Andrographis paniculata is a grass-shaped medicinal herb, traditionally used in Southeast Asia. The aim of this study was to evaluate the chemoprotective effects of A. paniculata on colorectal cancer. A. paniculata ethanol extract was tested on azoxymethane (AOM)-induced aberrant crypt foci (ACF) in vivo and in vitro. A. paniculata treated groups showed a significant reduction in the number of ACF of the treated rats. Microscopically, ACF showed remarkably elongated and stratified cells, and depletion of the submucosal glands of AOM group compared to the treated groups. Histologically, staining showed slightly elevated masses above the surrounding mucosa with oval or slit-like orifices. Immunohistochemically, expression of proliferating cell nuclear antigen (PCNA) and β-catenin protein were down-regulated in the A. paniculata treated groups compared to the AOM group. When colon tissue was homogenized, malondialdehyde (MDA) and nitric oxide (NO) levels were significantly decreased, whereas superoxide dismutase (SOD) activity was increased in the treated groups compared to the AOM group. A. paniculata ethanol extract showed antioxidant and free radical scavenging activity, as elucidated by the measure of oxidative stress markers. Further, the active fractions were assessed against cell lines of CCD841 and HT29 colon cancer cells.  相似文献

Colorectal cancer (CRC) is the second most common cause of death worldwide. According to the theory by Vogelstein, colorectal carcinogenesis involves a series of successive changes in the normal colonic mucosa, starting with excessive proliferation and focal disorders of intestinal crypts, followed by adenoma and its subsequent malignant transformation. The first identifiable changes in CRC carcinogenesis are aberrant crypt foci (ACF). ACF are invisible during routine colonoscopy yet are well identifiable in chromoendoscopy using methylene blue or indigo carmine. High-resolution colonoscopes are used for assessment of ACF. The aim of the present study was to evaluate the usefulness of standard-resolution colonoscopy for identification of rectal ACF. The following parameters were evaluated: duration of chromoendoscopy of a given rectal segment, type of ACF, sensitivity and specificity of endoscopy combined with histopathological evaluation. The mean duration of colonoscopy and chromoendoscopy was 26.8 min. In the study population, typical ACF were found in 73 patients (p = 0.489), hyperplastic ACF in 49 (p = 0.328), and dysplastic ACF in 16 patients (p = 0.107). Mixed ACF were observed in 11 individuals (p = 0.073). The sensitivity of the method was found to be 0.96 whereas its specificity 0.99.Identification of rectal ACF using standard-resolution colonoscopy combined with rectal mucosa staining with 0.25% methylene blue is characterised by high sensitivity and specificity.  相似文献

Annona muricata has been used in folk medicine for the treatment of cancer and tumors. This study evaluated the chemopreventive properties of an ethyl acetate extract of A. muricata leaves (EEAML) on azoxymethane-induced colonic aberrant crypt foci (ACF) in rats. Moreover, the cytotoxic compound of EEAML (Annomuricin E) was isolated, and its apoptosis-inducing effect was investigated against HT-29 colon cancer cell line using a bioassay-guided approach. This experiment was performed on five groups of rats: negative control, cancer control, EEAML (250 mg/kg), EEAML (500 mg/kg) and positive control (5-fluorouracil). Methylene blue staining of colorectal specimens showed that application of EEAML at both doses significantly reduced the colonic ACF formation compared with the cancer control group. Immunohistochemistry analysis showed the down-regulation of PCNA and Bcl-2 proteins and the up-regulation of Bax protein after administration of EEAML compared with the cancer control group. In addition, an increase in the levels of enzymatic antioxidants and a decrease in the malondialdehyde level of the colon tissue homogenates were observed, suggesting the suppression of lipid peroxidation. Annomuricin E inhibited the growth of HT-29 cells with an IC50 value of 1.62 ± 0.24 μg/ml after 48 h. The cytotoxic effect of annomuricin E was further substantiated by G1 cell cycle arrest and early apoptosis induction in HT-29 cells. Annomuricin E triggered mitochondria-initiated events, including the dissipation of the mitochondrial membrane potential and the leakage of cytochrome c from the mitochondria. Prior to these events, annomuricin E activated caspase 3/7 and caspase 9. Upstream, annomuricin E induced a time-dependent upregulation of Bax and downregulation of Bcl-2 at the mRNA and protein levels. In conclusion, these findings substantiate the usage of A. muricata leaves in ethnomedicine against cancer and highlight annomuricin E as one of the contributing compounds in the anticancer activity of A. muricata leaves.  相似文献

To express a human lactoferrin (hLF) gene in Bacteroides uniformis, a member of the anaerobic microflora in the colon, we constructed a recombinant plasmid, pVLFK, by subcloning hLF cDNA to an Escherichia coli–Bacteroides shuttle vector, pVAL-1. The plasmid pVLFKwas transferred to B. uniformis strain BU1001 by the filter mating procedure creating strain TCTK101. The lactoferrin protein in B. uniformis strain TCTK101 was detected by Western blot analysis with an anti-human lactoferrin monoclonal antibody. A culture of strain TCTK101 inhibited the growth ofE. coli strain HB101 in vitro compared to a culture of strain TCTK11, which is a B. uniformis strain-carrying plasmid pVAL-1, suggesting that the lactoferrin produced from strain TCTK101 possesses biological activity. To determine the effect of lactoferrin-producing B. uniformis on the formation of azoxymethane-induced aberrant crypt foci (ACF), putative neoplastic lesions, overnight cultures of strains TCTK11 and TCTK101 were given to rats as drinking water. The numbers of ACF and ACF having more than three crypts per focus five weeks after the beginning of the experiment significantly increased in the group treated by a culture of strain TCTK11 compared with those in the non-treated water group. However, rats treated with a culture of strain TCTK101 carrying plasmid pVLFK showed a significantly lower number of ACF than rats with a culture of strain TCTK11 (34% reduction), suggesting that hLF which is produced in a prokaryotic expression system prevents formation of ACF in the rat colon.  相似文献

目的:探讨益气健脾方对大肠癌模型异常腺窝病灶(ACF)的抑制效应及其相关机制。方法:50只Wistar大鼠随机分为5组:中药低剂量组(将中药溶液稀释3倍后,5 mL/kg日灌胃量)、中药中剂量组(5 mL/kg日灌胃量)、中药高剂量组(15 m L/kg日灌胃量)、模型组、正常组,每组10只大鼠,采用免疫组化法、美兰染色后镜下观察大肠组织。结果:各中药组ACF及大型ACF的数量较模型组减少(P0.05),并且中药中剂量组ACF数量减少最为明显,差异具有统计学意义(P0.05)。造模后24h、48h中药组肠腺细胞PCNA增殖指数低于模型组,差异具有统计学意义(P0.05)。造模后24 h、48 h中药组肠腺细胞凋亡指数高于模型组,差异具有统计学意义(P0.05)。中药组β-catenin异位表达均低于模型组,并且中药组β-catenin异位表达中高剂量组最高,低剂量组次之,中剂量组最低(P0.05)。中药组MMP-7的表达均低于模型组,而且各中药组中低剂量MMP-7表达最高,高剂量组次之,中剂量组最低(P0.05)。结论:益气健脾方可以明显降低Wistar大鼠ACF的发生数量,抑制大肠癌发生中Wnt信号的激活,在一定程度上减少了大肠癌的发生,起到了预防作用。  相似文献

Chemical investigation of Acanthus ilicifolius var. xiamenensis led to the isolation of eleven compounds, and their structures were identified to be 2-benzoxazolinone (1), 2-hydroxy-2H-1,4-benzoxazin-3(4H)-one (2), (2R)-2-O-β-d-glucopyranosyl-2H-1,4-benzox azin-3(4H)-one (3), (2R)-2-O-β-d-glucopyranosyl-4-hydroxy-2H-1,4-benzoxazin-3(4H)-one (4), (2R)-2-O-β-d-glucopyranosyl-7-hydroxy-2H-1,4-benzoxazin-3(4H)-one (5), lyoniside (6), 3′-methoxy-luteolin-7-O-β-d-glucopyranoside (7), β-sitosterol-3-O-β-d-glucopyranoside (8), stigmasterol octadecanoate (9), β-sitosterol octadecanoate (10), stigmasterol-3-O-β-d-glucopyranoside (11) on the basis of mass and NMR spectra. This is the first report on the occurrence of compound 6 and 7 in Acanthaceae. This work also represents the first phytochemical work on the roots of A. ilicifolius var. xiamenensis.  相似文献

Evidence suggests that selenium has cancer preventive properties that are largely mediated through selenoproteins. Our previous observations demonstrated that targeted down-regulation of the 15 kDa selenoprotein (Sep15) in murine colon cancer cells resulted in the reversal of the cancer phenotype. The present study investigated the effect of Sep15 knockout in mice using a chemically-induced colon cancer model. Homozygous Sep15 knockout mice, and wild type littermate controls were given four weekly subcutaneous injections of azoxymethane (10 mg/kg). Sep15 knockout mice developed significantly (p<0.001) fewer aberrant crypt foci than controls demonstrating that loss of Sep15 protects against aberrant crypt foci formation. Dietary selenium above adequate levels did not significantly affect aberrant crypt foci formation in Sep15 knockout mice. To investigate molecular targets affected by loss of Sep15, gene expression patterns in colonic mucosal cells of knockout and wild type mice were examined using microarray analysis. Subsequent analyses verified that guanylate binding protein-1 (GBP-1) mRNA and protein expression were strongly upregulated in Sep15 knockout mice. GBP-1, which is expressed in response to interferon-γ, is considered to be an activation marker during inflammatory diseases, and up-regulation of GBP-1 in humans has been associated with a highly significant, increased five-year survival rate in colorectal cancer patients. In agreement with these studies, we observed a higher level of interferon-γ in plasma of Sep15 knockout mice. Overall, our results demonstrate for the first time, that Sep15 knockout mice are protected against chemically-induced aberrant crypt foci formation and that Sep15 appears to have oncogenic properties in colon carcinogenesis in vivo.  相似文献

在温室内用自动控制装置模拟正规半日潮淹浸作用对老鼠簕种子发芽与幼苗生长的影响,设淹水0、3、6、9、12、15、18、21和24h·d-1共9个淹水时间梯度,人工海水盐度为2%,测定不同淹水时间下老鼠簕种子萌发、幼苗株高、叶面积、生物量以及幼苗叶脯氨酸含量、丙二醛(MDA)含量、叶绿素含量和超氧化物歧化酶(SOD)活性等生理生化指标.结果表明,长时间淹水(≥15 h·d-1)会推迟老鼠簕种子萌发的时间,并使发芽率和成苗率降低,淹水<15 h·d-1适宜老鼠簕种子的萌发.随着淹水时间的增长,脯氨酸含量、MDA含量和SOD活性均先下降后上升,叶绿素含量则逐渐降低.淹水≥3 h·d-1,幼苗生物量、单株叶面积、高生长和叶绿素含量与淹水时间均呈负相关.淹水0~3h·d-1,老鼠簕总叶绿素含量、根、茎、地上部分和总生物量积累均显著大于其他处理,淹水≥18 h·d-1,老鼠筋幼苗总生物量、叶面积、叶绿素含量显著降低,根冠比开始上升.在上述人工培养条件下,淹水0~3h·d-1是老鼠筋幼苗生长的最佳淹水时间,18h·d-1是老鼠簕幼苗生长的一个临界淹水时间,淹水≥18 h·d-1老鼠筋幼苗生长显著受到抑制.  相似文献

【目的】比较不同红树林地区的老鼠簕内生放线菌的地理分布,了解内生放线菌与其所处环境的相关性。【方法】分别从5个不同地点的红树林采集老鼠簕全株植物,采用9种分离培养基,从植株不同部位分离内生放线菌,用16S rRNA基因序列分析鉴定到属,用添加不同NaCl浓度的ISP 2液体培养基进行耐盐度测试,用无氮基础培养基进行固氮活性测试。【结果】共分离得到内生放线菌52株,其中从叶、茎和根部分别获得5株、2株和45株,花和果中未分离到。52株内生放线菌分别属于小单孢菌属(47株),链霉菌属(3株),疣孢菌属(1株)和继生菌属(1株)。48株菌表现出耐盐或嗜盐特征,其中18株最高耐盐度20%,4株不能在无盐条件下生长,12株菌可在含有3.3%NaCl的培养基上生长良好。4株菌可在无氮培养基下生长。【结论】对47株内生小单孢菌的地理分布分析表明,老鼠簕内生小单孢菌的类群因不同地理位置有很大差异。耐盐和固氮活性测试结果表明了老鼠簕内生放线菌对环境的适应性。  相似文献

Abstract

Effect of cadmium (Cd) on the primary metabolic activities and elemental distribution in roots was explored in Acanthus ilicifolius L., a halophyte with phytostabilization potential. The rate of photosynthesis decreased in the CdCl2 treated plants and this reduction was mainly attributed to the reduction of leaf area, photosynthetic pigments, impaired gaseous exchange caused by the stomatal closure and tissue water status. However, respiration rate was significantly higher in the CdCl2 treated plants which aid the plant with additional energy required for the metabolic activities. Distribution of essential elements in the roots exhibited significant differences from that of control, which indicate the nutritional adaptation developed by A. ilicifolius under the influence of toxic metal ions. Thus, Cd toxicity is neutralized through the resource allocation from the growth process to processes that increase the fitness of the plant to encounter adverse environmental condition. In addition, the absorbed Cd is retained in the cortical cells of root thereby preventing the upward movement to shoot thereby making the plant a potential candidate for phytostabilization of Cd.  相似文献

An apical membrane ouabain-sensitive H-K exchange and a barium-sensitive basolateral membrane potassium channel are present in colonic crypt cells and may play a role in both K absorption and intracellular pH (pHi) regulation. To examine the possible interrelationship between apical membrane H-K exchange and basolateral membrane K movement in rat distal colon in the regulation of pHi, experiments were designed to assess whether changes in extracellular potassium can alter pHi. pHi in isolated rat crypts was determined using microspectrofluorimetric measurements of the pH-sensitive dye BCECF-AM (2′,7′-bis(carboxyethyl-5(6)-carboxy-fluorescein acetoxy methylester). After loading with the dye, crypts were superfused with a Na-free solution which resulted in a rapid and reversible fall in pHi (7.36 ± 0.02 to 6.98 ± 0.03). Following an increase in extracellular [K] to 20 mm, in the continued absence of Na, there was a further decrease in pHi (0.20 ± 0.02, P < 0.01). K-induced acidification was blocked both by 2 mm bath barium, a K channel blocker, and by 0.5 mm lumen ouabain. K-induced acidification was also observed when intracellular acidification was induced by a NH4Cl prepulse. These observations suggest that increased basolateral K movement increases intracellular [K] resulting in a decrease in pHi that is mediated by a ouabain-sensitive apical membrane H,K-ATPase. Our results demonstrate an interrelationship between basolateral K movement and apical H-K exchange in the regulation of pHi and apical K entry in rat distal colon. An endophytic fungus (Talaromyces stipitatus SK-4) was isolated from the leaves of a mangrove plant Acanthus ilicifolius. Its crude extract exhibited significant antibacterial activity and was purified to afford two new depsidones, talaromyones A and B (1 and 2), along with five known depsidone analogs (3–7). Their structures, including absolute configuration, were elucidated through extensive spectroscopic data analysis and modified Mosher's method. Compound 2 showed antibacterial activity against Bacillus subtilis with an MIC value of 12.5 μg/mL. In the inhibitory assay against α-glucosidase, compounds 2, 4 and 5 displayed moderate activities with IC50 values ranging from 48.4 to 99.8 μM.  相似文献

The use of architectural and morphological characteristics of cells for establishing prognostic indicators by which individual pathologies are assigned grade and stage is a well-accepted practice. Advances in automated micro- and macroscopic image acquisition and digital image analysis have created new opportunities in the field of prognostic assessment; but, one area in experimental pathology, animal models for colon cancer, has not taken advantage of these opportunities. This situation is primarily due to the methods available to evaluate the colon of the rodent for the presence of premalignant and malignant pathologies. We report a new method for the excision and processing of the entire colon of the rat and illustrate how this procedure permitted the quantitative assessment of aberrant crypt foci (ACF), a premalignant colon pathology, for characteristics consistent with progression to malignancy. ACF were detected by methylene blue staining and subjected to quantitative morphometric analysis. Colons were then restained with high iron diamine–alcian blue for assessment of mucin depletion using an image overlay to associate morphometric data with mucin depletion. The subsequent evaluation of ACF for beta-catenin staining is also demonstrated. The methods described are particularly relevant to the screening of compounds for cancer chemopreventive activity.  相似文献

One, three, seven, and six of polymorphic chloroplast microsatellite (cpSSR) markers were developed from four mangrove species, Acanthus ilicifolius, Aegiceras corniculatum, Avicennia marina, and Lumnitzera racemosa, respectively. Characterization of 229, 509, 369, and 216 individuals of A. ilicifolius, A. corniculatum, A. marina, and L. racemosa, collected from different natural mangrove populations (A. ilicifolius, 6; A. corniculatum, 14; A. marina, 10; L. racemosa, 6) in the southern coastline of China showed that these loci provide cpSSR markers with polymorphisms ranging from two to four alleles per locus and gene diversity between 0.005 and 0.675. Combining the polymorphic cpSSR loci of each species, 3, 5, 11, and 4 of cpSSR haplotypes were separately detected in populations of A. ilicifolius, A. corniculatum, A. marina, and L. racemosa in the southern coastline of China. These cpSSR markers will be useful for analyzing the maternal lineage distributions and population genetic structures of these four species.  相似文献

 Mangroves, the intertidal ecosystems occurring primarily in the tropical regions of the world, are valuable natural resources with high productivity and unique habitat value. However, the genetic structure of plant species within the mangrove ecosystem is poorly understood. The present communication is the first report on the use of molecular markers in assessing intra-site and intra-specific polymorphism in one of the mangrove species, Acanthus ilicifolius, for identifying/ detecting distinct genotypes for long-term conservation. Random amplified polymorphic DNAs (RAPDs) and restriction fragment length polymorphisms (RFLPs) were used to elucidate the intra- and inter-population variability in this widely distributed mangrove species. In all, 48 genotypes representing eight distinct populations were analysed. A low level of polymorphism was detected at the intra-population level through both RAPD (3.8–7.3%) and RFLP (3.2–9.1%) analyses. At the inter-population level, 25 of the 73 RAPD loci (34%) detected through the use of 13 random primers and 44 of the 96 RFLP loci (45.8%) revealed through 15 probe/enzyme combinations were polymorphic. RFLP analyses were carried out using genomic clones developed from the same species. The somatic cells of the species displayed 48 chromosomes, with no numerical changes at either intra- or inter- population levels. Received: 5 June 1996 / Accepted: 8 November 1996  相似文献

We examined effects of a plant polyphenolic compound, curcumin, against fluoride-induced oxidative stress in the rat brain. Five experimental groups of male rats (10 animals each) were compared. Animals of these experimental groups were treated with curcumin (10 and 20 mg/kg body mass), vitamin C (10 mg/kg), and sample solvent (0.5 ml) for a week prior to sodium fluoride intoxication. After treatment, rats of the experimental groups, except for the normal control group, were intoxicated with sodium fluoride (600 ppm through drinking water) for a week. Then, brains were collected and homogenized, and activities of superoxide dismutase and catalase and levels of reduced glutathione and lipid peroxidation final products were evaluated in the brain tissue homogenates. Treatment with curcumin prior to fluoride intoxication significantly normalized the above biochemical parameters; the intensity of protective effects of 20 mg/kg curcumin was close to that of vitamin C.  相似文献

Three new β-resorcylic acid derivatives, compounds 1–3, along with six known analogues (49) were isolated from an endophytic fungus Lasiodiplodia sp. ZJ-HQ1 derived from medicinal plant Acanthus ilicifolius. Their structures were elucidated by 1D and 2D NMR spectroscopic analysis, high resolution mass spectrometric (HREIMS) data, and X-ray crystallography. The absolute configurations of compounds 1 and 2 were determined by the modified Mosher’s method. Compounds 1–7 showed more potent inhibitory effects against α-glucosidase activity than the clinical α-glucosidase inhibitor acarbose.  相似文献
