Macroalgae has bloomed in the brackish lake of Shenzhen Bay, China continuously from 2010 to 2014. Gracilaria tenuistipitata was identified as the causative macroalgal species. The aim of this study was to explore the outbreak mechanism of G. tenuistipitata, by studying the effects of salinity and nitrogen sources on growth, and the different nitrogen sources uptake characteristic. Our experimental design was based on environmental conditions observed in the bloom areas, and these main factors were simulated in the laboratory. Results showed that salinity 12 to 20 ‰ was suitable for G. tenuistipitata growth. When the nitrogen sources'' (NH4 +, NO3 ) concentrations reached 40 µM or above, the growth rate of G. tenuistipitata was significantly higher. Algal biomass was higher (approximately 1.4 times) when cultured with NH4 + than that with NO3 addition. Coincidentally, macroalgal bloom formed during times of moderate salinity (∼12 ‰) and high nitrogen conditions. The NH4 + and NO3 uptake characteristic was studied to understand the potential mechanism of G. tenuistipitata bloom. NH4 + uptake was best described by a linear, rate-unsaturated response, with the slope decreasing with time intervals. In contrast, NO3 uptake followed a rate-saturating mechanism best described by the Michaelis-Menten model, with kinetic parameters Vmax = 37.2 µM g−1 DM h−1 and Ks = 61.5 µM. Further, based on the isotope 15N tracer method, we found that 15N from NH4 + accumulated faster and reached an atom% twice than that of 15N from NO3 , suggesting when both NH4 + and NO3 were available, NH4 + was assimilated more rapidly. The results of the present study indicate that in the estuarine environment, the combination of moderate salinity with high ammonium may stimulate bloom formation.  相似文献

We compared growth kinetics of Prorocentrum donghaiense cultures on different nitrogen (N) compounds including nitrate (NO3 ), ammonium (NH4 +), urea, glutamic acid (glu), dialanine (diala) and cyanate. P. donghaiense exhibited standard Monod-type growth kinetics over a range of N concentraions (0.5–500 μmol N L−1 for NO3 and NH4 +, 0.5–50 μmol N L−1 for urea, 0.5–100 μmol N L−1 for glu and cyanate, and 0.5–200 μmol N L−1 for diala) for all of the N compounds tested. Cultures grown on glu and urea had the highest maximum growth rates (μm, 1.51±0.06 d−1 and 1.50±0.05 d−1, respectively). However, cultures grown on cyanate, NO3 , and NH4 + had lower half saturation constants (Kμ, 0.28–0.51 μmol N L−1). N uptake kinetics were measured in NO3 -deplete and -replete batch cultures of P. donghaiense. In NO3 -deplete batch cultures, P. donghaiense exhibited Michaelis-Menten type uptake kinetics for NO3 , NH4 +, urea and algal amino acids; uptake was saturated at or below 50 μmol N L−1. In NO3 -replete batch cultures, NH4 +, urea, and algal amino acid uptake kinetics were similar to those measured in NO3 -deplete batch cultures. Together, our results demonstrate that P. donghaiense can grow well on a variety of N sources, and exhibits similar uptake kinetics under both nutrient replete and deplete conditions. This may be an important factor facilitating their growth during bloom initiation and development in N-enriched estuaries where many algae compete for bioavailable N and the nutrient environment changes as a result of algal growth.  相似文献

To date, few studies are conducted to quantify the effects of reduced ammonium (NH4 +) and oxidized nitrate (NO3 ) on soil CH4 uptake and N2O emission in the subtropical forests. In this study, NH4Cl and NaNO3 fertilizers were applied at three rates: 0, 40 and 120 kg N ha−1 yr−1. Soil CH4 and N2O fluxes were determined twice a week using the static chamber technique and gas chromatography. Soil temperature and moisture were simultaneously measured. Soil dissolved N concentration in 0–20 cm depth was measured weekly to examine the regulation to soil CH4 and N2O fluxes. Our results showed that one year of N addition did not affect soil temperature, soil moisture, soil total dissolved N (TDN) and NH4 +-N concentrations, but high levels of applied NH4Cl and NaNO3 fertilizers significantly increased soil NO3 -N concentration by 124% and 157%, respectively. Nitrogen addition tended to inhibit soil CH4 uptake, but significantly promoted soil N2O emission by 403% to 762%. Furthermore, NH4 +-N fertilizer application had a stronger inhibition to soil CH4 uptake and a stronger promotion to soil N2O emission than NO3 -N application. Also, both soil CH4 and N2O fluxes were driven by soil temperature and moisture, but soil inorganic N availability was a key integrator of soil CH4 uptake and N2O emission. These results suggest that the subtropical plantation soil sensitively responses to atmospheric N deposition, and inorganic N rather than organic N is the regulator to soil CH4 uptake and N2O emission.  相似文献

The nitrogen (N) uptake kinetic parameters for Microcystis field assemblages collected from the San Francisco Bay Delta (Delta) in 2012 and non-toxic and toxic laboratory culture strains of M. aeruginosa were assessed. The 15N tracer technique was used to investigate uptake of ammonium (NH4+), nitrate (NO3), urea and glutamic acid over short-term incubations (0.5–1 h), and to study inhibition of NO3, NH4+ and urea uptake by NH4+, NO3 and NH4+, respectively. This study demonstrates that Delta Microcystis can utilize different forms of inorganic and organic N, with the greatest capacity for NH4+ uptake and the least for glutamic acid uptake, although N uptake did not always follow the classic Michaelis–Menten hyperbolic relationship at substrate concentrations up to 67 μmol N L−1. Current ambient N concentrations in the Delta may be at sub-saturating levels for N uptake, indicating that if N loading (especially NH4+) were to increase, Delta Microcystis assemblages have the potential for increased N uptake rates. Delta Microcystis had the highest specific affinity, α, for NH4+ and the lowest for NO3. In culture, N uptake by non-toxic and toxic M. aeruginosa strains was much higher than from the field, but followed similar N utilization trends to those in the field. Neither strain showed severe inhibition of NO3 uptake by NH4+ or inhibition of NH4+ uptake on NO3, but both strains showed some inhibition of urea uptake by NH4+.  相似文献

The net influx (uptake) rates of NO3, NH4+, NO2, and urea into roots of wheat (Triticum aestivum cv Yecora Rojo) seedlings from complete nutrient solutions containing all four compounds were monitored simultaneously. Although urea uptake was too slow to monitor, its presence had major inhibitory effects on the uptake of each of the other compounds. Rates of NO3, NH4+, and NO2 uptake depended in a complex fashion on the concentration of all four N compounds. Equations were developed which describe the uptake rates of each of the compounds, and of total N, as functions of concentrations of all N sources. Contour plots of the results show the interactions over the range of concentrations employed. The coefficients of these equations provide quantitative values for evaluating primary and interactive effects of each compound on N uptake.  相似文献

Assimilation of NO3 and NH4+ by perennial ryegrass (Lolium perenne L.) turf, previously deprived of N for 7 days, was examined. Nitrogen uptake rate was increased up to four- to five-fold for both forms of N by N-deprivation as compared to N-sufficient controls, with the deficiency-enhanced N absorption persisting through a 48 hour uptake period. Nitrate, but not NH4+, accumulated in the roots and to a lesser degree in shoots. By 48 hours, 53% of the absorbed NO3 had been reduced, whereas 97% of the NH4+ had been assimilated. During the early stages (0 to 8 hours) of NO3 uptake by N-deficient turf, reduction occurred primarily in the roots. Between 8 and 16 hours, however, the site of reduction shifted to the shoots. Nitrogen form did not affect partitioning of the absorbed N between roots (40%) and shoots (60%) but did affect growth. Compared to NO3, NH4+ uptake inhibited root, but not shoot, growth. Total soluble carbohydrates decreased in both roots and shoots during the uptake period, principally the result of fructan metabolism. Ammonium uptake resulted in greater total depletion of soluble carbohydrates in the root compared to NO3 uptake. The data indicate that N assimilation by ryegrass turf utilizes stored sugars but is also dependent on current photosynthate.  相似文献

Ricinus communis L. plants were grown in nutrient solutions in which N was supplied as NO3 or NH4+, the solutions being maintained at pH 5.5. In NO3-fed plants excess nutrient anion over cation uptake was equivalent to net OH efflux, and the total charge from NO3 and SO42− reduction equated to the sum of organic anion accumulation plus net OH efflux. In NH4+-fed plants a large H+ efflux was recorded in close agreement with excess cation over anion uptake. This H+ efflux equated to the sum of net cation (NH4+ minus SO42−) assimilation plus organic anion accumulation. In vivo nitrate reductase assays revealed that the roots may have the capacity to reduce just under half of the total NO3 that is taken up and reduced in NO3-fed plants. Organic anion concentration in these plants was much higher in the shoots than in the roots. In NH4+-fed plants absorbed NH4+ was almost exclusively assimilated in the roots. These plants were considerably lower in organic anions than NO3-fed plants, but had equal concentrations in shoots and roots. Xylem and phloem saps were collected from plants exposed to both N sources and analyzed for all major contributing ionic and nitrogenous compounds. The results obtained were used to assist in interpreting the ion uptake, assimilation, and accumulation data in terms of shoot/root pH regulation and cycling of nutrients.  相似文献

The effect of exogenous NH4+ on NO3 uptake and in vivo NO3 reductase activity (NRA) in roots of Phaseolus vulgaris L. cv Witte Krombek was studied before, during, and after the apparent induction of root NRA and NO3 uptake. Pretreatment with NH4Cl (0.15-50 millimolar) affected neither the time pattern nor the steady state rate of NO3 uptake.

When NH4+ was given at the start of NO3 nutrition, the time pattern of NO3 uptake was the same as in plants receiving no NH4+. After 6 hours, however, the NO3 uptake rate (NUR) and root NRA were inhibited by NH4+ to a maximum of 45% and 60%, respectively.

The response of the NUR of NO3-induced plants depended on the NH4Cl concentration. Below 1 millimolar NH4+, the NUR declined immediately and some restoration occurred in the second hour. In the third hour, the NUR became constant. In contrast, NH4+ at 2 millimolar and above caused a rapid and transient stimulation of NO3 uptake, followed again by a decrease in the first, a recovery in the second, and a steady state in the third hour. Maximal inhibition of steady state NUR was 50%. With NO3-induced plants, root NRA responded less and more slowly to NH4+ than did NUR.

Methionine sulfoximine and azaserine, inhibitors of glutamine synthetase and glutamate synthase, respectively, relieved the NH4+ inhibition of the NUR of NO3-induced plants. We conclude that repression of the NUR by NH4+ depends on NH4+ assimilation. The repression by NH4+ was least at the lowest and highest NH4+ levels tested (0.04 and 25 millimolar).

The influence of NH4+, in the external medium, on fluxes of NO3 and K+ were investigated using barley (Hordeum vulgare cv Betzes) plants. NH4+ was without effect on NO3 (36ClO3) influx whereas inhibition of net uptake appeared to be a function of previous NO3 provision. Plants grown at 10 micromolar NO3 were sensitive to external NH4+ when uptake was measured in 100 micromolar NO3. By contrast, NO3 uptake (from 100 micromolar NO3) by plants previously grown at this concentration was not reduced by NH4+ treatment. Plants pretreated for 2 days with 5 millimolar NO3 showed net efflux of NO3 when roots were transferred to 100 micromolar NO3. This efflux was stimulated in the presence of NH4+. NH4+ also stimulated NO3 efflux from plants pretreated with relatively low nitrate concentrations. It is proposed that short term effects on net uptake of NO3 occur via effects upon efflux. By contrast to the situation for NO3, net K+ uptake and influx of 36Rb+-labeled K+ was inhibited by NH4+ regardless of the nutrient history of the plants. Inhibition of net K+ uptake reached its maximum value within 2 minutes of NH4+ addition. It is concluded that the latter ion exerts a direct effect upon K+ influx.  相似文献

A prolific maize (Zea mays L.) genotype was grown to physiological maturity under greenhouse conditions to examine the effects of reproductive sink demand on (a) the remobilization of N accumulated during vegetative growth, and (b) the partitioning of N accumulated concurrent with ear development. One- and two-eared plants were treated with either a NO3 or NH4+ source of 15N-labeled N during reproductive growth. Plants with two ears enhanced grain production, N remobilization from the stalk and roots, and N translocation to the grain from concurrently assimilated N. But, remobilization of leaf-N was unaffected by ear number. In addition, N uptake and total dry matter accumulation during the reproductive period were also unaffected, although P uptake was greater in the two-eared plants. Less than 15% of the total K+ uptake was accumulated after silking while during this time more than 40% of the total N and more than 50% of the total P were absorbed. The data also indicate that with NO3 nutrition, internal recirculation of K+ between shoots and roots may play a prominent role in the transport of nitrogenous solutes during grain development. N source had no effect on dry matter production and N uptake of both one- and two-eared plants. However, slightly greater partitioning of labeled-N from the NH4+ source to the grain was observed in the two-eared plants.  相似文献

Following an invasion of exotic annual grasses into California oak woodlands, grass species dominance shifted from native perennials to exotic annuals. In combination with other ecosystem and species characteristics, species-specific N preferences may influence species coexistence and dominance. If species N preferences follow dominance patterns in California oak woodlands, then the more dominant exotic grasses should prefer the most abundant inorganic soil N form (NH4+), while the subordinate native grasses should prefer the less available inorganic (NO3?) or organic (glycine) soil N forms. To investigate this prediction, we applied 15N-labeled NH4+, NO3?, and glycine to soil and measured % 15N recovery by two dominant annual grasses (Bromus diandrus and Bromus hordeaceus) and two subordinate perennial grasses (Elymus glaucus and Nassella pulchra). As expected, shoots of B. diandrus recovered more 15N-NH4+ (74%) than either 15N-NO3? (51%) or 15N-glycine (39%). B. diandrus also captured at least 3.2 times more 15N-NH4+ than subordinate grasses. Dominant B. hordeaceus, however, demonstrated no N form preferences. As hypothesized, shoots of subordinate E. glaucus and N. pulchra recovered 2.1–2.3 times more 15N-NO3? than 15N-NH4+ and increased %N by 4.8–5.7% in response to the application of 15N-NO3?. Both subordinate grasses did not prefer 15N-glycine over 15N-NH4+, suggesting that the importance of this N form in structuring species coexistence in California oak woodlands is minimal. These results support our theory that species N preferences follow dominance patterns in California oak woodlands. To further understand the role of these species-specific N preferences in structuring dominance, the importance of N form versus such characteristics as rooting distribution and species phenologies needs to be explored in the presence of interspecific competition.  相似文献

At root temperature below 14 C the absorption of 15N from NH4+ greatly exceeded that from NO2 by tillers of Lolium multiflorum and Lolium perenne under conditions where pH, external concentration, plant N status, and pretreatment temperature were varied. There was a marked increase in the temperature sensitivity of NO3 transport below 14 C, irrespective of the temperature at which plants were grown previously. A marked increase in the temperature sensitivity was also seen for NH4+ transport, but this occurred at the lower temperature of 10 C. Pretreatment of roots at 8 C lowered this still further to 5 C. Above and below these transition temperatures the Q10 values for NO3 and NH4+ transport were similar. Thus, the increased absorption of NH4+ relative to NO3 at low temperatures seems to be related primarily to the difference in transition temperatures.  相似文献

Nitrogen (N) is an essential nutrient in the sea and its distribution is controlled by microorganisms. Within the N cycle, nitrite (NO2) has a central role because its intermediate redox state allows both oxidation and reduction, and so it may be used by several coupled and/or competing microbial processes. In the upper water column and oxygen minimum zone (OMZ) of the eastern tropical North Pacific Ocean (ETNP), we investigated aerobic NO2 oxidation, and its relationship to ammonia (NH3) oxidation, using rate measurements, quantification of NO2-oxidizing bacteria via quantitative PCR (QPCR), and pyrosequencing. 15NO2 oxidation rates typically exhibited two subsurface maxima at six stations sampled: one located below the euphotic zone and beneath NH3 oxidation rate maxima, and another within the OMZ. 15NO2 oxidation rates were highest where dissolved oxygen concentrations were <5 μM, where NO2 accumulated, and when nitrate (NO3) reductase genes were expressed; they are likely sustained by NO3 reduction at these depths. QPCR and pyrosequencing data were strongly correlated (r2=0.79), and indicated that Nitrospina bacteria numbered up to 9.25% of bacterial communities. Different Nitrospina groups were distributed across different depth ranges, suggesting significant ecological diversity within Nitrospina as a whole. Across the data set, 15NO2 oxidation rates were decoupled from 15NH4+ oxidation rates, but correlated with Nitrospina (r2=0.246, P<0.05) and NO2 concentrations (r2=0.276, P<0.05). Our findings suggest that Nitrospina have a quantitatively important role in NO2 oxidation and N cycling in the ETNP, and provide new insight into their ecology and interactions with other N-cycling processes in this biogeochemically important region of the ocean.  相似文献

An experiment was conducted to investigate the relative changes in NO3 assimilatory processes which occurred in response to decreasing carbohydrate availability. Young tobacco plants (Nicotiana tabacum [L.], cv NC 2326) growing in solution culture were exposed to 1.0 millimolar 15NO3 for 6 hour intervals during a normal 12 hour light period and a subsequent period of darkness lasting 42 hours. Uptake of 15NO3 decreased to 71 to 83% of the uptake rate in the light during the initial 18 hours of darkness; uptake then decreased sharply over the next 12 hours of darkness to 11 to 17% of the light rate, coincident with depletion of tissue carbohydrate reserves and a marked decline in root respiration. Changes also occurred in endogenous 15NO3 assimilation processes, which were distinctly different than those in 15NO3 uptake. During the extended dark period, translocation of absorbed 15N out of the root to the shoot varied rhythmically. The adjustments were independent of 15NO3 uptake rate and carbohydrate status, but were reciprocally related to rhythmic adjustments in stomatal resistance and, presumably, water movement through the root system. Whole plant reduction of 15NO3 always was limited more than uptake. The assimilation of 15N into insoluble reduced-N in roots remained a constant proportion of uptake throughout, while assimilation in the shoot declined markedly in the first 18 hours of darkness before stabilizing at a low level. The plants clearly retained a capacity for 15NO3 reduction and synthesis of insoluble reduced-15N even when 15NO3 uptake was severely restricted and minimal carbohydrate reserves remained in the tissue.  相似文献

Net uptakes of K+ and NO3 were monitored simultaneously and continuously for two barley (Hordeum vulgare) cultivars, Prato and Olli. The cultivars had similar rates of net K+ and NO3 uptake in the absence of NH4+ or Cl. Long-term exposure (over 6 hours) to media which contained equimolar mixtures of NH4+, K+, Cl, or NO3 affected the cultivars very differently: (a) the presence of NH4+ as NH4Cl stimulated net NO3 uptake in Prato barley but inhibited net NO3 uptake in Olli barley; (b) Cl inhibited net NO3 uptake in Prato but had little effect in Olli; and (c) NH4+ as (NH4)2SO4 inhibited net K+ uptake in Prato but had little effect in Olli. Moreover, the immediate response to the addition of an ion often varied significantly from the long-term response; for example, the addition of Cl initially inhibited net K+ uptake in Olli barley but, after a 4 hour exposure, it was stimulatory. For both cultivars, net NH4+ and Cl uptake did not change significantly with time after these ions were added to the nutrient medium. These data indicate that, even within one species, there is a high degree of genotypic variation in the control of nutrient absorption.  相似文献

Nitrogen-14 and nitrogen-15 nuclear magnetic resonance (NMR) spectra were recorded for freshly dissected buds of Picea glauca and for buds grown for 3, 6 and 9 weeks on shoot-forming medium. Resonances for Glu (and other αNH2 groups), Pro, Ala, and the side chain groups in Gln, Arg, Orn, and γ-aminobutyric acid could be detected in in vivo15N NMR spectra. Peaks for α-amino groups, Pro, NO3 and NH4+ could also be identified in 14N NMR spectra. Perfusion experiments performed for up to 20 hours in the NMR spectrometer showed that 15N-labeled NH4+ and NO3 are first incorporated into the amide group of Gln and then in the αNH2 pool. Subsequently, it also emerges in Ala and Arg. These data suggest that the glutamine synthetase/ glutamate synthase pathway functions under these conditions. The assimilation of NH4+ is much faster than that of NO3. Consequently after 10 days of growth more than 70% of the newly synthesized internal free amino acid pool derives its nitrogen from NH4+ rather than NO3. If NH4+ is omitted from the medium, no NO3 is taken up during 9 weeks and the buds support limited growth by utilizing their endogenous amino acid pools. It is concluded that NH4+ and NO3 are both required for the induction of nitrate- and nitrite reductase.  相似文献

Many reports have shown that plant growth and yield is superior on mixtures of NO3 and NH4+ compared with provision of either N source alone. Despite its clear practical importance, the nature of this N-source synergism at the cellular level is poorly understood. In the present study we have used the technique of compartmental analysis by efflux and the radiotracer 13N to measure cellular turnover kinetics, patterns of flux partitioning, and cytosolic pool sizes of both NO3 and NH4+ in seedling roots of rice (Oryza sativa L. cv IR72), supplied simultaneously with the two N sources. We show that plasma membrane fluxes for NH4+, cytosolic NH4+ accumulation, and NH4+ metabolism are enhanced by the presence of NO3, whereas NO3 fluxes, accumulation, and metabolism are strongly repressed by NH4+. However, net N acquisition and N translocation to the shoot with dual N-source provision are substantially larger than when NO3 or NH4+ is provided alone at identical N concentrations.  相似文献

Background and Aims

Orchid mycorrhizas exhibit a unique type of mycorrhizal symbiosis that occurs between fungi and plants of the family Orchidaceae. In general, the roots of orchids are typically coarse compared with those of other plant species, leading to a considerably low surface area to volume ratio. As a result, orchids are often ill-adapted for direct nutrient acquisition from the soil and so mycorrhizal assocaitions are important. However, the role of the fungal partners in the acquisition of inorganic and organic N by terrestrial orchids has yet to be clarified.

Methods

Inorganic and amino acid N uptake by non-mycorrhizal and mycorrhizal Cymbidium goeringii seedlings, which were grown in pots in a greenhouse, was investigated using a 15N-labelling technique in which the tracer was injected at two different soil depths, 2·5 cm or 7·5 cm. Mycorrhizal C. goeringii seedlings were obtained by inoculation with three different mycorrhizal strains isolated from the roots of wild terrestrial orchids (two C. goeringii and one C. sinense).

Key Results

Non-mycorrhizal C. goeringii primarily took up NO3 from tracers injected at 2·5-cm soil depth, whereas C. goeringii inoculated with all three mycorrhiza primarily took up NH4+ injected at the same depth. Inoculation of the mycorrhizal strain MLX102 (isolated from adult C. sinense) on C. goeringii roots only significantly increased the below-ground biomass of the C. goeringii; however, it enhanced 15NH4+ uptake by C. goeringii at 2·5-cm soil depth. Compared to the uptake of tracers injected at 2·5-cm soil depth, the MLX102 fungal strain strongly enhanced glycine-N uptake by C. goeringii from tracers injected at 7·5-cm soil depth. Cymbidium goeringii inoculated with CLB113 and MLX102 fungal strains demonstrated a similar N uptake pattern to tracers injected at 2·5-cm soil depth.

Conclusions

These findings demonstrate that mycorrhizal fungi are able to switch the primary N source uptake of a terrestrial orchid, in this case C. goeringii, from NO3 to NH4+. The reasons for variation in N uptake in the different soil layers may be due to possible differentiation in the mycorrhizal hyphae of the C. goeringii fungal partner.  相似文献

The effect of the exogenous and endogenous NO3 concentration on net uptake, influx, and efflux of NO3 and on nitrate reductase activity (NRA) in roots was studied in Phaseolus vulgaris L. cv. Witte Krombek. After exposure to NO3, an apparent induction period of about 6 hours occurred regardless of the exogenous NO3 level. A double reciprocal plot of the net uptake rate of induced plants versus exogenous NO3 concentration yielded four distinct phases, each with simple Michaelis-Menten kinetics, and separated by sharp breaks at about 45, 80, and 480 micromoles per cubic decimeter.

Influx was estimated as the accumulation of 15N after 1 hour exposure to 15NO3. The isotherms for influx and net uptake were similar and corresponded to those for alkali cations and Cl. Efflux of NO3 was a constant proportion of net uptake during initial NO3 supply and increased with exogenous NO3 concentration. No efflux occurred to a NO3-free medium.

The net uptake rate was negatively correlated with the NO3 content of roots. Nitrate efflux, but not influx, was influenced by endogenous NO3. Variations between experiments, e.g. in NO3 status, affected the values of Km and Vmax in the various concentration phases. The concentrations at which phase transitions occurred, however, were constant both for influx and net uptake. The findings corroborate the contention that separate sites are responsible for uptake and transitions between phases.

Beyond 100 micromoles per cubic decimeter, root NRA was not affected by exogenous NO3 indicating that NO3 uptake was not coupled to root NRA, at least not at high concentrations.

Aerobic and anaerobic groundwater continuous-flow microcosms were designed to study nitrate reduction by the indigenous bacteria in intact saturated soil cores from a sandy aquifer with a concentration of 3.8 mg of NO3-N liter−1. Traces of 15NO3 were added to filter-sterilized groundwater by using a Darcy flux of 4 cm day−1. Both assimilatory and dissimilatory reduction rates were estimated from analyses of 15N2, 15N2O, 15NH4+, and 15N-labeled protein amino acids by capillary gas chromatography-mass spectrometry. N2 and N2O were separated on a megabore fused-silica column and quantified by electron impact-selected ion monitoring. NO3 and NH4+ were analyzed as pentafluorobenzoyl amides by multiple-ion monitoring and protein amino acids as their N-heptafluorobutyryl isobutyl ester derivatives by negative ion-chemical ionization. The numbers of bacteria and their [methyl-3H]thymidine incorporation rates were simultaneously measured. Nitrate was completely reduced in the microcosms at a rate of about 250 ng g−1 day−1. Of this nitrate, 80 to 90% was converted by aerobic denitrification to N2, whereas only 35% was denitrified in the anaerobic microcosm, where more than 50% of NO3 was reduced to NH4+. Assimilatory reduction was recorded only in the aerobic microcosm, where N appeared in alanine in the cells. The nitrate reduction rates estimated for the aquifer material were low in comparison with rates in eutrophic lakes and coastal sediments but sufficiently high to remove nitrate from an uncontaminated aquifer of the kind examined in less than 1 month.  相似文献
