internally branched monomethylalkanes and dimethylalkanes with backbones of 28 and 29 carbons. Multiple compounds within these classes appear to be diagnostic for PSHB and TSHB, respectively.  相似文献

The accumulation and biosynthesis of cuticular and internal hydrocarbons in the Southern armyworm, Spodoptera eridania, were examined at closely timed intervals during larval and pupal development. Gas chromatography-mass spectrometry (GC-MS) was used to identify n-alkanes, monomethylalkanes, and dimethylalkanes ranging in chain length from 23 to 35 carbons. The amount of cuticular hydrocarbon stayed relatively constant during each stadium, while the amount of internal hydrocarbon increased dramatically during the first half of each larval stadium, presumably to replace the cuticular hydrocarbon lost on the shed cast skin with each molt. The accumulation of internal hydrocarbon was mirrored by large increases in the rate of incorporation of labeled acetate into the hydrocarbon fraction. Hydrocarbon production fell to very low rates during the latter part of the fourth and fifth larval stadia. Relatively high rates of hydrocarbon production were observed during the first and last one-third of the pupal stage and essentially all of the hydrocarbons produced during this stage remained internal. These data document large changes in the rates of hydrocarbon production during development in S. eridania and suggest that most of the hydrocarbon produced during each stage was stored internally and then transported to the cuticle of the next stage.  相似文献

Supercooling points (SCPs) and low temperature survival were determined for diapausing and nondiapausing larvae of the ectoparasitoid Nasonia vitripennis. Neither nondiapausing nor diapausing larvae could survive tissue freezing. The SCP profiles were nearly identical for nondiapause-destined (-27 degrees C) and diapausing larvae (-25 degrees C), but these values were not indicative of the lower limits of tolerance in either type of larvae: larvae were killed by chilling at temperatures well above the SCP. Diapausing larvae could withstand low temperature exposures 3-8 times longer than their nondiapausing counterparts. Low temperature survival was enhanced in diapausing and nondiapausing larvae by their encasement within the puparium of the host flesh fly, SARCOPHAGA CRASSIPALPIS: the LT(50)s determined for nondiapausing and diapausing larvae enclosed by fly puparia were 2-3 times higher than values calculated for larvae removed from the puparia. Additional low temperature protection was gained through acquisition of host cryoprotectants during larval feeding: nondiapausing parasitoid larvae that fed on diapausing flesh fly pupae with high levels of glycerol were able to survive exposure to a subzero temperature 4-9 times longer than wasps reared on nondiapausing fly pupae that contained lower quantities of glycerol. Alanine may also contribute to the cold hardiness of N. vitripennis, as evidenced by the fact that larvae feeding on diapausing fly pupae both contained higher concentrations of alanine and exhibited greater cold hardiness. The results thus demonstrate that several critical features of cold hardiness in the wasp are derived from biochemical and physical attributes of the host.  相似文献

Abstract Progeny of Sarcophaga bullata produced from mothers with a history of short day will not enter pupal diapause even if they are reared in a strong diapause-inducing environment (LD 12:12 h at 20oC). Short-day exposure and diapause commitment are normally inseparable, but this maternal effect provides a tool for examining separately the effect of photoperiod and diapause commitment. Duration of the wandering period of the third instar is longer in diapause-destined larvae than in non-diapause-destined larvae, and fecundity of flies that have experienced pupal diapause is lower than in long-day flies that have not been through diapause. The puparia of diapausing pupae contain more hydrocarbons than puparia of nondiapausing pupae, and this contributes to higher rates of net transpiration for the nondiapausing flies. Flies showing the maternal effect (short-day experience but no diapause) show an intermediate response: length of wandering, fecundity rate and quantities of puparial hydrocarbon are between the extremes observed in the other two groups of flies. Thus, the maternal effect switches the developmental programme to nondiapause, but the progeny retain some characteristics of diapause. Evidence from reciprocal crosses indicates that the photoperiodic history of the female, rather than the male, is responsible for the influence on fecundity.  相似文献

The major components of the egg hydrocarbons were dimethylalkanes (40%) and trimethylalkanes (24%) in which the first methyl branch was on carbon 2. The major dimethylalkanes were an approximately 2:1 mixture of 2,10- and 2,6-dimethyloctacosanes in females and eggs. The major trimethylalkanes were a mixture of 2,10,16- and 2,10,18-trimethyloctacosanes. 2,x- and 2,x,y-methyl-branched alkanes with an odd-numbered carbon backbone were proposed to have an even number of carbon atoms between the first and second methyl branch points indicating that their biosynthesis started with a primer derived from leucine. 13,17,21,25-Tetramethylheptatriacontane was the only tetramethylalkane identified. Females and eggs had more hydrocarbons with a 2-methyl branch point than did the males. The eggs had the lowest amount of internally-branched dimethylalkanes but the largest amount of 2,x-dimethylalkanes in their surface hydrocarbons. Only trace amounts of n-alkanes and alkenes were detected in the surface hydrocarbons of adult males and females, larvae and eggs, of the Colorado potato beetle.  相似文献

One critical aspect of an insect's ability to overwinter successfully is the effective management of its water resources. Maintenance of adequate water levels during winter is challenging because of the prevailing low relative humidity at that time of year and the short supply of environmental water that is not in the form of ice. These issues are further exacerbated for insects overwintering as pupae, comprising an immobile stage that is unable to move to new microhabitats if conditions deteriorate. The present study compares the water balance attributes of diapausing and nondiapausing pupae of the corn earworm Helicoverpa zea Boddie, aiming to identify the mechanisms used by diapausing pupae to maintain water balance during winter. Diapausing pupae are 10% larger than nondiapausing individuals. Water loss rates for nondiapausing pupae are low (0.21 mg h?1) and are suppressed (0.01 mg h?1) in diapausing pupae. Cuticular lipids, which serve to waterproof the cuticle and thus suppress cuticular water loss, are more than two‐fold more abundant on the surface of diapausing pupae, and oxygen consumption rates during diapause drop to almost one‐third the rate observed in nondiapausing pupae. Water gain can be accomplished only when atmospheric water content is near saturation or during contact with free water. At moderate relative humidities (20–40%), water loss rates are very low for diapausing pupae, suggesting that these moth pupae have robust mechanisms for combating water loss. The exceptional ability of H. zea to suppress water loss during diapause is probably a result of the combined effects of increased size, more abundant cuticular lipids and decreased metabolic rates.  相似文献

Gas chromatography of cuticular hydrocarbons was used to assess genetic similarity and heterozygosity between 18 sympatric populations of adult Ixodes ricinus ticks collected from Poland. Sixteen n-alkanes, 45 monomethylalkanes, and 36 dimethylalkanes were identified. Forty-one compounds were detected in 90% of the specimens and nine hydrocarbons were present in all the specimens studied. Visual inspection of chromatograms revealed major differences in the relative abundance of hydrocarbons between populations of I. ricinus. The average proportion of heterozygotic hydrocarbons was 50.68%, while the average genetic similarity among all populations was 0.967. Two other methods were used to measure the intrapopulation variability: the number of isomeric forms for each hydrocarbon and the range of hydrocarbon abundance. It was found that some hydrocarbons manifested an unexpectedly high number of isomeric patterns, such as nonacosane (21 patterns), triacontane (23), pentacosane (26), and hexacosane (28). Our data suggest that I. ricinus ticks from Poland consist of several interbreeding, non-isolated populations, with a high degree of genetic flow between them.  相似文献

Gas chromatography-mass spectrometry was used to determine the structures of the methyl branched alkanes of the grasshoppers Melanoplus differentialis (Thomas), M. packardii (Scudder) and M. sanguinipes (Fabricius). The branched hydrocarbons were composed of 2- and 3-methylalkanes and internally branched mono- and di-methylalkanes. The major dimethylalkanes were a homologous series from C33 to C55 and had nine methylenes between the branch points rather than three as reported by Soliday et al. (1974) for M. packardii and M. sanguinipes. Unexpectedly, the homologous series with nine methylenes between branch points did not extend to the C31 dimethylalkanes. The C31 dimethylalkanes were identified as 9,21-dimethylhentriacontane (11 methylenes between the branch points) in M. packardii and M. Sanguinipes and 11,19-dimethylhentriacontane (seven methylenes between the branch points) in M. differentialis.  相似文献

Abstract The age‐dependent cold hardiness profile of Ostrinia nubilalis is compared between nondiapausing and diapausing larvae, as well as with field‐collected larvae. The results suggest that both cold tolerance and accumulation of cryoprotectants depends upon the age of O. nubilalis larva. Late fifth‐instar nondiapausing larvae are more cold tolerant than younger fifth‐instars because they show enhanced ability to withstand sub‐zero temperatures. No appreciable difference is observed between the experimental groups of diapausing larvae as far as their supercooling ability and tolerance at sub‐zero temperatures above the supercooling point. In general, both field‐collected and diapausing larvae are more cold tolerant than nondiapausing larvae, indicating a direct link between diapause and cold hardiness. The age of diapausing larvae affects the ability to accumulate glycerol. Glycerol levels of 45‐day‐old diapausing larvae are significantly higher (2.7‐fold) compared with 90‐day‐old diapausing larvae. Moreover, diapausing larvae display a five‐ to 13‐fold higher glycerol content compared with nondiapausing larvae. There is a trend for an age‐dependent cold hardiness profile in O. nubilalis and further tests that could demonstrate a causal relationship between age and cold tolerance are needed.  相似文献

Abstract  The ecdysteroid levels in hemolymph, ovary, synganglion and whole body of diapausing female Dermacentor niveus were detected by HPLC, and compared with the results of nondiapausing female. It is revealed that the ecdysteroid levels in hemolymph and ovary of diapausing female are similar basically to that of nondiapausing female in the first few days after engorgement. From the 10th day after engorgement, the ecdysteroid levels of diapausing female decreased and even became distinctly lower than that of nondiapausing female. The paucity of ecdysteroids in these individuals would influence the normal development of oocytes. In order to explore the effect of ecdyateroids on the diapausing female, we injected 20-hydroxyecdysone with different dosages at different time into the ticks, and found that after just complete engorgement the injection with large dosages (10000 and 1375 ng/tick) caused death of the ticks. From 10th to 20th day after engorgement the ecdysteroid levels of diapausing female are lower than that of nondiapausing one before oviposition, the injection with certain dosages 50, 70 and 100 ng/tick> of 20E can accelerate vitellogenesis and terminate reproductive diapause, but the amount of eggs produced by them is less than that produced by nondiapausing female. The termination of diapause in female of ixcdid tick by exogenous ecdysteroids is reported for the first time.  相似文献

Three varieties of midseason oranges, viz. Jaffa, Homosassa and Queen, were examined for their saturated and mono-unsaturated hydrocarbon composition in juice sacs. Hydrocarbons were isolated by lipid extraction of the juice sac powders followed by column, thin-layer and AgNO3-TLC. After hydrogenation, the mono-unsaturated fraction and the saturated fraction were analyzed by GC. In the saturated fraction, the dominant linear hydrocarbon was C23 while C25 predominated in the monoene fraction. iso- And anteiso-branched hydrocarbons comprised between 53 and 63 % of the saturated fraction and only 20–26 % of the monoene fraction. Queen differed from Homosassa and Jaffa in the accumulation of higher percentages of saturated iso- and anteiso-branched hydrocarbons and conversely, showed lower percentages for these branched structures in the monoene fraction. Based on the total relative percentages of the three isomeric hydrocarbons, Homosassa could not be differentiated from Jaffa. The overall profiles for these two oranges, however. showed noticeable differences.  相似文献

Abstract A proportion of Helicoverpa armigera collected from fields in Okayama Prefecture (Western Japan; 34.6°N, 134.1°E) does not enter diapause when reared under a short days at 20 °C during the larval stages. However, diapause in such photo‐insensitive individuals can be induced when they are reared at moderately low temperatures, such as 15 °C, regardless of photoperiod. To determine whether such photo‐insensitive individuals can survive overwintering in fields, the present study compares the cold hardiness and sugar content between nondiapausing and diapausing pupae of photo‐insensitive individuals selected over several generations at 20 °C under a short day photoperiod (LD 10 : 14 h). Diapausing and nondiapausing pupae are obtained under the short days by rearing at 15 and 20 °C, respectively, during larval and pupal stages. These pupae are stepwise acclimated at a reduction of 5 °C every 5 days to 0 °C. Maximum survival periods of nondiapausing and diapausing pupae at 0 °C are approximately 30 and 90 days, respectively. Trehalose content in diapausing pupae increases, reaches a maximum level (1.95 mg 100 mg?1 in males and 2.1 mg 100 mg?1 in females) 28 days after exposure to 0 °C and then decreases. On the other hand, glucose content in diapausing pupae increases (maximum level: 0.32 mg 100 mg?1 in males and 0.21 mg 100 mg?1 in females) with decreasing trehalose content 42 days after exposure to 0°C. The decrease in trehalose content and the increase in glucose content may be linked to termination of diapause in H. armigera. These results suggest that, in Japan, the photo‐insensitive individuals can only survive in the mild winters of southern regions, and not in the severe winters of northern regions.  相似文献

Abstract. To understand overwintering of the cotton boll worm Helicoverpa armigera, cold hardiness and sugar content are compared between diapausing and nondiapausing pupae. Diapausing and nondiapausing pupae reared at 20 °C under short and long photoperiods are acclimatized with a reduction of 5 °C per 5 days to 0 °C. When the acclimation temperature reaches 0 °C, the survival of diapausing pupae is assessed. The survival gradually decreases as the period of treatment progresses and approximately half survive for 112 days. However, nondiapausing pupae survive only 14 days after exposure to 0 °C. The surpercooling points of nondiapausing, diapausing and acclimatized pupae are approximately −17 °C. The major sugars contained in pupae are trehalose and glucose. Even though trehalose contents in diapausing pupae (initial level: 0.6 mg 100 mg−1 fresh weight) increase significantly during cold acclimation and continue increasing until 58 days after exposure to 0 °C (maximum level: 1.8 mg 100 mg−1), glucose is maintained at low levels (0.02 mg 100 mg−1) for 56 days at 0 °C. However, glucose contents increase (maximum level: 0.8 mg 100 mg−1) with decreasing contents of trehalose 84 days after exposure to 0 °C. Glycogen content gradually decreases during cold acclimation. When nondiapausing pupae are acclimatized with a reduction of 5 °C per 5 days to 5 °C from the beginning of pupation until the eyespots move, trehalose content increases (maximum level: 1.0 mg 100 mg−1). Glucose contents in nondiapausing pupae increase before eclosion (0.09 mg 100 mg−1). From these results, diapausing pupae of H. armigera can overwinter in regions where average winter temperatures are higher than 0 °C, but nondiapausing pupae cannot.  相似文献

The pentane extract of the social spider, Anelosimus eximius (Araneae, Theridiidae), contains hydrocarbons, fatty acids and their methyl esters, and a series of novel propyl esters of long-chain methyl-branched fatty acids. The propyl esters comprise almost three-fourths of the extract and consist predominantly of odd-numbered carbon chain components. Mass spectrometric analyses of the propyl esters, their methyl esters and cyanide derivatives showed that mono-, di- and trimethyl branched components with methyl branches on even numbered carbons predominate. The major components are propyl 4,20- and 4,30-dimethylhentriacontanoate and propyl 6,20- and 6,30-trimethylhentriacontanoate. The hydrocarbon fraction consists of n-, monomethyl- and dimethylalkanes, containing a relatively high proportion of even-numbered carbon chain components. The abundance of even-numbered carbon chain length alkanes and odd-numbered carbon chain length fatty acyl groups, along with abundant methyl-branches suggest that the propionyl-CoA and its carboxylated product, methylmalonyl-CoA, play important roles in the biosynthesis of these unique waxes. De novo synthesis of contact female sex pheromone and hydrocarbons in Blattella germanica was examined using short in vivo incubations. Accumulation of pheromone on the epicuticular surface and the internal pheromone titer were related to age-specific changes in hydrocarbon synthesis and accumulation in normal and allatectomized females. The incorporation of radiolabel from [1-14C]propionate into the cuticular methyl ketone pheromone fraction was positively related to corpora allata activity during two gonotrophic cycles. During peak pheromone production the total internal lipid fraction contained greater titers of pheromone than the cuticular surface, and it too exhibited a cycle internally, preceding the rise in external pheromone. This suggests that synthesis and accumulation of pheromone internally are followed by transport of pheromone to the epicuticular surface where it accumulates. Radiolabel was incorporated efficiently into both cuticular and internal hydrocarbons after the imaginal molt and until the peak of pheromone synthesis, but it declined to lower levels before ovulation and throughout pregnancy. The internal hydrocarbon titer decreased 58% after oviposition, suggesting deposition in the egg case. It remained relatively unchanged during pregnancy and increased again during the second gonotrophic cycle. In allatectomized females, hydrocarbon synthesis was reduced relative to control females until oviposition in the latter. However, subsequent rates of hydrocarbon synthesis in allatectomized females (without oothecae) exceeded the rates in sham-operated females (with oothecae). In the absence of ovarian uptake of hydrocarbons, the internal titer increased without the decline found in control females at oviposition. As internal hydrocarbons increased, so did cuticular hydrocarbons and both internal and cuticular methyl ketone pheromones. These patterns corresponded well with feeding patterns in sham-operated and allatectomized females, suggesting that pheromone production is normally regulated by stage-specific feeding-induced hydrocarbon synthesis (precursor accumulation internally) and juvenile hormoneinduced conversion of hydrocarbon to pheromone. They also suggest that both the cuticle and the ovaries might be target sites for hydrocarbon and possibly methyl ketone deposition. © 1994 Wiley-Liss, Inc.  相似文献

Sodium [1-14C]acetate, sodium [1-14C]propionate, sodium [2-14C]propionate, sodium [3-14C]propionate and sodium [methyl-14C]methylmalonate were readily incorporated into the cuticular hydrocarbons of nymphal stages of the cockroach Periplaneta fuliginosa both in vivo and in vitro, whereas no incorporation of [methyl-14C]methionine was observed. The alkanes of the nymphal stages of this insect are 25+% n-alkanes, 14% 3-methylalkanes, and 59+% internally branched monomethylalkanes, principally 13-methylpentacosane. Sodium [1-14C]acetate was incorporated into each class of alkane at about its percentage composition. In contrast, labeled sodium propionate and sodium methylmalonate were preferentially incorporated into the branched fractions. Radio-gas-liquid chromatography showed that sodium [1-14C]propionate was incorporated almost exclusively into 3-methyltricosane and 13-methylpentacosane, whereas sodium [1-14C]acetate was incorporated into each glc peak at about its percentage composition. These data suggest that propionate, incorporated during chain elongation, serves as the branching methyl group donor for both the 3-methyl and the internally branched monomethylalkanes in insects. The location of hydrocarbon synthesis in P. fuliginosa was studied using an in vitro tissue slice system. Excised cuticle slices, with adhering fat body tissue removed, gave good incorporation of labeled substrates into the hydrocarbon fraction. No hydrocarbon synthesis was observed in fat body preparations.  相似文献

Analysis of cuticular hydrocarbons from mated and unmated male houseflies, Musca domestica, indicated that only minor changes in composition occurred during the post-eclosion period. In contrast, female hydrocarbon composition changed considerably during the post-eclosion period and was affected by age, mating, and sexual maturation. The cuticular hydrocarbons from sexually mature female houseflies, both mated and unmated, were attractive to male houseflies. Hydrocarbons from unmated male houseflies were not attractive to male houseflies. Hydrocarbons from males held to maturity with females were somewhat attractive to males, probably as a result of physical transfer of the attractant from the females.  相似文献

The cuticular lipids of nine species of beetles (Coleoptera) belonging to different superfamilies were analysed and found to consist of aliphatic hydrocarbons: n-alkanes, 2-, 3-, and other monomethylalkanes as well as dimethylalkanes and alkenes. The hydrocarbon pattern is qualitatively constant at the family level but varies from family to family and thus may be of use in the taxonomy of the insects.  相似文献
